Abstract: BACKGROUND AND AIMS: Alagille syndrome (ALGS) is characterized by chronic cholestasis with associated pruritus and extrahepatic anomalies. Maralixibat, an ileal bile acid transporter inhibitor, is an approved pharmacologic therapy for cholestatic pruritus in ALGS. Since long-term placebo-controlled studies are not feasible or ethical in children with rare diseases, a novel approach was taken comparing 6-year outcomes from maralixibat trials with an aligned and harmonized natural history cohort from the G lobal AL agille A lliance (GALA) study. APPROACH AND RESULTS: Maralixibat trials comprise 84 patients with ALGS with up to 6 years of treatment. GALA contains retrospective data from 1438 participants. GALA was filtered to align with key maralixibat eligibility criteria, yielding 469 participants. Serum bile acids could not be included in the GALA filtering criteria as these are not routinely performed in clinical practice. Index time was determined through maximum likelihood estimation in an effort to align the disease severity between the two cohorts with the initiation of maralixibat. Event-free survival, defined as the time to first event of manifestations of portal hypertension (variceal bleeding, ascites requiring therapy), surgical biliary diversion, liver transplant, or death, was analyzed by Cox proportional hazards methods. Sensitivity analyses and adjustments for covariates were applied. Age, total bilirubin, gamma-glutamyl transferase, and alanine aminotransferase were balanced between groups with no statistical differences. Event-free survival in the maralixibat cohort was significantly better than the GALA cohort (HR, 0.305; 95% CI, 0.189-0.491; p <0.0001). Multiple sensitivity and subgroup analyses (including serum bile acid availability) showed similar findings. Abstract: Wir Menschen sind auf funktionierende und widerstandsfähige Ökosysteme angewiesen. Der Zustand der Natur in Deutschland hat sich in den letzten Jahrzehnten weiter verschlechtert. Ergänzend zum Schutz der verbliebenen Natur sollte stärker als bisher der Zustand geschädigter Ökosysteme verbessert werden. SRU, WBBGR und WBW geben mit dieser Stellungnahme Empfehlungen für eine wirksame Renaturierungspolitik in Deutschland. Am 23. Mai 2024 wurden die Kernempfehlungen der Stellungnahme in einer öffentlichen Online-Veranstaltung präsentiert und diskutiert. Die Dokumentation ist hier abrufbar. Das Anliegen, die Erholung eines degradierten Ökosystems zu unterstützen, ist weit zu verstehen und umfasst neben der Verbesserung des Zustands von Schutzgebieten auch naturverträgliche Formen der Landnutzung. Ökosysteme sollen sich durch Renaturierung in Richtung naturnäherer Strukturen entwickeln, sodass sie langfristig vielfältige Leistungen erbringen können. Wie andere Vorhaben benötigen Renaturierungsmaßnahmen Flächen. Ökosysteme wiederherzustellen wird daher auch Konflikte um Flächennutzung, wirtschaftliche Aspekte und unser Konsumverhalten mit sich bringen. Die es zu moderieren und zu entscheiden gilt. Renaturierung ist eine gesellschaftliche Aufgabe, die nicht nur den Naturschutz betrifft, sondern vor allem auch die landnutzenden Sektoren, neben der Stadt- und Regionalplanung insbesondere die Landwirtschaft und die Waldbewirtschaftung. Daher zielt diese Stellungnahme darauf ab, Grundzüge einer Renaturierungspolitik zu entwickeln, die Synergien zwischen Naturschutz und Landnutzungsinteressen erzeugt sowie Zielkonflikte minimiert.

