Abstract

Transient delivery of CRISPR-Cas9 ribonucleoproteins (RNPs) into the central nervous system (CNS) for therapeutic genome editing could avoid limitations of viral vector-based delivery including cargo capacity, immunogenicity, and cost. Here, we tested the ability of cell-penetrant Cas9 RNPs to edit the mouse striatum when introduced using a convection-enhanced delivery system. These transient Cas9 RNPs showed comparable editing of neurons and reduced adaptive immune responses relative to one formulation of Cas9 delivered using AAV serotype 9. The production of ultra-low endotoxin Cas9 protein manufactured at scale further improved innate immunity. We conclude that injection-based delivery of minimally immunogenic CRISPR genome editing RNPs into the CNS provides a valuable alternative to virus-mediated genome editing.

Abstract

Surface texturing is a highly effective method for enhancing the tribological performance of engineering materials. Laser surface texturing (LST) has generated significant interest owing to its exceptional flexibility, excellent texturing precision, and effective controllability. This study examines the surface characteristics of two polymers, PA66 and PA66 reinforced with glass microspheres, after the laser surface texturing. The samples to be textured were obtained by injection moulding. According to microscopic surface analysis, the laser texturing technique produces uniform and controllable surfaces characterized by well-defined patterns, and the material behavior is influenced by factors such as the number of passes and the thermal distribution during the process. Microindentation analysis reveals that material structure, pattern type and number of passes significantly influence surface hardness and penetration depth. The addition of glass microspheres improves the hardness, and the tests demonstrate the reliability of the methodology and its relevance for optimizing the texturization of polymer surfaces. Scratch tests highlight variations in A-COF depending on material, pattern and number of passes. PA6.6 and reinforced PA6.6 react differently, indicating influences on roughness and hardness. Abstract

Die Dissertation untersucht zeitgenössische Wohnhäuser in Osttibet und bietet neue Einblicke in die moderne Wohnwelt der tibetischen Bevölkerung. Die Arbeit zeigt den Entwicklungsstand der Wohnhausarchitekturen in den regionalen Einheiten Dege, Kandze, Drango und Nyarong im Bezirk Ganzi der chinesischen Provinz Sichuan auf und diskutiert anhand von präsentierten Beispielen Wandel und Kontinuität in der osttibetischen Baukunst. Die Leitfrage der Untersuchung lautet: Wie wohnen die Tibeter heute und worauf kommt es ihnen an? Durch die Analyse von überwiegend in den 2010er Jahren errichteten Wohnhäusern sowie einigen Beispielen aus den 1990er oder 1980er Jahren wurde die Veränderung und Kontinuität verdeutlicht. Die Dissertation zeigt, dass die intensive Bauaktivität in dem untersuchten Teil der osttibetischen Kulturregion von Sichuan in der letzten Dekade dazu geführt hat, dass ein Großteil der Wohnhäuser der Bauern durch Neubauten ersetzt wurde, wobei die lokale Bevölkerung weiterhin primär mit den traditionellen Baumaterialien Lehm und Holz baute. Im Kontrast zu anderen Regionen Tibets hatten die Bauern im untersuchten Gebiet uneingeschränkte Gestaltungsfreiheit und konnten ihre Häuser gemäß ihren Wünschen und Bedürfnissen selbstbestimmt gestalten. Die Gestaltung und Ausführung der Bauten wurden nicht von gesetzlichen Regelungen und Vorschriften beeinflusst, sondern überwiegend von den Bauern selbst bestimmt. Die Arbeit bietet somit Einblicke in eine Wohnhausarchitektur, die von den Anforderungen und Bedürfnissen der Eigentümer geprägt ist und in der das lokale Handwerk und die vernakulare Architektur bis ins 21. Jahrhundert ungehindert weiterentwickelt werden konnten., The dissertation investigates contemporary residential buildings in eastern Tibet and provides new insights into the modern living environment of the Tibetan population. The work presents the state of development of residential architecture in the regional units of Dege, Kandze, Drango, and Nyarong in Ganzi Prefecture of China's Sichuan Province and discusses the changes and continuity in eastern Tibetan architecture based on presented examples. The main question of the study is: How do Tibetans live today and what is important to them? By analyzing mainly residential buildings built in the 2010s as well as some from the 1990s or 1980s, the dissertation highlights the changes and continuity in residential architecture. The study shows that the intensive construction activity in the investigated part of the eastern Tibetan cultural region of Sichuan in the last decade has led to the replacement of a large proportion of rural dwellings with new buildings, with the local population continuing to primarily use traditional building materials such as mud and wood. In contrast to other regions in Tibet, the farmers in the studied area had unrestricted creative freedom and were able to design their homes according to their wishes and needs. The design and execution of the buildings were not influenced by legal regulations and requirements but were primarily determined by the farmers themselves. This work provides insights into a residential architecture shaped by the requirements and needs of the owners and in which local craftsmanship and vernacular architecture could continue to develop freely into the 21st century.

Abstract

Poly(dimethyl siloxane) (PDMS) is one of the most widely used materials in the biomedical field. Despite its numerous advantages, its hydrophobic character promotes bacterial adhesion and biofilm formation. For breast implants, biocompatibility is challenged due to the biofilm formed around the implant that can degenerate to severe capsular contracture over time. Thus, the laboratory has set up strategies to prevent bacterial contamination by grafting covalently hydrophilic bioactive polymers on the surface of implants. In this study, poly(methacrylic acid) (PMAc) and poly(acrylic acid) (PAAc) were chosen as non-toxic and biocompatible bioactive polymers known for reducing bacteria adhesion. These polymers are also good candidates to lend reactivity on the surface for further functionalization. X-ray photoelectron Spectroscopy (XPS) and Fourier-Transform Infrared spectroscopy (FTIR) analysis have highlighted the covalent grafting of these polymers. Apparent water contact angle measurements have shown the change in hydrophilicity on the surface, and a colorimetric assay allowed us to assess the grafting rate of PMAc and PAAc. Tensile strength assays were performed to ensure that the functionalization process does not significantly alter the material's mechanical properties. Analyses of the surface aspect and roughness by Scanning Electron Microscope (SEM) and optical profilometer allow us to formulate hypotheses to approach the understanding of the behavior of the polymer once grafted. 45. Managerial Intuition across Cultures: Beyond a 'West-East Dichotomy'

Abstract

Purpose: Studies investigating intuition from a cultural and cross-cultural perspective have a long tradition in various disciplines but, due to the increased internationalization of business, an understanding of the mental lives of other cultures became one of the priorities of management practitioners and theoreticians. Cultures of Anglo-American and East Asian origins have drawn particular attention. However, an analysis of management research studies and those in other disciplines shows that the former do not back up the "intuitive East versus analytical West" pattern set by the latter. The article aims at discussing this discrepancy, its origins as well as the implication for the managerial practice. Design/methodology/approach: The article provides an overview of the various concepts of intuition and methods of measuring it across cultures. It seeks to understand how various epistemological assumptions incorporated in the research affected the results obtained. An aspect under particular scrutiny in the article is the concept of cognitive style, which is employed frequently in management studies. Findings: One explanation for inconsistent results is a premature effort of management science to test a hypothesis generated by other disciplines, applying different conceptualizations, definitions, methodologies as well as populations. The conclusion is that the cross-cultural research in management is in need of a more exploratory research, paying attention to the specificity of management context and local knowledge. Research limitations/implications: The article proposes that a methodology that potentially has a capacity to meet these requirements is the narrative approach. Possible implications of this observation are discussed in the context of multicultural teams management practice. Originality/value: The article provides useful information on managerial intuition and methods of measuring it across cultures.
