We establish stability and characteristics of two-dimensional (2D) vortex ring-shaped quantum droplets (QDs) formed by binary Bose-Einstein condensates (BECs). The system is modeled by the Gross-Pitaevskii (GP) equation with the cubic term multiplied by a logarithmic factor (as produced by the Lee-Huang-Yang correction to the mean-field theory) and a potential which is a periodic function of the radial coordinate. Narrow vortex rings with high values of the topological charge, trapped in particular circular troughs of the radial potential, are produced. These results suggest an experimentally relevant method for the creation of vortical QDs (thus far, only zero-vorticity ones have been reported). The 2D GP equation for the narrow rings is approximately reduced to the 1D form, which makes it possible to study the modulational stability of the rings against azimuthal perturbations. Full stability areas are delineated for these modes. The trapping capacity of the circular troughs is identified for the vortex rings with different winding numbers (WNs). Stable compound states in the form of mutually nested concentric multiple rings are constructed too, including ones with opposite signs of the WNs. Other robust compound states combine a modulationally stable narrow ring in one circular potential trough and an azimuthal soliton performing orbital motion in an adjacent one. 4. Research on carbon emissions during the construction process of prefabricated buildings based on BIM and LCA

The goal of double carbon reflects the new goals and requirements of China’s entry into a new development stage. In this regard, the construction industry, as a large producer of carbon emissions, should focus on emission reduction. In the whole life cycle of buildings, the carbon emission intensity in the physicochemical stage is the largest, so we should focus on the carbon emission in the physicochemical stage. Compared with the cast-in-situ concrete floor slab, the bidirectional prestressed precast hollow slab (BPPHS) has the characteristics of environment-friendly under the condition of meeting the building stress, and the manufacturing process of precast floor slab is more standardized, which can give full play to the performance of components. Based on BIM (Building Information Modeling) and LCA (Life Cycle Assessment) theory, and in the case of tram transportation, this paper studies the carbon emission of prefabricated hollow floor slab used in prefabricated buildings in the building stage. The research shows that the use of BPPHS reduces the carbon emission of 197,332.81 kg in the whole physicochemical stage, and the emission reduction rate is 46.52% compared with the cast-in-place slab. Compared with the ordinary prefabricated laminated plate, the emission reduction rate reaches 45.40%; The carbon emission sources in the materialization stage of prefabricated buildings are divided into production stage, transportation stage and construction stage. The carbon emission reduction in each stage accounts for 42.29%, 46.18% and 51.08%, respectively, in each stage of cast-in-situ floor slab. Compared with ordinary precast slab, the emission reduction rates of each stage are 34.67%, 53.60% and 47.84%, respectively. The emission reduction potential of BPPHS is very considerable. It can not only promote the development of prefabricated buildings but also have lower carbon than conventional precast slab. The study provides new ideas for reducing building carbon emissions, which will also provide reference for industry entrepreneurs and relevant governments to realize low-carbon economy and sustainable development.

5. Changes in the land-use landscape pattern and ecological network of Xuzhou planning area

Abstract Ongoing rapid urbanization has triggered significant changes in land use, rendering landscape patterns adversely impacted and certain habitat patches degraded. Ecological networks have consequently contracted overall. As such, an investigation into how land-use landscape patterns and ecological networks change over time and space is of major significance for ecological restoration and regional sustainability. Taking Xuzhou Planning Area as a case study, we examined spatiotemporal changes and features of the landscape pattern by employing the land-use change degree, the land-use transition matrix, and quantified landscape pattern indices. An ecological network analysis, which studies the changes in network connectivity and robustness, as well as their causes and contributors, was undertaken to probe into the features and trends of spatiotemporal changes in the land-use landscape pattern and ecological network amid expeditious urbanization. Analysis results unveiled the following: (1) From 1985 to 2020, there was a decline in the area of farmland, forest, and grassland, accompanied by an increase in land for construction, water bodies, and unused land. The southwestern research area witnessed farmland substantially give way to land for construction for this period, and the most dramatic change in land use occurred between 2000 and 2010. (2) The area of dominant patches in the research area shrank, along with more fragmented, complex landscapes. The land for construction was emerging as the dominant landscape by area, whereas patches of farmland, forest, grassland, and water bodies became less connected. (3) The ecological network was densely linked in the northeast, with sparser connections in the southwest. Spatial shrinkage was observed in the research area’s southwestern and central ecological corridors. Overall, the number of ecological sources and corridors rose and subsequently dropped before a rebound. (4) The ecological network grew more connected and robust from 1985 through 1990, as portions of farmland were converted into water bodies, which led to an increase in ecological sources. Given a reduction in ecological sources and corridors in the southwestern and central regions between 1990 and 2010, network connectivity and robustness declined, which was reversed from 2010 onward with the addition of two ecological sources—Pan’an Lake and Dugong Lake. With an optimal ecological network in 1990, however, it deteriorated significantly by 2010. The research area saw the minimum value of its network connectivity indices of network stability index (α), evenness index (β), and connectivity index (γ), in 2010, when its ecological network was highly fragmented and vulnerable, attributing to a strong contrast between the maximal connected subgraph’s relative size and connectivity robustness. The research findings can lay scientific groundwork for addressing ecological issues, restoring landscape patterns, and developing ecological networks amid urbanization.

