Abstract

Background: Sustained yet intractable immunosuppression is commonly observed in septic patients, resulting in aggravated clinical outcomes. However, due to the substantial heterogeneity within septic patients, precise indicators in deciphering clinical trajectories and immunological alterations for septic patients remain largely lacking. Methods: We adopted cross-species, single-cell RNA sequencing (scRNA-seq) analysis based on two published datasets containing circulating immune cell profile of septic patients as well as immune cell atlas of murine model of sepsis. Flow cytometry, laser scanning confocal microscopy (LSCM) imaging and Western blotting were applied to identify the presence of S100A9+ monocytes at protein level. To interrogate the immunosuppressive function of this subset, splenic monocytes isolated from septic wild-type or S100a9−/− mice were co-cultured with naïve CD4+ T cells, followed by proliferative assay. Pharmacological inhibition of S100A9 was implemented using Paquinimod via oral gavage. Results: ScRNA-seq analysis of human sepsis revealed substantial heterogeneity in monocyte compartments following the onset of sepsis, for which distinct monocyte subsets were enriched in disparate subclusters of septic patients. We identified a unique monocyte subset characterized by high expression of S100A family genes and low expression of human leukocyte antigen DR (HLA-DR), which were prominently enriched in septic patients and might exert immunosuppressive function. By combining single-cell transcriptomics of murine model of sepsis with in vivo experiments, we uncovered a similar subtype of monocyte significantly associated with late sepsis and immunocompromised status of septic mice, corresponding to HLA-DRlowS100Ahigh monocytes in human sepsis. Moreover, we found that S100A9+ monocytes exhibited profound immunosuppressive function on CD4+ T cell immune response and blockade of S100A9 using Paquinimod could partially reverse sepsis-induced immunosuppression. Conclusions: This study identifies HLA-DRlowS100Ahigh monocytes correlated with immunosuppressive state upon septic challenge, inhibition of which can markedly mitigate sepsis-induced immune depression, thereby providing a novel therapeutic strategy for the management of sepsis. 37. An improved density peak algorithm for micro-learning unit text clustering based on LSA model.

Abstract

With the explosive growth of micro-learning resources, a large number of unprocessed fragmented text resources bring great inconvenience to learners. In order to help learners to find suitable contents from fragmented resources for personalized learning, it is necessary to cluster micro-learning resources in the form of text. Therefore, this paper attempts to apply an improved density peak algorithm to micro-learning unit text clustering. Aiming at the problems of high dimensional sparse vector space, insufficient global consistency, cutoff distance sensitivity, and supervised selection of density peak centers when the density peak algorithm perform clustering in its field, this paper proposes two ap-proaches based on Latent Semantic Analysis (LSA) model. Firstly, a new definition of local density is proposed according to clustering requirements, density sensitive distance is used as the clustering criteria, and the global consistency problem of clustering is solved by solving the problem of cutoff distance sensitivity. Secondly, outliers are found by linear fitting to automatically find the density peak centers in order to realize unsupervised selection problem of peak centers. Experimental results on real data sets of micro-learning units show that the proposal is more suitable for text clustering of micro-learning units than the original algorithm and other classical clustering algorithms. Abstract

Adulterate sodium silicon apatite cement (s-AC) porous scaffolds for bone repair were prepared by us- ing the mixed powders of tetracalcium phosphate and dicalcium phosphate anhydrous, 5wt% sodium silicate water- solution as cement liquid, and NaCl particles as progens. The result shows that the composition of the s-AC scaf- folds is sodium silicon apatite. The macroporous sizes of the scaffolds were in the range from 200 μm to 600 μm, the porosity of the scaffolds is from 58% to 75%, and the macropores were interconnected with each other. The compressive strength of the s-AC scaffolds is from 1.6MPa to 3.8MPa, and the degradability of sodium silicon doped s-AC scaffolds improves significantly as compared with AC in Tris-HCl solution. When the s-AC scaffolds are implanted in femur bone defect of rabbits, the histological analysis results show that the new bone tissue is formed on the surfaces and ingrew into the interior of the scaffolds, the well interconnected macropores promoted the bone tissue ingrowth into the scaffolds. In conclusion, the s-AC scaffolds exhibit good biocompatibility, de- gradability and osteogenesis in vivo, which will be a good biomaterial for bone repair.

41. Comparative Analysis of Typical Low Rank Coal Pyrolysis Technology Based on a Nonlinear Programming Model

Abstract

A nonlinear programming (NLP) model has been established to describe the process of low rank coal pyrolysis, and the model was validated through comparison with pilot or semi-industry technologies. Results showed good agreement with actual production values, suggesting that the model can be regarded as a general one for simulating low rank coal pyrolysis. On the basis of this model, the products’ distributions and energy penalties can be obtained from the coal pyrolysis process. In this study, the NLP model was used to simulate typical five pyrolysis processes such as the Toscoal process, Lurgi–Ruhrgas process, Char Oil Energy Development technology, Dalian University of Technology process, and Multistage Rotary Furnace technology from China Coal Research Institute, and Hulunbuir lignite was taken as a coal sample. Combined with the simulation results from the NLP model, effects of the key parameters on the performance of the pyrolysis technologies are investigated and a comprehensive evaluation in terms of techno-economic performance is completed, from which the most suitable pyrolysis technologies for the Hulunbuir lignite feedstock are the Toscoal and Lurgi–Ruhrgas processes. All of the above work is beneficial for assisting the selection of the optimal pyrolysis technology for a coal feedstock with different composition features.

The kinetic processes of the photoionization and dissociation of 4H-furan have been investigated by using synchrontron radiation. The IP of 4H-furan and APs of other ions are gotten from the spectra aka possible channels or dissociative photo ionization have been analyzed. A serices of stable clusters composed of (THF)(n)(H2O)(n-2)H+ (n=2,3,4,5) are present in the MS and the possible structures of the clusters are given.

Abstract

Adaptive stepping sliding mode control strategy for the overload control of anti-ship missiles using Lyapunov's stability theory is proposed. In the first few steps, we assume there exists a virtual actuator, the virtual actuator concept can make the desired value can be easily traced, in the last step, a sliding mode controller is applied to guarantee the robustness of the system. At the same time, an adaptive controller is adopted to attenuate the unknown parameters and uncertainties; and because the switching term in the sliding mode control can cause the chattering phenomenon, a fuzzy controller is adopted to adjust it. The simulation shows validity of the proposed method.

Abstract

Both direct and sensitized irradiations of the endo title compound lead to a decarbonylation product, 4-isopropylbiphenyl, via Norrish Type 1 reaction, while the exo isomer gives a carbene intermediate and subsequently formaldehyde 2-biphenylyl isopropyl hemiacetal, which in turn gradually decomposes to dimethyl phenylbenzyl alcohol at room temperature. The triplet pathway dominates the photoreaction of the exo isomer. The singlet and triplet lifetimes of the reactant are found to be 0.29 ns and 5.4 μs, respectively, 3(n, x*), 1(n, x*), 3(x, x*) electronic configurations of the exo isomer and 3(x, x*) of the endo isomer are photochemically active, but 1(x, x*) of the endo isomer shows indirect photoreactivity.
