Abstract Cadmium (Cd) is a biologically non-essential heavy metal, a major soil pollutant, and extremely harmful to plants. The phytohormone methyl jasmonate (MeJA) plays an important role in plant heavy-metal resistance. However, the understanding of the effects of MeJA supply level on alleviating Cd toxicity in plants is limited. Here, we investigated how MeJA regulated the development of physiological processes and cell wall modification in Cosmos bipinnatus. We found that low concentrations of MeJA increased the dry weight of seedlings under 120 µM Cd stress by reducing the transport of Cd from roots to shoots. Moreover, a threshold concentration of exogenous MeJA increased the activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) in plant roots, the concentration of Cd in the root cell wall, and the contents of pectin and hemicellulose 1 polysaccharides, through converting Cd into pectin-bound forms. These results suggested that MeJA mitigated Cd toxicity by modulating root cell wall polysaccharide and functional group composition, especially through pectin polysaccharides binding to Cd, with effects on Cd transport capacity, specific chemical forms of Cd, and homeostatic antioxidant systems in C. bipinnatus.

Plant lead (Pb) tolerance and accumulation are key characteristics affecting phytoremediation efficiency. Bermudagrass is an excellent candidate for the remediation of Pb-polluted soil, and it needs to be mowed regularly. Here, we explored the effect of different mowing frequencies on the remediation of Pb-contaminated soil using bermudagrass. Mowing was found to decrease the biomass and photosynthetic efficiency of bermudagrass under Pb stress, thereby inhibiting its growth. Although mowing exacerbated membrane peroxidation, successive mowing treatments alleviated peroxidation damage by regulating enzymatic and nonenzymatic systems. A comprehensive evaluation of Pb tolerance revealed that all the mowing treatments reduced the Pb tolerance of bermudagrass, and a once-per-month mowing frequency had a less negative effect on Pb tolerance than did more frequent mowing. In terms of Pb enrichment, mowing significantly increased the Pb concentration, total Pb accumulation, translocation factor (TF), and bioenrichment factor (BCF) of bermudagrass. The total Pb accumulation was greatest under the once-a-month treatment, while the TF and BCF values were greatest under the three-times-a-month mowing treatment. Additionally, the decrease in soil pH and DOC were significantly correlated with the soil available Pb content and plant Pb accumulation parameters. The results showed that changes in the rhizosphere are crucial factors regulating Pb uptake in bermudagrass during mowing. Overall, once-a-month mowing minimally affects Pb tolerance and maximizes Pb accumulation, making it the optimal mowing frequency for soil Pb remediation by bermudagrass. This study provides a novel approach for the remediation of Pb-contaminated soil with bermudagrass based on mowing.

Abstract The complexity of the tumor microenvironment (TME) is a crucial factor in lung adenocarcinoma (LUAD) progression. To gain deeper insights into molecular mechanisms of LUAD, we perform an integrative single-cell RNA sequencing (scRNA-seq) data analysis of 377,574 cells from 117 LUAD patient samples. By linking scRNA-seq data with bulk gene expression data, we identify a cluster of prognostic-related UPP1high tumor cells. These cells, primarily situated at the invasive front of tumors, display a stronger association with the immunosuppressive components in the TME. Our cytokine array analysis reveals that the upregulation of UPP1 in tumor cells leads to the increased release of various immunosuppressive cytokines, with TGF-β1 being particularly prominent. Furthermore, this UPP1 upregulation also elevates the expression of PD-L1 through the PI3K/AKT/mTOR pathway, which contributes to the suppression of CD8 + T cells. Cytometry by time-of-flight (CyTOF) analysis provides additional evidence of the role of UPP1 in shaping the immunosuppressive nature of the TME. Using patient-derived organoids (PDOs), we discover that UPP1high tumors exhibit relatively increased sensitivity to Bosutinib and Dasatinib. Collectively, our study highlights the immunosuppressive role of UPP1 in LUAD, and these findings may provide insights into the molecular features of LUAD and facilitate the development of personalized treatment strategies.

Lodging reduces maize yield and quality. The improvement in maize lodging resistance has proven to be instrumental in maximizing the yield potential of maize varieties under high-density planting. Tillage practices accommodate larger groups by enhancing soil conditions. This study aimed to elucidate the impact of subsoil tillage in reducing the maize stalk lodging rate. The maize cultivars Xianyu 335 (XY335) and Zhongdan2 (ZD2) were selected for field experiments including two tillage methods, shallow rotary (RT) and subsoil (SS), and two densities, 75,000 plants ha−1 (D1) and 105,000 plants ha−1 (D2), were set up to investigate and analyze the changes of maize lodging rate and the related indexes of lodging resistance under SS and RT conditions. The findings revealed that under high density, as compared to rotary tillage, SS tillage decreased the plant and ear height by 9.01–9.20 cm and 3.50–4.90 cm, respectively. The stalk dry matter accumulation was enhanced by 8.98%–24.98%, while stalk diameter between two and seven internodes increased by 0.47– 4.15 mm. Stalk cellulose increased by 11.83% –12.38%, hemicellulose increased by 6.7%–15.97%, and lignin increased by 9.86%–15.9%. The rind puncture and crushing strength improved by 3.11%–20.06% and 11.90%–27.07%, respectively. The bending strength increased by 6.25%–27.96% and the lodging rate decreased by 1.20%–6.04%. Yield increased by 7.58%–8.17%. At SS tillage when density increased, the index changes in ZD2 were mostly less than those in XY335. The rind penetration strength, bending strength, crushing strength, stalk diameter, and dry matter accumulation all had a negative correlation with the lodging rate. It suggested that SS tillage was beneficial to lodging resistance and, in combination with stalk lodging-resistant varieties, can effectively alleviate the problem of stalk lodging after increased planting density.

Abstract Background Drought stress can substantially restrict maize growth and productivity, and global warming and an increasing frequency of extreme weather events are likely to result in more yield losses in the future. Therefore, unraveling the molecular mechanism underlying the response to drought stress is essential for breeding drought-resilient crops. Results In this study, we subjected the 3-leaf-period plants of two maize inbred lines, a drought-tolerant line (si287) and a drought-sensitive line (X178), to drought stress for seven days while growing in a chamber. Subsequently, we measured physiological traits and analyzed transcriptomic and metabolic profiles of two inbred lines. Our KEGG analysis of genes and metabolites revealed significant differences in pathways related to glycolysis/gluconeogenesis, flavonoid biosynthesis, starch and sucrose metabolism, and biosynthesis of amino acids. Additionally, our joint analysis identified proline, tryptophan and phenylalanine are crucial amino acids for maize response to drought stress. Furthermore, we concentrated on tryptophan (Trp), which was found to enhance tolerance via IAA-ABA signaling, as well as SA and nicotinamide adenine dinucleotide (NAD) consequent reactive oxygen species (ROS) scavenging. We identified three hub genes in tryptophan biosynthesis, indole-3-acetaldehyde oxidase (ZmAO1, 542,228), catalase 1 (ZmCAT1, 542,369), and flavin-containing monooxygenase 6 (ZmYUC6, 103,629,142), High expression of these genes plays a significant role in regulating drought tolerance. Two metabolites related to tryptophan biosynthesis, quinolinic acid, and kynurenine improved maize tolerance to drought stress by scavenging reactive oxygen species. Conclusions This study illuminates the mechanisms underlying the response of maize seedlings to drought stress. Especially, it identifies novel candidate genes and metabolites, enriching our understanding of the role of tryptophan in drought stress. The identification of distinct resistance mechanisms in maize inbred lines will facilitate the exploration of maize germplasm and the breeding of drought-resilient hybrids.

Abstract Due to the ongoing global warming, maize production worldwide is expected to be heavily inflicted by droughts. The grain yield of maize hybrids is an important factor in evaluating their suitability and stability. In this study, we utilized the AMMI model and GGE biplot to analyze grain yield of 20 hybrids from the three tested environments in Inner Mongolia in 2018 and 2019, aiming at selecting drought-tolerant maize hybrids. AMMI variance analysis revealed highly significant difference on main effects for genotype, environment, and their interaction. Furthermore, G11 (DK159) and G15 (JKY3308) exhibited favorable productivity and stability across all three test environments. Moreover, G10 (LH1) emerged as the most stable hybrid according to the AMMI analysis and the GGE biplot. Bayannur demonstrated the highest identification ability among the three tested sites. Our study provides accurate identification for drought-resilient maize hybrids in different rain-fed regions. These findings can contribute to the selection of appropriate hybrids that exhibit productivity, stability, and adaptability in drought-prone conditions.

