Immune response is an important factor in determining the fate of bone replacement materials, in which macrophages play an important role. It is a new idea to design biomaterials with immunomodulatory function to reduce inflammation and promote bone integration by regulating macrophages polarization. In this work, the immunomodulatory properties of CaP Zn-Mn-Li alloys and the specific mechanism of action were investigated. We found that the CaP Zn0.8Mn0.1Li alloy promoted the polarization of macrophages towards M2 and reduced inflammation, which could effectively up-regulate osteogenesis-related factors and promote new bone formation, indicating the important role of macrophages polarization in biomaterial induction of osteogenesis. In vivo studies further demonstrated that CaP Zn0.8Mn0.1Li alloy could stimulate osteogenesis better than other Zn-Mn-Li alloys implanttions by regulating macrophages polarization and reducing inflammation. In addition, transcriptome results showed that CaP Zn0.8Mn0.1Li played an important regulatory role in the life process of macrophages, activating Toll-like receptor signaling pathway, which participated in the activation and attenuation of inflammation, and accelerated bone integration. Thus, by preparing CaP coatings on the surface of Zn-Mn-Li alloys and combining the bioactive ingredient with controlled release, the biomaterial will be imbibed with beneficial immunomodulatory properties that promote bone integration.

Purpose: Nonampullary duodenal adenocarcinoma (NADA) is a rare disease. Although several prognostic factors have been reported for this disease, they remain controversial due to their rarity. In this study, we retrospectively analyzed 54 cases of invasive NADA, focusing on the microsatellite instability (MSI) phenotype, programmed cell death-ligand 1 (PD-L1) expression, and prognostic factors. Methods: Expression of the PD-L1 protein and cell differentiation markers in tumors was detected by immunohistochemistry. Microsatellite markers (NR-21, NR-22, NR-24, BAT-25, and BAT-26) were amplified for MSI assessment by PCR. Results: The incidence of MSI in invasive NADA was 35.2%. No significant correlation between the MSI phenotype and clinicopathological factors was observed. Positive expression of PD-L1 by immune cells was common in advanced-stage disease (p = 0.054), and positive expression of PD-L1 in cancer cells correlated significantly with the histologically undifferentiated type (p = 0.016). Kaplan-Meier survival analysis demonstrated a significantly better overall survival (OS) in patients with MSI (p = 0.013) and at early-stage disease (p = 0.000) than in those with microsatellite-stable or at late tumor stages. Univariate and multivariate analyses showed that MSI (hazard ratio [HR]: 0.282, 95% confidence interval [CI]: 0.106–0.751, p = 0.011) and early tumor stage (stage I-II) (HR: 8.81, 95% CI: 2.545–30.500, p = 0.001) were independent better prognostic factors of OS. Conclusions: MSI and early tumor stage (stage I-II) were independent better prognostic factors of OS. A high proportion of MSI phenotypes and positive PD-L1 expression may be helpful for identifying immune checkpoint inhibitors as a novel therapeutic strategy.

A potential K-ion solid-state electrolyte K[B3H7NH2BH2NH2B3H7] was synthesized and its relatively high K+ conductivity of 1.01 × 10−4 S cm−1 at 75 °C was obtained probably due to the increased electrostatic potential and size of the anions.

Improved methods for the synthesis of KB3H8, NH3B3H7, and N-alkyl analogues of NH3B3H7 have been developed based on previous works. KB3H8 was synthesized by the reaction of metallic potassium (K) with borane dimethyl sulfide ((CH3)2S·BH3) with high yield and atom-economy. In the preparation of NH3B3H7 and its N-alkyl analogues, KB3H8 served as a starting material and was converted to THF·B3H7 first through reactions with HCl diethyl ether solution or oxidation agent CoCl2. Then, the formed THF·B3H7 in situ reacted with the corresponding ammonia or amines to form the amine borane final products. This work paves an alternative way for preparing organic-inorganic hybrid materials containing B, N, and C atoms.

