Background: and purpose Prostate cancer is an comparatively prevalent clinical malignant tumor in men, impacting the lives of millions of men globally. This study measured the expression of Karyopherin Subunit Beta 1 (KPNB1) in prostate cancer cells, and made an effort to investigate how astragaloside IV affects the biological behavior, tumor growth, and mechanism of action of prostate cancer through KPNB1. Methods: Human prostate cancer and normal cells were obtained and KPNB1 expression levels in the two cells were determined using qPCR and WB. Prostate cancer cells were grouped according to the addition of astragaloside IV, KPNB1 inhibitor (importazole) alone and in combination. KPNB1, NF-κB, and cycle-related proteins were detected to be expressed at different levels in each group's cells by WB. MTT to assess the viability of the cells. To identify the cell cycle, use flow cytometry, and sphere formation experiment to observe sphere formation ability. Nude mice were purchased and subcutaneously inoculated with prostate cancer cells to establish a prostate cancer model, and grouped by tail vein injection of astragaloside IV and importazole. Tumor size was measured. KPNB1 and NF-κB expression in tumor tissues were detected by WB. The expression of proteins relevant to the cycle is observed by immunohistochemical methods. Abstract Background Congenital heart defects (CHDs) are the most common birth defects. Assessment of the incidence, distribution, disease spectrum, and genetic deficits of fetal CHDs in China is urgently needed. Methods A national echocardiography screening program for fetal CHDs was implemented in 92 prenatal screening–diagnostic centers in China. Findings A total of 18,171 fetal CHD cases were identified from 2,452,249 pregnancies, resulting in 7·4/1,000 as the national incidence rate of fetal CHD. The incidences of fetal CHD in the six geographical regions, the southern, central, eastern, southwestern, northern, and northwestern, were 7·647 (CI: 7·383–7·915), 7·839 (CI: 7·680–8·000), 7·647 (CI: 7·383–7·915), 7·562 (CI: 7·225–7·907), 5·618 (CI: 5·337–5·906), and 4·716 (CI: 4·341–5·108), respectively, per 1,000 pregnancies. Overall, ventricular septal defect was the most common fetal CHD, accounting for 17.04% of screened pregnancies nationwide, and tetralogy of Fallot, the most common anomaly in the major defect of fetal CHD, was the second most common, accounting for 9.72%. A total of 76.24% cases of fetal CHD were found to be an isolated intracardiac single defect. The remaining 23.76% of cases of fetal CHD had multiple heart defects. Among all extracardiac malformations, the central nervous system (CNS) was the most common tissue with extracardiac anomalies associated with CHD, accounting for 22.89% of fetal CHD cases. Chromosomal karyotyping identified trisomy 18 as the most common chromosomal abnormality in fetal CHD. We also documented that CHD-containing syndromes could be identified with a comprehensive approach integrating prenatal ultrasound, MRI, pathological autopsy, and cytogenetics and molecular genetics. Conclusion Implementation of prenatal echocardiography as a practically feasible platform to screen fetal CHD will reduce the financial and emotional burden of CHD, which may facilitate intrauterine and neonatal intervention of CHD.

Abstract The aim of this study was to understand the dynamics of multidimensional poverty and its determinants among mid-aged and older adults. We used 2011, 2013, 2015, and 2018 data from the China Health and Retirement Longitudinal Study. We utilised basic descriptive statistics, the poverty transition matrix, Kaplan–Meier estimates, and the discrete-time proportional hazards model for data analysis. From 2011 to 2018, the incidence of multidimensional poverty among mid-aged and older people basically decreased, but the average poverty intensity remained stable. Most mid-aged and older adults had transient multidimensional poverty. The longer an individual remained multidimensionally poor, the smaller the probability of exit from poverty; The longer an individual remained nonpoor after escaping from poverty, the smaller the probability of returning to poverty. As to other factors, individual characteristics, family structure, living arrangements, social capital, and living areas significantly affected the risks of multidimensional poverty exit and reentry. Based on these results, the government should implement targeted interventions for frail older adults with the identified characteristics to prevent them from persistent multidimensional poverty or return to poverty.

Background: ：Post-stroke insomnia（PSI）is one of the common complications after stroke and it is a chronic problem and hampers the patient's recovery. Some treatments have been shown to be effective in treating post-stroke insomnia. However, it is not clear which treatment is more effective. Methods: In this study, we searched CNKI, PubMed, and Cochrane Library for appropriate keywords with a deadline of October 2022 to select 23 randomized controlled trials（RCTs）. The mean difference between different treatments was assessed and summarized as mean and 95 % confidence interval (CI), resulting in a Bayesian network meta-analysis. Results: By meta-analysis of Bayesian networks, we found acupuncture(MD, −2.49; 95 % CI, [-3.63, −1.31]) and herbal (MD, −2.79; 95 % CI, [-4.9, −0.69]) were significantly better than estazolam in terms of PSQI score change, and the difference was statistically significant. Dexzopiclone, electrics stimulation, rTMS and zopiclone were not statistically significant with other treatments. Conclusion: Herbal, zopiclone, and acupuncture were more effective in improving PSQI scores in patients with post-stroke insomnia, followed by rTMS and dexzopiclone. However, the effectiveness between herbal, zopiclone and acupuncture was not statistically significant. Acupuncture and herbal are promising for the treatment of PSI, and more research remains to be invested.

Exploring the influence of farmland transfer on poverty among the mid-aged and elderly rural households is of great value in preventing them from returning to poverty and in solving the problem of rural poverty. Based on the propensity score matching with differences in differences approach, this paper measures and compares the effects of farmland transfer-in and transfer-out on the vulnerability to poverty of the mid-aged and elderly rural households by using the tracking survey of China Health and Retirement Longitudinal Study (CHARLS) in 2015 and 2018. The findings reveal that: (1) The anti-poverty effect of farmland transfer is “asymmetric”. Farmland transfer-in can significantly reduce the vulnerability to poverty of the lessee households, but farmland transfer-out has no such effect. (2) Obvious regional and household differences exist in the anti-poverty effect of farmland transfer. The anti-poverty effects of farmland transfer-in in the eastern and central regions are higher than in other regions. The greater vulnerability to poverty is associated with the more obvious anti-poverty effect of farmland transfer-in. These results benefit the government to strengthen the reforms related to farmland transfer and anti-poverty.