Corn straw is an important renewable resource, which could improve the quality of saline–alkali cultivated land. However, the slow decomposition of crop residues in cold, arid, and saline–alkali soils can lead to serious resource waste and ecological crises. The use of beneficial microorganisms with degradation functions could solve these problems. In this study, three types of saline–alkali soil with low, medium, and high salinity levels were used in the straw-returning experiment. The experiment was conducted with four treatments: GF2 (Klebsiella sp.), GF7 (Pseudomonas sp.), GF2+GF7, and CK (control without bacteria). Microbial characteristics, straw degradation efficiency, element release rate, and soil factors were compared, and random forest linear regression and partial least squares path modeling analysis methods were utilized. The results indicated that the degradation of bacterial metabolites, the efficiency of corn stover degradation, the efficiency of component degradation, and the release rates of elements (C, N, P, and K) initially increased and then decreased with the increase in salinity. At the maximum value of moderately saline–alkali soil, the effect of GF2+GF7 treatment was significantly better than that of other treatments (p < 0.05). Given the interactive effects of saline–alkali soil and microbial factors, the application of exogenous degrading bacteria could significantly increase soil enzyme activity and soil available nutrients, as well as regulate the salt–alkali ion balance in soil. The cation exchange capacity (9.13%, p < 0.01) was the primary driving force for the degradation rate of straw in saline–alkali soil with different degrees of salinization under the influence of exogenous degrading bacteria. Straw decomposition directly affected the soil chemical properties and indirectly affected soil enzyme activity. The results of this study would provide new strategies and insights into the utilization of microbial resources to promote straw degradation.

Compound pollution with cadmium (Cd) and zinc (Zn) is common in nature. The effects of compounded Cd and Zn on the growth and development of Iris pseudacorus in the environment and the plant’s potential to remediate heavy metals in the environment remain unclear. In this study, the effects of single and combined Cd and Zn stress on I. pseudacorus growth and the enrichment of heavy metals in I. pseudacorus seedlings were investigated. The results showed that under Cd (160 μM) and Zn (800 μM) stress, plant growth was significantly inhibited and photosynthetic performance was affected. Cd+Zn200 (160 μM + 200 μM) reduced the levels of malondialdehyde, hydrogen peroxide, and non-protein thiols by 31.29%, 53.20%, and 13.29%, respectively, in the aboveground tissues compared with levels in the single Cd treatment. However, Cd+Zn800 (160 μM + 800 μM) had no effect. Cd and Zn800 inhibited the absorption of mineral elements, while Zn200 had little effect on plants. Compared with that for Cd treatment alone, Cd + Zn200 and Cd+Zn800 reduced the Cd content in aboveground tissues by 54.15% and 49.92%, respectively, but had no significant effect on Cd in the root system. Zn significantly reduced the Cd content in subcellular components and limited the content and proportion of Cd extracted using water and ethanol. These results suggest that a low supply of Zn reduces Cd accumulation in aboveground tissues by promoting antioxidant substances and heavy metal chelating agents, thus protecting the photosynthetic systems. The addition of Zn also reduced the mobility and bioavailability of Cd to alleviate its toxicity in I. pseudacorus.

Maize leaf diseases significantly impact yield and quality. However, recognizing these diseases from images taken in natural environments is challenging due to complex backgrounds and high similarity of disease spots between classes.This study proposes a lightweight multi-level attention fusion network (LFMNet) which can identify maize leaf diseases with high similarity in natural environment. The main components of LFMNet are PMFFM and MAttion blocks, with three key improvements relative to existing essential blocks. First, it improves the adaptability to the change of maize leaf disease scale through the dense connection of partial convolution with different expansion rates and reduces the parameters at the same time. The second improvement is that it replaces a adaptable pooling kernel according to the size of the input feature map on the original PPA, and the convolution layer to reshape to enhance the feature extraction of maize leaves under complex background. The third improvement is that it replaces different pooling kernels to obtain features of different scales based on GMDC and generate feature weighting matrix to enhance important regional features. Experimental results show that the accuracy of the LFMNet model on the test dataset reaches 94.12%, which is better than the existing heavyweight networks, such as ResNet50 and Inception v3, and lightweight networks such as DenseNet 121,MobileNet(V3-large) and ShuffleNet V2. The number of parameters is only 0.88m, which is better than the current mainstream lightweight network. It is also effective to identify the disease types with similar disease spots in leaves.

Indocalamus plants are low-growing shrubby bamboos with growth advantages, such as high biomass and strong resistance, and they are rich in germplasm resources in southern China. This study conducted soil lead (Pb) stress experiments on Indocalamus latifolius (Keng) McClure (LA), Indocalamus hunanensis B.M. Yang (HU), Indocalamus chishuiensis Y.L. Yang and Hsueh (CH) and Indocalamus lacunosus Wen (LC). Five Pb treatments (0, 500, 1000, 1500 mg·kg−1 Pb, and 1000 mg·kg−1 Pb + 1000 mg·kg−1 ethylenediamine tetraacetic acid (EDTA)) were established. EDTA was applied to explore the tolerance mechanism of different Indocalamus species after absorbing large amounts of heavy metals. The results were as follows: (1) under Pb treatment, the total relative biomass of LA, HU and LC was 100%; (2) after applying EDTA, the bioconcentration coefficient, translocation factor, and free proline content of the four Indocalamus species increased; and (3) the Pb mobility and distribution rates of the underground parts of the four Indocalamus species were consistently greater than those of the aboveground parts. The Pb mobility and distribution rates in the stems increased after applying EDTA, while those in the leaves decreased, as the plants tended to transfer Pb to their stems, which have lower physiological activity than their leaves.

Bone is a major tissue for uranium deposition in human body. Considering mesenchymal stem cells (MSCs) play a vital role in bone formation and injury recovery, studying the mechanism of MSCs responding to uranium poisoning can benefit the understanding of bone damage and repair after uranium exposure. Cellular structural alterations were analyzed via transmission electron microscopy (TEM). Changes in cellular behaviors were assessed through cellular viability, apoptosis, and the production of DNA double-strand breaks (DSBs). In addition, the influence of gap junctional intercellular communication (GJIC) on uranium toxicity was assessed. The disruption of MSCs was elevated with the increase in uranyl nitrate concentration, as shown by TEM micrograph. This was verified by the results of cellular viability and DSB production. Interestingly, the results of apoptosis assay indicated significant apoptosis occurred, which was accompanied with an obvious disruption of cellular membranes. Furthermore, closely contacted cell confluence groups exhibited resistant to uranium poisoning in contrast to sparse growth groups, which can be eliminated with the pretreatment of a GJIC inhibitor in the close connection group. To verify the association between GJIC and cytotoxic effects of uranyl nitrate, GJIC function was evaluated by wound healing and cellular migration. The results showed an inhibition of the healing ratio and migration ability induced by the exposure of uranyl nitrate. The low transfer efficiency of the dye coupling experiment and depressed expression of gap functional protein connexins confirmed the impairment of GJIC function. These results suggest that uranium toxicity is involved with GJIC dysfunction.

To achieve high maize (Zea mays L.) yields and quality grain, it is necessary to develop stress-resistant cultivars and related cultivation practices, aiming to maximize efficiency. Thus, our objectives were (i) to investigate the impact of tillage practices and maize hybrids (which have improved over time) on yield and its components, and (ii) to characterize the response pattern of maize hybrid grain nutrient quality components to subsoiling. To achieve this, we conducted field trials with five maize hybrids from different eras under two tillage practices: rotary tillage and subsoiling. We compared grain yield, nutritional quality, and other indicators across different tillage conditions from the 1970s to the 2010s. The main results of this study are as follows: under rotary tillage conditions, the 2010s hybrid (DH618) significantly increased yields (9.37–55.89%) compared to hybrids from the 1970s–2000s. After subsoiling, the physiologically mature grains of all hybrids exhibited minimal changes in crude protein and fat content, while there was a significant reduction in the total soluble sugar content of the grains. After subsoiling, there was a substantial 8.14 to 12.79 percent increase in total starch accumulation in the grain for all hybrids during the period of 47–75 days post-anthesis. Furthermore, during the period of 47–75 days after anthesis, the consumption of grain crude protein significantly contributed to the accumulation of total starch in the grains. Ultimately, subsoiling significantly increased the yield of each hybrid and enhanced the total grain starch content at physiological maturity of all hybrids, with the 2010s hybrid (DH618) performing exceptionally well.

