Reinforcement configuration of Al composites has recently been at the center of attention. In this study, through the analysis of microstructure and tensile properties, effects of Cu content on the reinforcing behaviors of AlN network were investigated in details. Results indicated that the increasing amount of Cu would form micron Al2Cu which would decorated on the AlN network. With the increasing amount of Cu addition from 1.5% to 6%, the particle size of Al2Cu increased from 1 μm to 10 μm. Besides the micron Al2Cu, Cu elements were also existed in θ′ precipitates in the region where were poor of AlN and segregated on the surface of AlN. Given the microstructure changes with different Cu addition, the tensile properties at room temperature increased with Cu addition while the average tensile strength at 350 °C were decreased from 102 MPa to 85 MPa. The addition of elements would affect the bearing capacity of the 8.2AlN/Al–Cu composites at 350 °C. Firstly, the micron Al2Cu phase would suffer the load and get stress concentration. It would lead to the premature imitation of voids and strongly impair the strengthening behavior of AlN network. Secondly, the nano θ′ precipitates would become coarsen and the hinder effects of dislocation would be enhanced at 350 °C. Finally, the segregated Cu atoms on the AlN surface could increase the deformation coordination of AlN network and help decrease the stress concentration.

Background: College is a high-risk period for depressive symptoms. Anxiety sensitivity (AS) is a major predictor of depressive symptoms during college years, but the specific mechanisms remain unclear. This study investigated the relationship using a chain mediation model. Methods: A sample of 1261 eligible students (mean age of 20.20 ± 1.37 years) residing in Liaoning Province, located in the northeastern part of China, were selected through a convenient sampling method for the purpose of carrying out a comprehensive survey. The self-report questionnaires used in the study encompassed a range of parameters, including the general information questionnaire, anxiety sensitivity index-3 (ASI-3), resilience scale-14 (RS-14), depression, anxiety, and stress scale (DASS-21). Pearson correlation analysis, structural equation model testing, and Bootstrap deviation correction percentile analysis were employed to analyze the survey data using the R 4.3.2 statistical software. Results: The total AS score and its three lower-order dimensions were positively correlated with stress and depressive symptoms. Resilience was negatively correlated with AS, stress and depressive symptoms. In addition to the direct influence of AS on depressive symptoms, stress and resilience play a partial mediating role in the relationship between AS physical and cognitive concerns and depressive symptoms, the total indirect effects accounted for 91.5 % and 92.1 %, respectively, and fully mediate the relationship between AS social concerns and depressive symptoms. Conclusions: The study's outcomes indicate that practical actions can be taken to decrease AS and stress, thereby enhancing resilience and reducing depressive symptoms in adolescents.

In order to study the relieving effect of extracellular polysaccharides (EPS-A,EPS-B) which were extracted and purified from long shelf-life yogurt fermented by long shelf-life yogurt culture A and culture B, their molecular characteristics, structural morphology and monosaccharide composition were determined, and the mouse weight, influence of colon histopathology, the expression of inflammatory factors, the content of myeloperoxidase (MPO) and tight junction proteins (ZO-1, Occludin) in mice were also analyzed through constructing an animal model of colonic inflammation in mice by dextran sulfate sodium (DSS). The results showed that there were significantly differences on the molecular morphology and monosaccharide compositions between EPS-A and EPS-B. The EPS-A was slightly loose in structure, composed of three monosaccharides (galactosamine hydrochloride, galactose and glucose) with a molar ratio of 0.345:0.21:0.435. The structure of EPS-B was compact and it composed of four monosaccharides (galactosamine hydrochloride, glucosamine hydrochloride, galactose and glucose) with a molar ratio of 0.421:0.05:0.207:0.322. The weight of IBD mice was recovered in all the exopolysaccharide treatment groups, but there was no significant decrease in the disease activity index (DAI) score. The colon morphology of the high dose group of EPS-A was close to that of the blank control group, and there was no significant difference in colon length and colon coefficient between the two groups (P>0.05), the same below). Combined with the expression of inflammatory factor (TNF-α, IL-1β, IL-10) in different treatment groups, the high dose group of EPS-A had the best response to those inflammatory factors, and there was no significant difference between it and the drug treatment group (P>0.05). The expression of ZO-1 and occludin protein in the high-dose and low-dose groups of EPS-A was significantly improved and same as the drug treatment group (P0.05). In summary, it could be seen that EPS-A can significantly improve the enteritis of mice, and its effect was better than that of EPS-B.

This investigation involved the extraction of a novel polysaccharide from the spore fermentation broth of Tremella fuciformis using a method that combined ultrasonic and enzymatic assistance with ethanol precipitation. It was then compared with enzymatic and ultrasonic extraction methods. The objective of this research is to offer a reference point for expanding the application of ultrasonic-assisted enzymatic extraction technology in T. fuciformis polysaccharides (TFPs). Based on single-factor experiments, Box-Behnken was used to optimize the extraction conditions of TFPs by ultrasonic-enzymatic-assisted ethanol precipitation extraction. The results revealed an optimal combination of enzymes, with a cellulase-to-papain ratio of 2:1, an enzyme addition of 4000U/100 mL, an enzymolysis temperature of 49 °C, ultrasonic power at 3 W/mL and an ultrasonic time of 20 min. The extraction rate of TFPs and α- amylase inhibition rates were 23.94 % and 61.44 %, respectively. Comparing the physicochemical properties, structural characterization and in vitro activity of TFPs extracted through different methods, the results showed that ultrasonic treatment significantly influences the apparent morphology of polysaccharide and could enhance its in vitro biological activity. However, different extraction techniques exhibit insubstantial impact on the chemical composition, glycosidic bonds or glycosidic ring configurations within the polysaccharides. Among them, ultrasonic-enzymatic-assisted ethanol precipitation extraction of polysaccharide has the highest extraction rate and the lowest viscosity. It has significant effects on ABTS+ scavenging activity, α- amylase inhibition rate and glucose dialysis retardation index, polysaccharide treated with ultrasonic-enzymatic showed the best performance. These findings suggest that ultrasonic-enzymatic-assisted ethanol precipitation extraction can enhance the activities of TFPs, thereby providing a valuable insight for their future development and application.

Abstract Safflower is an annual herb of Compositae, which has great economic value. To explore the impact of invasive weed Xanthium italicum Moretti on the economic crop safflower, field experiments were conducted, the growth-related characters and the relative intensity of competition between the two species was explored. The results showed that under monoculture conditions, the stem height, crown width, stem diameter and the biomass of X. italicum root, stem and leaves were 1.14, 1.96, 1.82, 4.42, 4.21 and 3.99 times as high as those of safflower, respectively. When the two species coexisted, the growth related characters of X. italicum were further significantly improved, while the growth related characters of safflower were significantly decreased. When coexisted with X. italicum, the corolla biomass, hydroxysafflor yellow A content of corolla, seed yields, 100-seed weight, and seed oil content of safflower in the interplanted treatment 90.04%, 33.11%, 63.89%, 40.58%, and 25.61% lower than those in the monocultured treatment, respectively. Relative yield (RY) and Competitive balance index (CB) of X. italicum and safflower showed that the interspecific competitiveness of X. italicum was significantly higher than that of safflower. Under the competitive inhibition of X. italicum, not only the vegetative growth, but also the reproductive growth, yield, and quality of the economic organs of safflower were significantly negatively impacted. Together, our findings provide important scientific basis for evaluating the invasion risks and consequences of safflower’s cropland ecosystem by X. italicum.