The increasing Municipal Solid Waste (MSW) poses great pressure on cities, especially for emerging countries. This paper presents a mathematical programming model to optimise MSW management. A disjunctive fuzzy optimisation approach is used in this work for several MSW treatment technologies, resulting in multi-objective optimization. The proposed model is used to choose the suitable treatment technologies and optimal distribution of MSW between sources and sinks to minimise the total cost of MSW management and greenhouse gas emission. The optimisation problem is formulated as a mixed-integer linear programming model using generalised disjunctive programming to select suitable MSW treatment technologies and optimal distribution networks. A case study of Hefei, China is presented to illustrate the proposed approach. The multiple objectives of total cost and GHG emissions for MSW management are analysed based on fuzzy optimisation yielding a compromise solution of 192.99 M$/y for total cost and 1,120.48 t/y for emissions. A compromised solution suggested in this study can effectively minimize both total cost and carbon emission and properly deal with the MSW management problem. This work provides a decision tool for the selection of MSW technologies during MSW management.

For the purpose of investigating the contact erosion characteristics in breaking process of SF6 breaker, an erosion experiment was conducted on the tested SF6 circuit breaker. As a result, the relationship between the dynamic resistance and contact travel in various levels of degradation was acquired. The SF6 contact state parameters were obtained from the experimental results. Considering the parameters change, the follow-up conclusions can be sum up: (1) The contact travel’s minimum value is deduced to be 14.2 mm; (2) The numerical value of contact resistance has an acceptable range, which is 50∼150μΩ for main contact and 200∼400μΩ for arcing contact, respectively. (3) The measured value of arcing contact attenuate length is 0.318 mm, which remains unchanged in each erosion test. Taking all the mentioned above into consideration, we research on the contact material erosion process during the opening operation and the effect on the circuit breaker property. In the meantime, the theoretical calculation formula of length decrement in arcing contact is deduced. Based on the formula, the calculated value is 0.268 mm for each trial, which is close to the measured value of 0.318 mm. At last, the prediction methods for the SF6 circuit breaker’s electrical life is summarized.

Similar to the onshore wind power integrated through AC transmission, the offshore wind power integrated through the modular multilevel converter (MMC) based HVDC also lays remarkable impacts on the stability of the power system. According to grid codes, offshore wind power should be capable of adjusting its active power fast. This paper proposes a fast regulation method at the onshore point of common coupling for offshore wind power by utilizing the energy stored in the distributed capacitors in the MMC. By carefully designing the curve of capacitor energy, the active power can vary expectedly. Meanwhile, no disturbance is placed on the DC-side active power or DC voltage. The effectiveness of the proposed method is verified by electromagnetic simulation.

The high cost of the silver catalyst for ethylene epoxidation to ethylene oxide (EO) requires the development of the catalyst with a lower silver loading and a longer catalyst life span. Preventing silver particles from clustering could help maintain the activity for EO. In this work, we applied NH4F solution for an etching-treatment of the α-Al2O3 to improve the roughness of its internal surface before loading silver. The catalyst with etching-treated support showed close activity and EO selectivity initially when compared to the one that was prepared with untreated support while in the meantime its stability was greatly enhanced. The silver catalyst with untreated support lost its activity for EO to nearly zero after 22 days of test at 260 °C and 2 MPa(g), while the one with etching-treated support maintained stable activity from day 10 to 22. The detailed morphology study for the catalysts suggests that the loss of EO activity is mainly attributed to the migration of silver particles from the plane of the support flakes to the gaps that formed by the flakes and deep oxidation of ethylene was largely enhanced by the active sites crowded in the flake gaps. The support treatment effectively slowed down the formation of larger silver clusters and helped maintain a better dispersion of the silver particles on the support.

Carbon nano peapods, with their electronic properties and spintronics, have attracted great attention regarding their potential applications when combined with fullerenes or their derivatives encapsulated inside. Herein, we have designed and synthesized a series of fullerene derivatives with different functional groups, which are then encapsulated into single-walled carbon nanotubes (SWCNTs). Accurate morphological characterization with high-resolution TEM reveals a clear correlation between the filling ratio of the peapods and the steric bulk of the functionalized groups. Further spectroscopic characterizations reveal diameter-selective interactions between the fullerene derivatives and SWCNTs, which, in turn, influence the electronic structures of the nanotubes. Our results have shed new light on the controlled synthesis and property-tuning of nano peapods.

