Abstract On-chip optical power monitors are indispensable for functional implementation and stabilization of large-scale and complex photonic integrated circuits (PICs). Traditional on-chip optical monitoring is implemented by tapping a small portion of optical power from the waveguide, which leads to significant loss. Due to its advantages like non-invasive nature, miniaturization, and complementary metal-oxide-semiconductor (CMOS) process compatibility, a transparent monitor named the contactless integrated photonic probe (CLIPP), has been attracting great attention in recent years. The CLIPP indirectly monitors the optical power in the waveguide by detecting the conductance variation of the local optical waveguide caused by the surface state absorption (SSA) effect. In this review, we first introduce the fundamentals of the CLIPP including the concept, the equivalent electric model and the impedance read-out method, and then summarize some characteristics of the CLIPP. Finally, the functional applications of the CLIPP on the identification and feedback control of optical signal are discussed, followed by a brief outlook on the prospects of the CLIPP. Superconducting nanowire single-photon detectors (SNSPDs) have become a mainstream photon-counting technology that has been widely applied in various scenarios. So far, most multi-channel SNSPD systems, either reported in literature or commercially available, are polarization sensitive, that is, the system detection efficiency (SDE) of each channel is dependent on the state of polarization of the to-be-detected photons. Here, we reported an eight-channel system with fractal SNSPDs working in the wavelength range of 930 to 940 nm, which are all featured with low polarization sensitivity. In a close-cycled Gifford-McMahon cryocooler system with the base temperature of 2.2 K, we installed and compared the performance of two types of devices: (1) SNSPD, composed of a single, continuous nanowire and (2) superconducting nanowire avalanche photodetector (SNAP), composed of 16 cascaded units of two nanowires electrically connected in parallel. The highest SDE among the eight channels reaches 96−5+4%, with the polarization sensitivity of 1.02 and a dark-count rate of 13 counts per second. The average SDE for eight channels for all states of polarization is estimated to be 90 ± 5%. It is concluded that both the SNSPDs and the SNAPs can reach saturated, high SDE at the wavelength of interest, and the SNSPDs show lower dark-count (false-count) rates, whereas the SNAPs show better properties in the time domain. With the adoption of this system, we showcased the measurements of the second-order photon-correlation functions of light emission from a single-photon source based on a semiconductor quantum dot and from a pulsed laser. It is believed that this work will provide new choices of systems with single-photon detectors combining the merits of high SDE, low polarization sensitivity, and low noise that can be tailored for different applications.

While many studies have compared global precipitation datasets at national, continental, and global scales, few have evaluated these data at river basin scales. This study explored differences in precipitation estimates and trends of 12 widely applied precipitation datasets, including gauge-, satellite-, and reanalysis-based products, for the world's 6,292 river basins. Results showed that disparities between 12 precipitation datasets were considerable. A total of 3,125 river basins, with a land area of 5,989.1×104 km2, had differences in estimated annual average precipitation exceeding 500 mm yr−1, and these basins were mainly distributed in Greenland, Africa, Oceania, and West Asia. Disparities between the precipitation datasets were particularly large during the dry season when the percentage difference between the highest and lowest precipitation estimates exceeded 500% in 1,390 river basins (4,839.7×104 km2) expected due to numerical reasons. Differences in rainfall trends also varied markedly between data sources. The data products do not agree on precipitation trends for all river basins. These findings illustrate the importance of accurate precipitation data to ensure effective policy and planning in term of hydropower generation, domestic water supply, flood protection, and drought relief at river basin scales and highlight the uncertainty that exists in current global precipitation data. Abstract Silicon sub-bandgap photodetectors can detect light at the infrared telecommunication wavelengths but with relatively weak photo-response. In this work, we demonstrate the enhancement of sub-bandgap photodetection in silicon by helium-ion implantation, without affecting the transparency that is an important beneficial feature of this type of photodetectors. With an implantation dose of 1 × 1013 ions/cm2, the minimal detectable optical power can be improved from − 33.2 to − 63.1 dBm, or, by 29.9 dB, at the wavelength of 1550 nm, and the photo-response at the same optical power (− 10 dBm) can be enhanced by approximately 18.8 dB. Our work provides a method for strategically modifying the intrinsic trade-off between transparency and strong photo-responses of this type of photodetectors. Graphical Abstract

Food provides essential nutrients and energy for wildlife and directly influences the compositional patterns of the gut microbiota in natural reservoirs. Little is known about the interspecific differences and correlations between diet and microbial communities in sympatric herbivores. We conducted chloroplast trnL and 16S rRNA genes amplicon sequencing of 90 faecal samples from sika deer (Cervus nippon kopschi), Reeve’s muntjac (Muntiacus reevesi), and a Chinese hare (Lepus sinensis). Diet analysis revealed that sika deer predominantly foraged on Rubus (36.5 %), Loropetalum (25.5 %), and Eurya (13.4 %) at the genus level; Reeve’s muntjac favoured Rubus (51.8 %), Eurya (11.0 %), and Euonymus (6.1 %); Chinese hare showed dominance in Rubus (15.8 %), Smilax (15.6 %), and Rhus (10.6 %) consumption, with all these forage plants being shrubs. The gut microbiota of the three herbivores was characterised by Firmicutes, Bacteroidetes, and Proteobacteria, although the relative abundances of these taxa differed across herbivore species. Alpha diversity indicated no significant differences between sika deer and Reeve’s muntjac, whereas alpha indices showed significant differences between sika deer and Chinese hare (p < 0.05). Beta diversity analysis indicated significant differences between the Chinese hare and sika deer, as well as between the Chinese hare and Reeve’s muntjac (p < 0.05), with no significant difference found between the two cervid species, which may be attributed to the influence of interspecific genetics and evolutionary factors. Due to seasonal shifts, the food availability for herbivores decreased, resulting in the convergent adaptation of shrubs consumed by the three species. Furthermore, PICRUSt functional profile prediction indicated that the gut microbiota of the three species were mainly enriched in metabolic functions. Mantel correlation analysis suggested that the gut microbiota of sika deer showed a significant positive correlation with five genera of plants, one genus of Reeve’s muntjacs, and three genera of Chinese hare (p < 0.01), suggesting that the gut microbiota of sika deer may be more susceptible to changes in food abundance. Our study emphasises how the winter climate affects the foraging strategies and microbial communities of the three herbivores, as well as their correlations. These results may further contribute to the food selection, regulation of gut microbiota, disease prevention, conservation, and management of sika deer and other sympatric herbivores.

Partitioning of evapotranspiration (ET) into soil evaporation (E) and plant transpiration (T) poses a significant challenge. This study proposed a novel approach that combines eddy covariance, micro-lysimeter, and high-resolution unmanned aerial vehicle (UAV) images within the flux footprint to optimize the canopy and soil resistances of the Shuttleworth-Wallace model (S-W) using a multi-objective optimization scheme. A two-year experiment was conducted at the Yucheng and Luancheng sites to investigate the effectiveness of the proposed method for the summer maize-wheat winter system in the North China Plain. Our results showed that the S-W model with multi-objective optimization by using high-resolution UAV images within the flux footprint significantly improved the estimation accuracy of ET components compared with the traditional one- and two-objective optimization schemes. The estimation of ET, T, and E of our method had an average root mean square error of 0.67 mm day−1, 0.66 mm day−1, and 0.28 mm day−1, respectively. The optimized S-W model using high-resolution UAV data within the flux footprint produced better ET partitioning than that optimized using Moderate Resolution Imaging Spectroradiometer (MODIS) data. We then applied the proposed method to estimate ET and ET components in the North China Plain. This study highlighted the importance of utilizing multi-source data (especially high-resolution UAV images) and multi-objective optimization for accurate ET partitioning.