Abstract The Omicron transmission has infected nearly 600,000 people in Shanghai from March 26 to May 31, 2022. Combined with different control measures taken by the government in different periods, a dynamic model was constructed to investigate the impact of medical resources, shelter hospitals and aerosol transmission generated by clustered nucleic acid testing on the spread of Omicron. The parameters of the model were estimated by least square method and MCMC method, and the accuracy of the model was verified by the cumulative number of asymptomatic infected persons and confirmed cases in Shanghai from March 26 to May 31, 2022. The result of numerical simulation demonstrated that the aerosol transmission figured prominently in the transmission of Omicron in Shanghai from March 28 to April 30. Without aerosol transmission, the number of asymptomatic subjects and symptomatic cases would be reduced to 130,000 and 11,730 by May 31, respectively. Without the expansion of shelter hospitals in the second phase, the final size of asymptomatic subjects and symptomatic cases might reach 23.2 million and 4.88 million by May 31, respectively. Our results also revealed that expanded vaccination played a vital role in controlling the spread of Omicron. However, even if the vaccination rate were 100%, the transmission of Omicron should not be completely blocked. Therefore, other control measures should be taken to curb the spread of Omicron, such as widespread antiviral therapies, enhanced testing and strict tracking quarantine measures. This perspective could be utilized as a reference for the transmission and prevention of Omicron in other large cities with a population of 10 million like Shanghai.

Abstract Background It is critical to understand the sensitivity, response direction and magnitude of carbohydrates and secondary compounds to warming for predicting the structure and function of the tundra ecosystem towards future climate change. Results Open-top chambers (OTCs) were used to passively increase air and soil temperatures on Changbai Mountain alpine tundra. After seven years’ continuous warming (+ 1.5 °C), the vegetation coverage, nonstructural carbohydrates (soluble sugars and starch) and secondary compounds (total phenols, flavonoids and triterpenes) of leaves and roots in three dominant dwarf shrubs, Dryas octopetala var. asiatica, Rhododendron confertissimum and Vaccinium uliginosum, were investigated during the growing season. Warming did not significantly affect the concentrations of carbohydrates but decreased total phenols for the three species. Carbohydrates and secondary compounds showed significantly seasonal pattern and species-specific variation. No significant trade-off or negative relationship between carbohydrates and secondary compounds was observed. Compared to Dr. octopetala var. asiatica, V. uliginosum allocated more carbon on secondary compounds. Warming significantly increased the coverage of Dr. octopetala var. asiatica, did not change it for V. uliginosum and decreased it for Rh. confertissimum. Rh. confertissimum had significantly lower carbohydrates and invested more carbon on secondary compounds than the other two species. Conclusions Enhanced dominance and competitiveness of Dr. octopetala var. asiatica was companied by increased trend in carbohydrate concentrations and decreased ratio of secondary compounds to total carbon in the warming OTCs. We, therefore, predict that Dr. octopetala var. asiatica will continue to maintain dominant status, but the competition ability of V. uliginosum could gradually decrease with warming, leading to changes in species composition and community structure of the Changbai tundra ecosystem under future climate warming.

Frequent injections at high concentrations are often required for many therapeutic proteins due to their short in vivo half-life, which usually leads to unsatisfactory therapeutic outcomes, adverse side effects, high cost, and poor patient compliance. Herein we report a supramolecular strategy, self-assembling and pH regulated fusion protein to extend the in vivo half-life and tumor targeting ability of a therapeutically important protein trichosanthin (TCS). TCS was genetically fused to the N-terminus of a self-assembling protein, Sup35p prion domain (Sup35), to form a fusion protein of TCS-Sup35 that self-assembled into uniform spherical TCS-Sup35 nanoparticles (TCS-Sup35 NP) rather than classic nanofibrils. Importantly, due to the pH response ability, TCS-Sup35 NP well retained the bioactivity of TCS and possessed a 21.5-fold longer in vivo half-life than native TCS in a mouse model. As a result, in a tumor-bearing mouse model, TCS-Sup35 NP exhibited significantly improved tumor accumulation and antitumor activity without detectable systemic toxicity as compared with native TCS. These findings suggest that self-assembling and pH responding protein fusion may provide a new, simple, general, and effective solution to remarkably improve the pharmacological performance of therapeutic proteins with short circulation half-lives.

The effect of nitrogen-modified atmosphere storage (NS) on peanut lipid oxidation was investigated in this paper. Non-targeted lipidomics was employed to detect the lipid metabolites in peanuts with the aim of exploring the mechanism of lipid oxidation in peanuts under different storage conditions. The results showed that compared with conventional storage (CS), NS significantly (p < 0.05) delayed the increase in acid value, carbonyl value, and 2-thiobarbituric acid value and the decrease in vitamin E content. However, the storage time has a much greater effect on lipid oxidation than the oxygen level in the storage environment. Lipidomics analysis revealed that there were significant differences in metabolite changes between CS and NS. NS reduced the decline of most glycerophospholipids by regulating lipid metabolism in peanuts. NS maintained higher levels of Diacylglycerol (DAG), sulfoquinovosyl diacylglycerol (SQDG), lysophophatidylcholine (LPC), lysophosphatidylethanolamine (LPE) and phosphatidylinositol (PI) compared to CS. This work provided a basis for the application of NS technology to peanut storage.

γ-Polyglutamic acid (γ-PGA) is a kind of polymer material with good biocompatibility and degradability, which is widely used in food, medicine, environmental protection, cosmetics and agriculture. As γ-PGA is produced by microbial fermentation, its separation, purification and quantitative detection are very important. In this study, the separation methods such as organic solvent precipitation, copper salt precipitation and membrane separation precipitation were mainly discussed, and the quantitative determination methods such as spectrophotometer, enzyme method and viscosity method of γ-PGA were discussed and compared. Furthermore, Molecular imprinting as a new method for the separation of γ-PGA has also been analyzed and discussed in order to provide a reference for the separation and purification of γ-PGA.

Abstract Manipulating the polarizations of electroagnetic waves by flexible and diverse means is desirable for myriad microwave systems. More recently, metasurfaces have emerged as promising alternatives to conventional polarization manipulation components because the flexibility of their geometry means that they can be arbitrarily customized. In this context, a bilayered metasurface is presented to simultaneously manipulate the polarized states of reflected and transmitted microwaves. Regardless of whether an incident electromagnetic wave is x-polarized or y-polarized, the reflected and transmitted waves are converted into their orthogonal waves at the operating frequency. The designed metasurface has a high polarization conversion rate, above 90%, for both normal and oblique incidences. Experimental results verify the correctness of the simulated results. Finally, the axial ratio and surface current distributions are employed to reveal the physics of the polarization manipulation. The proposed metasurface will be beneficial in the design of flexible and versatile polarization converters, has great potential for applications in polarization-controlled devices and is believed to be extendable to higher frequency regimes.