Porcine circovirus-like virus (Po-Circo-like virus, PCLV), as a recently discovered virus, may be associated with diarrhea in pigs. To investigate the epidemic profile and genetic characteristics of the virus, one hundred and seventy-five clinical samples (141 intestinal samples, 17 blood samples and 17 fecal samples) were collected from diseased piglets during outbreaks of diarrhea on 33 pig farms in 19 cities of Henan and Shanxi Provinces of China between 2016 and 2021, and were screened by PCR for the presence of PCLV. The results showed that 32% (56/175) of diarrheal piglets were positive for PCLV, and 60.6% (20/33) of the pig farms and 57.9% (11/19) of the cities were positive for PCLV, which varied from 5.88–44.12% between 2016 and 2021. It was also found that PCLV might be co-infected with PRRSV, PCV2, PCV3, PCV4, and PEDV, but no nucleic acids were detected for TGEV, PDCoV and PRoV in piglets with diarrhea. Notably, only PCLV was detected in 13 diarrheal piglets from four of these farms, whereas other pathogens were negative. The findings suggest that PCLV has been circulating in piglets in Henan and Shanxi Provinces of China, and may be a diarrheal virus. The complete genomes of thirteen PCLV strains were sequenced, and these strains shared 35.4–91.0% genome nucleotide identity with those available in GenBank. Phylogenetic analysis demonstrated that the 13 PCLV strains in this study were clustered in Group 1, which closely related to 8 Chinese PCLV strains, Bo-Circo-like virus CH, two American strains 21 and 22, and two Hungary strains 288_4 and 302_4, but differed genetically from 7 other foreign PCLV strains. In addition, the whole genome and Rep gene of 13 PCLV strains in this study were 72.2% − 82% and 83.8% − 89.7% identity with those of Bo-Circo-like virus CH strain respectively, which provided further evidence for the hypothesis that PCLV is a novel virus in pigs that may be involved in cross-species transmission. Furthermore, a cross-recombination event was found in the genome and Rep region of the sequenced 13 PCLV strains. This research enriched the epidemiological data of PCLV infection in pigs in China and laid a foundation for further study on the pathogenesis of PCLV.

Objective To analyze and predict hematopoietic injury caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection and potential therapeutic drugs, and to provide theoretical basis for clinical treatment of the hematopoietic injury. Methods The gene expression omnibus (GEO) database was used to screen the whole genome expression data related to SARS-CoV-2 infection. The R language package was used for differential expression analysis and the Kyoto Encyclopedia of Genes and Genomes (KEGG) and Gene Ontology (GO) enrichment analysis. The core genes were screened by protein-protein interaction (PPI) network analysis using STRING online analysis website. Then the self-developed apparent precision therapy prediction platform (EpiMed) was used to analyze diseases, drugs and related target genes. Results A total of 222 differential genes were screened, including 172 up-regulated and 50 down-regulated. GO enrichment analysis suggested that gene is mainly related to type I interferon response, cell cycle regulation, inflammatory cell migration, innate immune response, secretion of blood particles and vesicles, chemokines and their receptors. KEGG enrichment analysis suggested that gene is mainly related to viral infection, myocardial injury, complement and coagulation cascade, cell chemotaxis, platelet activation, acute inflammation, immune response, cellular signal transduction and so on. Ten core genes such as STAT1, IL-6, IRF7, TNF, MX1, ISG15, IFIH1, IRF9, DDX58 and GBP1were screened by PPI network analysis. EpiMed screened 10 drugs with potential intervention effects, including Rabdosia rubescens, sirolimus, glucocorticoid, Houttuynia cordata, Polygonum multiflorum, Red peony, tretinoin, Glycyrrhiza, cyclosporine A, fluvastatin and so on. Conclusions SARS-CoV-2 infection can damage the hematopoietic system by changing the expression of a series of genes. The potential intervention drugs screened from this have certain reference significance for the basic and clinical research of coronavirus disease 2019 (COVID-19). Herein, a highly regioselective propargylic benzylation with propargylic carbonates and benzyl 1,4-dihydropyridine derivatives was developed via a photoredox/palladium dual-catalyzed process, which represents a novel catalytic model for non-terminal propargylic functionalization. The reaction showed excellent regioselectivity and functional group compatibility. A radical coupling mechanism between the propargylic radical and benzyl radical was proposed.

