Abstract Metal-organic frameworks (MOFs) with diverse chemistry, structures, and properties have emerged as appealing materials for miniaturized solid-state devices. The incorporation of MOF films in these devices, such as the integrated microelectronics and nanophotonics, requires robust patterning methods. However, existing MOF patterning methods suffer from some combinations of limited material adaptability, compromised patterning resolution and scalability, and degraded properties. Here we report a universal, crosslinking-induced patterning approach for various MOFs, termed as CLIP-MOF. Via resist-free, direct photo- and electron-beam (e-beam) lithography, the ligand crosslinking chemistry leads to drastically reduced solubility of colloidal MOFs, permitting selective removal of unexposed MOF films with developer solvents. This enables scalable, micro-/nanoscale (≈70 nm resolution), and multimaterial patterning of MOFs on large-area, rigid or flexible substrates. Patterned MOF films preserve their crystallinity, porosity, and other properties tailored for targeted applications, such as diffractive gas sensors and electrochromic pixels. The combined features of CLIP-MOF create more possibilities in the system-level integration of MOFs in various electronic, photonic, and biomedical devices.

Abstract Background Light is a critical factor in plant growth and development, particularly in controlled environments. Light-emitting diodes (LEDs) have become a reliable alternative to conventional high pressure sodium (HSP) lamps because they are more efficient and versatile in light sources. In contrast to well-known specialized LED light spectra for vegetables, the appropriate LED lights for crops such as cotton remain unknown. Results In this growth chamber study, we selected and compared four LED lights with varying percentages (26.44%–68.68%) of red light (R, 600–700 nm), combined with other lights, for their effects on growth, leaf anatomy, and photosynthesis of cotton seedlings, using HSP lamp as a control. The total photosynthetic photon flux density (PPFD) was (215 ± 2) μmol·m−2·s−1 for all LEDs and HSP lamp. The results showed significant differences in all tested parameters among lights, and the percentage of far red (FR, 701–780 nm) within the range of 3.03%–11.86% was positively correlated with plant growth (characterized by leaf number and area, plant height, stem diameter, and total biomass), palisade layer thickness, photosynthesis rate (P n), and stomatal conductance (G s). The ratio of R/FR (4.445–11.497) negatively influenced the growth of cotton seedlings, and blue light (B) suppressed stem elongation but increased palisade cell length, chlorophyll content, and P n. Conclusion The LED 2 was superior to other LED lights and HSP lamp. It had the highest ratio of FR within the total PPFD (11.86%) and the lowest ratio of R/FR (4.445). LED 2 may therefore be used to replace HPS lamp under controlled environments for the study of cotton at the seedling stage.

Chilo sacchariphagus (Lepidoptera: Pyralidae) is an economically important sugarcane pest. Although numerous studies were conducted on the physiological responses in C. sacchariphagus, little is known regarding the genes regulating these physiological processes. Gene expression analysis by qRT-PCR can offer a significant indication for functional gene studies. To our knowledge, the reference genes of C. sacchariphagus have not been screened or evaluated, which hinders the functional gene study. In the present study, the stability of seven reference genes (β-ACT, GAPDH, BTF3, 28S, RPL7, EF1α, and SDHA) was evaluated in C. sacchariphagus under different experimental conditions, including tissues (antenna, head, thorax, abdomen, leg, and wing), temperatures (4 °C, 25 °C, and 37 °C) and sexes (male and female), through RefFinder, which integrates four algorithms (Normfinder, BestKeeper, ΔCt method, and geNorm). The findings suggested that the combination of β-ACT and RPL7 is ideal to analyze gene expressions in different tissues and at distinct temperatures, and EF1α and SDHA were suitable reference genes for comparing gene expressions between sexes. Finally, the expression profiles of CsacPBP1 gene were evaluated, and the outcomes further confirm the importance of selecting fitting reference genes for normalization of qRT-PCR data. This study represents the first kind in screening out suitable reference genes for gene expression analysis in C. sacchariphagus. Information from this study is poised to galvanize future inquiry into the gene expression of C. sacchariphagus, an economically important pest of sugarcane.

Using creativity to promote recreational services is crucial. Accordingly, creative linguistic landscapes (CLLs) are being used to improve visitors' experiences in some recreational zones. However, relevant research is still in its early stages. Therefore, this study was conducted. It summarized the leisure function categories and function evaluation indicators of CLLs in recreational zones respectively based on image materials and related online reviews. The leisure function outcomes of all CLL types were ranked using the fuzzy PROMETHEE method; based on this ranking, a CLL configuration optimization mode was suggested. The findings reveal the following. (1) Currently, there are mainly nine leisure function types of CLL in practice, although the type structure is severely imbalanced; there are 12 primary corresponding function evaluation indicators, although each of them draws significantly different attention. (2) There are notable variations among the outcomes of different types of functions of CLL: mood adjustment is the most advantageous function of CLL for leisure services, followed by emotional guidance and cognitive building functions; (3) According to the study findings, in the configuration of CLL, which aims at leisure function optimization, the "function focusing and coordinating mode (the superior functions of CLL are focused on and its various functions are coordinated)" should be adopted. The results provide meaningful lessons for the establishment of rational and effective CLL in recreational zones.

The increasing worldwide demand for traditional herbs has been met by growing cultivated herbs. It is undoubtedly very important to seek a reasonable cultivation mode for the yield, quality and long-term production stability of traditional herbs. Glycyrrhiza is a commercial medicinal crop that is widely cultivated with high water and fertilizer application, in the arid areas of northwest China. Rational irrigation and fertilizer application are key factors for successful crop management. Studying water–fertilizer coupling effects in a drip irrigation system is critical for sustainable agricultural development in arid areas, such as that of Xinjiang in China, to find out the optimized water and fertilizer management. Therefore, a four-year field experiment was conducted to find out how the combination of four levels of irrigation quotas and four levels of fertilizer amounts would affect the different parts liquiritin contents of the licorice, data on different parts liquiritin contents of the licorice were collected from individual glycyrrhizin for each treatment during 2019 to 2022. The application of fertilizer significantly increased the liquiritin contents of the licorice under the same irrigation conditions. However, the liquiritin contents of the licorice were reduced in the excess water treatments (W4F1 and W4F4) and in the excess fertilizer treatments (W1F4 and W4F4) in contrast to the optimum W2F2 treatment. These results indicate that the combination of water and fertilizer management can potentially increase the accumulation the liquiritin contents of the licorice, which can be used to promote the cultivation of Glycyrrhiza.

Abstract Background Osteosarcoma (OS) mainly happens in children and youths. Surgery, radiotherapy and chemotherapy are the common therapies for osteosarcoma treatment but all their anti-tumor effects are limited. In recent years, a new cellular therapy, CAR-T, a cellular immunotherapy with genetically engineered T cells bearing chimeric antigen receptor targeting specific tumor-associated antigen, has been proved to be an effective therapy against acute lymphoblastic leukemia. Thus, CAR-T is a potentially effective therapy for osteosarcoma treatment. Methods A CAR gene targeting B7-H3 antigen was constructed into lentiviral vector through molecular biology techniques. Then, the CAR gene was transferred to T cells through lentiviral delivery system, and the CAR-T cells were largely expanded using in vitro culture technology. The in vitro anti-tumor effect of CAR-T cells was evaluated through Real Time Cell Analysis system (RTCA) and ELISA assay. The in vivo anti-tumor capabilities of CAR-T cells were evaluated using the patient-derived xenografts (PDX) model of osteosarcoma. Results The third-generation CAR-T cells we constructed could target the B7-H3 antigen, and the phenotype of CAR-T cells was consistent with normal T cells; The CAR-T cells showed superior antitumor effects both in vitro and in vivo. Conclusion Our study showed that B7-H3 targeted CAR-T cells had high anti-tumor efficacy against osteosarcoma both in vitro and in vivo, which proved that B7-H3 targeted CAR-T therapy is potentially effective for osteosarcoma treatment.