Background Gut dysbiosis in peritoneal dialysis (PD) patients causes chronic inflammation and metabolic disorders which result in a series of complications, probably playing an important role in PD technique failure. The reduction in gut microbial diversity was a common feature of gut dysbiosis. The objective was to explore the relationship between gut microbial diversity and technique failure in PD patients.Methods The gut microbiota was analyzed by 16s ribosomal RNA gene amplicon sequencing. Cox proportional hazards models were used to identify association between gut microbial diversity and technique failure in PD patients.Results In this study, a total of 101 PD patients were enrolled. During a median follow-up of 38 months, we found that lower diversity was independently associated with a higher risk of technique failure (hazard ratio [HR], 2.682; 95% confidence interval [CI], 1.319–5.456; p = 0.006). IntroductionAs global urbanization intensifies, the physical and mental stressors of modern life have led to the growing prevalence of suboptimal health conditions. Spending time in a forest benefits human health and well-being. In this context, based on the forest spatial types (forest interior and forest edge spaces), landscape elements (architecture, water and roads) and forest stands (coniferous, broadleaf and bamboo forests), this study investigated the effects of different forest spatial landscape characteristics on the restorative potential for college students, aesthetic preference and eye movement behavior (total fixation duration and fixation count).MethodsIn this study, a total of 60 subjects were exposed to 42 photographs depicting typical forest landscapes acquired through field studies. The Short-version Revised Restoration and Preference Scale and eye-tracking technology, were employed to study the recovery efficiency and visual attraction of forest spatial of different forest spatial types, element compositions and forest stands.Results(1) The restorative potential and aesthetic preference score of forest edge spaces were significantly higher than those of forest interior spaces. (2) The restorative potential of bamboo forests was significantly higher than those of coniferous and broadleaf forests. (3) In terms of forest interior space, the restorative potential of “forest + 1 element” composition and “forest + 2 elements” composition was significantly higher than that of pure forest, and the restorative potential of interior space of bamboo forest was significantly higher than those of coniferous and broadleaf forests. (4) In terms of forest edge space, the restorative potential of “forest + 2 elements” composition was significantly higher than that of pure forest, and the restorative potential of pure forests was significantly higher than that of the “forest + 1 element” composition. (5) The restorative potential of forest spatial landscape characteristics positively correlated with aesthetic preference and negatively correlated with total fixation duration and fixation count. These results can provide a reference for future forest landscape research, construction and management.

Abstract Background Perioperative hypotension increases postoperative complication rates and prolongs postoperative recovery time. Whether Passive Leg Raising test (PLR) and Subclavian Vein Diameter (DSCV) can effectively predict post-anesthesia hypotension remains to be tested. This study aimed to identify specific predictors of General Anesthesia (GA)induced hypotension by measuring DSCV in the supine versus PLR position. Methods A total of 110 patients who underwent elective gynecological laparoscopic surgery under general anesthesia, were enrolled in this study. Before anesthesia, DSCV and theCollapsibility Index of DSCV(DSCV-CI) were measured by ultrasound, and the difference in maximal values of DSCV between supine and PLR positions was calculated, expressed as ΔDSCV. Hypotension was defined as Mean Blood Pressure (MBP) below 60mmhg or more than 30% below the baseline. Patients were divided into two groups according to the presence (Group H) or absence (Group N) of postanesthesia hypotension. The area under the receiver operating characteristic curve (ROC) and logistic regression analyses were used to evaluate the predictability of DSCV and other parameters for predicting preincision hypotension. Results Three patients were excluded due to unclear ultrasound scans, resulting in a total of 107 patients studied. Twenty-seven (25.2%) patients experienced hypotension. Numerous viruses manipulate host factors for viral production. We demonstrated that human enterovirus A71 (EVA71), a primary causative agent for hand, foot, and mouth disease (HFMD), increased the level of the DNA damage response (DDR) marker γ-H2AX. DDR is primarily mediated by the ataxia telangiectasia mutated (ATM), ATM and Rad3-related (ATR), or DNA-dependent protein kinase (DNA-PK) pathways. Upregulation of γ-H2AX by EVA71 was dependent on the ATR but not the ATM or DNA-PK pathway. As a nuclear factor, there is no previous evidence of cytoplasmic distribution of γ-H2AX. However, the present findings demonstrated that EVA71 encouraged the localization of γ-H2AX to the cytoplasm. Of note, γ-H2AX formed a complex with structural protein VP3, non-structural protein 3D, and the viral genome. Treatment with an inhibitor or CRISPR/Cas9 technology to decrease or silence the expression of γ-H2AX decreased viral genome replication in host cells; this effect was accompanied by decreased viral protein expression and virions. In animal experiments, caffeine was used to inhibit DDR; the results revealed that caffeine protected neonatal mice from death after infection with EVA71, laying the foundation for new therapeutic applications of caffeine. More importantly, in children with HFMD, γ-H2AX was upregulated in peripheral blood lymphocytes. The consistent in vitro and in vivo data on γ-H2AX from this study suggested that caffeine or other inhibitors of DDR might be novel therapeutic agents for HFMD.

Acute kidney injury (AKI) is a serious condition with high mortality. The most common cause is kidney ischemia/reperfusion (IR) injury, which is thought to be closely related to pyroptosis. Disulfiram is a well-known alcohol abuse drug, and recent studies have shown its ability to mitigate pyroptosis in mouse macrophages. This study investigated whether disulfiram could improve IR-induced AKI and elucidated the possible molecular mechanism. We generated an IR model in mouse kidneys and a hypoxia/reoxygenation (HR) injury model with murine tubular epithelial cells (MTECs). The results showed that IR caused renal dysfunction in mice and triggered pyroptosis in renal tubular epithelial cells, and disulfiram improved renal impairment after IR. The expression of proteins associated with the classical pyroptosis pathway (Nucleotide-binding oligomeric domain (NOD)-like receptor protein 3 (NLRP3), apoptosis-related specific protein (ASC), caspase-1, N-GSDMD) and nonclassical pyroptosis pathway (caspase-11, N-GSDMD) were upregulated after IR. Disulfiram blocked the upregulation of nonclassical but not all classical pyroptosis pathway proteins (NLRP3 and ASC), suggesting that disulfiram might reduce pyroptosis by inhibiting the caspase-11-GSDMD pathway. In vitro, HR increased intracellular ROS levels, the positive rate of PI staining and LDH levels in MTECs, all of which were reversed by disulfiram pretreatment. Furthermore, we performed a computer simulation of the TIR domain of TLR4 using homology modeling and identified a small molecular binding energy between disulfiram and the TIR domain. We concluded that disulfiram might inhibit pyroptosis by antagonizing TLR4 and inhibiting the caspase-11-GSDMD pathway.

Cadmium (Cd) pollution is a global problem affecting soil ecology and plant growth. Abscisic acid (ABA) acts as a growth and stress hormone, regulates cell wall synthesis, and plays an important role in plant responses to stress. There are few studies on the mechanisms behind abscisic acid alleviation of cadmium stress in Cosmos bipinnatus, especially in regards to regulation of the root cell wall. This study examined the effects of different concentrations of abscisic acid at different concentrations of cadmium stress. Through adding 5 μmol/L and 30 μmol/L cadmium, followed by spraying 10 μmol/L and 40 μmol/L ABA in a hydroponic experiment, it was found that under two concentrations of cadmium stress, low concentration of ABA improved root cell wall polysaccharide, Cd, and uronic acid content. Especially in pectin, after the application of low concentration ABA, the cadmium concentration was significantly increased by 1.5 times and 1.2 times compared with the Cd concentration under Cd5 and Cd30 treatment alone, respectively. Fourier-Transform Infrared spectroscopy (FTIR) demonstrated that cell wall functional groups such as -OH and -COOH were increased with exposure to ABA. Additionally, the exogenous ABA also increased expression of three kinds of antioxidant enzymes and plant antioxidants. The results of this study suggest that ABA could reduce Cd stress by increasing Cd accumulation, promoting Cd adsorption on the root cell wall, and activating protective mechanisms. This result could help promote application of C. bipinnatus for phytostabilization of cadmium-contaminated soil.

