Deep coalbed methane (DCBM), an unconventional gas reservoir, has undergone significant advancements in recent years, sparking a growing interest in assessing pore pressure dynamics within these reservoirs. While some production data analysis techniques have been adapted from conventional oil and gas wells, there remains a gap in the understanding of pore pressure generation and evolution, particularly in wells subjected to large-scale hydraulic fracturing. To address this gap, a novel technique called excess pore pressure analysis (EPPA) has been introduced to the coal seam gas industry for the first time to our knowledge, which employs dual-phase flow principles based on consolidation theory. This technique focuses on the generation and dissipation for excess pore-water pressure (EPWP) and excess pore-gas pressure (EPGP) in stimulated deep coal reservoirs. Equations have been developed respectively and numerical solutions have been provided using the finite element method (FEM). Application of this model to a representative field example reveals that excess pore pressure arises from rapid loading, with overburden weight transferred under undrained condition due to intense hydraulic fracturing, which significantly redistributes the weight-bearing role from the solid coal structure to the injected fluid and liberated gas within artificial pores over a brief timespan. Furthermore, field application indicates that the dissipation of EPWP and EPGP can be actually considered as the process of well production, where methane and water are extracted from deep coalbed methane wells, leading to consolidation for the artificial reservoirs. Moreover, history matching results demonstrate that the excess-pressure model established in this study provides a better explanation for the declining trends observed in both gas and water production curves, compared to conventional practices in coalbed methane reservoir engineering and petroleum engineering. This research not only enhances the understanding of DCBM reservoir behavior but also offers insights applicable to production analysis in other unconventional resources reliant on hydraulic fracturing.

【Objective】Replenishing the power supply system of an underwater communication system is usually very expensive and impractical. The underwater Simultaneous Light-wave Information and Power Transmission (SLIPT) communication systems based on solar cells are powerful solutions. However, the communication bandwidth of the silicon solar cell receiver is very limited, and it is easy to have the phenomenon of deep fading of the Signal-to-Noise Ratio (SNR) due to the underwater light attenuation effect.【Methods】For these problems, this manuscript employs a negative-biased solar cell light receiver scheme that increases the-3 dB bandwidth of silicon solar cells from 420 kHz to 768 kHz. Aiming at the deep fading of SNR caused by various degradation effects in the water environment, a Discrete Fourier Transfor (DFT) extended Orthogonal Frequency Division Multiplexing (OFDM) modulation scheme with a low peak-average power ratio is adopted to offset the deep fading phenomenon in the system.【Results】The performance of DFT Spread OFDM (DFT-S-OFDM) and OFDM modulation systems in water environments with different turbidity (absorption and scattering characteristics) is compared. It is shown that the DFT-S-OFDM modulation and demodulation system is more robust.【Conclusion】Finally, experiments have shown that the total battery power efficiency of the energy harvesting system can be increased by 1.87 times under continuous illumination of white LEDs for 3 h, realizing synchronized energy harvesting in the communication process.

Abstract Background Plant cell walls have evolved precise plasticity in response to environmental stimuli. The plant heterotrimeric G protein complexes could sense and transmit extracellular signals to intracellular signaling systems, and activate a series of downstream responses. dep1 (Dense and Erect Panicles 1), the gain-of-function mutation of DEP1 encoding a G protein γ subunit, confers rice multiple improved agronomic traits. However, the effects of DEP1 on cell wall biosynthesis and wall-related agronomic traits remain largely unknown. Results In this study, we showed that the DEP1 mutation affects cell wall biosynthesis, leading to improved lodging resistance and biomass saccharification. The DEP1 is ubiquitously expressed with a relatively higher expression level in tissues rich in cell walls. The CRISPR/Cas9 editing mutants of DEP1 (dep1-cs) displayed a significant enhancement in stem mechanical properties relative to the wild-type, leading to a substantial improvement in lodging resistance. Cell wall analyses showed that the DEP1 mutation increased the contents of cellulose, hemicelluloses, and pectin, and reduced lignin content and cellulose crystallinity (CrI). Additionally, the dep1-cs seedlings exhibited higher sensitivity to cellulose biosynthesis inhibitors, 2,6-Dichlorobenzonitrile (DCB) and isoxaben, compared with the wild-type, confirming the role of DEP1 in cellulose deposition. Moreover, the DEP1 mutation-mediated alterations of cell walls lead to increased enzymatic saccharification of biomass after the alkali pretreatment. Furthermore, the comparative transcriptome analysis revealed that the DEP1 mutation substantially altered expression of genes involved in carbohydrate metabolism, and cell wall biosynthesis. Conclusions Our findings revealed the roles of DEP1 in cell wall biosynthesis, lodging resistance, and biomass saccharification in rice and suggested genetic modification of DEP1 as a potential strategy to develop energy rice varieties with high lodging resistance.

Abstract Background Malaria is an infectious malady caused by Plasmodium parasites, cerebral malaria standing out as one of its most severe complications. Clinical manifestation include elevated body temperature, loss of consciousness, and seizures. However, reports of cerebral malaria presenting as nonconvulsive status epilepticus are extremely rare. The case presented involves psychiatric symptoms, with the electroencephalogram indicated nonconvulsive status epilepticus associated with cerebral malaria. Case presentation A 53-year-old male, was urgently admitted, due to confusion and abnormal behaviour for 10 h. The patient returned to China after developing a fever while working in Tanzania two months ago. The blood smear revealed Plasmodium vivax and Plasmodium falciparum, and he was diagnosed with malaria. He recovered following anti-malarial treatment. After admission, the patient was confused, unable to communicate normally, and unwilling to cooperate with the physical examination. Plasmodium was not found in the blood smear, but the DNA sequence of P. falciparum was discovered using metagenomic next-generation sequencing of cerebrospinal fluid. Brain MRI revealed no significant abnormalities. Continuous electroencephalogram monitoring revealed that the patient had non-convulsive status epilepticus, which was treated with diazepam and levetiracetam. The patient had normal consciousness and behaviour. He received anti-malarial treatment for two weeks and fully recovered. Conclusions This case demonstrates that nonconvulsive status epilepticus can be a manifestation of cerebral malaria. It is imperative for attending physicians to heighten vigilance when encountering patients with a history of travel to malaria-endemic regions or a prior malaria infection, especially in the presence of unusual clinical presentations.

Abstract Background Nasopharyngeal carcinoma (NPC) is prevalent in southern China. EBV DNA is the most useful biomarker in NPC. However, the value of EBV DNA in posttreatment NPC patients infected with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) remains unclear. Methods Sixty-four eligible NPC patients were enrolled between December 2022 and February 2023. Patients who met the following criteria were included: had non-metastatic NPC, completed radical treatment, were first firstly infected with SARS-CoV-2 and their EBV DNA changed from undetectable to detectable. Results At the end of follow-up, 81.25% (52/64) of patients were confirmed not to relapse with undetectable EBV DNA (no-relapse). In addition, 18.75% (12/64) of patients experienced relapse with consistent detection of EBV DNA (yes-relapse). For all 64 patients, the average time from diagnosis of coronavirus disease 2019 (COVID-19) to detection of detectable EBV DNA was 35.41 days (2 to 139 days). For 52 no-relapse patients, the average time from EBV DNA changing from detectable to undetectable was 63.12 days (6 to 147 days). The levels of EBV DNA were greater in yes-relapse patients than that in no-relapse patients, and the average of EBV DNA levels were 1216 copies/ml and 53.18 copies/ml, respectively. Using 62.3 copies/mL as the threshold, the area under the curve for EBV DNA was 0.88 for distinguishing yes-relapse patients from no-relapse patients. The sensitivity and specificity were 81.97% (95% CI 0.71–0.95) and 86.67% (95% CI 0.70–0.95), respectively. Conclusion For NPC patients infected with SARS-CoV-2, EBV DNA alone is insufficient for monitoring relapse after radical therapy. Long-term follow-up and underlying mechanistic investigations of EBV DNA changes are urgently needed.