7. METTL3-dependent m6A modification of PSEN1 mRNA regulates craniofacial development through the Wnt/β-catenin signaling pathway

Abstract Craniofacial malformations, often associated with syndromes, are prevalent birth defects. Emerging evidence underscores the importance of m6A modifications in various bioprocesses such as stem cell differentiation, tissue development, and tumorigenesis. Here, in vivo, experiments with zebrafish models revealed that mettl3-knockdown embryos at 144 h postfertilization exhibited aberrant craniofacial features, including altered mouth opening, jaw dimensions, ethmoid plate, tooth formation and hypoactive behavior. Similarly, low METTL3 expression inhibited the proliferation and migration of BMSCs, HEPM cells, and DPSCs. Loss of METTL3 led to reduced mRNA m6A methylation and PSEN1 expression, impacting craniofacial phenotypes. Co-injection of mettl3 or psen1 mRNA rescued the level of Sox10 fusion protein, promoted voluntary movement, and mitigated abnormal craniofacial phenotypes induced by mettl3 knockdown in zebrafish. Mechanistically, YTHDF1 enhanced the mRNA stability of m6A-modified PSEN1, while decreased METTL3-mediated m6A methylation hindered β-catenin binding to PSEN1, suppressing Wnt/β-catenin signaling. Pharmacological activation of the Wnt/β-catenin pathway partially alleviated the phenotypes of mettl3 morphant and reversed the decreases in cell proliferation and migration induced by METTL3 silencing. This study elucidates the pivotal role of METTL3 in craniofacial development via the METTL3/YTHDF1/PSEN1/β-catenin signaling axis.

The continuous supply of reliable and affordable energy sources is crucial for driving a nation's economic activities. The phenomenon of continuous energy supply at affordable prices is called energy security. To date, more than two-thirds of the global energy demand is met by non-renewable energy derived from natural resources. The empirical connection between natural resource depletion and energy security risk remains unexplored in the prior literature. Thus, this study analyzes the empirical linkage between natural resource depletion and energy security risk by applying the CS-ARDL and PMG-ARDL models. As per our findings, natural resource depletion increases long-term energy security risks Globally and in Asia, America, Europe, and Africa. In short run, the role of natural resource depletion is mostly insignificant in energy security risk. In contrast, the GDP, financial development, trade openness, and technological development help reduce long-run energy security risk in all regions except Africa. In the short run, only GDP and trade openness help reduce energy security risks in most regions. Therefore, policymakers should focus on efficiently utilizing natural resources to mitigate energy security risks.

9. Breeding a new Ganoderma lucidum strain with increased contents of individual ganoderic acids by mono–mono crossing of genetically modified monokaryons

Ganoderic acids (GAs) are major functional components of Ganoderma lucidum. The study aimed to breed a new G. lucidum strain with increased contents of individual GAs. Two mating-compatible monokaryotic strains, G. 260125 and G. 260124, were successfully isolated from the dikaryotic G. lucidum CGMCC 5.0026 via protoplast formation and regeneration. The Vitreoscilla hemoglobin gene (vgb) and squalene synthase gene (sqs) were overexpressed in the monokaryotic G. 260124 and G. 260125 strain, respectively. Mating between the G. 260124 strain overexpressing vgb and the G. 260125 strain overexpressing sqs resulted in the formation of the new hybrid dikaryotic G. lucidum strain sqs-vgb. The maximum contents of ganoderic acid (GA)-T, GA-Me, and GA-P in the fruiting body of the mated sqs-vgb strain were 23.1, 15.3, and 39.8 μg/g dry weight (DW), respectively, 2.23-, 1.75-, and 2.69-fold greater than those in G. lucidum 5.0026. The squalene and lanosterol contents increased 2.35- and 1.75-fold, respectively, in the fruiting body of the mated sqs-vgb strain compared with those in the G. lucidum 5.0026. In addition, the maximum expression levels of the sqs and lanosterol synthase gene (ls) were increased 3.23- and 2.13-fold, respectively, in the mated sqs-vgb strain. In summary, we developed a new G. lucidum strain with higher contents of individual GAs in the fruiting body by integrating genetic engineering and mono–mono crossing.