Abstract Triphenyltin (TPT) is a widespread synthetic chemical used in many fields and its potential risk to organisms has been comprehensively investigated using different animal models and species. Currently, little is known about the effects of TPT exposure on microbial midgut diversity, therefore we explored these effects in the lepidopterous silkworm model using 16S rDNA sequencing. In total, 5273 and 5065 operational taxonomic units (OTUs) were identified in control and TPT-exposure group samples, ranging from 424 to 728 OTUs/sample. Alpha-diversity analyses revealed that TPT exposure induced the fluctuations of gut microbial diversity and abundance while beta-diversity analyses identified a distinct impact on major gut microbiota components. In our microbiome analyses, 23 phyla and 353 genera were recognized in the control group, while 20 phyla and 358 genera were recognized in the TPT exposure group. At the genus level, midgut microbiota were composed of several predominant bacterial genera, including Muribaculaceae, Lactobacillus, and UCG-010. In the TPT exposure group, o__Bacillales, f__Bacillaceae, and f__Caldicoprobacteraceae abundance was relatively high, while f__Oscillospiraceae, f__Fusobacteriaceae, and f__SC_I_84 abundance was relatively high in the control group. Gene function analyses in silkworm microbiota after TPT exposure showed that biosynthesis of ansamycins, fructose and mannose metabolism, glycerolipid metabolism, type II diabetes mellitus, glycolysis/gluconeogenesis, lipid metabolism, translation proteins, atrazine degradation, DNA repair and recombination proteins, nicotinate and nicotinamide metabolism were significantly increased. Collectively, our silkworm model identified gut microbial diversity risks and the adverse effects from TPT exposure, which were similar to other aquatic animals. Therefore, TPT levels in environmental samples must be monitored to prevent ecological harm.

The demand for freshwater is increasing with population growth and rapid socio-economic development. It is more and more important for refined irrigation water management to conduct research on crop evapotranspiration (ET) data with a high spatiotemporal resolution in agricultural regions. We propose the unmixing–weight ET image fusion model (UWET), which integrates the advantages of the unmixing method in spatial downscaling and the weight-based method in temporal prediction to produce daily ET maps with a high spatial resolution. The Landsat-ET and MODIS-ET datasets for the UWET fusion data are retrieved from Landsat and MODIS images based on the surface energy balance model. The UWET model considers the effects of crop phenology, precipitation, and land cover in the process of the ET image fusion. The precision evaluation is conducted on the UWET results, and the measured ET values are monitored by eddy covariance at the Luancheng station, with average MAE values of 0.57 mm/day. The image results of UWET show fine spatial details and capture the dynamic ET changes. The seasonal ET values of winter wheat from the ET map mainly range from 350 to 660 mm in 2019–2020 and from 300 to 620 mm in 2020–2021. The average seasonal ET in 2019–2020 is 499.89 mm, and in 2020–2021, it is 459.44 mm. The performance of UWET is compared with two other fusion models: the Spatial and Temporal Adaptive Reflectance Fusion Model (STARFM) and the Spatial and Temporal Reflectance Unmixing Model (STRUM). UWET performs better in the spatial details than the STARFM and is better in the temporal characteristics than the STRUM. The results indicate that UWET is suitable for generating ET products with a high spatial–temporal resolution in agricultural regions.

BackgroundPreoperative imaging for some unusual lesions in the sellar region can pose challenges in establishing a definitive diagnosis, impacting treatment strategies.MethodsThis study is a retrospective analysis of eight cases involving unusual sellar region lesions, all treated with endoscopic endonasal transsphenoidal surgery (EETS). We present the clinical, endocrine, and radiological characteristics, along with the outcomes of these cases.ResultsAmong the eight cases, the lesions were identified as follows: Solitary fibrous tumor (SFT) in one case, Lymphocytic hypophysitis (LYH) in one case, Cavernous sinus hemangiomas (CSH) in one case, Ossifying fibroma (OF) in two cases; Sphenoid sinus mucocele (SSM) in one case, Pituitary abscess (PA) in two cases. All patients underwent successful EETS, and their diagnoses were confirmed through pathological examination. Postoperatively, all patients had uneventful recoveries without occurrences of diabetes insipidus or visual impairment.ConclusionOur study retrospectively analyzed eight unusual lesions of the sellar region. Some lesions exhibit specific imaging characteristics and clinical details that can aid in preoperative diagnosis and inform treatment strategies for these unusual sellar diseases.

Polymerases encoded by segmented negative-strand RNA viruses cleave 5'-m7G-capped host transcripts to prime viral mRNA synthesis ("cap-snatching") to generate chimeric RNA, and trans-splicing occurs between viral and cellular transcripts. Bombyx mori cytoplasmic polyhedrosis virus (BmCPV), an RNA virus belonging to Reoviridae, is a major pathogen of silkworm (B. mori). The genome of BmCPV consists of 10 segmented double-stranded RNAs (S1-S10) from which viral RNAs encoding a protein are transcribed. In this study, chimeric silkworm-BmCPV RNAs, in which the sequence derived from the silkworm transcript could fuse with both the 5' end and the 3' end of viral RNA, were identified in the midgut of BmCPV-infected silkworms by RNA_seq and further confirmed by RT-PCR and Sanger sequencing. A novel chimeric RNA, HDAC11-S4 RNA 4, derived from silkworm histone deacetylase 11 (HDAC11) and the BmCPV S4 transcript encoding viral structural protein 4 (VP4), was selected for validation by in situ hybridization and Northern blotting. Interestingly, our results indicated that HDAC11-S4 RNA 4 was generated in a BmCPV RNA-dependent RNA polymerase (RdRp)-independent manner and could be translated into a truncated BmCPV VP4 with a silkworm HDAC11-derived N-terminal extension. Moreover, it was confirmed that HDAC11-S4 RNA 4 inhibited BmCPV proliferation, decreased the level of H3K9me3 and increased the level of H3K9ac. These results indicated that during infection with BmCPV, a novel mechanism, different from that described in previous reports, allows the genesis of chimeric silkworm-BmCPV RNAs with biological functions.

Chiroptical signals, including optical rotation (OR) and circular dichroism (CD), are widely utilized as potent probes for investigating the structure and properties of chiral molecules. However, acquiring both coexisting weak OR and CD signals simultaneously in a single measurement process with a high precision remains a challenge for conventional setups. In this article, a high-precision yet easy-to-set-up method for simultaneous detection of OR and CD signals based on weak measurement is theoretically and experimentally demonstrated. In addition, the chiroptical signals are detected using two new intensity-contrast-ratio pointers, which provide an expanded measurement range compared with the shift of the mean value pointer. The proposed method has been validated through a spin Hall effect light based experiment, with precision reaching the order of 10−7 and 10−6 rad for the detection of OR and CD, respectively. These results may serve as a catalyst for further studies of fast, multi-parameter biosensing technologies with ultra-precision.

Pit mud (PM), as an important source of microorganisms, is necessary for Chinese strong-flavor baijiu (CSFB) production. Although it has been revealed that the PM prokaryotic community diversities are influenced by its quality, product area, ages, etc., the characteristics and assembly process of the prokaryotic community in PMs across a pH gradient are still unclear. In this study, the regular changes of α- and β-diversities of the prokaryotic community across a pH gradient in PMs were revealed, which could be divided into “stable”, “relatively stable”, and “drastically changed“ periods. A total of 27 phyla, 53 classes, and 381 genera were observed in all given samples, dominated by Firmicutes, Bacteroidetes, Proteobacteria, Lactobacillus, Caproiciproducens, Proteiniphilum, etc. Meanwhile, the complexity of the network structure of the prokaryotic microbial communities is significantly influenced by pH. The community assembly was jointly shaped by deterministic and stochastic processes, with stochastic process contributing more. This study was a specialized report on elucidating the characteristics and assembly of PM prokaryotic communities across a pH gradient, and revealed that the diversity and structure of PM prokaryotic communities could be predictable, to some degree, which could contribute to expanding our understanding of prokaryotic communities in PM.