Abstract Positron emission tomography/magnetic resonance imaging (PET/MRI) is a hybrid imaging technique that quantitatively combines the metabolic and functional data from positron emission tomography (PET) with anatomical and physiological information from MRI. As PET/MRI technology has advanced, its applications in cancer care have expanded. Recent studies have demonstrated that PET/MRI provides unique advantages in the field of radiotherapy and has become invaluable in guiding precision radiotherapy techniques. This review discusses the rationale and clinical evidence supporting the use of PET/MRI for radiation positioning, target delineation, efficacy evaluation, and patient surveillance. Critical relevance statement This article critically assesses the transformative role of PET/MRI in advancing precision radiotherapy, providing essential insights into improved radiation positioning, target delineation, efficacy evaluation, and patient surveillance in clinical radiology practice. Due to the limited bioavailability of inorganic trace minerals, their utilization in poultry production has led to problems such as environmental contamination and inefficient resource utilization. It was investigated whether replacing inorganic trace minerals (ITM) with a blend of organic small peptide-chelated trace minerals (MIX) would improve production performance, selected biochemical parameters, antioxidant capacity, mineral deposition in liver, heart, and tibia, as well as mineral content in feces of broilers. A total of 432 healthy 21-day-old 817 broilers were randomly divided into 4 groups with 6 replicates per group and 18 chickens per replicate. The control group received a basal diet supplemented with 1,000 mg/kg of inorganic trace minerals as sulfate. The experimental groups received basal diets supplemented with 200, 400, and 600 mg/kg of mixed trace mineral elements (50% sulfate +50% small peptide-chelate) for a trial period of 30 days, divided into two stages: 21–35 days and 36–50 days. The results indicate that on the 50th day, compared with the 1,000 mg/kg ITM group, the levels of serum cholesterol, urea nitrogen, and malondialdehyde in the 200, 400, and 600 mg/kg MIX groups decreased (p 0.05). This study shows that replacing inorganic minerals with low-dose MIX (200, 400, and 600 mg/kg) can reduce the levels of zinc and manganese in feces, with no negative impact on growth and slaughter performance.

BackgroundAlthough Shen Gui capsules (SGCP) are widely used as an adjuvant treatment for chronic heart failure (CHF), their clinical efficacy and safety remain controversial.PurposeTo assess the efficacy and safety of SGCP in the treatment of CHF through a systematic review and meta-analysis, to provide high-quality evidence for evidence-based medicine.MethodsSeven databases were searched for randomized controlled trials (RCTs) assessing SGCP for CHF, from inception to 9 January 2023. RCT quality of evidence was evaluated using the Cochrane Handbook for the Evaluation of Intervention Systems to assess risk of bias and Grading of Recommendations Assessment, Development, and Evaluation. A meta-analysis with subgroup and sensitivity analyses was performed using Review Manager 5.4 and Stata 12.ResultsNine RCTs representing 888 patients with CHF were included in the review. Meta-analysis revealed that SGCP combined with conventional heart failure therapy is more advantageous for improving left ventricular ejection fraction [LVEF; mean difference (MD) = 5.26, 95% confidence interval (CI) (3.78, 6.74), p < 0.0000] and increasing effective rate [relative risk (RR) = 1.21, 95%CI (1.14, 1.29), p < 0.001] compared with conventional therapy alone. The experimental treatment also reduced brain natriuretic peptide [MD = −100.15, 95%CI (−157.83, −42.47), p = 0.0007], left ventricular end-diastolic diameter [MD = −1.93, 95%CI (−3.22, −0.64), p = 0.003], and hypersensitive C-reactive protein [MD = −2.70, 95%CI (−3.12,−2.28), p < 0.001] compared with the control group. However, there was not a statistically significant difference in tumor necrosis factor-α [MD = −14.16, 95%CI (−34.04, 5.73), p = 0.16] or left ventricular end-systolic diameter [MD = −1.56, 95%CI (−3.13, 0.01), p = 0.05]. Nor was there a statistically significant between-groups difference in incidence of adverse events (p > 0.05).ConclusionSGCP combined with conventional heart failure therapy can improve LVEF and increase the effective rate to safely treat patients with CHF. However, further high-quality studies are needed to confirm these findings, due to the overall low quality of evidence in this literature.Clinical Trial Registration:https://www.crd.york.ac.uk/PROSPERO/logout.php, PROSPERO [CRD42023390409].

ObjectiveProbiotics are beneficial to the intestinal barrier, but few studies have investigated probiotics from giant pandas. This study aims to explore the preventive effects of giant panda-derived Clostridium butyricum on dextran sodium sulfate (DSS)-induced colitis in mice.MethodsClostridium butyricum was administered to mice 14 days before administering DSS treatment to induce enteritis.ResultsClostridium butyricum B14 could more effectively prevent colitis in mice than C. butyricum B13. C. butyricum B14 protected the mouse colon by decreasing the histology index and serum interleukin-6 (IL-6) and tumor necrosis factor alpha (TNF-α) levels, which improved intestinal inflammation-related symptoms. In addition, the treatment led to the regulation of the expression of Tifa, Igkv12-89, and Nr1d1, which in turn inhibited immune pathways. The expression of Muc4, Lama3, Cldn4, Cldn3, Ocln, Zo1, Zo2, and Snai is related the intestinal mucosal barrier. 16S sequencing shows that the C. butyricum B14 significantly increased the abundance of certain intestinal probiotics. Overall, C. butyricum B14 exerted a preventive effect on colitis in mice by inhibiting immune responses, enhancing the intestinal barrier and increasing the abundance of probiotic species. Thus, C. butyricum B14 administration helps regulate the balance of the intestinal microecology. It can suppress immune pathways and enhance barrier-protective proteins.

Abstract Lichen symbiosis is centered around a relationship between a fungus and a photosynthetic microbe, usually a green alga. In addition to their main photosynthetic partner (the photobiont), lichen symbioses can contain additional algae present in low abundance. The biology of these algae and the way they interact with the rest of lichen symbionts remains largely unknown. Here we present the first genome sequence of a non-photobiont lichen-associated alga. Coccomyxa viridis was unexpectedly found in 12% of publicly available lichen metagenomes. With few exceptions, members of the Coccomyxa viridis clade occur in lichens as non-photobionts, potentially growing in thalli endophytically. The 45.7 Mbp genome of C. viridis was assembled into 18 near chromosome-level contigs, making it one of the most contiguous genomic assemblies for any lichen-associated algae. Comparing the C. viridis genome to its close relatives revealed the presence of traits associated with the lichen lifestyle. The genome of C. viridis provides a new resource for exploring the evolution of the lichen symbiosis, and how symbiotic lifestyles shaped evolution in green algae.

