Abstract Background and purpose The complications of radical surgery for cervical cancer can increase patient suffering and affect their quality of life. This retrospective study assessed the safety of radical hysterectomy (RH) with pelvic lymph node dissection (PLND) by observing the complications of patients with cervical cancer who underwent this procedure in a single centre over 10 years. Our findings may provide experience and evidence for preventing and reducing complications. Methods A total of 2226 cervical cancer patients who met the inclusion criteria were enrolled. All patients underwent RH + PLND. Intraoperative injury to adjacent tissues and short-term and long-term complications were recorded to analyze factors associated with the occurrence of complications. Results Postoperative complications occurred in 34.41% (766/2226) of patients, including 7.68% of patients with injury to adjacent tissues, 31.45% with short-term complications, and 2.96% with long-term complications. Age, tumor size, invasion depth, parametrial invasion, lymph vascular space invasion (LVSI), lymph node metastasis, International Federation of Gynaecology and Obstetrics (FIGO) stage, and surgical procedure were closely associated with the postoperative complications of RH + PLND (P IB2, and who underwent open surgery were more prone to complications.

Abstract Objective This study aimed to evaluate the clinical outcomes for patients with stage IB2/IIA2 cervical squamous cell carcinoma treated with neoadjuvant chemotherapy combined with radical surgery. Methods A total of 68 patients with cervical squamous cell carcinoma were randomly divided into the experimental group (n = 35) and the control group (n = 33). The patients in the experimental group received paclitaxel plus cisplatin neoadjuvant chemotherapy for two cycles, then underwent radical hysterectomy and bilateral adnexectomy at 2 weeks post-chemotherapy. The control group only underwent radical hysterectomy and bilateral adnexectomy after the diagnosis of cervical squamous cell carcinoma. The toxic and side effects of chemotherapy in the experimental group were observed. Also, the operation method, operation time, blood loss, grade of wound healing, complications, and postoperative pathology were noted in the two groups. Primary foci and pelvic lymph node recurrence and distant metastasis were observed, and 3-year and 5-year survival rates were calculated. Results Only one patient in the experiment had grade III bone marrow suppression; no other grade III and IV chemotherapy toxic reactions were observed. The operation was successfully completed in all patients. The operation time, intraoperative blood loss, placement of the ureteral catheter, bladder injury, ureteric injury, postoperative urinary tub, pelvic drainage tube indwelling time, anal exhaust time, postoperative complications, and metastatic ratio of lymph nodes were not significantly different between the two groups (P > 0.05). The number of dissected lymph nodes, deep myometrial invasion, and vascular tumor emboli showed a significant difference in the experimental group compared with the control group (P < 0.05). The 3-year disease-free survival (82.9% vs 81.9%), 5-year disease-free survival (71.4% vs 60.6%), 3-year overall survival (91.4% vs 87.8%), and 5-year overall survival (82.9% vs 75.6%) were not statistically significantly different between the experimental group and the control group (P > 0.05). Conclusions Neoadjuvant chemotherapy in IB2/IIA2 stage cervical squamous cell carcinoma showed low toxic side effects. Radical surgery after chemotherapy is safe and feasible. It plays a coordinating role in reducing the tumor infiltration depth of the deep muscle layer and the incidence of vascular tumor emboli, reducing the use of postoperative adjuvant therapy, and improving the quality of life of patients, but does not improve the 3-year/5-year survival rate.

In this work, Plackett-Burman (PB) and Box-Behnken (BB) experiment design were used to investigate the medium components of fermentation forth of ginsenoside Re bioconversion to ginsenoside Rh1. Firstly, nine variables including cultivation time, temperature, pH, reactant concentration, inoculum’s size, peptone, MgSO4*7H2O, K2HPO4 and NaNO3 were screened. The most important effective factors were estimated according to the statistical regression. Subsequently, the steepest ascent experiment was used to approach the optimum response range for each effective factor. Then Box-Benhnken Design (BBD) was finally used to analyze the response surface of the screened factors for further optimization. The optimized conditions for the influential factors were: ginsenoside Re 1.0 g/L, pH 4.5 and 7 days. The bioconversion of ginsenoside Re to Rh1 could reach up to 51.76 %.

