Background: For decades, Wrightia tinctoria (W. tinctoria) has been important in the field of traditional medicine. The identification and isolation of numerous chemically potent and pharmaceutically advantageous phytochemicals isolated from this plant extract has contributed to its resurrection into evidence-based modern medicine. This plant is inexpensive and indigenous to the Indian subcontinent. Materials and Method: An in vitro study was carried out using KB (KERATIN-forming tumour cell line HeLa) oral cancer cell line. The control drug doxorubicin along with the test drug W. tinctoria was tested for anti-cancer potential by using MTT (3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide) assay and for apoptotic activity by using DNA fragmentation assay and western blotting assay. Results: The study was conducted using MTT assay to determine the cytotoxicity of plant extract against oral cancer cell line by determining the IC50 values at the end of 24 hours by the MTT calorimetric assay. The IC50 value of 48.89 and 9.62 was arrived for the W. tinctoria extract and doxorubicin, respectively, by using MTT calorimetric assay. DNA fragmentation assay showed a ladder pattern when treated with 59 µg/mL extract of W. tinctoria, and western blot analysis revealed the presence of cleaved caspase 3, confirming the apoptotic potential of the extract. Conclusion: The current study demonstrated that the plant extract has effective anti-cancer and apoptotic potential and can be used as a natural source for the production of new anti-cancer and cytotoxic agents against oral cancer. A small peak at 425 nm suggested the presence of silver nanoparticles. In a later part of the study, the inhibitory effect of green synthesized silver nanoparticles on the growth of E. coli and S. aureus was monitored. W. tinctoria, an Indian herb Indrajao, has significant therapeutic potential. While studies have highlighted the anti-inflammatory potential of the leaves and bark of this plant, similar efficacy of the seed-pods remains unexplored. We demonstrate significant anti-inflammatory effects of the hexane fraction (Fr-B) of ethyl acetate extract of the seedpods in reducing lipopolysaccharide and palmitate mediated inflammation in RAW264.7 macrophages and HepG2 cells. GC-MS and NMR profiling of Fr-B revealed the existence of hexadecanoic acid, ethyl hexadecanoate, 9,12-octadecanoic acid, 9,12,15-octadecatrienoic acid, 9,12,15-octadecatrienoic acid ethyl ester, ethyl linoleate and octadecanoic acid ethyl esters. Natural fibers derived from cellulose and ligno-celluloses materials have many advantages, such as being renewable, low density, inexhaustible, and cheap rather than synthetic fibers. Researchers and scientists are searching for a new fiber source that can be processed environmentally sustainable. The aim is to produce an organic and Eco-friendly product. The present investigation aims to extract and characterize ligno-cellulosic fiber from the seedpod of Wrightia tinctoria (WT) and Cebia pentandra (CP) plants. The extraction of WT fibers (WTFs) and CP fibers (CPFs) was carried out using the hand-stripping method. The structural and functional Characterization of WTFs and CPFs were determined using Scanning Electron Microscope (SEM), Fourier Transform Infrared (FT-IR) spectroscopy, Chemical analysis, X-ray diffraction studies (XRD), and the thermal behavior of fibers determined by using Thermo Gravimetric Analysis (TGA). The results indicated that WTFs composed of 75% cellulose, 14% lignin, and 0.55% wax content were, as the CPFs were composed of 38% cellulose, 15% lignin, and wax content of 2.34%. The SEM micrograph confirms that both fibers were hollow structures with thin cell walls and luminous because of the wax content presence on the surface of the fiber. The crystallinity percentage of WTFs and CPFs was calculated from XRD studies and is valued at 62% and 52%. Thermo gravimetric analysis revealed that WTFs and CPFs were thermally stable up to 460°C and 350°C. Concentrations of heavy metals (Cr, Cu, Fe, Mn, Ni, Pb, and Zn) accumulation were studied in the leaves of two medicinal plant species, namely Holarrhena pubescens and Wrightia tinctoria, from two industrial areas and a control area. Our comparison study revealed that industrialization significantly increased the accumulation of heavy metals in both plant species. A comparison study in control and industrial areas exhibited that heavy metal accumulation was higher in the industrially affected area than in the control area. Heavy metal concentration exceeded the permissible limit recommended by the WHO in both species of two industrial areas. However, both species accumulated the least heavy metal concentration in the control area. Biochemical investigation specifies that in response to heavy metal accumulation, both species increased the activity of hydrogen peroxide (H2O2), malondialdehyde content, the activity of enzymatic (superoxide dismutase and peroxidase) and nonenzymatic (ascorbic acid) antioxidant, but decreased the primary (soluble carbohydrate and total protein), secondary metabolites (phenol and flavonoid) content and free radical