15. Clinical efficacy of integrated traditional Chinese and western medicine in the treatment of oligoasthenospermia in male infertility of kidney-yang deficiency type

This study examines the clinically comprehensive effects of Wuzi Shenqi Decoction, an acupoint application that combines traditional Chinese medicine with western medicine on male infertility with oligoasthenospermia of kidney-yang deficiency type. A total of 184 male infertiles with oligoasthenospermia who came to our hospital from April 2022 to June 2023 were randomly assigned to a study group (n = 92) and an experimental group (n = 92). The experimental group was treated with L-carnitine oral solution, while the study group was treated with Wuzi Shenqi Decoction combined with acupoint application based on the experimental group. Before and after treatment, clinical efficacy, semen parameters (semen volume, sperm density, sperm survival rate, sperm motility), sexual hormone parameters (follicle stimulating hormone (FSH), luteinizing hormone (LH), testosterone (T), prolactin (PRL), estradiol), and TCM (Traditional Chinese Medicine) syndrome scores were observed and compared between both groups. Before treatment, there was no significant difference in semen parameter index, sexual hormone index and TCM syndrome score between both groups (p > 0.05); after treatment, the semen parameter index and sexual hormone index in both groups were higher than before treatment, and the TCM syndrome score was diminished with a significant difference (p < 0.05). The overall response rate of the study group was 92.39%, which was significantly higher than that of the experimental group (72.83%) (p < 0.05). For male infertility patients with oligoasthenospermia of kidney-yang deficiency type, the use of Wuzi Shenqi Decoction combined with traditional Chinese medicine acupoint application plus L-carnitine oral solution can effectively improve the clinical symptoms of patients, improve semen quality, regulate sex hormone levels, which is worthy of clinical application.

Abstract The maternal-fetal interface shares similarities with tumor tissues in terms of the immune microenvironment. Normal pregnancy is maintained due to the immunosuppressed state, but pyroptosis induced by MITA can trigger the body’s immune response and disrupt the immunosuppressed state of the maternal-fetal interface, leading to abortion. In this study, we explored the role of MITA and TRIM38 in regulating pyroptosis and maintaining the immune tolerance of the maternal-fetal interface during pregnancy. Our findings show that the interaction between MITA and TRIM38 plays a crucial role in maintaining the immunosuppressed state of the maternal-fetal interface. Specifically, we observed that TRIM38-mediated K48 ubiquitination of MITA was higher in M2 macrophages, leading to low expression levels of MITA and thus inhibiting pyroptosis. Conversely, in M1 macrophages, the ubiquitination of K48 was lower, resulting in higher expression levels of MITA and promoting pyroptosis. Our results also indicated that pyroptosis played an important role in hindering the transformation of M1 to M2 and maintaining the immunosuppressed state of the maternal-fetal interface. These discoveries help elucidate the mechanisms that support the preservation of the immune tolerance microenvironment at the maternal-fetal interface, playing a vital role in ensuring successful pregnancy.

Objective: To evaluate the efficacy and prognosis of percutaneous vertebroplasty/kyphoplasty (PVP/PKP) in patients with newly diagnosed multiple myeloma (NDMM). Methods: Clinical data of NDMM patients who underwent PVP/PKP during front-line regimen at Peking Union Medical College Hospital from January 1, 2003, to June 30, 2023, were analyzed. Patients with comparable bone diseases not receiving orthopedic surgery were selected as controls. Visual analogue scale (VAS) score, progression-free survival (PFS), and overall survival (OS) were compared. Results: Baseline characteristics were matched between the surgical group (n = 51 with 56 surgeries) and non-surgical group (n = 102), including demographics, tumor load, International Staging System (ISS), bone diseases, cytogenetic abnormalities, first-line treatment, and autologous stem-cell transplantation (ASCT). Bone lesions for PVP/PKP were located at thoracic vertebrae (53.6 %, 30/56) or lumbosacral vertebrae (46.4 %, 26/56). Tumor-induced osteomalacia (TIO), also known as oncogenic osteomalacia, is very rare, with about 1000 reported cases globally. Removing most TIO culprit tumors requires the evaluation and intervention of orthopedic doctors. However, orthopedic doctors often have a poor understanding of the optical treatment of TIO due to its rarity. In addition, most TIO patients lack specific clinical manifestations. Also, the clinical localization and qualitative diagnosis of TIO are difficult and thus can easily be misdiagnosed and mistreated. Furthermore, the true incidence rate of TIO may be underestimated. Although many breakthroughs have been made in exploring the pathogenesis, clinical diagnosis, and treatment of TIO, rational and standardized orthopedic surgical treatment experience summary and sorting for TIO patients are lacking. In this article, the recent experience and progress in the field of orthopedic surgical treatment for TIO globally have been summarized, providing a theoretical basis and new clinical practice guidance for the rational treatment of TIO patients.