Abstract Background Renal collecting duct carcinoma (CDC) is a rare and lethal subtype of renal cell carcinoma (RCC). The genomic profile of the Chinese population with CDC remains unclear. In addition, clinical treatments are contradictory. In this study, we aimed to identify the genomic mutation spectrum of CDC in the Chinese population. Methods Whole-exome sequencing was performed using the Illumina Novaseq™ 6000 platform. MuTect2 detects single-nucleotide variants (SNVs) and small scale insertions/deletions (INDELs). The identified mutations were annotated with ANNOVAR and validated by Sanger sequencing. Control-FREEC was used to detect copy number variation (CNV), and GISTIC was applied to detect frequently mutated altered regions. These data were compared with associated The Cancer Genome Atlas cohorts. Results Ten normal-matched CDC patients were included. The mean tumour mutation burden was 1.37 Mut/Mb. Six new recurrent somatic mutated genes were identified, including RBM14, MTUS1, GAK, DST, RNF213 and XIRP2 (20% and 2 of 10, respectively), and validated by Sanger sequencing. In terms of common mutated genes, SETD2 was altered in both CDC and other RCC subtypes but not in bladder urothelial carcinoma (BLCA); CDKN2A was a driver gene in both CDC (SNV: 10%, 1 of 10) and BLCA but not in other RCC subtypes. Next, 29 amplifications and 6 deletions of recurrent focal somatic CNVs were identified by GISTIC2.0, which displayed differences from kidney renal clear cell carcinoma (KIRC), kidney renal papillary cell carcinoma (KIRP) and BLCA cohorts. Of note, CDKN2A (CNV alteration: 30%, 3 of 10) and CDKN2A-AS1 were the only overlapping genes of these four cohorts. Importantly, the CDKN2A mutation in our cohort differed from previous studies in urinary carcinomas. Moreover, CDKN2A-altered cases had significantly worse overall survival than wild-type cases in both KIRC and KIRP cohorts. In addition, the most frequently altered genomic pathway of our CDC cohort was the CDKN2A-mediated p53/RB1 pathway. Conclusions Our study offers the first genomic spectrum of the Chinese population with CDC, which differs from that of the Western population. The altered CDKN2A-mediated p53/RB1 pathway might provide new insight into potential therapeutic targets for CDC patients.

The small-signal stability problem of power electronic converters has always been a critical concern for safe operation. This paper conducts a comprehensive analysis of the small-signal stability for the hybrid modular multilevel converter (MMC), which is promising in the application of future over-head-line high-voltage-direct-current transmission. The impacts of the strong and weak AC system connection are comparatively researched through eigenvalue-based analysis. The newly emerging factors that hamper the stability are identified for the weak AC system integration. The work in this paper provides valuable guidance for robust control parameter configuration of the hybrid MMC.

The accurate extraction of cropland distribution is an important issue for precision agriculture and food security worldwide. The complex characteristics in southern China pose great challenges to the extraction. In this study, for the objective of accurate extraction and mapping of cropland parcels in multiple crop growth stages in southern China, we explored a method based on unmanned aerial vehicle (UAV) data and deep learning algorithms. Our method considered cropland size, cultivation patterns, spectral characteristics, and the terrain of the study area. From two aspects—model architecture of deep learning and the data form of UAV—four groups of experiments are performed to explore the optimal method for the extraction of cropland parcels in southern China. The optimal result obtained in October 2021 demonstrated an overall accuracy (OA) of 95.9%, a Kappa coefficient of 89.2%, and an Intersection-over-Union (IoU) of 95.7%. The optimal method also showed remarkable results in the maps of cropland distribution in multiple crop growth stages, with an average OA of 96.9%, an average Kappa coefficient of 89.5%, and an average IoU of 96.7% in August, November, and December of the same year. This study provides a valuable reference for the extraction of cropland parcels in multiple crop growth stages in southern China or regions with similar characteristics.

The integrated energy system (IES) is an important energy supply method for mitigating the energy crisis. A station-and-network–coordinated planning method for the IES, which considers the integrated demand responses (IDRs) of flexible loads, electric vehicles, and energy storage is proposed in this work. First, based on load substitution at the user side, an energy-station model considering the IDR is established. Then, based on the characteristics of the energy network, a collaborative planning model is established for the energy station and energy network of the IES, considering the comprehensive system investment, operation and maintenance, and clean energy shortage penalty costs, to minimize the total cost. This can help optimize the locations of the power lines and natural gas pipelines and the capacities of the equipment in an energy station. Finally, simulations are performed to demonstrate that the proposed planning method can help delay or reduce the construction of new lines and energy-station equipment, thereby reducing the investment required and improving the planning economics of the IES.

Abstract Objectives We performed an environmental sampling study to investigate the environmental contamination of SARS-CoV-2 by COVID-19 patients with prolonged PCR positive status of clinical samples. Methods We sampled the air from rooms for nine COVID-19 patients with illness or positive PCR > 30 days, before and after nasopharyngeal/oropharyngeal swabbing and before and after nebulization treatment. We also sampled patients’ surroundings and healthcare workers’ personal protection equipment (PPE) in a non-ICU ward. SARS-CoV-2 was detected by PCR. Results Eighty-eight samples were collected from high-touch surfaces and floors in patient rooms and toilets, with only the bedsheets of two patients and one toilet positive for SARS-CoV-2. All air samples (n = 34) were negative for SARS-CoV-2. Fifty-five samples collected from PPE were all negative. Conclusion Contamination of near-patient surroundings was uncommon for COVID-19 patients with prolonged PCR positive status if environmental cleaning/disinfection were performed rigorously. Airborne transmission of SARS-CoV-2 was unlikely in these non-ICU settings.

Small cell carcinoma (SCC)/neuroendocrine prostate cancer (NEPC) is a rare and highly aggressive subtype of prostate cancer associated with an AR(androgen receptor)-null phenotype and visceral metastases. This study presents a 44-year-old man originally diagnosed with metastatic hormone-sensitive prostatic adenocarcinoma. After 6-month androgen deprivation therapy (ADT) combined with docetaxel, the patient developed paraplegia. Laminectomy was performed, and a thoracic vertebral biopsy revealed neuroendocrine differentiation and mixed adenocarcinoma. The patient developed liver metastases and experienced stable disease for 4 months following etoposide combined with cisplatin and pembrolizumab. Seminal vesicle biopsy after chemotherapy revealed small-cell cancer. The prostate biopsy specimen also indicated pure SCC. We witnessed the dynamic evolution from pure adenocarcinoma to fully differentiated SCC, leading to obstruction and death. In addition, whole-exome sequencing was performed on both biopsy specimens of the thoracic vertebra at the beginning of castration resistance and that of seminal vesicle after multiple lines of treatment failure. Utilizing phylogenetic reconstruction, we observed that both samples shared a common ancestor clone harboring aberrations in the TP53, RB1, and NF2 genes. We also discovered that driver events in the private subclones of both samples, such as alterations in CDC27 and RUNX1, might have played a significant role in tumor progression or even neuroendocrine differentiation. Tumor biopsy and IHC assessment must be repeated at different stages of progression, because of intrapatient spatial and temporal heterogeneity of adenocarcinoma versus SCC/NEPC. Although, typical treatments including ADT, docetaxel, etoposide, cisplatin, and pembrolizumab provided temporary response, the patient still had a poor prognosis.

