Cytoarchitecture, the organization of cells within organs and tissues, serves as a crucial anatomical foundation for the delineation of various regions. It enables the segmentation of the cortex into distinct areas with unique structural and functional characteristics. While traditional 2D atlases have focused on cytoarchitectonic mapping of cortical regions through individual sections, the intricate cortical gyri and sulci demands a 3D perspective for unambiguous interpretation. In this study, we employed fluorescent micro-optical sectioning tomography to acquire architectural datasets of the entire macaque brain at a resolution of 0.65 μm × 0.65 μm × 3 μm. With these volumetric data, the cortical laminar textures were remarkably presented in appropriate view planes. Additionally, we established a stereo coordinate system to represent the cytoarchitectonic information as surface-based tomograms. Utilizing these cytoarchitectonic features, we were able to three-dimensionally parcel the macaque cortex into multiple regions exhibiting contrasting architectural patterns. The whole-brain analysis was also conducted on mice that clearly revealed the presence of barrel cortex and reflected biological reasonability of this method. Leveraging these high-resolution continuous datasets, our method offers a robust tool for exploring the organizational logic and pathological mechanisms of the brain’s 3D anatomical structure.

Glycyrrhizic acid (GA) is a natural edible chiral triterpene saponin that can undergo hierarchical self-assembly in water to form supramolecular hydrogels. Customizing the properties of GA supramolecular hydrogels is crucial for their wide food and biomedical applications, and the incorporation of specific metal ions can be an attractive way for this purpose. Herein, this study investigates the effects of metal ions with different valence states and concentrations on the linear and nonlinear rheological properties and network structures of GA supramolecular hydrogels. Monovalent metal ions (Na+ and K+), exhibiting weak binding affinity to GA, rely on the electrostatic screening effects at high concentrations (e.g., 50 mM) to enhance the GA interfibrillar interactions and thus the formation of a more compact and ordered gel network, which displays a pronounced nonlinear rheological behavior with a typical transition from elastic to viscous response. Polyvalent metal ions (Ca2+, Zn2+, and Al3+), owing to their greater charge density and stronger binding with GA, significantly enhance the network strength of the hydrogels at low concentrations (e.g., 5 mM). Nonetheless, at high ion concentrations (e.g., 50 mM), the GA-Mn+ forms discrete aggregated network structures due to excessively strong cation-carboxylate complexation, leading to the irregular nonlinear rheological responses and the lower resistance to large deformations. These findings can deepen our understanding of the highly tunable rheological behaviors and network structures of GA hydrogels, which demonstrates a new possibility for the design and development of responsive natural supramolecular hydrogels with controlled properties through the strategy of metal ion complexation.

Abstract Subsonic jet nozzles, commonly used in passenger aircrafts, generate significant noise that travels both downstream and upstream due to large-scale or fine-scale turbulence in the jet plume. To reduce jet noise, a novel wall treatment method, termed the wavy inner wall (WIW), is proposed. With this method, the smooth inner wall near the exit of the nozzle is replaced by treated walls that carry small wavy patterns. Numerical simulations were conducted to investigate the effects of the WIW treatment. Large eddy simulations (LES) were used to predict the unsteady flow field and the far-field noise, followed by the analogy method proposed by Ffowcs Williams and Hawkings. To better understand the mechanism behind the noise reduction achieved by the WIW treatment, the shear-layer instability, radial and azimuthal auto-correlation functions, turbulent kinetic energy, and acoustic source term from the Tam-Auriault (TA) jet-noise model were analyzed. Results indicated that the WIW treatment advances the onset of jet flow instability in the shear-layer, leading to the early breakdown of jet shear-layer and production of different scales of downstream turbulent structures. As a result, the distribution and production of turbulent kinetic energy are affected, and the generation and emission of jet noise are controlled. The WIW treatment enables the control of fine scale turbulence, resulting in the reduction of mid- to high-frequency noise in the far field, while ensuring a low thrust loss. This feature makes the WIW method a promising approach for jet noise control.

This study aimed to analyze the effect of lipid peroxidation on the allergenicity and functional properties of soybean β-conglycinin (7S) and glycinin (11S). Oxidation complexes were determined using the lipid peroxidation method. Functional properties were analyzed based on emulsifying and foaming properties. The potential allergenicity was evaluated by in vitro and in vivo methods. The results found that oxidation altered structures of the proteins and resulted in the formation of cross-linked protein polymers. The emulsion and foaming properties of the proteins were improved after oxidation. The IgE-binding capacity of 7S and 11S reduced after oxidation. KU812 cell assays showed that both histamine and IL-4 release decreased after oxidation treatment. A mouse model showed that oxidation reduced the IgE, IgG, and IgG1 levels, as well as reduced histamine and mMCP-1 release in serum, which might suppress the allergic reaction. In conclusion, the lipid peroxidation treatment likely causes changes to the functional properties of soybean, decreasing the potential allergenicity of 7S and 11S.