20. Surgical treatment for pelvic haemophilic pseudotumour: a retrospective analysis of 21 casesResearch in context

Summary: Background: Due to the rarity of pelvic haemophilic pseudotumour (PHPT) and demanding surgical technique for PHPT excision, no study reports the mid-term follow-up outcomes of surgical treatment of PHPT in a relatively large cohort. PHPT with varying degrees of bony pelvic involvement and infection status necessitates different operative procedures, yet there is currently no classification system for PHPT based on surgical practice. Methods: The study was conducted between June 25, 2004 and July 18, 2023, in Peking Union Medical College Hospital and Nanfang Hospital in China. We performed a retrospective analysis involving 21 patients with 24 PHPTs with a mean follow-up period of 7.1 years. The demographic information, PHPT characteristics, surgical data, and perioperative complications were analysed. Findings: 21 consecutive male patients with 24 PHPTs (21 primary PHPTs and three recurrent PHPTs) that underwent surgical treatment were involved in the study. A classification system including four subtypes was introduced as (I) PHPT confined to soft tissue; (II) PHPT involving bony pelvic without pelvic discontinuity; (III) PHPT causing pelvic discontinuity; (IV) Infectious PHPT. Of the 24 PHPTs, 11 (45.8%) were identified as Type I, five (20.8%) as Type II, three (12.5%) as Type III, and five (20.8%) as Type IV. At the time of surgery, the patients had a mean age of 37.0 ± 9.5 years (Range, 24–52 years). The mean maximum diameter of PHPTs upon surgery was 17.0 ± 7.7 cm (Range, 4.3–40.0 cm). The mean surgical duration was 192 ± 77 min (Range, 60–330 min) and the median intraoperative blood loss was 400 mL (IQR, 225–950 mL, Range, 100–3000 mL). One patient (4.8%) underwent intraoperative cardiopulmonary arrest and expired the following week. Four PHPTs (16.7%) presented postoperative wound infections and poor wound healing. During the follow-up period, five PHPTs (20.8%) experienced pseudotumour recurrence. Interpretation: Our findings suggest that surgical treatment for PHPTs is feasible and relatively safe. Symptomatic and progressive PHPTs should undergo surgical intervention as early as possible to minimise the surgical risks. Intraoperative use of abundant gelatin sponges in PHPT excision draws attention to severe embolism complications. Funding: There are no sources of funding for this manuscript.

Objectives Anthracycline-induced cardiotoxicity is a debilitating cardiac dysfunction for which there are no effective treatments, making early prevention of anthracycline-induced subclinical cardiotoxicity (AISC) crucial. High-density lipoprotein cholesterol (HDL-C) plays a role in cardioprotection, but its impact on AISC remains unclear. Our study aims to elucidate the protective capacity of HDL-C in AISC in patients with diffuse large B-cell lymphoma (DLBCL) treated with R-CHOP (cyclophosphamide, vincristine, doxorubicin, prednisone and rituximab).Design Prospective observational study.Setting Conducted in China from September 2020 to September 2022.Participants 70 chemotherapy-naïve patients newly diagnosed with DLBCL who were scheduled to receive the standard dose of R-CHOP; 60 participants included in a case–control study (DOI: 10.1186/s12885-022-10085-6).Primary outcome measures Serum biomarkers, 2D speckle tracking echocardiography and conventional echocardiography were measured at baseline, at the end of the third and sixth cycles of R-CHOP and 6 and 12 months after chemotherapy.Results 24 patients experienced AISC, while 10 did not. 36 patients were lost to follow-up and death. Cox regression analysis showed that higher levels of HDL-C were associated with a significantly lower risk of AISC (unadjusted HR=0.24, 95% CI 0.09 to 0.67, p=0.006; adjusted HR=0.27, 95% CI 0.09 to 0.79, p=0.017). Patients without AISC had a more stable and higher HDL-C level during the follow-up period. HDL-C levels significantly decreased from the end of the third cycle of chemotherapy to the end of the sixth cycle of chemotherapy in all patients (p=0.034), and particularly in the AISC group (p=0.003). The highest level of HDL-C was significantly higher in patients without AISC than in those with AISC (1.52±0.49 vs 1.22±0.29, p=0.034).Conclusions Our study suggests that higher HDL-C levels may associate with lower AISC risk in patients with DLBCL treated with R-CHOP. HDL-C could be a cardioprotective target, but further research is needed to confirm its benefits and limitations.Study registration number Study registration number: ChiCTR2100054721