Parkinson’s disease (PD) is the most common neurodegenerative movement disorder, and it is characterized by the selective loss of dopaminergic (DA) neurons in the substantia nigra pars compacta (SNpc), as well as the presence of intracellular inclusions with α-synuclein as the main component in surviving DA neurons. Emerging evidence suggests that the imbalance of proteostasis is a key pathogenic factor for PD. Endoplasmic reticulum (ER) stress-induced unfolded protein response (UPR) and autophagy, two major pathways for maintaining proteostasis, play important roles in PD pathology and are considered as attractive therapeutic targets for PD treatment. However, although ER stress/UPR and autophagy appear to be independent cellular processes, they are closely related to each other. In this review, we focused on the roles and molecular cross-links between ER stress/UPR and autophagy in PD pathology. We systematically reviewed and summarized the most recent advances in regulation of ER stress/UPR and autophagy, and their cross-linking mechanisms. We also reviewed and discussed the mechanisms of the coexisting ER stress/UPR activation and dysregulated autophagy in the lesion regions of PD patients, and the underlying roles and molecular crosslinks between ER stress/UPR activation and the dysregulated autophagy in DA neurodegeneration induced by PD-associated genetic factors and PD-related neurotoxins. Finally, we indicate that the combined regulation of ER stress/UPR and autophagy would be a more effective treatment for PD rather than regulating one of these conditions alone.

Hydrogen is one of the exhaust gases produced by nuclear power stations. Due to the potential danger of incomplete combustion and the emission of hydrogen, hydrogen catalytic combustion is introduced to ensure the safety of nuclear power stations. Palladium is a widely used catalyst for hydrogen catalytic combustion. H2 catalytic combustion on a Pd(111) slab model and Pd38 cluster model were simulated using density functional theory (DFT), in order to analyze the H2 oxidation mechanism on the catalyst surface.

