An evolutionarily conserved region of the TDP-43 low-complexity domain (LCD) twenty residues in length can adopt either an α-helical or β-strand conformation. When in the latter conformation, TDP-43 self-associates via the formation of a labile, cross-β structure. Self-association can be monitored via the formation of phase-separated protein droplets. Exposure of droplets to hydrogen peroxide leads to oxidation of conserved methionine residues distributed throughout the LCD. Oxidation disassembles the cross-β structure, thus eliminating both self-association and phase separation. Here, we demonstrate that this process reciprocally enables formation of α-helical structure in precisely the same region formerly functioning to facilitate β-strand-mediated self-association. We further observe that the α-helical conformation allows interaction with a lipid-like detergent and that exposure to lipids enhances the β-to-α conformational switch. We hypothesize that regulation of this oxidative switch will prove to be important to the control of localized translation within vertebrate cells. The experimental observations reported herein were heavily reliant on studies of 1,6-hexanediol, a chemical agent that selectively dissolves labile structures formed via the self-association of protein domains of low sequence complexity. This aliphatic alcohol is shown to exert its dissociative activity primarily via hydrogen-bonding interactions with carbonyl oxygen atoms of the polypeptide backbone. Members of the nucleobase/ascorbic acid transporter (NAT) gene family are found in all kingdoms of life. In mammals, the concentrative uptake of ascorbic acid (vitamin C) by members of the NAT family is driven by the Na+ gradient, while the uptake of nucleobases in bacteria is powered by the H+ gradient. Here, we report the structure and function of PurTCp, a NAT family member from Colwellia psychrerythraea. The structure of PurTCp was determined to 2.80 Å resolution by X-ray crystallography. PurTCp forms a homodimer, and each protomer has 14 transmembrane segments folded into a transport domain (core domain) and a scaffold domain (gate domain). A purine base is present in the structure and defines the location of the substrate binding site. Functional studies reveal that PurTCp transports purines but not pyrimidines and that purine binding and transport is dependent on the pH. Mutation of a conserved aspartate residue close to the substrate binding site reveals the critical role of this residue in H+-dependent transport of purines. Comparison of the PurTCp structure with transporters of the same structural fold suggests that rigid-body motions of the substrate-binding domain are central for substrate translocation across the membrane. We present analytical and numerical analyses of a yet unseen lensing paradigm that is based on a solid metamaterial slab in which the wave excitation source is attached. We propose and demonstrate sub-diffraction-limited acoustic focusing induced by surface resonant states in doubly negative metamaterials. The enhancement of evanescent waves across the metamaterial slab produced by their resonant coupling to surface waves is evidenced and quantitatively determined. The effect of metamaterial parameters on surface states, transmission, and wavenumber bandwidth is clearly identified. Based on this concept consisting of a wave source attached on the metamaterial, a high resolution of λ/28.4 is obtained with the optimum effective physical parameters, opening then an exciting way to design acoustic metamaterials for ultrasonic focused imaging. We report theoretically and numerically on the sound transmission loss performance through a thick plate-type acoustic metamaterial made of spring-mass resonators attached to the surface of a homogeneous elastic plate. Two general analytical approaches based on plane wave expansion were developed to calculate both the sound transmission loss through the metamaterial plate (thick and thin) and its band structure. The first one can be applied to thick plate systems to study the sound transmission for any normal or oblique incident sound pressure. The second approach gives the metamaterial dispersion behavior to describe the vibrational motions of the plate, which helps to understand the physics behind sound radiation through air by the structure. Computed results show that high sound transmission loss up to 72 dB at 2 kHz is reached with a thick metamaterial plate while only 23 dB can be obtained for a simple homogeneous plate with the same thickness. Such plate-type acoustic metamaterial can be a very effective solution for high performance sound insulation and structural vibration shielding in the very low-frequency range. Low complexity (LC) head domains 92 and 108 residues in length are, respectively, required for assembly of neurofilament light (NFL) and desmin intermediate filaments (IFs). As studied in isolation, these IF head domains interconvert between states of conformational disorder and labile, β-strand-enriched polymers. Solid-state NMR (ss-NMR) spectroscopic studies of NFL and desmin head domain polymers reveal spectral patterns consistent with structural order. A combination of intein chemistry and segmental isotope labeling allowed preparation of fully assembled NFL and desmin IFs that could also be studied by ss-NMR. Assembled IFs revealed spectra overlapping with those observed for β-strand-enriched polymers formed from the isolated NFL and desmin head domains. Phosphorylation and diseasecausing mutations reciprocally alter NFL and desmin head domain self-association yet commonly impede IF assembly. These observations show how facultative structural assembly of LC domains via labile, β-strand-enriched self-interactions may broadly influence cell morphology. [ABSTRACT FROM AUTHOR]

A methionine-rich low complexity (LC) domain is found within a C-terminal region of the TDP43 RNA-binding protein. Self-association of this domain leads to the formation of labile cross-ß polymers and liquid-like droplets. Treatment with H2O2 caused phenomena of methionine oxidation and dropletmelting thatwere reversed upon exposure of the oxidized protein to methionine sulfoxide reductase enzymes. Morphological features of the cross-β polymers were revealed by H2O2-mediated footprinting. Equivalent TDP43 LC domain footprints were observed in polymerized hydrogels, liquid-like droplets, and living cells. The ability of H2O2 to impede cross-β polymerization was abrogated by the prominent M337V amyotrophic lateral sclerosis-causing mutation. These observations may offer insight into the biological role of TDP43 in facilitating synapse-localized translation as well as aberrant aggregation of the protein in neurodegenerative diseases. A modulated metamaterial that exhibits both time-periodic stiffness and mass simultaneously is presented. The metamaterial element includes a primary body that undergoes infinitesimal motion, and is connected to a dynamic-mechanism structure, involving a rotational