Improved methods for the synthesis of KB

Porcine circovirus 4 (PCV4), a novel porcine circovirus identified in pigs, has recently been proved to be pathogenic to piglets. However, little is known about its cross-species transmission, and demonstration of PCV4 in dairy cows is lacking. To explore whether the PCV4 genome exists in dairy cows, 1170 fecal samples were collected from dairy farms in 7 cities in Henan Province of China during 2012-2021, and screened by qPCR for the presence of PCVs (PCV2-PCV4). The detection results showed that the positive rate of PCV4 in dairy cows was 2.22 % (26/1170), but all fecal samples were negative for PCV2 and PCV3. Three full-length and five partial genomes of PCV4 strains were acquired, of which two PCV4 strains (NY2012-DC and XC2013-DC) were achieved from 2012 and 2013, indicating that PCV4 has been circulating in dairy cows in Henan Province of China for at least 10 years. The three PCV4 strains sequenced in this study shared high identity (97.5-99.5 %) with reference strains at the genome level. In phylogenetic analysis, three genotypes (PCV4a, PCV4b and PCV4c) were temporarily confirmed by analyzing 44 strains, and one amino acid variation in Rep (V239L) and three amino acid variations in Cap (N27S, R28G and M212L) were considered as a conserved genotype specific molecular marker. Analyzed from three perspectives (cross-time, cross-species and transboundary), the high nucleotide homology of PCV4 strains indicated the PCV4 evolutionary rate might be slow. Overall, this study was the first to report the detection of PCV4 in dairy cows and conducted a long-term retrospective investigation of PCV4 in Henan Province of China, which has important implications for understanding the genetic diversity and cross-species transmission of the ongoing PCV4 cases.

Porcine circovirus 4 (PCV4) was identified as a novel porcine circovirus in China in 2019. To investigate the prevalence and genetic characteristics of PCV2 and PCV4, 133 clinical samples (103 tissue samples and 30 serum samples) were collected from 30 different pig farms in Henan province of China, and a SYBR Green I-based duplex quantitative real-time polymerase chain reaction assay was established to detect PCV2 and PCV4 genomes simultaneously. The complete genome sequences of 20 PCV2 and 6 PCV4 strains from 19 and 6 clinical samples respectively were sequenced and analyzed. The results showed the detection limits of this assay were 80.2 copies/μL for PCV2 and 58.6 copies/μL for PCV4. The detection results of clinical samples revealed the PCV2 positive rate was 63.16% (84/133), the PCV4 positive rate was 33.33% (45/133), and the PCV2 and PCV4 co-infection positive rate was 21.05% (28/133). Among 20 PCV2 strains, 6 belonged to PCV2a, 6 belonged to PCV2b and 8 belonged to PCV2d. Co-infection with JZ1 (PCV2b) and JZ2 (PCV2d) strains was identified in one sample (JZ-1). Eleven putative recombination events were found through the recombination analysis, suggesting that the new PCV2 variant strains had circulated in Henan province, which contributes to our understanding of evolutionary characteristics of PCV2 in China. The possible genotypes of PCV4 strains were determined based on genomic sequences of 6 PCV4 strains in this study and 29 PCV4 reference strains available at GenBank. According to three different phylogenetic trees (ORF1, ORF2 and complete genome), all 35 PCV4 strains were clustered into two major genotypes (PCV4a and PCV4b), and 6 PCV4 strains in this study belonged to PCV4a. Additionally, the functional regions of PCV4 strains were predicted by comparison with other circoviruses, which are conducive to the further study of the biological functions of PCV4 genome.

We have synthesized the tetrabutylammonium ([Bu 4 N] + ), tetraethylammonium ([Et 4 N] + ), guanidinium ([C(NH 2 ) 3 ] + ), and methylguanidinium ([C(N 3 H 5 CH 3 )] + ) salts of the [BH 3 (NH 2 BH 2 ) 2 H] - anion, a five-membered B/N anionic chain, in high yield by the metathesis reactions of Na[BH 3 (NH 2 BH 2 ) 2 H] with the corresponding halides and characterized them by NMR ( 11 B, 11 B{ 1 H }, 1 H, 1 H{ 11 B}, 13 C), IR, elemental analysis, TGA-DSC, and TGA-MS. These complexes behave like ionic liquids (ILs), in which the melting point of the [Bu 4 N][BH 3 (NH 2 BH 2 ) 2 H] is the lowest (-51 o C). The dehydrogenation of these ILs have been studied through the thermal decomposition and catalytic hydrolysis in aqueous solution using the noble or non-noble metals or their salts as catalysts, and the results indicate that these ILs of five-membered B/N anionic chain are promising hydrogen storage materials.

