Abstract

Isotope fractionation of metals/metalloids in biological systems is an emerging research area that demands the application of state-of-the-art analytical chemistry tools and provides data of relevance to life sciences. In this work, Se uptake and Se isotope fractionation were measured during the biofortification of baker's yeast (Saccharomyces cerevisiae)—a product widely used in dietary Se supplementation and in cancer prevention. On the other hand, metabolic labeling with 15N is a valuable tool in mass spectrometry-based comparative proteomics. For Se-yeast, such labeling would facilitate the assessment of Se impact on yeast proteome; however, the question arises whether the presence of 15N in the microorganisms affects Se uptake and its isotope fractionation. To address the above-mentioned aspects, extracellularly reduced and cell-incorporated Se fractions were analyzed by hydride generation–multi-collector inductively coupled plasma–mass spectrometry (HG MC ICP–MS). It was found that extracellularly reduced Se was enriched in light isotopes; for cell-incorporated Se, the change was even more pronounced, which provides new evidence of mass fractionation during biological selenite reduction. In the presence of 15N, a weaker preference for light isotopes was observed in both, extracellular and cell-incorporated Se. Furthermore, a significant increase in Se uptake for 15N compared to 14N biomass was found, with good agreement between hydride generation microwave plasma–atomic emission spectrometry (HG MP–AES) and quadrupole ICP–MS results. Biological effects observed for heavy nitrogen suggest 15N-driven alteration at the proteome level, which facilitated Se access to cells with decreased preference for light isotopes. Abstract

The feasibility of raw data acquired by liquid chromatography – high resolution mass spectrometry for the separation of four tequila corresponding to different maturation stages was demonstrated using partial least squares regression (PLS). Five samples of silver, rested, aged, and extra-aged tequilas from different manufacturers were analyzed each in two technical replicates. Prior to the analysis, the samples were four-fold diluted with 0.1% trifluoroacetic acid. Rectangle bucketing was performed for raw data, with a retention time width of 60 s and m/z width of 1. The prepared dataset was submitted to principal component analysis (PCA) which allowed for the detection of tequila contaminants identified to be N-lauryl diethanolamine and phthalate esters. Next, PLS was performed on depurated data, annotating tequila categories as follows: 0 - silver, 1 - rested, 2 - aged and 3 - extra aged. In the computed model, two principal components accounted for 64% and 98% of X-data and Y-data variability, respectively. Cross-validation revealed similar analytical performance in calibration and in prediction with root mean square errors of 0.1851 and 0.2082, respectively. The assignation of category's numerical value (0-3) was attained for eight randomly selected tequilas (two per category). Statistical differences between predicted values were obtained for each pair of categories except for the aged versus extra-aged liquors (ANOVA, p < 0.05). Overall, the assignation of tequila category was achieved with no need for the detection/identification nor quantification of individual compounds. [ABSTRACT FROM AUTHOR]

Abstract

Interactions of mineral elements in crops strongly affect their nutritional value and the yield. Within the context of ruminants feeding, ensuring Cu:Mo molar ratio in the range from 2:1 up to 20:1 has been considered. In this work, the impact of different Cu and Mo concentrations applied in nutrient solution of hydroponically grown Helianthus annuus was evaluated in terms of the seedlings growth and the status of Cu, Mo, Fe, Zn, Mg, Ca, K, Na. Nine groups of plants were obtained using concentration range 0.1–10 μmol L−1 for Cu and 0.1–50 μmol L−1 for Mo. Biomass in each treatment was assessed and eight elements were determined in roots and in aerial parts by microwave plasma atomic emission spectrometry (MP-AES). Several synergic and antagonistic interactions of Cu and/or Mo with other six mineral nutrients during uptake and root-to-shoot transport were observed. Regarding the feasibility of tuning Cu and Mo in medium to meet ruminants requirements, each of eight treatments was compared against control plants that were obtained using typical Cu and Mo concentrations in medium (0.44 and 0.70 μmol L−1, respectively). For ten-fold increased Cu concentration in medium (treatment 6), the most favorable Cu:Mo ratio was afforded (8.8:1 in roots and 17:1 in aerial parts) when compared with the control plants (1.8:1 and 20:1, respectively). Further, the concentrations of both elements in aerial parts were within ranges reported for forage; seedlings growth was not affected and concentrations of six other elements were practically unchanged when compared with controls.