Abstract Background Potassium (K) deficiency has become a common field production problem following the widespread adoption of Bacillus thuringiensis (Bt) transgenic cotton (Gossypium hirsutum L.) worldwide. The purpose of this study was to clarify whether the introduction of Bt gene directly reduces the K-use efficiency of cotton to induce K deficiency. Results The cotton variety, Jihe 321 (wild type, WT) and its two Bt (Cry1Ac)-transgenic overexpression lines (OE-29317, OE-29312) were studied in field with low soil-test K+ (47.8 mg·kg−1). In the field with low soil-test K+, only OE-29317 had less biomass and K+ accumulation than the WT at some growth stages. Both Bt lines produced similar or even greater seed cotton yield than WT in the field. When the Bt gene (~ 70%) in OE-29317 and OE-29312 plants was silenced by virus-induced gene silencing (VIGS), the VIGS-Bt plants did not produce more biomass than VIGS-green fluorescent protein (control) plants. Conclusions The introduction of Bt gene did not necessarily hinder the K use efficiency of the cotton lines under this study.

Defoliation and accelerating ripening are important measures for cotton mechanization, and judging the time of defoliation and accelerating the ripening and harvest of cotton relies heavily on the boll opening rate, making it a crucial factor to consider. The traditional methods of cotton opening rate determination are time-consuming, labor-intensive, destructive, and not suitable for a wide range of applications. In this study, the relationship between the change rate of the vegetation index obtained by the unmanned aerial vehicle multi-spectrum and the ground boll opening rate was established to realize rapid non-destructive testing of the boll opening rate. The normalized difference vegetation index (NDVI) and green normalized difference vegetation index (GNDVI) had good prediction ability for the boll opening rate. NDVI in the training set had an R2 of 0.912 and rRMSE of 15.387%, and the validation set performance had an R2 of 0.929 and rRMSE of 13.414%. GNDVI in the training set had an R2 of 0.901 and rRMSE of 16.318%, and the validation set performance had an R2 of 0.909 and rRMSE of 15.225%. The accuracies of the models based on GNDVI and NDVI were within the acceptable range. In terms of predictive models, random forests achieve the highest accuracy in predictions. Accurately predicting the cotton boll opening rate can support decision-making for harvest and harvest aid spray timing, as well as provide technical support for crop growth monitoring and precision agriculture.

Abstract Background Cotton production in China is challenged by high labor input including manual topping (MT). Recently, to replace MT in the Xinjiang cotton region of China, mepiquat chloride (MC) was applied once more than the traditional multiple-application; this was designated as chemical topping (CT), but it is unclear whether the amount of irrigation needs to be adjusted to accommodate CT. Results The main plots were assigned to three drip irrigation amounts [300 (I1), 480 (I2), and 660 (I3) mm], and the subplots were assigned to the CT treatments [450 (MC1), 750 (MC2), and 1 050 (MC3) mL·hm−2 25% MC] with MT as a control that was performed after early bloom. The optimum drip irrigation amount for CT was explored based on leaf photosynthesis, chlorophyll fluorescence, biomass accumulation, and yield. There were significant influences of drip irrigation, topping treatments and their interaction on chlorophyll fluorescence characteristics, gas exchange parameters and biomass accumulation characteristics as well as yield. The combination of I2 and MC2 (I2MC2) performed best. Compared with I2MT, the net photosynthetic rate (Pn), stomatal conductance (Gs), transpiration rate (Tr), and photochemical quenching coefficient (qP) of I2MC2 significantly increased by 4.0%∼7.2%, 6.8%∼17.1%, 5.2%∼17.6%, and 4.8%∼9.6%, respectively, from the peak flowering to boll opening stages. Moreover, I2MC2 showed fast reproductive organ biomass accumulation and the highest seed cotton yield; the latter was 6.6%∼12.8% higher than that of I2MT. Further analysis revealed that a 25% MC emulsion in water (MCEW) application resulted in yield improvement by increasing Pn, φPSII, and qP to promote biomass accumulation and transport to reproductive organs. Conclusion The results showed that the 480 mm drip irrigation combined with 750 mL·hm−2 MC increased the rate of dry matter accumulation in reproductive organs by increasing Pn, φPSII, and qP to improve photosynthetic performance, thus achieving higher yield.

Patient-derived tumor xenograft (PDX)/organoid (PDO), driven by cancer stem cells (CSC), are considered the most predictive models for translational oncology. Large PDX collections reflective of patient populations have been created and used extensively to test various investigational therapies, including population-trials as surrogate subjects in vivo. PDOs are recognized as in vitro surrogates for patients amenable for high-throughput screening (HTS). We have built a biobank of carcinoma PDX-derived organoids (PDXOs) by converting an existing PDX library and confirmed high degree of similarities between PDXOs and parental PDXs in genomics, histopathology and pharmacology, suggesting "biological equivalence or interchangeability" between the two. Here we demonstrate the applications of PDXO biobank for HTS "matrix" screening for both lead compounds and indications, immune cell co-cultures for immune-therapies and engineering enables in vitro/in vivo imaging. This large biobank of >550 matched pairs of PDXs/PDXOs across different cancers could become powerful tools for the future cancer drug discovery.

Insect odorant-binding proteins (OBPs) are significant in binding and transporting odorants to specific receptors. Our previous study demonstrated that BminOBP3 exhibited a strong affinity with undecanol. However, the binding mechanism between them remains unknown. Here, using homology modeling and molecular docking, we found that the C-terminus (I116-P122), especially the hydrogenbonds formed by the last three amino acid residues (V120, F121, and P122) of the C-terminus, is essential for BminOBP3′s ligand binding. Mutant binding assays showed that the mutant T-OBP3 that lacks C-terminus (I116-P122) displayed a significant decrease in affinity to undecanol (Ki = 19.57 ± 0.45) compared with that of the wild-type protein BminOBP3 (Ki = 11.59 ± 0.51). In the mutant 3D2a that lacks F121 and P122 and the mutant V120A in which V120 was replaced by alanine, the bindings to undecanol were completely abolished. In conclusion, the C-terminus plays a crucial role in the binding interactions between BminOBP3 and undecanol. Based on the results, we discussed the ligand-binding process of BminOBP3.