Lotus flower polysaccharide (LFP) and lotus seedpod polysaccharide (LSP) were separated by water extract–alcohol precipitation, and their structures and biological activities were investigated. The results of monosaccharide composition showed that LFP and LSP were composed of nine monosaccharides, fucose, rhamnose, arabinose, glucose, galactose, mannose, fructose, galacturonic acid, and glucuronic acid, with the molar percentages of 0.18: 0.43: 2.26: 45.22: 32.14: 4.28: 8.20: 6.28: 1.01 and 2.70: 1.02: 8.15: 45.63: 20.63: 1.44: 2.59: 16.45. LSP and LFP exhibited molecular weights of 9.37 × 104 Da and 1.24 × 106 Da, respectively. SEM showed that LFP and LSP have similar structures; XRD analysis showed that both polysaccharides had crystalline structure and amorphous structure. The results of ABTS+, DPPH, hydroxyl radical scavenging experiment, and a reducing power experiment showed that LFP and LSP had good antioxidant capacity. Cell viability findings showed that polysaccharide concentrations of lotus flower and lotus seedpod could enhance cellular proliferation ranging from 25 to 400 μg/mL without cytotoxicity. By inducing the production of crucial proteins in the TLR4/NF-κB pathway, LFP and LSP were able to induce autophagy in RAW264.7, according to the results of the RT-PCR and Western blotting assays.

A single-layer differential-fed (DF) wideband metasurface (MTS) antenna is proposed in this paper. As the prototype, a three-by-three MTS formed by identical rectangular patches is investigated at first. We observe that there are many unwanted higher-order modes (HOMs) resonating near the wanted fundamental mode. Two probes with differential signals feed MTS on its centerline to suppress the majority of HOMs. The remaining HOM can be removed from the discussed frequency range by modifying the prototype MTS to a nonuniform structure. Then, the optimal feeding positions (FPs) are determined by a quantitative prediction of the impendence bandwidth (IBW) without any physical feeds involved. The processes of HOMs suppression and FPs determination are based on characteristic mode analysis with the virtual probes. Moreover, two interdigital capacitor plates are loaded on the probes to improve the impedance matching of the antenna. Finally, the proposed DF MTS antenna is fabricated and measured. The measured −10-dB IBW is 18.4% (4.93 to 5.93 GHz) with broadside radiation, stable high gains, and front-to-back ratios better than 21 dB.

In this work, a transmission polarizer is described by using a frequency-selective surface to transform linearly polarized waves into circularly polarized waves. The linear to circular (LTC) polarizer consists of four layers. Two types of unit cells are designed in the LTC polarizer to improve design freedom. As a result, two orthogonal polarized components of transmission waves display nearly 90° phase differences while maintaining nearly high transparency for the generation of CPWs. The less than 3 dB axial ratio with a fractional width of over 76.8% from 9.20 to 20.67 GHz for this LTC polarizer is obtained. Even when the incident angle reaches 20°, its operating frequency band covers 9.69 to 20.21 GHz. Compared with the converters proposed before, the one proposed in this paper has a wider bandwidth. In order to evaluate the design strategy, a prototype is manufactured and tested. The results of the simulation and the experiment are in good accord.

Evidence suggests that exposure to coal dust increases immunoglobulin concentration. However, there is a paucity of data reporting immunoglobulin G (IgG) subclass in coal workers’ pneumoconiosis (CWP). Therefore, this study intended to evaluate potential diagnostic biomarkers for the disease. CWP patients, dust-exposed workers without pneumoconiosis (DEW), and matched healthy controls (HCs) presented to the General Hospital of Datong Coal Mining Group and Occupational Disease Prevention and Treatment Hospital of Datong Coal Mining Group between May 2019 and September 2019 were recruited. The serum immunoglobulin concentration was determined by the multiplex immunoassay technique. Totally, 104 CWP patients, 109 DEWs, and 74 HCs were enrolled. Heavy metals released in the environment pose a huge threat to soil and water quality, food safety and public health. Additionally, humans and other mammals may also be directly exposed to heavy metals or exposed to heavy metals through the food chain, which seriously threatens the health of animals and humans. Chromium, especially hexavalent chromium [Cr (VI)], as a common heavy metal, has been shown to cause serious environmental pollution as well as intestinal damage. Thus, increasing research is devoted to finding drugs to mitigate the negative health effects of hexavalent chromium exposure. Seaweed polysaccharides have been demonstrated to have many pharmacological effects, but whether it can alleviate gut microbial dysbiosis caused by hexavalent chromium exposure has not been well characterized. Here, we hypothesized that seaweed polysaccharides could alleviate hexavalent chromium exposure-induced poor health in mice. Mice in Cr and seaweed polysaccharide treatment group was compulsively receive K2Cr2O7. At the end of the experiment, all mice were euthanized, and colon contents were collected for DNA sequencing analysis. Results showed that seaweed polysaccharide administration can restore the gut microbial dysbiosis and the reduction of gut microbial diversity caused by hexavalent chromium exposure in mice. Hexavalent chromium exposure also caused significant changes in the gut microbial composition of mice, including an increase in some pathogenic bacteria and a decrease in beneficial bacteria. However, seaweed polysaccharides administration could ameliorate the composition of gut microbiota. In conclusion, this study showed that seaweed polysaccharides can restore the negative effects of hexavalent chromium exposure in mice, including gut microbial dysbiosis. Meanwhile, this research also lays the foundation for the application of seaweed polysaccharides.

The isolation of Stropharia rugosoannulata polysaccharide (SRP) by three-phase extraction was optimized, and its structure and biological activities were identified. The optimal extraction conditions were: mass fraction of ammonium sulfate, 20%; volume ratio of sample solution to t-butanol, 1:1.5; extraction temperature, 35°C. Under these conditions, the yield of SRP was 6.85% ± 0.13%. SRP was found to be composed of glucose (35.79%), galactose (26.80%), glucuronic acid (9.92%), fructose (8.65%), xylose (7.92%), fucose (4.19%), arabinose (3.46%) and rhamnose (3.26%), with the molecular weight of 27.52 kDa. The results of DPPH, hydroxyl, ABTS+ radical scavenging and reducing power tests showed that SRP had good antioxidant capacities. SRP had no cytotoxic effect on RAW264.7 macrophages at the concentrations of 25-200 μg/mL, and could significantly promote phagocytosis activity and cell migration according to CCK-8 assay, phagocytosis assay and cell scratch experiment. SRP can significantly stimulate the transcript expression levels of TNF-α, IL-1β and IL-6, as determined by RT-PCR and Western blot assays. SRP activated the TLR4/NF-κB signaling pathway, and autophagy also occurred. These results suggest that SRP is a safe antioxidant and immunomodulator, and that it can be used in the development of functional foods and/or pharmaceuticals.

IntroductionLong-chain fatty acids in breast milk are affected by the mother’s diet and play an important role in the growth, development, and immune construction of infants. This study aims to explore the correlation between maternal diet, breast milk fatty acids (FAs), and the infant intestinal flora.MethodsWe enrolled 56 paired mothers and their infants; both breast milk samples and infants’ fecal samples were collected to determine the long-chain FA content of breast milk by ultra-performance liquid chromatography–tandem mass spectrometry (UPLC-MS), and metagenomic technology was applied to determine the microbial composition of infant feces. The maternal diet was also investigated using a 24-h dietary recall.ResultsThe results indicated that the fat contribution rates of edible oils in the maternal diet are significantly positively correlated with the contents of certain long-chain fatty acids (C16:0, C18:1, C16:1, and C22:4) in breast milk, which mainly regulate the abundance of Lacticaseibacillus rhamnosus, Lacticaseibacillus fermentum, and Lacticaseibacillus paracasei in the infant gut. Through KEGG pathway analysis, our data revealed that the long-chain FAs in different groups of breast milk were significantly correlated with the pathways of biotin metabolism, glycerolipid metabolism, and starch and sucrose metabolism.DiscussionThe results of this study suggest a pathway in which the diets of lactating mothers may affect the composition of the infant intestinal microbiota by influencing breast milk FAs and then further regulating infant health.

