Abstract

We are Studying the magnetic cloud event of January 6 - 11, 1997. Specifically, we have investigated the response of the magnetosphere to the shock wave in front of the magnetic cloud on January 10, 1997 using data from WIND, GEOTAIL and POLAR spacecraft as well as ground magnetometer data. The WIND spacecraft, which was located as about 104 Re upstream from the Earth (85.1, -55.2, -22.1) Re(sub GSM), observed the arrival of the shock wave front at 0050 UT. Geotail was located at the equatorial magnetopause (approx. 8.7 Re, 10.7 MLT, -7.46 MLAT), while POLAR was located in the northern dawn sector above the auroral zone at 8.4 Re, 6.1 MLT and 61.1 MLAT. A magnetic signature was nearly simultaneously observed at about 0104 UT at the POLAR and Geotail spacecraft. The Geotail spacecraft entered from the magnetosphere into the magnetosheath. Particle density increases were observed on WIND and Geotail, but not on POLAR. Two instruments on the Polar spacecraft (TIDE and TIMAS) actually observed a slight reduction in energy, density and temperature. The UV aurora shows a dawnside intensification. The shock wave did not cause an auroral substorm and therefore was not geoeffective.

11. 18F-Labeled phosphopeptide-cell-penetrating peptide dimers with enhanced cell uptake properties in human cancer cells

Abstract

Introduction Phosphopeptides represent interesting compounds to study and elucidate cellular protein phosphorylation/dephosphorylation processes underlying various signal transduction pathways. However, studies of phosphopeptide action in cells are severely constrained by the negatively charged phosphate moiety of the phosphopeptide resulting in poor transport through the cell membrane. The following study describes the synthesis and radiopharmacological evaluation of two 18F-labeled phosphopeptide-cell-penetrating peptide dimers. The polo-like kinase-1-binding hexaphosphopeptide H-Met-Gln-Ser-pThr-Pro-Leu-OH was coupled to cell-penetrating peptides (CPPs), either sC18, a cathelicidin-derived peptide, or the human calcitonin derivative hCT(18-32)-k7. Methods Radiolabeling was accomplished with the prosthetic group N-succinimidyl 4-[18F]fluorobenzoate ([18F]SFB) using both, conventional and microfluidic-based bioconjugation of [18F]SFB to N-terminal end of phosphopeptide part of the peptide dimers. Cellular uptake studies in human cancer cell lines HT-29 and FaDu cells at 4 °C and 37 °C and small animal PET in BALB/c mice were utilized for radiopharmacological characterization. Results Isolated radiochemical yields ranged from 2% to 4% for conventional bioconjugation with [18F]SFB. Significantly improved isolated radiochemical yields of up to 26% were achieved using microfluidic technology. Cellular uptake studies of radiolabeled phosphopeptide and phosphopeptide-CPP dimers indicate enhanced internalization of 50% ID/mg protein after 2 h for both phosphopeptide dimers compared to the phosphopeptide alone (< 1% ID/mg protein). In vivo biodistribution of 18F-labeled peptide dimers was determined with small animal PET revealing a superior biodistribution pattern of sC18-containing peptide dimer MQSpTPL-sC18 [18F]4. Conclusion [18F]SFB labeling of the phosphopeptide-CPP dimers using a microfluidic system leads to an improved chemoselectivity towards the N-terminal NH2 group compared to the conventional labeling approach. Cell-penetrating peptide sC18 can be considered as an ideal molecular shuttle for intracellular delivery of the Plk1-PBD-binding hexaphosphopeptide as demonstrated by its favourable radiopharmacological profile.