Background: To explore the effect of continuous renal replacement therapy (CRRT) in patients with sepsis-associated acute kidney injury (SA-AKI) with the Acute Kidney Injury Network Classification III and its effect on inflammatory mediators and coagulation function. Methods: We evaluated 90 patients who were diagnosed with sepsis and treated at our hospital. Forty patients received CRRT (group A) and the remainder received routine therapy (group B). We compared the renal function indices, represented by blood urea nitrogen (BUN) and serum creatinine (Scr), the urinary levels of kidney injury molecule 1, and the curative effect indices between the two groups. The incidence of major adverse cardiovascular events was compared between both groups. Further, the therapeutic effect (total effective rate) was evaluated and compared. With the shift of social interaction to online venues, do women still conform to existing gender norms? This article examines the online performance of professional women in urban China and their interactions with workplace colleagues on WeChat, a popular Chinese social media app that is an important venue for workplace interaction. As interviews showed, workplace interactions on WeChat perpetuated traditional gender norms of hegemonic masculinity, and professional women accommodated to those existing gender norms by using particular “personas.” Three major personas were identified: one that emphasized professional identity and downplayed gender identity; another that accentuated femininity and downplayed professional identity; and a third that performed femininity to please male workplace supervisors by confirming their masculinity. Persona is used as an analytical term to capture the WeChat activities of these professional women because, compared to online self-presentation, the persona represented a strategic conformity to existing norms and the women distanced themselves from their assumed personas. The use of social media, therefore, reproduced and reinforced conformity to existing gender norms because the online gendered persona constituted a comprehensive and enduring constraint.

Ribonuclease P/MRP Subunit P40 (RPP40), a component of ribonuclease P and multimeric ribonuclease P complex, was reported as one of the promoting factors for the chemoresistance of acute myeloid leukemia and a recurrence predictor of early-stage triple-negative breast cancer. However, the functional role of RPP40 in uterine corpus endometrial carcinoma (UCEC) is unclear. In this study, comprehensive bioinformatic analyses were conducted to explore the predictive role of RPP40 on UCEC diagnosis and prognosis, as well as the underlying mechanism. Differential analyses of multiple databases showed that both messenger RNA (mRNA) and the protein expression of RPP40 were significantly upregulated in UCEC tumor tissues. Furthermore, the RPP40 mRNA expression level was significantly correlated with the clinicopathological characteristics of UCEC patients, including the clinical stage, primary therapy outcome, histological type, histologic grade, overall survival event, disease-specific survival event, and progression-free interval event. Receiver operating characteristic (ROC) analysis showed that RPP40 was a reliable predictor for UCEC diagnosis with an area under the curve (AUC) of 0.775, a sensitivity of 0.829, and a specificity of 0.719. Kaplan–Meier, Cox regression, and nomogram analyses showed that high RPP40 expression was an independent prognostic factor for the 1-year, 3-year, and 5-year survival of UCEC patients. In addition, the enrichment analysis of RPP40-associated differentially expressed genes and correlation analyses showed that the expression of RPP40 was correlated with the regulation of extracellular matrix and immune cell infiltration. In conclusion, the upregulation of RPP40 is significantly correlated with the poor survival and tumor microenvironment of UCEC, suggesting that RPP40 is a promising biomarker of poor prognosis and a potential target of chemotherapy or immunotherapy in UCEC.

K+ channels of the Shaker family have been shown to play crucial roles in K+ uptake and transport. Cotton (Gossypium hirsutum) is an important cash crop. In this study, the 24 Shaker family genes were identified in cotton. Phylogenetic analysis suggests that they were assigned to five clusters. Additionally, their chromosomal location, conserved motifs and gene structure were analyzed. The promoter of cotton Shaker K+ channel genes comprises drought-, low-temperature-, phytohormone-response elements, etc. As indicated by qRT-PCR (quantitative real-time PCR), cotton Shaker K+ channel genes responded to low K+ and NaCl, and especially dehydration stress, at the transcript level. Moreover, one of the Shaker K+ channel genes, GhKAT1aD, was characterized. This gene is localized in the plasma membrane and is predicted to contain six transmembrane segments. It restored the growth of the yeast mutant strain defective in K+ uptake, and silencing GhKAT1a via VIGS (virus-induced gene silencing) resulted in more severe symptoms of K+ deficiency in cotton leaves as well as a lower net K+ uptake rate. The results of this study showed the overall picture of the cotton Shaker K+ channel family regarding bioinformatics as well as the function of one of its members, which provide clues for future investigations of cotton K+ transport and molecular insights for breeding K+-efficient cotton varieties.

The use of 1,1-dimethyl-piperidinium chloride (DPC) in Xinjiang is one of the necessary measures for regulating cotton populations and optimizing canopy structure. However, mechanical spraying involves operation and energy consumption, which can alternatively be achieved via drip application. In the present study, to investigate the effect of drip application, two types of hydrophobic nano-released DPCs were used to regulate plant type at different dosages. DPC dripingation reduced plant height by more than 10 cm and plant width by more than 4 cm, and the effect improved with increasing concentration and times. The main effect of height control was at the 6th–10th nodes of the main stem, and the effect of width control was observed at the 1st and 2nd nodes of the fruit branch. The SPAD value was higher than that in the control group during the initial stages. An irregular downward trend was observed in the subsequent stages. The proline content was higher than that of the control and increased with higher concentrations of DPC. There was no significant difference in the soil DPC content at the different sites. The DPC content decreased by more than 30% at the seventh day after dripping, and the content was 2–4 μg·g−1 in each treatment. At 15 days after application, the soil DPC was lower than 1 μg·g−1 in all treatments. The number of harvested plants was more than 150,000 plants·hm−2, and the number of bolls per plant increased at least by one, compared with the control. The final yield of seed cotton increased by at least 300 kg·hm−2, with a maximum increase of 1672.01 kg·hm−2. Considering the different types of DPCs, nano-released types worked better than the conventional type in terms of plant height, plant width, degradation in soil and boll formation. Nano-released DPC can play an even better role than conventional DPC in chemical regulation with drip irrigation.

Thidiazuron (TDZ) is a widely used chemical defoliant in cotton and can stimulate the production of ethylene in leaves, which is believed to be the key factor in inducing leaf abscission. Ethephon (Eth) can also stimulate ethylene production in leaves, but it is less effective in promoting leaf shedding. In this study, the enzyme-linked immunosorbent assays (ELISA) and RNA-seq were used to determine specific changes at hormonal levels as well as transcriptomic mechanisms induced by TDZ compared with Eth. The TDZ significantly reduced the levels of auxin and cytokinin in cotton leaves, but no considerable changes were observed for Eth. In addition, TDZ specifically increased the levels of brassinosteroids and jasmonic acid in the leaves. A total of 13 764 differentially expressed genes that specifically responded to TDZ were identified by RNA-seq. The analysis of KEGG functional categories suggested that the synthesis, metabolism, and signal transduction of auxin, cytokinin, and brassinosteroid were all involved in the TDZ-induced abscission of cotton leaves. Eight auxin transport genes (GhPIN1-c_D, GhPIN3_D, GhPIN8_A, GhABCB19-b_A, GhABCB19-b_D, GhABCB2-b_D, GhLAX6_A, and GhLAX7_D) specifically responded to TDZ. The pro35S::GhPIN3a::YFP transgenic plants showed lower defoliation than the wild type treated with TDZ, and YFP fluorescence in leaves was almost extinguished after treatment with TDZ rather than Eth. This provides direct evidence that GhPIN3a is involved in the leaf abscission induced by TDZ. We found that 959 transcription factors (TFs) specifically responded to TDZ, and a co-expression network analysis (WGCNA) showed five hub TFs (GhNAC72, GhWRKY51, GhWRKY70, GhWRKY50, and GhHSF24) during chemical defoliation with TDZ. Our work sheds light on the molecular basis of TDZ-induced leaf abscission in cotton.