Acute respiratory illness caused by human adenovirus have been increasing in morbidity and mortality in recent years. Currently, isolation of symptomatic infected individuals is the primary means of controlling outbreaks in closed spaces such as schools and military camps. However, the disease is still spreading despite the implementation of control measures. To reveal the underlying mechanism of this phenomenon, we propose a dynamic model that considers invisible transmission and isolated confirmed cases. By calculating and analyzing the control reproduction number, it is found that asymptomatic infected individuals play an important role in the spread of the epidemic. Therefore, in the absence of specific vaccines, non-pharmaceutical interventions such as quarantine of exposed individuals are effective means to mitigate severity. The results show that the earlier the control of invisible transmission is implemented, the lower the peak and the shorter the duration of the outbreak. These findings will provide the theoretical basis and recommendations for prevention and control of human adenovirus transmission in closed spaces.

Yeast β-glucan (YG) possess an extensive range of biological activities, such as the inhibition of oxidation, but the poor water solubility of macromolecular YG limits its application. In this study, through the combined degradation of ultrasonic waves and H2O2, and the optimization of the main process parameters for solubilizing YG by response surface methodology (RSM), a new product of YGUH was generated. The molecular weight, structural characteristics and degradation kinetics before and after solubilization were evaluated. The results showed that the optimal solubilization conditions were reaction time: 4 h, ultrasonic power: 3 W/mL, H2O2 concentration: 24 %. Under these conditions, ultrasound-assisted H2O2 increased the solubility (from 13.60 % to 70.00 %) and reduced molecular weight (from 6.73 × 106 Da to 1.22 × 106 Da). Fourier transform infrared spectroscopy (FTIR), nuclear magnetic resonance (NMR), Congo red (CR), scanning electron microscopy (SEM) revealed that ultrasound-assisted H2O2 increased the conformation's flexibility greatly, without changing the main structure of YG. More importantly, solubilization of YG improved free radical scavenging activity with YGUH exhibiting the highest levels of DPPH and ABTS+ free radical scavenging activity. These results revealed that ultrasound-assisted H2O2 degradation could be a suitable way to increase the solubility of YG for producing value-added YG.

Microbial inoculants are widely used in wastewater treatment, soil remediation, and biological control. Safety and compliance for active constituents are considered to be the most important measures of imported microbial inoculants. Microbial inoculants composition was commonly identified by phenotypic culture, which is time-consuming and labor intense with occasionally false negative results provided, and can only be tested for specific species. High-throughput sequencing (HTS), known for its non-targeted detection of unknown species composition in samples, is suitable for composition consistency identification and biosafety analysis of imported microbial inoculants. In this study, the application of HTS for microflora distribution and resistance gene was verified in microbial inoculants for environmental protection and then applicated in imported microbial inoculants. Both Illumina- and Nanopore-based HTS methods identified the same dominant bacterial species successfully in the imported microbial inoculants. The main component of bacterial species was Bacillus subtilis, Bacillus amyloliquefaciens, Bacillus licheniformis, and Enterococcus faecium, and further confirmed with traditional methods. The antibiotic resistance genes Bacillus subtilis mprF, bcrA, blt, lmrB, rphB, tet(L), tmrB, vmlR, ykkC, and ykkD were detected in all samples. Our results indicated that HTS processes the application potential to identify the active ingredients of microbial inoculants. Therefore, rapid and accurate identification of the microbial compositions in microbial formulation products is of high importance for port biosafety supervision.

The outcomes of 80 patients with hematologic malignancies who received haploidentical peripheral blood stem cell transplantation (haplo-PBSCT) combined with unrelated cord blood (UCB) from March 2017 to June 2020 were analyzed in this retrospective study. Anti-thymocyte globulin(ATG) was administered at a dose of 7.5 mg/kg. The median time for neutrophil and platelet engraftment was 13(range: 8-22) days and 14(range: 8-103) days, respectively. The 30-day cumulative incidence of neutrophil engraftment was 100%, and the 100-day cumulative incidence of platelet engraftment was 95%. All patients achieved complete haploidentical peripheral blood stem cell engraftment, and no cord blood chimerism was observed. The cumulative incidence of grades II-IV and grades III-IV acute graft-versus-host disease (aGVHD) on 100-day was 26.3%(95%CI: 17.2%–36.3%) and 5.0%(95%CI: 1.6%–11.4%), respectively. The estimated cumulative incidence of chronic GVHD (cGVHD) and moderate-severe cGVHD at 3-year was 43.3%(95%CI: 31.6%–54.4%) and 16.0%(95%CI: 8.7%–25.2%), respectively. The estimated 3-year cumulative incidence of relapse and non-relapse mortality was 18.8%(95%CI: 10.0%–29.7%) and 17.8%(95%CI: 9.9%–27.5%), respectively. The estimated 3-year probabilities of overall survival, disease-free survival, GVHD/relapse-free survival were 77.6%(95%CI: 68.3%–88.1%), 63.4%(95%CI: 52.6%–76.5%), and 55.5%(95%CI: 44.8%–68.7%), respectively. These satisfying results suggested that haplo-PBSCT combined with UCB is an alternative transplantation protocol for hematologic malignancies.

The structure and moisture retention of Tremella polysaccharide fermented from GCMCC5.39 (FTP) were evaluated. After UV, infrared spectrum analysis, HPAEC-PAD, HPSEC and 1 D NMR analysis, the composition of the purified FTP was determined. Purified components of fermented Tremella polysaccharide (FTPS) was made of galactose, mannose, glucose, galactosmine, glucosamine, and contain a large amount of hydroxyl, carbonyl and amino groups. FTPS was α -neutral pyranose without uronic acid. FTPS-1 and FTPS-2 were obtained after purification by DEAE-Sepharose Fast Flow Column. The molecular weights of FTPS-1 and FTPS-2 were 25 722 and 177 263 Da. FTPS-2 had a better ability to prevent moisture loss, and the optimal moisture retention period was 0–4 h. FTPS-2 could significantly increase the moisture content of the skin epidermis and showed a dose-concentration relationship. The effect of FTPS-2 on the expression of different moisturizing genes was evaluated in a human skin keratinocyte model. The results showed that FTPS-2 has no cytotoxicity, and could significantly promote AQP3, TGM1, CASP14, HYAL2, FLG gene expression level in HaCaT cells. It has the most significant influence at HYAL2 protein expression on 50 μg/mL.

