Catalyzing polysulfides conversion for lithium‐sulfur batteries is an efficient strategy to overcome the sluggish kinetics of polysulfides conversion as well as its serious shuttling effect. Due to the multistep and complicated phase transformation of sulfur species, the monofunctional catalyst can hardly promote the overall polysulfides redox process. Herein, a molybdenum‐based heterostructure is proposed, that facilitates the entire reduction process by tandemly catalyzing liquid‐liquid conversion and liquid‐solid conversion. It is uncovered that the MoC physiochemically immobilizes the soluble long‐chain polysulfide and accelerates the conversion between S8 to Li2S4 through adsorbing Li2S8 and extending its S─S bond distance. Then, the kinetics of Li2S precipitation is enhanced by facilitating the migration of Li2S4 from MoC to MoSe2. This is driven by the internal electric field at the heterogeneous interface and the low diffusion energy barrier on MoSe2 for Li2S4. Moreover, MoC‐MoSe2 exhibits the smallest degree of Li2S2 disproportionation throughout the reduction process. Consequently, the cell with MoC‐MoSe2/C/S cathode delivers an initial discharge‐specific capacity of 841.1 mAh g−1 and long‐term cycling stability with a capacity attenuation rate of 0.08% per cycle at 1.0 C. Porcine reproductive and respiratory syndrome (PRRS) is one of the most important swine diseases affecting pig industry worldwide. Sialoadehesin (Sn) and CD163 are the two specific receptors for PRRSV infection of porcine alveolar macrophages. Our previous study showed that the soluble Sn receptor Sn4D-Fc and soluble CD163 receptor SRCR59-Fc expressed by the two recombinant adenoviral (rAd) vectors have an additive anti-PRRSV effect in vitro . In the present study, rAd-Sn4D-Fc and rAd-SRCR59-Fc were inoculated into pigs, and the efficient expression of Sn4D-Fc and SRCR59-Fc proteins was detected by ELISA. Then, PRRSV-naïve pigs were inoculated with rAd-Sn4D-Fc and/or rAd-SRCR59-Fc before contagious infection with different PRRSV strains. Among the three rAd inoculation groups, simultaneous inoculation with the two rAd vectors provided the best protection against highly pathogenic JXA1 strain PRRSV, followed by rAd-SRCR59-Fc inoculation and rAd-Sn4D-Fc inoculation. Clinical observation and quantitative RT-PCR analyses showed that all of the double rAd-inoculated pigs (n = 9) survived from the contagious infection with highly pathogenic JXA1, JS07 or SH1705 strain PRRSV with significantly alleviated clinical scores, viremia, fecal viral emission and tissue virus loads. Chicken egg yolk immunoglobulin (IgY) is a superior alternative to mammalian immunoglobulin, but its practical application is limited due to the complex purification procedure. In this study, the C2 domain of streptococcal protein G (SPG) with the binding affinity for chicken IgY was expressed in E. coli as an elastin-like polypeptide (ELP) fusion protein, and purified to a high purity by inverse transition cycling (ITC). Binding experiments showed that chicken IgY could bind to and eluted off the ELP-C2 fusion protein in pH-, temperature- and/or time-dependent manners. By using the ELP-C2 protein, a simple chicken IgY purification method was developed, and its purification performance was compared with that of ammonium sulfate precipitation and ethanol fractionation. Quantitative SDS-PAGE analysis showed that the ELP-C2 binding capture method provided a chicken IgY purity of 96.3% and a recovery of 64%, both of which were significantly higher than that of the two traditional methods The ELP-C2 binding capture method could be accomplished within 3 h, in contrast to 30.3 h for ammonium sulfate precipitation or 4.3 h for ethanol fractionation. • VC 0.75 /Co-CoO was applied as a host in Li-S battery for the first time. • The catalytic activity towards polysulfides is regulated. • The bifunctional catalytic effect of VC 0.75 /Co-CoO is analyzed. The shuttle effect and sluggish redox kinetics of lithium polysulfides lead to a short lifespan and poor rate capability of lithium-sulfur (Li-S) batteries. Introducing electrocatalysts into sulfur host materials has been deemed a promising strategy to overcome these issues in Li-S batteries. Herein, an efficient catalytic sulfur host material (i.e., VC 0.75 /Co-CoO) has been fabricated by simply pyrolyzing the electrospun vanadium and cobalt salt-based polymer fibers. Electrochemical measurements demonstrated that the as-formed heterostructure of Co-CoO with VC 0.75 evidently strengthens the redox kinetics of polysulfides due to the enhanced adsorption ability to polysulfides and adjusts the electronic structure of heterostructure via the introduction of VC 0.75. Further theoretical computation discloses that Co-CoO mainly plays