Abstract: Organismal functional strategies form a continuum from slow- to fast-growing organisms, in response to common drivers such as resource availability and disturbance. However, whether there is synchronisation of these strategies at the entire community level is unclear. Here, we combine trait data for >2800 above- and belowground taxa from 14 trophic guilds spanning a disturbance and resource availability gradient in German grasslands. The results indicate that most guilds consistently respond to these drivers through both direct and trophically mediated effects, resulting in a ‘slow-fast’ axis at the level of the entire community. Using 15 indicators of carbon and nutrient fluxes, biomass production and decomposition, we also show that fast trait communities are associated with faster rates of ecosystem functioning. These findings demonstrate that ‘slow’ and ‘fast’ strategies can be manifested at the level of whole communities, opening new avenues of ecosystem-level functional classification.

Abstract: Background and Aims: Alagille syndrome (ALGS) is a multisystem disorder, characterized by cholestasis. Existing outcome data are largely derived from tertiary centers, and real-world data are lacking. This study aimed to elucidate the natural history of liver disease in a contemporary, international cohort of children with ALGS. Approach and Results: This was a multicenter retrospective study of children with a clinically and/or genetically confirmed ALGS diagnosis, born between January 1997 and August 2019. Native liver survival (NLS) and event-free survival rates were assessed. Cox models were constructed to identify early biochemical predictors of clinically evident portal hypertension (CEPH) and NLS. In total, 1433 children (57% male) from 67 centers in 29 countries were included. The 10 and 18-year NLS rates were 54.4% and 40.3%. By 10 and 18 years, 51.5% and 66.0% of children with ALGS experienced ≥1 adverse liver-related event (CEPH, transplant, or death). Children (>6 and ≤12 months) with median total bilirubin (TB) levels between ≥5.0 and <10.0 mg/dl had a 4.1-fold (95% confidence interval [CI], 1.6-10.8), and those ≥10.0 mg/dl had an 8.0-fold (95% CI, 3.4-18.4) increased risk of developing CEPH compared with those <5.0 mg/dl. Median TB levels between ≥5.0 and <10.0 mg/dl and >10.0 mg/dl were associated with a 4.8 (95% CI, 2.4-9.7) and 15.6 (95% CI, 8.7-28.2) increased risk of transplantation relative to <5.0 mg/dl. Median TB <5.0 mg/dl were associated with higher NLS rates relative to ≥5.0 mg/dl, with 79% reaching adulthood with native liver (p < 0.001). Conclusions: In this large international cohort of ALGS, only 40.3% of children reach adulthood with their native liver. A TB <5.0 mg/dl between 6 and 12 months of age is associated with better hepatic outcomes. These thresholds provide clinicians with an objective tool to assist with clinical decision-making and in the evaluation of therapies.

Abstract: Background: Chemotherapy crosses the placenta, however, it remains unclear to what extent it affects fetal growth. The current literature suggests up to 21% of the offspring of women receiving chemotherapy are small for gestational age (SGA, birth weight <10th percentile). Limiting research to birth weights only might misjudge fetal growth restriction (FGR) in this high-risk population with multiple risk factors for impaired fetal growth. Moreover, the role of the duration of chemotherapy and gestational age at initiation of chemotherapy in fetal growth is yet poorly understood. Objective: This retrospective cohort study evaluates fetal growth and neonatal birthweights in pregnant women receiving chemotherapy. Study design: All pregnant patients, registered by the International Network of Cancer, Infertility and Pregnancy (INCIP), treated with chemotherapy with at least two ultrasounds reporting on fetal growth, were eligible for this study. Duration and gestational age at initiation of chemotherapy were our major determinants, followed by cancer type and stage, maternal characteristics (parity, BMI, ethnicity hypertension, and diabetes) and individual cytotoxic agents (anthracycline, taxanes, and platinum). Fetal growth outcomes were described using the following mutually exclusive groups (1) FGR, based on a Delphi consensus (2016); (2) “low risk SGA” (birth weight below the 10th percentile), but an estimated growth above the 10th percentile; (3) “fetal growth disturbance”, which did not meet all FGR criteria; (4) “non-FGR”. Obstetric and oncological characteristics were compared between the growth impaired groups and non-FGR group. The aim of this study was to identify if chemotherapy affects the maternal ovaries during pregnancy and whether these effects depend on type of chemotherapy and duration of exposure. METHODS: Pregnant 8-week-old female BL6 mice were exposed to 6 different single chemotherapeutic agents (carboplatin, cisplatin, paclitaxel, epirubicin, doxorubicin, or cyclophosphamide) or saline at gestational day (GD) 13.5. The mice were sacrificed at GD 15.5 or GD 18.5. Ovaries were assessed by histopathology and immunohistochemistry. Follicle count was determined per follicle stage and per treatment modality. RESULTS: Maternal ovarian damage was demonstrated by the presence of apoptosis and necrosis in preantral follicles. The extent of this damage depends upon type of chemotherapy and duration of exposure (2 or 5 days). After short exposure, 81% of ovaries showed histopathologic signs of damage compared to 36% after long exposure, which might suggest a transient effect. Loss of primordial follicles (PMFs) was observed after both short and long exposure, with a reduction of more than 70%. Evidence of DNA damage, as demonstrated by phospho-H2AX expression, was present in 23% (range 0-89%) of PMFs exposed to chemotherapy, but only in the short exposure group. Overall, the least damage was seen after administration of paclitaxel. CONCLUSION: Despite physiological ovarian function suppression during gestation, chemotherapy-induced damage of the ovaries occurs in pregnant mouse models, potentially affecting future fertility.