Abstract Lithium‐ion batteries (LIBs) are widely used as energy storage devices. Accurately estimating the state of charge (SOC) is critical for the safe operation of LIBs, but the complex internal characteristics of LIBs make this a difficult task. In this study, an algorithm called BPNN‐UKF, which combines the backpropagation neural network (BPNN) and unscented Kalman filtering (UKF), is proposed. First, a fusion model construction method incorporating AdaBoost and recursive least squares (RLS) is developed to enhance the accuracy and generalization of the battery model. Next, the fusion model is applied to create BPNN‐UKF, which reduces the model dependence of UKF and the training requirements of BPNN. In this method, UKF estimates the SOC, and then BPNN learns the nonlinear relationship between SOC estimation error and process variables in UKF preliminary estimation. The BPNN output is used to update the state vector in real time during the new UKF estimation process. Experimental results reveal that the proposed method can drastically improve estimation accuracy compared with the UKF and the simple combined BPNN‐UKF, and it exhibits considerable generalization to common disturbances in SOC estimation and various battery working conditions.

A new method, named array fluorescent micro-optical sectioning tomography (array-fMOST), is developed for identifying the three-dimensional information at single-cell resolution from multi-samples.

In this study, a functional composite membrane was facilely fabricated by using a dual nanofibril system of bacterial cellulose (BC) and chitin (CH) nanofibrils as bio-based building blocks. The BC-CH membranes with enhanced antibacterial activity were constructed by incorporation of all-natural bioactive nanoparticles (GBTPs), which were formed by spontaneous molecular interactions of three naturally occurring active small molecules, i.e., glycyrrhizic acid (GA), berberine (BR), and tannic acid (TA). The microstructure, physicochemical properties, and antibacterial behaviors of the resulting BC-CH-GBTPs nanocomposites were then characterized. The obtained results showed that the GBTPs with a diameter of around 50–100 nm and membrane matrix were bound by non-covalent interactions, and the addition of GBTPs did not compromise the structural integrity and thermal stability of the composites, which retained good mechanical properties. Furthermore, the addition of GBTPs led to a rougher surface structure and increased the water contact angle of the membrane surfaces from 48.13° to 59.80°. The antimicrobial tests indicate that the BC-CH-GBTPs nanocomposites exhibited significant inhibitory effects against Escherichia coli and Staphylococcus aureus, showing a satisfactory antibacterial ability. These results suggest that the BC-CH-GBTPs nanocomposites based on all-natural, plant-based building blocks, hold promising potentials as active packaging materials for sustainable applications.

Cells are the basic unit of human organs that are not fully understood. The revolutionary advancements of optical imaging allowed us to observe single cells in whole organs, revealing the complicated composition of cells with spatial information. Therefore, in this review, we revisit the principles of optical contrast related to those biomolecules and the optical techniques that transform optical contrast into detectable optical signals. Then, we describe optical imaging to achieve three-dimensional spatial discrimination for biological tissues. Due to the milky appearance of tissues, the spatial information blurred deep in the whole organ. Fortunately, strategies developed in the last decade could circumvent this issue and lead us into a new era of investigation of the cells with their original spatial information.

Compared to young people and adults, there are differences in the ability of elderly people to resist diseases or injuries, with some noticeable features being gender-dependent. However, gender differences in age-related viscera alterations are not clear. To evaluate a potential possibility of gender differences during the natural aging process, we used three age groups to investigate the impact on spleens, kidneys, and adrenal glands. The immunofluorescence results showed that male-specific p21 proteins were concentrated in the renal tubule epithelial cells of the kidney. Histological staining revealed an increase in the frequencies of fat vacuoles located in the renal tubule epithelial cells of the cortex, under the renal capsule in the kidneys of male mice with age. In female mice, we found that the width of the globular zone in the adrenal gland cortex was unchanged with age. On the contrary, the male displayed a reduction in width. Compared to females, the content of epinephrine in adrenal gland tissue according to ELISA analysis was higher in adults, and a greater decline was observed in aged males particularly. These data confirmed the age-dependent differences between female and male mice; therefore, gender should be considered one of the major factors for personalized treatment in clinical diagnosis and treatment.