Objective: To assess the feasibility of single-port transperitoneal robotic-assisted laparoscopic radical prostatectomy (spRALP) and discuss its surgical technique. Methods: A 60-year-old male was admitted with an elevated prostate-specific antigen (PSA) level of 13.89 ng/mL and confirmed with prostate cancer on biopsy showing three of 22 positive cores with a Gleason score of 3 + 4 = 7. Multiparametric magnetic resonance (MR) and bone scintigraphy showed organ-confined disease. spRALP was performed using da Vinci Si HD surgical system, with access of a quadri-channel laparoscopic port placed supraumbilically. Two drainage tubes were placed before wound closure. The surgical procedure was largely in consistence with a conventional robotic-assisted laparoscopic radical prostatectomy. Results: The surgery was successfully carried out with a duration of 152 min and an estimated blood loss of 100 mL. The patient was discharged on postoperative Day 4 after removal of both pelvic drainage tubes. Foley catheter was removed on postoperative Day 14. No major complications were encountered. Postoperative pathology showed a Gleason score of 3 + 4 = 7 with no extraprostatic extension and negative surgical margins. Conclusion: Single-port robotic prostatectomy is feasible using the currently available robotic instruments in most Chinese robotic urological centers. Meticulous preoperative planning and careful patient selection are mandatory. Further studies concerning perioperative complications and pentafecta outcome compared with the conventional multi-port robotic prostatectomy is required. Objectives: Tuberculosis (TB) remains a global public health problem. Recent studies have implicated long non-coding RNAs and their variants as possibly playing important roles in TB. The aim of this study was to assess the clinical relevance of lnc-AC145676.2.1-6 and lnc-TGS1-1 and their variants in a western Chinese population. Methods: This case–control study included 467 TB patients and 473 healthy controls from West China Hospital. The expression levels of lnc-AC145676.2.1-6 and lnc-TGS1-1 were analyzed by reverse transcriptase quantitative real-time PCR. Single-nucleotide polymorphism genotyping was performed using a custom-designed 2 × 48-Plex SNPscan kit. Results: It was observed that lnc-AC145676.2.1-6 and lnc-TGS1-1 expression levels were both obviously down-regulated in TB patients. In addition, a lower expression level of lnc-TGS1-1 was associated with the presence of thrombocytopenia in TB patients during anti-TB treatment, and the homozygous CC genotype of rs4737420 correlated with a decreased risk of leukopenia, compared with individuals with the T allele (TT/CT genotype), in the dominant mode. Conclusions: For the first time, potential TB-associated promoting effects were identified for the decreased expression levels of lnc-AC145676.2.1-6 and lnc-TGS-1, while lnc-TGS1-1 and its variant rs4737420 may be predictive indicators of anti-TB drug-induced adverse drug reactions. Background Tuberculosis (TB) is difficult to diagnose under complex clinical conditions as electronic health records (EHRs) are often inadequate in making an affirmative diagnosis. As exosomal miRNAs emerged as promising biomarkers, we investigated the potential of using exosomal miRNAs and EHRs in TB diagnosis. Methods: A total of 370 individuals, including pulmonary tuberculosis (PTB), tuberculous meningitis (TBM), non-TB disease controls and healthy state controls, were enrolled. Exosomal miRNAs were profiled in the exploratory cohort using microarray and miRNA candidates were selected in the selection cohort using qRT-PCR. EHRs and follow-up information of the patients were collected accordingly. miRNAs and EHRs were used to develop diagnostic models for PTB and TBM in the selection cohort with the Support Vector Machine (SVM) algorithm. These models were further evaluated in an independent testing cohort. Findings: Six exosomal miRNAs (miR-20a, miR-20b, miR-26a, miR-106a, miR-191, miR-486) were differentially expressed in the TB patients. Three SVM models, ''EHR+miRNA'', ''miRNA only'' and ''EHR only'' were compared, and ''EHR + miRNA'' model achieved the highest diagnostic efficacy, with an AUC up to 0.97 (95% CI 0.80–0.99) in TBM and 0.97 (0.87–0.99) in PTB, respectively. However, ''EHR only'' model only showed an AUC of 0.67 (0.46–0.83) in TBM. After 2-month anti-tuberculosis therapy, overexpressed miRNAs presented a decreased expression trend (p= 4.80 × 10−5). Interpretation: Our results showed that the combination of exosomal miRNAs and EHRs could potentially improve clinical diagnosis of TBM and PTB. Introduction Patients with locally advanced prostate cancer are at high risk of recurrence after definitive treatment. There are emerging data that radical prostatectomy can delay the progression of castration resistance and potentially prolong survival. Neoadjuvant radiation therapy improves local control and has shown survival benefit with favourable toxicity profiles in several other malignancies. We have designed this trial to investigate whether this combination, which theoretically maximises local control, is a safe and feasible approach for treating locally advanced prostate cancer.Methods and analysis This study is a phase I, open-label study to investigate the safety and feasibility of neoadjuvant hormone and radiation therapy followed by robot-assisted radical prostatectomy by a traditional 3+3 dose-escalation design with four planned radiation dose levels (39.6 Gy/22F, 45 Gy/25F, 50.4 Gy/28F and 54 Gy/30F). Locally advanced prostate cancer patients with positive pelvic and/or retroperitoneal lymph nodes will be recruited. The primary objective is to determine the adverse events and maximal tolerable dose (MTD) of neoadjuvant radiotherapy. Toxicity will be assessed using the National Cancer Institute Common Toxicity Criteria V.5.0.Ethics and dissemination This protocol was approved by the Institutional Review Board of Shanghai Changhai Hospital (ref. CHEC2019-070 and CHEC2019-082). The study will be performed in compliance with applicable local legislation and in accordance with the ethical principles developed by the World Medical Association in the Declaration of Helsinki 2013. Background: Factors influencing the kidney function of patients after renal transplantation include both recipient-related factors and donor-related factors. To gain a better understanding of these factors and to improve clinical decision-making, we performed a retrospective study of southwestern Chinese people receiving kidney transplantation.Methods: In this retrospective analysis, a total of 2,462 recipients receiving allogeneic kidney transplantation in West China Hospital of Sichuan University from December 13, 2008 to January 10, 2018 were included. Data on recipient and donor characteristics were extracted from the Transplant Center Database and stratified by discrete time points after kidney transplantation. Univariate and multivariate logistic regression analyses were carried out on the study variables, and kidney function of postoperative patients was monitored using cystatin C (CysC) as the outcome indicator.Results: From the univariate analysis, several factors showed statistically significant short-term impact on kidney function based on CysC after kidney transplantation, including age, ethnicity, body mass index (BMI), and HLA A-B-DR-DQ loci mismatch. Gender of recipients and gender-consistency between donors and recipients revealed both short-term and long-term influence. Younger donors had significantly better medium-and-long-term influence on kidney function. From the multivariate logistic regression analysis, recipient gender, ethnicity, BMI, and donor age were independent factors affecting postoperative CysC recovery at discrete time points.Conclusion: Several factors of recipients related to renal function after kidney transplantation, such as gender, ethnicity, BMI and donor's age should be paid more attention to. Moreover, female and non-Han recipients decreased the risk of poor outcome during postoperative kidney function recovery while large BMI of recipients and higher donor age increased the risk. It is useful to predict the postoperative renal function earlier according to corresponding factors, and improve the patient's quality of life.

Abstract In order to identify the potential reaction paths of C2H4 and their product distribution in Fischer‐Tropsch synthesis (FTS), a series of experiments were designed over a Co/TiO2 catalyst in the absence of CO. C2H4 did quickly react with H2 to produce C1‐6 products under Fischer‐Tropsch (FT) reaction conditions. Although the dominant reaction is C2H4 hydrogenation to ethane, changing the reaction conditions (temperature and partial pressure of reactants) can lead to the other reaction pathways being enhanced, resulting in varying product selectivity to both linear and branch olefins and paraffins. Possible reaction pathways had been summarized and discussed, which including C2H4 reaction to ethylidene followed by dimerization; C2H4 insertion into C2 surface species and dimerization and C4 decomposition and/or direct C2 hydrogenolysis. Furthermore, the products obtained from C2H4 reactions were fit to a typical FTS product distribution, which indicate that both the chain growth initiators and monomers are not necessarily only derived from hydrogenation of CO but also from the secondary reactions of olefins.

In the era of big data, it is challenging to efficiently retrieve the required images from the vast amount of data. Therefore, a content-based image retrieval system is an important research direction to address this problem. Furthermore, a multi-feature-based image retrieval system can compensate for the shortage of a single feature to a certain extent, which is essential for improving retrieval system performance. Feature selection and feature fusion strategies are critical in the study of multi-feature fusion image retrieval. This paper proposes a multi-feature fusion image retrieval strategy with adaptive features based on information entropy theory. Firstly, we extract the image features, construct the distance function to calculate the similarity using the information entropy proposed in this paper, and obtain the initial retrieval results. Then, we obtain the precision of single feature retrieval based on the correlation feedback as the retrieval trust and use the retrieval trust to select the effective features automatically. After that, we initialize the weights of selected features using the average weights, construct the probability transfer matrix, and use the PageRank algorithm to update the initialized feature weights to obtain the final weights. Finally, we calculate the comprehensive similarity based on the final weights and output the detection results. This has two advantages: (1) the proposed strategy uses multiple features for image retrieval, which has better performance and more substantial generalization than the retrieval strategy based on a single feature; (2) compared with the fixed-feature retrieval strategy, our method selects the best features for fusion in each query, which takes full advantages of each feature. The experimental results show that our proposed method outperforms other methods. In the datasets of Corel1k, UC Merced Land-Use, and RSSCN7, the top10 retrieval precision is 99.55%, 88.02%, and 88.28%, respectively. In the Holidays dataset, the mean average precision (mAP) was 92.46%.