To overcome the drawbacks of the existing sinusoidal map and tent map, this paper proposes the design of a sinusoidal–tent (ST) map. The test results indicate that the new chaotic system exhibits more significant advantages in chaos control. Compared with the sinusoidal map and tent map, the proposed sinusoidal–tent map performs better in terms of bifurcation diagram and Lyapunov exponents. The trajectories of the sinusoidal–tent map can occupy all the phase planes over (0,4), while those of the two classic maps only occupy a small phase space, and the Lyapunov exponents of the ST map are all positive within the range of control parameters, higher than those of seed maps. Simultaneously, a novel quantum scrambling operation is devised based on the sinusoidal–tent map to avoid the periodicity of the quantum Arnold scrambling method. Initially, two chaotic sequences are generated to scramble the pixel positions of the watermark image, further enhancing the security of the watermarking algorithm. Subsequently, the host image is processed by the quantum discrete cosine transform, and finally, the scrambled watermark image is inserted into the medium-frequency band of the transformed host image, ensuring the invisibility of the watermarking. According to the simulation results, the quantum watermarking algorithm has excellent invisibility and robustness.

23. Influencing factors of anthracycline-induced subclinical cardiotoxicity in acute leukemia patients

Abstract Background Current treatment of acute leukemia is based on anthracycline chemotherapy. Anthracyclines, despite improving patient survival, have serious cardiotoxicity and therefore cardiac monitoring should be a priority. The purpose of this study is to explore the possible early predictors of anthracycline-induced subclinical cardiotoxicity(AISC)in acute leukemia patients. Methods We conducted a prospective observational study involving 51 patients with acute leukemia treated with anthracycline. Demographic data, clinical variables, echocardiography variables and biochemical variables were collected at baseline and after 3 cycles of chemotherapy. Patients were divided into the AISC and No-AISC groups according to changes of global longitudinal peak systolic strain. Regression models and receiver operating characteristic curve analysis were used to explore the relationship between the variables and AISC. Result 17 of the patients suffered subclinical cardiotoxicity after 3 cycles of anthracycline treatment. Multiple logistic regression analysis showed a significant association of DBil (OR 0.612, 95% CI 0.409–0.916, p = 0.017), TBil (OR 0.841, 95% CI 0.717–0.986, p = 0.033), PLT (OR 1.012, 95% CI 1.002–1.021, p = 0.016) and Glu (OR 1.873, 95% CI 1.009–3.475, p = 0.047) with the development of AISC. After 3 cycles of chemotherapy, there was a significant difference in PLT between the AISC and NO-AISC groups. Moreover, the dynamic changes in PLT from baseline to after 3 cycles of chemotherapy were each statistically significant in the AISC and NO-AISC groups. The combination of PLT and N-terminal pro–B-type natriuretic peptide (NT-proBNP) had the highest area under curves (AUC) for the diagnosis of AISC than PLT and NT-proBNP alone (AUC = 0.713, 95%CI: 0.56–0.87, P = 0.017). Conclusion Total bilirubin (TBil), direct bilirubin (DBil), platelets (PLT) and blood glucose (Glu) are independent influencing factors for AISC in acute leukemia patients receiving anthracycline therapy. Bilirubin may be a protective factor and PLT may be a contributing factor for AISC. The combination of baseline PLT and baseline NT-proBNP shows satisfactory predictive ability for AISC in acute leukemia cases treated with 3 cycles of chemotherapy.

Abstract Background Tooth agenesis is a common dental anomaly that can substantially affect both the ability to chew and the esthetic appearance of patients. This study aims to identify possible genetic factors that underlie various forms of tooth agenesis and to investigate the possible molecular mechanisms through which human dental pulp stem cells may play a role in this condition. Results Using whole-exome sequencing of a Han Chinese family with non-syndromic tooth agenesis, a rare mutation in FGFR1 (NM_001174063.2: c.103G > A, p.Gly35Arg) was identified as causative and confirmed by Sanger sequencing. Via GeneMatcher, another family with a known variant (NM_001174063.2: c.1859G > A, p.Arg620Gln) was identified and diagnosed with tooth agenesis and a rare genetic disorder with considerable intrafamilial variability. Fgfr1 is enriched in the ectoderm during early embryonic development of mice and showed sustained low expression during normal embryonic development of Xenopus laevis frogs. Functional studies of the highly conserved missense variant c.103G > A showed deleterious effects. FGFR1 (c.103G > A) was overexpressed compared to wildtype and promoted proliferation while inhibiting apoptosis in HEK293 and human dental pulp stem cells. Moreover, the c.103G > A variant was found to suppress the epithelial-mesenchymal transition. The variant could downregulate ID4 expression and deactivate the TGF-beta signaling pathway by promoting the expression of SMAD6 and SMAD7. Conclusion Our research broadens the mutation spectrum associated with tooth agenesis and enhances understanding of the underlying disease mechanisms of this condition.

