Abstract Background Older subjects have a higher risk for vertebral compression fracture. Maintaining a higher bone mineral density (BMD) at this age can protect individuals from osteoporosis-related events. Body mass index (BMI) has been found to have a robust association with BMD. However, excessive BMI is detrimental to bone health and may cause systemic disorders. Therefore, the present study aimed to determine the association between BMI and BMD, and identify a reasonable BMI range. Methods A total of 961 participants were recruited from community-dwelling residents between August 2021 and May 2022. A weighted multivariate linear regression model was applied to identify the relationship between BMI and BMD. Meanwhile, subgroup stratified analysis by BMI quartile and gender was also performed. A non-linear relationship and threshold value were determined based on the smooth curve fittings and threshold effects analysis model. Results A robust relationship was found between BMI and BMD, which remained significant in subgroups stratified by gender and BMI quartile. The BMI inflection point values in lumbar BMD and femoral neck BMD were 25.2 kg/m2 and 27.3 kg/m2, respectively. Abstract Osteosarcomas are intricate cellular ecosystems, where heterotypic interactions significantly influence disease progression and therapeutic outcomes. Despite their importance, a detailed understanding of their cellular composition and organizational structure remains elusive. In this study, we provide a comprehensive single-cell and spatially resolved transcriptomics analysis of human osteosarcomas. We construct a cellular meta-map to dissect spatial transcriptomic data, unveiling a detailed atlas of osteosarcoma compositional subgroups. We meticulously characterize the unique gene signatures and functional states of each subgroup and investigate the impact of chemotherapy on these cellular subpopulations. Additionally, our spatial transcriptomics analysis identifies a distinct spatial niche, located at the forefront of tumor necrotic zones, potentially associated with chemotherapy resistance. We also delve into the crosstalk between different cellular subgroups. This study furnishes a comprehensive transcriptional atlas of osteosarcoma's cellular architecture, enriching our comprehension of its complexity and laying the groundwork for more targeted therapeutic approaches.

Abstract AlGaN-based light-emitting diodes (LEDs) operating in the deep-ultraviolet (DUV) spectral range (210–280 nm) have demonstrated potential applications in physical sterilization. However, the poor external quantum efficiency (EQE) hinders further advances in the emission performance of AlGaN-based DUV LEDs. Here, we demonstrate the performance of 270-nm AlGaN-based DUV LEDs beyond the state-of-the-art by exploiting the innovative combination of bandgap engineering and device craft. By adopting tailored multiple quantum wells (MQWs), a reflective Al reflector, a low-optical-loss tunneling junction (TJ) and a dielectric SiO2 insertion structure (IS-SiO2), outstanding light output powers (LOPs) of 140.1 mW are achieved in our DUV LEDs at 850 mA. The EQEs of our DUV LEDs are 4.5 times greater than those of their conventional counterparts. This comprehensive approach overcomes the major difficulties commonly faced in the pursuit of high-performance AlGaN-based DUV LEDs, such as strong quantum-confined Stark effect (QCSE), severe optical absorption in the p-electrode/ohmic contact layer and poor transverse magnetic (TM)-polarized light extraction. Furthermore, the on-wafer electroluminescence characterization validated the scalability of our DUV LEDs to larger production scales. Our work is promising for the development of highly efficient AlGaN-based DUV LEDs.

Abstract The conversion of biomass to chemicals/fuels has emerged as a valuable solution that offers both environmental and economic benefits, with the transformation of carbohydrate into formic acid garnering escalating scholar interest. However, the relative limited efficiency of catalyzed-oxidation or expensive cost of H2O2 and alkali in wet hydrothermal oxidation impose limitations on industrialization. This paper proposed a new idea for formic acid production by O2-H2O2 co-oxidation of carbohydrate. A two-step reaction method was developed, where the initial step is engineered to regulate the carbon chain cleavage of carbohydrates to augment the production of active intermediate. Oxygen was employed in the subsequent step as effective oxidant through free radical mechanism, resulting in a formic acid yield of 82.6%. Theoretical calculation, intermediates detection and real time EPR confirmed the reaction mechanism. Finally, the universality of the reaction was verified by using disaccharides and polysaccharides such as cellulose as substrates.

Abstract In August 2023, Xiaomi unveiled the Redmi K60 Ultra, the first multi-frequency smartphone integrated with BeiDou-3 Navigation Satellite System Precise Point Positioning (PPP-B2b) services and employing PPP technology as the primary positioning method. The positioning enhancement service is provided by the Assisted Global Navigation Satellite System (A-GNSS) location platform developed by the China Academy of Information and Communications Technology. The signaling interaction between the server and the users strictly adheres to the Third Generation of Mobile Communications Technology Partnership Project Long-Term Evolution Positioning Protocol and the Open Mobile Alliance Secure User Plane Location framework. To comprehensively evaluate the Redmi K60 Ultra’s capabilities, this study designed six distinct experimental scenarios and conducted comprehensive research on multi-frequency and multi-GNSS observation noise, Time to First Fix (TTFF), as well as the performance of both GNSS-based and network-based positioning. Experimental results indicate that the GNSS chipset within the Redmi K60 Ultra has achieved a leading position in the consumer market concerning supported satellite constellations, frequencies, and observation accuracy, and is comparable to some low-cost GNSS receivers. A-GNSS positioning can reduce the TTFF from 30 to under 5 s, representing an improvement of over 85% in the cold start speed compared to a standalone GNSS mode. The positioning results show that the A-GNSS PPP-B2b service can achieve positioning performance with RMS errors of less than 1.5 m, 2.5 m, and 4 m in open-sky, realistic, and challenging urban environments. Compared to GNSS-based positioning, cellular network-based Observed Time Difference of Arrival (OTDOA) positioning achieves an accuracy ranging from tens to hundreds of meters in various experimental scenarios and currently functions primarily as coarse location determination. Additionally, this study explores the potential of the Three-Dimensional Mapping-Aided (3DMA) GNSS algorithm in detecting Non-Line-of-Sight signals and enhancing positioning performance. The results indicate that 3DMA PPP, as compared to conventional PPP, can significantly accelerate PPP convergence and improve positioning accuracy by over 30%. Consequently, 3D city models can be utilized as future assistance data for the A-GNSS location platform.

Abstract Mounting evidence showed that HER2-Low breast cancer patients could benefit from the novel anti-HER2 antibody–drug conjugates (ADCs) treatment, which pointed the way towards better therapy for HER2-Low patients. The purpose of this study was to describe the clinicopathological features, along with chemotherapeutic effects and survival outcomes of HER2-Low and HER2-Zero in TNBC who received neoadjuvant chemotherapy (NACT). We retrospectively evaluated 638 triple-negative breast cancer patients who were treated with neoadjuvant chemotherapy between August 2014 and August 2022. Pathologic complete response (pCR) and survival outcomes were analyzed in HER2-Low cohort, HER2-Zero cohort and the overall patients, respectively. In the entire cohort, 342 (53.6%) patients were HER2-Low and 296 (46.4%) patients were HER2-Zero. No significant difference was found between HER2-Low and HER2-Zero patients based on all the clinical–pathological characteristics. 143 cases (22.4%) achieved pCR after NACT in the overall TNBC patients. The pCR rate of the HER2-Low patients and the HER2-Zero patients was 21.3% and 23.6%, respectively, exhibiting no statistical difference (p = 0.487). The survival of pCR group after NACT significantly improved compared to non-pCR group either in HER2-Low patients or in HER2-Zero patients. Although we found that patients with HER2-Low had longer DFS than patients with HER2-Zero, there was no considerable difference (p = 0.068). However, HER2-Low patients had a dramatically longer OS than HER2-Zero patients (p = 0.012). The data from present study confirmed the clinical importance of HER2-Low expression in TNBC. Further effort is needed to determine whether HER2-Low could be a more favorable prognostic marker for individual treatment.

