BackgroundCoronavirus disease 2019 (COVID-19), an infectious disease caused by severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2), has caused a global pandemic. Gastric cancer (GC) poses a great threat to people’s health, which is a high-risk factor for COVID-19. Previous studies have found some associations between GC and COVID-19, whereas the underlying molecular mechanisms are not well understood.MethodsWe employed bioinformatics and systems biology to explore these links between GC and COVID-19. Gene expression profiles of COVID-19 (GSE196822) and GC (GSE179252) were obtained from the Gene Expression Omnibus (GEO) database. After identifying the shared differentially expressed genes (DEGs) for GC and COVID-19, functional annotation, protein-protein interaction (PPI) network, hub genes, transcriptional regulatory networks and candidate drugs were analyzed.ResultsWe identified 209 shared DEGs between COVID-19 and GC. Functional analyses highlighted immune-related pathways as key players in both diseases. Ten hub genes (CDK1, KIF20A, TPX2, UBE2C, HJURP, CENPA, PLK1, MKI67, IFI6, IFIT2) were identified. The transcription factor/gene and miRNA/gene interaction networks identified 38 transcription factors (TFs) and 234 miRNAs. More importantly, we identified ten potential therapeutic agents, including ciclopirox, resveratrol, etoposide, methotrexate, trifluridine, enterolactone, troglitazone, calcitriol, dasatinib and deferoxamine, some of which have been reported to improve and treat GC and COVID-19.ConclusionThis research offer valuable insights into the molecular interplay between COVID-19 and GC, potentially guiding future therapeutic strategies.

IntroductionThe coronavirus disease 2019 (COVID-19) pandemic, stemming from the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), has persistently threatened the global health system. Meanwhile, tuberculosis (TB) caused by Mycobacterium tuberculosis (M. tuberculosis) still continues to be endemic in various regions of the world. There is a certain degree of similarity between the clinical features of COVID-19 and TB, but the underlying common pathogenetic processes between COVID-19 and TB are not well understood.MethodsTo elucidate the common pathogenetic processes between COVID-19 and TB, we implemented bioinformatics and systematic research to obtain shared pathways and molecular biomarkers. Here, the RNA-seq datasets (GSE196822 and GSE126614) are used to extract shared differentially expressed genes (DEGs) of COVID-19 and TB. The common DEGs were used to identify common pathways, hub genes, transcriptional regulatory networks, and potential drugs.ResultsA total of 96 common DEGs were selected for subsequent analyses. Functional enrichment analyses showed that viral genome replication and immune-related pathways collectively contributed to the development and progression of TB and COVID-19. Based on the protein-protein interaction (PPI) network analysis, we identified 10 hub genes, including IFI44L, ISG15, MX1, IFI44, OASL, RSAD2, GBP1, OAS1, IFI6, and HERC5. Subsequently, the transcription factor (TF)–gene interaction and microRNA (miRNA)–gene coregulatory network identified 61 TFs and 29 miRNAs. Notably, we identified 10 potential drugs to treat TB and COVID-19, namely suloctidil, prenylamine, acetohexamide, terfenadine, prochlorperazine, 3′-azido-3′-deoxythymidine, chlorophyllin, etoposide, clioquinol, and propofol.ConclusionThis research provides novel strategies and valuable references for the treatment of tuberculosis and COVID-19.