AIM: To investigate the clinical efficacy of diquafosol sodium(DQS)eye drops combined with Qingrun Yangmu oral liquid in the treatment of dry eye after phacoemulsification and intraocular lens implantation.METHODS:A total of 57 patients(65 eyes)with dry eye after cataract surgery, who were admitted to the ophthalmology department of our hospital from September 2020 to January 2021, were selected as the research objects. They were divided into a control group(28 cases, 32 eyes, treated with 3% DQS eye drops)and an observation group(29 cases, 33 eyes, treated with 3% DQS eye drops combined with the Qingrun Yangmu oral liquid)based on a random number table method. Indicators, such as ocular surface disease index(OSDI), non-invasive tear film break-up time(NIBUT), Schirmer I test(SIt), corneal fluorescein sodium staining(CFS), and TCM symptom score, were compared and analyzed between the two groups 1d before, 1wk after(before treatment), and 1mo after(after treatment)surgery.RESULTS:The total effective rates of the observation group and the control group after treatment were 88% and 75%, respectively. Over the last few decades, policy changes have resulted in changes in family structure and cyclical changes within families. The structure of multi-child families will have a significant influence on housing demand and supply, necessitating a thorough demand study. This study examines the requirements of 739 multi-child families in the Yangtze River Delta (YRD) region at different stages and graphically displays the variables influencing their level of housing satisfaction, offering a scientific reference for the design and optimization of housing. Firstly, the residential elements that impact families with multiple children were categorized into 3 categories, 14 subcategories, and 65 influencing elements based on expert consultation and word frequency data. Secondly, 739 families in the YRD region were chosen for investigation, and importance–performance analysis (IPA) was employed to study and assess the residential elements of multi-child families. The IPA study findings were compared to those obtained from on-site surveys and network data crawling. Finally, the residential elements to be optimized were quantitatively determined, and the relevant optimization techniques were provided in conjunction with door-to-door interviewing. This study examines the needs of multi-child families at various phases, as well as the factors that impact their residential satisfaction, and provides optimization solutions for the long-term use and renewal of residential structures.

Abstract Purpose Among high‐risk acute lymphoblastic leukemia (ALL) patients undergoing allogeneic hematopoietic stem cell transplantation (allo‐HSCT), those with positive minimal residual disease (MRD) are susceptible to poor outcomes. Therefore, it is necessary to determine the most suitable preparatory regimen for these patients. Methods Data were analyzed from 141 patients who received allo‐HSCT and were diagnosed with high‐risk ALL. These patients underwent intensified conditioning regimens, including either total marrow and lymphoid irradiation (TMLI)‐etoposide (VP16)‐cyclophosphamide (CY) or busulfan (BU)‐VP16‐CY between October 2016 and November 2022. A total of 141 individuals were in complete remission (CR) before transplantation and, among all patients, 90 individuals exhibited a negative MRD status and 51 patients had a positive MRD status. Results In patients who tested negative for MRD, the incidence of relapse within a 2‐year timeframe was 25.0% (24.8%–25.5%), compared with 32.2% (31.2%–33.2%) in MRD‐positive patients; however, this difference was not statistically significant. There were no significant differences in the 2‐year disease‐free survival (DFS) and 2‐year overall survival (OS) rates between the MRD‐negative and MRD‐positive groups (DFS: 67.2% (57.9%–78.1%) vs. 55.5% (42.6%–72.3%); OS: 69.0% (61.9%–88.2%) vs. 66.7% (53.9%–82.5%)). Furthermore, no notable variations were observed in the occurrence of transplant‐related mortality (TRM) and graft‐versus‐host disease (GVHD) across the two groups. Conclusion This study reveals the benefits of TMLI‐VP16‐CY and BU‐VP16‐CY conditioning regimens in high‐risk ALL patients with CR and MRD‐positive status. A large‐scale prospective clinical trial is warranted in the future.

Breast cancer is one of the most common female malignant tumors today and represents a serious health risk for women. Although the survival rate and quality of life of patients with breast cancer are improving with the continuous development of medical technology, metastasis, recurrence, and drug resistance of breast cancer remain a significant problem. Huaier, a traditional Chinese medicine (TCM) fungus, is a type of Sophora embolism fungus growing on old Sophora stems. The polysaccharides of Trametes robiniophila Murr (PS-T) are the main active ingredient of Huaier. There is increasing evidence that Huaier has great potential in breast cancer treatment, and its anti-cancer mechanism may be related to a variety of biological activities, such as the inhibition of cell proliferation, metastasis, tumor angiogenesis, the promotion of cancer cell death, and regulation of tumor-specific immunity. There is growing evidence that Huaier may be effective in the clinical treatment of breast cancer. This review systematically summarizes the basic and clinical studies on the use of Huaier in the treatment of breast cancer, providing useful information to guide the clinical application of Huaier and future clinical studies.