Objective: This study aimed to examine the latent profile of nurses' mental workload (MWL) and explore the influencing factors via a person-centred approach. Methods: From March to July 2023, a quantitative cross-sectional study was carried out to investigate 526 Chinese clinical nurses from five tertiary hospitals in Sichuan Province, China, by using demographic information, the Perceived Social Support Scale, Simplified Coping Skill Questionnaire, and NASA-Task Load Index. Latent profile analyses were performed using Mplus 7.3 software. Pearson’s chi-squared and logistic regression analysis was done using SPSS 24.0 software. Results: Three profiles of mental workload were identified based on the nurses’ responses to the mental workload assessment, designated as “low MWL-high self-rated (n = 70, 13.3%)”, “moderate MWL (n = 273, 51.9%)”, and “high MWL-low self-rated (n = 183, 34.8%)”. Abstract Background Hypoxic pulmonary hypertension (HPH) is a challenging lung arterial disorder with remarkably high incidence and mortality rates, and the efficiency of current HPH treatment strategies is unsatisfactory. Endothelial-to-mesenchymal transition (EndMT) in the pulmonary artery plays a crucial role in HPH. Previous studies have shown that lncRNA-H19 (H19) is involved in many cardiovascular diseases by regulating cell proliferation and differentiation but the role of H19 in EndMT in HPH has not been defined. Methods In this research, the expression of H19 was investigated in PAH human patients and rat models. Then, we established a hypoxia-induced HPH rat model to evaluate H19 function in HPH by Echocardiography and hemodynamic measurements. Moreover, luciferase reporter gene detection, and western blotting were used to explore the mechanism of H19. Results Here, we first found that the expression of H19 was significantly increased in the endodermis of pulmonary arteries and that H19 deficiency obviously ameliorated pulmonary vascular remodelling and right heart failure in HPH rats, and these effects were associated with inhibition of EndMT. Moreover, an analysis of luciferase activity indicated that microRNA-let-7 g (let-7 g) was a direct target of H19. H19 deficiency or let-7 g overexpression can markedly downregulate the expression of TGFβR1, a novel target gene of let-7 g. Furthermore, inhibition of TGFβR1 induced similar effects to H19 deficiency. Conclusions In summary, our findings demonstrate that the H19/let-7 g/TGFβR1 axis is crucial in the pathogenesis of HPH by stimulating EndMT. Our study may provide new ideas for further research on HPH therapy in the near future.

Abstract This qualitative study investigates the factors that affect the job satisfaction of home caregivers. It draws upon a thematic analysis of data from in-depth interviews with home caregivers in the UK and finds that both visible and invisible factors have an effect. Overall factors pertaining to the way the caregiving agencies are run and behave have a significant influence on caregivers’ job satisfaction. Identifying these factors will assist both policymakers and professional care agencies to be aware of the needs of caregivers in practice. These experiences can be used as references to handle the challenges of an ageing society in other countries with similar care industry characteristics.

Plasmon-induced hot-electron transfer from metal nanostructures is being intensely pursed in current photocatalytic research, however it remains elusive whether molecular-like metal clusters with excitonic behavior can be used as light-harvesting materials in solar energy utilization such as photocatalytic methanol steam reforming. In this work, we report an atomically precise Cu13 cluster protected by dual ligands of thiolate and phosphine that can be viewed as the assembly of one top Cu atom and three Cu4 tetrahedra. The Cu13H10(SR)3(PR’3)7 (SR = 2,4-dichlorobenzenethiol, PR’3 = P(4-FC6H4)3) cluster can give rise to highly efficient light-driven activity for methanol steam reforming toward H2 production.

Abstract This study explores the representation of gender through the social interactions presented in four sets of English textbooks used in China. By examining the representation of both gender stereotypes and changing roles, the results reveal that although traditional stereotypes exist in Chinese secondary school-level English textbooks, women’s changing social roles are acknowledged in that they are now beginning to be shown engaging in non-traditional female jobs. However, in family scenarios, women are still depicted as taking sole responsibility for housekeeping duties without any participation from adult males, showing the incomplete transformation of gender roles. No male characters are shown participating in non-traditional jobs, which reinforces the idea of the masculine role being ‘superior’ to feminine or non-masculine roles. The continued presence of gender stereotypes shows textbook editors’ tacit agreement with the gender norms. Drawing on gender as a fluid concept and social interactions of gender, this study acknowledges the variety and diversity of inputs of gender roles through social interactions in the learning community and suggests that students can be given access to a more gender-progressive education by improving the quality of social interactions between genders in textbooks and presenting a more equal view on gender roles.

Caudofoveata is a class of worm-like molluscs (aplacophorans) that typically have an infaunal lifestyle, burrowing in soft bottoms in a wide range of marine habitats from shallow to deep waters. Here, we describe a very large new species of caudofoveate from South China Sea methane seeps growing up to 154 mm in length: Chaetoderma shenloong sp. nov. It is the first caudofoveate to be named from a chemosynthetic ecosystem and the first aplacophoran mollusc associated with seeps. Our new species stands out from other Pacific Chaetoderma species by its large size, a wide body relative to its length, a barely sclerotised radula, and the presence of isosceles-triangular sclerites. Phylogenetic reconstruction using the mitochondrial cytochrome c oxidase subunit I (COI) gene placed it within a paraphyletic clade comprising Chaetodermatidae and Limifossoridae, in line with a previous phylogenetic analysis. This also revealed that C. shenloong sp. nov. is conspecific with a Chaetoderma sp. whose whole genome was recently sequenced and assembled but remained undescribed until now. The most closely related species with an available COI sequence was C. felderi, the largest caudofoveate species recorded. Our discovery suggests caudofoveates may be present in other seeps globally but so far neglected; a potential example is C. felderi from the Gulf of Mexico, where seeps are abundant but whose exact habitat remains unclear.

Abstract Bacteroidales (syn. Bacteroidetes) are prominent members of the human gastrointestinal ecosystem mainly due to their efficient glycan-degrading machinery, organized into gene clusters known as polysaccharide utilization loci (PULs). A single PUL was reported for catabolism of high-mannose (HM) N-glycan glyco-polypeptides in the gut symbiont Bacteroides thetaiotaomicron, encoding a surface endo-β-N-acetylglucosaminidase (ENGase), BT3987. Here, we discover an ENGase from the GH18 family in B. thetaiotaomicron, BT1285, encoded in a distinct PUL with its own repertoire of proteins for catabolism of the same HM N-glycan substrate as that of BT3987. We employ X-ray crystallography, electron microscopy, mass spectrometry-based activity measurements, alanine scanning mutagenesis and a broad range of biophysical methods to comprehensively define the molecular mechanism by which BT1285 recognizes and hydrolyzes HM N-glycans, revealing that the stabilities and activities of BT1285 and BT3987 were optimal in markedly different conditions. BT1285 exhibits significantly higher affinity and faster hydrolysis of poorly accessible HM N-glycans than does BT3987. We also find that two HM-processing endoglycosidases from the human gut-resident Alistipes finegoldii display condition-specific functional properties. Altogether, our data suggest that human gut microbes employ evolutionary strategies to express distinct ENGases in order to optimally metabolize the same N-glycan substrate in the gastroinstestinal tract.