Immunotherapy has revolutionized cancer treatment, with the anti-PD-1/PD-L1 axis therapy demonstrating significant clinical efficacy across various tumor types. However, it should be noted that this therapy is not universally effective for all PD-L1-positive patients, highlighting the need to expedite research on the second ligand of PD-1, known as Programmed Cell Death Receptor Ligand 2 (PD-L2). As an immune checkpoint molecule, PD-L2 was reported to be associated with patient’s prognosis and plays a pivotal role in cancer cell immune escape. An in-depth understanding of the regulatory process of PD-L2 expression may stratify patients to benefit from anti-PD-1 immunotherapy. Our review focuses on exploring PD-L2 expression in different tumors, its correlation with prognosis, regulatory factors, and the interplay between PD-L2 and tumor treatment, which may provide a notable avenue in developing immune combination therapy and improving the clinical efficacy of anti-PD-1 therapies.

The purpose of this paper is to analyze and research the practical activities of ideological and political education in colleges and universities using causal thinking design. Firstly, it studies the meaning and characteristics of ideological and political education and proposes a causal hypothesis. Secondly, the structural causal model is used to complete the basic structure of the causal thinking model, and the structural causal model is optimized using the HSIC Lasso method. Then, based on the Bayesian generative causal directional algorithm, the functional causal model is constructed, and the model is optimized. Based on the model, the Interaction Network Method for Civic Education Practice in Colleges and Universities is framed. Finally, based on the SCM algorithm, the learning effect of Civics in colleges and universities is analyzed. The Y → X and X → Y causal inference accuracy of the SCM algorithm is 100% and 90%, respectively. More than 70% of the students think that causal thinking exercises help to improve the learning effect of the Civics course.

Abstract Fungal cell walls undergo continual remodeling that generates β-1,3-glucan fragments as products of endo-glycosyl hydrolases (GHs), which can be recognized as pathogen-associated molecular patterns (PAMPs) and trigger plant immune responses. How fungal pathogens suppress those responses is often poorly understood. Here, we study mechanisms underlying the suppression of β-1,3-glucan-triggered plant immunity by the blast fungus Magnaporthe oryzae. We show that an exo-β-1,3-glucanase of the GH17 family, named Ebg1, is important for fungal cell wall integrity and virulence of M. oryzae. Ebg1 can hydrolyze β-1,3-glucan and laminarin into glucose, thus suppressing β-1,3-glucan-triggered plant immunity. However, in addition, Ebg1 seems to act as a PAMP, independent of its hydrolase activity. This Ebg1-induced immunity appears to be dampened by the secretion of an elongation factor 1 alpha protein (EF1α), which interacts and co-localizes with Ebg1 in the apoplast. Future work is needed to understand the mechanisms behind Ebg1-induced immunity and its suppression by EF1α.

To achieve the goals of carbon peaking and carbon neutrality under the backgrounds of poor resource endowments, weak theoretical basis and other factors, the development of the coalbed methane industry of China faces many bottlenecks and challenges. This paper systematically analyzes the coalbed methane resources, key technologies and progress, exploration effect and production performance in China and abroad. The main problems are summarized as low exploration degree, low technical adaptability, low return on investment and small development scale. This study suggests that the coalbed methane industry in China should follow the “two-step” (short-term and long-term) development strategy. The short-term action before 2030, can be divided into two stages: (1) From the present to 2025, to achieve new breakthroughs in theory and technology, and accomplish the target of annual production of 10 billion cubic meters; (2) From 2025 to 2030, to form the technologies suitable for most geological conditions, further expand the industry scale, and achieve an annual output of 30 billion cubic meters, improving the proportion of coalbed methane in the total natural gas production. The long-term action after 2030 is to gradually realize an annual production of 100 billion cubic meters. The strategic countermeasure to achieve the above goals is to adhere to “technology+management dual wheel drive”, realize the synchronous progress of technology and management, and promote the high-quality development of the coalbed methane industry. Technically, the efforts will focus on fine and effective development of coalbed methane in the medium to shallow layers of mature fields, effective development of coalbed methane in new fields, extensive and beneficial development of deep coalbed methane, three-dimensional comingled development of coalbed methane, applying new technologies such as coalbed methane displacement by carbon dioxide, microwave heating and stimulation technology, ultrasonic stimulation, high-temperature heat injection stimulation, rock breaking by high-energy laser. In terms of management, the efforts will focus on coordinative innovation of resource, technology, talent, policy and investment, with technological innovation as the core, to realize an all-round and integrated management and promote the development of coalbed methane industry at a high level.

Ticks and tick-borne diseases have negative impacts on the health of wild animals including endangered and vulnerable species. The giant panda (Ailuropoda melanoleuca), a vulnerable and iconic flagship species, is threatened by tick infestation as well. Not only can ticks cause anemia and immunosuppression in the giant panda, but also bacterial and viral diseases. However, previous studies regarding tick infestation on giant pandas were limited in scope as case reports from sick or dead animals. In this study, an investigation focusing on the tick infestation of a reintroduced giant panda at the Daxiangling Reintroduction Base in Sichuan, China was conducted. Ticks were routinely collected and identified from the ears of the giant panda from March to September in 2021. A linear model was used to test the correlation between tick abundance and climate factors. All ticks were identified as Ixodes ovatus. Tick abundance was significantly different among months. Results from the linear model showed temperature positively correlated to tick abundance, while air pressure had a negative correlation with tick abundance. To the best of our knowledge, this study is the first reported investigation of tick species and abundance on a healthy giant panda living in the natural environment, and provides important information for the conservation of giant pandas and other species sharing the same habitat.

AbstractPurpose: To assess the impact of neoadjuvant chemotherapy (NACT) on the tumor immune microenvironment (TIME) in gynaecological tumors, with a focus on understanding the potential for enhanced combination therapies.Methods: We systematically queried the PubMed, Embase, and Cochrane databases, encompassing reviews, clinical trials, and case studies, to undertake a thorough analysis of the impact of NACT on the TIME of gynaecological tumors.Results: NACT induces diverse immune microenvironment changes in gynaecological tumors. In cervical cancer, NACT boosts immune-promoting cells, enhancing tumor clearance. Ovarian cancer studies yield variable outcomes, influenced by patient-specific factors and treatment regimens. Limited research exists on NACT’s impact on endometrial cancer’s immune microenvironment, warranting further exploration. In summary, NACT-induced immune microenvironment changes display variability. Clinical trials highlight personalized immunotherapy’s positive impact on gynaecological tumor prognosis, suggesting potential avenues for future cancer treatments. However, rigorous investigation is needed to determine the exact efficacy and safety of combining NACT with immunotherapy.Conclusion: This review provides a solid foundation for the development of late-stage immunotherapy and highlights the importance of therapeutic strategies targeting immune cells in TIME in anti-tumor therapy.