Field manual labor behavior recognition is an important task that applies deep learning algorithms to industrial equipment for capturing and analyzing people’s behavior during field labor. In this study, we propose a field manual labor behavior recognition network based on an enhanced SlowFast architecture. The main work includes the following aspects: first, we constructed a field manual labor behavior dataset containing 433,500 fast-track frames and 8670 key frames based on the captured video data, and labeled it in detail; this includes 9832 labeled frames. This dataset provides a solid foundation for subsequent studies. Second, we improved the slow branch of the SlowFast network by introducing the combined CA (Channel Attention) attention module. Third, we enhanced the fast branch of the SlowFast network by introducing the ACTION hybrid attention module. The experimental results show that the recognition accuracy of the improved SlowFast network model with the integration of the two attention modules increases by 7.08%. This implies that the improved network model can more accurately locate and identify manual labor behavior in the field, providing a more effective method for problem solving.

Abstract Drought stress and the scarcity of nitrogen fertilizer are two of the important abiotic factors affecting maize growth. Bio-char can enhance the maize yield. Therefore, two field experiments were carried out in the 2 years (2019–2020) to study the effects of nitrogen fertilizer at three levels and four levels of bio-char on endogenous protective enzymes, dry matter accumulation, and yield of the maize ‘Xianyu 335’ under two different irrigation methods. A split-plot system in three replications was established to conduct the field trials. Two irrigation methods (Regular irrigation and Irregular irrigation) were in the main plots, three nitrogen fertilization levels (0, 150, 300 kg h−1 m2) were in sub-plots, and four bio-char levels (0, 8, 16, 24 t h−1 m−2) were in the sub-sub plots. Each sub-plot consisted of 9 rows with 5 m length and 0.6 m width, and each sub-plot area was 30 m2 in the 2 years. The results indicated that the irrigation methods, the nitrogen, and bio-char supply significantly affected the maize endogenous protective enzymes, dry matter accumulation, and yield in the 2 years. Under the same irrigation method, nitrogen fertilizer and bio-char significantly improved the endogenous protective enzyme activity, dry matter accumulation, and yield of maize compared to the treatment without nitrogen fertilizer and bio-char. The above characteristics improved with increased bio-char supply and nitrogen fertilization at 150 kg h−1 m−2. The treatment of C24N150 recorded the highest values for the parameters of maize endogenous protective enzymes activity, dry matter accumulation, and yield under different irrigation methods during the two harvest seasons.

Plumbago indica L. is a perennial herb with ornamental and anticancer medicinal functions widely distributed in the tropics. It is affected by temperature and cannot bloom normally in colder subtropical regions, which seriously affects its ornamental value. To create low-temperature resistance mutants and enrich new germplasm resources, this study used tissue culture and chemical reagent (0.5 mmol/L NaN3) and low-temperature stress (0°C, full darkness for 48h) induction to target and screen for cold-resistance mutants. The results showed that the ISSR band polymorphism ratio of the 24 suspected mutant materials was 87.5%. The DNA profiles of the 9 mutants initially identified were altered. The content of plumbagin in the stems and leaves of the mutants was examined, and it was found that the accumulation in the leaves of the mutant SA24 could be as high as 3.84 times that of the control, which was 0.5991%. There were significant differences in the anatomical structures of roots, stems and leaves. The mutants mostly exhibited reduced root diameter (only 0.17-0.69 times that of CK), increased stem diameter (up to 2.19 times that of CK), enlarged mesophyll cells, increased thickness (up to 1.83 times that of CK) and high specificity, which are thought to be important for the different cold resistance obtained by the mutants. In the cold resistance experiment, four cold-tolerant mutants were successfully screened according to their morphological characteristics and physiological indexes, and the mutagenesis efficiency could be as high as 2.22% and did not affect the accumulation of plumbagin in their stems and leaves, even higher than CK. The responses of the screened mutants SA15, SA19, SA23 and SA24 to low temperature showed slower leaf wilting, higher light energy conversion efficiency, less accumulation of MDA content, increased enzymatic activities of antioxidant enzymes (SOD, CAT, POD) and more accumulation of soluble sugars and proline content. These characteristics are consistent with the response of cold-resistance plants to low temperatures. The cold- resistance mutants cultivated in soil were observed of agronomic and ornamental traits for one year, mainly manifested as delayed flowering and delayed entry into the senescence stage. This study provides a more rapid and accurate technique for identifying and screening cold-tolerant mutants, and lays the foundation for future experiments on the creation of new cold-resistant varieties.

Background Wenyang-Yiqi Granule (WYYQ) is a four-component herbal formula, widely used to treat heart failure in China. It is known to regulate autophagy, but the mechanism(s) are unknown.

Drought is among the most important abiotic stresses on plants, so research on the physiological regulation mechanisms of plants under drought stress can critically increase the economic and ecological value of plants in arid regions. In this study, the effects of drought stress on the growth status and biochemical indicators of Iris japonica were explored. Under drought stress, the root system, leaves, rhizomes, and terrestrial stems of plants were sequentially affected; the root system was sparse and slender; and the leaves lost their luster and gradually wilted. Among the physiological changes, the increase in the proline and soluble protein content of Iris japonica enhanced the cellular osmotic pressure and reduced the water loss. In anatomical structures, I. japonica chloroplasts were deformed after drought treatment, whereas the anatomical structures of roots did not substantially change. Plant antioxidant systems play an important role in maintaining cellular homeostasis; but, as drought stress intensified, the soluble sugar content of terrestrial stems was reduced by 55%, and the ascorbate peroxidase, glutathione reductase, and monodehydroascorbate reductase (MDHAR) activities of leaves and the MDHAR activity of roots were reduced by 29%, 40%, 22%, and 77%, respectively. Overall, I. japonica was resistant to 63 days of severe drought stress and resisted drought through various physiological responses. These findings provide a basis for the application of I. japonica in water-scarce areas.

Maize is essential in ensuring food security in China as a primary food and feed crop. One of the main ways to increase yield in maize production systems is to increase planting density as appropriate. Clarifying the relationship between planting density and grain nutritional quality can provide theoretical guidance for high-yielding high-quality maize cultivation and management practices. To this end, five representative high-yielding maize varieties from the 1970s to the 2010s in China were used as experimental material, and two planting densities of 45,000 plants hm−2 and 105,000 plants hm−2 were set to analyze the changing patterns of yield traits and grain nutrient quality of maize varieties in different eras, as well as their responses to densification conditions. The results showed that, under low-density conditions, the grain nutrient quality components of the 2010s’ variety (DH618) were all different 75 days after anthesis compared with the 1970s’–2000s’ varieties and yields also significantly increased by 11.15% to 19.18% (p < 0.05). The increase in planting density led to a rise in total grain starch and soluble sugar content 75 days post-anthesis in all varieties from the 1970s to the 2010s, with increases of 0.65–1.65% and 39.44–69.01%, and a decrease in crude grain protein and crude fat content, with reductions of 4.15–8.50% and 3.00–11.18%. The increase in total grain starch content 75 days post-anthesis was mainly due to the rise in grain starch accumulation between 23 and 47 days post-anthesis in the 1970s’–2010s’ varieties, with an increase of 7.72–9.19% in all varieties. The higher accumulation of crude fat and soluble sugar in the 0–23 days post-anthesis period also contributed to the increase in total starch accumulation in the 23–47 days post-anthesis period. Ultimately, densification conditions also contributed to a significant increase in yield across all eras of the varieties based on changes in grain nutritional quality, with a more significant increase in yield due to densification and a smaller decrease in grain crude fat content due to densification 75 days after anthesis in the 2010s’ variety (DH618). Therefore, in cultivation and production processes that do not have specific requirements for the nutritional quality components of maize grain, we suggest that the use of a representative high-yielding maize variety (DH618) from the 2010s, together with appropriate planting at close planting distances, can significantly increase maize yields based on an increase in the total starch content of the grain at physiological maturity.

Rosa rugosa (Rosaceae) is an important functional plant used in food products, tea, and aromatherapy. Characteristics of R. rugosa varieties based on the biological traits and floral fragrant components were studied by applying an analytic hierarchy process, headspace solid-phase microextraction gas chromatography–mass spectrometry, and metabolomic analysis. The 77 Rosa accessions (comprising 27 R. rugosa varieties, 43 scented R. hybrida cultivars, and seven fragrant R. species) were grouped into nine classes based on 17 morphological characters and 16 targeted fragrant substances by cluster analysis. Three R. rugosa cultivars differing in fragrance type were selected for volatile metabolomics analysis at four stages of flower development. In total, 156 differential volatile organic compounds (VOC) were detected and the VOC content patterns were further investigated in two important metabolic pathways (the monoterpenoid biosynthetic pathway, and the phenylalanine, tyrosine, and tryptophan biosynthesis pathway). The results provide a foundation for efficient use of Rosa germplasm and insights into the utilization of R. rugosa as a functional flower.