CircRNAs are a class of single-stranded RNAs characterized by covalently looped structures. Emerging advances have promoted our understanding of circRNA biogenesis, nuclear export, biological functions, and functional mechanisms. Roles of circRNAs in diverse diseases have been increasingly recognized in the past decade, with novel approaches in bioinformatics analysis and new strategies in modulating circRNA levels, which have made circRNAs the hot spot for therapeutic applications. Moreover, due to the intrinsic features of circRNAs such as high stability, conservation, and tissue-/stage-specific expression, circRNAs are believed to be promising prognostic and diagnostic markers for diseases. Aiming cardiovascular disease (CVD), one of the leading causes of mortality worldwide, we briefly summarize the current understanding of circRNAs, provide the recent progress in circRNA functions and functional mechanisms in CVD, and discuss the future perspectives both in circRNA research and therapeutics based on existing knowledge.

In order to solve the existing problem of low efficiency of multi-layer and multi-pass welding methods for thick stainless steel clad plate (SSCP), this paper proposes high-power laser-arc hybrid welding (HP-LAHW) single-pass weld thick SSCP. The effects of wire feeding rate (WFR) on the weld's composition distribution, microstructure and mechanical properties were investigated. The weld morphology is nail-shaped, consisting of a wide arc zone (ArcZ) within the stainless steel layer and a long narrow laser zone (LaserZ), mainly in the carbon steel layer. With the increase of WFR, the dilution rate of the carbon steel layer to the ArcZ decreases first and then increases. The influence of WFR on the microstructure and mechanical properties of the weld is observed mainly in the ArcZ. When the WFR reaches 6 m/min, the composition of the microstructure within the ArcZ will change from martensite, ferrite, and a small amount of granular austenite to ferrite and a small amount of austenite, resulting in the microhardness of the ArcZ decreasing rapidly from 398 HV to 210 HV. The solidification type and microstructure transformation mechanisms within the weld were elucidated based on quantitative calculations of the chromium and nickel equivalent ratios. The microstructure of the LaserZ in all welds is predominantly martensitic due to the rapid cooling rate, which makes the tensile properties of the weld higher than the base metal (BM). The results of this study provide technical support for realizing high-efficiency and high-performance single-pass HP-LAHW of thick SSCP.

The mudstone within the Upper Triassic Adula Formation constitutes a crucial hydrocarbon source rock reservoir in the Qiangtang Basin, yet there is considerable debate regarding its paleo-depositional environment. Through systematic studies, including petrology, organic geochemistry, and elemental geochemistry analyses of the Adula Formation at the Eertuolongba section in the Quemocuo area of the eastern Qiangtang Basin, this study investigated the paleo-depositional environment of the Adula Formation source rocks and its impact on hydrocarbonsource rock development. The total organic carbon (TOC) content of the Adula Formation mudstone ranges from 0.27% to 3.46%, with an average of 1.60%, indicating overall favorable source rocks with locally deve-loped high-quality source rocks. The shelf facies mudstone in the lower section of the Adula Formation was formed during a phase of sea-level rise, characterized by deeper, oxygen-poor water, a semi-arid to semi-humid paleoclimate, moderate chemical weathering, minimal terrigenous input, and higher salinity. The deposition of the upper section occurred during a phase of basin contraction and demise, suggesting a transition from shelf facies to delta facies. This period was marked by relatively shallower, oxygen-rich water and a shift from arid to semi-arid to semi-humid paleoclimate, with increased terrigenous input and a saline to semi-saline water environment influenced by freshwater input. The source rocks of the Adula Formation are felsic volcanic rocks, primarily from a continental island arc tectonic setting, likely sourced from the island arc source domain in the Jinsha River suture zone during the Early to Middle Triassic. In the lower section of the Adula Formation, the TOC content of shelf facies mudstone shows a robust positive correlation with redox condition indicators, suggesting that source rock development was mainly controlled by oxygen-poor water conditions during sea-level rise period. In contrast, no significant correlation was observed between TOC content and paleo-environmental parameters in the upper shelf delta facies mudstones. Mudstones with TOC content greater than 2% were likely deposited during periods of high terrigenous input and relatively humid paleoclimate, indicating that source rock development was influenced by multiple factors such as paleoclimate and terrigenous input.

Abstract Background Acute respiratory distress syndrome (ARDS) is a severe and fatal disease. Although mesenchymal stem cell (MSC)-based therapy has shown remarkable efficacy in treating ARDS in animal experiments, clinical outcomes have been unsatisfactory, which may be attributed to the influence of the lung microenvironment during MSC administration. Extracellular vesicles (EVs) derived from endothelial cells (EC-EVs) are important components of the lung microenvironment and play a crucial role in ARDS. However, the effect of EC-EVs on MSC therapy is still unclear. In this study, we established lipopolysaccharide (LPS) - induced acute lung injury model to evaluate the impact of EC-EVs on the reparative effects of bone marrow-derived MSC (BM-MSC) transplantation on lung injury and to unravel the underlying mechanisms. Methods EVs were isolated from bronchoalveolar lavage fluid of mice with LPS - induced acute lung injury and patients with ARDS using ultracentrifugation. and the changes of EC-EVs were analysed using nanoflow cytometry analysis. In vitro assays were performed to establish the impact of EC-EVs on MSC functions, including cell viability and migration, while in vivo studies were performed to validate the therapeutic effect of EC-EVs on MSCs. RNA-Seq analysis, small interfering RNA (siRNA), and a recombinant lentivirus were used to investigate the underlying mechanisms. Results Compared with that in non-ARDS patients, the quantity of EC-EVs in the lung microenvironment was significantly greater in patients with ARDS. EVs derived from lipopolysaccharide-stimulated endothelial cells (LPS-EVs) significantly decreased the viability and migration of BM-MSCs. Furthermore, engrafting BM-MSCs pretreated with LPS-EVs promoted the release of inflammatory cytokines and increased pulmonary microvascular permeability, aggravating lung injury. Mechanistically, LPS-EVs reduced the expression level of isocitrate dehydrogenase 2 (IDH2), which catalyses the formation of α-ketoglutarate (α-KG), an intermediate product of the tricarboxylic acid (TCA) cycle, in BM-MSCs. α-KG is a cofactor for ten-eleven translocation (TET) enzymes, which catalyse DNA hydroxymethylation in BM-MSCs. Conclusions This study revealed that EC-EVs in the lung microenvironment during ARDS can affect the therapeutic efficacy of BM-MSCs through the IDH2/TET pathway, providing potential strategies for improving the therapeutic efficacy of MSC-based therapy in the clinic.