Abstract Background Prior drought stress may change plants response patterns and subsequently increase their tolerance to the same condition, which can be referred to as “drought memory” and proved essential for plants well-being. However, the mechanism of transcriptional drought memory in psammophytes remains unclear. Agriophyllum squarrosum, a pioneer species on mobile dunes, is widely spread in Northern China’s vast desert areas with outstanding ability of water use efficiency. Here we conducted dehydration-rehydration treatment on A. squarrosum semi-arid land ecotype AEX and arid land ecotype WW to dissect the drought memory mechanism of A. squarrosum, and to determine the discrepancy in drought memory of two contrasting ecotypes that had long adapted to water heterogeneity. Result Physiological traits monitoring unveiled the stronger ability and longer duration in drought memory of WW than that of AEX. A total of 1,642 and 1,339 drought memory genes (DMGs) were identified in ecotype AEX and WW, respectively. Furthermore, shared DMGs among A. squarrosum and the previously studied species depicted that drought memory commonalities in higher plants embraced pathways like primary and secondary metabolisms; while drought memory characteristics in A. squarrosum were mainly related to response to heat, high light intensity, hydrogen peroxide, and dehydration, which might be due to local adaptation to desert circumstances. Heat shock proteins (HSPs) occupied the center of the protein-protein interaction (PPI) network in drought memory transcription factors (TF), thus playing a key regulatory role in A. squarrosum drought memory. Co-expression analysis of drought memory TFs and DMGs uncovered a novel regulating module, whereby pairs of TFs might function as molecular switches in regulating DMG transforming between high and low expression levels, thus promoting drought memory reset. Conclusion Based on the co-expression analysis, protein-protein interaction prediction, and drought memory metabolic network construction, a novel regulatory module of transcriptional drought memory in A. squarrosum was hypothesized here, whereby recurrent drought signal is activated by primary TF switches, then amplified by secondary amplifiers, and thus regulates downstream complicated metabolic networks. The present research provided valuable molecular resources on plants’ stress-resistance basis and shed light on drought memory in A. squarrosum.

With regard to China's vigorous promotion of environmental governance and ecological restoration, farmers around mines are victims of environmental damage and major participants in ecological restoration. Their willingness to participate is related to whether the government's environmentalpolicies can play a good role. This study developed a structural equation model of the impact of environmental perception on pro-environmental behavior willingness and conducted an empirical study using survey data of farmers around the Dabaoshan tailings area in Shaoguan in 2020. Value perception includes "economic value" and "ecology value", which was measured by the two observable variables "increasing income" and "improving the surrounding environmental quality". The results showed that value perception had a positive impact on farmers' willingness to be environmentally friendly, and that the standardization coefficient of the "increase in income" variable was higher, which meant that if farmers felt that environmental governance in the tailings area could bring them economic benefits or improve the ecological environment, they were more inclined to participate in environmental governance activities in tailings areas, and local farmers paid more attention to economic interests. Endowment perception represented farmers' self-efficacy, which was measured by "energy perception" and "ability perception". The above two variables strongly influenced farmers' willingness to be pro-environmental. The results showed that the higher the self-efficacy evaluation of farmers, the stronger their willingness to participate in environmental governance of the tailings area. Policy perceptions including exemplary norms and command-line norms were measured by "government attitudes", "attitudes of miner" and "attitudes of neighbor and friend". The results showed that policy perception had a direct impact on farmers' willingness to pro-environmental behavior, but this was due to an indirect impact by affecting farmers' value perception and policy perception, and "mining enterprise attitude" was the policy perception where the important variables were explained. Therefore, improving farmers' value perception and endowment perception through policy perception could also indirectly improve their willingness to exhibit pro-environmental behavior. Based on the above results, this study puts forward relevant policy implications. First, traditional media, newspapers, and TV should combine with emerging online platforms such as Douyin and Kuaishou to expand publicity channels. As rational economic people, farmers naturally attach great importance to the value brought about by participating in environmental governance in tailing areas. Therefore, it is necessary to publicize the economic and ecological value of environmental governance in the tailings area in a simple and easy-to-understand way and to make the publicity more targeted. Second, it is necessary to strengthen employment guidance and vocational education, improve the technical literacy of local farmers, lower the threshold and cost for farmers to participate in environmental governance, and encourage them to make scientific decisions. Last but not the least, it is necessary to build an ecological restoration exchange platform, strengthen the demonstrative role of neighborhoods, break the information barriers between the government and farmers, and explore effective leading models.

【Objective】The chicken fibroblast cell line (DF-1) of stably expressing Cas9 protein was screened, and combined with the reporter vector system based on single strand annealing (SSA) repair mechanism, the Cas9 nuclease activity in the established DF-1 cell line was detected.【Method】The lentiviral vector plasmid carrying Cas9 protein was transfected into 293T cells packaging lentivirus together with the helper plasmid. DF-1 cells were infected with lentivirus Cas9 supernatant, and DF-1 cells with stable Cas9 protein expression were obtained through antibiotic screening. The expression of Cas9 protein in positive screened DF-1 cells was verified by polymerase chain reaction (PCR) and Western blot. The sgRNA sequence of chicken ovalbumin (OVA) gene was selected and and the annealing products were cloned into pYP152 to construct sgRNA expression vector. Primers were designed at both ends of the sgRNA target site to amplify the target fragment of OVA gene. The target fragment was cloned into mCherry-SSA reporter vector pCMV-SSA-mCherry-Hind Ⅲ to destroy the expression of mCherry protein. Then the sgRNA expression vector and mCherry-SSA reporter vector were co-transfected into DF-1 cells of stably expressing Cas9 protein. Finally, the repair of mCherry expression by different stable transformants was analyzed by fluorescence microscope.【Result】27 DF-1 cell lines of stably expressing Cas9 protein were obtained after antibiotic screening, cell genome was stably transfected by PCR amplification, and the results showed that these cells contained Cas9 protein sequences. The results of Western blot assay showed that all the stable transfection cell lines expressed Cas9 protein; after co-transformation of sgRNA expression vector and mCherry-SSA reporter vector, fluorescence microscope observation showed that all the selected stable transfection cell lines could restore the expression of mCherry protein in the report vector.【Conclusion】The DF-1 cell line of stably expressing Cas9 protein with cleavage activity was successfully established, which could provide basic materials for the subsequent research on chicken functional genes on the DF-1 cell line of stably expressing Cas9 protein.

Abstract Purpose To evaluate the anticancer activities of lenvatinib in ICC and its possible molecular mechanisms. Methods Patients-derived xenograft (PDX) model and cell line-derived xenograft (CDX) model were both used for the in vivo study. For in vivo work, ICC cell lines were applied to analyze the effect of Lenvatinib on cell proliferation, cell cycle progression, apoptosis, and the molecular mechanism. Results In the present study, we found that lenvatinib dramatically hindered in vivo tumor growth in ICC patient-derived xenograft models. In addition, by using in vitro experiments in ICC cell lines, we found that lenvatinib dose- and time-dependently inhibited the proliferation of ICC cells and induced cell cycle arrest in the G0/G1 phase. Transcriptional profiling analysis further applied indicated that lenvatinib might inhibit cell proliferation through the induction of cell-cycle arrestment via activating of Gadd45a, it was evidenced by that the knockout of Gadd45a significantly attenuated the cycle arrest induced by lenvatinib, as well as the inhibitory effect of lenvatinib on ICC. Conclusion Our work first found that lenvatinib exerted an excellent antitumor effect on ICC, mainly via inducing Gadd45a-mediated cell cycle arrest. Our work provides evidence and a rationale for the future use of lenvatinib in the treatment of ICC.