the role in stretching the S S bond in polysulfides to facilitate the reduction of polysulfide, extending the Li-S bond in Li 2 S and reducing energy barriers of Li 2 S decomposition to promote the oxidation process of Li 2 S. Accordingly, the collaboration of Co-CoO and VC 0.75 synergetically facilitates the redox kinetics of polysulfides, prolonging the lifespan of Li-S batteries. Impressively, VC 0.75 /Co-CoO composites endow Li-S batteries with a discharge capacity decay rate as low as 0.02 % per cycle at the current density of 0.2 C. The specific capacity reaches 626 mAh/g at the high current density of 1.0 C even after 450 cycles. A downlink non‐orthogonal multiple access with QPSK input constellations based on virtual channel optimisation is presented to address the similar channel conditions scenario. The novel idea is to construct a uniquely decodable sum constellation by maximising the minimum Euclidean distance among the points of the superposed constellation. An optimal angle difference is derived to make the proposed scheme easy to implement. With a fixed angle pair setup, a non‐orthogonal multiple access system without the knowledge of channel state information is proposed. Mycoplasma synoviae (MS) is an important avian pathogen that has brought substantial economic losses to the global poultry industry. Fast and accurate diagnosis is one of the critical factors for the control of MS infection. This study established a simple, rapid and visual detection method for MS using a recombinase-aided amplification (RAA) combined with a lateral flow dipstick (LFD). The reaction temperature and time of the RAA-LFD assay were optimized after selecting the primers and probe, and the specificity and sensitivity rates were analyzed. The results showed that RAA could amplify the target gene in 20 min at a constant temperature of 38°C, and the amplification products could be visualized by LFD within 5 min. There was no cross-reaction with Mycoplasma gallisepticum (MG), Pasteurella multocida ( P. multocida ), Escherichia coli ( E. coli ), Newcastle disease virus (NDV), infectious bursal disease virus (IBDV), infectious bronchitis virus (IBV), and avian reovirus (ARV). Furthermore, the RAA-LFD assay exhibited high sensitivity with a detection limit of 10 copies/μL. A total of 128 clinical samples with suspected infection of MS were tested by RAA-LFD, PCR, and real-time fluorescence quantitative PCR (RFQ-PCR). The coincidence rate of the detection results was 95.3% between RAA-LFD and PCR, and 98.4% between RAA-LFD and RFQ-PCR. These results suggested that the RAA-LFD method established in the present study was easy to use and was associated with strong specificity and high sensitivity. Poly(l -lactic acid) (PLLA) has been attracted increasing interesting in biomedicine and tissue engineering. However, its hydrophobicity can bring about some adverse effects on protein adsorption and downstream cell adhesion. Poly(N-isopropylacrylamide) (PNIPAm) is a well-known water-soluble polymer which exhibits a hydrophobic-hydrophilic phase transition in aqueous solution upon heating. In this work, a series of P(NIPAm-co-VAc) polymers have been synthesized by using NIPA and vinyl acetate (VAc) as raw materials to insert into PLLA. The results show that the lower critical solution temperature (LCST) of synthesized polymer is 33–34 °C. When the molar ratio of NIPA to VAc and the blend ratio of synthesized polymer to PLLA are 0.76/1 and 20 wt%, respectively, the hydrophilicity of composite enhances significantly at room temperature. In particular, the composite exhibits thermoresponsive wettability below and above the LCST. Although the water contact angle presents a systematic increase with temperature increasing, it doesn't pass through the real hydrophobic domain because the conformational transition of wettable PNIPAm segment is subjected to intramolecular PVAc segment and extramolecular PLLA chain. Moreover, the synthesized polymer also offsets the brittleness of PLLA. So the method provides thermoresponsive wettability for PLLA and builds a platform for developing intelligent PLLA. Colon cancer is the most common malignant tumor of the gastrointestinal tract, and approximately 80%–90% of colon cancers are colon adenocarcinomas (COADs). This study aimed to screen key microRNAs (miRNAs) associated with COAD. Differentially expressed (DE) miRNAs were screened between COAD and adjacent cancer samples based on the Gene Expression Omnibus (GEO) and the Cancer Genome Atlas obtained from datasets. The miRNAs of interest were validated using quantitative real-time polymerase chain reaction. Moreover, the effects of hsa-miR-135b-5p on the biological behavior of COAD cells were observed. To obtain the target genes of