Summary: The rationality of block scale and structure is the guarantee of vitality and humanization quality, facing complex and diverse problems, and the structure reconstruction of urban built-up areas is one of the most difficult challenges in the process of promoting the block system. The traditional planning-construction strategy, as practice in recent years has demonstrated, faces challenges in terms of construction costs, demolition costs, property rights, and jurisprudence. Based on the interaction of the block structure and external space, the study presents a “implant-reconstruction” model. It investigates possibilities of implanting elements and graphically depicts the potential impact of implant-reconstruction using Space Syntax. The findings indicate that the external space implantation mode may actively encourage the construction of small-scale blocks and has benefits in terms of texture respect, low impact, and ease of operation. The simulation and pre-judgment dynamically illustrate the viable path and gives a scientific reference for block alteration.

CRISPR-Cas12a has been used for genome editing and molecular diagnosis. The well-studied Cas12a orthologs have a T-rich PAM and are usually categorized as non-thermally stable enzymes. Here, we identified a new Cas12a ortholog from Clostridium thermobutyricum, which survives at 60 °C. This Cas12a ortholog is named as CtCas12a and exhibits low sequence similarity to the known Cas12a family members. CtCas12a is active in a wide temperature range from 17 to 77 °C. Moreover, this ortholog has a relaxed PAM of YYV (Y=C or T, V = A or C or G). We optimized the conditions for trans-cleavage and enabled its detection of nucleic acids. CtCas12a executed genome editing in human cells and generated up to 26% indel formation in the EGFP locus. With the ability to be active at high temperatures as well as having a relaxed PAM sequence, CtCas12a holds potential to be further engineered for pathogen detection and editing a wide range of genomic sequences.

ABSTRACTThe Nav1.9 channel is a voltage-gated sodium channel. It plays a vital role in the generation of pain and the formation of neuronal hyperexcitability after inflammation. It is highly expressed in small diameter neurons of dorsal root ganglions and Dogiel II neurons in enteric nervous system. The small diameter neurons in dorsal root ganglions are the primary sensory neurons of pain conduction. Nav1.9 channels also participate in regulating intestinal motility. Functional enhancements of Nav1.9 channels to a certain extent lead to hyperexcitability of small diameter dorsal root ganglion neurons. The hyperexcitability of the neurons can cause visceral hyperalgesia. Intestinofugal afferent neurons and intrinsic primary afferent neurons in enteric nervous system belong to Dogiel type II neurons. Their excitability can also be regulated by Nav1.9 channels. The hyperexcitability of intestinofugal afferent neurons abnormally activate entero-enteric inhibitory reflexes. The hyperexcitability of intrinsic primary afferent neurons disturb peristaltic waves by abnormally activating peristaltic reflexes. This review discusses the role of Nav1.9 channels in intestinal hyperpathia and dysmotility.

Abstract Differential evolution (DE) is one of the most efficient evolution algorithms (ES) for dealing with nonlinear, complicated and difficult global optimization problems. The main contribution of this paper can be summarized in three directions: Firstly, a novel crossover rate (CR) generation scheme based on the zscore value of fitness, named fcr, is introduced. For a minimization problem, the proposed CR generation strategy always assigns a smaller CR value to individual with smaller fitness value. Therefore, the parameters of individuals with better fitness are inherited by their offspring with high probability. In the second direction, the control parameters are adjusted by unused bimodal settings in which each parameter setting is selected according to the evolution status of individual. The third direction of our work is introducing the L1 norm distance as the weights for updating the mean value of crossover rate and scale factor. Theoretically, compared with L2 norm, L1-norm is more efficient to suppress outliers in the difference vector. These modifications are first integrated with the mutation strategy of JADE, then a modified version, named JADEfcr, is proposed. In addition, to improve the optimization ability further, another variant LJADEfcr by using a linear population reduction mechanism is considered. So as to confirm and examine the performance of JADEfcr and LJADEfcr, numerical experiments are conducted on 29 optimization problems defined by CEC2017 benchmark. For JADEfcr, its experimental results are made a comparison with twelve state-of-the-art algorithms. The comparative study demonstrates that in terms of robustness, stability and solution quality, JADEfcr are better and highly competitive with these well-known algorithms. For LJADEfcr, its results are compared with JADEfcr and other nine powerful algorithms including four recent algorithms and five top algorithms on CEC2017 competition. Experimental results indicate that LJADEfcr is superior and statistically competitive with these excellent algorithms in terms of robustness, stability and the quality of the obtained solutions.

