1. Structure and immunostimulating activity of polysaccharides derived from the roots and leaves of dandelion

Abstract

Abstract Two polysaccharides were obtained from dandelion roots (DPR) and dandelion leaves (DPL) via water extraction and ethanol precipitation. Both DPR and DPL were an acid heteropolysaccharide, with a molecular weight of 5.89 × 104 and 28.2 × 104 Da, respectively. Monosaccharide composition results showed that both DPR and DPL contained Man, Glc, Rha, GalA, Glc, Gal, and Ara with a molar ratios of 2.98:1.00:3.67:6.41:2.98:16.70:2.48 and 1.23:1.00:1.06:5.74:2.88:8.59:1.84, respectively. Methylation and NMR analysis showed DPR and DPL, with triple-helix conformations, were mainly composed of 4-ɑ-Galp and 4-ɑ-GalAp, terminated with t-ɑ-Araf, t-ɑ-Rhap, t-ɑ-Glcp and t-ɑ-Manp. DPL showed an immunoprotective effect in cyclophosphamide (Cy)-induced black-bone silky chickens by improving chicken growth performance, increasing the spleen, thymus, and bursa of Fabricius indices, and promoting blood lymphocyte proliferation, the secretion of cytokines (IL-2, IL-6, and INF-γ) and serum immunoglobulin (IgA, IgG, and IgM) levels in a dose-dependent manner. Moreover, the oxidative stress damage in immunosuppressed chickens was significantly reformed after DPL treatment. These findings provide useful information on the potential for application of dandelion polysaccharides as natural nutrients to enhance chicken immune and antioxidant functions. Graphical Abstract

Abstract

Dryland rearing on netting floors (DRNF) is a new rearing method for ducks, which could prevent duck excreta from polluting water bodies. However, the influence of DRNF on duck production and immune performance remains poorly understood. In this study, 2,280 Shaoxing ducks, an egg-type breed of Sheldrake in China, were chosen and randomly divided into 2 groups to investigate the effects of DRNF on duck farming. During the experimental laying rates, feed-egg ratios, and mortality rates of the 2 groups were calculated and recorded. Serum immune parameters, including thymus index, spleen index, levels of immunoglobulin G (IgG), interleukin 1β (IL-1β), and tumor necrosis factor α (TNF-α), were determined. Sequencing of the 16S rRNA gene was used to analyze the variability of gut microbiota in the duck ileum and cecum. The results showed that DRNF significantly reduced the mortality rate of the ducks and increased the thymus index (P0.05), compared to the control. No other significant differences were detected in productional and immune indices (P0.05). The 16S rRNA sequencing results revealed differentially enriched microbial compositions in the ileum and cecum, which might be responsible for the improved immune function of Shaoxing ducks. For example, an increase in Lactobacillaceae (family), Anaerotruncus (genus), Saccharibacteria (phylum), Flavobacteriaceae (family), and a reduction in Anaerobiospirillum (genus), Lachnospiraceae (family), Blautia (genus) was revealed in the DRNF ducks. In conclusion, DRNF could alter gut microflora, enhance duck immune system, and reduce mortality in Shaoxing ducks.

Brucellosis is a zoonotic animal disease causing a serious obstacle to public health, social and economic progress, food security and food safety. The present study was to investigate the prevalence of Brucella infection in goats in Hubei province, China. A total 2007serum samples were obtained from 6 counties in Hubei province in 2014 and 2015 and were tested for antibodies to Brucella by serum agglutination test. The results showed that the seroprevalence of Brucella in goats was 0.25% (95% CI= 0.1-0.6). In different genders, the prevalence were 0.21% (95% CI= 0-0.8) and 0.28% (95% CI= 0.1-0.8) in male and female goats, respectively. In different seasons, the prevalence ranged from 0 to 0.38% with the distribution of that 0 (95 CI: 0.0, 0.1) in spring; 0.35% (95 CI: 0.1, 0.3) in summer; 0.38% (95 CI: 0.1, 0.6) in autumn and 0 (95 CI: 0.0, 1.3) in winter, respectively. The present study herein indicated the existence of Brucella infection in goats in Hubei province in serology for the first time.