To address the functional failure caused by low-surface-tension liquid wetting and adhesion in superhydrophobic materials, in this study, we constructed a bilayer coating with superamphiphobic properties using an epoxy (Ep) bonding layer and an organosilane-grafted attapulgite (OG-ATP) layer through spray-coating technique. The prepared superamphiphobic bilayer coating exhibited high contact angles (CAs) and low sliding angles (SAs) for cold water (25 °C), hot water (85 °C), glycerol, ethylene glycol, peanut oil, and even n-hexadecane (CA = 150.4 ± 0.3° and SA = 7 ± 2°) with surface tension of 27.5 mN m−1. Surface morphology and elemental analysis confirmed the coating's unique micro-nano hierarchical structure and low surface energy characteristics. Electrochemical impedance spectroscopy (EIS) results showed that the surface charge transfer resistance (Rct) and low-frequency modulus (|Z|0.01Hz) were enhanced by 6–7 orders of magnitude compared to the bare aluminum alloy substrate. The anti-corrosion performance remained stable even after 5 days of 3.5 wt% NaCl immersion. Additionally, icing experiments with water droplets at −10 °C and −15 °C revealed a significant delay in ice formation on the substrate protected by the bilayer coating. The development of this protective material, combining superamphiphobicity, self-cleaning, corrosion resistance, and anti-icing functions, will provide advanced technological reserves in areas such as marine, aviation, transportation, and manufacturing.

Abstract Spectrum feature extraction plays a crucial role in identifying seismic events and calculating structural response parameters. However, the criteria for identifying effective modal components in Variational Mode Decomposition (VMD) are not well-defined, resulting in inaccurate spectrum feature extraction. To address this issue, we propose a novel spectrum feature extraction method that combines Allan variance, VMD, and power spectral density (PSD). Firstly, VMD is applied to filter noise components from triaxial accelerometer observations and add effective signals. Secondly, PSD is utilized to extract three groups of seismic frequencies (tri-axial accelerometers). Finally, the Allan method is introduced to identify the group of accelerometer observations with the highest reliability as the vibration frequency caused by the seismic excitation. We validate the effectiveness of our method by analyzing a Mw 2.6 micro-seismic event that occurred in Huairou, Beijing in 2022. The result shows that our proposed method accurately extracts spectrum features of the Great Wall. Specifically, the seismic excitation vibration frequencies at four monitoring stations were found to be 26.95 Hz, 12.89 Hz, 12.89 Hz, and 12.5 Hz. These findings underscore our method's utility in evaluating the Great Wall's structural response to seismic loading, which has significant implications for the conservation and protection of heritage structures.

Abstract To improve their research performance in international league tables, many universities in non-English language dominant settings recruit academic returnees in the hope that they will increase the quantity and quality of articles published in English-medium internationally indexed journals. This study explores Chinese early career returnee academics’ motivations for writing and publishing in English. Utilising ecological systems theory, the findings show that the microsystem is reflected in the early career returnee academics’ interaction with collaborators, while national policies constitute the exosystem. The academic culture has a noteworthy impact at the macrosystem level. This study contributes to the understanding of early career returnee academics’ motivations to write and publish in English which will assist policymakers and university administrators to create a more beneficial environment to promote the accomplishments of academic returnees.

Abstract Objective To evaluate the antibacterial effectiveness of a combination of ε-poly-L-lysine (ε-PL), funme peptide (FP) as well as domiphen against oral pathogens, and assess the efficacy of a BOP® mouthwash supplemented with this combination in reducing halitosis and supragingival plaque in a clinical trial. Abstract Even though deep learning is fascinated in fields of coronary vessel segmentation in X-ray angiography and achieves prominent progresses, most of those models probably bring high false and missed detections due to indistinct contrast between coronary vessels and background, especially for tiny sub-branches. Image improvement technique is able to better such contrast, while boosting extraneous information, e.g., other tissues with similar intensities and noise. If incorporating features derived from original and enhanced images, the segmentation performance is improved because those images comprise complementary information from different contrasts. Accordingly, inspired from advantages of contrast improvement and encoding-decoding architecture, a dual multi-scale feature aggregation network (named DFA-Net) is introduced for coronary vessel segmentation in digital subtraction angiography (DSA). DFA-Net integrates the contrast improvement using exponent transformation into a semantic segmentation network that individually accepts original and enhanced images as inputs. Through parameter sharing, multi-scale complementary features are aggregated from different contrasts, which strengthens leaning capabilities of networks, and thus achieves an efficient segmentation. Meanwhile, a risk cross-entropy loss is enforced on the segmentation, for availably decreasing false negatives, which is incorporated with Dice loss for joint optimization of the proposed strategy during training. Experimental results demonstrate that DFA-Net can not only work more robustly and effectively for DSA images under diverse conditions, but also achieve better performance, in comparison with state-of-the-art methods. Consequently, DFA-Net has high fidelity and structure similarity to the reference, providing a way for early diagnosis of cardiovascular diseases.

Abstract With the rapid development of national infrastructure projects, there has been a significant increase in intersecting lines in transportation construction. As a result, rotating bridges are increasingly used in engineering projects that span existing railway lines. In order to study the spatial response characteristics and vibration wave transmission mechanisms of the rotating bridge structure under the loading of existing railway trains, field experiments and numerical analyses were conducted. The response characteristics of these bridges were investigated under different types and speeds of adjacent existing lines. A comprehensive methodology has been proposed, integrating the time domain spectrum and the Hilbert–Huang Transform (HHT) energy spectrum for signal processing and vibration analysis. The analysis was carried out using MATLAB 2018a software. This methodology was applied to analyze the test data. The results show that significant resonance phenomenon occurs in the girders of the rotating bridge under the loading of trains on the existing line. The low-frequency component f 1 (2–5 Hz) is the primary factor contributing to the amplification of the acceleration response in the rotating bridge, while f 3 (10–13 Hz) plays a secondary role. The frequency distribution characteristics of vibration waves caused by train loads on the existing line have a significant influence on the acceleration response of the rotating bridge's girders. The predominant frequency of vibration waves at each measuring point along the transmission path shows a trend of decreasing → increasing → decreasing. The impact on the rotating bridge structure of vibration waves generated by low-speed freight trains on existing railways is greater. The research findings are of great importance for studying the dynamic response of rotating bridges adjacent to existing railway lines.

Abstract Background Medial patellar ligament reconstruction (MPFL-R) in combination with derotational distal femoral osteotomy (DDFO) for treating recurrent patellar dislocation (RPD) in the presence of increased femoral anteversion is one of the most commonly used surgical techniques in the current clinical practice. However, there are limited studies on the clinical outcomes of MPFL-R in combination with DDFO to treat RPD in the presence of increased femoral anteversion. Purpose To study the role of MPFL-R in combination with DDFO in the treatment of RPD in the presence of increased femoral anteversion. Methods A systematic review was performed according to the PRISMA guidelines by searching the Medline, Embase, Web of Science, and Cochrane Library databases through June 1, 2023. Studies of patients who received MPFL-R in combination with DDFO after presenting with RPD and increased femoral anteversion were included. Methodological quality was assessed using the MINORS (Methodological Index for Nonrandomized Studies) score. Each study’s basic characteristics, including characteristic information, radiological parameters, surgical techniques, patient-reported outcomes, and complications, were recorded and analyzed. Results A total of 6 studies with 231 patients (236 knees) were included. Sample sizes ranged from 12 to 162 patients, and the majority of the patients were female (range, 67-100%). The mean age and follow-up ranges were 18 to 24 years and 16 to 49 months, respectively. The mean femoral anteversion decreased significantly from 34° preoperatively to 12° postoperatively. In studies reporting preoperative and postoperative outcomes, significant improvements were found in the Lysholm score, Kujala score, International Knee Documentation Committee score, and visual analog scale for pain. Postoperative complications were reported in all studies, with an overall reported complication rate of 4.7%, but no redislocations occurred during the follow-up period. Conclusion For RPD with increased femoral anteversion, MPFL-R in combination with DDFO leads to a good clinical outcome and a low redislocation rate. However, there was no consensus among researchers on the indications for MPFL-R combined with DDFO in the treatment of RPD.