Recent studies have shown that haploidentical hematopoietic stem cell transplantation supported by third-party cord blood (haplo-cord-HSCT) results in rapid hematopoietic recovery, low incidences of graft-versus-host disease (GVHD), and relapse of hematologic malignancies. However, few reports on haploidentical peripheral blood stem cell transplantation supported by third-party cord blood (haplo-cord-PBSCT) have been published. To evaluate the outcomes of patients who underwent haplo-cord-PBSCT or human leukocyte antigen (HLA)-matched sibling donor peripheral blood stem cell transplantation (MSD-PBSCT), we retrospectively reviewed the clinical data of patients with hematologic malignancies who underwent haplo-cord-PBSCT (n = 93) or MSD-PBSCT (n = 72) in our hospital from March 2017 to December 2020. In the haplo-cord-PBSCT and MSD-PBSCT groups, the median time for neutrophil and platelet engraftment was 13 vs. 12 days (p = 0.07) and 16 vs. 13 days (p = 0.06), respectively. The 30-day cumulative incidences of neutrophil engraftment were 100.0% and 98.6% (p = 0.12). The 100-day cumulative incidences of platelet engraftment were 96.8% and 98.6% (p = 0.01). The 100-day cumulative incidences of grade II–IV and grade III–IV acute GVHD were 29.1% vs. 23.6% (p = 0.42) and 9.7% vs. 4.2% (p = 0.18). The cumulative incidences of total and moderate/severe chronic GVHD at 1 year were 26.5% vs. 17.4% and 8.1% vs. 4.5%, respectively, and at 3 years were 34.7% vs. 34.3% (p = 0.60) and 13.6% vs. 10.6% (p = 0.49), respectively. The cumulative incidences of relapse at 1 year were 9.3% and 7.2% and at 3 years were 17.0% and 17.0% (p = 0.98). Non-relapse mortality (NRM) at 1 year was 14.6% and 8.6% and at 3 years was 17.4% and 8.6% (p = 0.13) in two groups. The probabilities of overall survival (OS), disease-free survival (DFS), and GVHD-free/relapse-free survival (GRFS) at 1 year were 81.7% vs. 88.6%, 76.1% vs. 84.2%, and 71.7% vs. 79.7%, respectively, and at 3 years were 78.7% vs. 79.0%, 65.6% vs. 74.4%, and 55.5% vs. 63.6%, respectively, in the corresponding group, p > 0.05. In conclusion, for patients with acute myeloid leukemia/myelodysplastic syndrome (AML/MDS) and acute lymphoid leukemia (ALL), haplo-cord-PBSCT results in similar outcomes compared with MSD-PBSCT, and it may be a valid alternative transplantation method.

Coix seed extract (CSE) and probiotics have been reported to regulate glycolipid metabolism via different modes of action. We tested the effects of CSE, Bifidobacterium BPL1, and their combination to determine their effects on glycolipid metabolism in obese mice. Male C57BL/6J mice were fed a high-fat diet for 8 weeks to establish an obesity model. Obese mice were selected and divided into four groups: the model control group and three intervention groups. After 10 weeks of continuous gavage intervention, the mice in the intervention groups exhibited lower body weight (lower about 2.31 g, vs. HFD mice 42.23 g) and epididymal (lower about 0.37 g, vs. HFD mice 2.5 g) and perirenal fat content (lower about 0.47 g, vs. HFD mice 0.884 g); decreased fasting blood glucose, total cholesterol, triglycerides, and VLDL; and increased HLDL, respiratory exchange ratio, energy expenditure, and amount of exercise performed. CSE, BPL1 and their combination can effectively control the weight gain in obese mice, reduce fat content, and regulate blood lipids and abnormal blood sugar. These results may be related to reduce the chronic inflammatory states, improve energy metabolism, exercise, relieve insulin sensitivity, and reduce lipid synthesis via the intervention of CSE, BPL1 and their combination. Compared with the single use of CSE alone, the combination of CSE + BPL1 can better exert the regulation function of intestinal flora, and change in the abundance of bacteria that could improve the level of inflammatory factors, such as increasing Bifidobacterium, reducing Lactococcus. Compared with the use of BPL1 alone, the combination of CSE and BPL1 can better regulate pancreatic islet and improve blood sugar. CSE may act directly on body tissues to exert anti-inflammatory effects. BPL1 and CSE + BPL1 may improve the structure and function of the intestinal flora, and reduce tissue inflammation.

The purpose of this study was to investigate the effect of ultrasound-assisted enzymolysis on modified solubilization of yeast β-glucan and its related mechanism. The depolymerization effects of this system on the physicochemical properties and structural features of the degraded fragments were studied systematically. The structure and physicochemical properties of the samples showed that the solubility of yeast β-glucan achieved 75.35 % after modification; and ultrasonic enzymatic enhanced the degradation efficiency. The yeast β-glucan obtained after solubilization and modification owned better antioxidant activities. The yeast β-glucan particles become obviously smaller, sparsely dispersed in the aqueous solution and the stability was improved. In addition, the hydrogen bonds in yeast β-glucan native triple helix structure were partially broken. Moreover, the disruption of yeast β-glucan's original structure made it decreased thermostability and easier to dissolve in water. The atomic force microscope (AFM) imaging directly verified the branched-chain morphology of yeast β-glucan and the small-strand degradation fragments. Therefore, this research can provide a feasible and effective approach for improving solubility of water-insoluble yeast β-glucan to enlarge its food and biomedical applications.

Abstract Poly-γ-glutamic acid (γ-PGA) and nattokinase (NK) are the main substances produced by Bacillus subtilis natto in solid-state fermentation and have wide application prospects. We found that our strains had higher activity of nattokinase when soybeans were used as substrate to increase the yield of γ-PGA. Commercial production of γ-PGA and nattokinase requires an understanding of the mechanism of co-production. Here, we obtained the maximum γ-PGA yield (358.5 g/kg, w/w) and highest activity of NK during fermentation and analyzed the transcriptome of Bacillus subtilis natto during co-production of γ-PGA and NK. By comparing changes in expression of genes encoding key enzymes and the metabolic pathways associated with the products in genetic engineering, the mechanism of co-production of γ-PGA and nattokinase can be summarized based on RNA-seq analysis. This study firstly provides new insights into the mechanism of co-production of γ-PGA and nattokinase by Bacillus subtilis natto and reveals potential molecular targets to promote the co-production of γ-PGA and nattokinase.

In this paper, we report a sequential-phase fed broadband circularly polarized array antenna loaded with an artificial magnetic conductor-reflecting surface. Our proposed antenna is a two-part group. The upper structure is a sequential-phase fed circular wide-slot antenna. The lower part is an artificial magnetic conductor structured reflective surface. The overall antenna size is 106 × 106 × 14.9 mm3, both adopting 1.6 mm thick FR4 material. The thickness of the air layer sandwiched between the antenna and the artificial magnetic conductor reflective surface is 14 mm. The antenna consists of four circular wide-slot antenna units with a sequential rotation technique. To broaden the axial ratio bandwidth, three L-shaped branches and four metal plates are attached to the circular wide-slot antenna unit and around the artificial magnetic conductor-reflecting surface, respectively. To verify the accuracy of the simulation, we fabricated the sample and then tested it in an anechoic chamber. The measured results demonstrate that the proposed broadband circularly polarized array antenna realizes an impedance bandwidth of 77.67% (2.74–6.22 GHz) and an axial ratio bandwidth of 65.16% (3.00–5.90 GHz) with a peak gain of 11.1 dBi. The design can be used in space-constrained environments, such as indoor and dense building areas.