48. L-carnitine promotes liver regeneration after hepatectomy by enhancing lipid metabolism

Abstract Background Lipid metabolism plays an important role in liver regeneration, but its regulation still requires further research. In this study, lipid metabolites involved in mouse liver regeneration at different time points were sequenced and analyzed to study their influence on liver regeneration and its mechanism. Methods Our experiment was divided into two parts. The first part examined lipid metabolites during liver regeneration in mice. In this part, lipid metabolites were sequentially analyzed in the livers of 70% mouse hepatectomy models at 0, 1, 3and 7 days after operation to find the changes of lipid metabolites in the process of liver regeneration. We screened L-carnitine as our research object through metabolite detection. Therefore, in the second part, we analyzed the effects of carnitine on mouse liver regeneration and lipid metabolism during liver regeneration. We divided the mouse into four groups: control group (70% hepatectomy group); L-carnitine group (before operation) (L-carnitine were given before operation); L-carnitine group (after operation)(L-carnitine were given after operation) and L-carnitine + perhexiline maleate (before operation) group. Weighing was performed at 24 h, 36 and 48 h in each group, and oil red staining, HE staining and MPO staining were performed. Tunnel fluorescence staining, Ki67 staining and serological examination. Results Sequencing analysis of lipid metabolites in 70% of mouse livers at different time points after hepatectomy showed significant changes in carnitine metabolites. The results showed that, compared with the control group the mouse in L-carnitine group (before operation) at 3 time points, the number of fat drops in oil red staining was decreased, the number of Ki67 positive cells was increased, the number of MPO positive cells was decreased, the number of Tunnel fluorescence positive cells was decreased, and the liver weight was increased. Serum enzymes were decreased. Compared with control group, L-carnitine group (after operation) showed similar trends in all indexes at 36 and 48 h as L-carnitine group (before operation). L-carnitine + perhexiline maleate (before operation) group compared with control group, the number of fat drops increased, the number of Ki67 positive cells decreased, and the number of MPO positive cells increased at 3 time points. The number of Tunnel fluorescent positive cells increased and serum enzyme increased. However, both liver weights increased. Conclusion L-carnitine can promote liver cell regeneration by promoting lipid metabolism and reduce aseptic inflammation caused by excessive lipid accumulation.

49. Numerical optimization on differential adjusting measures for asymmetrical degenerated warm permafrost railway embankment

Affected by climate warming and shady–sunny slope effect, the permafrost foundation under the railway is undergoing a significant asymmetric degradation process; research on the corresponding regulating or strengthening measures is urgently needed. Based on a well developed of numerical model, we analyzed the ‘thermal repair’ effect of three redesigned differential thermal reinforcement on the partially degraded permafrost embankment with steps. The results show that, when used as a strengthening measure, the method of laying crushed rock revetment (CRR) with different thicknesses on the sunny and shady slopes cannot completely eliminate the residual warm permafrost regions formed in the previous thermal erosion. When the TCRR scheme is adopted for reinforcement, although horizontal region from the evaporation section of thermosyphones (TPCTs) to the right side of the embankment is remarkably cooled, permafrost under the sunny slope is still deteriorating. In the long run, both ETCRR and TCECRR reinforcement schemes can produce a large range of ‘cold core’ right below the whole embankment. However, from the perspective of the simultaneous functioning range and cold storage capacity, the TCECRR scheme is signally more efficient. The thermal insulation materials in the above composite strengthening measures can not only resist the heat input in the warm season, but also improve their working efficiency by maintaining the temperature difference between the evaporation section of the heat pipe and the external environment in cold season. The addition of crushed rock revetment in TCECRR has played its thermal semiconductor effect and further improved the comprehensive thermal repair efficiency of the composite measures.

50. Thermally stable threshold selector based on CuAg alloy for energy-efficient memory and neuromorphic computing applications

Abstract As a promising candidate for high-density data storage and neuromorphic computing, cross-point memory arrays provide a platform to overcome the von Neumann bottleneck and accelerate neural network computation. In order to suppress the sneak-path current problem that limits their scalability and read accuracy, a two-terminal selector can be integrated at each cross-point to form the one-selector-one-memristor (1S1R) stack. In this work, we demonstrate a CuAg alloy-based, thermally stable and electroforming-free selector device with tunable threshold voltage and over 7 orders of magnitude ON/OFF ratio. A vertically stacked 64 × 64 1S1R cross-point array is further implemented by integrating the selector with SiO2-based memristors. The 1S1R devices exhibit extremely low leakage currents and proper switching characteristics, which are suitable for both storage class memory and synaptic weight storage. Finally, a selector-based leaky integrate-and-fire neuron is designed and experimentally implemented, which expands the application prospect of CuAg alloy selectors from synapses to neurons.