The liver, a crucial metabolic organ in animals, is responsible for the synthesis, breakdown, and transport of lipids. However, the regulatory mechanisms involving both coding and noncoding RNAs that oversee the development of the goose liver remain elusive. This study aimed to fill this knowledge gap by conducting RNA-seq to profile the expression of circular RNAs (circRNAs) and microRNAs (miRNAs) during goose liver development. We analyzed circRNAs in liver samples from Sichuan white geese at three developmental stages: posthatching day 0, 10 weeks (fast growth stage), and 30 weeks (sexual maturation stage). Our findings revealed 11,079 circRNAs and 994 miRNAs, among which the differentially expressed circRNAs and miRNAs were significantly enriched in pathways such as fatty acid biosynthesis, degradation, and metabolism. Further analysis of the target genes of the differentially expressed miRNAs revealed enrichment in pathways related to fatty acid biosynthesis, metabolism, PPAR signaling, DNA replication, and the cell cycle. We also established circRNA–miRNA–mRNA regulatory networks, identifying key regulatory factors and miRNAs. In conclusion, our study offers valuable insights into the complex interplay of circRNA–miRNA–mRNA interactions during goose liver development, and illuminates the molecular pathways that regulate this vital life function.

The parameters of the GR4J-CemaNeige coupling model (GR4neige) are typically treated as constants. However, the maximum capacity of the production store (parX1) exhibits time-varying characteristics due to climate variability and vegetation coverage change. This study employed differentiable parameter learning (dPL) to identify the time-varying parX1 in the GR4neige across 671 catchments within the United States. We built two types of dPL, including static and dynamic parameter networks, to assess the advantages of the time-varying parameter. In the dynamic parameter network, we evaluated the impact of potential evapotranspiration (PET), precipitation (P), temperature (T), soil moisture (SM), and normalized difference vegetation index (NDVI) datasets on the performance of dPL. We then compared dPL with the empirical functional method (fm). The results demonstrated that the dynamic parameter network outperformed the static parameter network in streamflow estimation. There were differences in streamflow estimation among the dynamic parameter network driven by various input features. In humid catchments, simultaneously incorporating all five factors, including PET, P, T, SM, and the NDVI, achieved optimal streamflow simulation accuracy. In arid catchments, it was preferable to introduce PET, T, and the NDVI separately for improved performance. dPL significantly outperformed the empirical fm in estimating streamflow and uncalibrated intermediate variables, like evapotranspiration (ET). Both the derived parX1 from dPL and the empirical fm exhibited significant spatiotemporal variation across 671 catchments. Notably, compared to parX1 obtained through the empirical fm, parX1 derived from dPL exhibited a distinct spatial clustering pattern. This study highlights the potential of dPL in enhancing model accuracy and contributes to understanding the spatiotemporal variation characteristics of parX1 under the influence of climate factors, soil conditions, and vegetation change.

BackgroundThe continuous exploration of oligometastatic disease has led to the remarkable achievements of local consolidative therapy (LCT) and favorable outcomes for this disease. Thus, this study investigated the potential benefits of LCT in patients with single-organ metastatic pancreatic ductal adenocarcinoma (PDAC).MethodsPatients with single-organ metastatic PDAC diagnosed between 2010 - 2019 were identified from the Surveillance, Epidemiology, and End Results (SEER) database. Propensity score matching (PSM) was performed to minimize selection bias. Factors affecting survival were assessed by Cox regression analysis and Kaplan-Meier estimates.ResultsA total of 12900 patients were identified from the database, including 635 patients who received chemotherapy combined with LCT with a 1:1 PSM with patients who received only chemotherapy. Patients with single-organ metastatic PDAC who received chemotherapy in combination with LCT demonstrated extended median overall survival (OS) by approximately 57%, more than those who underwent chemotherapy alone (11 vs. 7 months, p < 0.001). Furthermore, the multivariate Cox regression analysis revealed that patients that received LCT, younger age (< 65 years), smaller tumor size (< 50 mm), and lung metastasis (reference: liver) were favorable prognostic factors for patients with single-organ metastatic PDAC.ConclusionThe OS of patients with single-organ metastatic pancreatic cancer who received LCT may be prolonged compared to those who received only chemotherapy. Nevertheless, additional prospective randomized clinical trials are required to support these findings.

Recently, bismuth (Bi)-based photocatalysts have been a well-deserved hotspot in the field of photocatalysis owning to their photoelectrochemical properties driven by the distortion of the Bi 6 s orbital, while their narrow band gap and poor quantum efficiency still restrict their application. With the development of ultrasonic technology, it is expected to become a broom to clear the application obstacles of Bi-based photocatalysts. The special forces and environmental conditions brought by ultrasonic irradiation play beneficial roles in the preparation, modification and performance releasement of Bi-based photocatalysts. In this review, the role and influencing factors of ultrasound in the preparation and modification of Bi-based photocatalysts were introduced. Crucially, the mechanism of the improving the performance for various types of Bi-based photocatalysts by ultrasound in the whole process of photocatalysis was deeply analyzed. Then, the application of ultrasonic synergistic Bi-based photocatalysts in contaminants treatment and energy conversion was briefly introduced. Finally, based on an unambiguous understanding of ultrasonic technology in assisting Bi-based photocatalysts, the future directions and possibilities for ultrasonic synergistic Bi-based photocatalysts are explored.

IntroductionPosterior communicating artery (Pcom) aneurysm has unique morphological characteristics and a high recurrence risk after coil embolization. This study aimed to evaluate the relationship between the recurrence-related morphology characteristics and hemodynamics.MethodA total of 20 patients with 22 Pcom aneurysms from 2019 to 2022 were retrospectively enrolled. The recurrence-related morphology parameters were measured. The hemodynamic parameters were simulated based on finite element analysis and computational fluid dynamics. The hemodynamic differences before and after treatment caused by different morphological features and the correlation between these parameters were analyzed.ResultSignificant greater postoperative inflow rate at the neck (Qinflow), relative Qinflow, inflow concentration index (ICI), and residual flow volume (RFV) were reported in the aneurysms with wide neck (>4 mm). Significant greater postoperative RFV were reported in the aneurysms with large size (>7 mm). Significant greater postoperative Qinflow, relative Qinflow, and ICI were reported in the aneurysms located on the larteral side of the curve. The bending angle of the internal carotid artery at the initiation of Pcom (αICA@PCOM) and neck diameter had moderate positive correlations with Qinflow, relative Qinflow, ICI, and RFV.ConclusionThe morphological factors, including aneurysm size, neck diameter, and αICA@PCOM, are correlated with the recurrence-inducing hemodynamic characteristics even after fully packing. This provides a theoretical basis for evaluating the risk of aneurysm recurrence and a reference for selecting a surgical plan.