Peritoneal dialysis (PD) is a renal replacement therapy for end-stage renal disease. Gut microbiota-derived uremic solutes, indoxyl sulfate (IS), p-cresyl sulfate (PCS), and trimethylamine-N-oxide (TMAO) accumulate in PD patients. The objective was to explore the gut microbiota and their influence on uremic toxins in PD patients and healthy controls (HC). Fecal samples were collected from PD patients (n = 105) and HC (n = 102). 16S rRNA gene regions were sequenced for gut microbiota analysis. IS, PCS, and TMAO levels were measured using HPLC-MS. PD patients exhibited lower alpha diversity and altered gut microbiota composition compared to HC. At the genus level, PD patients showed increased abundance of opportunistic pathogenic bacteria, and decreased abundance of beneficial bacteria. Three Operational Taxonomic Units discriminated PD patients from HC. Phenylalanine metabolism increased in PD, whereas tryptophan metabolism was unaltered. Low serum PCS did not necessarily mean healthier due to the loss of alpha diversity, increased Proteobacteria and opportunistic pathogenic bacteria. High serum PCS was mainly caused by elevated p-cresol-producing bacteria, enriched amino acid related enzymes, and enhanced sulfur metabolism, rather than declined residual renal function. In patients with different urine volumes, the gut microbiota alpha diversity and composition were unaltered, but serum IS and TMAO were significantly elevated in anuric patients. In conclusion, the gut microbiota abundance, composition, and function were altered in PD patients, which increased the PCS levels. We provided a better understanding of the microbiota-metabolite-kidney axis in PD patients. Targeting certain bacteria could decrease the PCS levels, whereas preserving the residual renal function could reduce the IS and TMAO levels.

Lead (Pb) is one of the most harmful, toxic pollutants to the ecological environment and humans. Centipedegrass, a fast-growing warm-season turfgrass, is excellent for Pb pollution remediation. Exogenous low-molecular-weight organic acid (LMWOA) treatment is a promising approach for assisted phytoremediation. However, the effects of this treatment on the tolerance and Pb accumulation of centipedegrass are unclear. This study investigated these effects on the physiological growth response and Pb accumulation distribution characteristics of centipedegrass. Applications of 400 μM citric acid (CA), malic acid (MA) and tartaric acid (TA) significantly reduced membrane lipid peroxidation levels of leaves and improved biomass production of Pb-stressed plants. These treatments mainly increased peroxidase (POD), catalase (CAT) and ascorbate peroxidase (APX) activities and enhanced free protein (Pro), ascorbic acid (AsA) and phytochelatins (PCs) contents, ultimately improving the Pb tolerance of centipedegrass. Their promoting effects decreased as follows: TA>CA>MA. All the treatments decreased root Pb concentrations and increased stem and leaf Pb concentrations, thus increasing total Pb accumulation and TF values. MA had the best and worst effects on Pb accumulation and Pb transportation, respectively. CA had the best and worst effects on Pb transportation and Pb accumulation, respectively. TA exhibited strong effects on both Pb accumulation and transport. Furthermore, all treatments changed the subcellular Pb distribution patterns and distribution models of the chemical forms of Pb in each tissue. The root Pb concentration was more highly correlated with the Pb subcellular fraction distribution pattern, while the stem and leaf Pb concentrations were more highly correlated with the distribution models of the chemical forms of Pb. Overall, TA improved plant Pb tolerance best and promoted both Pb absorption and transportation well and is considered the best candidate for Pb-contaminated soil remediation with centipedegrass. This study provides a new idea for Pb-contaminated soil remediation with centipedegrass combined with LMWOAs.

Abstract Background: The immune senescence marked by inflation of memory T cell is established in end-stage renal disease (ESRD) patients with peritoneal dialysis(PD). These patients suffer high incidence of infectious disease, which has been relevant to immune dysfunction. However the association of immune senescence with infection in PD patients is not clear. This prospective study is aim to investigate relationship between proportion of T cell subsets and infection event in patients on PD. Methods: We enrolled patients on PD> 6 months from January 1,2016 to December 30,2016 and followed until April 30,2020. Baseline T cell subsets from blood were collected at the time of recruitment. The primary end point was infection event including peritonitis, exit site infection, pneumonia, urinary tract infection and other infection. Results: There were 94 patients (46 male) with a mean age of 56.1±14.9 years old enrolled during follow-up period, and 26 patients suffered infection events. Higher proportion of effector memory(EM) CD8+ T cell was found in patients with infection than those without infection. There was no difference of distribution of EM CD8+ T cell between PD-related and non-dialysis infection. Increased level of EM CD8+ T cell was risk factor for first infection event in PD patients. Conclusion: High level of EM CD8+ T cell could be a significant predictor of infection event in patients on PD.

Abstract Background Epithelial-mesenchymal transition (EMT) of mesothelial cells is a key step in the peritoneal fibrosis (PF). Recent evidence indicates that signal transducer and activator of transcription 3 (STAT3) might mediate the process of renal fibrosis, which could induce the expression of hypoxia-inducible factor-1α (HIF-1α). Here, we investigated the effect of STAT3 activation on HIF-1α expression and the EMT of mesothelial cells, furthermore the role of pharmacological blockade of STAT3 in the process of PF during peritoneal dialysis (PD) treatment. Methods Firstly, we investigated the STAT3 signaling in human peritoneal mesothelial cells (HPMCs) from drained PD effluent. Secondly, we explored the effect of STAT3 signaling activation on the EMT and the expression of HIF-1α in human mesothelial cells (Met-5A) induced by high glucose. Finally, peritoneal fibrosis was induced by daily intraperitoneal injection with peritoneal dialysis fluid (PDF) so as to explore the role of pharmacological blockade of STAT3 in this process. Results Compared with the new PD patient, the level of phosphorylated STAT3 was up-regulated in peritoneal mesothelial cells from long-term PD patients. High glucose (60 mmol/L) induced over-expression of Collagen I, Fibronectin, α-SMA and reduced the expression of E-cadherin in Met-5A cells, which could be abrogated by STAT3 inhibitor S3I-201 pretreatment as well as by siRNA for STAT3. Furthermore, high glucose-mediated STAT3 activation in mesothelial cells induced the expression of HIF-1α and the profibrotic effect of STAT3 signaling was alleviated by siRNA for HIF-1α. Daily intraperitoneal injection of high-glucose based dialysis fluid (HG-PDF) induced peritoneal fibrosis in the mice, accompanied by the phosphorylation of STAT3. Immunostaining showed that phosphorylated STAT3 was expressed mostly in α-SMA positive cells in the peritoneal membrane induced by HG-PDF. Administration of S3I-201 prevented the progression of peritoneal fibrosis, angiogenesis, macrophage infiltration as well as the expression of HIF-1α in the peritoneal membrane induced by high glucose. Conclusions Taken together, these findings identified a novel mechanism linking STAT3/HIF-1α signaling to peritoneal fibrosis during long-term PD treatment. It provided the first evidence that pharmacological inhibition of STAT3 signaling attenuated high glucose-mediated mesothelial cells EMT as well as peritoneal fibrosis.