Infectious bronchitis virus (IBV) causes infectious bronchitis in chicken, an acute, highly contagious respiratory infection. Because of genetic mutations and recombination, IBV forms many subtypes, which makes it difficult to treat the disease and apply commercial vaccines. Therefore, to detect IBV in time and stop the virus from spreading, a novel and convenient IBV detection technology based on reverse transcription recombinase-aided amplification (RT-RAA) was established in this study. According to the S1 gene of IBV CH I–V and Mass genotypes and S1 gene of IBV CH VI genotype, a set of optimal primers were designed and selected to establish a real-time dual fluorescence RT-RAA method. The lowest detection line was 10 copies/μL of RNA molecules and the method exhibited no cross-reactivity with avian reticuloendotheliosis virus (REV), infectious bursal disease virus (IBDV), avian leukosis virus (ALV), Newcastle disease virus (NDV), chicken infectious anemia virus (CIAV), infectious laryngotracheitis virus (ILTV), Marek's disease virus (MDV), and H9N2 avian influenza virus (H9N2), demonstrating high specificity. When compared to qPCR detection results, our method achieved a sensitivity of 96.67%, a specificity of 90%, and a Kappa value of 0.87 for the IBV CH I–V and Mass genotypes, and achieved a sensitivity of 100%, a specificity of 97.73%, and a Kappa value of 0.91 for the IBV CH VI genotype. This paper attempts to analyze and assess Yongchuan Xiuya tea quality quickly, accurately, and digitally. The sensory evaluation method was first used to assess Yongchuan Xiuya tea quality, and then near infrared spectroscopy (NIRS) was obtained, and standard methods were applied to the testing of the chemical components. Next, principal component analysis (PCA) and the correlation coefficient method were used to comprehensively screen out the representative components. Finally, NIRS combined with partial least squares regression (PLSR) and back propagation artificial neural network (BP-ANN) methods were applied to build quality evaluation models for Yongchuan Xiuya tea, respectively, and external samples were employed to examine the practical application results of the best model. The cumulative variance contribution rate of the first three principal components of the ingredients in tea was 97.73%. Seven components closely related to tea quality were screened out, namely, amino acids, total catechin, epigallocatechin gallate (EGCG), tea polyphenols, water extracts, epicatechin gallate (ECG), and epigallocatechin (EGC) (p < 0.01). Between the two models established to predict the tea quality, the model built by the PLS method had the better results, whose coefficient of determination of prediction (Rp2) and root mean square error of prediction (RMSEP) were 0.7955 and 1.2263, respectively, and the best results were obtained by the nonlinear BP-ANN model, whose Rp2 and RMSEP were 0.9315 and 0.6787, respectively. The 10 external Yongchuan Xiuya samples were employed to test the best BP-ANN model, and the results of R2 and RMSEP were 0.9579 and 0.6086, respectively, meaning that the model has good robustness. Therefore, the model established by NIRS combined with the BP-ANN method can be used to assess Yongchuan Xiuya tea quality rapidly, accurately, and digitally, and it can also provide new ideas and methods for evaluating the quality of other teas. Introduction: Progressive accumulation of amyloid-β (Aβ) is a pathological trait of Alzheimer's disease (AD). Amyloid-β increases free radical production in neuronal cells, leading to neuronal cell death. Hormone replacement therapy can reduce the incidence of AD, and oestrogen significantly improves the clinical signs in patients with AD. However, the long-term use of oestrogen causes a variety of diseases. Phytoestrogens have been reported to bind and activate oestrogen receptors in mammals and humans to produce oestrogen-like or anti-oestrogen-like effects. Kaempferol is a flavonoid phytoestrogen that can produce a certain protective effect in neurons. However, the molecular mechanism of kaempferol in AD is unclear.Material and Methods: This study used pheochromocytoma (PC-12) cells that were damaged by Aβ25-35 as an in vitro model of AD, and oestradiol was a positive control. The cells were incubated with kaempferol alone or in combination with fulvestrant (an antagonist of ER) and U0126 (an inhibitor of ERK) in Aβ25-35 culture. Cell activity was measured by the MTT method. Cell apoptosis was evaluated by flow cytometry. Gene and protein expression levels were tested by qRT-PCR and Western blotting.Results: This study demonstrated that kaempferol protected PC-12 cells from Aβ25-35-induced cell death and apoptosis in a dose-dependent manner. Treatment with fulvestrant (an antagonist of ER) and U0126 (an inhibitor of ERK) significantly increased the apoptosis of PC-12 cells. Moreover, kaempferol promoted the expression of anti-apoptotic molecules and inhibited the expression of pro-apoptotic molecules, which were blocked by fulvestrant and U0126.Conclusions: Kaempferol protected PC-12 cells against Aβ25-35-induced cell apoptosis through the ER/ERK/MAPK signalling pathway.