In this issue of Chem, Kondo and co-workers report a facile soft-chemical method for the preparation of layered, electrically conductive hydrogen boride, which is short-range order but amorphous as a result of the geometrical frustration of the terminal B–H bonds. This work offers an alternative route to two-dimensional (2D) boron materials.

A facile synthesis of heavy alkali metal octahydrotriborates (MB3 H8 ; M=K, Rb, and Cs) has been developed. It is simply based on reactions of the pure alkali metals with THF⋅BH3 , does not require the use of electron carriers or the addition of other reaction media such as mercury, silica gel, or inert salts as for previous procedures, and delivers the desired products at room temperature in very high yields. However, no reactions were observed when pure Li or Na was used. The reaction mechanisms for the heavy alkali metals were investigated both experimentally and computationally. The low sublimation energies of K, Rb, and Cs were found to be key for initiation of the reactions. The syntheses can be carried out at room temperature because all of the elementary reaction steps have low energy barriers, whereas reactions of LiBH4 /NaBH4 with THF⋅BH3 have to be carried out under reflux. The high stability and solubility of KB3 H8 were examined, and a crystal structure thereof was obtained for the first time.

Different mechanisms for [NH3BH2NH3]+[BH4]− → NH3BH3 + [NH2BH2]n and [NH4]+[BH3NH2BH3]− → NH3BH2NH2BH3 have been elucidated, which leads to the proposal of a general mechanism as below.

Herein, a novel photo-induced amine-free radical reductive dehalogenation of inactivated aryl/alkyl bromides and chlorides with a palladium complex is described, which reveals excellent functional group compatibility and broad substrate scope. Extensional transformations for reductive cyclization, dehalogenative deuteration, and intra- and intermolecular radical addition can be achieved smoothly. Mechanistic studies indicate a single-electron photoredox catalytic system with inactivated solvent as the hydrogen atom donor.

An efficient synthetic method for MB3H8 (M = Li and Na) has been developed with improved yields on the basis of the investigation of the interconversion between THFB3H7 and corresponding MB3H8. The mechanism was tentatively proposed based on the understanding of the nucleophilicity of the B-H bonding pair electrons.

Octahydridoborate, i.e. [B3H8](-) containing compounds, have recently attracted interest for hydrogen storage. In the present study, the structural, hydrogen storage, and ion conductivity properties of KB3H8 have been systematically investigated. Two distinct polymorphic transitions are identified for KB3H8 from a monoclinic (alpha) to an orthorhombic (alpha ') structure at 15 degrees C via a second-order transition and eventually to a cubic (beta) structure at 30 degrees C by a first-order transition. The beta-polymorph of KB3H8 displays a high degree of disorder of the [B3H8](-) anion, which facilitates increased cation mobility, reaching a K+ conductivity of similar to 10(-7) S cm(-1) above 100 degrees C. beta-KB3H8 starts to release hydrogen at similar to 160 degrees C, simultaneously with the release of B5H9 and trace amounts of B2H6. KBH4 and K-3(BH4)(B12H12) are identified as crystalline decomposition products above 200 degrees C, and the formation of a KBH4 deficient structure of K3-x(BH4)(1-x)(B12H12) is observed at elevated temperature. The hydrogen-uptake properties of a KB3H8-2KH composite have been examined under 380 bar H-2, resulting in the formation of KBH4 at T >= 150 degrees C along with higher metal hydridoborates, i.e. K2B9H9, K2B10H10, and K2B12H12.

In this paper, CuO hollow microspheres with different shell thickness and porosity have been synthesized using carbonaceous saccharide microspheres as templates according to a modified literature method. These CuO hollow microspheres were characterized and their catalytic properties in the hydrolytic dehydrogenation of ammonia borane (NH3BH3, AB) were examined. A kinetic study indicated that a maximum hydrogen generation rate of 294 mL H2 min−1 (g catalyst)−1 can be achieved at 45 ± 0.2 °C in the present system, which is comparable with that for other reported Cu based catalysts.