body, and spring with a large-scale motion, which is designed to produce a time-modulated linear momentum and elastic constraint for the primary body. The non-reciprocal wave propagation is then investigated in a space-time lattice metamaterial that is constructed by coupling doubly time-modulated elements with linear springs of constant stiffness. The dispersion property shows the frequency degeneracy occurring at the center or edge of the Brillouin zone, and the unidirectional bandgap at certain frequencies. This phenomenon represents a unique property of the doubly modulated metamaterials compared to the singly modulated ones, thus may provide more promising applications to the design of non-reciprocal devices. Platelet-derived growth factor receptor β (PDGFRβ) dynamically changes after brain injury, possibly mediating the neuroprotective role of soluble homodimers of the platelet-derived growth factor β subunit (PDGF-BB) that is secreted by microcirculation cells. The aim of this study was to determine whether binding of PDGF-BB to astrocytic PDGFRβ enhanced crosstalk among the various components of the neurovascular unit, leading to synaptic recovery after subarachnoid hemorrhage (SAH). The soluble PDGF-BB from the cerebrospinal fluid (CSF) of patients with SAH was measured. The relationship between PDGF-BB treatment and astrocytic PDGFRβ signaling was further explored in vivo and in vitro in experimental SAH models. Compared with the levels in the control samples, the PDGF-BB protein levels in the CSF of patients with SAH were significantly increased. After the generation of experimental SAH, astrocyte activation markers were markedly induced by the binding of PDGF-BB to astrocytic PDGFRβ, accompanied by improved levels of synaptic recovery and cognitive function. Soluble PDGF-BB and astrocytic PDGFRβ signaling are essential for the neuroprotective effect in the hippocampus and the coculture system in vitro after SAH that otherwise leads to cognitive dysfunction and neuronal damage. Human genetic studies have given evidence of familial, diseasecausing mutations in the analogous amino acid residue shared by three related RNA binding proteins causative of three neurological diseases. Alteration of aspartic acid residue 290 of hnRNPA2 to valine is believed to predispose patients to multisystem proteinopathy. Mutation of aspartic acid 262 of hnRNPA1 to either valine or asparagine has been linked to either amyotrophic lateral sclerosis or multisystem proteinopathy. Mutation of aspartic acid 378 of hnRNPDL to either asparagine or histidine has been associated with limb girdle muscular dystrophy. All three of these aspartic acid residues map to evolutionarily conserved regions of low-complexity (LC) sequence that may function in states of either intrinsic disorder or labile self-association. Here, we present a combination of solid-state NMR spectroscopy with segmental isotope labeling and electron microscopy on the LC domain of the hnRNPA2 protein. We show that, for both the wild-type protein and the aspartic acid 290-to-valine mutant, labile polymers are formed in which the LC domain associates into an in-register cross-β conformation. Aspartic acid 290 is shown to be charged at physiological pH and immobilized within the polymer core. Polymers of the aspartic acid 290-to-valine mutant are thermodynamically more stable than wild-type polymers. These observations give evidence that removal of destabilizing electrostatic interactions may be responsible for the increased propensity of the mutated LC domains to self-associate in diseasecausing conformations. Electrochemiluminescence (ECL) has been engineered to perform various tasks in the area of immunoassays and molecular diagnosis. However, there is still substantial potential for developments of ECL assay with high efficiency to achieve trace analysis. Herein, we demonstrate a polymer-amplified ECL assay via construction of linear Ru(bpy)32+ polymer. This new polymer material compensates for the relatively low ECL intensity from single ECL luminophore and realizes a stable and controllable labeling process. The polymer-amplified ECL assay achieved a remarkable sensitivity of 100 amol. The wide-ranging applications of the polymer-amplified ECL assay for Hepatitis B virus, carcinoembryonic antigen, 16sRNA, and thrombin also demonstrate its superiority. Hence, the polymer-amplified ECL assay possesses the potential to create a new paradigm in amplified ECL assays that could provide outstanding performance for biomedical analysis. The present study investigated whether erythromycin (ERY) reduces cigarette smoke (CS)-induced emphysema in rats and aimed to determine the anti-inflammatory effect of ERY, which may identify potential treatments for chronic obstructive pulmonary disease. Furthermore, the current study focused on the potential effects on the imbalance between matrix metalloprotease (MMP) and anti-MMP activity, the phosphorylation of mitogen-activated protein kinases (MAPKs) and the nuclear factor-κB (NF-κB) signaling pathway. Wistar rats were divided into the following three groups (n=12 each): control (ERY vehicle only, without any CS exposure), CS (animals were exposed to CS for 12 weeks) and CS + ERY (animals were exposed to CS for 12 weeks and received 100 mg/kg/day ERY). The recruitment of inflammatory cells into the bronchoalveolar lavage fluid (BALF) and the histopathology of lung tissue from all groups was evaluated to grade the severity of the emphysema. The expression of MMP-2, MMP-9 and tissue inhibitor of metalloproteinase-1 was evaluated by immunohistochemistry and western blotting. The activation of MAPKs, NF-κB and inhibitor of NF-κB (IκBα), in lung tissues was examined by western blotting. Treatment with ERY resulted in fewer inflammatory cells and cytokines in the BALF, and fewer emphysema-associated changes in the lungs compared with control. The stimulus of CS promoted the phosphorylation of extracellular signal-regulated kinase (ERK)1/2 and p38, but not c-Jun NH2-terminal kinase, thereby inducing the activation of the ERK/MAPK signaling pathway in rats. Furthermore, CS exposure increased the expression of NF-κB and decreased the expression of IκBα. The levels of phosphorylated ERK1/2 and p38 were significantly reduced in rats with CS-induced emphysema when treated with ERY compared with the CS group. The results of the present study therefore indicate that oral administration of ERY may suppress CS-induced emphysema by regulating inflammatory cytokines and the MMP/anti-MMP imbalance via the MAPK/NF-κB pathway. The authors study the wave propagation in continuum acoustic metamaterials whose all or not all of the principal elements of the mass tensor or the scalar compressibility can be negative due to wave dispersion. Their time-domain wave characteristics are particularly investigated by the finite-difference time-domain (FDTD) method, in which algorithms for the Drude and Lorentz dispersion pertinent to acoustic metamaterials are provided