Species co-existence depends on how organisms utilize their environment and resources. Little is known about the winter diet composition and sympatric co-existence of South China sika deer and its companion species in Taohongling. In this study, high-throughput sequencing and metabarcoding trnL were used to study the diet composition and interspecific relationship including sika deer, Reeve’s muntjac, and Chinese hare. Our results show that 203 genera in 90 families are contained in the diet of sika deer, 203 genera in 95 families for Reeve’s muntjac, and 163 genera in 75 families for Chinese hare. Sika deer fed on Rubus chingii, Loropetalum chinense, and Eurya japonica in winter, accounting for 75.30%; Reeve’s muntjac consumed mainly R. chingii, E. japonica, and Euonymus grandiflorus, accounting for 68.80%, and Chinese hare mainly fed on R. chingii, Smilax china, and Rhus chinensis, accounting for 41.98%. The Shannon index showed no significant difference between groups (p > 0.05). The NMDS analysis found considerable overlap among three species. Sika deer and Reeve’s muntjac consumed similar forage plants but varied greatly in Chinese hare, which occupied the widest choice in winter, resulting in higher diet breadth and increased dietary divergence, thereby reducing competition and facilitating coexistence. The diet niche overlap index among them, as represented by Pianka’s index, ranging from 0.62 between sika deer and Chinese hare to 0.83 between sika deer and Reeve’s muntjac, which indicated a more similar niche and potential competition in closely related species. Our findings provide a new diet perspective of three herbivores, leading to a more comprehensive understanding of resource partitioning and species coexistence.

ABSTRACT Some insect viruses encode suppressors of RNA interference (RNAi) to counteract the antiviral RNAi pathway. However, it is unknown whether Bombyx mori cytoplasmic polyhedrosis virus (BmCPV) encodes an RNAi suppressor. In this study, the presence of viral small interfering RNA (vsiRNA) in BmN cells infected with BmCPV was confirmed by small RNA sequencing. The Dual-Luciferase reporter test demonstrated that BmCPV infection may prevent firefly luciferase (Luc) gene silencing caused by particular short RNA. It was also established that the inhibition relied on the nonstructural protein NSP8, which suggests that NSP8 was a possible RNAi suppressor. In cultured BmN cells, the expressions of viral structural protein 1 (vp1) and NSP9 were triggered by overexpression of nsp8, suggesting that BmCPV proliferation was enhanced by NSP8. A pulldown assay was conducted with BmCPV genomic double-stranded RNA (dsRNA) labeled with biotin. The mass spectral detection of NSP8 in the pulldown complex suggests that NSP8 is capable of direct binding to BmCPV genomic dsRNA. The colocalization of NSP8 and B. mori Argonaute 2 (BmAgo2) was detected by an immunofluorescence assay, leading to the hypothesis that NSP8 interacts with BmAgo2. Coimmunoprecipitation further supported the present investigation. Moreover, vasa intronic protein, a component of RNA-induced silencing complex (RISC), could be detected in the coprecipitation complex of NSP8 by mass spectrum analysis. NSP8 and the mRNA decapping protein (Dcp2) were also discovered to colocalize to processing bodies (P bodies) for RNAi-mediated gene silencing in Saccharomyces cerevisiae. These findings revealed that by interacting with BmAgo2 and suppressing RNAi, NSP8 promoted BmCPV growth. IMPORTANCE It has been reported that the RNAi pathway is inhibited by binding RNAi suppressors encoded by some insect-specific viruses belonging to Dicistroviridae, Nodaviridae, or Birnaviridae to dsRNAs to protect dsRNAs from being cut by Dicer-2. However, it is unknown whether BmCPV, belonging to Spinareoviridae, encodes an RNAi suppressor. In this study, we found that nonstructural protein NSP8 encoded by BmCPV inhibits small interfering RNA (siRNA)-induced RNAi and that NSP8, as an RNAi suppressor, can bind to viral dsRNAs and interact with BmAgo2. Moreover, vasa intronic protein, a component of RISC, was found to interact with NSP8. Heterologously expressed NSP8 and Dcp2 were colocalized to P bodies in yeast. These results indicated that NSP8 promoted BmCPV proliferation by binding itself to BmCPV genomic dsRNAs and interacting with BmAgo2 through suppression of siRNA-induced RNAi. Our findings deepen our understanding of the game between BmCPV and silkworm in regulating viral infection.

Abstract Mesenchymal stem cell‐derived extracellular vesicle (MSC‐EV) is shown to promote cardiac repair, however, it still falls short in initiating myocardia proliferation restart. In this regard, ROS‐induced DNA damage and responses are the culprit of cellcycle arrest. Here, this work constructs a hybrid cell‐derived extracellular vesicle that is composed of MSC and macrophage membranes and encompasses MitoN, a ROS scavenger, to boost the healing of the heart. The MitoN, a NAD(P)H mimic, could target the mitochondrial to eliminate the ROS resuming the arrested cell cycle. The hybrid extracellular vesicle (N@MEV) could respond to the inflammatory signals generated during myocardial injury and thus enable superior targeting and enrichment to the location of the damage. L‐arginine, which could be catalyzed by NOS and ROS into NO and SO provide a driving force, is immobilized within the vesicle (NA@MEV) to further enhance the N@MEV's potential to penetrate the cardiac stroma. In combination with multiple mechanisms, NA@MEV increased heart function 1.3‐fold EF% versus MSC‐EV in mouse myocardial injury model. A more in‐depth mechanistic study found that the NA@MEV could modulate M2 macrophage; promote angiogenesis; reduce DNA damage and response, and thereby restart cardiomyocyte proliferation. Thus, this combined therapy shows synthetic effects in heart repair and regeneration.

Aiming at the problem of asynchronous multi-target tracking, this paper studies the AA fusion optimization problem of multi-sensor networks. Firstly, each sensor node runs a PHD filter, and the measurement information obtained from different sensor nodes in the fusion interval is flood communicated into composite measurement information. The Gaussian component representing the same target is associated with a subset by distance correlation. Then, the Bayesian Cramér–Rao Lower Bound of the asynchronous multi-target-tracking error, including radar node selection, is derived by combining the composite measurement information representing the same target. On this basis, a multi-sensor-network-optimization model for asynchronous multi-target tracking is established. That is, to minimize the asynchronous multi-target-tracking error as the optimization objective, the adaptive optimization design of the selection method of the sensor nodes in the sensor network is carried out, and the sequential quadratic programming (SQP) algorithm is used to select the most suitable sensor nodes for the AA fusion of the Gaussian components representing the same target. The simulation results show that compared with the existing algorithms, the proposed algorithm can effectively improve the asynchronous multi-target-tracking accuracy of multi-sensor networks.

Due to the special tectonic background and complex geological evolution characteristics of the South China Sea, reef dolomite reservoirs (sush as Well Xike 1) are widely developed. Based on the drilling core data of Well Xike 1, the structure and geochemical characteristics of dolomite reservoirs, including carbon, oxygen, hydrogen isotopes and REE were systematically studied using geochemical and petrological methods. It is found that the geochemical characteristics of REE show that the main diagenetic environment of dolomites is a low-temperature alkaline semi open oxidation environment; the carbon and oxygen isotopes of the dolomites are generally lack of correlation, the δD value is significantly lower than the hydrogen isotope value of seawater. Meanwhile, the oxygen isotope value of deep dolomites is negatively biased, which may be due to the increase or decrease of pore water temperature caused by deep thermal convection that related to the regional tectonic movements of the South China Sea. The δ18O value is also consistent with the geological reality of increasing saddle dolomite content in deep dolomites. The distribution of the δ13C value indicates that the dolomite inherited the carbon of the original limestone during dolomitization, while the characteristic of the δD value shows that it may be affected by the mixing of atmospheric precipitation and concentrated seawater in the quasi contemporaneous period. Based on the comprehensive analysis of the geochemical characteristics of the Well Xike 1, it is considered that the higher diagenetic temperature could be an important factor leading to the huge differences between the diagenetic model of deep and shallow dolomites. The geochemical characteristics of the shallow dolomites show that it is mainly reflux infiltration dolomitization under the micro evaporation and concentration sea water environments, while the deep dolomite is transformed by the hot water fluids in the epigenetic diagenetic evolution stage.