A general approach for the synthesis of covalent and ionic amine borane complexes containing trinitromethyl fragments has been developed through metathesis reactions between amine chloroborane complexes and potassium salt of trinitromethyl (K[C(NO2)3]). Five covalent and ionic trinitromethyl amine borane complexes have been synthesized in good yields with high purity and it is found that the ionic complex, [H2B(NH3)2][C(NO2)3], might be a promising energetic material on the basis of the investigation of its thermal decomposition behaviour.

Palladium catalyzed visible-light-mediated borylation of inactivated aryl and alkyl halides is reported; the method provided high yields and excellent functional group compatibility. Furthermore, arylsilicates were synthesized selectively using dimethylphenylsilyl boronic ester via changing the reaction conditions. Finally, the possible reaction mechanism is determined through fluorescence quenching and turn on/off experiments.

Background Burkitt lymphoma is a kind of non-Hodgkin B-cell-derived malignancy, derived from germinal center B cells. FAM54A has been proved to be involved in various physiological and pathological processes of cancers, but the biological function of FAM54A in Burkitt lymphoma remains unclear. Thus, the aim of our research was to elucidate the roles of FAM54A in proliferation, apoptosis and cell cycle of Burkitt lymphoma.Methods Burkitt lymphoma cell line (Namalwa) was chosen to perform the following experiments. FAM54A-shRNA and negative control-shRNA lentivirus that were synthesized, respectively by Qiagen were used to transfect targeted cells in order to knockdown FAM54A or as negative control. Then, cell proliferation, cell cycle and cell apoptosis were detected by using MTS assay, propidium iodide staining and Annexin V-APC staining, respectively.Results Our results showed that high expression of FAM54A protein was found in Namalwa cell line. Furthermore, MTS analysis exhibited that knockdown of FAM54A obviously inhibited cell proliferation in Namalwa cells. What’s more, cell cycle analysis showed that knockdown of FAM54A induced Namalwa cell apoptosis and arrested cell cycle in G2/M phase.Conclusions These findings suggest that FAM54A is essential for Namalwa cell proliferation and may be a potential therapeutic target for the treatment of Burkitt lymphoma.

Boron compounds are well-known electrophiles. Much less known are their nucleophilic properties. By recognition of the nucleophilicity of the B–H bond, the formation mechanism of octahydrotriborate (B3H8–) was elucidated on the bases of both experimental and computational investigations. Two possible routes from the reaction of BH4– and THF·BH3 to B3H8– were proposed, both involving the B2H6 and BH4– intermediates. The two pathways consist of a set of complicated intermediates, which can convert to each other reversibly at room temperature and can be represented by a reaction circle. Only under reflux can the B2H6 and BH4– intermediates be converted to B2H5– and BH3(H2) via a high energy barrier, from which H2 elimination occurs to yield the B3H8– final product. The formation of B2H6 from THF·BH3 by nucleophilic substitution of the B–H bond was captured and identified, and the reaction of B2H6 with BH4– to produce B3H8– was confirmed experimentally. Long-sought mechanisms for the conversion of diammoniate of diborane ([NH3BH2NH3]+[BH4]-) into NH3BH3 and [NH2BH2]n as well as ammonium aminodiborane ([NH4]+[BH3NH2BH3]-) into a butane analogue, NH3BH2NH2BH3, have been elucidated on the basis of extensive experimental and theoretical studies. The [NH4]+ ammonium cation and the (η2-H2)BH2R moiety are found to be critical in B/N chain expansion.