scavenging (DPPH) activity. Psoriasis, a widespread skin condition impacting over 100 million individuals globally, is characterised by uncontrolled hyperproliferation of keratinocytes, abnormal apoptosis, and excessive secretion of inflammatory cytokines and angiogenic factors. Traditional use of Alstonia scholaris (L.) R.Br. , Wrightia tinctoria (Roxb.) R.Br. and Solanum xanthocarpum Schrad. & Wendl. in Ayurveda and Siddha medicinal systems have shown promising anti-inflammatory and wound-healing properties. However, underlying mechanisms of their phytoactivity in addressing psoriasis-like skin inflammation on human keratinocytes remain largely unexplored. The study was aimed to investigate anti-psoriatic potential of ethyl acetate and ethanolic extracts of A. scholaris, W. tinctoria and S. xanthocarpum in human keratinocyte cell line (HaCaT). Ethyl acetate and ethanolic extracts of A. scholaris (ASEA and ASE), W. tinctoria (WTEA and WTE) and S. xanthocarpum (SXEA and SXE) were first subjected to phytochemical screening through high-performance liquid chromatography (HPLC) using their marker compound loganin, kaempferol and chlorogenic acid, respectively. The proliferation inhibition efficiency of these extracts was measured using MTT assay on HaCaT cell line. Subsequently, the apoptotic effect of these extracts on HaCaT cell line was determined by JC-1 and Annexin V assays. Furthermore, IL-8 and RANTES levels were measured in TNF-alpha-induced HaCaT cell line post-treatment with these extracts to determine their anti-inflammatory properties. ASEA, ASE, WTEA, WTE, SXEA and SXE significantly inhibited proliferation of keratinocytes (HaCaT cells) and resulted in the induction of apoptotic markers (mitochondrial membrane potential and phosphatidyl serine externalization). Additionally, pro-inflammatory markers (IL-8 and RANTES levels) were downregulated in HaCaT cells. The anti-proliferative effects were particularly distinct at higher concentrations (200 μg/mL), with inhibition rates reaching over 85% for W. tinctoria and S. xanthocarpum extracts. In apoptotic assays, notable increases in late apoptotic or necrotic cell populations and significant losses in mitochondrial membrane potential were observed. All extracts markedly reduced the secretion of inflammatory mediators IL-8 and RANTES. All three plants exerted an anti-psoriatic effect at the cellular level via multiple parameters (anti-proliferative, pro-apoptotic, anti-inflammatory effect). This study provides insight into the mechanism of action of ASEA, ASE, WTEA, WTE, SXEA and SXE and highlights their promising potential for development as herbal therapeutic agents for psoriasis. Wrightia tinctoria, an important traditional medicinal plant is exploited for treating several diseases. The study intends to reveal the presence of phytochemicals and test the antibacterial activity of W. tinctoria leaf extracts on nonwoven fabrics to find its suitability for wound dressings. The methodology includes identification and collection of W. tinctoria leaves, preparation of leaf powder, determination of physicochemical analysis, extraction using different solvents, preliminary phytochemical screening, quantitative estimation of phytoconstituents, yield of the extracts and determination of antibacterial activity on plant extract treated fabrics. Results revealed the presence of more active metabolites in the ethanol plant extract of W. tinctoria leaves which may be the reason for the promising antibacterial potential against the bacterial strains. In the present paper we have described the importance of the degree of agua-phobia characteristics of Psorolin B ointment and how such formulation would exceptionally be useful in treating Psoriasis. The ecosystem outside of is high in water (in the form of humidity) and skin also has the property of excreting various waste metabolites through sweat which would eventually both hinder and remove water soluble constituents from skin easily whereas such removal would be low if the constituents are agua-phobic. This work aims at research on characterization of Wrightia tinctoria fiber, such as fiber content, fiber longitudinal view, cross-sectional view, fiber diameter, moisture content, SEM analysis, surface resistivity, swelling, and also mechanical properties of fiber, such as fiber strength, elongation, linear density, etc. The fiber was used to produce carded webs using the pilot carding machine from blends of Wrightia tinctoria fiber (W) with cotton (C), bamboo (B), and polyester (P) in the blend ratios of 25:75 and 50:50 for required GSM. The produced webs were bonded using spunlace bonding technique to produce required non-woven medical apparel. The produced nonwoven were tested for dressing mass and thickness, absorbency of wound dressing, the dehydration rate of dressing, wettability, vertical wicking, air permeability, potential of hydrogen, water vapor permeability, and anti-bacterial. The test results of the studies were investigated in detail and observed that the product development from Wrightia tinctoria fiber has immense potential for curing various skin diseases as revealed from the test results.

