Abstract

In this article, the antibiofilm activity and physicochemical properties of graphene oxide (GO) nanoribbons, which have been among the most exciting materials, were studied by measuring the ratio of killed to alive bacteria incubated with these nanomaterials. Our objective was to determine the related physicochemical and antibiofilm properties of graphene oxide nanoribbons. We hypothesized that the physicochemical properties of graphene oxide nanoribbons could affect their antibiofilm activity. A combination of spectroscopic and microscopic measurements of the samples allowed us to determine their physicochemical properties affecting the biofilms. Our work includes information on the surface properties of these materials related to their incubation with the biofilms. The Fourier transform infrared spectroscopy showed the vibrations of OH groups of water molecules adsorbed on graphene oxide nanoribbons. The results show the high antibiofilm activity of the graphene oxide nanoribbons. The fluorescence confocal microscopy revealed that 50 % ± 3 % of the total number of bacteria were killed with these nanomaterials. The incubation of graphene oxide nanoribbons with bacterial biofilms resulted in the appearance of the NO2–, NO3– peaks in the negative mode mass spectrum. The attenuation of the O– and OH– peaks were attributed to the interactions of the samples with the biofilms. Our study gives more evidence of the practical value of graphene oxide nanoribbons in killing bacteria related to their surface physical properties and the potential of these nanomaterials for materials science and biomedical applications.

Abstract

Background: At the time of AtTEnd trial design, standard treatment for advanced or recurrent endometrial cancer included carboplatin and paclitaxel chemotherapy. This trial assessed whether combining atezolizumab with chemotherapy might improve outcomes in this population. Methods: AtTEnd was a multicentre, double-blind, randomised, placebo-controlled, phase 3 trial done in 89 hospitals in 11 countries across Europe, Australia, New Zealand, and Asia. Enrolled patients were aged 18 years or older, and had advanced or recurrent endometrial carcinoma or carcinosarcoma, an Eastern Cooperative Oncology Group performance status of 0–2, and received no previous systemic chemotherapy for recurrence. Patients were randomly assigned (2:1) using an interactive web response system (block size of six) to either atezolizumab 1200 mg or placebo given intravenously with chemotherapy (carboplatin at area under the curve of 5 or 6 and paclitaxel 175 mg/m2 intravenously on day 1 every 21 days) for 6–8 cycles, then continued until progression. Stratification factors were country, histological subtype, advanced or recurrent status, and mismatch repair (MMR) status. Participants and treating clinicians were masked to group allocation. The hierarchically tested co-primary endpoints were progression-free survival (in patients with MMR-deficient [dMMR] tumours, and in the overall population) and overall survival (in the overall population). Primary analyses were done in the intention-to-treat population, defined as all randomly assigned patients who gave their full consent to participation in the study and data processing. Safety was assessed in all patients included in the intention-to-treat population who received at least one dose of study treatment. Here, we report the primary progression-free survival and the interim overall survival results. This study is ongoing and is registered with ClinicalTrials.gov, NCT03603184. Abstract

B cells are frequently found in the margins of solid tumours as organized follicles in ectopic lymphoid organs called tertiary lymphoid structures (TLS)1,2. Although TLS have been found to correlate with improved patient survival and response to immune checkpoint blockade (ICB), the underlying mechanisms of this association remain elusive1,2. Here we investigate lung-resident B cell responses in patients from the TRACERx 421 (Tracking Non-Small-Cell Lung Cancer Evolution Through Therapy) and other lung cancer cohorts, and in a recently established immunogenic mouse model for lung adenocarcinoma3. We find that both human and mouse lung adenocarcinomas elicit local germinal centre responses and tumour-binding antibodies, and further identify endogenous retrovirus (ERV) envelope glycoproteins as a dominant anti-tumour antibody target. ERV-targeting B cell responses are amplified by ICB in both humans and mice, and by targeted inhibition of KRAS(G12C) in the mouse model. ERV-reactive antibodies exert anti-tumour activity that extends survival in the mouse model, and ERV expression predicts the outcome of ICB in human lung adenocarcinoma. Finally, we find that effective immunotherapy in the mouse model requires CXCL13-dependent TLS formation. Conversely, therapeutic CXCL13 treatment potentiates anti-tumour immunity and synergizes with ICB. Our findings provide a possible mechanistic basis for the association of TLS with immunotherapy response.