necessarily. Wave propagation nature of anisotropic acoustic metamaterials with all admissible material parameters are analyzed in a general manner. It is found that anomalous negative refraction phenomena can appear in several dispersion regimes, and their unique time-domain signatures have been discovered by the FDTD modeling. It is further proposed that two different metamaterial layers with specially assigned dispersions could comprise a conjugate pair that permits wave propagation only at specific points in the wave vector space. The time-domain pulse simulation verifies that acoustic directive radiation capable of modulating radiation angle with the wave frequency can be realized with this conjugate pair. The study provides the detailed analysis of wave propagation in anisotropic and dispersive acoustic mediums, which makes a further step toward dispersion engineering and transient wave control through acoustic metamaterials. Previous prototypes of acoustic hyperlens consist of rigid channels, which are unable to adapt in shape to the object under detection. We propose to overcome this limitation by employing soft plastic tubes that could guide acoustics with robustness against bending deformation. Based on the idea of soft-tube acoustics, acoustic magnifying hyperlens with planar input and output surfaces has been fabricated and validated experimentally. The shape-adaption capability of the soft-tube hyperlens is demonstrated by a controlled experiment, in which the magnifying super-resolution images remain stable when the lens input surface is curved. Our study suggests a feasible route toward constructing the flexible channel-structured acoustic metamaterials with the shape-adaption capability, opening then an additional degree of freedom for full control of sound. Genetic analysis is of great importance for the detection of pathogenic bacteria. Bacterial identification must become simpler, less expensive, and more rapid than the traditional methods. In this study, a lowcost, label-free, and wireless paper-based bipolar electrode electrochemiluminescence (pBPE-ECL) analysis system was constructed for the rapid and sensitive genetic detection of pathogenic bacteria. Wax-screen printing was used to form hydrophilic channels on filter paper, and a carbon ink-based bipolar electrode and driving electrodes were screen-printed into the channels. The "light-switch" molecule [Ru(phen)2dppz]2+ (phen = 1,10- phenanthroline; dppz = dipyridophenazine) was used to intercalate into the base pairs of the double-stranded DNA PCR amplification products, and the complexs were then applied to the paper-based bipolar electrode to perform the ECL assays; the ECL of [Ru(phen)2dppz]2+ is quenched in aqueous solution, but this molecule displays intense ECL when intercalated into double-stranded DNA Under optimized experimental conditions, as little as 10 copies/μL of the genomic DNA of Listeria monocytogenes could be detected. Additionally, the system could also specifically distinguish Listeria monocytogenes from Salmonella, Escherichia coli 0157:H7, and Staphylococcus aureus. Seleniumis applied as the cathode material of lithium/selenium(Li–Se) battery, which has high theoretical volumetric capacitydensity (3253 mA h·cm–3) and high output voltages.However, it has a low melting point (494 K) and a large sensitivityto thermal treatment, which often result in the phase transition betweencrystalline Se (trigonal phase) and amorphous Se during the charge/dischargecycles of Li–Se battery as reported in literatures. In orderto clarify the different chemical (de)lithiation mechanisms betweenthem in Li–Se battery, in this work large-area amorphous selenium(a-Se) nanowires (NWs) have been successfully prepared first througha facile high-energy ball-milling method. Subsequently the crystalline(c) and crystalline/amorphous (c/a) selenium NWs have also been preparedthrough annealing the above as-obtained amorphous products, respectively.The affirmative composition and morphology of the as-obtained Se nanostructureshave been demonstrated by the XRD, SEM, TEM, HRTEM and Raman spectrameasurements. And their specific surface area and pore size distributionhave also been analyzed by BET measurements. Finally, it is provedthat the as-obtained NWs used as the cathode material of Li–Sebattery displayed different chemical reaction processes with Li+and the related various storage capacities (a-Se: 755 mAh·g–1; c/a-Se: 705 mAh·g–1; c-Se:250 mAh·g–1). Thermosolutocapillary convection within a rectangular cavity with dynamic free surface is numerically investigated in the absence of gravitational effects. Both the temperature and solute concentration gradients are applied horizontally. The free surface deformation is captured by the level set method. Two cases of the ratio of thermal to solutal Marangoni number Rσ < -1 and Rσ = -1 are considered. For Rσ < -1, the free surface bulges out near the left wall and bulges in near the right wall; with the increase of Marangoni number, the free surface deformation decreases and with the increase of capillary number and aspect ratio, it increases. For Rσ = -1, the free surface bulges out near the left and right walls and bulges in at the central zone; with the increase of Marangoni number, the free surface deformation mode is changed and with the increase of capillary number and aspect ratio, the free surface deformation increases. We propose a solid phononic crystal lens capable of acoustic superfocusing beyond the diffraction limit. The unit cell of the crystal is formed by four rigid cylinders in a hosting material with a cavity arranged in the center. Theoretical studies reveal that the solid lens produces both negative refraction to focus propagating waves and surface states to amplify evanescent waves. Numerical analyses of the superfocusing effect of the considered solid phononic lens are presented with a separated source excitation to the lens. In this case, acoustic superfocusing beyond the diffraction limit is evidenced. Compared to the fluid phononic lenses, the solid lens is more suitable for ultrasonic imaging applications. Visceral leishmaniasis patients have been reported to have a urine concentration defect. Concentration of urine by the renal inner medulla is essentially dependent on a transcription factor, NFAT5/TonEBP, because it activates expression of osmoprotective genes betaine/glycine transporter 1 (BGT1) and sodium/myo-inositol transporter (SMIT), and water channel aquaporin- 2, all of which are imperative for concentrating urine. Leishmania parasites evade macrophage immune defenses by activating protein tyrosine phosphatases, among which SHP-1 is critical. We previously demonstrated that SHP-1 inhibits tonicity-dependent activation of NFAT5/TonEBP in HEK293 cells through screening a genome-wide small interfering (si) RNA library against phosphatases (Zhou X, Gallazzini M, Burg MB, Ferraris JD. We sought to examine whether Leishmania can activate SHP-1 and inhibit NFAT5/TonEBP activity in