Abstract Mask-free multi-photon lithography enables the fabrication of arbitrary nanostructures low cost and more accessible than conventional lithography. A major challenge for multi-photon lithography is to achieve ultra-high precision and desirable lateral resolution due to the inevitable optical diffraction barrier and proximity effect. Here, we show a strategy, light and matter co-confined multi-photon lithography, to overcome the issues via combining photo-inhibition and chemical quenchers. We deeply explore the quenching mechanism and photoinhibition mechanism for light and matter co-confined multiphoton lithography. Besides, mathematical modeling helps us better understand that the synergy of quencher and photo-inhibition can gain a narrowest distribution of free radicals. By using light and matter co-confined multiphoton lithography, we gain a 30 nm critical dimension and 100 nm lateral resolution, which further decrease the gap with conventional lithography.

Abstract Translational control is crucial for protein production in various biological contexts. Here, we use Ribo-seq and RNA-seq to show that genes related to oxidative phosphorylation are translationally downregulated during heart regeneration. We find that Nat10 regulates the expression of Uqcr11 and Uqcrb mRNAs in mouse and human cardiomyocytes. In mice, overexpression of Nat10 in cardiomyocytes promotes cardiac regeneration and improves cardiac function after injury. Conversely, treating neonatal mice with Remodelin—a Nat10 pharmacological inhibitor—or genetically removing Nat10 from their cardiomyocytes both inhibit heart regeneration. Mechanistically, Nat10 suppresses the expression of Uqcr11 and Uqcrb independently of its ac4C enzyme activity. This suppression weakens mitochondrial respiration and enhances the glycolytic capacity of the cardiomyocytes, leading to metabolic reprogramming. We also observe that the expression of Nat10 is downregulated in the cardiomyocytes of P7 male pig hearts compared to P1 controls. The levels of Nat10 are also lower in female human failing hearts than non-failing hearts. We further identify the specific binding regions of Nat10, and validate the pro-proliferative effects of Nat10 in cardiomyocytes derived from human embryonic stem cells. Our findings indicate that Nat10 is an epigenetic regulator during heart regeneration and could potentially become a clinical target.

Abstract Background Mycoplasma pneumoniae infections have increased in China recently, causing some evidence of familial clustering. The purpose of this study was to compare the clinical features of parents and children in cases of familial clustering of Mycoplasma pneumoniae infection. Methods A retrospective analysis was performed on the cases of familial clustering of Mycoplasma pneumoniae infection, and the clinical characteristics of parents and children were compared. Results We identified 63 families, of these, 57 (65.5%) adults and 65 (94.2%) children required hospitalization. Fifty-seven adults (mean age 35.1 ± 4.6 years, 80.7% female) and 55 children (mean age 6.3 ± 3.9 years, 54.5% female) were included in the analysis. The incidence of mycoplasma infection in adults had increased gradually over the past year, while the rate in children had spiked sharply since June 2023. The clinical symptoms were similar in the two groups, mainly fever and cough. The peak temperature of children was higher than that of adults (39.1 ± 0.7℃ vs 38.6 ± 0.7℃, p = 0.004). Conventional horizontal twin-roll casting (TRC) with steel rollers generally possesses slow casting speed. In this study, the inclined TRC method with Cu rollers was proposed, which significantly increases the casting efficiency. A three-dimensional model of the inclined TRC was established, which revealed the temperature field, solid fraction, velocity distribution, and TRC microstructure of the AA6005 aluminum (Al) alloys by the finite element simulation. We considered the various effects of the pouring temperature, roll gap, and rolling speed on the outlet temperature of the inclined cast rolling zone, and determined the appropriate range of processing parameters. The casting speed of the inclined TRC process could reach ∼5.8–7.9 m/min when the roll gap was 2 and 3 mm. Due to the different contact areas between the melt and two rollers, the temperature field and the shape of the liquid hole were asymmetric, and the isotherm and solidification end point (Kiss point) shifted to the direction of the upper roller at the steady inclined TRC state. The columnar/equiaxed grain structure and the upper shift of central segregation in the inclined TRC sheets were successfully simulated by the finite element method, which was in good agreement with the experiments. Moreover, the inclined TRC method with Cu rollers has high cooling intensity and the potential to eliminate central segregation. The results of this study can effectively reduce the trial-and-error effort, and provide a new insight into the development of the TRC Al alloys.

This study investigates the phenomenon of “music fever” in the Chinese immigrant community in the UK, a relatively neglected research field in the English language literature. It explores the attitudes of Chinese immigrant parents towards their children’s music learning through three key concepts in Bourdieu’s cultural theory: capital, habitus, and field. Although the interviewees generally displayed a positive attitude towards their children’s music learning, it was conditional on it not interfering with their children’s academic studies. This study extends existing research on East Asian immigrants’ and parents’ perceptions of children’s music education, and contributes to the application of Bourdieu’s theoretical tool in research on East Asian cultural groups in both Western and non-Western contexts.

Abstract Precise diagnostic biomarkers of anticitrullination protein antibody (ACPA)‐negative and early‐stage RA are still to be improved. We aimed to screen autoantibodies in ACPA‐negative patients and evaluated their diagnostic performance. The human genome‐wide protein arrays (HuProt arrays) were used to define specific autoantibodies from the sera of 182 RA patients and 261 disease and healthy controls. Statistical analysis was performed with SPSS 17.0. In Phase I study, 51 out of 19,275 recombinant proteins covering the whole human genome were selected. In Phase II validation study, anti‐ANAPC15 (anaphase promoting complex subunit 15) exhibited 41.8% sensitivity and 91.5% specificity among total RA patients. There were five autoantibodies increased in ACPA‐negative RA, including anti‐ANAPC15, anti‐LSP1, anti‐APBB1, anti‐parathymosin, and anti‐UBL7. Abstract Magnetosonic (MS) waves with frequencies above the proton gyrofrequency can be locally generated by ring‐beam protons in the Martian heavy ion‐rich ionosphere. In this study, we conduct a parametric analysis to investigate the effects of heavy ion concentrations, energy (Erb), and angle (αrb) of ring‐beam protons, and wave normal angle on the excitation features of Martian ionospheric MS waves. We find the growth rates and frequency range of MS waves decrease by including O+ and O2+ ions but are insensitive to their relative concentrations. With increasing Erb or αrb, the growth rates of MS waves show a general dropping tendency. Meanwhile, their frequency and wavenumber range are almost unaffected by increasing Erb but shrink to a narrower range mainly distributed in high frequencies by increasing αrb. Unstable MS waves expand to a wider wavenumber range but shrink to a narrower frequency range as they become more oblique.