Objective There have been no recent improvements in the glioblastoma multiforme (GBM) outcome, with median survival remaining 15 months. Consequently, the need to identify novel biomarkers for GBM diagnosis and prognosis, and to develop targeted therapies is high. This study aimed to establish biomarkers for GBM pathogenesis and prognosis. Methods In total, 220 overlapping differentially expressed genes (DEGs) were obtained by integrating four microarray datasets from the Gene Expression Omnibus database (GSE4290, GSE12657, GSE15824, and GSE68848). Then a 140-node protein–protein interaction network with 343 interactions was constructed. Results The immune response and cell adhesion molecules were the most significantly enriched functions and pathways, respectively, among DEGs. The designated hub genes ITGB5 and RGS4 , which have a high degree of connectivity, were closely correlated with patient prognosis, and GEPIA database mining further confirmed their differential expression in GBM versus normal tissue. We also determined the 20 most appropriate small molecules that could potentially reverse GBM gene expression, Prestwick-1080 was the most promising and had the highest negative scores. Conclusions This study identified ITGB5 and RGS4 as potential biomarkers for GBM diagnosis and prognosis. Insights into molecular mechanisms governing GBM occurrence and progression will help identify alternative biomarkers for clinical practice.

ABSTRACT The contamination of patients’ surroundings by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) remains understudied. We sampled the surroundings and the air of six negative-pressure non-intensive care unit (non-ICU) rooms in a designated isolation ward in Chengdu, China, that were occupied by 13 laboratory-confirmed coronavirus disease 2019 (COVID-19) patients who had returned from overseas travel, including 2 asymptomatic patients. A total of 44 of 112 (39.3%) surface samples were positive for SARS-CoV-2 as detected by real-time PCR, suggesting extensive contamination, although all of the air samples were negative. In particular, in a single room occupied by an asymptomatic patient, four sites were SARS-CoV-2 positive, highlighting that asymptomatic COVID-19 patients do contaminate their surroundings and impose risks for others with close contact. Placement of COVID-19 patients in rooms with negative pressure may bring a false feeling of safety, and the importance of rigorous environment cleaning should be emphasized. IMPORTANCE Although it has been well recognized that the virus SARS-CoV-2, the causative agent of COVID-19, can be acquired by exposure to fomites, surprisingly, the contamination of patients’ surroundings by SARS-CoV-2 is largely unknown, as there have been few studies. We performed an environmental sampling study for 13 laboratory-confirmed COVID-19 patients and found extensive contamination of patients’ surroundings. In particular, we found that asymptomatic COVID-19 patients contaminated their surroundings and therefore imposed risks for other people. Environment cleaning should be emphasized in negative-pressure rooms. The findings may be useful to guide infection control practice to protect health care workers.

Session-based recommendations aim to predict a user’s next click based on the user’s current and historical sessions, which can be applied to shopping websites and APPs. Existing session-based recommendation methods cannot accurately capture the complex transitions between items. In addition, some approaches compress sessions into a fixed representation vector without taking into account the user’s interest preferences at the current moment, thus limiting the accuracy of recommendations. Considering the diversity of items and users’ interests, a personalized interest attention graph neural network (PIA-GNN) is proposed for session-based recommendation. This approach utilizes personalized graph convolutional networks (PGNN) to capture complex transitions between items, invoking an interest-aware mechanism to activate users’ interest in different items adaptively. In addition, a self-attention layer is used to capture long-term dependencies between items when capturing users’ long-term preferences. In this paper, the cross-entropy loss is used as the objective function to train our model. We conduct rich experiments on two real datasets, and the results show that PIA-GNN outperforms existing personalized session-aware recommendation methods.

This article investigates the feasibility of interconnecting LCC-HVDC and VSC-HVDC through an un-interrupted DC autotransformer (DC AUTO). The interconnected system aims at taking advantage of two HVDC technologies. To begin with, the system configuration and the potential benefits are illustrated. Then, the corresponding control strategies towards LCC-HVDC, VSC-HVDC, and the un-interrupted DC AUTO are proposed. The installed controllers enable the system stable operation during both normal time and faulted conditions. Finally, a four-terminal test system is built on PSCAD/EMTDC platform to validate the technical feasibility of the interconnected system and the effectiveness of the proposed control strategy.

To integrate bulk wind power, using modular multilevel converter (MMC) based DC grids is an effective solution to improve the flexibility and reliability of the power grid. This paper analyses a bipolar four-terminal MMC based DC grid to integrate wind farms with overhead lines. Since the overhead lines are prone to DC faults, the hybrid MMC is adopted in the system to ride through DC faults. The fault ride-through control strategies are designed, which remain continuous operation of wind turbines under DC faults. Besides, an adaptive dissipation control strategy associated with chopper resistances is designed to absorb surplus energy. Finally, simulations based on PSCAD/EMTDC platform verify the fault ride through control of hybrid MMC based DC grid and proves the effectiveness of the energy dissipation strategies.

DC grid has a prospect of application in collecting and transmitting large-scale renewable energy over long distance. However, the randomness and fluctuation of the renewable power may hamper the stable operation of DC grid. Energy storage device is able to actively absorb or supplement active power, which can be used to smooth the power fluctuation. The paper investigates a DC grid topology with energy storage and proposes a coordinated control strategy. The proposed strategy enables coordinated control of the renewable energy output power, pumped storage power and the grid consuming power. During the steady state, the control strategy enables the system DC voltage stable and power flow of the grid under zero-error control. During the faulted time, the coordinate control strategy enables the converter fast response and maintains the system DC voltage within safety margin. The effectiveness of the coordinated control strategy is verified by simulation in PSCAD/EMTDC.