Dehydration of maize grains in the field can reduce costs associated with drying after harvest. A delayed harvest approach after physiological maturity, in which plants stand in the field to allow the stems to dry, has been widely adopted in maize production. However, it remains unclear how harvesting at different time points during the dehydration stage may affect grain yield and quality. Objective This study aimed to retrospectively analyse the influence of epidural labour analgesia (ELA) on delivery and maternal and neonatal outcomes in nulliparous women with gestational diabetes mellitus (GDM) using propensity score-matched analysis.Design Retrospective cohort analysis.Setting Primary care practices in a teaching hospital from March 2018 to October 2021.Participants A total of 816 delivery records of nulliparous women with GDM were collected and retrospectively analysed.Interventions ELA and non-ELA (NELA) cohorts were assessed.Main outcome measure The primary outcome assessed was delivery type (spontaneous, assisted vaginal or caesarean). Ethylenediaminetetraacetic acid (EDTA) is one of the most effective chelating agents for enhancing lead (Pb) accumulation in various plant organs. However, it has a higher risk of causing secondary pollution than other chelating agents. To reduce such environmental risks and increase remediation efficiency, EDTA can be combined with degradable chelating agents for use in phytoremediation, but there are few reports on the combination of EDTA and nitrilotriacetic acid (NTA). This study evaluated the effects of combined EDTA and NTA application at different concentrations (900, 1200, or 1500 mg/kg) and with different methods (1 application or 3 applications) on dwarf bamboo (Sasa argenteostriata (Regel) E.G. Camus) growth and phytoremediation efficiency and on the soil environment in pot experiments with Pb-contaminated soil. Applying EDTA and NTA together resulted in lower soil water-soluble Pb concentrations than applying EDTA alone and therefore resulted in lower environmental risk. The increased availability of soil Pb produced a stress response in the dwarf bamboo plants, which increased their biomass significantly. Moreover, under the chelating treatments, the soil Pb availability increased, which promoted Pb translocation in plants. The Pb content in the aerial parts of the dwarf bamboo increased significantly in all treatments (translocation factors increased by 300~1500% compared with that in CK). The Pb content increase in the aerial parts caused high proline accumulation in dwarf bamboo leaves, to alleviate Pb toxicity. Maximum Pb accumulation was observed in the EN1500 treatment, which was significantly higher than that in the other treatments except the EN900 treatment. This study elucidates the choice of remediation techniques and the physiological characteristics of the plants used in such studies. In conclusion, the EN900 treatment resulted in the lowest environmental risk, greatest biomass production, and highest phytoremediation efficiency of all treatments, indicating that it has great potential for application in phytoremediation with dwarf bamboo in Pb-contaminated soil.

Quantifying the relationships of maize yield and nitrogen use efficiency (NUE) to inherent soil productivity (ISP) could provide a theoretical basis for implementing strategies that concurrently narrow the yield gap, increase NUE, and improve soil quality. A field study under irrigation conditions was conducted at five locations with large differences in ISP (estimated by maize grain yield without using fertilizers) across various ecological regions in Inner Mongolia, China. Our results showed that the changes in maize yield and nitrogen partial factor productivity (PFPN) to ISP followed a linear-plateau model, with a common inflection point. When ISP was below 8.0 t ha–1, maize yield and PFPN were stagnating at a low level, due to a trade-off between the contribution of soil and chemical fertilizers to yield. When ISP exceeded 8.0 t ha–1, the contribution rate of soil to yield stabilized at 80%, resulting in a simultaneous increase in maize yield by 1.2 t ha–1 and PFPN by 4.6 kg kg–1, for increasing ISP at each t ha–1. Our results indicated that while keeping other management strategies unchanged, to increase maize yield and PFPN by 15% or further 30%, it is necessary to increase ISP to 9.3 and 10.4 t ha–1, respectively. In this scenario, N input will be reduced by 33.5 and 47.5%, and apparent N losses will be reduced by 11.7 and 21.5%, respectively. Therefore, enhancing ISP could lead to a simultaneous improvement in yield and NUE as well as strongly support the green development of maize production.

In this study, a ternary blend modifier (TBM) was prepared with RPE, RPP, and various dosages of PP-g-MAH (i.e., 0%, 1%, 3%, 5%, 7%, and 9% by weight of RPE and RPP) by a twin-screw extruder. The performance of TBM (4% by weight of asphalt) modified asphalt was studied, with the increase of PP-g-MAH content, the penetration decreases gradually, but softening point and viscosity increases and ductility exists minimum value at 7% content, with the increasing of PP-g-MAH percentage, the dynamic rheological properties show that |G*| increases, while the δ and |G*|cosδ decrease, and the fatigue performance (|G*|sinδ) of TBM modified asphalt is worse than virgin asphalt. Dynamic stability test shows that TBM shows the prominent high-temperature properties of a modified (asphalt mixture. BBR test indicates that the m-value of asphalt modified with TBM does not fulfill the requirements (m-value>0.3) except at 5% and 7%, but the S meets the standard requirements (S < 300 MPa), and increases from 0% to 7%, then decreases slightly. In order to illuminate possible reasons, a chemical reaction (ester compounds generated) and physical twines of concerned polymer molecules were put forward, and the results were confirmed by infrared analysis.

Solid bacterial agents are required to accelerate stover degradation in low-temperature areas. However, the laboratory-to-practice translation of bioprocessing techniques is hindered by high cost, poor practicality, and short shelf life. Using corn stover powder, starch, and bran as additives, we screened Pseudomonas putida and Acinetobacter lwoffii, which effectively degrades corn stover at low temperatures, to develop a sustainable and low-cost bacterial agent formula that ensures bacterial viability in low-temperature soil and storage. The optimal formulation included precipitates and additives at a 1:4 ratio, including corn stover powder, starch, and bran at a 4:3:9 ratio. The viable bacterial count with this formulation reached 7.5 × 1010 colony-forming units/g, with high lignocellulase activities. The degradation effect of the optimal formulation on stover and its components, in both lab soil culture simulation and the field environment, was significantly higher than that without bacterial agent application. This formulation had an outstanding effect on lignin. The optimal storage conditions included vacuum packing under 10% water content at 4 °C; the survival rate of viable bacteria reached 85.33% after 180 d. Given the global value of stover-return agriculture, our results offer a valuable strategy for application in low-temperature soils where stover degradation rates are otherwise low.

To clarify the effect of tillage methods on saline–alkali land improvement and maize growth in cropland salinized to different degrees, we set up two treatments (shallow rotation (15 cm depth; CK) and deep straw return (35 cm depth; DPR)) in land characterized by three different salinization degrees and analyzed the effects of the two treatments on soil nutrient content, salinity index, chlorophyll fluorescence, growth status, and yield at three salinization levels. The results show that (1) compared with CK, alkaline N, total N, Olsen P, exchangeable K, and organic matter in saline soils were all significantly improved, and total salt and pH values were reduced by 34.01–50.79% and 2.56–7.54%, respectively, under deep straw return conditions, representing the largest values in moderately saline–alkali land. (2) Compared with CK, chlorophyll fluorescence was significantly improved, and maximum photochemical efficiency (Fv/Fm), photochemical quenching (qP), and effective quantum yield of PSII (ΦPSII) were significantly increased by 8.09–15.41%, 9.13–17.93%, and 38.79–70.83% following deep straw return treatment; these increases were the largest ones observed in moderately saline–alkali land. (3) Deep straw return promoted the growth of maize and significantly increased the yield of maize. Plant height, leaf area index, and yield increased the most in moderately saline–alkali land and increased by 6.84–21.79%, 0.59–2.28 units, and 12.78–28.07%, respectively. The yield increased by 33.89 %, which was mainly due to the increase in 1000-grain weight. The results provide a theoretical basis for taking straw return measures to improve soil and increase maize yield in saline–alkali land.

ObjectiveTo investigate the effects of glial cell-derived neurotrophic factor (GDNF), GDNF family receptor alpha 1 (GFRα1), and glial fibrillary acidic protein (GFAP) on colonic motility in a mouse model of intestinal neuronal dysplasia by intervention with Bifidobacterium and to explore the influence of Bifidobacterium on enteric glial cells (EGCs).MethodsWestern blotting and qRT-PCR were employed to detect the expression of GFRα1 and GFAP in colonic tissues of mice with or without Tlx2 mutations, and ELISA was used to detect the expression of GDNF in serum. IHC was used to detect the appearance of the ganglion cells. Subsequently, Tlx2 homozygous mutant (Tlx2−/−) mice were treated with Bifidobacterium. Colonic motility was measured before and after intervention by measuring the glass bead expelling time. The variations in abdominal circumference and GDNF, GFRα1, and GFAP expression were measured. In addition, 16SrRNA gene sequencing was performed to detect the abundance of the intestinal microbiota.ResultsThe mRNA and protein expression of GFRα1 and GFAP was decreased in the colonic tissues of Tlx2−/− mice and GDNF expression was decreased in serum compared with Tlx2+/− and WT mice. After confirming the colonization of Bifidobacterium by 16S rRNA gene sequencing, the expelling time and abdominal distension were ameliorated, and the expression of GFAP, GDNF, and GFRα1 was increased.ConclusionsThe expression of GDNF, GFRα1, and GFAP is associated with colonic motility. The altered expression of EGC-related factors suggested that Bifidobacterium may be involved in the EGC activation process. The amelioration of IND symptoms after intervention with Bifidobacterium prompted the elicitation of adjuvant therapy.