Chemical modification with acetic anhydride is an effective method to stabilise wood against dimensional movement and improve decay resistance of wood. It has also been reported to retard UV light induced color darkening on wood surface. Coatings of wood surfaces with certain nano metal oxides have also shown promise as UV screening agent. In this work, we have evaluated synergistic effect of acetylation of wood and application of nano material on wood surfaces to enhance their UV stability. Photo-stability of acetylated Wrightia tinctoria wood coated with zinc oxide (ZnO) nanodispersion was evaluated by irradiating these wood specimens to UV radiation in an accelerated weathering tester. Acetylation of wood with acetic anhydride exhibited photo-bleaching and inhibited light-induced color darkening observed in unmodified wood. Coating of unmodified and modified wood surfaces with ZnO nanodispersion reduced photo-discolouration and degradation of wood polymers. The present study deals with the importance of herbal particle size in the AYUSH oil preparations for psoriasis in order to achieve greater absorption and therapeutic value. More often the viscosity of the oil, deep colour and the density of herbal constituents are associated with superior therapeutic value. But our study has clearly demonstrated that the dermal absorbable herbal constituents (under 40 μm) was less than 10% in most of the Wrightia tinctoria based AYUSH oil preparations sold for the treatment of psoriasis in India whereas Dr. JRK's 777 oil contain more than 60% of the herbal constituents below 40 μm size. Further the role of particle size besides dermal absorption; in the therapeutic value also established by oxygen species scavenging assay. Findings reveal that smaller the particles greater was the antioxidant effect and hence such herbal constituents not only would offer superior therapeutic effect but also protection to the oil from going rancid. In this study, eco-friendly natural dye sensitizers were extracted from the dried leaves of Wrightia tinctoria R.Br. (commonly known as "Pala indigo" or "Dyer's oleander") by cold methanolic and soxhlet extraction and used in dye sensitized solar cells (DSSC). The acidification of the extracted pigments was carried out using 0.1 N HCl. The absorption characteristics were studied by UV–visible spectroscopy, and the functional groups were identified by Fourier transform infrared spectroscopy. DSSCs constructed using these sensitizers were evaluated in terms of current density–voltage characteristics and electrochemical response. The DSSC fabricated with natural dye extracted by cold methanol exhibited maximum power conversion efficiency of 0.19%. The efficiency of the acidified cold methanolic extract-based DSSC was 0.06%. Wrightia tinctoria stem proteases were partially purified for the first time through a non-chromatographic technique, three phase partitioning (TPP), to concentrate the milk clotting proteases. Various parameters like salt and solvent concentration that affect the partitioning of the protease were examined. Maximum recovery and purification fold of the protease activity were found in the interfacial phase (IP) with 60% ammonium sulphate and 1:1 crude enzyme to t -butanol. Optimum pH and temperature of the enzyme fraction were found to be 7.5 and 50 °C respectively. Inhibition studies revealed its serine nature. Non-denaturing PAGE, Zymography and 2D PAGE of IP revealed presence of three different caseinolytic proteases of molecular weights 95.62 kDa, 91.11 kDa and 83.23 kDa with p I 3.89, 5.45 and 5.43 respectively. Both aqueous and lyophilized form of IP were remarkably stable retaining complete activity at 4 °C for 3 weeks. Electrophoretic analysis of casein hydrolysate by IP at different incubation time indicated a time dependent substrate subunit specificity with hydrolysis of κ-casein commencing after 10 min followed by α and β caseins. This pattern was found similar to that by commercial vegetable coagulant, Enzeco®. Wrightia tinctoria Roxb. (R.Br.) is an Ayurvedic remedy, ethnomedically used in the treatment of various ailments. The present work was carried out to evaluate the anticancer and antioxidant activity as well as total phenolic and phytochemical contents of W. tinctoria bark methanolic extract (WTBM) by high-performance liquid chromatography (HPLC)-diode array detector. Antiproliferative activity of WTBM was evaluated against MDA-MB-231 and MCF-7 cancer cells by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay, colony formation, and Hoechst staining. In addition, the antioxidant potential was determined by 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity and 2,2- azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical cation decolorization assay. Total phenolic content was assessed by Folin-Ciocalteu method. The results demonstrated that WTBM exhibited significant antiproliferative effect against MDA-MB-231 (IC 50 = 88.9 ± 1.27 μg/ml) and MCF-7 (IC 50 = 45.71 ± 7.74 μg/ml) cancer cells in time- and dose-dependent manner. WTBM significantly suppresses colony formation and induces apoptosis in both MDA-MB-231 and MCF-7 cells as evident by morphological assessment, clonogenic assay, and Hoechst staining. The total phenolic content of WTBM was found to be 30.3 gallic acid equivalent mg/g dry weight of bark extract while IC 50 value for DPPH and ABTS radical scavenging activity was 72.2 ± 2.8 μg/ml and 45.16 ± 1.95 μg/ml, respectively. HPLC analysis showed the presence of gallic acid, rutin, and quercetin in WTBM. These findings demonstrated that WTBM significantly inhibited proliferation of breast cancer cells and induced apoptosis, suggesting the potential chemopreventive activity of W. tinctoria bark.