Introduction: The severe acute respiratory syndrome coronavirus 2 (SARS-COV-2) has posed a significant challenge to individuals’ health. Increasing evidence shows that patients with metabolic unhealthy obesity (MUO) and COVID-19 have severer complications and higher mortality rate. However, the molecular mechanisms underlying the association between MUO and COVID-19 are poorly understood.Methods: We sought to reveal the relationship between MUO and COVID-19 using bioinformatics and systems biology analysis approaches. Here, two datasets (GSE196822 and GSE152991) were employed to extract differentially expressed genes (DEGs) to identify common hub genes, shared pathways, transcriptional regulatory networks, gene-disease relationship and candidate drugs.Results: Based on the identified 65 common DEGs, the complement-related pathways and neutrophil degranulation-related functions are found to be mainly affected. The hub genes, which included SPI1, CD163, C1QB, SIGLEC1, C1QA, ITGAM, CD14, FCGR1A, VSIG4 and C1QC, were identified. From the interaction network analysis, 65 transcription factors (TFs) were found to be the regulatory signals. Some infections, inflammation and liver diseases were found to be most coordinated with the hub genes. Importantly, Paricalcitol, 3,3′,4,4′,5-Pentachlorobiphenyl, PD 98059, Medroxyprogesterone acetate, Dexamethasone and Tretinoin HL60 UP have shown possibility as therapeutic agents against COVID-19 and MUO.Conclusion: This study provides new clues and references to treat both COVID-19 and MUO.

Abstract Severe acute respiratory syndrome coronavirus 2 (SARS‐CoV‐2) has been suggested to purpose threats to health of mankind. Alcoholic hepatitis (AH) is a life‐threatening acute and chronic liver failure that takes place in sufferers who drink excessively. During the epidemic, AH has an increasing incidence of severe illness and mortality. The intrinsic relationship of molecular pathogenesis, as well as common therapeutic strategies for two diseases are still poorly understood. The transcriptome of the COVID‐19 and AH has been compared to obtain the altered genes and hub genes were screened out through protein–protein interaction (PPI) network analysis. Via gene ontology (GO), pathway enrichment, and transcription regulator analysis, a deeper appreciation of the interplay mechanism between hub genes were established. Finally, gene‐disease and gene–drug analysis were displayed to instruct the clinical treatments. With 181 common differentially expressed genes (DEGs) of AH and COVID‐19 were obtained, 10 hub genes were captured. Follow‐up studies located that these 10 genes typically mediated the diseases occurrence by regulating the activities of the immune system. Other results suggest that the common pathways of the two ailments are enriched in regulating the function of immune cells and release of immune molecules. The top 10 drug candidates have been chosen primarily, some of which have been proved effective in treating AH sufferers infected with COVID‐19. This study reveals the common pathogenesis of COVID‐19 and AH and assist to discover necessary therapeutic targets to combat the ongoing pandemic induced via SARS‐CoV‐2 infection and acquire promising remedy strategies for the two diseases.

Extended-spectrum beta-lactamase-producing Gram-negative bacteria are common in the community and hospitals. To monitor ESBLs mediated by the CTX-M genotype, we collected clinical ESBL pathogenic strains from a hospital in central China and observed a strain of Escherichia coli, namely Ec15103 carrying blaCTX–M–14, blaCTX–M–64 and blaTEM–1, isolated from the blood of a 7-day-old infant in 2015. Strain Ec15103 contains two drug resistance plasmids: pEc15103A, an IncFI-type plasmid that cannot be conjugatively transferred and carries the drug resistance genes blaTEM–1, aacC2, aadA5, sul1, mph(A), sul2, strAB, and tetA(A); and pEc15103B, an IncK2/Z-type plasmid that carries the conjugation transfer gene and blaCTX–M–14. In addition, blaCTX–M–64 is located on the chromosome of Ec15103, and it is the first report of pathogen with blaCTX–M–64 located on its chromosome (the search terms used “blaCTX-M-64” and “chromosome”). blaCTX–M–14 and blaCTX–M–64 are carried by ISEcp1-mediated transposon Tn6503a and Tn6502, respectively. The conjugation transfer ability of pEc15103B was significantly inhibited by zidovudine (AZT) and linoleic acid (LA) and that expression of blaCTX–M–14, blaCTX–M–64 and blaTEM–1 at the mRNA level did not change based on the concentration of cefotaxime or ampicillin. Co-occurrence of blaCTX–M–14 and blaCTX–M–64 in a single isolate will enhance the drug resistance of bacteria, and the presence of blaCTX–M–64 in the chromosome may make the resistance more maintain. This fact will facilitate its dissemination and persistence under different antimicrobial selection pressures. It is essential to prevent these strains from further spreading in a hospital environment.