hsa-miR-135b-5p, transcriptome sequencing of the SW480 cells was performed, followed by protein-protein interaction (PPI) network and hsa-miR-135b-5p-target gene regulatory network construction and prognostic analysis. Downregulation of hsa-miR-135b-5p significantly inhibited SW480 cell proliferation, migration, and invasion and significantly facilitated apoptosis (P  < 0.05). A total of 3384 DEmRNAs were screened, and enrichment analysis showed that the upregulated mRNAs were enriched in 25 Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways and 326 Gene Ontology Biological Processes (GO-BPs) while the downregulated mRNAs were enriched in 20 KEGG pathways and 276 GO-BPs. A PPI network was then constructed, and H2BC14, H2BC3, and H4C11 had a higher degree. In addition, a total of 352 hsa-miR-135b-5p-gene regulatory relationships were identified. Prognostic analysis showed that FOXN2 , NSA2 , MYCBP , DIRAS2 , DESI1 , and RAB33B had prognostic significance (P  < 0.05). African swine fever (ASF) is one of the most severe diseases caused by the ASF virus (ASFV), causing massive economic losses to the global pig industry. Serological tests are important in ASF epidemiological surveillance, and more antigen targets are needed to meet market demand for ASFV antibody detection. In the present study, ASFV p15 protein was fusion-expressed in Escherichia coli (E. coli) with elastin-like polypeptide (ELP), and the ELP-p15 protein was purified using a simple inverse transition cycling (ITC) process. The ELP tag was cleaved off using tobacco etch virus protease (TEVp), resulting in a tag-free p15 protein. Western blot analysis demonstrated that the p15 protein reacted strongly with ASFV-positive serum. The p15 protein was used as a coating antigen in an indirect ELISA (iELISA) for detecting ASFV antibodies. The p15-iELISA method demonstrated high specificity to ASFV-positive sera, with a maximum detection dilution of 1:1600. Moreover, the method exhibited good reproducibility, with less intra-assay and inter-assay CV values than 10%. Environmental surveillance of porcine reproductive and respiratory syndrome virus (PRRSV) represents a key issue in control of the disease. CD151 has recently been recognized as one of several receptors for PRRSV. We describe here a novel method for concentration of PRRSV from the environmental samples by CD151-binding capture. After fusion to self-aggregating peptide ELK16, the large extracellular loop (LEL) of porcine CD151 and its two segments (namely N63 and C63) were expressed in E. coli as protein aggregates. The three fusion proteins were purified to high purities by regular centrifugation and washing with Triton X-100. Viral binding assay showed that the C63-ELK16 protein, but not ELK16-N63 protein, had the specific binding affinity for PRRSV. The C63-ELK16 protein could bind to, and eluted from, PRRSV in pH-, temperature-, and time-dependent manners with a final virus recovery of 44.7%. By using PRRSV-spiked and experimentally infected pig fecal slurry samples, the C63-ELK16 binding capture-combined quantitative RT-PCR was shown to have higher detection sensitivity than the conventional RT-PCR. Although the viral RNA could be detected in the experimentally infected pig samples with or without C63-ELK16 binding capture, infectious PRRSV was not isolated without C63-ELK16 binding capture. (1) Purpose: Challenges remain in dose accumulation for cervical cancer radiotherapy combined with external beam radiotherapy (EBRT) and brachytherapy (BT) as there are many large and complex organ deformations between different treatments. This study aims to improve deformable image registration (DIR) accuracy with the introduction of multi-metric objectives for dose accumulation of EBRT and BT. (2) Materials and methods: Twenty cervical cancer patients treated with EBRT (45–50 Gy/25 fractions) and high-dose-rate BT (≥20 Gy in 4 fractions) were included for DIR. The multi-metric DIR algorithm included an intensity-based metric, three contour-based metrics, and a penalty term. Nonrigid B-spine transformation was used to transform the planning CT images from EBRT to the first BT, with a six-level resolution registration strategy. To evaluate its performance, the multi-metric DIR was compared with a hybrid DIR provided by commercial software. The DIR accuracy was measured by the Dice similarity coefficient (DSC) and Hausdorff distance (HD) between deformed and reference organ contours. The accumulated maximum dose of 2 cc (D2cc) of the bladder and rectum was calculated and compared to simply addition of D2cc from EBRT and BT (ΔD2cc). (3) Results: The mean DSC of all organ contours for the multi-metric DIR were significantly higher than those for the hybrid DIR (p ≤ 0.011). In total, 70% of patients had DSC > 