the renal inner medulla in a murine model of visceral leishmaniasis by injection of female BALB/c mice with a single intravenous dose of 5 X 105 L. chagasi metacyclic promastigotes. We found that SHP-1 is expressed in the kidney inner medulla. L. chagasi activates SHP-1 with an increase in stimulatory phosphorylation of SHP-1-Y536 in the region. L. chagasi reduces expression of NFAT5/TonEBP mRNA and protein as well as expression of its targeted genes: BGT1, SMIT, and aquaporin-2. The culture supernatant from L. chagasi metacyclic promastigotes increases SHP-1 protein abundance and potently inhibits NFAT5 transcriptional activity in mIMCD3 cells. However, L. chagasi in our animal model has no significant effect on urinary concentration. We conclude that L. chagasi, most likely through its secreted virulence factors, activates SHP-1 and reduces NFAT5/ TonEBP gene expression, which leads to reduced NFAT5/TonEBP transcriptional activity in the kidney inner medulla. A holey-structured metamaterial is proposed for near-field acoustic imaging beyond the diffraction limit. The structured lens consists of a rigid slab perforated with an array of cylindrical holes with periodically modulated diameters. Based on the effective medium approach, the structured lens is characterized by multilayered metamaterials with anisotropic dynamic mass, and an analytic model is proposed to evaluate the transmission properties of incident evanescent waves. The condition is derived for the resonant tunneling, by which evanescent waves can completely transmit through the structured lens without decaying. As an advantage of the proposed lens, the imaging frequency can be modi?ed by the diameter modulation of internal holes without the change of the lens thickness in contrast to the lens due to the Fabry-Pérot resonant mechanism. In this experiment, the lens is assembled by aluminum plates drilled with cylindrical holes. The imaging experiment demonstrates that the designed lens can clearly distinguish two sources separated in the distance below the diffraction limit at the tunneling frequency. Background: Drs2p was known to be stimulated by phosphatidylinositol 4-phosphate (PI(4)P), but the mechanism wasunclear. Results: Loss of the C-terminal tail, or presence of PI(4)P, stimulates the activity of Drs2p. Conclusion: Inhibition of Drs2p activity by its C-terminal tail is relieved by PI(4)P. Activation of the transcription factor NFAT5 by high NaCl involves changes in phosphorylation. By siRNA screening, we previously found that protein targeting to glycogen (PTG), a regulatory subunit of protein phosphatase (PP1), contributes to regulation of high NaClinduced NFAT5 transcriptional activity. The present study addresses the mechanism involved. We find that high NaCl-induced inhibition of PTG elevates NFAT5 activity by increasing NFAT5 transactivating activity, protein abundance, and nuclear localization. PTG acts via a catalytic subunit PP1γ. PTG associates physically with PP1γ, and NaCl reduces both this association and remaining PTG-associated PP1γ activity. High NaCl-induced phosphorylation of p38, ERK, and SHP-1 contributes to activation of NFAT5. Knockdown of PTG does not affect phosphorylation of p38 or ERK. However, PTG and PP1γ bind to SHP-1, and knockdown of either PTG or PP1γ increases high NaCl-induced phosphorylation of SHP-1-S591, which inhibits SHP-1. Mutation of SHP-1-S591 to alanine, which cannot be phosphorylated, increases inhibition of NFAT5 by SHP-1. Thus high NaCl reduces the stimulatory effect of PTG and PP1γ on SHP-1, which in turn reduces the inhibitory effect of SHP-1 on NFAT5. Oxidative stress damages cells. NaCl and urea are high in renal medullary interstitial fluid, which is necessary to concentrate urine, but which causes oxidative stress by elevating reactive oxygen species (ROS). Here, we measured the antioxidant enzyme superoxide dismutases (SODs, MnSOD, and Cu/ ZnSOD) and catalase in mouse kidney that might mitigate the oxidative stress. MnSOD protein increases progressively from the cortex to the inner medulla, following the gradient of increasing NaCl and urea. MnSOD activity increases proportionately, but MnSOD mRNA does not. Water restriction, which elevates renal medullary NaCl and urea, increases MnSOD protein, accompanied by a proportionate increase in MnSOD enzymatic activity in the inner medulla, but not in the cortex or the outer medulla. In contrast, Cu/ZnSOD and TNF-α (an important regulator of MnSOD) do not vary between the regions of the kidney, and expression of catalase protein actually decreases from the cortex to the inner medulla. Water restriction increases activity of mitochondrial enzymes that catalyze production of ROS in the inner medulla, but reduces NADPH oxidase activity there. We also examined the effect of high NaCl and urea on MnSOD in Madin-Darby canine kidney (MDCK) cells. High NaCl and high urea both increase MnSOD in MDCK cells. This increase in MnSOD protein apparently depends on the elevation of ROS since it is eliminated by the antioxidant N-acetylcysteine, and it occurs without raising osmolality when ROS are elevated by antimycin A or xanthine oxidase plus xanthine. Specific and sensitive detection of proteins in biotechnological applications and medical diagnostics is one of the most important goals for the scientific community. In this study, a new protein assay is developed on the basis of hairpin probe and nicking enzyme assisted signal amplification strategy. The metastable state hairpin probe with short loop and long stem is designed to contain a protein aptamer for target recognition. A short Black Hole Quencher (BHQ)-quenching fluorescence DNA probe (BQF probe) carrying the recognition sequence and cleavage site for the nicking enzyme is employed for fluorescence detection. Introduction of target protein into the assay leads to the formation change of hairpin probe from hairpin shape to open form, thus faciliating the hybridization between the hairpin probe and BQF probe. The fluorescence signal is amplified through continuous enzyme cleavage. Thrombin is used as model analyte in the current proof-of-concept experiments. This method can detect thrombin specifically with a detection limit as low as 100 pM. Additionally, the proposed protein detection strategy can achieve separation-free measurement, thus eliminating the washing steps. Mouse embryonic cells isolated from focal adhesion kinase (FAK)--null animals at embryonic day 7.5 display multiple defects in focal adhesion remodeling, microtubule dynamics, mechanotransduction, proliferation, directional motility, and invasion. To date, the ability of FAK to modulate cell function has been ascribed largely to its control of posttranscriptional signaling cascades in this embryonic cell population. In this paper, we demonstrate that FAK unexpectedly exerts control over an epithelial--mesenchymal transition (EMT) program that commits embryonic FAK-null cells to an epithelial status highlighted by the expression of