Five new biflorane-type diterpenoids, biofloranates E–I (1–5), and two new bicyclic diterpene glycosides, lemnaboursides H–I (6–7), along with the known lemnabourside, were isolated from the South China Sea soft coral Lemnalia bournei. Their chemical structures and stereochemistry were determined based on extensive spectroscopic methods, including time-dependent density functional theory (TDDFT) ECD calculations, as well as a comparison of them with the reported values. The antibacterial activities of the isolated compounds were evaluated against five pathogenic bacteria, and all of these diterpenes and diterpene glycosides showed antibacterial activities against Staphylococcus aureus and Bacillus subtilis, with MICs ranging from 4 to 64 µg/mL. In addition, these compounds did not exhibit noticeable cytotoxicities on A549, Hela, and HepG2 cancer cell lines, at 20 μM.

The petroleum and natural gas industries exhibit a high dependency on lifting equipment for oil and gas. Any malfunction in these devices can lead to severe economic losses. Therefore, continuous and timely monitoring of the status of pumping equipment is of paramount importance to proactively prevent potential issues. In an effort to enhance this monitoring process, this study delves into multi-source data images at the well site and extends traditional information analysis methods. It introduces an improved YOLOv7 method based on switchable atrous convolution. While the YOLOv7 algorithm achieves a balance between speed and accuracy, its robustness in non-standard environments is suboptimal. To address this limitation, we propose the utilization of a switchable atrous convolution method for enhancement, thereby augmenting the adaptability of the model. Images of pumping units from diverse scenarios are actively collected and utilized to construct training, validation, and test sets. Different models, including YOLOv7SAC, YOLOv7, and YOLOv5-n, undergo testing, and their detection performances are systematically compared in complex environments. Experimental findings demonstrate that YOLOv7SAC consistently attains optimal detection results across various scenes. In conclusion, the study suggests that the combination of the YOLOv7 model with switchable atrous convolution proves effective for detecting pumping unit equipment in complex scenarios. This provides robust theoretical support for the detection and identification of pumping equipment issues under challenging conditions.

The surface quality of components fabricated via laser-based powder bed fusion (L-PBF) is highly dependent on the post-machining technique. This study aims to investigate the microstructure evolution of the turning-affected subsurface layer of the IN625 superalloy manufactured by L-PBF with the assistance of the finite element analysis (FEA). A finite element model with integrated user-defined subroutine VUSDFLD was created for numerical modeling of the dislocation density and grain size evolution due to turning operation. The simulation results regarding grain size and the depth of the affected layer were validated against the experiments. This study shed some light on the metallurgical behavior evolution when turning of L-PBF of nickel-based superalloy based on the proposed material microstructure model.

ABSTRACT: A total of 480 one-day-old male yellow-feathered broilers were randomly divided into 4 groups with 6 replicates of 20 chicks per replicate. A basal diet was administered to the control group (CON), whereas CML350, CML500, and CML1000 groups were fed with basal diet supplemented with 350, 500, and 1,000 mg/kg of lauric acid monoglyceride and cinnamaldehyde complex, respectively. However, adding 500 mg/kg of lauric acid monoglyceride and cinnamaldehyde complex improved weight gain (P < 0.01), enhanced intestinal morphology, increased serum total protein and albumin content, and total antioxidant capacity (P < 0.01), and significantly increased the Chao1 and Ace indices (P < 0.01), indicating an increase in the richness of the gut microbiota. At the phylum level, CML500 group reduced the abundance of Fusobacteriota at 21 d and Proteobacteria at 42 d (P < 0.01). At the genus level, CML500 group increased the abundance of Faecalibacterium and Alistipes at 42 d (P < 0.01) and decreased the abundance of Escherichia–Shigella (P < 0.01). At the species level, CML500 group reduced the abundance of Escherichia coli at 42 d (P < 0.01) and increased the abundance of Alistipes_sp_CHKCI003 at 42 d (P < 0.01). According to these results, adding 500 mg/kg of lauric acid monoglyceride and cinnamaldehyde complex in feed can improve the growth performance, intestinal morphology, and gut microbiota of yellow-feathered broilers.

Toxoplasma gondii is a worldwide-distributed zoonotic protozoan parasite which causes toxoplasmosis and has a significant effect on public health. In the giant panda (Ailuropoda melanoleuca), toxoplasmosis can cause asymptomatic infections, reproductive disorder and even death, which poses a serious threat to the conservation of this rare protected species. Therefore, serological investigation of T. gondii is essential to understanding its risk to giant pandas, however, there are no specific testing kits for giant pandas. Previous research has used MAT as the reference method for screening T. gondii, to investigate this further, this study focused on the agreement comparing of MAT with ELISA and IHA tests for detecting T. gondii antibodies in 100 blood samples from 55 captive giant pandas in Chengdu, China. The results showed 87.0%, 87.0%, 84.0%, samples were sero-positive for T. gondii using ELISA (kits a, b, c), respectively, while MAT and IHA tests were 84.0% and 9.0% sero-positive, respectively. BACKGROUND The analysis of selenium enrichment characteristics in various crops planted in a certain area and investigation and evaluation of selenium content in selenium fertilizer crops can provide a basis for the development of selenium-enriched characteristic agriculture. It is important to find out the influencing factors of selenium content in different crops for developing selenium-enriched agricultural products. OBJECTIVES To understand the distribution and enrichment characteristics of Se in soil and crops in Luoyang City. METHODS The selenium resources detailed investigation project area and other agricultural planting areas in Luoyang City, Henan Province were chosen as the research object. The selenium content of 22 kinds of crops and their root soil were measured by ICP-MS, and the selenium enrichment characteristics of different crops were studied. RESULTS The average value of soil selenium in the study area was close to the standard of selenium-enriched soil in Henan Province. The selenium-enriched sites in acidic soil, neutral soil, alkaline soil, and overall soil accounted for 28.61%, 43.72%, 41.99%, and 40.56%, respectively. According to the evaluation of selenium enrichment coefficient of various crops, the study area was suitable for the development of selenium-enriched beans, oil-bearing crops, wheat, millet, sweet potato and other characteristic crops and selenium-enriched traditional Chinese medicine Stachys floridana Schuttl.ex Benth, but not suitable for the development of selenium-enriched corn. The correlation of selenium content in different crops with soil selenium content and pH showed that soil selenium content was the decisive factor of selenium content in crops, followed by soil pH. CONCLUSIONS In order to promote the development of selenium-enriched agricultural products in the study area, selenium fertilizer can be applied to improve the selenium content of crops. Crops planted in low selenium background soil, especially fruits, are suitable for applying selenium fertilizer, while millet planted in high selenium background soil is not suitable for applying selenium fertilizer.