Abstract Emerging evidence has demonstrated the profound impact of the gut microbiome on cardiovascular diseases through the production of diverse metabolites. Using an animal model of myocardial ischemia–reperfusion (I/R) injury, we found that the prophylactic administration of a well‐known probiotic, Bifidobacterium infantis (B. infantis), exhibited cardioprotective effects in terms of preserving cardiac contractile function and preventing adverse cardiac remodeling following I/R and that these cardioprotective effects were recapitulated by its metabolite inosine. Transcriptomic analysis further revealed that inosine mitigated I/R‐induced cardiac inflammation and cell death. Mechanistic investigations elucidated that inosine suppressed the production of pro‐inflammatory cytokines and reduced the numbers of dendritic cells and natural killer cells, achieved through the activation of the adenosine A2A receptor (A2AR) that when inhibited abrogated the cardioprotective effects of inosine. Additionally, in vitro studies using C2C12 myoblasts revealed that inosine attenuated cell death by serving as an alternative carbon source for adenosine triphosphate (ATP) generation through the purine salvage pathway when subjected to oxygen‐glucose deprivation/reoxygenation that simulated myocardial I/R injury. Likewise, inosine reversed the I/R‐induced decrease in ATP levels in mouse hearts. Taken together, our findings indicate that B. infantis or its metabolite inosine exerts cardioprotective effects against I/R by suppressing cardiac inflammation and attenuating cardiac cell death, suggesting prophylactic therapeutic options for acute ischemic cardiac injury.

BackgroundRecent advancements in artificial intelligence (AI) and large language models (LLMs) have shown potential in medical fields, including dermatology. With the introduction of image analysis capabilities in LLMs, their application in dermatological diagnostics has garnered significant interest. These capabilities are enabled by the integration of computer vision techniques into the underlying architecture of LLMs. ObjectiveThis study aimed to compare the diagnostic performance of Claude 3 Opus and ChatGPT with GPT-4 in analyzing dermoscopic images for melanoma detection, providing insights into their strengths and limitations. MethodsWe randomly selected 100 histopathology-confirmed dermoscopic images (50 malignant, 50 benign) from the International Skin Imaging Collaboration (ISIC) archive using a computer-generated randomization process. The ISIC archive was chosen due to its comprehensive and well-annotated collection of dermoscopic images, ensuring a diverse and representative sample. Images were included if they were dermoscopic images of melanocytic lesions with histopathologically confirmed diagnoses. Each model was given the same prompt, instructing it to provide the top 3 differential diagnoses for each image, ranked by likelihood. Primary diagnosis accuracy, accuracy of the top 3 differential diagnoses, and malignancy discrimination ability were assessed. The McNemar test was chosen to compare the diagnostic performance of the 2 models, as it is suitable for analyzing paired nominal data. ResultsIn the primary diagnosis, Claude 3 Opus achieved 54.9% sensitivity (95% CI 44.08%-65.37%), 57.14% specificity (95% CI 46.31%-67.46%), and 56% accuracy (95% CI 46.22%-65.42%), while ChatGPT demonstrated 56.86% sensitivity (95% CI 45.99%-67.21%), 38.78% specificity (95% CI 28.77%-49.59%), and 48% accuracy (95% CI 38.37%-57.75%). The McNemar test showed no significant difference between the 2 models (P=.17). For the top 3 differential diagnoses, Claude 3 Opus and ChatGPT included the correct diagnosis in 76% (95% CI 66.33%-83.77%) and 78% (95% CI 68.46%-85.45%) of cases, respectively. IntroductionBreeding rice with drought tolerance for harsh environments is crucial for agricultural sustainability. Understanding the genetic underpinnings of drought tolerance is vital for developing resilient rice varieties. Genome-wide association studies (GWAS) have emerged as pivotal tools in unravelling the complex genetic architecture of traits like drought tolerance, capitalizing on the natural genetic diversity within rice germplasm collections.MethodsIn this study, a comprehensive panel of 210 rice varieties was phenotyped over ten days in controlled conditions, subjected to simulated drought stress using 20% PEG 6000 in petri dishes. Throughout the stress period, crucial traits such as germination percentage (GP), germination rate index (GRI), mean germination time (MGT), and seedling percentage (SP) were meticulously monitored.ResultsThe GWAS analysis uncovered a total of 38 QTLs associated with drought tolerance traits, including novel loci like qMGT-5.2, qSP-3, qSP7.2, and qGP-5.2. Additionally, RNA-seq analysis identified ten genes with significant expression differences under drought stress conditions. Notably, haplotype analysis pinpointed elite haplotypes in specific genes linked to heightened drought tolerance.DiscussionOverall, this study underscores the importance of GWAS in validating known genes while unearthing novel loci to enrich the genetic resources for enhancing drought tolerance in rice breeding programs.

Background: Most ovarian tumors exhibit a pure histological characteristic. Nevertheless, a combination of tumors with the same histogenetic origin but different histologic subtypes is relatively common. Additionally, co-occurrence of tumors with different histogenetic origins is very rare. Typically, these mixed tumors include mixed epithelial tumors, mixed epithelial-stromal tumors, mixed germ cell-sex cord-stromal tumors, and mixed germ cell tumors. However, mixed epithelial-sex cord stromal-lymphohematopoietic system tumors are rare. Currently, clinicians have limited knowledge of this type of tumor, and the epidemiology, diagnosis, and treatment of this disease are yet to be established. Case presentation: We report a case of a 73-year-old woman with abdominal distension and pain for three months. Imaging evaluation revealed a large pelvic mass, with ultrasound suggesting a benign ovarian cyst along with leiomyoma. Furthermore, computed tomography (CT) and magnetic resonance imaging (MRI) revealed a malignant tumor. Blood tests showed significant increases in CA125 and CA199 levels. The patient underwent a total abdominal hysterectomy and bilateral salpingo-oophorectomy. During the surgery, a large multinodular cystic solid mass was observed in the right ovary, and the maximum nodular diameter was 14.2 cm. The solid areas of the mass appeared gray-white and taupe, whereas the cystic areas contained clear liquid with smooth walls 0.2 cm thick and no intracystic solid areas. The left ovary had solitary nodules, the largest being 4 cm in diameter. Microscopic examination of the right ovary revealed three different cell types. The first type of cell area was analogous, round, fusiform, and staggered mixed cells with unclear boundaries and rare nucleolus or mitosis. The second type of cell area was the cystic dilatation area. The cyst wall was covered with a single layer of flat epithelium, rich eosinophilic cytoplasm, uniform nuclear chromatin, and no papillary structures. The third type was a diffuse lymphoid region with uniform medium-sized cells, rough nuclear chromatin and evident nucleoli and mitosis. The morphology of the left ovarian cell was single, which was consistent with the first type of cell area in the right ovary. Immunohistochemistry of the right ovary indicated that the first region expressed vimentin, inhibin-α, calretinin, SF-1, WT-1 and CD56, with Ki-67 at 5 %, and no CKpan expression. The second region expressed CKpan, with Ki-67 at 1 %. The third region expressed CD20, Pax-5, Bcl-6, Bcl-2, MUM1, CD45, and C-myc, with Ki-67 at 70 %, and positive IGH clonal gene rearrangement. Lastly, the pathological diagnosis was a mixed ovarian tumor in the right ovary, comprising thecoma-fibroma, serous cystadenoma, diffuse large B-cell lymphoma, and a thecoma-fibroma in the left ovary. A follow-up examination of the patient after 15 months showed no mass or lymph node enlargement in other parts of the body, and no recurrence or metastasis was observed. Conclusions: We present a case of a postmenopausal woman with a rare combination of thecoma-fibroma, serous cystadenoma and diffuse large B-cell lymphoma in the ovary. To the best of our knowledge, this is the first reported case of such a combination. Typical pathological morphology and immunohistochemistry are crucial for the diagnosis of this disease. Owing to the limited knowledge of the disease, its pathogenesis and tissue origin are unknown. Clinicians should be careful about such patients. We believe this case report may provide some novel insights into the diagnosis and therapy of patients with this type of tumor.