Fiber optic distributed acoustic sensing (DAS) technology is widely used in security surveillance and geophysical survey applications. The response of the DAS system to external vibrations varies with different types of fiber optic cable connections. The mechanism of mutual influence between the cable’s characteristics and DAS measurement results remains unclear. This study proposed a dynamic model of the interaction between the optical cable and the soil, analyzed the impact of the dynamic parameters of the optical cable and soil on the sensitivity of the DAS system, and validated the theoretical analysis through experiments. The findings suggest that augmenting the cable’s bending stiffness 5.5-fold and increasing its unit mass 4.2-fold result in a discernible reduction of the system’s response to roughly 0.15 times of its initial magnitude. Cables with lower unit mass and bending stiffness are more sensitive to vibration signals. This research provides a foundation for optimizing vibration-enhanced fiber optic cables and broadening the potential usage scenarios for DAS systems.

H7N9 avian influenza virus (AIV) has caused huge losses in the poultry industry and impacted human public health security, and still poses a potential threat. Currently, immune prevention and control of avian influenza relies on traditional inactivated vaccines; however, they have some limitations and genetically engineered avian influenza subunit vaccines may be potential candidate vaccines. In this study, a T169A mutation in the HA protein derived from H7N9 AIV A/Chicken/Guangdong/16876 (H7N9-16876) was generated using the baculovirus expression system (BVES). The results showed that the mutant (HAm) had significantly increased thermostability compared with the wild-type HA protein (HA-WT). Importantly, immunizing chickens with HAm combined with ISA 71VG elicited higher cross-reactive hemagglutination inhibition (HI) antibody responses and cytokine (IFN-γ and IL-4) secretion. After a lethal challenge with heterologous H7N9 AIV, the vaccine conferred chickens with 100% (10/10) clinical protection and effectively inhibited viral shedding, with 90% (9/10) of the chickens showing no virus shedding. The thermostability of HAm may represent an advantage in practical vaccine manufacture and application. In general, the HAm generated in this study represents a promising subunit vaccine candidate for the prevention and control of H7N9 avian influenza.

Hepatitis B virus (HBV) produces circular RNA (circRNA), whose functions have not yet been clearly elucidated. In this study, a novel circRNA HBV_circ_1 produced by HBV was identified in HBV-positive HepG2.2.15 cells and HBV-related hepatocellular carcinoma (HCC) tissue (HCCT). Microarray analysis of 68 HCCT samples showed that HBV_circ_1 abundance was significantly higher than that in paracancerous tissues. In addition, survival rate of HBV_circ_1-positive patients was significantly lower compared with HBV_circ_1-negative patients. Transient expression indicated that HBV_circ_1 enhanced cell proliferation, migration, and invasion and inhibited apoptosis in vitro. Furthermore, ectopical HBV_circ_1 expression increased tumor size in vivo. HBV_circ_1 was confirmed to interact with cyclin-dependent kinase 1 (CDK1) to regulate cell proliferation. These results suggest that HCC progression may be promoted by interaction of HBV_circ_1 with CDK1. Our data not only showed a novel clue to understand carcinogenesis and progress of HBV-related HCC but also provided a new target for the development of therapeutic drugs.

Introduction: Intrahepatic cholangiocarcinoma (iCCA) is a heterogeneous entity with diverse etiologies, morphologies, and clinical outcomes, but our knowledge of its epidemiology and carcinogenesis is very limited.Materials and methods: The expression patterns of circRNAs were explored in iCCA tissues and corresponding adjacent normal ones, denoted by (iCCA) and (iCCAP), respectively, using high-throughput sequencing.Results: A total of 117 differential expressed (DE) circRNAs were identified. Based on the parental transcripts of circRNAs, these DE circRNAs were related to several important GO terms and were enriched in important pathways. Two circRNA-mediated ceRNA networks were constructed and many important metabolic pathways related to mRNAs were regulated by DE circRNAs via miRNAs.Conclusion: Our study revealed the DE circRNAs in the iCCA tissues compared with iCCAP ones, suggesting that circRNAs may play crucial roles in the pathogenesis of iCCA.

Circular RNAs (circRNAs) as novel regulatory molecules have been recognized in diverse species, including viruses. The virus-derived circRNAs play various roles in the host biological process and the life cycle of the viruses. This review summarized the circRNAs from the DNA and RNA viruses and discussed the biogenesis of viral and host circRNAs, the potential roles of viral circRNAs, and their future perspective. This review will elaborate on new insights gained on viruses encoded circRNAs during virus infection.

Preparation of a high-sensitive and uniform surface-enhanced Raman spectroscopy (SERS) active substrate structure usually faces complex processes and high costs. Here, porous Au–Ag composite nanostructures that are conventional fabricated by the deposition of a multilayer Au–Ag, annealing, and dealloying process are proposed for high-performance SERS. By annealing at a suitable temperature, nanopores could be firmly distributed on the surface, which serves as hot spots. The electric field distribution was also performed by the finite difference time domain. The experiment results exhibited excellent uniformity and high sensitivity of SERS detection. The enhancement factor of the R6G molecules detected by the SERS substrate reached 1.37 × 107, and the relative standard deviation was as low as 4.9%. The minimum detection concentration of R6G molecules by the Au–Ag composite nanostructures with bottom Au mirror could reach 10−13 M. The proposed Au–Ag composite nanostructures and the fabrication process have great potential in preparation of a high-sensitive and uniform SERS substrate.

Circular DNAs derived from single-stranded RNA viruses play important roles in counteracting viral infection. However, whether double-stranded RNA viruses generate functional circular DNAs is still unknown. Using circDNA sequencing, divergent PCR, DNA in situ hybridization and rolling circular amplification, we presently confirmed that in silkworm, Bombyx mori cytoplasmic polyhedrosis virus (BmCPV), a double-stranded RNA virus belonging to cypovirus, is prone to produce a BmCPV-derived circular DNA termed as vcDNA-S7. We have also found that vcDNA-S7 formation is mediated by endogenous reverse transcriptase (RT), and the proliferation of BmCPV can be inhibited by vcDNA-S7 in vitro and in vivo. Moreover, we have discovered that the silkworm RNAi immune pathway is activated by vcDNA-S7, while viral small interfering RNAs (vsiRNAs) derived from transcribed RNA by vcDNA-S7 can be detected by small RNA deep sequencing. These results suggest that BmCPV-derived vcDNA-S7, mediated by RT, can serve as a template for the biogenesis of antiviral siRNAs, which may lead to the repression of BmCPV infection. To our knowledge, this is the first demonstration that a circular DNA, produced by double stranded RNA viruses, is capable of regulating virus infection.

Bombyx mori nucleopolyhedrovirus (BmNPV) is one of the most serious pathogens and causes serious economic losses in sericulture. At present, there is no epigenetic modification of BmNPV transcripts, especially of m6A, and this modification mediates diverse cellular and viral functions. This study showed that m6A modifications are widespread in BmNPV transcripts in virally infected cells and the identified m6A peaks with a conserved RRACH sequence. m6A sites predominantly appear in the coding sequences (CDS) and the 3′-end of CDS. About 37% of viral genes with m6A sites deleted from the viral genome did not produce any infectious virions in KOV-transfected cells. Among the viral genes related to replication and proliferation, ie-1 mRNA was identified with a higher m6A level than other viral genes. The m6A sites in the ie-1 mRNA may be negatively related to the protein expression. Viral replication was markedly inhibited in cells overexpressed with BmYTHDF3 in a dose-dependent manner, and a contrary effect was found in si-BmYTHDF3-transfected cells. Collectively, the identification of putative m6A modification in BmNPV transcripts provides a foundation for comprehensively understanding the viral infection, replication, and pathobiology in silkworms.