Bluetooth positioning is an important and challenging topic in indoor positioning. Although a lot of algorithms have been proposed for this problem, it is still not solved perfectly because of the instable signal strengths of Bluetooth. To improve the performance of Bluetooth positioning, this article proposes a coarse-to-fine positioning method based on weighted K-nearest neighbors and adaptive bandwidth mean shift. The method first employs weighted K-nearest neighbors to generate multi-candidate locations. Then, the testing position is obtained by applying adaptive bandwidth mean shift to the multi-candidate locations, which is used to search for the maximum density of the candidate locations. Experimental result indicates that the proposed method improves the performance of Bluetooth positioning.

With the rapid development of information storage technology and the spread of the Internet, large capacity image databases that contain different contents in the images are generated. It becomes imperative to establish an automatic and efficient image retrieval system. This paper proposes a novel adaptive weighting method based on entropy theory and relevance feedback. Firstly, we obtain single feature trust by relevance feedback (supervised) or entropy (unsupervised). Then, we construct a transfer matrix based on trust. Finally, based on the transfer matrix, we get the weight of single feature through several iterations. It has three outstanding advantages: (1) The retrieval system combines the performance of multiple features and has better retrieval accuracy and generalization ability than single feature retrieval system; (2) In each query, the weight of a single feature is updated dynamically with the query image, which makes the retrieval system make full use of the performance of several single features; (3) The method can be applied in two cases: supervised and unsupervised. The experimental results show that our method significantly outperforms the previous approaches. The top 20 retrieval accuracy is 97.09%, 92.85%, and 94.42% on the dataset of Wang, UC Merced Land Use, and RSSCN7, respectively. The Mean Average Precision is 88.45% on the dataset of Holidays.

Atrial septal defect (ASD) is the third most frequent type of congenital heart anomaly, featuring shunting of blood between the two atria. Gene-environment interaction remains to be an acknowledged cause for ASD occurrence. A recent European genome-wide association study (GWAS) of congenital heart disease (CHD) identified 3 susceptibility SNPs at chromosome 4p16 associated with ASD: rs870142, rs16835979 and rs6824295. A Chinese-GWAS of CHD conducted in the corresponding period did not reveal the 3 susceptibility SNPs, but reported 2 different risk SNPs: rs2474937 and rs1531070. Therefore, we aimed to investigate the associations between the 3 European GWAS-identified susceptibility SNPs and ASD risk in the Han population in southwest China. Additionally, to increase the robustness of our current analysis, we conducted a meta-analysis combining published studies and our current case-control study. We performed association, linkage disequilibrium, and haplotype analysis among the 3 SNPs in 190 ASD cases and 225 age-, sex-, and ethnicity-matched healthy controls. Genotype and allele frequencies among the 3 SNPs showed statistically significant differences between the cases and controls. Our study found that individuals carrying the allele T of rs870142, the allele A of rs16835979, and the allele T of rs6824295 had a respective 50.1% (odds ratio (OR) = 1.501, 95% confidence interval (CI) = 1.122-2.009, PFDR-BH = 0.018), 48.5% (OR = 1.485, 95%CI = 1.109-1.987, PFDR-BH = 0.012), and 38.6% (OR = 1.386, 95%CI = 1.042-1.844, PFDR-BH = 0.025) increased risk to develop ASD than wild-type allele carriers in our study cohort. In the haplotype analysis, we identified a disease-risk haplotype (TAT) (OR = 1.540, 95%CI = 1.030-2.380, PFDR-BH = 0.016). Our meta-analysis also showed that the investigated SNP was associated with ASD risk (combined OR (95%CI) = 1.35 (1.24-1.46), P < 0.00001). Our study provides compelling evidence to motivate better understanding of the etiology of ASD.

Lipid synthesis is required for cell growth and is subject to pharmacologic regulation. Keratinocyte growth factor (KGF) stimulates proliferation and lipogenesis in H292 cells, a pulmonary epithelial cancer cell line, but the signaling pathways are not known. KGF stimulated the expression of the transcription factors sterol-regulatory element binding protein-1 (SREBP-1), CCAAT/enhancer binding protein α (C/EBPα), and C/EBPδ and two key enzymes involved in lipogenesis, FAS and stearoyl coenzyme A desaturase-1 (SCD-1). We found that KGF induced rapid activation of Akt, p70 S6K, JNK, and extracellular signal-regulated (ERK). Induction of SREBP-1, SCD-1, and FAS by KGF was inhibited by the JNK inhibitor SP600125 and the phosphatidylinositol 3-kinase (PI3K) inhibitor LY294002 but not by the ERK inhibitor PD98059. Using FAS and SCD-1-luciferase promoter constructs, we observed that KGF stimulated the transcription of these promoters and that exogenous cholesterol inhibited the induction. Mutation of the SREBP-1 binding site in the SCD-1 promoter abolished the effect of KGF on SCD-1 transcription. In addition, overexpression of active SREBP-1 directly stimulated SCD-1 and FAS. Conversely, adenovirus-mediated overexpression of a dominant negative form of SREBP-1 inhibited the KGF effect on FAS and SCD-1 expression.In summary, we conclude that KGF requires both PI3K and JNK signaling pathways to induce SREBP-1, which in turn induces SCD-1 and FAS expression in H292 cells.

Face verification for unrestricted faces in the wild is a challenging task. This paper proposes a method based on two deep convolutional neural networks (CNN) for face verification. In this work, we explore using identification signals to supervise one CNN and the combination of semi-verification and identification to train the other one. In order to estimate semi-verification loss at a low computation cost, a circle, which is composed of all faces, is used for selecting face pairs from pairwise samples. In the process of face normalization, we propose using different landmarks of faces to solve the problems caused by poses. In addition, the final face representation is formed by the concatenating feature of each deep CNN after principal component analysis (PCA) reduction. Furthermore, each feature is a combination of multi-scale representations through making use of auxiliary classifiers. For the final verification, we only adopt the face representation of one region and one resolution of a face jointing Joint Bayesian classifier. Experiments show that our method can extract effective face representation with a small training dataset and our algorithm achieves 99.71% verification accuracy on Labeled Faces in the Wild (LFW) dataset.