E-cadherin, desmoplakin, and cytokeratins. FAK rescue reestablished the mesenchymal characteristics of FAK-null embryonic cells to generate committed mouse embryonic fibroblasts via an extracellular signal--related kinase--and Akt-dependent signaling cascade that triggered Snail1 gene expression and Snail1 protein stabilization. These findings indentify FAK as a novel regulator of Snail1-dependent EMT in embryonic cells and suggest that multiple defects in FAK-/- cell behavior can be attributed to an inappropriate commitment of these cells to an epithelial, rather than fibroblastic, phenotype. Background: Hepatitis C virus (HCV) 6a accounts for 23.6% of all HCV infections of the general population and 58.5% of intravenous drug users in Hong Kong. However, the geographical origin of this highly predominant HCV subgenotype is largely unknown. This study explores a hypothesis for one possible transmission route of HCV 6a to Hong Kong. Methods: NS5A sequences derived from 26 HCV 6a samples were chosen from a five year period (1999-2004) from epidemiologically unrelated patients from Hong Kong. Partial-NS5A sequences (513-bp from nt 6728 to 7240) were adopted for Bayesian coalescent analysis to reconstruct the evolutionary history of HCV infections in Hong Kong using the BEAST v1.3 program. A rooted phylogenetic tree was drawn for these sequences by alignment with reference Vietnamese sequences. Demographic data were accessed from "The Statistic Yearbooks of Hong Kong". Results: Bayesian coalescent analysis showed that the rapid increase in 6a infections, which had increased more than 90- fold in Hong Kong from 1986 to 1994 correlated to two peaks of Vietnamese immigration to Hong Kong from 1978 to 1997. The second peak, which occurred from 1987 through 1997, overlapped with the rapid increase of HCV 6a occurrence in Hong Kong. Phylogenetic analyses have further revealed that HCV 6a strains from Vietnam may be ancestral to Hong Kong counterparts. Separate reports that hypertonicity activates p38 via a Rac1-OSM-MEKK3-MKK3-p38 pathway and that p38a contributes to activation of TonEBP/OREBP led us to the hypothesis that Rac1 might activate TonEBP/OREBP via p38. The present studies examine that possibility. High NaCl is hypertonic. We find that siRNA knockdown of Rac1 reduces high NaCl-induced increase of TonEBP/OREBP transcriptional activity (by reducing its transactivating activity but not its nuclear localization). Similarly, siRNA knockdown of osmosensing scaffold for MEKK3 (OSM) also reduces high NaCl-dependent TonEBP/OREBP transcriptional and transactivating activities. Simultaneous siRNA knockdown of Rac1 and OSM is not additive in reduction of TonEBP/OREBP transcriptional activity, indicating a common pathway. However, siRNA knockdown of MKK3 does not reduce TonEBP/OREBP transcriptional activity, although siRNA knockdown of MKK6 does. Nevertheless, the effect of Rac1 on TonEBP/OREBP is also independent of MKK6 because it occurs in MKK6-null cells. Furthermore, we find that siRNA knockdown of Rac1 or OSM actually increases activity (phosphorylation) of p38, rather than decreasing it, as previously reported. Thus, the effect of Rac1 on TonEBP/OREBP is independent of p38. We find instead that phospholipase C-γ1 (PLC-γ1) is involved. When transfected into PLC-γ1-null mouse embryonic fibroblast cells, catalytically active Rac1 does not increase TonEBP/OREBP transcriptional activity unless PLC-γ1 is reconstituted. Similarly, dominant-negative Rac1 also does not inhibit TonEBP/OREBP in PLC-γ1-null cells unless PLC-γ1 is reconstituted. A new protein assay based on fluorescence crosscorrelation spectroscopy (FCCS) and aptamer probe is developed. In this assay, two spectrally distinct fluorophores labeled aptamer probes are used to recognize and detect thrombin through a sandwich reaction. The sandwich complexes are diffused through a confocal detection volume. The cross-correlation signals can be observed only at the presence of the aptamer probes-protein sandwich complexes. Thrombin is selected as a target to validate the assay. The whole detection process can be completed within an hour with low-nanomolar sensitivity and high specificity. The recently developed DNA-gold nanoparticle (DNA- GNP) biobarcode assay provides polymerase chain reaction (PCR)-like sensitivity for nucleic acid and protein targets without a need for enzymatic amplification. However, application of the conventional assay is challenged by its complex, expensive, time-consuming, and labor-intense procedure. Herein, we present a new electrochemiluminescence (ECU) biobarcode method based on cysteaxnine-GNP conjugates. In this strategy, an ECU nanoprobe is fabricated that relies on GNP that is modified with tris.(2,2'.bipyridyl) ruthenium (TBR) labeled cysteamine to boost ECL signals and single strand DNA for target recognition. Specifically, a sandwich complex that consists of a biotin labeled capture probe, target DNA, and cysteamine-GNP conjugate is captured by magnetic microparticles (MMPs) and subsequently identified by the ECU signals from loaded TBR. With the use of the developed probe, a limit of detection as low as 100 fM can be achieved and the assay exhibits excellent selectivity for single-mismatched DNA detection even in human serum. Hypertonicity activates the transcription factor TonEBP/OREBP, resulting in increased expression of osmoprotective genes, including those responsible for accumulation of organic osmolytes and heat-shock proteins. Phosphorylation of TonEBP/OREBP contributes to its activation. Several of the kinases that are involved were previously identified, but the phosphatases were not. In the present studies we screened a genomewide human phosphatase siRNA library in human embryonic kidney (HEK)293 cells for effects on TonEBP/OREBP transcriptional activity. We found that siRNAs against 57 phosphatases significantly alter TonEBP/OREBP transcriptional activity during normotonicity (290 mosmol/kg) or hypertonicity (500 mosmol/kg, NaCl added) or both. Most siRNAs increase TonEBP/OREBP activity, implying that the targeted phosphatases normally reduce that activity. We further studied in detail SHP-1, whose knockdown by its specific siRNA increases TonEBP/OREBP transcriptional activity at 500 mosmol/kg. We confirmed that SHP-1 is inhibitory by overexpressing it, which reduces TonEBP/OREBP transcriptional activity at 500 mosmol/kg. SHP-1 dephosphorylates TonEBP/OREBP at a known regulatory site, Y143, both in vivo and in vitro. It inhibits TonEBP/OREBP by both reducing TonEBP/OREBP nuclear localization, which is Y143 dependent, and by lowering high NaCl-induced TonEBP/OREBP transactivating activity. SHP-1 coimmunoprecipitates with TonEBP/OREBP and vice versa, suggesting that they are physically associated in the cell. High NaCl inhibits the effect of SHP-1 on TonEBP/OREBP by increasing phosphorylation of SHP-1 on Ser591, which reduces its phosphatase activity and localization to the nucleus. Thus, TonEBP/OREBP is extensively regulated by