Abstract: Across the tree of life, organismal functional strategies form a continuum from slow- to fast-growing organisms, in response to common drivers such as resource availability and disturbance. However, the synchronization of these strategies at the entire community level is untested. We combine trait data for >2800 above- and belowground taxa from 14 trophic guilds spanning a disturbance and resource availability gradient in German grasslands. Most guilds consistently respond to these drivers through both direct and trophically-mediated effects, resulting in a "slow-fast" axis at the level of the entire community. Fast trait communities were also associated with faster rates of whole ecosystem functioning. These findings demonstrate that "slow" and "fast" strategies can be manifested at the level of whole ecosystems, opening new avenues of ecosystem-level functional classification.

Abstract: Ecological intensification has been embraced with great interest by the academic sector but is still rarely taken up by farmers because monitoring the state of different ecological functions is not straightforward. Modelling tools can represent a more accessible alternative of measuring ecological functions, which could help promote their use amongst farmers and other decision-makers. In the case of crop pollination, modelling has traditionally followed either a mechanistic or a data-driven approach. Mechanistic models simulate the habitat preferences and foraging behaviour of pollinators, while data-driven models associate georeferenced variables with real observations. Here, we test these two approaches to predict pollination supply and validate these predictions using data from a newly released global dataset on pollinator visitation rates to different crops. We use one of the most extensively used models for the mechanistic approach, while for the data-driven approach, we select from among a comprehensive set of state-of-The-Art machine-learning models. Moreover, we explore a mixed approach, where data-derived inputs, rather than expert assessment, inform the mechanistic model. We find that, at a global scale, machine-learning models work best, offering a rank correlation coefficient between predictions and observations of pollinator visitation rates of 0.56. In turn, the mechanistic model works moderately well at a global scale for wild bees other than bumblebees. Biomes characterized by temperate or Mediterranean forests show a better agreement between mechanistic model predictions and observations, probably due to more comprehensive ecological knowledge and therefore better parameterization of input variables for these biomes. Abstract: Seventy five percent of the world's food crops benefit from insect pollination. Hence, there has been increased interest in how global change drivers impact this critical ecosystem service. Because standardized data on crop pollination are rarely available, we are limited in our capacity to understand the variation in pollination benefits to crop yield, as well as to anticipate changes in this service, develop predictions, and inform management actions. Here, we present CropPol, a dynamic, open, and global database on crop pollination. It contains measurements recorded from 202 crop studies, covering 3,394 field observations, 2,552 yield measurements (i.e., berry mass, number of fruits, and fruit density [kg/ha], among others), and 47,752 insect records from 48 commercial crops distributed around the globe. CropPol comprises 32 of the 87 leading global crops and commodities that are pollinator dependent. Malus domestica is the most represented crop (32 studies), followed by Brassica napus (22 studies), Vaccinium corymbosum (13 studies), and Citrullus lanatus (12 studies). The most abundant pollinator guilds recorded are honey bees (34.22% counts), bumblebees (19.19%), flies other than Syrphidae and Bombyliidae (13.18%), other wild bees (13.13%), beetles (10.97%), Syrphidae (4.87%), and