Abstract

Several vineyard techniques have been proposed to delay grape maturity in light of the advanced maturation driven by increasingly frequent water and heat stress events that are detrimental to grape quality. These studies differ in terms of their experimental conditions, and in the present work we have attempted to summarize previous observations in a quantitative, data-driven systematic review. A meta-analysis of quantitative data gathered across 43 relevant studies revealed the overall significance of the proposed treatments and evaluated the impact of different experimental conditions on the outcome of antitranspirants, delayed pruning and late source limitation. Antitranspirants were most effective when applied twice and closer to veraison, while di-1-p-menthene increased the ripening delay by about 1 °Brix compared to kaolin. Larger ripening delays were achieved with delayed pruning of low-yielding vines or by pruning at later stages of apical bud development. Late defoliation or shoot trimming delayed ripening in high-yielding vines and represent suitable solutions for late-harvested varieties, but became ineffective where the treatment decreased yield. This quantitative meta-analysis of 242 primary observations uncovers factors affecting the efficacy of vineyard practices to delay ripening, which should be carefully considered by grape growers attempting to achieve this outcome.

Abstract

In silico studies’ refers to using computer modelling and simulation to undertake virtual experiments (chips are largely made of silicon, hence the name). This report will focus on in silico technologies applied to the life sciences or healthcare to facilitate the discovery, development, optimisation or regulatory evaluation of medicines or healthcare products. In silico studies utilise computational modelling to simulate or predict cellular, molecular, or even subatomic interactions, such as DNA replication, protein folding, and RNA splicing. However, more recently, methods underpinned by very similar fundamental approaches have been used to model tissues, organs, full organisms and entire populations in health and disease. In silico differs from in vivo (meaning “in life”) and in vitro (“in glass”). In vivo refers to studies conducted on living organisms, while in vitro refers to studies conducted in test tubes or other laboratory settings outside a living organism. In silico approaches emerged to overcome limitations inherent to lab experimentation with living organisms, cells and specimens. Primarily, it reduces the need for extensive lab work and expensive clinical trials, accelerates regulatory approval, and allows experimentation to be done on a much faster and larger scale. In some form, these techniques have been used for around 30 years, often in combination with in vitro research methods. However, in silico approaches have increased over the last few years thanks to upgrades in computational power and innovations in machine learning and computational sciences. They are becoming an increasing part of research and innovation in life sciences and healthcare thanks to the increasing and vast availability of digital data. This report focuses on UK SMEs using in silico technologies to support a range of outcomes in the life sciences and medical technology sectors. These companies range from those using in silico technologies for drug discovery purposes to those using the technology to test medical devices and those are developing in silico technologies as their primary proposition. The data in the report pertains to 77 UK headquartered in silico companies that are either currently active, independent private companies or have historically been part of this category but have exited via acquisition or IPO or have ceased operations. The rapid development of COVID-19 vaccines shows how rapidly new and effective drugs can be created. In silico technologies promise to improve further and accelerate our ability to tackle disease and health problems with safer and more effective therapeutic practices, drugs, and medical devices., InnovateUK KTN and the Royal Academy of Engineering Chair in Emerging Technology (INSILEX CiET1819/19) funded this report. 31. Prediction models for individual-level healthcare costs associated with cardiovascular events in the UK