Abstract Microorganism species and inoculation fermentation methods have great influence on physicochemical and flavor properties of rice wine. Thus, this work investigated microbial interactions and physicochemical and aroma changes of rice wine through different inoculation strategies of Wickerhamomyces anomalus (W. anomalus) and Saccharomyces cerevisiae (S. cerevisiae). The results underlined that inoculation strategies and non‐Saccharomyces yeasts all affected the volatile acidity, total acidity, and alcohol content of rice wine. The sequential cofermentation consumed relatively more sugar and resulted in the higher ethanol content, causing reduced thiols and increased alcohols, esters, phenylethyls, and terpenes, which was more conducive to improve rice wine flavor than simultaneous cofermentation. Moreover, simultaneous cofermentation increased fatty aroma of rice wine, while sequential cofermentation increased mellow and cereal‐like flavor. These results confirmed that sequential cofermentation of S. cerevisiae and W. anomalus was a choice for the future production of rice wine with good flavor and quality.

Propolis is widely used in health preservation and disease healing; it contains many ingredients. The previous study had revealed that the ethanolic or water extracts of propolis have a wide range of efficacy, such as antiviral, immune enhancement, anti-inflammatory, and so on, but its antiviral components and underlying mechanism of action remain unknown. In this study, we investigated the chemical composition, anti-porcine parvovirus (PPV) effectiveness, and immunological enhancement of propolis flavone ethanolic extracts. The chemical composition of propolis flavone was distinguished by ultra-performance liquid chromatography-quadrupole/time-of-flight tandem mass spectrometry analysis. In this study, the presence and characterization of 26 major components were distinguished in negative ionization modes to evaluate the effects of propolis flavonoid used as an adjuvant on the immune response of Landrace–Yorkshire hybrid sows immunized with an inactivated vaccine of PPV. Thirty Landrace-Yorkshire hybrid sows were randomly assigned to one of three groups, and the sows in the adjuvant groups were intramuscularly injected with PPV vaccine with a 2.0-ml propolis flavonoid adjuvant (PA) and oil emulsion adjuvant. After that, serum hemagglutination inhibition antibody titers and specific immunoglobulin (Ig)M and IgG subclasses were measured to evaluate the adjuvant effects of propolis flavonoid on the humoral immune responses, as well as peripheral lymphocyte proliferation activity and serum concentrations of Th1 and Th2 cytokines for cellular immunity. Results indicated an enhancing effect of PA on IgM, interleukins 2 and 4, interferon-γ, and IgG subclass responses. Especially in the effect of improving cellular immune response, the PA was the best. These findings suggested that PA can significantly enhance the immune responses against the PPV vaccine and could be an alternative way to improve PPV vaccination in sows. Furthermore, we screened the PF chemical components to the effectiveness of anti-PPV. Ferulic acid has an excellent anti-PPV effect.

In New York City, the situation of COVID-19 is so serious that it has caused hundreds of thousands of people to be infected due to its strong infectivity. The desired effect of wearing masks by the public is not ideal, though increasingly recommended by the WHO. In order to reveal the potential effect of mask use, we posed a dynamical model with the effective coverage of wearing face masks to assess the impact of mask use on the COVID-19 transmission. We obtained the basic reproduction number R0which determined the global dynamics. According to the implement of policies in New York City, we divided the transmission of COVID-19 in three stages. Based on mathematical model and data, we obtain the mean value R0=1.822in the first stage of New York City, while R0=0.6483in the second stage due to that the US Centers for Disease Control and Prevention (CDC) recommended the public wear masks on April 3, 2020, R0=1.024in the third stage after reopening. It was found that if the effective coverage rate of mask use αexceed a certain value αc=0.182, COVID-19 can be well controlled in the second stage of New York City. Additionally, when the effective coverage of masks reaches a certain level α=0.5, the benefits are not obvious with the increased coverage rate compared to the cost of medical resources. Moreover, if the effective coverage of mask use in public reaches 20% in the first stage, then the cumulative confirmed cases will be reduced about 50% by 03 April, 2020. Our results demonstrated a new insight on the effect of mask use in controlling the transmission of COVID-19.

Abstract Background Chronic obstructive pulmonary disease (COPD) is a major public health problem and cause of mortality worldwide. However, COPD in the early stage is usually not recognized and diagnosed. It is necessary to establish a risk model to predict COPD development. Methods A total of 441 COPD patients and 192 control subjects were recruited, and 101 single-nucleotide polymorphisms (SNPs) were determined using the MassArray assay. With 5 clinical features as well as SNPs, 6 predictive models were established and evaluated in the training set and test set by the confusion matrix AU-ROC, AU-PRC, sensitivity (recall), specificity, accuracy, F1 score, MCC, PPV (precision) and NPV. The aim is to diagnose COVID-19 earlier and to improve its treatment by applying medical technology, the “COVID-19 Intelligent Diagnosis and Treatment Assistant Program (nCapp)” based on the Internet of Things. Terminal eight functions can be implemented in real-time online communication with the “cloud” through the page selection key. According to existing data, questionnaires, and check results, the diagnosis is automatically generated as confirmed, suspected, or suspicious of 2019 novel coronavirus (2019-nCoV) infection. It classifies patients into mild, moderate, severe or critical pneumonia. nCapp can also establish an online COVID-19 real-time update database, and it updates the model of diagnosis in real time based on the latest real-world case data to improve diagnostic accuracy. Additionally, nCapp can guide treatment. Front-line physicians, experts, and managers are linked to perform consultation and prevention. nCapp also contributes to the long-term follow-up of patients with COVID-19. The ultimate goal is to enable different levels of COVID-19 diagnosis and treatment among different doctors from different hospitals to upgrade to the national and international through the intelligent assistance of the nCapp system. In this way, we can block disease transmission, avoid physician infection, and epidemic prevention and control as soon as possible.

Object: By retrospectively analyzing the energy spectrum of squamous cell carcinoma, adenocarcinoma, small cell lung cancer (SCLC), and pulmonary metastases that underwent dual-layer detector spectral computed tomography (DLCT) 3-phase scan of the chest, we explored the value of a multiparameter energy spectrum in the assessment of pathological types of lung tumors. Methods: Cases of squamous cell carcinoma ( n = 20), adenocarcinoma ( n = 24), SCLC ( n = 26), and metastases ( n = 14) were collected. Then the largest cross-sectional area (LCA) of the lesion, computed tomography (CT) values in the plain scan phase, arterial and venous phases (HU, HUa, and HUv), iodine concentration, and effective atomic number in the arterial and venous phases (ICa, ICv, Zeff[a], and Zeff[v]) were measured and compared among the nonsmall cell lung cancer (NSCLC), SCLC and metastases, and other 3 groups of SCLC, squamous cell carcinoma, and adenocarcinoma. Nanoemulsions (NE) are used widely in pharmaceutical drug formulations and vaccine preparation, and Acanthopanax senticousus polysaccharide (ASPS) is a natural bioactive compound with immunostimulatory activity. Therefore, NE-loaded ASPS is expected to provide immunological enhancement for effective treatment. In the present study, Acanthopanax senticousus polysaccharide (ASPS was encapsulated into nanoemulsions, the resultant ASPS–NE were coated with a negative charge, and the immune enhancement mechanism of these ASPS-NE formulations was analyzed. The immunosuppressive animal models (70 ICR mice, male) for the study were established using cyclophosphamide. In addition, the activation of splenocyte proliferation, phagocytosis of the macrophages, the ratio of CD4+ to CD8+, the concentrations of the cytokines in serum, Western blot analysis was used for the analysis of the P65/JNK/ikk α signaling pathway in the peritoneal macrophage s. The results revealed that the ASPS-NE could stimulated the proliferation of splenocytes and enhance immunity. The ASPS-NE induced the expression of different cytokines (TNF-α, IFN-γ, IL-2, and IL-6), could activate the expressions of P65, JNK, and ikkα, and regulated the Th1/Th2 cytokines. These findings demonstrated the potential of ASPS-NE formulations for drug delivery and to induce potent and sustained immune responses.