Green supply chain has developed rapidly due to the advocacy of ecological civilization, and choosing a proper green supplier is a crucial issue. Considering the fuzziness of evaluation information and the psychological states of decision makers (DMs) in selecting process, a novel TODIM based on prospect theory with q-rung orthopair fuzzy set (q-ROFS) is proposed. The novel TODIM concerns both the perceived transformed probability weighting function and the differences in risk attitudes. A new distance, which concerns the herd mentality, is carried out to measure the perceived difference of the q-ROFS. Besides, a new systematic evaluation index system, named as PCEM (Product, Cooperation ability, Environment, Market), has been established. A case related to pork supplier companies is presented and fully demonstrates the effectiveness of the novel TODIM when compared with the extended one, the intuitionistic fuzzy TODIM, the Pythagorean fuzzy TODIM as well as the TOPSIS with q-ROFS. Finally, a series of comparative analyses illustrate the advantages of the proposed TODIM. First published online 4 September 2020

Since the approval of chimeric antigen receptor (CAR) T cell therapy targeting CD19 by the FDA, CAR-T cell therapy has received increasing attention as a new method for targeting tumors. Although CAR-T cell therapy has a good effect against hematological malignancies, it has been less effective against solid tumors. In the present study, we selected mesothelin (MSLN/MESO) as a target for CAR-T cells because it is highly expressed by solid tumors but only expressed at low levels by normal tissues. We engineered a third generation MSLN-CAR comprising a single-chain variable fragment (scFv) targeting MSLN (MSLN-scFv), a CD8 transmembrane domain, the costimulatory domains from CD28 and 4-1BB, and the activating domain CD3ζ. In vitro, MSLN-CAR-T cells killed various solid tumor cell lines, demonstrating that it could specifically kill MSLN-positive cells and release cytokines. In vivo, we investigated the effects of MSLN-CAR-T cell therapy against ovarian, breast, and colorectal cancer cell-line-derived xenografts (CDX) and MSLN-positive colorectal and gastric cancer patient-derived xenografts (PDX). MSLN-CAR decreased the growth of MSLN-positive tumors concomitant with significantly increased T cells and cytokine levels compared to the control group. These results indicated that modified MSLN-CAR-T cells could be a promising therapeutic approach for solid tumors.

Abstract Background Epidemiological and clinical features of patients with corona virus disease 2019 (COVID-19) were well delineated. However, no researches described the patients complicated with pleural effusion (PE). In the present study, we aimed to clinically characterize the COVID-19 patients complicated with PE and to create a predictive model on the basis of PE and other clinical features to identify COVID-19 patients who may progress to critical condition. Methods This retrospective study examined 476 COVID-19 inpatients, involving 153 patients with PE and 323 without PE. The data on patients’ past history, clinical features, physical checkup findings, laboratory results and chest computed tomography (CT) findings were collected and analyzed. LASSO regression analysis was employed to identify risk factors associated with the severity of COVID-19. Results Laboratory findings showed that patients with PE had higher levels of white blood cells, neutrophils, lactic dehydrogenase, C-reactive protein and D-dimer, and lower levels of lymphocytes, platelets, hemoglobin, partial pressure of oxygen and oxygen saturation. Meanwhile, patients with PE had higher incidence of severe or critical illness and mortality rate, and longer hospital stay time compared to their counterparts without pleural effusion. Moreover, LASSO regression analysis exhibited that pleural effusion, lactic dehydrogenase (LDH), D-dimer and total bilirubin (TBIL) might be risk factors for critical COVID-19. Conclusions Pleural effusion could serve as an indicator for severe inflammation and poor clinical outcomes, and might be a complementary risk factor for critical type of COVID-19.

Abstract Aims To explore the clinical characteristics and placental pathological changes of pregnant women with 2019 novel coronavirus (CoV) disease (COVID-19) in the third trimester, and to assess the possibility of vertical transmission. Methods and results The placenta tissues were evaluated by using immunohistochemistry for inflammatory cells and Hofbauer cells, and using severe acute respiratory syndrome (SARS) CoV-2 RNA Fluorescence In-Situ Hybridization (FISH) and SARS-CoV-2 spike protein immunofluorescence (IF) double staining. All eight placentas from the third trimester pregnancy women were studied. All patients were cured, no clinical or serological evidence pointed to vertical transmission of SARS-CoV-2. Features of maternal vascular malperfusion (MVM) such as increased syncytial knots were present in all 8 cases (8/8), and increased focal perivillous fibrin depositions were presented in 7 cases (7/8). No significate chronic histiocytic intervillositis was noted in the placenta. The number of macrophages and inflammatory cells such as T cells, B cells and plasma cells in the placental villous was not significantly increased in all cases. Moreover, all of eight cases demonstrated negative results by FISH using a SARS-CoV-2 virus RNA probe and by IF using a monoclonal antibody against SARS-CoV-2 spike protein. Conclusions We found no evidence of vertical transmission and adverse maternal-fetal outcomes in the placentas of third trimester COVID-19 pregnancy women, which provided further information for the clinical management of those women in the third trimester. However, further studies are still needed for patients with infections in different stage of gestation, especially in first and second trimester.

High-performance MgO-CaO-Fe2O3 clinker was prepared using magnesite from Xinjiang (with high calcium and low silica), calcium oxide, and ferric oxide as raw materials. Microstructural analysis and thermogravimetric analysis, combined with HSC chemistry 6 software simulations, were used to investigate the synthesis mechanism of MgO-CaO-Fe2O3 clinker and the effect of firing temperature on the properties of MgO-CaO-Fe2O3 clinker. The results show that MgO-CaO-Fe2O3 clinker with a bulk density of 3.42 g·cm−3, water absorption of 0.7%, and excellent physical properties can be formed by firing at 1600 °C for 3 h. In addition, the crushed and reformed specimens can be refired at temperatures of 1300 °C and 1600 °C to achieve compressive strengths of 17.9 MPa and 39.1 MPa, respectively. The main crystalline phase of the MgO-CaO-Fe2O3 clinker is the MgO phase; the 2CaO·Fe2O3 phase generated by the reaction is distributed between the MgO grains to form a cemented structure with a small quantity of 3CaO·SiO2 and 4CaO·Al2O3·Fe2O3 also distributed between the MgO grains. A series of decomposition and resynthesis chemical reactions occurred during the firing of the MgO-CaO-Fe2O3 clinker, and the liquid phase appeared in the system once the firing temperature exceeded 1250 °C. The presence of the liquid phase promoted intergranular mass transfer between the MgO grains, ensuring the continuous growth of the MgO grains and furthering the densification of the MgO-CaO-Fe2O3 clinker.

The balance between vegetative and reproductive growth is the central objective in the cotton production system, which is associated with cotton maturity and yield. In China, manual topping (MT) has been performed many years prior to or during the flowering period to inhibit vegetative growth and enhance reproductive growth. MT is gradually being replaced by chemical topping (CT) with mepiquat chloride (MC, 180 g ha−1, 98% soluble powder) due to labor shortages and increasing labor cost. To determine whether CT influences cotton maturity and yield relative to MT, we carried out field experiments at four locations in the Yellow River Valley of China during 2018–2020. The results showed that CT did not alter the boll age, and although it produced taller and slender spatial boll distribution under several environments, it had little effect on the accumulation of boll fraction (the number of bolls in a given period divided by total boll number) over time at the end of blooming across locations. As a result, there were no significant differences between MT and CT in boll opening percentage in the late season. CT did not influence yield or yield components, except under severe drought. Therefore, CT with MC (180 g/ha, 98% soluble powder) during the flowering period will not compromise the maturity or yield of cotton in the Yellow River Valley of China. Similar outcomes would be achieved in other areas with similar ecological conditions and social conditions that require an alternative to extensive manual labor.