Abstract

Objectives: To develop prediction models for the individual-level impacts of cardiovascular events on UK healthcare costs. Methods: In the UK Biobank, people 40-70 years old, recruited in 2006-2010, were followed in linked primary (N=192 983 individuals) and hospital care (N=501 807 individuals) datasets. Primary and hospital care costs’ (2020 UK£) regression models of annual costs associated with individual characteristics and experiences of myocardial infarction (MI), stroke, coronary revascularization, incident diabetes and cancer, and vascular and nonvascular death are reported. Results: For both people without and with previous CVD, primary care costs were modelled using onepart generalised linear models (GLMs) with identity link and Poisson distribution, and hospital costs with two-part models (part 1: logistic regression models probability of incurring costs; part 2: GLM with identity link and Poisson distribution models costs conditional on incurring any). In people without previous CVD, mean annual primary and hospital care costs were £360 and £514. The excess primary care costs were £190 and £360 following MI and stroke, respectively, whereas excess hospital costs decreased from £4340 and £5590, respectively, in the year of these events, to £190 and £410 two years after. People with previous CVD had more than twice higher annual costs, and incurred higher excess costs for cardiovascular events. Other characteristics associated with higher costs included older age, female sex, south Asian ethnicity, higher socioeconomic deprivation, smoking, lower level of physical activities, unhealthy body mass index, and comorbidities. Conclusions: These individual-level healthcare cost prediction models could inform assessments of the value of health technologies and policies to reduce cardiovascular and other disease risks and healthcare costs. An accompanying Excel calculator is available to facilitate the use of the models.

36. An Online Learning Space Facilitating Supervision Pedagogies in Science

Abstract

Quality research supervision leading to timely completion and student satisfaction involves explicit pedagogy and effective communication. This article describes the development within an action research cycle of an online learning space designed to achieve these goals. The research "spirals" involved interventions in the form of instructive resources scaffolding best practice and discussion starters facilitating quality interactions. These were evaluated by focus groups and surveys of supervisors, postgraduate coordinators and research students. The instructive resources were valued for their interactivity, conciseness, clarification of research issues, and development of confidence, while the discussion starters were valued for their access ease, unpacking of expectations, encouragement of independent reflection, and enhancement of confidence in supervision relationships. Although the framework and pedagogies for effective supervision in Science have been well described, this online learning space is the first to address the interaction between supervisors and research students and provide resources for facilitating supervision pedagogy. Abstract

Environmental context: Having appropriate and robust models used for developing water quality guidelines is critical for sound environmental management. Methods used to validate models have only been demonstrated appropriate for a small portion of data types used in these models. This study has found that models using certain data types would be more appropriately validated using alternative evaluation criteria. This study serves as an important reference for developing and evaluating robust models. Rationale: Bioavailability-based toxicity models for metals often have performance assessed by whether it can predict toxicity data within a factor of 2 of their paired observed toxicity data. This method has only been verified for median effect values (EC50) for acute fish and daphnia data, however toxicity models have been developed for a much broader range of effect levels (i.e. EC10/EC20) and species (e.g. microalga). This study tested whether the factor-of-2 rule is appropriate for a wider range of organisms and effect concentrations than previously studied. Methodology: Toxicity estimate data from repeated tests conducted under the same conditions were collated to assess variation in results and compare this variation to a range of 4 (a factor of 2 above and below the mean) and a range of 9 (a factor of 3 above and below the mean) to assess if a factor-of-3 rule may be more appropriate for some species and effect levels. Results and discussion: Overall, the factor-of-2 rule is broadly applicable for metal toxicity to a range of species for EC50 data. The EC10 datasets highlighted that larger variability exists in low effect levels and supported the use of a factor-of-3 rule, while the either the factor-of-2 or factor-of-3 rule could be applied to microalgae. The level of performance evaluation chosen may depend on the application of the bioavailability model. Abstract

One of the most concerning trends associated with increasing heat and water stress is advanced ripening of grapes, leading to harvesting fruit that is high in sugar concentrations but lacks optimal phenolic (i.e. color and mouthfeel) and aromatic maturity. Mitigation techniques for this phenomenon have been researcherd for the past decades and practices to delay sugar accumulation have been identified, including antitranspirants, delayed pruning and late source limitation techniques. Evaluation of the efficacy of these vineyard practices has occurred in a wide range of environments, vintages, varieties and growing conditions. To assess the efficacy of these three vineyard practices, which are easy-to-implement and cost-effective, a meta-analytic approach was adopted using data retrieved from 43 original studies. Statistical techniques of meta-analysis were applied to the data collected to evaluate the efficacy of antitranspirants, late pruning and late source limitation techniques.