This study explored changes in the microbial community structure during straw degradation by a microbial decomposer, M44. The microbial community succession at different degradation periods was analyzed using MiSeq high-throughput sequencing. The results showed that 14 days after inoculation, the filter paper enzyme and endoglucanase activities increased to 2.55 U·mL-1 and 2.34 U·mL-1. The xylanase, laccase, and lignin peroxidase activities rose to 9.86 U·mL-1, 132.16 U·L-1, and 85.43 U·L-1 after 28 d, which was consistent with changes in the straw degradation rate. The degradation rates of straw, lignin, cellulose, and hemicellulose were 31.43%, 13.67%, 25.04%, and 21.69%, respectively, after 28 d of fermentation at 15°C. Proteobacteria, Firmicutes, and Bacteroidetes were the main bacterial species in samples at different degradation stages. The dominant genera included Pseudomonas, Delftia, and Paenibacillus during the initial stage (1 d, 7 d) and the mid-term stage (14 d). The key functional microbes during the late stage (21 d, 28 d) were Rhizobium, Chryseobacterium, Sphingobacterium, Brevundimonas, and Devosia. Changes in the bacterial consortium structure and straw degradation characteristics during different degradation periods were clarified to provide a theoretical basis for the rational utilization of microbial decomposer M44.

Objective: The aim of this study was to explore the development of enteric glial cells (EGCs) in different segments of Hirschsprung's disease (HSCR).Methods: Colonic specimens from 35 children with HSCR were selected to analyze the relative expression of glial fibrillary acidic protein and S100 calcium-binding protein B using Western blotting and real-time fluorescence quantitative PCR. Immunofluorescence and immunohistochemical staining were performed to determine the distribution of myenteric EGCs and neuronal cells in different segments of HSCR.Results: There was a trend of diminished protein and mRNA expression of glial fibrillary acidic protein and S100 calcium-binding protein B from the proximal, dilated, and transitional segments to the aganglionic segment (p < 0.05). Immunofluorescence and immunohistochemistry showed that the EGCs in the aganglionic, transitional, and dilated colonic muscles were morphologically abnormal, which was consistent with the dysplasia of myenteric neurons.Conclusion: Aberrant development of myenteric EGCs was observed in the colon of HSCR, which may affect the survival of enteric neurons.

Meteorological factors are one of the important factors that determine maize kernel weight and grain nutritional quality. Analyzing the influence of meteorological factors before and after anthesis on maize kernel weight and nutritional quality components is of great significance for improving corn yield and quality. Therefore, five different maize hybrids and conducted continuous experiments from 2018 to 2021 were selected in this study, to explore the response of maize kernel weight and grain nutritional quality to meteorological factors in different growth periods, and to quantify the linear relationship between grain nutritional quality parameters, grain weight, and meteorological factors. The main results were as follows: the 100-grain weight reached the maximum value of 39.53 g in 2018; the contents of crude protein, total starch, and crude fat in grains reached the maximum in 2018, 2020, and 2018, respectively, which were 9.61%, 69.2%, and 5.1%. Meteorological factors significantly affected the maize grain weight (p < 0.05). Before anthesis, total sunshine duration, average temperature, relative humidity, and the accumulated temperature had strong effects on grain weight. After anthesis, average daily temperature, total rainfall, temperature difference, accumulated temperature, average daily highest temperature, and total sunshine hours had strong effects on grain weight. There was also a significant correlation between grain weight and grain nutritional quality components (p < 0.05). The multivariate polynomial equation analysis revealed that further potential for maize grain weight could be exploited by adjusting the content of each quality component of the kernels under the current test conditions. Meteorological elements can indirectly affect the 100-grain weight through their relationship with the nutritional quality of the grains, with accumulated temperature before anthesis, average temperature after anthesis, and accumulated temperature after anthesis having the greatest indirect effect on the 100-grain weight. Therefore, the effects of pre-anthesis accumulation temperature, post-anthesis average temperature, and post-anthesis accumulation temperature on the nutritional quality of the grains can be harmonized by the application of hybrids of different lengths of vegetation and by adjusting the sowing time in agricultural production. Ultimately, maize grain weight can be increased on the basis of optimizing the content of various quality components in the grains.

Hijacking host ubiquitin pathways is essential for the replication of diverse viruses. However, the role of deubiquitinating enzymes (DUBs) in the interplay between viruses and the host is poorly characterized. Here, we demonstrate that specific DUBs are potent inhibitors of viral proteins from HIVs/simian immunodeficiency viruses (SIVs) that are involved in viral evasion of host restriction factors and viral replication. In particular, we discovered that T cell-functioning ubiquitin-specific protease 8 (USP8) is a potent and specific inhibitor of HIV-1 virion infectivity factor (Vif)-mediated apolipoprotein B mRNA-editing enzyme catalytic polypeptide-like 3 (APOBEC3)G (A3G) degradation. Ectopic expression of USP8 inhibited Vif-induced A3G degradation and suppressed wild-type HIV-1 infectivity even in the presence of Vif. In addition, specific DUBs repressed Vpr-, Vpu-, and Vpx-triggered host restriction factor degradation. Our study has revealed a previously unrecognized interplay between the host’s DUBs and viral replication. Enhancing the antiviral activity of DUBs therefore represents an attractive strategy against HIVs/SIVs.

BackgroundStomach adenocarcinoma (STAD) is a common reason for tumor-related fatalities globally, as it results in distant metastasis. Methyltransferase-like 14 (METTL14), a notable RNA N6-adenosine methyltransferase (m6A), plays a significant role in the growth of tumor through controlling the RNA working. This study aims to highlight METTL14 in STAD’s biological function and molecular mechanism.MethodsBioinformatics and immunohistochemical (IHC) assays have been utilized for the detection of METTL14 expression in the STAD. METTL14’s biological function has been shown while making use of HGC-27 and AGS cells in vitro experiments. MeRIP-qPCR and luciferase reporter assays were employed for the exploration of METTL14’s mechanism modifying the target of phosphatase and tensin homologue (PTEN). Subcutaneous xeno transplantation model and STAD liver metastasis orthotopic tumor model were used to study METTL14 in STAD in vivo.ResultsMETTL14 expression was substantially downregulated in STAD reflecting contribution to major tumors, progressed TNM stage as well as poor overall survival (OS) in STAD. Moreover, METTL14’s inhibition of STAD cells proliferation, migration and invasion has been verified in vitro assays. Furthermore, an identification of PTEN being METTL14-mediated m6A modification’s substrate has been made. METTL14’s overexpression highly enhanced PTEN mRNA m6A variation, stabilized PTEN mRNA and increased protein expression. Further, it has been found out that METTL14-mediated STAD cells inhibition of proliferation and invasion dependent on PTEN. At last, we demonstrated that METTL14 inhibit STAD growth and metastasis in vivo models.ConclusionsMETTL14 inhibits tumor growth and metastasis of STAD via stabilization of PTEN mRNA expression. Therefore, METTL14 is a potential biomarker of prognosis and therapeutic targets for STAD.

Interpersonal Needs Questionnaire (INQ) is a self-report measure of perceived burdensomeness and thwarted belongingness with five versions in recent studies. There are five versions of INQ. But results from studies using different versions are quite different. Current suicide behavior among teenagers has attracted much attention. But which version is more suitable for teenage samples is still uncertain. It is important to compare the potential differences in different versions of INQ to identify the most psychometrically available version to predict teenagers' acquired capability for suicide and provide them with timely help to reduce teenagers' suicide rates. This study compared the construct validity, internal consistency, validity, and average test information of each version in the sample of teenagers. Results showed the 10-item version provided the most average test information in both thwarted belongingness subscale and perceived burdensomeness subscale, and the INQ-10 is more suitable for teenage samples.