This research aims to explore the multi-focus group method as an effective tool for systematically eliciting business requirements for business information system (BIS) projects. During the COVID-19 crisis, many businesses plan to transform their businesses into digital businesses. Business managers face a critical challenge: they do not know much about detailed system requirements and what they want for digital transformation requirements. Among many approaches used for understanding business requirements, the focus group method has been used to help elicit BIS needs over the past 30 years. However, most focus group studies about research practices mainly focus on a particular disciplinary field, such as social, biomedical, and health research. Limited research reported using the multi-focus group method to elicit business system requirements. There is a need to fill this research gap. A case study is conducted to verify that the multi-focus group method might effectively explore detailed system requirements to cover the Case Study business's needs from transforming the existing systems into a visual warning system. The research outcomes verify that the multi-focus group method might effectively explore the detailed system requirements to cover the business's needs. This research identifies that the multi-focus group method is especially suitable for investigating less well-studied, no previous evidence, or unstudied research topics. As a result, an innovative visual warning system was successfully deployed based on the multi-focus studies for user acceptance testing in the Case Study mine in Feb 2022. The main contribution is that this research verifies the multi-focus group method might be an effective tool for systematically eliciting business requirements. Another contribution is to develop a flowchart for adding to Systems Analysis & Design course in information system education, which may guide BIS students step by step on using the multi-focus group method to explore business system requirements in practice.

The SARS-CoV-2 coronavirus has caused worldwide disruption through the COVID-19 pandemic, providing a sobering reminder of the profound impact viruses can have on human well-being. Understanding virus life cycles and interactions with host cells lays the groundwork for exploring therapeutic strategies against virus-related diseases. Fluorescence microscopy plays a vital role in virus imaging, offering high spatiotemporal resolution, sensitivity, and spectroscopic versatility. In this opinion piece, we first highlight two recent techniques, SunTag and StayGold, for the in situ imaging of viral RNA translation and viral assembly. Next, we discuss a new class of genetically encoded fluorogenic protease reporters, such as FlipGFP, which can be customized to monitor SARS-CoV-2’s main (Mpro) or papain-like (PLpro) protease activity. These assays have proven effective in identifying potential antivirals through high-throughput screening, making fluorogenic viral protease reporters a promising platform for viral disease diagnostics and therapeutics.

The quick access recorder (QAR), as an airborne device used to monitor and record flight parameters, has been widely installed on various types of aircraft. Based on QAR data, research on runway overrun, a typical flight safety incident, has attracted widespread attention in recent years. However, existing runway overrun risk models generally suffer from oversimplified risk metrics or insufficient consideration of risk dynamics. In this paper, we propose a new dynamic runway overrun risk assessment model based on QAR data. We first consider the noise of aircraft trajectory data in the QAR parameters and present a landing trajectory correction method combining ground speed and runway position information. Second, to improve the accuracy of the risk assessment model, we design an algorithm to automatically recognize the aircraft autobrake level during the landing phase, based on which a new dynamic risk assessment model is developed. Finally, feature engineering is performed to extract the relevant contributing factors of runway overrun risk, based on which classification and regression models are applied to identify risky flights and predict the risk values. The proposed risk assessment model was evaluated using QAR data from an airline in China. The results show that the automatic deceleration rate, the way that the aircraft approaches the runway, the touchdown distance, and the kinetic energy at 50 ft are key factors in the risk of runway overrun during the landing phase.