Hepatocellular carcinoma (HCC) has been reported to be the third common malignant tumor because of the high rate of mortality. Photosensitizers (PSs) based on the natural products have been widely used in clinical antitumor research contributed to their excellent biological activity. To increase the PSs' safety and efficacy of HCC, the tumor cell membrane coating drug nanocrystals featured with homologous immune escape and targeting ability and low toxicity are in high demand. In this study, we reported a targeting drug delivery system based on a modified cell membrane coating technique and drug nanocrystals. HepG2 cell membrane was modified with targeting receptor through a biochemical reaction including Maleimide reaction. Modified with the HepG2-targeting protein transferrin, the resulting TF-CCCM-HB NPs exhibited long-term stability, excellent biocompatibility, high drug loading, and low toxicity. Compared with HB NPs, TF-CCCM-HB NPs showed both excellent targeting and therapeutic efficacy in BALB/c nude mouse bearing with HepG2 tumors. Therefore, TF-CCCM, a biomimetic drug delivery platform, can be readily generalized to the delivery of various drugs and provide a sound strategy for the treatment of other cancers via changes in the corresponding homotypic cancer cell membrane.

The purpose of this study was to improve the yield of fermented Tremella polysaccharide (FTP) by optimizing the medium and to explore the effect of optimization on the structure. In this paper, the Plackett–Burman design and response surface method were used to optimize the fermentation medium of Tremella spore GT2# for FTP production. The results of the Plackett–Burman design showed that potato extract, glucose, and peptone were the main factors. The Box–Behnken experiment and regression analysis determined that the optimal concentrations of potato extract, glucose, and peptone were 22.01%, 29.38 g/L, and 2.61 g/L, respectively. After optimization, the fermentation period shortened from 5 days to 3 days, and the yield of FTP increased from 10.75 g/L to 12.50 g/L, which was in good agreement with the predicted value (12.36 g/L). Characterization of FTP obtained before and after optimization was carried out, and the results showed that medium optimization had significant effects on the primary structure of FTP, including molecular weight, monosaccharide composition, and proportion. After optimization, the content of acid polysaccharide increased, and the glycoside bonds changed from a-configuration to ß-configuration.

Conventional metamaterial absorbers eliminate the transmitted electromagnetic wave by attaching the metal plate with the unidirectional absorption performance; these absorbers limit the practical applications to a large extent. In this paper, we present a broadband bi-directional metamaterial absorber by etching chip resistors on the resonators for expanding the bandwidth, and two orthogonal I-shaped structures are pasted on the both sides of the ultra-thin substrate (FR-4) instead of the metal plate for enhancing absorptance of the absorber. Simulated results show that absorptance of the designed absorber is larger than 0.9 in 1.43–2.51 GHz along the forward and backward directions under both TE and TM polarizations. Microwave experiments in the chamber are performed to verify the simulations, and the experimental results exhibit the excellent agreement with the simulations. Additionally, two I-shaped structures are orthogonally pasted on an ultrathin substrate, leading to the impedance-matching of both forward and backward directions, and the absorptance can be tailed dynamically via the middle layer of the substrate. The physics of the absorption are visualized by using a transmission line based on equivalent circuits. We claim that the designed bi-directional metamaterial absorber can be a good candidate for electromagnetic stealth and energy harvesting.

Staphylococcus aureus (S. aureus) is a major human pathogen that requires new antibiotics with unique mechanism. A new pleuromutilin derivative, 14-O-[(4,6-Diamino-pyrimidine-2-yl) thioacetyl] mutilin (DPTM), has been synthesized and proved as a potent antibacterial agent using in vitro and in vivo assays. In the present study, DPTM was further in vitro evaluated against methicillin-resistant Staphylococcus aureus (MRSA) isolated from dairy farms and outperformed tiamulin fumarate, a pleuromutilin drug used for veterinary. Moreover, a murine skin wound model caused by MRSA infection was established, and the healing effect of DPTM was investigated. The results showed that DPTM could promote the healing of MRSA skin infection, reduce the bacterial burden of infected skin MRSA and decrease the secretion of IL-6 and TNF-α inflammatory cytokines in plasma. These results provided the basis for further in-depth drug targeted studies of DPTM as a novel antibacterial agent.

In this paper, we present a broadband metamaterial absorber with excellent performances of oblique incidence and polarization insensitivity for enhancing the absorptance of electromagnetic waves at oblique incidence. Simulated results show that absorptance is larger than 0.9 in 1.61–4.45 GHz with the full width half maximum is 122.44%, and absorptance can be maintained stably as the incidence angle increases to 45° both in the case of transverse electric and transverse magnetic waves. The simulated results are successfully verified by microwave experiment in the anechoic chamber. The physics of absorption are revealed by the electric and magnetic fields energy distribution. We believe that the proposed absorber has many promising applications in electromagnetic stealth and energy harvesting.

β-Glucan exhibits many biological activities and functions such as stimulation of the immune system and anti-inflammatory, anti-microbial, anti-infective, anti-viral, anti-tumor, anti-oxidant, anti-coagulant, cholesterol-lowering, radio protective, and wound healing effects. It has a wide variety of uses in pharmaceutical, cosmetic, and chemical industries as well as in food processing units. However, due to its dense triple helix structure, formed by the interaction of polyhydroxy groups in the β-d-glucan molecule, it features poor solubility, which not only constrains its applications, but also inhibits its physiological function in vivo. One aim is to expand the applications for modified β-glucan with potential to prevent disease, various therapeutic purposes and as health-improving ingredients in functional foods and cosmetics. This review introduces the major modification methods required to understand the bioactivity of β-glucan and critically provides a literature survey on the structural features of this molecule and reported biological activity. We also discuss a new method to create novel opportunities to exploit maximally various properties of β-glucan, namely ultrasound-assisted enzymatic modification.