Bombyliidae (0.05%). Der Beitrag diskutiert Möglichkeiten zur Umsetzung dieser Regelung im Hinblick auf die Verwendung von Wildpflanzensaatgut (unter Ausklammerung der Gehölze) mit besonderer Berücksichtigung faunistischer Aspekte. Er entstand aus einem Expertinnen- und Expertenworkshop mit dem Ziel, eine reflektierte interdisziplinäre Debatte zur Berücksichtigung genetischer Vielfalt anzustoßen, welche die Auswirkungen für die Naturschutzpraxis aus ganzheitlicher Sicht beleuchtet. Es wird deutlich, dass bestehende Regelungen auf großen Unsicherheiten fußen und massive Einschränkungen für die Wirksamkeit von Renaturierungen und die Erreichbarkeit übergeordneter naturschutzfachlicher Ziele mit sich bringen. Das betrifft besonders die ausgeklammerte Relevanz von Phytodiversität für das Vorkommen artenreicher Tiergemeinschaften und damit für möglichst vollständige Biozönosen sowie die Renaturierung von Biotopen. Abstract: Background: Data on the long-term effects of prenatal exposure to maternal cancer and its treatment on child development are scarce. Methods: In a multicenter cohort study, the neurologic and cardiac outcomes of 6-year-old children born to women diagnosed with cancer during pregnancy were compared with the outcome of children born after an uncomplicated pregnancy. Assessment included clinical evaluation, comprehensive neuropsychological testing, electrocardiography and echocardiography. Results: In total, 132 study children and 132 controls were included. In the study group, 97 children (73.5%) were prenatally exposed to chemotherapy (alone or in combination with other treatments), 14 (10.6%) to radiotherapy (alone or in combination), 1 (0.8%) to trastuzumab, 12 (9.1%) to surgery alone and 16 (12.1%) to no treatment. Although within normal ranges, statistically significant differences were found in mean verbal IQ and visuospatial long-term memory, with lower scores in the study versus control group (98.1, 95% confidence interval [CI]: 94.5–101.8, versus 104.4, 95% CI: 100.4–108.4, P = 0.001, Q < 0.001 [Q refers to the false discovery rate adjusted P value], and 3.9, 95% CI: 3.6–4.3, versus 4.5, 95% CI: 4.1–4.9, P = 0.005, Q = 0.045, respectively). A significant difference in diastolic blood pressure was found, with higher values in chemotherapy-exposed (61.1, 95% CI: 59.0 to 63.2) versus control children (56.0, 95% CI 54.1 to 57.8) (P < 0.001, Q < 0.001) and in a subgroup of 59 anthracycline-exposed (61.8, 95% CI: 59.3 to 64.4) versus control children (55.9, 95% CI: 53.6 to 58.1) (P < 0.001, Q = 0.02). Conclusions: Children prenatally exposed to maternal cancer and its treatment are at risk for lower verbal IQ and visuospatial long-term memory scores and for higher diastolic blood pressure, but other cognitive functions and cardiac outcomes were normal at the age of 6 years. Abstract: Land-use intensification is a major driver of biodiversity loss. However, understanding how different components of land use drive biodiversity loss requires the investigation of multiple trophic levels across spatial scales. Using data from 150 agricultural grasslands in central Europe, we assess the influence of multiple components of local- and landscape-level land use on more than 4,000 above- and belowground taxa, spanning 20 trophic groups. Plot-level land-use intensity is strongly and negatively associated with aboveground trophic groups, but positively or not associated with belowground trophic groups. Meanwhile, both above- and belowground trophic groups respond to landscape-level land use, but to different drivers: aboveground diversity of grasslands is promoted by diverse surrounding land-cover, while belowground diversity is positively related to a high permanent forest cover in the surrounding landscape. These results highlight a role of landscape-level land use in shaping belowground communities, and suggest that revised agroecosystem management strategies are needed to conserve whole-ecosystem biodiversity.