To screen genes associated with poor prognosis of hepatocellular carcinoma (HCC) and to explore the clinical significance of these genes.The proper expression profile data of HCC was obtained from the Gene Expression Omnibus (GEO) database, and the differentially expressed genes (DEGs) were identified by differential expression analysis. The DAVID and String database were used for function enrichment analysis and to construct the protein-protein interaction (PPI) network respectively. The Cancer Genome Atlas (TCGA) database and the Cox Proportional Hazard Model were used for prognosis analysis of the DEGs.A eligible human HCC data set (GSE84402) met the requirements. A total of 1141 differentially expressed genes were identified, including 720 up-regulated and 421 down-regulated genes. The results of function enrichment analysis and PPI network performed that CDK1、CDC6、CCNA2、CHEK1、CENPE 、PIK3R1、RACGAP1、BIRC5、KIF11 and CYP2B6 were prognosis key genes. And the prognosis analysis showed that the expressions of CDC6、PIK3R1、KIF11 and RACGAP1 were increased, and the expression of CENPE was decreased, which was closely related to prognosis of HCC.CDC6、CENPE、PIK3R1、KIF11 and RACGAP1 may be closely related to poor prognosis of HCC, and can be used as molecular biomarkers for future research of HCC prognosis.

To investigate the prognosis-related miRNA histological features and clinical significance of lung adenocarcinoma.Using The Cancer Genome Atlas (TCGA) data, the miRNA expression profile data of human lung adenocarcinoma were searched for differential analysis, and the prognosis-related miRNAs were screened by Cox risk regression model. The targeted miRNAs were predicted by mirwalk analysis platform, KEGG functional enrichment analysis, and finally, predict the function of prognosis-related miRNAs.A total of 46 differential miRNAs in lung adenocarcinoma were screened, including 19 up-regulated and 27 down-regulated. Six prognostic-related miRNAs were screened by Cox survival analysis, namely hsa-mir-21, hsa-mir-142, hsa-mir-200a high expression, hsa-mir-101, hsa-let-7c, hsa-mir-378e low expression, hsa-mir-21 and hsa-mir-378e were associated with poor prognosis in patients with lung adenocarcinoma, and the survival time was shortened significantly (Hsa-mir-21 and hsa-mir-378e are associated with poor prognosis of lung adenocarcinoma, and may be used as a molecular marker for prognosis of lung adenocarcinoma after further clinical verification.

To analyze the molecular markers associated with occurrence, development and poor prognosis of acute myeloid leukemia (AML) by using the data of GEO and TCGA database, as well as multiomics analysis.The transcriptome data meeting requirements were down-loaded from GEO database, the differentially expressed genes were screened by using the R language limma package, and the GO function enrichment analysis and KEGG pathway analysis were performed for differentially expressed genes, at the same time, the protein interaction network was contracted by using STRING database and cytoscape software to screen out the hub gene, then the prognosis analysis was carried out for hub gene by combination with the clinical information affected in TCGA database.620 differentially expressed genes were screened out, among which 162 differentially expressed genes were up-regulated, and 458 differentially expressed genes were down-regulated. Based on the results of GO functional enrichment, the KEGG pathway enrichment and protein interaction network, CXCL4, CXCR4, CXCR1, CXCR2, CCL5 and JUN were selected as hub genes. The survival analysis showed that the high expression of CXCL4, CXCR1, and CCL5 was a risk factor for poor prognosis of patiants.CXCL4, CXCR1 and CCL5 can be used as biomarkers for the occurrence and development of AML, which relateds with the unfavorable prognosis and can provide a basis for further study.急性髓系白血病患者预后不良相关基因的多组学分析.利用GEO数据库和 TCGA数据库的数据，通过多组学分析方法分析与急性髓系白血病（acute myeloid leukemia，AML）发生、发展及不良预后相关的分子标志物。.从GEO数据库下载符合要求的转录组数据，运用R语言Limma程序包进行差异表达基因的筛选，并对差异表达基因进行GO功能富集分析和 KEGG 通路富集分析，同时使用STRING数据库数据，利用Cytoscape软件构建蛋白质相互作用网络，筛选出hub gene，结合TCGA 数据库附带的临床信息对hub gene进行预后分析。.共筛选出620个差异基因，上调的差异表达基因162个，下调的差异表达基因458个。综合GO功能富集、KEGG 通路富集分析及蛋白相互作用网络结果，筛选出CXCL4、CXCR4、CXCR1、CXCR2、CCL5、JUN为hub gene。生存分析显示，CXCL4、CXCR1、CCL5高表达是患者预后不良的危险因素。.CXCL4、CXCR1、CCL5可以作为AML发生、发展的相关生物标志物，且与不良预后相关，这可以为进一步研究提供依据。.