Compelling studies have implicated that the Wnt signaling pathway plays an important role in the development and progression of tuberculosis, however, there is little literature addressing the role of polymorphisms in Wnt pathway on tuberculosis. We took a pathway based candidate gene approach to investigate the possible correlation between genetic variants in Wnt pathway and tuberculosis. Three single nucleotide polymorphisms (SNPs) in Wnt pathway (rs4135385 in CTNNB1 gene, rs7832767 in SFRP1 gene, and rs11079571 in AXIN2 gene) were genotyped in 422 Chinese Han tuberculosis patients and 402 frequency matched (age, gender, and ethnicity) controls using high-resolution melting analysis. The genotype and allelic frequencies of rs4135385 and rs7832767 were significantly different among patients and controls. The dominant model of rs4135385 was significantly associated with an increased risk of tuberculosis (AG/GG versus AA: OR = 1.49, 95% CI = 1.06-2.09, p = 0.019). The recessive model of rs7832767 posed a significant higher risk for tuberculosis (TT versus TC/CC, OR = 2.70, 95% CI = 1.41-5.18, p = 0.002). These SNPs were further evaluated whether they were correlated with the site of tuberculosis and the level of inflammatory markers. Rs7832767 was significantly associated with the level of CRP (p = 0.014), and the patients carrying T allele might present with elevated CRP values (OR = 1.90, 95% CI = 1.21-2.96, p = 0.005). Our study provided the first evidence that rs4135385 and rs7832767 were associated with tuberculosis risk, and genetic variants in Wnt signaling pathway might participate in genetic susceptibility to tuberculosis in Chinese Han population. Further epidemiological and functional studies in larger populations are warranted to verify our results.

BACKGROUND: Non-reactive platelet counts elevation occurs mainly in myeloproliferative disorders (MPDs), which have been reported to be closely associated with JAK2 V617F mutation. Complete blood cell count (CBC) is essential in diagnosis of MPDs, however, the impact of JAK2 V617F mutation on the patients' hemogram variation remains not clear. METHODS: JAK2 V617F mutation was detected by allele specific real-time quantitative fluorescence PCR (AS-qPCR). RESULTS: Of the 402 non-reactive platelet elevating patients, JAK2 V617F mutation was detected in 222 (55.2%) patients. RBC counts, WBC counts, platelet-large contrast ratio (P-LCR), platelet distribution width (PDW) and mean platelet volume (MPV) were much higher in JAK2 V617F mutated patients, except platelet counts. In addition, when the patients were classified into subgroups by blood cell counts, it was found that JAK2 V617F mutation rate increased progressively with the increase of RBC counts and WBC counts, other than platelet counts. Furthermore, trilineage hyperplasia group showed highest JAK2 V617F mutation rate (93.26%), followed by the bilineage hyperplasia groups. Lastly, JAK2 V617F mutant allele burden was found much higher in polycythemia vera (PV) patients [median(P25-P75): 45.02%(35.12%-54.22%)] than in essential thrombocythemia (ET) patients [median(P25-P75): 28.23%(17.77%-41.66%)], and that it increased with WBC counts (r = 0.393, p = 0.000) and RBC counts(r = 0.215, p = 0.001), other than platelet counts (r = -0.051, p = 0.452). Further analysis revealed that in ET patients, JAK2 V617F mutant allele burden correlated with WBC counts and platelet counts positively, other than RBC counts, while in PV patients, it correlated with WBC counts and RBC counts positively, but not platelet counts. CONCLUSIONS: JAK2 V617F mutation occurs frequently in patients with non-reactive elevated platelet counts. The presence of JAK2 V617F mutation has great impact on hemogram variation, including RBC counts, WBC counts, platelet parameters and lineage hyperplasia, but not on platelet counts. Besides, JAK2 V617F mutant allele burden affects the blood cell proliferation pattern.

Members of the genus Xenorhabdus are entomopathogenic bacteria that associate with nematodes. The nematode-bacteria pair infects and kills insects, with both partners contributing to insect pathogenesis and the bacteria providing nutrition to the nematode from available insect-derived nutrients. The nematode provides the bacteria with protection from predators, access to nutrients, and a mechanism of dispersal. Members of the bacterial genus Photorhabdus also associate with nematodes to kill insects, and both genera of bacteria provide similar services to their different nematode hosts through unique physiological and metabolic mechanisms. We posited that these differences would be reflected in their respective genomes. To test this, we sequenced to completion the genomes of Xenorhabdus nematophila ATCC 19061 and Xenorhabdus bovienii SS-2004. As expected, both Xenorhabdus genomes encode many anti-insecticidal compounds, commensurate with their entomopathogenic lifestyle. Despite the similarities in lifestyle between Xenorhabdus and Photorhabdus bacteria, a comparative analysis of the Xenorhabdus, Photorhabdus luminescens, and P. asymbiotica genomes suggests genomic divergence. These findings indicate that evolutionary changes shaped by symbiotic interactions can follow different routes to achieve similar end points.

A computer code has been developed to simulate and study the evolution of ion charge states inside the trap region of an electron beam ion trap. In addition to atomic physics phenomena previously included in similar codes such as electron impact ionization, radiative recombination, and charge exchange, several aspects of the relevant physics such as dielectronic recombination, ionization heating, and ion cloud expansion have been included for the first time in the model. The code was developed using object oriented concepts with database support, making it readable, accurate, and well organized. The simulation results show a good agreement with various experiments, and give useful information for selection of operating conditions and experiment design.
