From accumulation mechanisms and the identification of effective faults, hydrocarbon distribution in slope areas of petroliferous basins is assessed. In this study, we superimpose two methods to develop a set of key methods, which can identify effective faults for hydrocarbon accumulation in slope areas. The first of this set of methods includes locating the distribution area of the connected sand bodies and paleotectonic ridges overlying the formation where the sand bodies are located, which aids determining the lateral migration path of hydrocarbons in slope areas. The second set of methods includes identifying the lateral sealing parts of faults that are oriented perpendicular or oblique to the channel sand bodies, thereby determining the faults that are effective for hydrocarbon accumulation. By applying these methods, we identify the effective faults in the first member of the Nantun Formation (K1n1) on the Wunan slope of the Wuerxun Depression in the Hailar Basin. The results show that the effective faults in K1n1 are located mainly in its middle and northern parts, with a few distributed in the south-central part. This is conducive to reservoir development, since hydrocarbons which were generated in the western trough source rocks of K1n1, accumulated in the nearby trap formed from a suitable configuration of the fault and sand body of K1n1. This is consistent with previous exploration data, and the observation that hydrocarbons discovered in K1n1 on the Wunan slope are mainly distributed in the middle and northern parts of the region, and a small amount occur in the south-central part. The results show that this set of methods is feasible for determining effective faults for hydrocarbon accumulation in slope areas.

Abstract Background Widely used restriction-dependent cloning methods are labour-intensive and time-consuming, while several types of ligase-independent cloning approaches have inherent limitations. A rapid and reliable method of cloning native DNA sequences into desired plasmids are highly desired. Results This paper introduces ABI-REC, a novel strategy combining asymmetric bridge PCR with intramolecular homologous recombination in bacteria for native DNA cloning. ABI-REC was developed to precisely clone inserts into defined location in a directional manner within recipient plasmids. It featured an asymmetric 3-primer PCR performed in a single tube that could robustly amplify a chimeric insert-plasmid DNA sequence with homologous arms at both ends. Intramolecular homologous recombination occurred to the chimera when it was transformed into E.coli and produced the desired recombinant plasmids with high efficiency and fidelity. It is rapid, and does not involve any operational nucleotides. We proved the reliability of ABI-REC using a double-resistance reporter assay, and investigated the effects of homology and insert length upon its efficiency. We found that 15 bp homology was sufficient to initiate recombination, while 25 bp homology had the highest cloning efficiency. Inserts up to 4 kb in size could be cloned by this method. The utility and advantages of ABI-REC were demonstrated through a series of pig myostatin (MSTN) promoter and terminator reporter plasmids, whose transcriptional activity was assessed in mammalian cells. We finally used ABI-REC to construct a pig MSTN promoter-terminator cassette reporter and showed that it could work coordinately to express EGFP. Conclusions ABI-REC has the following advantages: (i) rapid and highly efficient; (ii) native DNA cloning without introduction of extra bases; (iii) restriction-free; (iv) easy positioning of directional and site-specific recombination owing to formulated primer design. ABI-REC is a novel approach to DNA engineering and gene functional analysis.

This article underscores the need for future research to explore three-dimensional applications and consider user age, with potential expansions into medical and sports domains. The final particle moisture content of ceramic slurry spray drying affects the processing, product quality, and process energy consumption. While the real-time moisture content determination is very difficult for spray drying process due to nonlinear, severe lag, interference factors, and complexity of the systems, the high temperature, high humidity drying environment and short drying process. In current work, an improved moth optimization (IMFO) algorithm combined with backpropagation neural network (BPNN) was proposed to predict the moisture content of ceramic slurry particles during spray drying. The performance of the model is trained and tested. Results demonstrate that IMFO has better convergence ability and speed compared to other algorithms such as particle swarm optimization (PSO) and gravitational search algorithm (GSA). The IMFO-BPNN model achieves a MAE of 0.0293, RMSE of 0.0383, and R2 value of 0.9113, outperforming the prediction performance of BPNN and MFO-BPNN models. The absolute error rate of the IMFO-BPNN model (0–5%) is lower compared to BPNN (5–10%) and MFO-BPNN model (>10%), showcasing superior accuracy in predicting moisture content. This mathematical model established in the study provides an efficient, accurate, and nondestructive method for predicting the drying endpoint of ceramic slurry spray drying. [ABSTRACT FROM AUTHOR]

This study aimed to improve apple slices' color and drying kinetics by optimizing the hot-air drying process, utilizing machine and deep learning models. Different steam blanching times (30, 60, 90, and 120 s), drying temperatures (50, 55, 60, 65, and 70 °C), and humidity control methods (full humidity removal or temperature–humidity control) were examined. These factors significantly affected the quality of apple slices. 60 s blanching, 60 °C temperature, and full dehumidification represented the optimal drying conditions for apple slices' dehydration, achieving better drying kinetics and the best color quality. However, the fastest drying process (40 min) was obtained at a 60 °C drying temperature combined with complete dehumidification after 90 s blanching. Furthermore, machine and deep learning models, including backpropagation (BP), convolutional neural network–long short-term memory (CNN-LSTM), temporal convolutional network (TCN), and long short-term memory (LSTM) networks, effectively predicted the moisture content and color variation in apple slices. Among these, LSTM networks demonstrated exceptional predictive performance with an R2 value exceeding 0.98, indicating superior accuracy. This study provides a scientific foundation for optimizing the drying process of apple slices and illustrates the potential application of deep learning in the agricultural processing and engineering fields. In this study, a tunnel-type continuous multi-physical field drying equipment was developed to combine microwave, hot air, and infrared drying technologies. The microwave source arrangement and hot air equalizing plate were optimized based on numerical simulation to enhance the uniformity of the microwave field and hot air velocity distribution. Mode 4 and opening ratio 37.70% (r = 60 mm) were had the highest electric field distribution and hot air distribution. This equipment was further used to pre-dehydrate cowpeas and the effects of microwave, hot air, and infrared on food drying properties and quality of dried samples were investigated. The results showed that the moisture content was reduced by 20% and the ΔE value greater than 5 after pre-dehydration. Microwave treatment rapidly reduced the moisture content of cowpeas. The infrared treatment significantly improved the uniformity of temperature distribution and the texture profile of the dried sample. The hot air drying increased 10 °C for the overall temperature of the food material and balanced the internal water content of the food material. This study compared four drying methods, namely hot-air and vacuum combined drying (HAVCD), hot-air drying (HAD), vacuum drying (VD), and vacuum freeze-drying (VFD), with the aim to select a high-quality and efficient drying method to dry American ginseng slices. Drying kinetics and various physicochemical properties, such as color, rehydration ratio, total ginsenoside content, total ginsenoside residual rate, hardness, and microstructure were explored. An improved multi-indicator test formula method was used to score the experimental results quantitatively. HAVCD resulted in comprehensive advantages, with the highest product score for the dried American ginseng slices. Therefore, HAVCD was subjected to a response surface methodology (RSM) analysis and process optimization. The interaction of hot-air temperature and vacuum temperature on the comprehensive score of American ginseng drying was highly significant (p < 0.01). The optimized and validated process parameters obtained were a hot-air temperature of 57 °C, a vacuum temperature of 54 °C, and a moisture content at the conversion point of 39% (wet basis). Under these conditions, the best drying time was 170 min; the total ginsenoside content was 2.3 mg/100 g; the L* value was 91.68; the rehydration ratio was 3.10; and the comprehensive score was 96.77. The findings of this work indicated HAVCD as a promising drying method for American ginseng slices, considering the drying time and dried-product quality attributes. [ABSTRACT FROM AUTHOR]