Circular RNAs are widespread in various species and have important roles in myogenesis. However, the circular RNAs involved in breast muscle development in ducks have not yet been studied. Here, to identify circular RNAs during duck skeletal muscle development, three pectorales from Shan Ma ducks at E13 and E19, which represent undifferentiated and differentiated myoblasts, respectively, were collected and subjected to RNA sequencing. A total of 16,622 circular RNAs were identified, of which approximately 80% were exonic circular RNAs and 260 were markedly differentially expressed between E19 and E13. The parental genes of the differentially expressed circular RNAs were significantly enriched in muscle-related biological processes. Moreover, we found that the overexpression of circGAS2-2 promoted cell cycle progression and increased the proliferation viability of duck primary myoblasts; conversely, knockdown of circGAS2-2 retarded the cell cycle and reduced the proliferation viability of myoblasts. Taken together, our results demonstrate that circular RNAs are widespread and variously expressed during the development of duck skeletal muscle and that circGAS2-2 is involved in the regulation of myogenesis.

Abstract Excessive ultraviolet (UV) radiation exposure results in skin chronic photodamage via stimulating reactive oxygen species (ROS) and inflammation. Anthocyanins are a group of flavonoids frequently found in edible plants. Cyanidin‐3‐O‐glucoside (C3G) as a typical anthocyanin shows effective anti‐oxidative and anti‐inflammatory properties. This study aims to investigate whether the topical application of C3G moisturizing gel on mice can protect the skin from UVB‐induced chronic photodamage. The results of in vitro experiment showed that the active ingredient C3G can penetrate the mice skin. The dorsal of Kunming mice were treated with C3G moisturizing gel (100, 200, 300 μmol/L) after UVB exposure. The animal experiment demonstrated that C3G can reduce chronic photodamage caused by UVB. C3G could effectively ameliorate the UVB‐induced epidermal barrier dysfunction including an increase in the skin hydration and decrease in the transepidermal water loss, and have statistically significance. Besides, our results also indicated that C3G inhibited UVB‐induced epidermal hyperplasia, the destruction of collagen fibers, ROS levels, and the expression of COX‐2 and IL‐6. In brief, these results indicate that C3G can reduce UVB‐induced chronic photodamage by inhibiting oxidative stress and inflammation.

The continuous evolution of the H7N9 avian influenza virus suggests a potential outbreak of an H7N9 pandemic. Therefore, to prevent a potential epidemic of the H7N9 influenza virus, it is necessary to develop an effective crossprotective influenza vaccine. In this study, we developed H7N9 virus-like particles (VLPs) containing HA, NA, and M1 proteins derived from H7N9/16876 virus and a helper antigen HMN based on influenza conserved epitopes using a baculovirus expression vector system (BEVS). The results showed that the influenza VLP vaccine induced a strong HI antibody response and provided effective protection comparable with the effects of commercial inactivated H7N9 vaccines against homologous H7N9 virus challenge in chickens. Meanwhile, the H7N9 VLP vaccine induced robust crossreactive HI and neutralizing antibody titers against antigenically divergent H7N9 viruses isolated in wave 5 and conferred on chickens complete clinical protection against heterologous H7N9 virus challenge, significantly inhibiting virus shedding in chickens. Importantly, supplemented vaccination with HMN antigen can enhance Th1 immune responses; virus shedding was completely abolished in the vaccinated chickens. Our study also demonstrated that viral receptor-binding avidity should be taken into consideration in evaluating an H7N9 candidate vaccine. These studies suggested that supplementing influenza VLP vaccine with recombinant epitope antigen will be a promising strategy for the development of broad-spectrum influenza vaccines.

The taxonomy of the Blue-crowned Laughingthrush (Garrulax courtoisi) and its relationship with the Yellow-throated Laughingthrush (G. galbanus) and G. c. simaoensis, a range-restricted subspecies in China, has not been fully elucidated. So the taxonomic status and system evolution of the three taxa G. courtoisi, G. galbanus and G. c. simaoensis need to be reclarified. Two gene sequences myoglobin (MYO) and the mitochondrial cytochrome coxidase subunit I (COI) were combined to investigate the phylogenetic relationships among courtoisi, simaoensis and galbanus, genetic data, combining with morphological, ecological and acoustic data were used to comb out the classification status and divergence level of the three taxa. Significant genetic and morphological differentiations (body size and plumage coloration) were detected between courtoisi and galbanus. However, no notable and reliable differences between the courtoisi and simaoensis were detected. The courtoisi, simaoensis and galbanus are clearly isolated in geographical distribution as a result of differing altitudes, climate conditions and habitats. The courtoisi has characteristic preference for nest location compared with galbanus. In addition, the results of song analysis also indicated that there are differences in maximum frequency between courtoisi and galbanus. G. courtoisi was confirmed to be an independent species based on genetic, morphological, geographical, ecological and vocal characteristics, and the validity of simaoensis as a subspecies still need more evidence. This study further confirmed the high conservation value of Blue-crowned Laughingthrush. In addition, due to the genetic differences between Simao and Wuyuan populations, this should be fully considered in future protection strategies.

Purpose. To analyze the application value of multidisciplinary diagnosis and treatment (MDT) nursing mode based on doctor-nurse-integration for stroke patients undergoing emergency intervention surgery. Methods. In this study, a historical comparative study method was adopted. 118 stroke patients and medical staff (9 doctors and 11 nurses) who met the diagnosis and inclusion criteria of emergency intervention surgery admitted from July 2021 to February 2022 were treated clinically according to the traditional medical care mode (TMC group), 87 stroke patients and medical staff (9 doctors and 11 nurses) who met the diagnosis and inclusion criteria of emergency intervention surgery admitted from February 2022 to June 2022 were treated and cared according to the MDT nursing mode based on medical integration (MDT group). Comparison of perioperative time indicators, postoperative outcome indicators, treatment compliance, secondary complications and visit satisfaction between the two groups of patients, and comparison of cooperation satisfaction between the two groups of medical staff. ObjectiveThe aim of this study was to investigate the efficacy and safety of combined applications of local consolidative radiation therapy (LCRT) and first-line tyrosine kinase inhibitors (TKIs) for the treatment of primary tumors and oligometastatic sites in oligometastatic NSCLC harboring Epidermal Growth Factor Receptor (EGFR) activating mutations.Patients and MethodsElderly patients with oligometastatic NSCLC (≤5 metastases) harboring EGFR activating mutations at the time of diagnosis were identified. They were treated with first-line TKIs alone or in combination with LCRT. Progression‐free survival (PFS) and overall survival (OS) were estimated through the Kaplan–Meier method.ResultsA total of 122 elderly patients were enrolled between February 2010 and January 2018. Among them, 41.0% (n = 50) received TKIs combined with LCRT (TKIs + LCRT group), whereas 59.0% (n = 72) received TKIs monotherapy (TKIs alone group). Patients were followed up for a median length of 34 months (ranging from 7.0 to 64 months). The discharge of mine water from underground coal mines in arid areas is leading to extensive water loss and secondary pollution. To eliminate the water loss and environmental pollution, sealing the concentrated brine after the mine water treatment in the underground storage and pumping the clean water to the surface for recycling are effective methods. In this study, focusing on the Ling Xin mining area, China, a coupled model of concentrated brine flow and solute transport in underground reservoir was established. The migration patterns of concentrated brine under two simulation scenarios of long-term penetration and sudden leakage were analyzed. At the same time, it also initially revealed the essence of the environmental pollution caused by the penetration and leakage of the concentrated brine in the underground concentrated brine storage reservoir. Results show that the concentrated brine would penetrate the bottom aquifer in about 60 years in long-term penetration while approximately 40 days in sudden leakage. In addition, the storage time, reservoir permeability, and groundwater head difference were important factors affecting the migration of concentrated brine, where the influence of permeability varieties was the most significant. The results of this study provide technical options for the subsequent study of the environmental risk of underground concentrated brine reservoirs and have important technical significance for the study and engineering application of underground reservoirs in arid areas.