Abstract

Objectives: According to GLOBOSCAN 2008 the worldwide prostate cancer incidence rates will increase to about 1.7 million new cases per year in 2030. In recent years, bombesin and bombesin analogs have attracted much attention as high affinity and selectivity ligands for the gastrin-releasing peptide (GRP) receptor. The GRP receptor was found to be overexpressed and implicated in a variety of human tumors including prostate cancer. Radiolabeled bombesin and bombesin analogues belong to an interesting class of new diagnostic probes for molecular imaging of GRP receptor-expressing prostate cancer. This study describes the synthesis and radiopharmacological evaluation of a high affinity and metabolically stabilized 18F-labeled bombesin analog for PET imaging of GRP receptors in prostate cancer. Methods: Three modified bombesin analogs bearing an aminovaleric (BBN-1, BBN-2), or an aminooctanoic acid linker (BBN-3) were tested in a competitive binding assay against 125I-[Tyr4]-BBN for their binding to the GRP receptor. A calcium release assay in human prostate cancer cells (PC3) was performed to determine agonistic or antagonistic behaviour. The derivative with the highest affinity to GRP (BBN-2) was selected to be conjugated with the prosthetic labeling agent N-succinimidyl-4-[18F]fluorobenzoate ([18F]SFB) in borate buffer (pH=8.2) for 30 min at 40°C to synthesize the desired 18F-labeled bombesin analog. Tumor-targeting of [18F]BBN-2 was evaluated in PC3 tumor-bearing male nude mice with biodistribution experiments (mean ± SD) and dynamic small animal PET studies. Results: The competitive binding assay revealed IC50-values between 8.7 and 16.7 nM for BBN-1, BBN-2 and BBN-3 against 125I-[Tyr4]-BBN versus 3.0 nM for I-[Tyr4]-BBN. All three stabilized bombesin analogs are GRP receptor antagonists. 18F-labeled [18F]BBN-2 was prepared in 30% radiochemical yield (based upon [18F]SFB) within 80 min including HPLC purification, evaporation of HPLC eluent and formulation in 0.9% saline. The radiochemical purity exceeded 95%, and the specific activity was determined to be 20 GBq/µmol. [18F]BBN-2 showed reasonable metabolic stability in mouse blood resulting in 65% of intact radiolabeled peptide after 60 min p.i.. Uptake of [18F]BBN-2 into PC3 tumors was 2.75±1.82 %ID/g after 5 min and 2.45±1.25 %ID/g after 60 min p.i.. Radiolabeled BBN derivatives are promising diagnostic probes for molecular imaging of GRP receptor-expressing prostate cancer. This study describes the synthesis and radiopharmacological evaluation of various metabolically stabilized fluorobenzoylated bombesin analogues (BBN-1, BBN-2, BBN-3). Methods: Three fluorobenzoylated BBN analogues containing an aminovaleric (BBN-1, BBN-2), or an aminooctanoic acid linker (BBN-3) were tested in a competitive binding assay against 125I-[Tyr4]-BBN for their binding potency to the GRP receptor. Intracellular calcium release in human prostate cancer cells (PC3) was measured to determine agonistic or antagonistic profiles of fluorobenzoylated BBN derivatives. Bombesin derivative BBN-2 displayed the highest inhibitory potency toward GRP receptor (IC50 = 8.7 ± 2.2 nM) and was subsequently selected for radiolabeling with fluorine-18 (18F) through acylation with N-succinimidyl-4-[18F]fluorobenzoate ([18F]SFB). The radiopharmacological profile of 18F-labeled bombesin [18F]BBN-2 was evaluated in PC3 tumor-bearing NMRI nude mice involving metabolic stability studies, biodistribution experiments and dynamic small-animal PET studies. Results: All fluorobenzoylated BBN derivatives displayed high inhibitory potency toward the GRP receptor (IC50 = 8.7–16.7 nM), and all compounds exhibited antagonistic profiles as determined in an intracellular calcium release assay. Abstract

An experimental setup based on the constant volume technique is developed to measure the mass flow rate through microtubes under isothermal stationary flow conditions. Four different working gases (helium, nitrogen, argon, and carbon dioxide), and two surface materials (stainless steel and Sulfinert) are considered. The Knudsen number calculated for the experimental conditions varies from ~10-4 (hydrodynamic regime) to ~5 (transitional regime). In the reduced range (10-4-0.1) corresponding to the hydrodynamic and slip regimes, an approach based on the analytical solution of the Stokes equation subjected to a first order velocity slip boundary condition is used. The velocity slip coefficient and the tangential momentum accommodation coefficient are extracted from the experimental data of the mass flow rate using their analytical expressions. The results are summarized in the tables representing the accommodation coefficients for the corresponding gas-surface material combinations. The influence of the molecular mass on the tangential momentum accommodation coefficient is discussed. In addition, an original technique is proposed to extract the accommodation coefficient in the whole experimentally available range (10-4-5) of the gas rarefaction. Abstract