ObjectivesThe objective of this study is to investigate the indirect causalities between gut microbiota and sleep disorders.MethodsIn stage 1, we utilized 196 gut microbiota as the exposure factor and conducted a two-sample univariable Mendelian randomization (MR) analysis on five sleep disorders: insomnia, excessive daytime sleepiness (EDS), sleep-wake rhythm disorders (SWRD), obstructive sleep apnea (OSA), and isolated REM sleep behavior disorder (iRBD). In stage 2, we validated the MR findings by comparing fecal microbiota abundance between patients and healthy controls through 16S rDNA sequencing. In stage 3, we explored the indirect pathways by which the microbiota affects sleep, using 205 gut microbiota metabolic pathways and 9 common risk factors for sleep disorders as candidate mediators in a network MR analysis.ResultsIn stage 1, the univariable MR analysis identified 14 microbiota potentially influencing five different sleep disorders. In stage 2, the results from our observational study validated four of these associations. In stage 3, the network MR analysis revealed that the Negativicutes class and Selenomonadales order might worsen insomnia by increasing pain [mediation: 12.43% (95% CI: 0.47, 24.39%)]. Oxalobacter could raise EDS by disrupting adenosine reuptake [25.39% (1.84, 48.95%)]. Allisonella may elevate OSA risk via obesity promotion [36.88% (17.23, 56.54%)], while the Eubacterium xylanophilum group may lower OSA risk by decreasing smoking behavior [7.70% (0.66, 14.74%)].ConclusionTriangulation of evidence from the MR and observational study revealed indirect causal relationships between the microbiota and sleep disorders, offering fresh perspectives on how gut microbiota modulate sleep.

ObjectivePreoperative obstructive sleep apnea (OSA) is supposed to be the abnormally high occurrence of OSA the night before surgery under general anesthesia. This study aimed to evaluate the prevalence preoperative OSA using cardiopulmonary coupling (CPC) and its correlation with imbalance of sympathetic/parasympathetic nervous system.MethodsA total of 550 patients with plans to receive surgery under general anesthesia were enrolled. Soil is the basis of the existence of fruit tree and soil nutrients plays a crucial role in plant growth and berry quality. To investigate the characteristics and interrelationships between soil nutrients and berry quality in Cabernet Gernischet vineyards, this study focused on seven representative vineyards in the eastern foothills of the Helan Mountains. Fifteen soil physicochemical factors and 10 berry quality factors were measured, followed by variation analysis, correlation analysis, multiple linear regression (MLR), partial-least squares regression (PLSR), principal component analysis (PCA), and systematic cluster analysis. We identified the main soil nutrient indicators influencing berry quality and developed linear regression equations. Utilizing PCA, a comprehensive evaluation model for berry quality was constructed, which enabled the calculation and ranking of integrated berry quality scores. The results indicated that soil nutrients in the vineyards of the eastern foothills of the Helan Mountains are relatively deficient and alkaline. The coefficient of variation for soil nutrient factors ranged from 3.19 to 118.08% and for berry quality factors 2.41–26.37%. Correlation analysis revealed varying degrees of correlation between soil nutrient indicators and fruit quality indicators. PCA extracted four principal components with a cumulative contribution rate of 91.506%. Based on the scores of these components and their corresponding weights, a comprehensive model for evaluating the quality of Cabernet Gernischet berries was established. The vineyards were ranked from the highest to the lowest combined scores as Zhenbeibu (ZBB), Yuquanying (YQY), Dawukou (DWK), Beihaizi (BHZ), Shuxin (SX), Huangyangtan (HYT), and Hongde (HD). These findings provide insights into soil nutrient management and comprehensive quality assessment of vineyards in the eastern foothills of the Helan Mountains. In conclusion, this study offers a theoretical foundation for vineyard managers to enhance grape berries quality through soil nutrient management. This will aid in the diagnosis of vineyard soil nutrition and the efficient use of fertilizers, with critical practical and theoretical implications for the meticulous management of vineyards and the production of high-quality wines.

BackgroundKawasaki disease (KD) has been considered as the most common required pediatric cardiovascular diseases among the world. However, the molecular mechanisms of KD were not fully underlined, leading to a confused situation in disease management and providing precious prognosis prediction. The disorders of gut microbiome had been identified among several cardiovascular diseases and inflammation conditions. Therefore, it is urgent to elucidate the characteristics of gut microbiome in KD and demonstrate its potential role in regulating intravenous immunoglobulin (IVIG) resistance and coronary artery injuries.MethodsA total of 96 KD children and 62 controls were enrolled in the study. One hundred forty fecal samples had been harvested from KD patients, including individuals before or after IVIG treatment, with or without early coronary artery lesions and IVIG resistance. Fecal samples had been collected before and after IVIG administration and stored at −80°C. Then, metagenomic analysis had been done using Illumina NovaSeq 6000 platform. After that, the different strains and functional differences among comparisons were identified.ResultsFirst, significant changes had been observed between KD and their controls. We found that the decrease of Akkermansia muciniphila, Faecalibacterium prausnitzii, Bacteroides uniformis, and Bacteroides ovatus and the increase of pathogenic bacteria Finegoldia magna, Abiotrophia defectiva, and Anaerococcus prevotii perhaps closely related to the incidence of KD. Then, metagenomic and responding functional analysis demonstrated that short-chain fatty acid pathways and related strains were associated with different outcomes of therapeutic efficacies. Among them, the reduction of Bacteroides thetaiotaomicron, the enrichment of Enterococcus faecalis and antibiotic resistance genes had been found to be involved in IVIG resistance of KD. Moreover, our data also revealed several potential pathogenetic microbiome of that KD patients with coronary artery lesionsConclusionThese results strongly proved that distinct changes in the gut microbiome of KD and the dysfunction of gut microbiomes should be responsible for the pathogenesis of KD and significantly impact the prognosis of KD.