Abstract Background Mepiquat chloride (MC), a plant growth regulator, enhances root growth by promoting lateral root formation in cotton. However, the underlying molecular mechanisms of this phenomenon is still unknown. Methods In this study, we used 10 cotton (Gossypium hirsutum Linn.) cultivars to perform a seed treatment with MC to investigate lateral root formation, and selected a MC sensitive cotton cultivar for dynamic monitor of root growth and transcriptome analysis during lateral root development upon MC seed treatment. Results The results showed that MC treated seeds promotes the lateral root formation in a dosage-depended manner and the effective promotion region is within 5 cm from the base of primary root. MC treated seeds induce endogenous auxin level by altering gene expression of both gibberellin (GA) biosynthesis and signaling and abscisic acid (ABA) signaling. Meanwhile, MC treated seeds differentially express genes involved in indole acetic acid (IAA) synthesis and transport. Furthermore, MC-induced IAA regulates the expression of genes related to cell cycle and division for lateral root development. Conclusions Our data suggest that MC orchestrates GA and ABA metabolism and signaling, which further regulates auxin biosynthesis, transport, and signaling to promote the cell division responsible for lateral root formation.

Cotton constitutes 81% of the world’s natural fibers. Accurate and rapid cotton yield estimation is important for cotton trade and agricultural policy development. Therefore, we developed a remote sensing index that can intuitively represent cotton boll characteristics and support cotton yield estimation by extracting cotton boll pixels. In our study, the Density of open Cotton boll Pixels (DCPs) was extracted by designing different cotton boll indices combined with the threshold segmentation method. The relationship between DCP and field survey datasets, the Density of Total Cotton bolls (DTC), and yield were compared and analyzed. Five common yield estimation models, Linear Regression (LR), Support Vector Regression (SVR), Classification and Regression Trees (CART), Random Forest (RF), and K-Nearest Neighbors (KNN), were implemented and evaluated. The results showed that DCP had a strong correlation with yield, with a Pearson correlation coefficient of 0.84. The RF method exhibited the best yield estimation performance, with average R2 and rRMSE values of 0.77 and 7.5%, respectively (five-fold cross-validation). This study showed that RedGreenBlue (RGB) and Near Infrared Red (NIR) normalized, a normalized form index consisting of the RGB and NIR bands, performed best.

Abstract Background Soil salinity seriously affects cotton growth, leading to the reduction of yield and fiber quality. Recently, genetic engineering has become an efficient tool to increase abiotic stress tolerance in crops. Results In this study, isopentyl transferase (IPT), a key enzyme involved in cytokinin (CTK) biosynthesis from Agrobacterium tumefaciens, was selected to generate transgenic cotton via Agrobacterium-mediated transformation. A senescence-inducible SAG12 promoter from Arabidopsis was fused with the IPT gene. Ectopic-expression of SAG12::IPT significantly promoted seed germination or seedling tolerance to salt stress. Two IPT transgenic lines, OE3 as a tolerant line during seed germination, and OE8 as a tolerant line at seedling stage, were selected for further physiological analysis. The data showed that ectopic-expression of SAG12::IPT induced the accumulation of CTKs not only in leaves and roots, but also in germinating seeds. Moreover, ectopic-expressing IPT increased the activity of antioxidant enzymes, which was associated with the less reactive oxygen species (ROS) accumulation compared with control plants. Also, ectopic-expression of IPT produced higher K+/Na+ ratio in cotton shoot and root. Conclusion The senescence-induced CTK accumulation in cotton seeds and seedlings positively regulates salt stress partially by elevating ROS scavenging capability.

Background In December 2019, an outbreak of coronavirus disease 2019 (COVID-19) began in Wuhan, China, and led to a global epidemic. We aimed to compare the clinical and serological features of COVID-19 patients with positive and negative reverse transcriptase polymerase chain reaction (RT-PCR) tests. Methods This was a retrospective cohort study conducted from 9 February to 4 April 2020. COVID-19 patients at Leishenshan Hospital in Wuhan, China (125 total cases; 87 RT-PCR positive and 38 RT-PCR negative) were included. COVID-19 serology was assessed by colloidal gold assay. All cases were analyzed for demographic, clinical, and serological features. Results There were no significant differences in most demographic features, clinical symptoms, complications or treatments of RT-PCR positive and negative COVID-19 patients. Serum IgM/IgG was positive in 82 (94%) and 33 (87%) RT-PCR positive and negative cases, respectively. IgM was detectable as early as 3 days after symptom onset and was undetectable 60 days after symptom onset. By contrast, IgG could be detected only 10 days after symptom onset and reached its peak 60 days after symptom onset. Conclusions Serological tests performed during the appropriate time window of disease progression could be valuable auxiliary methods to RT-PCR in COVID-19 patients.

Thidiazuron (TDZ) is widely used as a defoliant to induce leaf abscission in cotton. However, the underlying molecular mechanism is still unclear. In this study, RNA-seq and enzyme-linked immunosorbent assays (ELISA) were performed to reveal the dynamic transcriptome profiling and the change of endogenous phytohormones upon TDZ treatment in leaf, petiole, and abscission zone (AZ). We found that TDZ induced the gene expression of ethylene biosynthesis and signal, and promoted ethylene accumulation earlier in leaf than that in AZ. While TDZ down-regulated indole-3-acetic acid (IAA) biosynthesis genes mainly in leaf and IAA signal and transport genes. Furthermore, the IAA content reduced more sharply in the leaf than that in AZ to change the auxin gradient for abscission. TDZ suppressed CTK biosynthesis genes and induced CTK metabolic genes to reduce the IPA accumulation for the reduction of ethylene sensitivity. Furthermore, TDZ regulated the gene expression of abscisic acid (ABA) biosynthesis and signal and induced ABA accumulation between 12–48 h, which could up-regulate ABA response factor genes and inhibit IAA transporter genes. Our data suggest that TDZ orchestrates metabolism and signal of ethylene, auxin, and cytokinin, and also the transport of auxin in leaf, petiole, and AZ, to control leaf abscission.

TODIM is a well-known multiple-criteria decision-making (MCDM) which considers the bounded rationality of decision makers (DMs) based on prospect theory (PT). However, in the classical TODIM, the perceived probability weighting function and the difference of the risk attitudes for gains and losses are not consistent with the original idea of PT. Moreover, probabilistic hesitant fuzzy information shows its superiority in handling the situation that the DMs hesitate among several possible values with different possibilities. Hence, a novel TODIM with probabilistic hesitant fuzzy information is proposed in this paper to simulate the perceptions of the DMs in PT. To show the advantages of the proposed method, a novel TODIM is combined with hesitant fuzzy information. Finally, a case study is carried out to demonstrate the feasibility of the proposed method, and a series of comparative analyses and the sensitivity analyses are used to show the stability of the proposed method.