0.8 using the multi-metric DIR, while 15% of patients had DSC > 0.8 using the commercial hybrid DIR. The mean ΔD2cc of the bladder and rectum for the multi-metric DIR were 3.25 ± 2.29 and 3.54 ± 2.02 GyEQD2, respectively, whereas those for the hybrid DIR were 2.68 ± 2.56 and 2.32 ± 3.25 GyEQD2, respectively. The multi-metric DIR resulted in a much lower proportion of unrealistic D2cc than the hybrid DIR (2.5% vs. 17.5%). (4) Conclusions: Compared with the commercial hybrid DIR, the introduced multi-metric DIR significantly improved the registration accuracy and resulted in a more reasonable accumulated dose distribution. Porcine epidemic diarrhea virus (PEDV) envelope protein (E) has been characterized as an important structural protein that plays critical roles in the interplay with its host to affect the virus life cycle. Stress granules (SGs) are host translationally silent ribonucleoproteins, which are mainly induced by the phosphorylation of eIF2α in the PERK/eIF2α signaling pathway. Our previous study found that PEDV E protein caused endoplasmic reticulum stress response (ERS)-mediated suppression of antiviral proteins' translation. However, the link and the underlying mechanism by which PEDV induces SGs formation and suppresses host translation remain elusive. In this study, our results showed that PEDV E protein significantly elevated the expression of GRP78, CANX, and phosphorylation of PERK and eIF2α, indicating that the PERK/eIF2α branch of ERS was activated. PEDV E protein localized to the ER and aggregated into puncta to reconstruct ER structure, and further induced SGs formation, which has been caused through upregulating the G3BP1 expression level. In addition, a significant global translational stall and endogenous protein translation attenuation were detected in the presence of E protein overexpression, but the global mRNA transcriptional level remained unchanged, suggesting that the shutoff of protein translation was associated with the translation, not with the transcription process. Collectively, this study demonstrates that PERK/eIF2α activation is required for SGs formation and protein translation stall. Background: Robotic linac is ideally suited to deliver hypo-fractionated radiotherapy due to its compact head and flexible positioning. The non-coplanar treatment space improves the delivery versatility but the complexity also leads to prolonged optimization and treatment time.Methods: In this study, we attempted to use the deep learning (pytorch) framework for the plan optimization of circular cone based robotic radiotherapy. The optimization problem was topologized into a simple feedforward neural network, thus the treatment plan optimization was transformed into network training. With this transformation, the pytorch toolkit with high-efficiency automatic differentiation (AD) for gradient calculation was used as the optimization solver. To improve the treatment efficiency, plans with fewer nodes and beams were sought. The least absolute shrinkage and selection operator (lasso) and the group lasso were employed to address the "sparsity" issue.Results: The AD-S (AD sparse) approach was validated on 6 brain and 6 liver cancer cases and the results were compared with the commercial MultiPlan (MLP) system. It was found that the AD-S plans achieved rapid dose fall-off and satisfactory sparing of organs at risk (OARs). Treatment efficiency was improved by the reduction in the number of nodes (28%) and beams (18%), and monitor unit (MU, 24%), respectively. The computational time was shortened to 47.3 s on average.Conclusions: In summary, this first attempt of applying deep learning framework to the robotic radiotherapy plan optimization is promising and has the potential to be used clinically. The serious shuttle effect, sluggish reduction kinetics of polysulfides and the difficult oxidation reaction of Li2S have hindered LiS battery practical application. Herein, a 3D hierarchical structure composed of NiMoO4 nanosheets in situ anchored on NS doped carbon clothes (NiMoO4@NSCC) as the free‐standing host is creatively designed and constructed for LiS battery. Dual transitional metal oxide (NiMoO4) increases the electrons density near the Fermi level due to the contribution of the incorporating molybdenum (Mo), leading to the smaller bandgap, and thus stronger metallic properties compared with NiO. Furthermore, as a bidirectional catalyst, NiMoO4 is proposed to facilitate reductions of polysulfides through lengthening the SS bond distance of Li2S4 and reducing the free energy of polysulfides conversion, meanwhile promote critical oxidation of insulative discharge product (Li2S) via lengthening LiS bond distance of Li2S and decreasing Li2S decomposition barrier. Therefore, after loading sulfur (2 mg cm−2), NiMoO4@NSCC/S as the self‐supporting cathode for the LiS battery exhibits impressive long cycle stability. • A copolymer is devised by PMVS, PTMG and PCL as main chain and PAA as side chain. • The synthetic endows PLLA with water-responsive shape memory characteristics. • The incorporated PLLA shows good repeatable shape memory and great flexibility. Poly(L-lactide) (PLLA) was one of the most well-known biodegradable polyesters. Although its thermo-responsive shape memory (SM) function has been studied by proper program, high transformation temperature is not suitable in some biomedical applications. Water-responsive SM polymer is an alternative choice and exhibits great potential because it is harmless to the cells and tissues of organisms. In the work, a novel PCL-(PMVS- s -PAA)-PCL-PTMG-PCL-(PMVS- s -PAA)-PCL copolymer was devised as an additive to endow PLLA with water-responsive SM capability. The incorporated PLLA was characterized by cross-section, water contact angle, differential scanning calorimetry (DSC) and dynamic mechanical analysis (DMA). Inserted PTMG segment and assisted ambient temperature were contributed to its SM behaviors. The shape fixation ratio and shape recovery ratio were 92.0% and 94.2%, respectively. The schematic diagram between internal microstructures and macroscopic properties was proposed. At the same time, the incorporated PLLA showed excellent toughness. Synergistic delivery of two drugs is a desirable choice to overcome drug resistance. Resveratrol (RES) and xanthohumol (XAN) which are both new drugs originated from plants exhibit great potential in the treatment of cancer, but there are few studies on combining them in a delivery system. In this work, a new core/shell fiber mesh containing RES and XAN was designed out via coaxial electrospinning, and the effect of drug content on the medicated fibers was discovered. RES/polyethylene oxide (PEO) could be well combined with XAN/Poly(lactide-glycolide) (PLGA) to make core/shell fibers, but fibrous morphology worsened with increasing drug content of the core or shell to some extent. The core/shell was amorphous nature, and changing drug content had little influence on the wetability of the core/shell fibers. The release accelerated with drug content increasing in the fibers, and RES and XAN showed gradient release, which the release rate of RES was slower than XAN. These two drugs could sustain release for 350 h. When the blend ratios of RES/PEO and XAN/PLGA were 50/50 and 10/90 in the fibers, the fiber mesh showed the best mechanical properties, of which the tensile strength and elongation at break were 2.85 ± 0.10 MPa and 55.23 ± 2.53%, respectively. Oligodendrocyte precursor cells (OPCs) undergo an extensive and coordinated migration in the developing CNS, using the pre-formed scaffold of developed blood vessels as their physical substrate for migration. While OPC association with vasculature is critical for dispersal, equally important for permitting differentiation and proper myelination of target axons is their appropriate and timely detachment, but regulation of this process remains unclear. Here we demonstrate a correlation between the developmental formation of astrocytic endfeet on vessels and the termination of OPC perivascular migration. Ex vivo and in vivo live imaging shows that astrocyte endfeet physically displace OPCs from vasculature, and genetic abrogation of endfoot formation hinders both OPC detachment from vessels and subsequent differentiation. Astrocyte-derived semaphorins 3a and 6a act to repel OPCs from blood vessels at the cessation of their perivascular migration and, in so doing, permit subsequent OPC differentiation by insulating them from a maturation inhibitory endothelial niche. Oligodendrocyte (OL) maturation arrest in human white matter injury contributes significantly to the failure of endogenous remyelination in multiple sclerosis (MS) and newborn brain injuries such as hypoxic ischemic encephalopathy (HIE) that cause cerebral palsy. In this study, we identify an oligodendroglial-intrinsic factor that controls OL maturation specifically in the setting of injury. We find a requirement for the ring finger protein Rnf43 not in normal development but in neonatal hypoxic injury and remyelination in the adult mammalian CNS. Rnf43, but not the related Znrf3, is potently activated by Wnt signaling in OL progenitor cells (OPCs) and marks activated OPCs in human MS and HIE. Rnf43 is required in an injury-specific context, and it promotes OPC differentiation through negative regulation of Wnt signal strength in OPCs at the level of Fzd1 receptor presentation on the cell surface. Inhibition of Fzd1 using UM206 promotes remyelination following ex vivo and in vivo demyelinating injury.