Purpose The objective of the present study was to analyze, with relatively high sensitivity and specificity, uptake properties of [11C]-choline in prostate cancer patients by means of positron-emission tomography (PET)/ computed tomography (CT) imaging using objectively defined pet parameters to test for statistically significant changes before, during, and after external-beam radiation therapy (EBRT) and to identify the time points at which the changes occur. Methods The study enrolled 11 patients with intermediate-risk prostate cancer treated with ebrt, who were followed for up to 12 months after EBRT. The [11C]-choline PET scans were performed before treatment (baseline); at weeks 4 and 8 of EBRT; and at 1, 2, 3, 6, and 12 months after EBRT. Results Analysis of [11C]-choline uptake in prostate tissue before treatment resulted in a maximum standardized uptake value (SUVMAX) of 4.0 ± 0.4 (n = 11) at 40 minutes after injection. During week 8 of EBRT, the SUVMAX declined to 2.9 ± 0.1 (n = 10, p < 0.05). At 2 and 12 months after EBRT, SUVMAX values were 2.3 ± 0.3 (n = 10, p < 0.01) and 2.2 ± 0.2 (n = 11, p < 0.001) respectively, indicating that, after EBRT, maximum radiotracer uptake in the prostate was significantly reduced. Similar effects were observed when analyzing the tumour:muscle ratio (TMR). The TMR declined from 7.4 ± 0.6 (n = 11) before EBRT to 6.1 ± 0.4 (n = 11, nonsignificant) during week 8 of EBRT, to 5.6 ± 0.03 (n = 11, p < 0.05) at 2 months after EBRT, and to 4.4 ± 0.4 (n = 11, p < 0.001) at 12 months after EBRT. Conclusions Our study demonstrated that intraprostatic [11C]- choline uptake in the 11 analyzed prostate cancer patients significantly declined during and after EBRT. The PET parameters SUVMAX and TMR also declined significantly. These effects can be detected during radiation therapy and up to 1 year after therapy. The prognostic value of these early and statistically significant changes in intraprostatic [11C]-choline pet avidity during and after EBRT are not yet established. Future studies are indicated to correlate changes in [11C]-choline uptake parameters with long-term biochemical recurrence to further evaluate [11C]-choline pet changes as a possible, but currently unproven, biomarker of response. 30. In vitro and in vivo evaluation of [(18)F]F-GAZ, a novel oxygen-mimetic azomycin-glucose conjugate, for imaging hypoxic tumor.

Abstract

Several F-18-labeled 2-nitroimidazole (azomycin) derivatives have been proposed for imaging hypoxia using positron emission tomography (PET). Their cell penetration is based on passive diffusion, which limits their intracellular concentration maxima. The purpose of this study was to investigate the uptake of N-(2-[(18)F]fluoro-3-(6-O-glucosyl)propyl-azomycin ([(18)F]F-GAZ), a new azomycin-glucose conjugate, in vitro and in vivo. [(18)F]F-GAZ was synthesized from its tetraacetyl nosylate precursor by nucleophilic radiofluorination. [(18)F]F-GAZ was evaluated in vivo in EMT-6 tumor-bearing Balb/C mice utilizing the PET and biodistribution analysis. In vitro uptake of [(18)F]FDG by EMT-6 cells was measured in the presence of unlabeled F-GAZ, 2-FDG, and D-glucose. [(18)F]F-GAZ was rapidly cleared from all tissues, including the blood pool and kidneys, with ultimate accumulation in the urinary bladder. Uptake of tracer doses of [(18)F]F-GAZ into EMT-6 tumors was fast, reaching a standardized uptake value of 0.66±0.05 within 5-6 minutes postinjection (p.i.), and decreased to 0.24±0.04 by 60 minutes p.i. (n=6). A tumor-muscle ratio of 1.87±0.18 was observed after 60 minutes. Total uptake of [(18)F]F-GAZ in tumors (60 minutes) amounted to 1.25%±0.15% ID/g versus 0.61%±0.14% ID/g (n=4) in muscle. Similar biodistribution and excretion were observed using carrier-added (100 mg/kg) doses of F-GAZ. In vitro, D-glucose and unlabeled 2-FDG were two orders of magnitude more potent than F-GAZ as competitive inhibitors of [(18)F]FDG uptake into EMT-6 cells. Besides its interaction with glucose transporters, F-GAZ seems to be not transported in the presence of glucose. Furthermore, [(18)F]F-GAZ is unlikely to be effective as a hypoxia imaging agent. The low in vivo toxicity and substantial retention in tumor observed at high doses of F-GAZ do provide rationale for further testing as a radiosensitizer for external beam radiation therapy of radioresistant, hypoxic tumors. 32. Radiolabeling and In Vivo Behavior of Copper-64-Labeled Cross-Bridged Cyclam Ligands