In this paper, we report the green synthesis of gold nanoparticles (AuNPs) using Wright tinctoria seed extract. The biomolecules present in the seed extract is responsible for the formation of AuNPs and they found to play dual role of both reducing as well as capping agents. Appearance of violet colour confirms the AuNPs formation. The surface nature of the synthesised nanoparticle is studied using SEM and TEM analysis. The high crystallinity of AuNPs is observed from the diffraction peaks in XRD profile. FTIR spectrum confirms the presence of different functional groups of biomolecules participated in encapsulating AuNPs. In this work, a possible mechanism of AuNPs formation was also proposed. The synthesised biogenic nano-scale gold particles by natural tribes are tested for its inborn antimicrobial activity. Objective: Psoriasis is a chronic immune-mediated disorder of the skin. The disease manifests itself with red or silvery scaly plaques distributing over the lower back, scalp, and extensor aspects of limbs. Several medications are available for the treatment of psoriasis; however, high rates of remission and side-effects still persist as a major concern. Siddha, one of the traditional systems of Indian medicine offers cure to many dermatological conditions, including psoriasis. The oil prepared from the leaves of Wrightia tinctoria is prescribed by many healers for the treatment of psoriasis. This work aims to decipher the mechanism of action of the W. tinctoria in curing psoriasis and its associated comorbidities. Design: The work integrates various pharmacology approaches such as drug-likeness evaluation, oral bioavailability predictions, and network pharmacology approaches to understand the roles of various bioactive components of the herb. Results: This work identified 67 compounds of W. tinctoria interacting with 238 protein targets. The compounds were found to act through synergistic mechanism in reviving the disrupted process in the diseased state. A serine protease was purified from the leaves of Wrightia tinctoria by sequential flow through method comprising screening, optimization, ammonium sulfate precipitation, gel filtration and ion exchange column chromatography. The yield and purification fold obtained were 11.58% and 9.56 respectively. A single band of serine protease was visualized on SDS-PAGE and 2-D gel electrophoretic analyses were revealed with the molecular mass of 38.5 kDa. Serine protease had an optimum pH of 8.0 and was stable at 45°C with high relative protease activity. The addition of metal ions such as Mg2+ and Mn2+ exhibits a high relative activity. Serine protease had a potent antibacterial activity against both Gram-positive and Gram-negative bacteria. A 10 μg/ml of serine protease was tested against S. aureus, M. luteus, P. aeruginosa and K. pneumoniae which had 21, 20, 18 and 17 mm of zone of inhibition respectively. Objective: The current study investigates the potential of alcoholic extract of Wrightia tinctoria leaves and its isolates in repairing the DNA damage induced by gamma radiation. Methodology: The ethanolic extract of Wrightia tinctoria (EEWT) the leaves was subjected to in vitro anti oxidant assay by DPPH radical scavenging and hydrogen peroxide scavenging methods. He EEWT was investigated to the Micronucleus assay, spleen colony assay and comet assay in Sprague Dawley rats wherein DNA damage had already been induced by irradiation. Results: The ethanolic extract of Wrightia tinctoria leaves having good antioxidant activity. In the micronucleus assay, a 2 Gy quantum of wholebody gamma radiation significantly enhanced the levels of micronucleated reticulocytes and this induction was substantially reversed in the extract treatment group. Results of the spleen colony assay pointed to the extract's ability to enhance the recovery of the hematopoietic system, in a dosedependent manner, following 6 Gy gamma irradiation. The decrease in cellular repair index due to enhanced DNA repair was evident from the comet assay parameters. Moreover, the isolates from ethanolic extract of Wrightia tinctoria, when imperiled to DNA protection by in vitro gamma radiation method in plasmid pBR322 DNA, abetted DNA repair at higher dose. Introduction: Wrightia tinctoria R.Br. (Family: Apocynaceae) commonly called "Indrajau" is well known in Indian traditional system for its traditional uses. Materials and Methods: The present investigation was carried out to determine the possible bioactive components of plant seed ethanolic extract, pet ether fraction using GC-MS analysis. 