Abstract To study the influence of diesel fuel blended with polyoxymethylene dimethyl ethers (PODEn), a new alternative fuel with a high oxygen content and large cetane number, on the combustion characteristics, fuel economies, and emission characteristics of heavy-duty diesel engines that meet China VI emission standards, relevant tests were conducted on a supercharged intercooled high-pressure common-rail diesel engine. The PODEn were blended with diesel fuel at three different ratios (volume fractions of 10%, 20%, and 30%). The test results showed that the PODEn could optimize the combustion process of diesel engines that met the China VI emission standards, effectively improve the thermal efficiencies of diesel engines, and reduce the emissions of hydrocarbon (HC), carbon monoxide (CO), and soot. With an increase in the PODEn blending ratio, the peak values of the in-cylinder pressure, average in-cylinder temperature, and instantaneous heat release rate gradually decreased, and each peak progressively moved forward. As the start of combustion gradually moved forward, the combustion duration was shortened by 0.7–2.8°CA, the heat release process became more concentrated, and the effective thermal efficiency was increased by up to 2.57%. The effective fuel consumption gradually increased, yet the equivalent effective fuel consumption gradually decreased, with the largest drop being as high as 4.55%. The nitrogen oxides (NO x ) emission increased slightly, and the emissions of HC, CO, and soot gradually decreased. The emissions of CO and soot declined significantly under high-speed and high-load conditions, with the highest reductions reaching 66.2% and 76.3%, respectively.

Abstract Shielded metal arc butt welded joints for 9%Ni steel using nickel-based filler metal were analyzed by optical microscopy, scanning electron microscope, as well as transmission electron microscopy. The nanoindentation method was used to study the correlation between the structure and micromechanical properties of the fusion welded joint. The results show that there is a microstructural evolution from melted deposit to 9%Ni base steel. A significant coarse lath martensite-bainite mixture has been obtained in 9%Ni steel close to the fusion boundary while the retained austenite distributing in grain boundary of lath martensite has reduced to an undetectable level. In fusion boundary, an intermetallic layer has been observed which corresponds to the epitaxial growth of weld metal. The analysis of stress-strain behavior using nanoindentation shows that the heat-affected zone of coarse grains exhibits ductility loss and quantitative plastic deformation failure. The fusion boundary has the lowest value of yield stress while the coarse grained heat affected zone has the maximum value of yield stress.

The respiratory disease COVID-19 is caused by the coronavirus SARS-CoV-2. Here we report the discovery of ethacridine as a potent drug against SARS-CoV-2 (EC50 ~ 0.08 μM). Ethacridine was identified via high-throughput screening of an FDA-approved drug library in living cells using a fluorescence assay. Plaque assays, RT-PCR and immunofluorescence imaging at various stages of viral infection demonstrate that the main mode of action of ethacridine is through inactivation of viral particles, preventing their binding to the host cells. Consistently, ethacridine is effective in various cell types, including primary human nasal epithelial cells that are cultured in an air-liquid interface. Taken together, our work identifies a promising, potent, and new use of the old drug via a distinct mode of action for inhibiting SARS-CoV-2.

As a major branch of Non-Photorealistic Rendering (NPR), image stylization mainly uses computer algorithms to render a photo into an artistic painting. Recent work has shown that the ex-traction of style information such as stroke texture and color of the target style image is the key to image stylization. Given its stroke texture and color characteristics, a new stroke rendering method is proposed. By fully considering the tonal characteristics and the representative color of the original oil painting, it can fit the tone of the original oil painting image into a stylized image whilst keeping the artist’s creative effect. The experiments have validated the efficacy of the proposed model in comparison to three state-of-the-arts. This method would be more suitable for the works of pointillism painters with a relatively uniform style, especially for natural scenes, otherwise, the results can be less satisfactory.