Abstract Background In order to efficiently determine genotypic differences in rooting patterns of crops, novel hardware and software are needed simultaneously to characterize dynamics of root development. Results We describe a prototype robotic monitoring platform—the RhizoChamber-Monitor for analyzing growth patterns of plant roots automatically. The RhizoChamber-Monitor comprises an automatic imaging system for acquiring sequential images of roots which grow on a cloth substrate in custom rhizoboxes, an automatic irrigation system and a flexible shading arrangement. A customized image processing software was developed to analyze the spatio-temporal dynamics of root growth from time-course images of multiple plants. This software can quantify overall growth of roots and extract detailed growth traits (e.g. dynamics of length and diameter) of primary roots and of individual lateral roots automatically. It can also identify local growth traits of lateral roots (pseudo-mean-length and pseudo-maximum-length) semi-automatically. Two cotton genotypes were used to test both the physical platform and the analysis software. Conclusions The combination of hardware and software is expected to facilitate quantification of root geometry and its spatio-temporal growth patterns, and therefore to provide opportunities for high-throughput root phenotyping in support of crop breeding to optimize root architecture.

Abstract Background Tissue factor pathway inhibitor (TFPI) is the main physiological inhibitor of TF-induced blood coagulation process, and may play essential roles in the pathogenesis of major adverse cardiac events. This study was designed to determine whether the variation of TFPI was related with coronary artery disease (CAD) in the Han Chinese populations. Methods A total of 1271 patients with coronary atherosclerosis and 1287 normal individuals from northern China were enrolled in the present study. Four tagging single-nucleotide polymorphisms (SNPs) (rs7586970, rs6434222, rs10153820 and rs8176528) from TFPI were selected and genotyped by direct sequencing. And the genotypes of the above SNPs were determined in all these participants. Results In the populations from Beijing and Harbin, no significant case-control differences in the frequencies of TFPI polymorphism (rs10153820 and rs8176528) were observed between CAD patients and controls. Meanwhile, two SNPs of TFPI (rs7586970 and rs6434222) were found to be associated with CAD in both groups. In stratified analyses based on gender, smoking, hypertension, diabetes mellitus and hyperlipidemia, we further determined that the investigated genetic variations of the TFPI genes seemed to be related with diabetes mellitus in CAD patients. Conclusions Genetic variations of the TFPI genes seem to be related with CAD, which likely cooperate with metabolic risk factor (diabetes mellitus) and play critical roles in the pathogenesis of coronary artery disease.

BackgroundLimited findings have been reported to systematically study miRNA and mRNA expression profiles in aged human atria. In this study, we aimed to identify miRNAs, genes, and miRNA-mRNA interaction networks for human atrial aging (AA).MethodsRight atrial appendages from twelve patients who received aortic valve replacement were subjected to miRNA-seq and RNA-seq. All the patients were in sinus rhythm (SR) and stratified by age into four groups. Differential expression analysis was carried out to identify miRNAs and genes for human AA. The miRNA-mRNA interactions for human AA were identified by Pearson correlation analysis and miRNA target prediction programs.ResultsSeven miRNAs (4 upregulation and 3 downregulation) and 42 genes (23 upregulation and 19 downregulation) were differentially expressed in human right atrial tissues between older samples and younger samples. Bioinformatic analysis identified 114 pairs of putative miRNA-mRNA interactions on AA and four types of correlation. Pathway enrichment analysis identified over 40 significant pathways and the top three pathways included rhythmic process (P = 7.5 × 10–5, Q = 0.034), senescence and autophagy in cancer (P = 9.0 × 10–5, Q = 0.034), and positive regulation of cytokine biosynthetic process (P = 1.1 × 10–4, Q = 0.034).ConclusionOur study revealed novel miRNA-mRNA interaction networks and signaling pathways for AA, providing novel insights into the development of human AA. Future studies are needed to investigate the potential significance of these miRNA-mRNA interactions in human AA or AA-related cardiovascular diseases.

Venture capitalists (VCs) have long been preoccupied by the issue of selecting a promising start-up firm, whereas, ranking the available start-up firms is an effective way to solve this issue. In this paper, the PROMETHEE is chosen to be the fundamental ranking method. Also, the hesitant fuzzy linguistic term set is a suitable tool to simulate VCs’ evaluation information. Additionally, as the deepening of social division of labor and specialization of individuals, group decision making is famous for improving decision-making quality. Moreover, in the decision-making process, VCs exhibit behavioral characteristics which is depicted well by prospect theory that VCs are risk averse for gains and risk seeking for losses and rely on the transformed probability to make their decisions rather than unidimensional probability. Thus, a group prospect PROMETHEE with hesitant fuzzy linguistic information is constructed for VCs to make a better decision. Then, the proposed method is applied to rank start-up firms and the comparative analyses are made as well. It confirms that the group prospect PROMETHEE is better in describing the common behavioral characteristics of VCs and in enhancing the quality of evaluation.

Short-chain fatty acids (SCFA) are important dietary and microbiome metabolites that can have roles in gut immunity as well as further afield. We previously observed that gut microbiome alteration via antibiotics led to attenuated lung inflammatory responses. The rationale for this study was to identify gut microbiome factors that regulate lung immune homeostasis. We first investigated key factors within mouse colonic lumen filtrates (CLF) which could elicit direct inflammatory effects in vitro. We identified lipopolysaccharide (LPS) and SCFAs as key CLF ingredients whose levels and inflammatory capacity changed after antibiotic exposure in mice. Specifically, the SCFA propionate appeared to be a key regulator of LPS responses in vitro. Elevated propionate: acetate ratios, as seen in CLF after antibiotic exposure, strongly blunted inflammatory responses in vitro. In vivo, exposure of lungs to high dose propionate, to mimic how prior antibiotic exposure changed SCFA levels, resulted in diminished immune containment of Staphylococcus aureus pneumonia. Finally, we discovered an enrichment of propionate-producing gut bacteria in mice with reduced lung inflammation following lung ischemia reperfusion injury in vivo. Overall, our data show that propionate levels can distinctly modulate lung immune responses in vitro and in vivo and that gut microbiome increased production of propionate is associated with reduced lung inflammation.

Radiation-induced lung injury (RILI) is one of the most common and fatal complications of thoracic radiotherapy. Inflammatory cell infiltration, imbalance of inflammatory cytokines, and oxidative damage were reported to be involved during RILI pathogenesis, especially in the early phase of RILI. Nuclear factor-erythroid 2-related factor 2 (Nrf2) is a key transcriptional regulator of antioxidative cascades, and regulates life span of mice after administration of thoracic irradiation. We investigated the effects of Nrf2 on RILI and inflammation using Nrf2-knockout, Nrf2-overexpression and wild-type mice with or without 15 Gy ionizing radiation to thorax. Our results showed that Nrf2 deficiency aggravated radiation-induced histopathological changes, macrophage and neutrophil infiltration, serum levels of pro-inflammatory cytokines (IL-6, MCP-1, IFN-γ, TNF, and IL-12p70), and the levels of peroxidation products in the mouse lung. Moreover, loss of Nrf2 reduced radiation-induced serum levels of anti-inflammatory cytokine, IL-10, and antioxidative proteins. Nrf2 overexpression significantly alleviated radiation-induced histopathological changes, macrophages and neutrophils infiltration, serum levels of pro-inflammatory cytokines, and the levels of peroxidation products in lung tissues. Nrf2 overexpression also increased the serum levels of IL-10 and antioxidative proteins. These results indicated that Nrf2 had a protective role against radiation-induced acute lung injury and inflammation, and that antioxidative therapy might be a promising treatment for RILI.