Abstract: As the incidence of cancer in pregnancy has been increasing in recent decades, more specialists are confronted with a complex oncologic-obstetric decision-making process. With the establishment of (inter)national registries, including the International Network on Cancer, Infertility and Pregnancy, and an increasing number of smaller cohort studies, more evidence on the management of cancer during pregnancy is available. As fetal, neonatal, and short-term pediatric outcomes after cancer treatment are reassuring, more women receive treatment during pregnancy. Prenatal treatment should adhere to standard treatment as much as possible to optimize maternal prognosis, always taking into account fetal well-being. In order to guarantee the optimal treatment for both mother and child, a multidisciplinary team of specialists with expertise should be involved. Apart from oncologic treatment, a well-considered obstetric and perinatal management plan discussed with the future parents is crucial. Results of non-invasive prenatal testing are inconclusive in women with cancer and alternatives for prenatal anomaly screening should be used. Especially in women treated with chemotherapy, serial ultrasounds are strongly recommended to follow-up fetal growth and cervical length. After birth, a neonatal assessment allows the identification of any cancer or treatment-related adverse events. In addition, placental histologic examination aims to assess the fetal risk of metastasis, especially in women with malignant melanoma or metastatic disease. Breastfeeding is discouraged when systemic treatment needs to be continued after birth. At least a 3-week interval between the last treatment and nursing is recommended to prevent any treatment-induced neonatal effects from most non-platinum chemotherapeutic agents.

Abstract: Soil organisms, including earthworms, are a key component of terrestrial ecosystems. However, little is known about their diversity, their distribution, and the threats affecting them. We compiled a global dataset of sampled earthworm communities from 6928 sites in 57 countries as a basis for predicting patterns in earthworm diversity, abundance, and biomass. We found that local species richness and abundance typically peaked at higher latitudes, displaying patterns opposite to those observed in aboveground organisms. However, high species dissimilarity across tropical locations may cause diversity across the entirety of the tropics to be higher than elsewhere. Climate variables were found to be more important in shaping earthworm communities than soil properties or habitat cover. The PREDICTS project—Projecting Responses of Ecological Diversity In Changing Terrestrial Systems (www.predicts.org.uk)—has collated from published studies a large, reasonably representative database of comparable samples of biodiversity from multiple sites that differ in the nature or intensity of human impacts relating to land use. We have used this evidence base to develop global and regional statistical models of how local biodiversity responds to these measures. We describe and make freely available this 2016 release of the database, containing more than 3.2 million records sampled at over 26,000 locations and representing over 47,000 species. We outline how the database can help in answering a range of questions in ecology and conservation biology. To our knowledge, this is the largest and most geographically and taxonomically representative database of spatial comparisons of biodiversity that has been collated to date; it will be useful to researchers and international efforts wishing to model and understand the global status of biodiversity.