Abstract The mechanical properties and corrosion performance of the N10276 alloy welded joint were investigated with nano-hardness testing and electrochemical measurement. The results showed that solidification mode changes from equiaxed crystal to columnar crystal from the center of the weldment to the fusion zone. With the increase of immersion times, the diameter of capacitance arc and the impedance modulus increase, and the corrosion resistance of HAZ decreases. The strengthening effect of welded metal was more obvious than that of heat affected zone for the nickel-based welded joint and especially in coarse grained heat affected zone, the hardening resulted from overheating was not apparent. Nickel-based weld metal with high content of alloying elements were often segregated at interdendritic regions or precipitated in grain interior under nonequilibrium solidification, which differ from conventional low alloy steel welded joint.

Abstract Micro-arc oxidation (MAO) treatment with various voltages was carried out on Ti alloys in 10% H2SO4 aqueous solution to enhance surface wettability for biological application. The effects of applied voltage on the surface morphology, phase constituents, functional group, roughness and wettability of the MAO coatings were investigated comprehensively. Increasing applied voltages significantly change phase constituents, surface morphology and wettability of the MAO coatings. Higher applied voltages are favorable to form higher fraction of rutile phase, larger micro-pores and higher surface roughness. The apparent contact angle decreases slightly after treated at 80 V but it decreases sharply after treated at 160 V or 200 V. The significant change in wettability of MAO coatings is attributed to hydroxyl group content and morphology of the MAO coatings. Differences in hydroxyl group content and surface morphology result in different contact interface types between water and MAO coatings, which is attributed to air captured or no air captured interface models.

Background: Anti-N-methyl-D-aspartate receptor (NMDAR) encephalitis is a cause of autoimmune encephalitis and is characterized by epileptic seizures, psychosis, and consciousness impairments. It mostly affects young adults with ovarian cancers. We herein reported a case of anti-NMDAR encephalitis associated with clear cell renal carcinoma.Case Presentation: A 54-year-old male with headache for 1 week and mood and behavioral changes for 3 days was presented, but his clinical presentation and poor response to antiviral treatment did not support a diagnosis of viral encephalitis. Positive anti-NMDAR antibodies in serum and cerebrospinal fluid confirmed autoimmune encephalitis. A subsequent evaluation revealed a paraneoplastic etiology of a renal mass, and this was then resected and pathologically confirmed as clear cell renal carcinoma. The patient's symptoms showed improvement after resection of the mass. The patient relapsed 6 months after discharge, and the symptoms completely disappeared after treatment with corticosteroids and intravenous immunoglobulin.Conclusion: Our findings suggested that NMDAR encephalitis might be associated with clear cell renal carcinoma. When patients present with unexplained seizures, neuropsychiatric disorder, or other brain symptoms, clinicians should be careful with paraneoplastic neurological disorders. Early diagnosis and treatment of primary tumors might show improvement.

To investigate the parameters of sucrose dust explosion, the minimum ignition energy (MIE) and minimum ignition temperature (MIT) were evaluated. The experiments tested the MIE of sucrose dust under different conditions of dust quantity, ignition delay time (IDT), and powder injection pressure (PIP). The experiments tested the MIT of different particle sizes. The results demonstrate that the MIE of sucrose powder under three conditions was an open-up quadratic polynomial. When the dust quantity, the IDT, and PIP were 0.5 g (417 g/m3), 90 ms, and 150 kPa, respectively, the MIE was 58.9 mJ, 62.6 mJ, and 52.4 mJ. The MIT was positively correlated with the particle size of sucrose dust, and the MIT was 340 °C. At the molecular level, the “O–H” bonds of the sucrose molecule hydroxyl groups were broken by the discharge of electrodes or high temperature to generate H2. The combustion of H2 caused the explosion to spread to the surrounding sucrose dust and made the deposited dust rise, forming an interlocking explosion. The explosion would not stop until the dust concentration dropped below the lowest explosion limit. The results of this study can provide guidance for sucrose enterprises to prevent dust explosion accidents.