Flow characteristics and phase distribution of concentrated brine storage in mining area have been core factors linking to the leakage risk assessment and ecological evaluation. Notably, saturation plays a crucial role in impacting the flow characteristics and distribution of brine, while the existence of oil left by mining machines and original reservoir and gas produced from coal bed gas and air has complicated the issue. In this work, we conducted the microfluidic visualization experiments to reveal the saturation distribution during brine storage in mining area. We applied machine learning model to extract saturation data from experimental images with over 95% accuracy. Eventually, we found that the existence of gas significantly impacts on the saturation distribution in micropores accounting for more than 80% contribution. We clarified that the gas production rate of median 200 μL/min impacts the least on saturation variation. Results in this research are of significance for deeper comprehension on three-phase saturation characteristics of concentrated brine storage in mining area.

ABSTRACT This study aimed to compare the fecal microbiome and antimicrobial resistance between captive and free-range sika deer with the same exposure to antibiotic anthelmintics. The taxonomic differences mainly involved significant changes in the dominant phyla, genera, and species. Linear discriminant analysis effect size (LEfSe) analysis revealed that 22 taxa were significantly different between the two groups. The KEGG analysis showed that the fecal microbiome metabolic function, and all level 2 categories in metabolism had higher abundance in the free-range deer. Based on the carbohydrate-active enzyme (CAZy) database analysis, glycoside hydrolases and carbohydrate-binding modules showed remarkable differences between the two groups. Regarding antibiotic resistance, tetQ and lnuC dominated the antibiotic resistance ontology (ARO) terms, and tetracycline and lincosamide resistance dominated the antimicrobial resistance patterns. Furthermore, the lnuC, ErmF, and tetW/N/W AROs and lincosamide resistance showed higher abundance in the captive deer, suggesting that captivity may yield more serious resistance issues because of the differences in greenfeed diet, breeding density, and/or housing environment. The results also revealed important associations between the phylum Proteobacteria, genus Prevotella, and major antibiotic resistance genes. Although the present study was a pilot study with a limited sample size that was insufficient control for some potential factors, it serves as the metagenomic study on the microbial communities and antimicrobial resistance in sika deer. IMPORTANCE We used a metagenomic approach to investigate whether and how captive and free-range impact the microbial communities and antimicrobial resistance in sika deer. The results provide solid evidence of the significant impacts on the microbial composition and function in captive and free-range sika deer. Interestingly, although the sika deer had the same exposure to antibiotic anthelmintics, the antimicrobial resistances were affected by the breeding environment.

Abstract Background The protective efficacy of avian influenza virus (AIV) vaccines is unsatisfactory due to the presence of various serotypes generated by genetic reassortment. Thus, immunization with a polyantigen chimeric epitope vaccine may be an effective strategy for protecting poultry from infection with different AIV subtypes. Methods Baculovirus has recently emerged as a novel and attractive gene delivery vehicle for animal cells. In the present study, a recombinant baculovirus BmNPV-CMV/THB-P10/CTLT containing a fused codon-optimized sequence (CTLT) of T lymphocyte epitopes from H1HA, H9HA, and H7HA AIV subtypes, and another fused codon-optimized sequence (THB) of Th and B cell epitopes from H1HA, H9HA, and H7HA AIV subtypes, driven by a baculovirus P10 promoter and cytomegalovirus CMV promoter, respectively, was constructed. Results Western blotting and cellular immunofluorescence demonstrated that the CTLT (THB) can be expressed in rBac-CMV/THB-P10/CTLT-infected silkworm cells (mammalian HEK293T cells). Furthermore, the recombinant virus, rBac-CMV-THB-CTLT, was used to immunize both chickens and mice. Conclusions The results of an indirect ELISA, immunohistochemistry, and T lymphocyte proliferation assay indicated that specific humoral and cellular responses were detected in both chicken and mice. These results suggest that rBac-CMV/THB-P10/CTLT can be developed as a potential vaccine against different AIV subtypes.

Compared with traditional machine tools, the micro machine tools have advantages of small volume, low cost, less energy consumption, high efficiency and high flexibility. Therefore, it is regarded as an important equipment for micro-cutting machining which has been used widely all over the world and. As a key component of the micro-cutting machine tools, the body structure directly influences the machining performance. Thus, an integral column and base structure for micro machining tools was proposed in this work, and the detailed structural parameters were designed based on parameter analysis. Besides, the static and dynamic performance of the proposed machine were analyzed and compared between the integral structure and the separated one. The deformation and stress of the proposed structures under typical working conditions were studied by numerical simulation, along with the natural frequencies, vibration modes and frequency response peaks. Further, optimization was performed on the integral body structure, the prototype of the micro-machine tool was trial-produced, and the positioning accuracy of each coordinate axis was qualitatively analyzed. In addition, the micro-milling test was carried out with 6061 aluminum alloy to show the performance of the novel cutting machine. The results revealed that the proposed integrated micro-machine bed structure is superior to the separated structure in terms of static deformation and harmonic response characteristics, with good comprehensive mechanical properties, greatly improved static and dynamic performance of the machine tool, significantly improved structural accuracy, improved processing quality of the specimen and good application value.

Doctors usually diagnose a disease by evaluating the pattern of abnormal blood vessels in the fundus. At present, the segmentation of fundus blood vessels based on deep learning has achieved great success, but it still faces the problems of low accuracy and capillary rupture. A good vessel segmentation method can guide the early diagnosis of eye diseases, so we propose a novel hybrid Transformer network (HT-Net) for fundus imaging analysis. HT-Net can improve the vessel segmentation quality by capturing detailed local information and implementing long-range information interactions, and it mainly consists of the following blocks. The feature fusion block (FFB) is embedded in the shallow levels, and FFB enriches the feature space. In addition, the feature refinement block (FRB) is added to the shallow position of the network, which solves the problem of vessel scale change by fusing multi-scale feature information to improve the accuracy of segmentation. Finally, HT-Net’s bottom-level position can capture remote dependencies by combining the Transformer and CNN. We prove the performance of HT-Net on the DRIVE, CHASE_DB1, and STARE datasets. The experiment shows that FFB and FRB can effectively improve the quality of microvessel segmentation by extracting multi-scale information. Embedding efficient self-attention mechanisms in the network can effectively improve the vessel segmentation accuracy. The HT-Net exceeds most existing methods, indicating that it can perform the task of vessel segmentation competently.