Abstract Background The composition and diversity of root microbial community are affected by plant genotypes and soil environment, which in turn affect plant growth and development. Grafting rootstock types of the apple tree can affect phenotypes in cultivation practice, but it is not clear whether grafting rootstock types can affect the composition and diversity of root microbial community and the resistance of apple tree to apple Valsa canker. Methods To explore root microbial differences and the correlation, 16S rRNA and ITS genes were sequenced using Novaseq technology. Results The results showed that the influence of grafting rootstock types on the composition of the root fungal community was greater than that of bacteria. And the bacterial community richness was higher in the healthy (OTUs: 1693) and dwarfing rootstock (OTUs: 1526) than in the disease (OTUs: 1181) and standard rootstock (OTUs: 1412), while the fungal community richness was the opposite. Moreover, the bacterial abundance of root zone, rhizosphere, and root endophytic microorganisms with the same grafting rootstock type exhibited a decreasing trend. Results of Nested PCR assay on soil and root tissue of Valsa mali showed that the content of V. mali in dwarfing rootstocks are lower than standard rootstocks. These results suggest that apple trees grafting with dwarfing rootstocks are more resistant to V. mali than standard rootstocks. Conclusions Under different grafting types, the effect on the composition of fungal community in apple tree root was greater than that of bacteria. The bacterial community in dwarfing rootstocks is more abundant and diverse, including more beneficial microorganisms. Therefore, dwarfing rootstock is more conducive to the resistance to apple Valsa canker from biological control.

Abstract Background Extended-spectrum β-lactamases (ESBL)-producing strains of Klebsiella pneumoniae remain a worldwide, critical clinical concern. However, limited information was available concerning ESBL-producing Klebsiella pneumoniae in giant pandas. The objective of this study was to characterize ESBL-producing Klebsiella pneumoniae isolates from captive giant pandas. A total of 211 Klebsiella pneumoniae isolates were collected from 108 giant pandas housed at the Chengdu Research Base of Giant Panda Breeding (CRBGP), China. Samples were screened for the ESBL-producing phenotype via the double-disk synergy test. Result A total of three (1.42%, n = 3/211) ESBL-producing Klebsiella pneumoniae strains were identified, and characterization of ESBL-producing Klebsiella pneumoniae isolates were studied by the detection of ESBL genes and mobile genetic elements (MGEs), evaluation of antimicrobial susceptibility and detection of associated resistance genes. Clonal analysis was performed by multi-locus sequencing type (MLST). Among the three ESBL-producing isolates, different ESBL-encoding genes, including bla CTX-M, and bla TEM, were detected. These three isolates were found to carry MGEs genes (i.e., IS903 and tnpU) and antimicrobial resistance genes (i.e., aac(6')-Ib, aac(6')-I, qnrA, and qnrB). Furthermore, it was found that the three isolates were not hypermucoviscosity, resistant to at least 13 antibiotics and belonged to different ST types (ST37, ST290, and ST2640). Conclusion Effective surveillance and strict infection control strategies should be implemented to prevent outbreaks of ESBL-producing Klebsiella pneumoniae in giant pandas.

ABSTRACT: This study aimed to assess the effect of inbreeding on production traits using a long-term closed-line population recorded for residual feed intake (RFI). The study first used data from a previously reported population to determine the appropriate period of divergent selection for RFI. The results showed that RFI had similar moderate heritability estimates (0.28–0.34) during the fast-growing period (7–12 wk), and RFI at 7 to 10 wk had the highest heritability (0.34). Therefore, divergent selection was performed in a Chinese broiler population for RFI at 7 to 10 wk; the total sample size from generations zero (G0) to 13 was 9050. The divergence between the 2 lines increased steadily throughout generations, resulting in G13 with average RFI values of 304.55 in high RFI (HRFI) males, −160.31 in low RFI (LRFI) males, 296.30 in HRFI females and −157.55 in LRFI females. The feed intake (FI) and feed conversion ratio were almost higher in HRFI broilers than in LRFI broilers, and the magnitude of the difference in FI increased from approximately 4% for both sexes in G1 to approximately 33% in G13. Body weight gain was irregular from G1 to G13 and higher in LRFI broilers than in HRFI broilers after G10. Indeed, the HRFI broilers consumed more food, but they were lighter than LRFI broilers. In G13, LRFI males had heavier slaughter weight, longer cecum length, more white blood cells (WBC), red blood cells (RBC) and hemoglobin (HGB), but triglycerides, lower dressed percentage, percentage of half eviscerated yield, and eviscerated yield than HRFI males. LRFI females had a higher percentage of breast muscle and gizzard yield, longer cecum length, and more WBCs, RBCs and HGB but less abdominal fat and serum total cholesterol than HRFI females. This study was the first to verify that long-term divergent selection for RFI in Chinese broiler chickens is positive and beneficial.

ABSTRACT: The use of antibiotics leads to antibiotic residues in livestock and poultry products, adversely affecting human health. Ciprofloxacin (CFX) is a broad-spectrum antibiotic shared between animals and humans that is useful in treatments besides infections. However, changes in the gut microbiota caused by CFX and the possible link with the elimination of CFX residues have not been investigated. Herein, we used the Silkie chicken model to study the changes in the gut microbiota during the entire CFX-metabolic repertoire. We detected CFX residues in different tissues and showed that the elimination time of CFX from different tissues was dissimilar (liver > kidney > chest muscle > skin). Analysis of liver and kidney injury biomarkers and plasma antioxidant indices indicated slight hepatotoxicity and nephrotoxicity in the Silkie chickens. Importantly, the changes in the gut microbial community predominantly occurred early in the metabolic process. Correlation analysis revealed that the particular bacterial microbiota were associated with the pharmacokinetics of CFX in different Silkie chicken tissues (e.g., aerobic bacteria, including Escherichia and Coprococcus, and anaerobic bacteria, including Fusobacterium, Ruminococcus, Bifidobacterium, and Eubacterium). Collectively, certain microbiota may boost antibiotic metabolism and participate in restoring the microbial consortia after CFX is metabolized. Therefore, regulating the core intestinal microbiota may reduce foodborne antibiotics and accelerate the development of drug resistance.

Abstract Background The red panda has been classified as an endangered species due to the decreased number in the world and disease is considered as a great threat to the health and survival of the cubs in captivity. Results This study analyzed 32 red panda cub mortalities (15 females and 17 males, age less than two months) through gross necropsy, microbiological examination, and histopathological observation at the Chengdu Research Base of Giant Panda Breeding, China, during 2014–2020. The results showed that screenings for canine distemper virus, canine parvovirus, rotavirus and parasite infection were all negative, however bacteria such as Klebsiella pneumoniae, Proteus mirabilis, Escherichia coli, Enterococcus faecalis, Pseudomonas were isolated from the tissue samples of some cubs. The major causes of death were respiratory (43.75%) and digestive system disease (28.13%), followed by cardiovascular disease (12.5%) and neonatal stillbirths (9.38%). Renal system diseases and trauma were also detected, at lower incidence (one case for each). The mortality rate within 15 days of birth was 68.75% and gradually decreased with age, there was no significant difference in gender. Conclusion This study can provide a scientific basis for the analysis of the cause of death among red panda cubs in captivity, so as to improve the survival rate, help build the captive population and further the ex-situ conservation management of this endangered species. Additionally, our research may also provide insights into the in-situ conservation of wild red pandas by identifying emerging disease threats within the wild population and potential treatment for rescued individuals.