48. Compensatory mechanisms of litter decomposition under alternating moisture regimes in tropical rice fields

A conversion from permanently flooded rice cropping systems to crop rotations that include non-flooded upland crops is heavily promoted in tropical regions to meet the challenges of sustainable food production while reducing water consumption and trace gas emissions. Shorter periods of flooding and manipulation of fertilizer inputs affect belowground community composition, biomass and functioning. However, there have been no previous studies of how such shifts in rice field management might affect soil biota and decomposition. Our objective was to examine how crop diversification, which demands different moisture regimes and nitrogen inputs, influences belowground invertebrate assemblages and their contribution to the decomposition of crop residues.We conducted a litterbag experiment in lowland paddy fields at the experimental field sites of the DFG-ICON project (Laguna, the Philippines) that were either continuously flooded, had seasonally alternating wet and dry periods, or were continuously dry. Additionally, subplot treatments within these crop rotations included different N fertilizer management practices. At a total of 36 plots we used litterbags with two different mesh sizes to assess decomposition with and without fauna over a period of 72 days. Furthermore, we sampled soil microfauna and mesofauna in both the wet (rainy) and dry seasons.Although we found no correlation between faunal abundance and the contribution by invertebrates to rice straw decomposition, we found that soil water content was the decisive factor determining the activity of decomposer invertebrates as well as the composition and abundance of the soil fauna in fields with alternating cropping regimes. The impact of invertebrates on rice straw decomposition was higher under anaerobic than aerobic soil conditions which compensated for reduced microbial decay rates during periods of flooding. In contrast, microbial decomposition rates were higher under aerobic conditions, whereas invertebrates had no apparent effects on the mass loss of rice straw despite their higher abundance in dry fields. Our results demonstrate that invertebrates are essential for the effective decay of rice straw residues under flooded soil conditions, and therefore play an important role in supplying nutrients to flooded rice.

49. An individual-based simulation model to link population, community and metacommunity dynamics

Abstract: Introduction The dynamical behavior of individuals in ecosystems involves a multifaceted set of interaction types and processes that take place at different hierarchical levels. We present an individual-based, stochastic model that considers species dynamics at three hierarchical levels: population, community, and metacommunity. We use an individual-based model to show how the consequences of mechanisms that are specific to each hierarchical level may interact with processes that belong to other hierarchical levels. The strength of these effects is quantified in terms of impacts on metapopulation sizes and spatial distribution of populations. Results indicate the following: (1) the cohesion of the social network structure among conspecific individuals heavily affects their feeding efficiency at food-web level; (2) more generalist feeding habits trigger homogeneous spatial distribution of species at the landscape scale; and (3) high frequency of migration movements limits the local success of a generalist species thus leading to small metapopulation sizes. We illustrate how such a hierarchical framework may contribute to understanding the emergence of macroscopic patterns (i.e., metapopulation size and spatial heterogeneity) starting from elementary, bottom–up rules defined at the individual level. Abstract: Individual-Based Modeling in Conservation Biology A major challenge for food-web research is studying diversity and variability more explicitly. This means a focus on individual-level variability in populations (Bolnick et al., 2011) that hopefully might help to better understand how structural properties predict dynamical behavior (Dunne, 2006). One reason why linking structure to dynamics is still a hard challenge can be that intrapopulation variability is relatively poorly considered in most models. Yet defining developmental stages as graph nodes is a step toward managing this challenge: for example, in many food-web models certain species are represented by separate graph nodes that include juveniles and adults. Trophic status and network dynamics can be quite sensitive to this kind of demographic aggregation (or resolution) of the web and are expected to be influenced by individual-level differences in terms of behavior and feeding habits. Individual-level variability includes genetic, demographic, and stochastic factors and to date it is not easy to incorporate all these in most modeling frameworks. Yet developing the methodological background of individual-based modeling seems to be very useful for future research and applications. Since individual-level differences are more important in smaller populations (Lande, 1988), studying their effects explicitly is relevant for conservation efforts. Using network metrics as proxies or predictors of food-web dynamics is an old but still open issue. For an example, network hubs are supposed to be species of key importance and hubbish networks are thought to be safe against errors but vulnerable against attacks (Montoya and Solé, 2002). We make structural predictions routinely but we are very poor in testing these on either real-time series or simulation models.