N6-Methyladenosine is a reversible epigenetic modification that influences muscle development. However, the m6A modification profile during poultry skeletal muscle development is poorly understood. Here, we utilized m6A-specific methylated RNA immunoprecipitation sequencing to identify m6A sites during two stages of breast muscle development in ducks: embryonic days 13 (E13) and E19. MeRIP-seq detected 19,024 and 18,081 m6A peaks in the E13 and E19 groups, respectively. Similarly to m6A distribution in mammalian transcripts, our results revealed GGACU as the main m6A motif in duck breast muscle; they also revealed that m6A peaks are mainly enriched near the stop codons. In addition, motif sequence analysis and gene expression analysis demonstrated that m6A modification in duck embryo skeletal muscles may be mediated by the methyltransferase-like 14. GO and KEGG analysis showed that m6A peaks containing genes at E19 were mainly enriched in muscle-differentiation- and muscle-growth-related pathways, whereas m6A peaks containing genes in E13 were mainly enriched in embryonic development and cell proliferation pathways. Combined analysis of MeRIP-seq and RNA-seq showed that the mRNA expression may be affected by m6A modification. Moreover, qRT-PCR analysis of the expression of METTL14 and its cofactors (WTAP, ZC3H13, RBM15 and VIRMA) during duck embryonic skeletal muscle development in breast and leg muscle samples revealed a significant downward trend as the developmental age progressed. Our results demonstrated that m6A mRNA methylation modifications control muscle development in ducks. This is the first study of m6A modification patterns in duck muscle tissue development, and it lays the foundation for the study of the effects of RNA modification on poultry skeletal muscle development.

Corn harvester row alignment is a critical factor to improve harvesting quality and reduce cob drop loss. In this paper, a corn harvester automatic row alignment system is designed with a self-propelled corn combine harvester, incorporating a front-end touch row alignment mechanism of the cutting table and a harvester steering control system. Corn stalk lateral deviation from the reference row during harvesting is detected by a front-end touch-row alignment mechanism that serves as an input to the automatic alignment system. The harvester steering control system consists of Hirschmann PLC controller, electric steering wheel, steering wheel deflection angle detection device, display module and mode selection module, etc. The adaptive fuzzy PID control algorithm is used to determine the desired turning angle of the harvester steering wheel by combining with the harvester kinematic model, and the model is simulated and analyzed by Matlab/Simulink software. The automatic row alignment system was mounted on a 4LZ-8 self-propelled corn harvester for field tests, and the test results showed that the average percentage of deviation of corn stalks from the center of the row alignment cutting path within ±15 cm during the automatic row alignment process was 95.4% at harvester speeds ranging from 0 to 4.6 km/h, which could meet the requirements of the corn harvester for row alignment harvesting. The test results meet the requirements of the corn harvester for row alignment and serve as a benchmark for the research on automatic row alignment of corn harvesters.

Background The Père David’s deer (Elaphurus davidianus) population was established from only a small number of individuals. Their genetic diversity is therefore relatively low and transmissible (parasitic) diseases affecting them merit further attention. Parasitic infections can affect the health, survival, and population development of the host. However, few reports have been published on the gastrointestinal parasites of Père David’s deer. The aims of this study were: (1) to identify the intestinal parasites groups in Père David’s deer; (2) to determine their prevalence and burden and clarify the effects of different seasons and regions on various indicators of Père David’s deer intestinal parasites; (3) to evaluate the effects of the Père David’s deer reproductive period on these parasites; (4) to reveal the regularity of the parasites in space and time. Methods In total, 1,345 Père David’s deer faecal samples from four regions during four seasons were tested using the flotation (saturated sodium nitrate solution) to identify parasites of different genus or group, and the McMaster technique to count the number of eggs or oocysts. Results Four groups of gastrointestinal parasites were found, of which strongyles were dominant; their prevalence and burden were significantly higher than other groups. Significant temporal and spatial effects on gastrointestinal parasitic infection were found. Parasite diversity, prevalence, parasite burden, and aggregation were the highest in summer. Among the four regions, parasite diversity, prevalence, and burden were the highest in the Dongting Lake area. In addition, parasite diversity and burden during the reproductive period of Père David’s deer was significantly higher than during the post-reproductive period. Conclusions The summer season and the reproductive period of Père David’s deer had great potential for parasite transmission, and there is a high risk of parasite outbreaks in the Dongting Lake area.

Abstract Background In our previous study, we identified four isoforms of the Bmovo gene, Bmovo-1, Bmovo-2, Bmovo-3 and Bmovo-4 from the silkworm ovary and verified that ovarian development was regulated by the BmOVO proteins. Results: To understand the regulatory mechanisms of ovarian development, the regulation of four BmOVO isoforms on the B. mori ovarian tumor (Bmotu) promoter activity was investigated with luciferase reporter assays. The results showed the Bmotu promoter activity was positively regulated by BmOVO-1, BmOVO-2, BmOVO-3 and BmOVO-4 in a dose-dependent manner, of which BmOVO-2 had the highest transcriptional activation. However, the first (A1) and third acidic domains (A3) at the N-terminus of BmOVO-1 are transcriptional repression domains, while the fourth (A4) and fifth acidic domains (A5) are transcriptional activation domains. A recombinant BmOVO zinc-finger domain was found to bind to the GTACCGTTGTA sequence located at the Bmotu promoter. Furthermore, the Bmotu promoter activity was negatively regulated by ‘Tal-like’ peptide, which can trigger BmOVO-1 degradation at the N-terminus. Conclusions These results will help us to further understand the regulatory mechanisms of BmOVO isoforms on Bmotu promoter activity and ovarian development in the silkworm.

The cardinal symptoms of some ophthalmic diseases observed through exceptional retinal blood vessels, such as retinal vein occlusion, diabetic retinopathy, etc. The advanced deep learning models used to obtain morphological and structural information of blood vessels automatically are conducive to the early treatment and initiative prevention of ophthalmic diseases. In our work, we propose a hierarchical dilation convolutional network (HDC-Net) to extract retinal vessels in a pixel-to-pixel manner. It utilizes the hierarchical dilation convolution (HDC) module to capture the fragile retinal blood vessels usually neglected by other methods. An improved residual dual efficient channel attention (RDECA) module can infer more delicate channel information to reinforce the discriminative capability of the model. The structured Dropblock can help our HDC-Net model to solve the network overfitting effectively. From a holistic perspective, the segmentation results obtained by HDC-Net are superior to other deep learning methods on three acknowledged datasets (DRIVE, CHASE-DB1, STARE), the sensitivity, specificity, accuracy, f1-score and AUC score are {0.8252, 0.9829, 0.9692, 0.8239, 0.9871}, {0.8227, 0.9853, 0.9745, 0.8113, 0.9884}, and {0.8369, 0.9866, 0.9751, 0.8385, 0.9913}, respectively. It surpasses most other advanced retinal vessel segmentation models. Qualitative and quantitative analysis demonstrates that HDC-Net can fulfill the task of retinal vessel segmentation efficiently and accurately.

Planar, nanopillar and Al nanosphere structure AlGaN-based deep-ultraviolet light-emitting diodes (DUV-LEDs) were numerically investigated via a three-dimensional finite difference time domain (3D FDTD) method. The three types of DUV-LEDs were compared and analyzed in terms of light extraction efficiency (LEE), Purcell factor (FP) and modulation bandwidth. The results showed that nanopillar structure DUV-LEDs with optimal nanopillar height, width and spacing can enhance transverse electric (TE)-polarized LEE to 39.7% and transverse magnetic (TM)-polarized LEE to 4.4%. The remarkable improvement was mainly due to the increased scattering effect, decreased absorption of the p-GaN layer and total internal reflection (TIR) effect. After adopting the Al nanospheres, the TE-polarized modulation bandwidth was increased by 71 MHz and the TM-polarized LEE was enhanced approximately 4.3-fold as compared to the nanopillar LED structure, while the Al nanosphere diameter was 120 nm. The reasons for promotion are mainly attributed to the coupling behavior of diploe and localized surface plasmon induced by Al nanospheres. The designed structures provide a meaningful solution for realization of high-efficiency DUV-LEDs.