ABSTRACT Detection of moving objects is an essential skill for animals to hunt prey, recognize conspecifics and avoid predators. The zebrafish, as a vertebrate model, primarily uses its elaborate visual system to distinguish moving objects against background scenes. The optic tectum (OT) receives and integrates inputs from various types of retinal ganglion cells (RGCs), including direction-selective (DS) RGCs and size-selective RGCs, and is required for both prey capture and predator avoidance. However, it remains largely unknown how motion information is processed within the OT. Here we performed in vivo whole-cell recording and calcium imaging to investigate the role of superficial interneurons (SINs), a specific type of optic tectal neurons, in motion detection of larval zebrafish. SINs mainly receive excitatory synaptic inputs, exhibit transient ON- or OFF-type of responses evoked by light flashes, and possess a large receptive field (RF). One fifth of SINs are DS and classified into two subsets with separate preferred directions. Furthermore, SINs show size-dependent responses to moving dots. They are efficiently activated by moving objects but not static ones, capable of showing sustained responses to moving objects and having less visual adaptation than periventricular neurons (PVNs), the principal tectal cells. Behaviorally, ablation of SINs impairs prey capture, which requires local motion detection, but not global looming-evoked escape. Finally, starvation enhances the gain of SINs’ motion responses while maintaining their size tuning and DS. These results indicate that SINs serve as a motion detector for sensing and localizing sized moving objects in the visual field.

Abstract Background The mechanism of winter survival for perennials involves multiple levels of gene regulation, especially cold resistance. Agropyron mongolicum is one important perennial grass species, but there is little information regarding its overwintering mechanism. We performed a comprehensive transcriptomics study to evaluate global gene expression profiles regarding the winter survival of Agropyron mongolicum. A genome-wide gene expression analysis involving four different periods was identified. Twenty-eight coexpression modules with distinct patterns were performed for transcriptome profiling. Furthermore, differentially expressed genes (DEGs) and their functional characterization were defined using a genome database such as NT, NR, COG, and KEGG. Result A total of 79.6% of the unigenes were characterized to be involved in 136 metabolic pathways. In addition, the expression level of ABA receptor genes, regulation of transcription factors, and a coexpression network analysis were conducted using transcriptome data. We found that ABA receptors regulated downstream gene expression by activating bZIP and NAC transcription factors to improve cold resistance and winter survival. Conclusion This study provides comprehensive transcriptome data for the characterization of overwintering-related gene expression profiles in A. mongolicum. Genomics resources can help provide a better understanding of the overwintering mechanism for perennial gramineae species. By analyzing genome-wide expression patterns for the four key stages of tiller bud development, the functional characteristics of the DEGs were identified that participated in various metabolic pathways and have been shown to be strongly associated with cold tolerance. These results can be further exploited to determine the mechanism of overwintering in perennial gramineae species.

The cell wall plays an important role in responses to various stresses. The cellulose synthase-like gene (Csl) family has been reported to be involved in the biosynthesis of the hemicellulose backbone. However, little information is available on their involvement in plant tolerance to low-temperature (LT) stress. In this study, a total of 42 Csls were identified in Musa acuminata and clustered into six subfamilies (CslA, CslC, CslD, CslE, CslG, and CslH) according to phylogenetic relationships. The genomic features of MaCsl genes were characterized to identify gene structures, conserved motifs and the distribution among chromosomes. A phylogenetic tree was constructed to show the diversity in these genes. Different changes in hemicellulose content between chilling-tolerant and chilling-sensitive banana cultivars under LT were observed, suggesting that certain types of hemicellulose are involved in LT stress tolerance in banana. Thus, the expression patterns of MaCsl genes in both cultivars after LT treatment were investigated by RNA sequencing (RNA-Seq) technique followed by quantitative real-time PCR (qPCR) validation. The results indicated that MaCslA4/12, MaCslD4 and MaCslE2 are promising candidates determining the chilling tolerance of banana. Our results provide the first genome-wide characterization of the MaCsls in banana, and open the door for further functional studies.