phosphatases, including SHP-1, whose inhibition by high NaCl increases phosphorylation of TonEBP/OREBP at Y143, contributing to the nuclear localization and activation of TonEBP/OREBP. The broad-spectrum expression of telomerase in most malignancies makes it a promising target for a cancer diagnostic and prognostic tool. Conventional polymerase chain reaction (PCR)-based telomerase activity assay is highly sensitive but susceptible to amplification-related errors. Here, we present a novel approach to telomerase activity detection. The detection of telomerase activity is accomplished by the hybridization of electrochemiluminescence (ECL) nanoprobes to telomerase reaction products, subsequent capture by magnetic beads, and in situ measurement of the light signal from ECL nanoprobes. The ECL intensity directly reflects the quantity of telomerase reaction products, thus telomerase activity. The high sensitivity afforded by the current magnetic bead and nanoparticle based ECL detection platform allows measuring of telomerase activity from as little as 500 cultured cancer cells in crude cell extracts without the PCR amplification of telomerase reaction products. In addition, a comparative study of the ECL nanoprobe and linear telomere antisense ECL probe was executed. Hollow fiber modules are commonly used to conveniently and efficiently remove cryoprotective agents (CPAs) from cryopreserved cell suspensions. In this paper, a steady-state model coupling mass transfers across cell and hollow fiber membranes is theoretically developed to evaluate the removal of CPAs from cryopreserved blood using hollow fiber modules. This steady-state model complements the unsteady-state model, which was presented in our previous study. The steady-state model, unlike the unsteady-state model, can be used to evaluate the effect of ultrafiltration flow rates on the clearance of CPAs. The steady-state model is validated by experimental results, and then is compared with the unsteady-state model. Using the steady-state model, the effects of ultrafiltration flow rates, NaCI concentrations in dialysate, blood flow rates and dialysate flow rates on CPA concentration variation and cell volume response are investigated in detail. According to the simulative results, the osmotic damage of red blood cells can easily be reduced by increasing ultrafiltration flow rates, increasing NaCl concentrations in dialysate, increasing blood flow rates, or decreasing dialysate flow rates. To understand the effects of magnetic fields on the evolution behaviours of thermocapillary convection in a two-layer fluid system under the absence of gravity, we conducted a series of unsteady three-dimensional numerical simulations of thermocapillary flows in a rectangular cavity. The results show that the convective cell merging process under x direction magnetic field is decelerated in the encapsulant, but that in the melt is accelerated; magnetic field in z direction decelerates the convective cells merging in the melt and encapsulant greatly; the effect of magnetic field in y direction on the convective cell merging is weak. The maximum vorticity magnitude of thermocapillary convection 58.3 s−1 at fully developed without magnetic field is reduced to 53.4, 54.4 and 20.7 s−1 under magnetic fields in x, y and z directions, respectively. Type-IV P-type ATPases (P4-ATPases) are putative phospholipid translocases, or flippases, that translocate specific phospholipid substrates from the exofacial to the cytosolic leaflet of membranes to generate phospholipid asymmetry. In addition, the activity of Drs2p, a P4-ATPase from Saccharomyces cerevisiae, is required for vesicle-mediated protein transport from the Golgi and endosomes, suggesting a role for phospholipid translocation in vesicle budding. Drs2p is necessary for translocation of a fluorescent phosphatidylserine analogue across purified Golgi membranes. However, a flippase activity has not been reconstituted with purified Drs2p or any other P4-ATPase, so whether these ATPases directly pump phospholipid across the membrane bilayer is unknown. Here, we show that Drs2p can catalyze phospholipid translocation directly through purification and reconstitution of this P4-ATPase into proteoliposomes. The noncatalytic subunit, Cdc50p, also was reconstituted in the proteoliposome, although at a substoichiometric concentration relative to Drs2p. In proteoliposomes containing Drs2p, a phosphatidylserine analogue was actively flipped across the liposome bilayer to the outer leaflet in the presence of Mg2+-ATP, whereas no activity toward the phosphatidylcholine or sphingomyelin analogues was observed. This flippase activity was mediated by Drs2p, because protein-free liposomes or proteoliposomes reconstituted with a catalytically inactive form of Drs2p showed no translocation activity. Perinuclear aggresome formation is a key mechanism to dispose of misfolded proteins that exceed the degradative capacity of ubiquitin-proteasome and autophagy-lysosome systems. Functional blockade of either degradative system leads to an enhanced aggresome formation. The tuberous sclerosis complex-Ras homologue enriched in brain-mammalian target of rapamycin (TSC-Rheb-mTOR) pathway is known to play a central role in modulating protein synthesis and autophagy. However, in spite of the constitutive activation of mTOR and the abrogated autophagy activity in TSC1- or TSC2-deficient cells, the TSC mutant cells are defective in aggresome formation and undergo apoptosis upon misfolded protein accumulation both in vitro and in vivo. High Rheb activity in TSC mutant cells inhibits aggresome formation and sensitizes cell death in response to misfolded proteins. Surprisingly, this previously unrecognized function of Rheb is independent of TOR complex 1. Active Rheb disrupts the interaction between dynein and misfolded protein cargos, and therefore blocks aggresome formation by inhibiting dynein-dependent transportation of misfolded proteins. Although packet delay and loss are two important parameters of the Internet performance, to the best of our knowledge, the evolution of large-scale IPv6 delay and loss performance has previously not been studied. In this paper, we analyze more than 600 end-to-end IPv6 paths between about 26 testboxes of RIPE Network Coordination Centre over two years, and compare the delay and loss performance over time with their IPv4 counterparts. We present and discuss the measurement methodologies and show that IPv6 paths have a higher delay and loss than their IPv4 counterparts. High NaCl rapidly activates p38 MAPK by phosphorylating it, the phosphorylation presumably being regulated by a balance of kinases and phosphatases. Kinases are known, but the phosphatases are uncertain. Our initial purpose was to identify the phosphatases. We find that in HEK293 cells transient overexpression of MAPK phosphatase-1 (MKP-1), a dual-specificity phosphatase, inhibits high NaCl-induced phosphorylation of p38, and that over-expression of a dominant negative mutant of