22 components were identified from pet ether fraction obtained from elution of ethanolic extract packed in silica column. Results: The prevailing compounds from fraction F6to F9 were [1,1'-Bicyclopropyl]-2-octanoic acid, 2'-hexyl-, methyl ester (21.39%) , Trilinolein (7.74%), 2-Myristynoyl pantetheine (18.07%), 9-Octadecen12-ynoic acid, methyl ester (4.46%), 1Hexadecanol,2-methyl (3.77%), Cyclopropane tetradecanoic acid, 2-octyl-, methyl ester (2.36%), 1b, 4a-Epoxy-2H-cyclopenta [3,4] cyclopropa [8,9]cycloundec [1,2-b]oxiren-5 (6H)-one, 7-(acetyloxy) decahydro-2,9,10-trihydroxy-3,6,8,8,10a-pentamethyl (38.91%), Geranyl isovalerate (23.58%), cis13-Octadecenoic acid (5.91%), Quassin (3.82%), cis-10-Heptadecenoic acid (3.08%), 9,12,15-Octadecatrienoic acid 2-phenyl-1, 3-dioxan-5-yl ester (31.50%), 9,12,15-Octadecatrienoic acid, (Z,Z,Z)-2,3-dihydroxypropyl ester (14.35%), Cyclopropanebutanoic acid, 2-[ [2-[ [2- [(2-pentylcyclopropyl) methyl] cyclopropyl] methyl] cyclopropyl] methyl]-, methyl ester (10.13%), 6,9,12,15-Docosatetraenoic acid, methyl ester (3.39%), 9,12-Octadecadienoic acid, (2-phenyl1,3-dioxolan-4-yl) methyl ester, trans-( 2.73%), 9,12-Octadecadienoic acid, (2-phenyl-1,3-dioxolan-4-yl) methyl ester, cis-(4.34%), Ursodeoxycholic acid (7.14%), Bufa-20,22-dienolide, 3-(acetyloxy)-14,15-epoxy-16-hydroxy-, (3á,5á,15á,16á)-(4.75%), 5H-Cyclopropa [3,4] benz [1,2-e]azulen-5-one, 9a (acetyloxy)-1,1a,1b,4,4a,7a,7b,8,9,9ade cahydro-4a,7b,9-trihydroxy-3-(hydroxymethyl)-1,1,6,8-tetramethyl-,[1aR-(1aà,1bá,4aá,7aà,7bà,8à,9á,9 aà)]-(6.59%), Docosahexaenoic acid, 1,2,3-propanetriyl ester (10.86%), Olean-12-ene-3,15,16,21,22,28-hexol, (3á,15à,16à,21á,22à)-( 4.40%) found as the major components. Introduction: Wrightia tinctoria R. Br. belongs to family Apocynaceae commonly called as sweet Indrajao, Pala indigo plant, Dyer's Oleander. "Jaundice curative tree" in south India. Materials and Methods: A method has been developed for different extracts of W. tinctoria for high-performance thin-layer chromatography (HPTLC) fingerprinting analysis for identification and quantification of the marker compound. Result: The satisfactory resolution was obtained in the solvent system toluene:ethyl acetate v/v (8:2) for petroleum ether extract, toluene:ethyl acetate v/v (7:3) for chloroform extract and toluene:ethyl acetate:formic acid v/v (7:3:0.1) for alcoholic extract. Objective: The objective of this study was to investigate the hypoglycemic activity of Wrightia tinctoria (Roxb) (R. Br. Linn., Family- Apocynaceae) total flavonoid (WTTF) fraction on alloxan induced hyperglycemia. Materials and Methods: Total flavonoid fraction from WTTF seed was separated and LD50 calculated as 1006.93 mg/kg, i.p., following Organization for Economic Co-operation and Development guideline 423. The effect of total flavonoid fraction was observed in three different dose levels 50, 75, and 100 mg/kg. Hypoglycemic activity was carried out using alloxan induced hyperglycemia model. The parameters observed were body weight, blood glucose, total cholesterol, triglycerides (TG), low density lipoprotein, high density lipoprotein, urea and creatinine along with relative organ weight of liver, kidney, and pancreas. Results: Total flavonoid fraction of W. tinctoria did not have any significant effect on reducing glucose level. WTTF had significantly reduced creatinine (P < 0.001), TG (P < 0.05) level and relative weight of liver in diabetic rat at the dose of 100 mg/kg. Conclusion: All the findings indicate absence of antidiabetic activity in W. tinctoria seed but possible presence of liver and kidney protective property. [ABSTRACT FROM AUTHOR]