Abstract

Macrocyclic chelators and their metal complexes have widespread applications in the biomedical sciences, including radiopharmaceutical chemistry. The use of copper radionuclides in radiopharmaceuticals is increasing. Macrocyclic chelators have been found to have enhanced in vivo stability over acyclic chelators such as ethylenediaminetetraacetic acid (EDTA) and diethylenetriaminepentaacetic acid (DTPA). The currently used chelators of choice for labeling copper radionuclides to biological molecules are analogues of TETA (1,4,8,11-tetraazacyclotetradecane-1,4,8,11-tetraacetic acid); however, recent reports have demonstrated evidence of in vivo instability of the radio-Cu(II)−TETA complexes. A new class of structurally reinforced macrocycles, the “cross-bridged” cyclam derivatives, form highly stable complexes with Cu(II) that are resistant to dissociation in strong acid. Here, we evaluate a series of 64Cu(II) cross-bridged macrocyclic complexes for biological stability and in vivo behavior. The ligands evaluated include the parent ligand, 1,4,8,11-tetraazabicyclo[6.6.2]hexadecane (1), and three 4,11-di-pendant arm derivatives:  4,11-bis(carboxymethyl)-1,4,8,11-tetraazabicyclo[6.6.2]hexadecane (2); 4,11-bis(N,N-diethyl-amidomethyl)-1,4,8,11-tetraazabicyclo[6.6.2]hexadecane (3); and 4,11-bis(amidoethyl)-1,4,8,11-tetraazabicyclo[6.6.2]hexadecane (4). Copper-64 formed complexes with ligands 1−4 in high radiochemical yields. The 64Cu−2 complex was neutral, while 64Cu complexes of 1, 3, and 4 were positively charged. All complexes showed no decomposition in rat serum out to 24 h. Biodistribution experiments in Sprague−Dawley rats indicated that 64Cu−1, −3, and −4 were taken up by the liver and kidney and cleared slowly over 24 h, whereas 64Cu−2 cleared rapidly from all tissues. The rapid clearance of the 64Cu−2 complex from the blood and liver, as well as liver metabolism experiments in rats, suggests that it is highly stable in vivo. A bifunctional chelator of 2 is a significant candidate for labeling copper radionuclides to biological molecules for diagnostic imaging and targeted radiotherapy.

Abstract

A simulated three-dimensional computer graphics display system was developed to address issues critical to the training of Air Force air weapons controllers; primarily those training issues related to air intercept tactics and geometry. The research and development system provided the capability to display and control up to three aircraft in a defined airspace, plus the addition of supplemental graphics to address specific training issues using full-color, raster-scan, animated microcomputer display technology. To transition this technology to the training environment, two video tapes were produced and delivered to the 325th Weapons Controller Training Squadron (formerly the USAF Interceptor Weapons School), where they are being incorporated into the training syllabus. Volume I of this document provides an overview and summary of the project (Final Report). Abstract

Background: The objective of this research was the development and evaluation of 203 Pb-labelled panitumumab ( 203 Pb-PSC-panitumumab) as an immuno-SPECT radioligand for the detection of EGFR + head and neck squamous cell carcinoma (HNSCC) in a patient-derived xenograft (PDX) mouse model. The 51.9 h physical half-life and favourable γ-emission (279 keV; 81%) of 203 Pb offer an excellent opportunity for developing immuno-SPECT radioligands. Moreover, 203 Pb has a complementary therapeutic radionuclide ( 212 Pb), making 203 Pb and 212 Pb an ideal matched radiotheranostic pair., Results: Radiolabeling of panitumumab was performed at a pH of 5.0 and room temperature for 5-10 min with [ 203 Pb]Pb(OAc) 2 , and the incorporation efficiency was determined using radio-TLC. 203 Pb-PSC-panitumumab (~ 10 MBq, 140 μl of saline) was injected into the tail vein of NRG mice bearing subcutaneous (s.c.) HNSCC patient-derived xenografts (PDX). SPECT/CT images were acquired at 48 and 120 h post-injection. For biodistribution studies, mice were euthanized five days after 203 Pb-panitumumab injection. The tumour and normal tissues were collected and weighed, and uptake of 203 Pb was measured in a γ-counter. The uptake was calculated as the percent injected dose per gram of each tissue (ID%/g). Blocking experiments were performed by pretreating a group of mice (n = 5) with 1 mg of panitumumab 1 h before administering 203 Pb-PSC-panitumumab. 4-5 chelators of a new lead-specific chelator (PSC) were attached per antibody; radiolabeling efficiency was 99.2 ± 0.7%. The isolated radiochemical yield of 203 Pb-PSC-panitumumab was 41.4 ± 8% (n = 5), and the molar activity was 1.2 ± 0.35 GB/mg. SPECT imaging and biodistribution confirmed high accumulation and retention of 203 Pb-PSC-panitumumab in the tumour (26% ID/g) at 120 h post-injection (p.i.), which could be reduced to 6.2%ID/g at 120 h p.i. by predosing with panitumumab (1 mg) confirming EGFR specificity of 203 Pb-PSC-panitumumab uptake., Conclusions: Panitumumab was successfully and reproducibly labelled with 203 Pb in high radiochemical purity using the chelator PSC-NCS. 203 Pb-PSC-panitumumab was specifically accumulated and retained in EGFR + tumours in NRG mice with s.c. HNSCC PDX. 203 Pb-PSC-panitumumab is a suitable immuno-SPECT radioligand for imaging EGFR + tumours and has great potential for combining with 212 Pb-PSC-panitumumab in a radiotheranostic strategy for imaging and treating HNSCC., Competing Interests: Declarations.