We assessed the effect of closing live poultry markets in China on influenza A(H7N9) virus detection and viability. Intensive sampling was carried out before, during, and after a 2-week citywide market closure; the markets were cleaned and disinfected at the beginning of the closure period. Swab samples were collected at different sites within the markets and tested for H7N9 by real-time reverse transcription PCR and culture. During the closure, H7N9 viral RNA detection and isolation rates in retail markets decreased by 79% (95% CI 64%–88%) and 92% (95% CI 58%–98%), respectively. However, viable H7N9 virus could be cultured from wastewater samples collected up to 2 days after the market closure began. Our findings indicates that poultry workers and the general population are constantly exposed to H7N9 virus at these markets and that market closure and disinfection rapidly reduces the amount of viable virus.

Banana (Musa spp.) production is seriously threatened by low temperature (LT) in tropical and subtropical regions. Xyloglucan endotransglycosylase/hydrolases (XTHs) are considered chief enzymes in cell wall remodelling and play a central role in stress responses. However, whether MaXTHs are involved in the low temperature stress tolerance in banana is not clear. Here, the identification and characterization of MaXTHs were carried out, followed by prediction of their cis-acting elements and protein--protein interactions. In addition, candidate MaXTHs involved in banana tolerance to LT were screened through a comparison of their responses to LT between tolerant and sensitive cultivars using RNA-Seq analysis. Moreover, immunofluorescence (IF) labelling was employed to compare changes in the temporal and spatial distribution of different types of xyloglucan components between these two cultivars upon stress. In total, 53 MaXTHs have been identified, and all were predicted to be located in the cell wall, 14 of them also in the cytoplasm. Only 11 MaXTHs have been found to interact with other proteins. Among 16 MaXTHs with LT responsiveness elements, MaXTH26/29/32/35/50 (Group I/II members) and MaXTH7/8 (Group IIIB members) might be involved in banana tolerance to LT stress. IF results suggested that the content of xyloglucan components recognized by CCRC-M87/103/104/106 antibodies might be negatively related to banana chilling tolerance. In conclusion, we have identified the MaXTH gene family and assessed cell wall re-modelling under LT stress. These results will be beneficial for banana breeding against stresses and enrich the cell wall-mediated resistance mechanism in plants to stresses. [ABSTRACT FROM AUTHOR]

Extended-spectrum beta-lactamase-producing Gram-negative bacteria are common in the community and hospitals. To monitor ESBLs mediated by the CTX-M genotype, we collected clinical ESBL pathogenic strains from a hospital in central China and observed a strain of Escherichia coli, namely Ec15103 carrying blaCTX-M-14, blaCTX-M-64 and blaTEM-1, isolated from the blood of a 7-day-old infant in 2015. Strain Ec15103 contains two drug resistance plasmids: pEc15103A, an IncFI-type plasmid that cannot be conjugatively transferred and carries the drug resistance genes blaTEM-1, aacC2, aadA5, sull, mph(A), sul2, strAB, and tetA(A); and pEc15103B, an IncK2/Z-type plasmid that carries the conjugation transfer gene and blaCTX-M-14. In addition, blaCTX-M-64 is located on the chromosome of Ec15103, and it is the first report of pathogen with blaCTX-M-64 located on its chromosome (the search terms used "blaCTX-M-64" and "chromosome"). blaCTX-M-14 and blaCTX-M-64 are carried by ISEcpl-mediated transposon Tn6503a and Tn6502, respectively. The conjugation transfer ability of pEc15103B was significantly inhibited by zidovudine (AZT) and linoleic acid (LA) and that expression of blaCTX-M-14, blaCTX-M-64 and blaTEM-1 at the mRNA level did not change based on the concentration of cefotaxime or ampicillin. Co-occurrence of blaCTX-M-14 and blaCTX-M-64 in a single isolate will enhance the drug resistance of bacteria, and the presence of blaCTX-M-64 in the chromosome may make the resistance more maintain. This fact will facilitate its dissemination and persistence under different antimicrobial selection pressures.