ObjectivesThe emergency response to the COVID-19 pandemic may disrupt hospital management activities of antimicrobial resistance (AMR). This study aimed to determine the changing AMR trend over the period in China when stringent COVID-19 response measures were implemented.MethodsThis retrospective study was conducted in a designated hospital for COVID-19 patients in Guangzhou, China from April 2018 to September 2021. The prevalence of 13 antimicrobial-resistant bacteria was compared before and after the COVID-19 responses through Chi-square tests. Interrupted time series (ITS) models on the weekly prevalence of AMR were established to determine the changing trend. Controlled ITS models were performed to compare the differences between subgroups.ResultsA total of 10,134 isolates over 1,265 days were collected. PurposeSARS-CoV-2 can invade the thyroid gland. This study was to delineate the risk of thyroid dysfunction amidst the prevalence of the Omicron variant, and to investigate the correlation between thyroid function and Coronavirus disease 2019 (COVID-19) outcomes. The study also aimed to ascertain whether thyroid dysfunction persisted during COVID-19 recovery phase.MethodsThis was a retrospective cohort study. COVID-19 patients from the Renmin Hospital of Wuhan University, China during the epidemic of Omicron variants were included, and their thyroid function were analyzed in groups.ResultsA history of thyroid disease was not associated with COVID-19 outcomes. COVID-19 can lead to a bimodal distribution of thyroid dysfunction. The severity of COVID-19 was inversely proportional to the levels of thyroid- stimulating hormone (TSH), free triiodothyronine (FT3) and free thyroxine (FT4), leading to a higher prevalence of thyroid dysfunction. Severe COVID-19 was a risk factor for euthyroid sick syndrome (ESS) (OR=22.5, 95% CI, 12.1 - 45.6). Neutrophil to lymphocyte ratio mediated the association between severe COVID-19 and ESS (mediation effect ratio = 41.3%, p < 0.001). ESS and decreased indicators of thyroid function were associated with COVID-19 mortality, while high levels of FT3 and FT4 exhibited a protective effect against death. This effect was more significant in women (p < 0.05). During the recovery period, hyperthyroidism was quite uncommon, while a small percentage of individuals (7.7%) continued to exhibit hypothyroidism.ConclusionCOVID-19 severity was linked to thyroid dysfunction. Severe COVID-19 increased the risk of ESS, which was associated with COVID-19 mortality. Post-recovery, hyperthyroidism was rare, but some individuals continued to have hypothyroidism.

The clinical use of JAK1 inhibitors upadacitinib and abrocitinib in the treatment of refractory rosacea has rarely been explored.Case reportWe presented two cases of patients who received the JAK1 inhibitor upadacitinib and four cases of patients who received the JAK1 inhibitor abrocitinib for the treatment of refractory rosacea.DiscussionThe JAK1 inhibitors upadacitinib and abrocitinib may be promising medical options for patients with refractory rosacea. However, the long-term safety and efficacy of upadacitinib and abrocitinib require prospective controlled studies to assess them more comprehensively.

Exploring the relationship between sensory perception and brain responses holds important theoretical and clinical implications. However, commonly used methodologies like correlation analysis performed either intra- or inter- individually often yield inconsistent results across studies, limiting their generalizability. Representational similarity analysis (RSA), a method that assesses the perception-response relationship by calculating the correlation between behavioral and neural patterns, may offer a fresh perspective to reveal novel findings. Here, we delivered a series of graded sensory stimuli of four modalities (i.e., nociceptive somatosensory, non-nociceptive somatosensory, visual, and auditory) to/near the left or right hand of 107 healthy subjects and collected their single-trial perceptual ratings and electroencephalographic (EEG) responses. We examined the relationship between sensory perception and brain responses using within- and between-subject correlation analysis and RSA, and assessed their stability across different numbers of subjects and trials. We found that within-subject and between-subject correlations yielded distinct results: within-subject correlation revealed strong and reliable correlations between perceptual ratings and most brain responses, while between-subject correlation showed weak correlations that were vulnerable to the change of subject number. In addition to verifying the correlation results, RSA revealed some novel findings, i.e., correlations between behavioral and neural patterns were observed in some additional neural responses, such as “γ-ERS” in the visual modality. RSA results were sensitive to the trial number, but not to the subject number, suggesting that consistent results could be obtained for studies with relatively small sample sizes. In conclusion, our study provides a novel perspective on establishing the relationship between behavior and brain activity, emphasizing that RSA holds promise as a method for exploring this pattern relationship in future research.

Abstract Introduction Triptolide (TP), a natural product derived from the herbal medicine Tripterygium wilfordii, exhibits potent immunosuppressive activity. However, the mechanisms underlying its effects in rheumatoid arthritis remain incompletely understood. Methods Collagen‐induced arthritis (CIA) model was induced in Sprague−Dawley rats by immunization with bovine type II collagen, and TP was administrated as treatment. The therapeutic effect of TP was evaluated based on paw swelling, histopathology, and serum levels of inflammatory factors. Exosomes isolated from rat serum were characterized by transmission electron microscopy, dynamic light scattering, and western blot analysis. Proteomic profiling of exosomes was analyzed by direct DIA quantitative proteomics analysis. Gene ontology and the Kyoto Encyclopedia of Genes and Genomes databases were employed for enrichment analysis related to molecular function, biological processes, and signaling pathways. Western blot analysis was used to analyze differentially expressed proteins. Results TP treatment ameliorated arthritic phenotypes in CIA rats as evidenced by reduced arthritis score, paw swelling, pathological injury severity scores, and serum levels of inflammatory cytokines. The proteomic analysis revealed that TP treatment significantly inhibited complement and coagulation cascades, interleukin‐17 signaling pathway, and cholesterol metabolism, which were reactivated in CIA rats. Importantly, lipocalin 2 (LCN2) and myeloperoxidase (MPO) levels were markedly upregulated in the CIA group but suppressed upon TP administration. Furthermore, in synovial tissues, LCN2 and MPO expression levels were also elevated in the CIA group but decreased following TP treatment. Conclusion Our findings demonstrate that TP alleviates CIA, possibly through modulation of exosomal LCN2 and MPO proteins.

Sarcopenia is a condition characterized by age-related loss of muscle mass and strength. Increasing evidence suggests that patients with sarcopenia have higher rates of coronavirus 2019 (COVID-19) infection and poorer post-infection outcomes. However, the exact mechanism and connections between the two is unknown. In this study, we used high-throughput data from the GEO database for sarcopenia (GSE111016) and COVID-19 (GSE171110) to identify common differentially expressed genes (DEGs). We conducted GO and KEGG pathway analyses, as well as PPI network analysis on these DEGs. Using seven algorithms from the Cytoscape plug-in cytoHubba, we identified 15 common hub genes. Further analyses included enrichment, PPI interaction, TF-gene and miRNA-gene regulatory networks, gene-disease associations, and drug prediction. Additionally, we evaluated immune cell infiltration with CIBERSORT and assessed the diagnostic accuracy of hub genes for sarcopenia and COVID-19 using ROC curves. In total, we identified 66 DEGs (34 up-regulated and 32 down-regulated) and 15 hub genes associated with sarcopenia and COVID-19. GO and KEGG analyses revealed functions and pathways between the two diseases. TF-genes and TF-miRNA regulatory network suggest that FOXOC1 and hsa-mir-155-5p may be identified as key regulators, while gene-disease analysis showed strong correlations with hub genes in schizophrenia and bipolar disorder. Immune infiltration showed a correlation between the degree of immune infiltration and the level of infiltration of different immune cell subpopulations of hub genes in different datasets. The ROC curves for ALDH1L2 and KLF5 genes demonstrated their potential as diagnostic markers for both sarcopenia and COVID-19. This study suggests that sarcopenia and COVID-19 may share pathogenic pathways, and these pathways and hub genes offer new targets and strategies for early diagnosis, effective treatment, and tailored therapies for sarcopenia patients with COVID-19.

Objectives: To evaluate the impact of pay-for-performance on antimicrobial consumption and antimicrobial expenditure in a large teaching hospital in Guangzhou, China. Methods: We collected data from hospital information system from January 2018 through September 2022 in the inpatient wards. Antimicrobial consumption was evaluated using antibiotic use density (AUD) and antibiotic use rate (AUR). The economic impact of intervention was assessed by antimicrobial expenditure percentage.