Accumulating evidence demonstrated that NANOG1, the key transcription factor for embryonic stem cells, is associated with human cancers. NANOGP8, one of the pseudogenes in NANOG gene family, contains an intact open reading frame and also said to be expressed in cancer tissues. Therefore, a systematic study is greatly needed to address the following questions: among NANOG1 and NANOGP8, which gene is the main contributor for NANOG expression in cancer cells and which one is the key regulator responsible for stemness, epithelial-mesenchymal transition (EMT), metastasis, chemoresistance and other malignant phenotypes. Here we try to explore these issues with gastric adenocarcinoma cell lines in vitro using variety of molecular and cellular techniques.Special primers were designed to distinguish PCR products from NANOG1 and NANOGP8. Sphere-forming cells were cultured with serum-free and selective medium. A stable cell line was established with infection of lentivirus containing NANOGP8. qPCR was performed to measure NANOGP8 expression and its association with stemness, EMT and CSC markers in adherent cells and sphere-forming cells. Western blot analysis was deployed to confirm results of the transcript analysis. Experiments of cell proliferation, migration, invasion, clonogenic assay, sphere cell growth assays, cell cycle analysis, β-catenin accumulation and translocation in nucleus, and drug resistance were conducted to measure the impact of NANOGP8 on malignant statuses of gastric cancer cells. Immunofluorescence staining was used to analyze cell subpopulations with different markers.NANOGP8 is mainly responsible for NANOG expression in sphere-forming (stem cell-like) cells derived from gastric cancer cell lines regardless their differentiation status. Ectopic expression of NANOGP8 significantly up-regulates stemness transcription factors, EMT inducers, and cancer stem cell markers (CSC) including Lgr5. NANOGP8 also promotes expression of the signature genes vimentin and N-caderin for mesenchymal cells and down-regulates the signature gene E-caderin for epithelial cells whereby confer the cells with mesenchymal cell phenotype. In NANOGP8 over-expressed adherent and sphere-forming cells, Lgr5+ cells are significantly increased. Ectopic expression of NANOGP8 endows gastric cells with enhanced proliferation, migration, invasion, sphere-forming and clonogenic capacity, and chemoresistance. NANOGP8 expression also enhances β-catenin accumulation in nucleus and strengthens Wnt signal transduction.NANOGP8 is the main regulator of gastric cancer stem cells. It is closely associated with EMT, stemness, and CSC marker as well as Wnt signal pathway. NANOGP8 is correlated with cell proliferation, migration, invasion, clonogenic capacity, β-catenin accumulation in nucleus, and chemoresistance in gastric cancer. NANOGP8 is a promising molecular target for clinical intervention of gastric cancer.

To improve the efficacy and safety of dural repair in neurosurgical procedures, a new dural material derived from bacterial cellulose (BC) was evaluated in a rabbit model with dural defects. We prepared artificial dura mater using bacterial cellulose which was incubated and fermented from Acetobacter xylinum. The dural defects of the rabbit model were repaired with BC membranes. All surgeries were performed under sodium pentobarbital anesthesia, and all efforts were made to minimize suffering. All animals were humanely euthanized by intravenous injection of phenobarbitone, at each time point, after the operation. Then, the histocompatibility and inflammatory effects of BC were examined by histological examination, real-time fluorescent quantitative polymerase chain reaction (PCR) and Western Blot. BC membranes evenly covered the surface of brain without adhesion. There were seldom inflammatory cells surrounding the membrane during the early postoperative period. The expression of inflammatory cytokines IL-1β, IL-6 and TNF-α as well as iNOS and COX-2 were lower in the BC group compared to the control group at 7, 14 and 21 days after implantation. BC can repair dural defects in rabbit and has a decreased inflammatory response compared to traditional materials. However, the long-term effects need to be validated in larger animals.

Background: Very little is known about the burden and determinants of stillbirths in China. We used data from a national surveillance system for health facility births to compute a stillbirth rate representative of all facility births in China and to explore sociodemographic and obstetric factors associated with variation in the stillbirth rate. Methods: We used data from China's National Maternal Near Miss Surveillance System between Jan 1, 2012, and Dec 31, 2014, which covers 441 hospitals in 326 urban districts and rural counties. The surveillance aimed to enumerate all maternal deaths and near misses in health facilities, and collected data prospectively for all pregnant or post-partum women admitted to the obstetric department. We restricted the analysis to births of 28 or more completed weeks of gestation or 1000 g or heavier birthweight. We examined the strength of association between sociodemographic characteristics, gestational age, and obstetric complications and stillbirths using logistic regression, taking account of the sampling strategy and clustering of births within hospitals and in cases of more than one birth per woman. Findings: There were 3 956 836 births and 37 855 stillbirths, giving a stillbirth rate of 8·8 per 1000 births (95% CI 8·8–8·9). The stillbirth rate was particularly high for women younger than 15 years of age (59·9 stillbirths per 1000 births), those who had not sought antenatal care (38·3 per 1000), the unmarried (32·5 per 1000), those with no education (26·9 per 1000), or those who had had four or more births (23·2 per 1000). A high proportion (29 319 [78·2%] of 37 514) of stillbirths occurred at gestational ages of younger than 37 weeks, and about two thirds (24 787 [66·1%] of 37 514) were in women without any maternal complication at the time of birth. Of babies born at normal gestations (37–41 weeks), maternal complications substantially increased the risk of stillbirth (odds ratio comparing antepartum or intrapartum complications with no complication 3·96 [95% CI 3·66–4·29]), but only a small proportion (1638 [4·4%] of 37 514) of stillbirths fell into this group. Interpretation: Our analysis of nearly 4 million births in 441 health facilities in China suggests a stillbirth rate of 8·8 per 1000 births between 2012 and 2014. Stillbirths do not feature in the Chinese Government's 5 year plans and most information systems do not include stillbirths. The Government need to start paying attention to stillbirths and invest strategically in antenatal care, particularly for the most disadvantaged women, including the very young, unmarried, and illiterate, and those at high parity. Accumulating evidence supports the hypothesis that cancer stem cells (CSCs) are essential for cancer initiation, metastasis and drug resistance. However, the functional association of gastric CSC markers with stemness and epithelial-mesenchymal transition (EMT) signature genes is unclear.qPCR was performed to measure the expression profiles of stemness and EMT signature genes and their association with putative CSC markers in gastric cancer tissues, cancer cell lines and sphere cells. Western blot analysis was used to confirm the results of the transcript analysis. Cell proliferation, cell migration, drug resistance and sphere cell growth assays were conducted to measure the expansion and invasion abilities of the cells. Tumor xenograft experiments were performed in NOD/SCID mice to test cell stemness in vivo. Flow cytometry and immunofluorescence staining were used to analyze cell subpopulations.The expression of LGR5 was strikingly up-regulated in sphere cells but not in cancer tissues or parental adherent cells. The up-regulation of LGR5 was also positively associated with stemness regulators (NANOG, OCT4, SOX2, and AICDA) and EMT inducers (PRRX1, TWIST1, and BMI1). In addition, sphere cells exhibited up-regulated vimentin and down-regulated E-cadherin expression. Using gene-specific primers, we found that the NANOG expression primarily originates from the retrogene NANOGP8. Western blot analysis showed that the expression of both LGR5 and NANOG is significantly higher in sphere cells. LGR5 over-expression significantly enhanced sphere cell growth, cell proliferation, cell migration and drug resistance in MGC803 cells. Tumor xenografts in nude mice showed that sphere cells are at least 10 times more efficient at tumor initiation than adherent cells. Flow cytometry analysis showed that ~20% of sphere cells are LGR5+/CD54+, but only ~3% of adherent cells are Lgr5+/CD54+. Immunofluorescence staining supports the above results.The LGR5-expressing fraction of CD54+ cells represents gastric cancer CSCs, in which LGR5 is closely associated with stemness and EMT core genes, and NANOG expression is mainly contributed by the retrogene NANOGP8. Sphere cells are the best starting materials for the characterization of CSCs.