High grain water content at harvest stage is the main limiting factor for realizing mechanized maize grain harvest in China. Under the background of yield gain by density increase, it is necessary to clarify the effect of planting density increase on maize grain dehydration characteristics, which would provide theoretical support for realizing mechanized grain harvest under high planting density conditions. Therefore, this study selects five main hybrids, ZD2, DY13, YD13, XY335, and DH618, released in different eras that were widely promoted in Inner Mongolia from the 1970s to 2010s. The experiment was conducted in the Chilechuan Modern Agriculture Expo Park, Tumed Right Banner, Baotou city, Inner Mongolia, in 2018 and 2019. Under the three densities of 45,000 plants ha−1 (low density), 75,000 plants ha−1 (medium density) and 105,000 plants ha−1 (high density), the indexes of grain dehydration, leaf stay-green, bract and cob dehydration of the different maize hybrids were measured and analyzed. The results show that MCpm (moisture content at physiological maturity) of hybrids in the 1970s and 1990s was significantly reduced by 1.57 and 1.14 percentage points, respectively, and MCh (moisture content at harvest time) in the harvest period of hybrids in the 1970s was significantly reduced by 0.99 percentage points, from a low to medium density. The GDRbm (rate of grain dehydration before maturation) and the GDRam (rate of grain dehydration after maturation) showed an increasing trend from a low to medium density. From a medium to high density, the MCpm from the 1980s to 2000s could be significantly reduced by 1.78, 1.53 and 1.88 percentage points; the MCh from the 1980s could be significantly reduced by 1.77 percentage points; and the GDRbm from the 1970s was significantly increased by 0.101%/d, but the improvement of GDRam was not significant. With the planting density increase, the decreased ratios of relative GLAD (green leaf area duration) and leaf SPAD (soil and plant analyzer development) per plant of old maize hybrids were more than that of modern maize hybrids, which promoted the decrease in grain water content and the rate increase in grain dehydration for old maize hybrids. There was a direct positive correlation between the bract and grain dehydration rates, but the cob dehydration rate had no significant effect on the grain dehydration rate. With the increase in planting density, the relative GLAD and leaf SPAD values of plants decreased, and the stay-green of plants worsened, and a significant increase in the dehydration rate of bracts in old and modern eras was an important reason for the decrease in grain moisture content and increase in dehydration rate.

Renal tubule-interstitial fibrosis is related to chronic kidney disease progression and a typical feature of the aging kidney. Epigenetic modifications of fibrosis-prone genes regulate the development of renal fibrosis. As a kind of “epigenetic diet”, soy isoflavone genistein was reported to have renal protective action and epigenetic-modulating effects. However, its renal protection role and underlying mechanisms are yet to be fully clarified. Herein, we showed that genistein exhibits a demonstrable anti-fibrotic effect on kidney in vivo UUO (unilateral ureteral occlusion) model and renal epithelial cells in vitro model. The mechanism is strongly associated with epithelial-to-mesenchymal transition and m6A RNA demethylase ALKBH5. Mouse fibrotic kidneys induced by UUO exhibited adverse expression of renal fibrosis-related proteins and significant increases in the total m6A level. As an eraser, ALKBH5 showed severer suppression in the renal fibrosis process. However, genistein pretreatment restored ALKBH5 loss remarkably and reduced renal fibrosis, abnormal protein, and inflammatory markers. The examination of possible mechanisms revealed that genistein promoted ALKBH5 and maybe induced the level of mRNA m6A methylation in some epithelial-to-mesenchymal transition-related transcription factors. We found snail was the critical regulator and critical for the protective role of genistein. To verify the relationship between ALKBH5 and snail, we generated knockdown and overexpression of ALKBH5 cells in vitro. ALKBH5 knockdown enhanced the mesenchymal phenotype marker α-smooth muscle actin and snail expression. In agreement, overexpression ALKBH5 increased epithelial adhesion molecule E-cadherin and reduced snail expression. In conclusion, genistein increased renal ALKBH5 expression in UUO-induced renal fibrosis and reduced RNA m6A levels and ameliorates renal damages.

Background: A few studies have shown that urinary kidney injury molecule-1 (uKIM-1) levels are increased in acute kidney injury (AKI); however, the correlation between uKIM-1 and histological tubular injury, which is considered to be the gold standard for evaluating renal damage and predicting prognosis, is still unclear. We performed this study to determine whether the predicted value of uKIM-1 is correlated with renal KIM-1 (tKIM-1) expression and tissue damage in AKI patients. Methods: This retrospective study recruited 14 healthy individuals and 27 biopsy-proven acute tubular injury (ATI) patients. uKIM-1 and plasma KIM-1 (sKIM-1) levels were measured by ELISA, and tKIM-1 expression was evaluated by immunohistochemistry. Results: Elevated levels of urinary, plasma, and renal KIM-1 were found in ATI patients. The uKIM-1 concentration was positively correlated with tKIM-1 expression and reflected the severity of renal histological injury. The outcome of ATI was associated with uKIM-1 expression: the ATI patients with higher uKIM-1 levels had an increased potential for an incomplete recovery of renal function during follow-up. Additionally, the level of KIM-1, regardless of source, was negatively related to the eGFR, and ROC curve analysis revealed that the ROC-AUC was 0.923 (p = 0.000) for the diagnosis of ATI based on a combination of high uKIM-1 and sKIM-1 levels. Conclusion: The uKIM-1 level corresponds with the severity of renal histological damage and can be a potential reliable predictor of adverse renal outcomes in ATI patients. Moreover, combining uKIM-1 and sKIM-1 can increase the sensitivity and specificity of the diagnosis of severe ATI.

Background/Aims: Even though delayed ischemic preconditioning (DIPC) has been reported to produce renal protection, the underlying mechanism remains poorly understood. We reported that a 15-minute renal ischemic preconditioning (IPC) 4 days before subsequent ischemia-reperfusion attenuated renal injury Kidney dendritic cells (DCs) are abundant in the renal tubulointerstitium and, depending on their status, can induce immune activation or tolerance. The aim of the present study was to investigate the role of DCs in IPC of the kidney. Methods: Mouse kidneys were challenged by transient brief episodes of sublethal ischemia followed by subsequent prolonged ischemia. DC abundance and maturation in the spleen and kidney were measured by flow cytometry and immunohistochemical staining. To confirm the function of mature DCs in the renoprotective effect of IPC on renal ischemia-reperfusion injury the A2 adenosine receptor (A2AR) antagonist SCH58261 was administered to stimulate DC maturation prior to assessment of renal functional and histological injury and the inflammatory reaction. Results: Compared with sham-operated animals, preconditioned mice had a reduced injury with less CD11c+ cells, lower levels of the pro-inflammatory cytokine IL-17 and reduced expression of the mature DC marker CCR7. Preconditioned mice also produced more of the anti-inflammatory cytokine IL-10. Both renal cells and splenocytes from these mice had more DCs (CD45+/CD11c+/F4/80-), but fewer of these DCs were mature (CD45+/CD11c+/ F4/80-/MHC-II+/CD80+) compared with those from sham-treated animals, suggesting that the immunomodulatory effect of renal ischemic preconditioning is both local and systemic. Additionally, injection of the A2AR antagonist SCH58261 reversed IPC-induced inhibition of DC maturation and mitigated the protective effect of preconditioning, suggesting that DC maturation contributes to immune cell-mediated ischemic preconditioning. Conclusion: Our results show that DIPC of the kidney provides local and systemic immunosuppression by inhibiting DC maturation and hence mediates a renal protective effect.

Background/Aims: Ischemia is one of the main causes of the high rate of absorption of transplanted autologous fat. Autophagy allows cells to survive by providing energy under starvation. Rapamycin has been found to play a role in promoting autophagy. In this study, we investigated whether rapamycin participates in the survival and adipogenesis of ischemia-challenged adipose-derived stem cells (ADSCs) by regulating autophagy. Methods: Before the cells were exposed to oxygen-glucose deprivation (OGD), a simulated ischemic microenvironment, the level of autophagy was reduced or increased by lentiviral transfection with short hairpin RNA targeting microtubule-associated protein 1-light chain 3 gene (shRNA-LC3) or treatment with rapamycin, respectively. The level of autophagy was assessed by western blotting, transmission electron microscopythen the apoptosis ratio was determined through terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling (TUNEL) and flow cytometry. Adipogenesis was further evaluated by oil red O staining and the expressions level of some specific proteins for adipocytes. Results: shRNA-LC3 and rapamycin treatment effectively decreased and improved the level of autophagy in cells with or without OGD challenge, respectively. In addition, autophagy inhibition increased the apoptosis rate and activated caspase-3 expression level in response to OGD, and these were markedly inhibited by rapamycin preconditioning. During adipogenesis, autophagy inhibition decreased not only oil droplet accumulation but also lipoprotein lipase (LPL) and peroxisome proliferator-activated receptor gamma (PPARγ) expression in cells with or without OGD challenge. However, autophagy promotion by rapamycin increased oil droplet accumulation and LPL and PPARγ expression. Conclusions: Rapamycin may promote the survival and adipogenesis of ischemia-challenged ADSCs by upregulating autophagy.