MKP-1 does the opposite. High NaCl lowers MKP-1 activity by increasing reactive oxygen species, which directly inhibit MKP-1, and by reducing binding of MKP-1 to p38. Because inhibition of p38 is reported to reduce hypertonicity-induced activation of the osmoprotective transcription factor, TonEBP/OREBP, we anticipated that MKP-1 expression might also. However, overexpression of MKP-1 has no significant effect on Ton EBP/OREBP activity. This paradox is explained by opposing effects of p38α and p38δ, both of which are activated by high NaCl and inhibited by MKP-1. Thus, we find that overexpression of p38α increases high NaCI-induced TonEBP/OREBP activity, but overexpression of p38δ reduces it. Also, siRNA-mediated knockdown of p38δ enhances the activation of TonEBP/OREBP. We conclude that high NaCl inhibits MKP-1, which contributes to the activation of p38. However, opposing actions of p38α and p38δ negate any effect on TonEBP/OREBP activity. Thus, activation of p38 isoforms by hypertonicity does not contribute to activation of TonEBP/OREBP because of opposing effects of p38α and p38δ, and effects of inhibitors of p38 depend on which isoform is affected, which can be misleading. A mass-spring system with negative effective mass is experimentally realized, and its transmission property is examined in the low-frequency range. The local resonance of the basic unit is observed and explained by Newton's theory. The negative effective mass is confirmed by experiments through the transmission properties of a finite periodic system composed of such basic units. In the negative mass range, low transmissions of the system are observed and it is well predicted by the theory. In addition, zero effective mass is discussed and experimentally investigated, which gives rise to no phase shifts in the system. Finally, the anti-vibration effect with a negative mass system is also analyzed. The relevant results are helpful for a better understanding of the resonant nature of metamaterials. Studies have shown that acoustic experiences significantly contribute to the functional shaping of the structural organization and signal processing capacities of the mammalian auditory system during postnatal development. Here, we show how an early epoch of exposure to structured noise influences temporal processing in the rat primary auditory cortex documented immediately after exposure and again in adulthood. Pups were continuously exposed to broadband-pulsed noise across the critical period for auditory system development. Immediately after cessation of exposure at postnatal day ≈35 (P35) or ≈55 days later (i.e., P90) in other rats, the temporal modulation-transfer functions of cortical neurons were documented. We found that pulsed noise exposure at a low modulation rate significantly decreased cortical responses to repetitive stimuli presented across a range of higher modulation rates. The highest temporal rate at which temporal modulation-transfer function was at half of its maximum was reduced when compared with naïve rats. Low-rate pulsed noise exposure also decreased cortical response synchronization at higher stimulus rates, as shown by vector strength and Rayleigh statistic measures. These postexposure changes endured into adulthood. These findings bear significant implications for the role of early sound experiences as contributors to the ontogeny of human auditory and language-related abilities and impairments. Corticofugal modulation of sub-cortical auditory selectivity has been shown previously in mammals for frequency, amplitude, time, and direction domains in separate studies. As such, these studies do not show if multi-parametric corticofugal modulation can be mediated through the same sub-cortical neuron. Here we specifically studied corticofugal modulation of best frequency (BF), best amplitude (BA), and best azimuth (BAZ) at the same neuron in the inferior colliculus of the big brown bat, Eptesicus fuscus, using focal electrical stimulation in the auditory cortex. Among 53 corticofugally inhibited collicular neurons examined, cortical electrical stimulation produced a shift of all three measurements (i.e., BF, BA, and BAZ) toward the value of stimulated cortical neuron in 13 (24.5%) neurons, two measurements (i.e., BF and BAZ or BA and BAZ) in 19 (36%) neurons, and one measurement in 16 (30%) neurons. Cortical electrical stimulation did not shift any of these measurements in the remaining five (9.5%) neurons. Corticofugally induced collicular BF shift was symmetrical, whereas the shift in collicular BA or BAZ was asymmetrical. The amount of shift in each measurement was significantly correlated with each measurement difference between recorded collicular and stimulated cortical neurons. However, shifts of three measurements were not correlated with each other. Furthermore, average measurement difference between collicular and cortical neurons was larger for collicular neurons with measurement shifts than for those without shifts. These data indicate that multi-parametric corticofugal modulation can be mediated through the same subcortical neuron based on the difference in auditory selectivity between subcortical and cortical neurons. The spectral, temporal, and intensive selectivity of neurons in the adult primary auditory cortex (A1) is easily degraded in early postnatal life by raising rat pups in the presence of pulsed noise. The nonselective frequency tuning recorded in these rats substantially endures into adulthood. Here we demonstrate that perceptual training applied in these developmentally degraded postcritical-period rats results in the recovery of normal representational fidelity. By using a modified go/no-go training strategy, structured noise-reared rats were trained to identify target auditory stimuli of specific frequency from a set of distractors varying in frequency. Target stimuli changed daily on a random schedule. Consistent with earlier findings, structured noise exposure within the critical period resulted in disrupted tonotopicity within A1 and in degraded frequency-response selectivity for A1 neurons. Tonotopicity and frequency-response selectivity were normalized by perceptual training. Changes induced by training endured without loss for at least 2 months after training cessation. The results further demonstrate the potential utility of perceptual learning as a strategy for normalizing deteriorated auditory representations in older (postcritical-period) children and adults. Hypertonicity activates the transcription factor tonicity-responsive enhancer/ osmotic response element binding protein (TonEBP/OREBP), resulting in increased expression of genes involved in osmoprotective accumulation of organic osmolytes, including glycine betaine, and in increased expression of osmoprotective heat shock proteins. Our previous studies showed that high NaCl increases reactive oxygen species (ROS), which contribute to activation of TonEBP/OREBP. Mitochondria are a major source of ROS. The purpose of the present study was to