Considering the risk aversion for gains and the risk seeking for losses of venture capitalists, the TODIM has been chosen as the decision-making method. Moreover, group decision is an available way to avoid the limited ability and knowledge etc. of venture capitalists. Simultaneously, venture capitalists may be hesitant among several assessed values with different probabilities to express their real perception because of the uncertain decision-making environment. However, the probabilistic hesitant fuzzy information can solve such problems effectively. Therefore, the TODIM has been extended to probabilistic hesitant fuzzy circumstance for the sake of settling the decision-making problem of venture capitalists in this paper. Moreover, due to the uncertain investment environment, the criteria weights are considered as probabilistic hesitant fuzzy information as well. Then, a case study has been used to verify the feasibility and validity of the proposed TODIM. Also, the TODIM with hesitant fuzzy information has been carried out to analysis the same case. From the comparative analysis, the superiority of the proposed TODIM in this paper has already appeared.

To properly understand cotton responses to potassium (K+) deficiency and how its shoot feedback regulates K+ uptake and root growth, we analyzed the changes in root transcriptome induced by low K+ (0.03 mM K+, lasting three days) in self-grafts of a K+ inefficient cotton variety (CCRI41/CCRI41, scion/rootstock) and its reciprocal grafts with a K+ efficient variety (SCRC22/CCRI41). Compared with CCRI41/CCRI41, the SCRC22 scion enhanced the K+ uptake and root growth of CCRI41 rootstock. A total of 1968 and 2539 differently expressed genes (DEGs) were identified in the roots of CCRI41/CCRI41 and SCRC22/CCRI41 in response to K+ deficiency, respectively. The overlapped and similarly (both up- or both down-) regulated DEGs in the two grafts were considered the basic response to K+ deficiency in cotton roots, whereas the DEGs only found in SCRC22/CCRI41 (1954) and those oppositely (one up- and the other down-) regulated in the two grafts might be the key factors involved in the feedback regulation of K+ uptake and root growth. The expression level of four putative K+ transporter genes (three GhHAK5s and one GhKUP3) increased in both grafts under low K+, which could enable plants to cope with K+ deficiency. In addition, two ethylene response factors (ERFs), GhERF15 and GhESE3, both down-regulated in the roots of CCRI41/CCRI41 and SCRC22/CCRI41, may negatively regulate K+ uptake in cotton roots due to higher net K+ uptake rate in their virus-induced gene silencing (VIGS) plants. In terms of feedback regulation of K+ uptake and root growth, several up-regulated DEGs related to Ca2+ binding and CIPK (CBL-interacting protein kinases), one up-regulated GhKUP3 and several up-regulated GhNRT2.1s probably play important roles. In conclusion, these results provide a deeper insight into the molecular mechanisms involved in basic response to low K+ stress in cotton roots and feedback regulation of K+ uptake, and present several low K+ tolerance-associated genes that need to be further identified and characterized.

It is critical to measure the roll angle of a spinning missile quickly and accurately. Magnetometers are commonly used to implement these measurements. At present, the estimation of roll angle parameters is usually performed with the unscented Kalman filter algorithm. In this paper, the two-step adaptive augmented unscented Kalman filter algorithm is proposed to calibrate the biaxial magnetometer and circuit measurements quickly, which allows accurate estimates of the missile roll angle. Unlike the existing algorithms, the state vector of the algorithm is based on the missile roll angle parameters and the error factors caused by the magnetometer and the measurement circuit errors. Next, a two-step fast fitting algorithm is used to fit the initial value. After satisfying the stop rule, the state vector of the filter is configured to estimate the roll angle parameters and the calibration parameters. This method is evaluated by running numerous simulations. In the experiment, the algorithm completes the calibration of the magnetometer and the measurement circuit 1 s after the missile launches, with a sampling rate of 1 ms and an output roll attitude angle with a 0.0015 rad precision. The conventional unscented Kalman filter algorithm requires more time to achieve such a high accuracy. The simulation results demonstrate that the proposed two-step augmented unscented Kalman filter outperforms the conventional unscented Kalman filter in its estimation accuracy and convergence characteristics.

Parasitoid wasps of the order Hymenoptera, the most diverse groups of animals, are important natural enemies of arthropod hosts in natural ecosystems and can be used in biological control. To date, only one neuropeptidome of a parasitoid wasp, Nasonia vitripennis, has been identified. This study aimed to identify more neuropeptides of parasitoid wasps, by using a well-established workflow that was previously adopted for predicting insect neuropeptide sequences. Based on publicly accessible databases, totally 517 neuropeptide precursors from 24 parasitoid wasp species were identified; these included five neuropeptides (CNMamide, FMRFamide-like, ITG-like, ion transport peptide-like and orcokinin B) that were identified for the first time in parasitoid wasps, to our knowledge. Next, these neuropeptides from parasitoid wasps were compared with those from other insect species. Phylogenetic analysis suggested the divergence of AST-CCC within Hymenoptera. Further, the encoding patterns of CAPA/PK family genes were found to be different between Hymenoptera species and other insect species. Some neuropeptides that were not found in some parasitoid superfamilies (e.g., sulfakinin), or considerably divergent between different parasitoid superfamilies (e.g., sNPF) might be related to distinct physiological processes in the parasitoid life. Information of neuropeptide sequences in parasitoid wasps can be useful for better understanding the phylogenetic relationships of Hymenoptera and further elucidating the physiological functions of neuropeptide signaling systems in parasitoid wasps.

Sterile lung injury is an important clinical problem that complicates the course of severely ill patients. Interruption of blood flow, namely ischemia–reperfusion (IR), initiates a sterile inflammatory response in the lung that is believed to be maladaptive. The rationale for this study was to elucidate the molecular basis for lung IR inflammation and whether it is maladaptive or beneficial. Using a mouse model of lung IR, we demonstrate that sequential blocking of inflammasomes [specifically, NOD-, LRR-, and pyrin domain-containing 3 (NLRP3)], inflammatory caspases, and interleukin (IL)-1β, all resulted in an attenuated inflammatory response. IL-1β production appeared to predominantly originate in conjunction with alveolar type 2 epithelial cells. Lung IR injury recruited unactivated or dormant neutrophils producing less reactive oxygen species thereby challenging the notion that recruited neutrophils are terminally activated. However, lung IR inflammation was able to limit or reduce the bacterial burden from subsequent experimentally induced pneumonia. Notably, inflammasome-deficient mice were unable to alter this bacterial burden following IR. Thus, we conclude that the NLRP3 inflammasome, through IL-1β production, regulates lung IR inflammation, which includes recruitment of dormant neutrophils. The sterile IR inflammatory response appears to serve an important function in inducing resistance to subsequent bacterial pneumonia and may constitute a critical part of early host responses to infection in trauma.