The objective of this study was to understand biological characteristics of one bacteria strain named as VPG which was isolated from multiple organs of a dead captive giant panda cub. Here, we use biochemical tests, 16S rRNA and gyrB genes for bacterial identification, the disk diffusion method for antibiotic resistance phenotype, smart chip real-time PCR for the antibiotic resistance genotype, multiplex PCR for determination of virulence genes, and the acute toxicity test in mice for testing the pathogenicity of isolates. The isolate was identified as A. veronii strain based on the biochemical properties and genetic analysis. We found that the strain carried 31 antibiotic resistance genes, revealed antimicrobial resistance phenotypically to several antibiotics including penicillin, ampicillin, oxacillin, amoxicillin, imipenem, and vancomycin, and carried virulence genes including aer, act, lip, exu, ser, luxs, and tapA. The main pathological changes in giant panda were congestion, necrotic lesions and a large number of bacteria in multiple organs. In addition, the LD50 in Kunming mice infected with strain VGP was 5.14 × 107 CFU/mL by intraperitoneal injection. Infection with strain VGP led to considerable histological lesions such as hemorrhage of internal organs, necrosis of lymphocytes and neurons in Kunming mice. Taken together, these results suggest that infection with strain VGP would be an important causes of death in this giant panda cub.

Crop plants have coevolved phytohormone-mediated defenses to combat and/or repel their colonizers. The present study determined the effects of jasmonic acid (JA) accumulation during aphid infestation on the preference and performance of Sitobion miscanthi Takahashi (Hemiptera: Aphididae), and its potential role in fine-tuning hormone-dependent responses in XN979 wheat cultivar seedlings was evaluated via the transcriptional profiles of marker genes related to JA- and salicylic acid (SA)-dependent responses. The preference experiment and the life table data reveal that direct foliage spraying of 2.5 mM methyl jasmonate (MeJA) exhibited weak negative or positive effects on the preferential selection and the population dynamics and oviposition parameters of S. miscanthi. The transcription level of phytohormone biosynthesis genes shows that foliage spraying of MeJA significantly upregulated the marker genes in the JA biosynthesis pathway while downregulating the SA pathway. In addition, either MeJA treatment or previous aphid infestation significantly induced upregulated transcription of the genes involved in the JA- and SA-dependent defense responses, and the transcription level of the tryptophan decarboxylase (TaTDC) gene, which facilitates the conversion of L-tryptophan to tryptamine, was rapidly upregulated after the treatments as well. The main products of tryptamine conversion could play a crucial role in suppressing SA-dependent defense responses. These results will provide more experimental evidence to enable understanding of the antagonistic interaction between hormone signaling processes in cereals under aphid infestation.

BACKGROUND Molybdenum is an essential and beneficial element for humans and crops and has an anti-cancer effect. The content of molybdenum and the pH of the soils are different in different regions, the content of molybdenum in crops varies greatly. At the same time, different crops have a different absorption ability of molybdenum. Studying the rules of molybdenum enrichment in different crops provides a basis for the development of healthy geology, the development of molybdenum-rich agricultural products, the development of functional agriculture, and the adjustment of planting structure. OBJECTIVES To investigate Mo content in different crops and their controlling factors. METHODS This study used the detailed survey area of selenium resources in Luoyang City and other agricultural planting areas as the research object. By collecting 22 kinds of crops and their root soils, inductively coupled plasma-mass spectrometry (ICP-MS) was used to determine the molybdenum content in soils and crops, and the characteristics and influencing factors of the content of molybdenum in different crops were studied. RESULTS The content of molybdenum in the soil of Luoyang City was relatively high, which was the characteristic area of molybdenum-rich soil in China. Mung bean, cowpea, black bean, yellow bean, red bean and peanuts were the main molybdenum enrichment crops, with average >9mg/kg molybdenum content and enrichment factor >500%, which belonged to the molybdenum hyperaccumulator. Sesame, long bean, millet, wheat, corns and rapeseed had high molybdenum content, which ranged from 0.446mg/kg to 2.437mg/kg, and the enrichment coefficient ranged from 40% to 300%, which were molybdenum rich crops. The content of molybdenum in chili, garlic, sweet potatoes and okra ranged from 0.1mg/kg to 0.3mg/kg, and the enrichment factor ranged from 10% to 30%, which were high-molybdenum crops. The content of molybdenum in apple, pear, grape, pomegranate, cherry and Stachys floridana Schuttl.ex Benth was less than 0.05mg/kg, and the enrichment factor was less than 5%, which was a low-molybdenum crop. The content of molybdenum in most crops was positively correlated with that in root soils, while the content of molybdenum in apple, grape, pomegranate, and cherry was negatively correlated within the molybdenum content in the root soils. CONCLUSIONS Studies have shown that molybdenum in soils is more easily absorbed by crops under an alkaline environment. Compared with other areas in China, the crop in the area is richer in molybdenum, so it is a favorable area for the development of a molybdenum-rich agricultural industry. According to the molybdenum content of different crops, mung bean, cowpea, black bean, yellow bean, red bean and peanuts were selected as the characteristic Mo-rich agricultural products in the study area; sesame, long bean, millet, wheat, corns and rapeseed were Mo-rich agricultural products; pepper, garlic, sweet potato and okra were high Mo agricultural products. The results provide a scientific basis for the development and adjustment of the molybdenum-rich agricultural product planting structure in the study area.

Abstract This article analyzes the self-enterprising mechanism of research & development (R&D) engineers based on a case study of labor control in a high-tech company. The concept of “self as enterprise” refers to the form of labor control by allowing workers to self-manage according to the market principle. It is neither a form of normative control built upon accepting and internalizing market-oriented values nor responsible autonomy aiming to enhance workers' organizational commitment. R&D engineers' reproduction of current living standards depends heavily on performance-based salaries, which forces them to pragmatically comply with market rules. When both the reproduction of labor and the labor market have been overmarketized, R&D engineers are at high risk of downgrading their living standards and facing double job insecurity. Therefore, they must manage their work and life in an enterprising way to maximize labor value and prevent it from being devalued.

IntroductionToxoplasma gondii, a globally zoonotic protozoan parasite, infects most warm-blooded animals including the giant panda, and poses a serious threat to the giant panda conservation. However, the seroprevalence and the risk factors for toxoplasmosis in giant pandas are unknown. Here we aimed to determine the seroprevalence of T. gondii in the captive population of giant pandas and analyze the factors associated with the increased risk of infection.MethodsA total of 203 serum samples were collected from 157 (95 females and 62 males) captive giant pandas from 2007 to 2022, antibodies against T. gondii were screened using commercial ELISA and MAT kits.ResultsThe results showed 56 (35.67%) giant pandas were seropositive, age and transfer history between institutions were identifified as risk factors for T. gondii infection. It is suggested that age-related seroprevalence was the main factor, and housing multiple species in the same environment may increase the chance of cross-infection of T. gondii.DiscussionThis study can provide research data for developing policies for the prevention and control of T. gondii and protecting the health of captive giant pandas and other wildlife.