examine whether mitochondria produce the ROS that contribute to activation of TonEBP/OREBP. We inhibited mitochondrial ROS production in HEK293 cells with rotenone and myxothiazol, which inhibit mitochondrial complexes I and III, respectively. Rotenone (250 nM) and myxothiazol (12 nM) reduce high NaCl-induced ROS over 40%, whereas apocynin (100 μM), an inhibitor of NADPH oxidase, and allopurinol (100 μM), an inhibitor of xanthine oxidase, have no significant effect. Rotenone and myxothiazol reduce high NaCl-induced increases in TonEBP/OREBP transcriptional activity (ORE/TonE reporter assay) and BGT1 (betaine transporter) mRNA abundance ranging from 53 to 69%. They inhibit high NaCl-induced TonEBP/OREBP transactivating activity, but not its nuclear translocation. Release of ATP into the medium on hypertonic stress has been proposed to be a signal that triggers cellular osmotic responses. However, we do not detect release of ATP into the medium or inhibition of high NaCl-induced ORE/TonE reporter activity by an ATPase, apyrase (20 U/ml), indicating that high NaCl-induced activation of TonEBP/OREBP is not mediated by release of ATP. The signaling pathways leading to high NaCl-induced activation of the transcription factor tonicity-responsive enhancer binding protein/osmotic response element binding protein (TonEBP/OREBP) remain incompletely understood. High NaCl has been reported to produce oxidative stress. Reactive oxygen species (ROS), which are a component of oxidative stress, contribute to regulation of transcription factors. The present study was undertaken to test whether the high NaCl-induced increase in ROS contributes to tonicity-dependent activation of TonEBP/OREBP. N-acetylcysteine (NAC), an antioxidant, and MnTBAP, an inhibitor of superoxide, reduce high NaCl-induced superoxide activity and suppress both high NaCl-induced increase in TonEBP/OREBP transcriptional activity and high NaCl-induced increase in expression of BGT1mRNA, a transcriptional target of TonEBP/OREBP. Catalase, which decomposes hydrogen peroxide, does not have these effects, whether applied exogenously or overexpressed within the cells. Furthermore, NAC and MnTBAP, but not catalase, blunt high NaCl-induced increase in TonEBP/OREBP transactivation. NG-monomethyl-L-arginine, a general inhibitor of nitric oxide synthase, has no significant effect on either high NaCl-induced increase in superoxide or TonEBP/OREBP transcriptional activity, suggesting that the effects of ROS do not involve nitric oxide. Ouabain, an inhibitor of Na-K-ATPase, attenuates high NaCl-induced superoxide activity and inhibits TonEBP/OREBP transcriptional activity. We conclude that the high NaCl-induced increase in ROS, including superoxide, contributes to activation of TonEBP/OREBP by increasing its transactivation. [ABSTRACT FROM AUTHOR]

Hydrogen peroxide mediates FK506-induced cytotoxicity in renal cells. Background. The nephrotoxicity induced by immunosuppressant FK506 remains a serious clinical problem, and the underlying mechanism has not been completely understood. The present study was undertaken to determine the role of hydrogen peroxide in FK506-mediated cytotoxicity in a porcine renal proximal tubular cell line, LLC-PK1 cells, and human embryonic kidney (HEK293) cells. Methods. Cytotoxicity was estimated by crystal violet and lactate dehydrogenase release assays. The activity of reactive oxygen species (ROS) was detected by flow cytometry. FK506-induced cell death was examined in the presence of the hydrogen peroxide scavenger, catalase, or a scavenger of hydroxyl radicals, sodium benzoate. As a control, FK506-induced cell death was also measured in the presence of superoxide anion inhibitor, 4,5-dihydroxy-1,2-benzene disulfonic acid (Tiron), TEMPO, or overexpressed human manganese superoxide dismutase (MnSOD). Catalase was also used in tumor necrosis factor-α (TNF-α)-induced cell injury to determine whether the enzyme specifically protected cells against FK506-mediated cytotoxicity. Results. FK506 induced cell death in a dose-dependent manner and coincided with a dose-dependent increase in ROS activity. Abrogation of FK506-mediated ROS by catalase and N-acetylcysteine blunted FK506-induced cell death. Furthermore, overexpression of catalase, sodium benzoate, and deferoxamine inhibited the cytotoxic effect of FK506. In contrast, Tiron, TEMPO, or overexpression of human MnSOD failed to show cytoprotection. In fact, TEMPO or expression of MnSOD enhanced the effect of FK506. Catalase did not significantly affect TNF-α–induced cell injury. Conclusion. The signaling pathway that transduces the stimulatory effect of low K[sup +] on the biosynthesis of Na,K-ATPase remains largely unknown. The present study was undertaken to examine whether reactive oxygen species (ROS) mediated the effect of low K[sup +] in Madin-Darby canine kidney (MDCK) cells. Low K[sup +] increased ROS activity in a time- and dose-dependent manner, and this effect was abrogated by catalase and N-acetylcysteine (NAC). To determine the role of ROS in low-K[sup +]-induced gene expression, the cells were first stably transfected with expression constructs in which the reporter gene chloramphenicol acetyl transferase (CAT) was under the control of the avian Na,K-ATPase α-subunit 1.9 kb and 900-bp 5'-flanking regions that have a negative regulatory element. Low K[sup +] increased the CAT expression in both constructs. Catalase or NAC inhibited the effect of low K[sup +]. To determine whether the increased CAT activity was mediated through releasing the repressive effect or a direct stimulation of the promoter, the cells were transfected with a CAT expression construct directed by a 96-bp promoter fragment that has no negative regulatory element. Low K[sup +] also augmented the CAT activity expressed by this construct. More importantly, both catalase and NAC abolished the effect of low K[sup +]. Moreover, catalase and NAC also inhibited low-K[sup +]-induced increases in the Na,K-ATPase α[sub 1]- and β[sub 1]l-subunit protein abundance and ouabain binding sites. The antioxidants had no significant effect on the basal levels of CAT activity, protein abundance, or ouabain binding sites. In conclusion, low K[sup +] enhances the Na,K-ATPase gene expression by a direct stimulation of the promoter activity, and ROS mediate this stimulation and also low-K[sup +]-induced increases in the Na,K-ATPase protein contents and cell surface molecules. A metamaterial slab of anisotropic mass with one diagonal component being infinity and the other being zero is demonstrated to behave as a superlens for acoustic imaging beyond the diffraction limit. The underlying mechanism for extraordinary transmission of evanescent waves is attributed to the zero mass effect. Microstructure design for such anisotropic lens is also presented. In contrast to the anisotropic superlens based on Fabry-Pérot resonant mechanism, the proposed lens operates without the limitation on lens thickness, thus more flexible in practical applications.