Mechanical harvesting has become inevitable for cotton production in China due to the rising labor cost in the country. It usually requires a moderate plant height and compact plant architecture. Correlation and stepwise regression were employed to analyze databases of our 24 field experiments between 2010 and 2017 in Hebei Province. The purpose is to identify the relationships between plant architecture traits and seed cotton yield within natural plant height range (58.6−163.2 cm) or preferred plant height range (80−120 cm) for mechanical harvesting in the Yellow River Valley of China, and define some important factors affecting seed cotton yield. Under natural plant height range across all experiments, there was a significantly negative correlation (r = −0.452) between plant height and yield. On limited plant height range desired for mechanical harvesting, the degree of this negative correlation decreased to r = −0.366. The correlation of plant height with seed cotton yield varied greatly with year, cultivar, plant density and mepiquat chloride (MC) application. Moreover, stepwise regression analysis picked internode length of the 1st (generally including 1st−7th mainstem node from bottom), 2nd (8th−12th node) and 4th (above 17th node) mainstem section and the length of lower fruiting branch (LFB) as significant factors influencing seed cotton yield under plant height range of 80−120 cm. The results have implications for precise control of cotton plant architecture preferred for mechanized harvesting in China.

Coronatine is a phytotoxin that affects the accumulation of defence-related metabolites in plants but information on how its effects may be mediated by environmental stress is scanty. An experiment was carried out to determine the changes in growth, gas exchange, relative water content, chlorophyll (Chl) content, antioxidant enzymes and lipid peroxidation in maize (Zea mays L., var. ‘Nongda 3138’) seedlings treated with coronatine under simulated drought stress. Seedlings raised hydroponically in a growth chamber with simulated drought for 8d (long-period drought) or 3d (short-period drought) were treated with or without coronatine at the three-leaf stage. Under the drought condition, treated with coronatine significantly increased the fresh weight and relative water content in leaves of seedling leaves. The increase was accompanied by increased rates of photosynthesis and transpiration, and the maintenance of Chl pigments. Coronatine had no effects on catalase (CAT), guaiacol peroxidase (POD) and glutathione reductase (GR) under normal condition, but it significantly enhanced activities of CAT, POD and GR in stressed seedlings under the long-period drought treatment. Under the short-period drought treatment, the POD and GR activity in the seedlings treated with coronatine were much higher than in those not treated. Malondialdehyde (MDA) increased sharply under drought condition, but treatment with coronatine significantly reduced it by 15%. The total Chl content of leaves under the drought condition was markedly increased by the treatment with coronatine. Seedlings subjected to a short-period drought had reduced water content, but recovered fairly well by the treatment with coronatine with negligible effects on most physiological and biochemical processes. The application of coronatine alleviated the drought stress in maize seedlings and enhanced their tolerance of water stress through changes in physiological and anti-oxidant enzyme activities.

In order to recognize the target of high speed quickly and accurately, an identification system was designed based on analysis of the distribution characteristics of the plume spectrum. In the system, the target was aligned with visible light tracking module, and the spectral analysis of the target’s plume radiation was achieved by interference module. The distinguishing factor recognition algorithm was designed on basis of ratio of multifeature band peaks and valley mean values. Effective recognition of the high speed moving target could be achieved after partition of the active region and the influence of target motion on spectral acquisition was analyzed. In the experiment the small rocket combustion was used as the target. The spectral detection experiment was conducted at different speeds 2.0 km away from the detection system. Experimental results showed that spectral distribution had significant spectral offset in the same sampling period for the target with different speeds, but the spectral distribution was basically consistent. Through calculation of the inclusion relationship between distinguishing factor and distinction interval of the peak value and the valley value at the corresponding wave-bands, effective identification of target could be achieved.

Coronary heart disease (CHD) is one of the leading causes of mortality and morbidity in China. Genetic factors that predispose individuals to CHD are unclear. In the present study, we aimed to determine whether the variation of FoxOs, a novel genetic factor associated with longevity, was associated with CHD in Han Chinese populations.1271 CHD patients and 1287 age-and sex-matched controls from Beijing and Harbin were included. We selected four tagging single nucleotide polymorphisms (SNPs) of FoxO1 (rs2755209, rs2721072, rs4325427 and rs17592371) and two tagging SNPs of FoxO3 (rs768023 and rs1268165). And the genotypes of these SNPs were determined in both CHD patients and non-CHD controls.For population from Beijing, four SNPs of FoxO1 and two SNPs of FoxO3 were found not to be associated with CHD (p>0.05). And this was validated in the other population from Harbin (p>0.05). After combining the two geographically isolated case-control populations, the results showed that the six SNPs did not necessarily predispose to CHD in Han Chinese(p>0.05). In stratified analysis according to gender, the history of smoking, hypertension, diabetes mellitus, hyperlipidemia and the metabolic syndrome, we further explored that neither the variants of FoxO1 nor the variants of FoxO3 might be associated with CHD (p>0.05).The variants of FoxO1 and FoxO3 may not increase the prevalence of CHD in Han Chinese population.

Defoliants can increase machine harvest efficiency of cotton (Gossypium hirusutum L.), prevent lodging and reduce the time from defoliation to harvest. Coronatine (COR) is a chlorosis-inducing non-host-specific phytotoxin that induces leaf and/or fruit abscission in some crops. The present study investigates how COR might induce cotton leaf abscission by modulating genes involved in cell wall hydrolases and ACC (ethylene precursor) in various cotton tissues. The effects of COR on cotton boll ripening, seedcotton yield, and seed development were also studied. After 14 d of treatment with COR, cells within the leaf abscission zone (AZ) showed marked differentiation. Elevated transcripts of GhCEL1, GhPG and GhACS were observed in the AZs treated with COR and Thidiazuron (TDZ). The relative expression of GhCEL1 and GhACS in TDZ treated plants was approximately twice that in plants treated with COR for 12 h. However, only GhACS expression increased in leaf blade and petiole. There was a continuous increase in the activity of hydrolytic enzymes such as cellulase (CEL) and polygalacturonase (PG), and ACC accumulation in AZs following COR and TDZ treatments, but there was greater increase in ACC activity of COR treated boll crust, indicating that COR had greater ripening effect than TDZ. Coronatine significantly enhanced boll opening without affecting boll weight, lint percentage and seed quality. Therefore, COR can be a potential cotton defoliant with different physiological mechanism of action from the currently used TDZ.

Secondary bacterial pneumonias are a frequent complication of influenza and other respiratory viral infections, but the mechanisms underlying viral-induced susceptibility to bacterial infections are poorly understood. In particular, it is unclear whether the host's response against the viral infection, independent of the injury caused by the virus, results in impairment of antibacterial host defense. Here, we sought to determine whether the induction of an "antiviral" immune state using various viral recognition receptor ligands was sufficient to result in decreased ability to combat common bacterial pathogens of the lung. Using a mouse model, animals were administered polyinosine-polycytidylic acid (poly I:C) or Toll-like 7 ligand (imiquimod or gardiquimod) intranasally, followed by intratracheal challenge with Streptococcus pneumoniae. We found that animals pre-exposed to poly I:C displayed impaired bacterial clearance and increased mortality. Poly I:C-exposed animals also had decreased ability to clear methicillin-resistant Staphylococcus aureus. Furthermore, we showed that activation of Toll-like receptor (TLR)3 and Retinoic acid inducible gene (RIG-I)/Cardif pathways, which recognize viral nucleic acids in the form of dsRNA, both contribute to poly I:C mediated impairment of bacterial clearance. Finally, we determined that poly I:C administration resulted in significant induction of type I interferons (IFNs), whereas the elimination of type I IFN signaling improved clearance and survival following secondary bacterial pneumonia. Collectively, these results indicate that in the lung, poly I:C administration is sufficient to impair pulmonary host defense against clinically important gram-positive bacterial pathogens, which appears to be mediated by type I IFNs.