ObjectivesEvidence from systematic reviews/meta-analyses about the efficacy and safety of Qishen Yiqi (QSYQ) dripping pills in chronic heart failure (CHF) remains unclear. This study comprehensively reviewed available systematic reviews on latest evidence to provide reliable information for the clinical use of QSYQ in CHF.MethodsThe systematic review was performed on studies retrieved from six major medical databases. Eligible studies were evaluated in terms of methodological quality and quality of evidence using the Assessment of Multiple Systematic review 2 (AMSTAR-2) tool, the Risk of Bias in Systematic Reviews (ROBIS) was used to assess the risk of bias, and the Preferred Reporting Items for Systematic Review and Meta-analyses (PRISMA) 2020 was utilized for assessing reporting quality. In addition, the Grading of Recommendations Assessment, Development, and Evaluation (GRADE) was used to determine primary outcome indicators’ evidence quality.ResultsA total of 14 systematic reviews were included in this study, based on which it could be concluded that QSYQ combined with conventional medicine (CM) treatment tended to be superior to CM treatment alone in terms of improving cardiac function-related indices (e.g., increasing the left ventricular ejection fraction [LVEF] and reducing the left ventricular end-diastolic dimension [LVEDD] and left ventricular end-systolic internal diameter [LVESD]), improving the total effective rate and 6-min walking distance (6MWD), and reducing N-terminal pro-brain natriuretic peptide (NT-proBNP). Overall, no serious QSYQ-related adverse events were observed. However, the GRADE results showed “very low” to “moderate” evidence for these outcomes, with no high-quality evidence supporting them. Unsatisfactory results were obtained in terms of methodological quality, risk of bias and reporting quality after assessment using the AMSTAR-2, ROBIS, and PRISMA 2020, limited mainly by deficiencies in the following areas: registration of study protocols, explanation of the inclusion of randomized controlled trials (RCTs), complete and detailed search strategy, list of excluded literature, description of funding sources for inclusion in RCTs, investigation of the impact of risk of bias on the results of meta-analysis, and reporting of potential conflicts of interest.ConclusionThe efficacy and safety of QSYQ adjuvant therapy in CHF remain to be further clarified due to the lack of high-quality evidence provided by current systematic reviews.

Due to the low bio-availability of inorganic trace minerals, its application in poultry production has been causing many problems such as environment pollution and waste of resources. The current study was designed to evaluate if replacing inorganic trace minerals (ITM) with small peptide chelate trace minerals (SPM) affects production performance, some biochemical parameters and antioxidant status, tibia mineral deposition, and fecal mineral content in 817 white-feathered broilers. A total of 432 broilers (21-day-old) were randomly divided into four groups with six replicates of 18 chicks each. The four groups included inorganic trace minerals group (addition of 1,000 mg/kg ITM; common practice by commercial poultry farms), three organic trace minerals groups with supplementation of 150, 300, and 500 mg/kg SPM, respectively. The experiment lasted for 30 days. The results showed that there was no significant difference in growth performance and slaughter performance among the four groups (p > 0.05). Total cholesterol in the SPM group was significantly lower than those in the ITM groups (p < 0.01). Compared with the ITM group, the serum urea nitrogen in 150 and 300 mg/kg SPM groups decreased significantly (p < 0.01). Among all SPM treatments, 300 mg/kg SPM groups had the highest serum glutathione peroxidase (GSH-Px) activity (p < 0.01). The activity of copper and zinc superoxide dismutase (Cu/Zn SOD) of liver in ITM group was the lowest among the four groups (p < 0.01). The catalase (CAT) activity of liver in the 150 mg/kg SPM group was significantly higher than the ITM group and 300 mg/kg SPM group (p < 0.05). Compared to the ITM group, the iron content of the tibia was significantly increased in 300 mg/kg SPM group (p < 0.05) and 500 mg/kg SPM group (p < 0.01). Compared to the ITM group, dietary supplementation with SPM significantly reduced fecal content of zinc and manganese (p < 0.01). The 150 mg/kg SPM and 300 mg/kg SPM group had significantly reduced content of iron (p < 0.05). This study demonstrated that replacing inorganic minerals with low doses of SPM (300 and 500 mg/kg) did not negatively affect growth and slaughter performance, as well as the antioxidant status of broiler chickens. In addition, SPM can also promote mineral content in the tibia and reduce mineral content in the feces.

Characterising the spreading of ideas within echo chambers is essential for understanding polarisation. In this article, we explore the characteristics of popular and viral content in climate change discussions on Twitter around the 2019 announcement of the Nobel Peace Prize, where we find the retweet network of users to be polarised into two well-separated groups of activists and sceptics. Operationalising popularity as the number of retweets and virality as the spreading probability inferred using an independent cascade model, we find that the viral themes echo and differ from the popular themes in interesting ways. Most importantly, we find that the most viral themes in the two groups reflect different types of bonds that tie the community together, yet both function to enhance ingroup connections while repulsing outgroup engagement. With this, our study sheds light, from an information-spreading perspective, on the formation and upkeep of echo chambers in climate discussions.

Agriophyllum squarrosum (L.) Moq., well known as sandrice, is an important wild forage in sandy areas and a promising edible and medicinal resource plant with great domestication potential. Previous studies showed flavonoids are one of the most abundant medicinal ingredients in sandrice, whereby isorhamnetin and isorhamnetin-3-glycoside were the top two flavonols with multiple health benefits. However, the molecular regulatory mechanisms of flavonoids in sandrice remain largely unclear. Based on a common garden trial, in this study, an integrated transcriptomic and flavonoids-targeted metabolomic analysis was performed on the vegetative and reproductive periods of six sandrice ecotypes, whose original habitats covered a variety of environmental factor gradients. Multiple linear stepwise regression analysis unveiled that flavonoid accumulation in sandrice was positively correlated with temperature and UVB and negatively affected by precipitation and sunshine duration, respectively. Weighted co-expression network analysis (WGCNA) indicated the bHLH and MYB transcription factor (TF) families might play key roles in sandrice flavonoid biosynthesis regulation. A total of 22,778 differentially expressed genes (DEGs) were identified between ecotype DL and ecotype AEX, the two extremes in most environmental factors, whereby 85 DEGs could be related to known flavonoid biosynthesis pathway. A sandrice flavonoid biosynthesis network embracing the detected 23 flavonoids in this research was constructed. Gene families Plant flavonoid O-methyltransferase (AsPFOMT) and UDP-glucuronosyltransferase (AsUGT78D2) were identified and characterized on the transcriptional level and believed to be synthases of isorhamnetin and isorhamnetin-3-glycoside in sandrice, respectively. A trade-off between biosynthesis of rutin and isorhamnetin was found in the DL ecotype, which might be due to the metabolic flux redirection when facing environmental changes. This research provides valuable information for understanding flavonoid biosynthesis in sandrice at the molecular level and laid the foundation for precise development and utilization of this functional resource forage.