Abstract Systemic chemotherapy is still the primary treatment for advanced-stage nasopharyngeal carcinoma (NPC), but only limited therapeutic success has been achieved in the past decade because of drug resistance and systemic toxicity. Curcumin (Cur) is an effective alternative to chemotherapeutics because it showed remarkable therapeutic potential in the treatment of NPC. However, lack of tissue specificity and poor penetration in solid tumors are the major obstacles to effective therapy. Therefore, in this work, a self-assembled sub-30 nm therapeutic lipid nanoparticle loaded with Cur, named as Cur@α-NTP-LN, was constructed, specifically targeting scavenger receptor class B member 1 (SR-B1) and enhancing its therapeutic effects on NPC in vivo. Our results showed that Cur@α-NTP-LNs were effective and superior to free Cur on NPC cell-specific targeting, suppressing cell proliferation and inducing cell apoptosis. In vivo and ex vivo optical imaging revealed that Cur@α-NTP-LNs exerted high targeting efficiency, specifically accumulating in NPC xenograft tumors and delivering Cur into the tumor center after systemic administration. Furthermore, Cur@α-NTP-LNs exhibited a remarkable inhibitory effect on the growth of NPC subcutaneous tumors, with over 71 and 47% inhibition compared to Cur- and α-NTP-LNs-treated groups, respectively. In addition, Cur@α-NTP-LNs almost blocked NPC metastasis in a lung metastasis model of NPC and significantly improved the survival rate. Thus, the sub-30 nm Cur@α-NTP-LNs enhanced the solubility of Cur and demonstrated the ability of targeted Cur delivery into the center of the solid NPC tumor, performing synergistic inhibitory effects on the growth of NPC tumor and its metastasis with high efficiency.

Obtaining various structures of the entire mature heart at single-cell resolution is highly desired in cardiac studies; however, effective methodologies are still lacking. Here, we propose a pipeline for labeling and imaging myocardial and vascular structures. In this pipeline, the myocardium is counterstained using fluorescent dyes and the cardiovasculature is labeled using transgenic markers. High-definition dual-color fluorescence micro-optical sectioning tomography is used to perform heart-wide tissue imaging, enabling the acquisition of whole-heart data at a voxel resolution of 0.32 × 0.32 × 1 μm3. Obtained structural data demonstrated the superiority of the pipeline. In particular, the three-dimensional morphology and spatial arrangement of reconstructed cardiomyocytes were revealed, and high-resolution vascular data helped determine differences in the features of endothelial cells and complex coiled capillaries. Our pipeline can be used in cardiac studies for examining the structures of the entire heart at the single-cell level.

The adsorption and foaming properties of an edible colloidal nanoparticle (EYPNs), self-assembled from the food-derived, amphiphilic egg yolk peptides, were investigated, with the aim of evaluating their potential as efficient particulate stabilizers for development of aqueous food foams. The influence of particle aggregation induced by the changes of environmental conditions (mainly the pH) on these properties of EYPN systems was determined. Our results showed that the EYPNs are a highly pH-responsive system, showing the pH-dependent particle aggregation behavior, which is found to strongly affect the interfacial adsorption and macroscopic foaming behaviors of systems. Compared to high pH (6.0–9.0), the EYPNs at low pH (2.0–5.0) showed higher surface activity with a lower equilibrated surface tension as well as a higher packing density of particles and particle aggregates at the interface, probably due to the reduced electrostatic adsorption barrier. Accordingly, the EYPNs at these low pH values exhibited significantly higher foamability and foam stability. The presence of large particle clusters and/or aggregates formed at low pH in the continuous phase may contribute to the foam stability of EYPNs. These results indicate that our edible peptide-based nanoparticle EYPNs can be used as a new class of Pickering-type foam stabilizer for the design of food foams with controlled material properties, which may have sustainable applications in foods, cosmetics, and personal care products.

Responsive dual-structured emulsions and gel emulsions have attracted more and more attention due to their complex microstructures, on-demand responsive properties, and controlled release of active cargoes. In this work, the effect of monoglyceride (MG)-based oil phase structuring on the formation and stability, structural properties, and thermoresponsive and cargo release behavior of gel emulsions stabilized by glycyrrhizic acid (GA) nanofibrils were investigated. Owing to the formation of GA fibrillar networks in the aqueous phase and MG crystalline networks in the oil phase, a stable dual-structured gel emulsion can be successfully developed. The microstructure of the dual-structured gel emulsions largely depended on the concentration of MG in the oil phase. At low MG concentrations (1–2 wt%), the larger formed and lamellar MG crystals may pierce the interfacial fibrillar film, inducing the formation of partially coalesced droplets. In contrast, at high MG concentrations (4 wt% or above), the smaller MG crystals with enhanced interfacial activity can lead to the formation of a bilayer shell of GA nanofibrils and MG crystals, thus efficiently inhibiting the interfacial film damage and forming a jamming structure with homogeneously distributed small droplets. Compared to pure GA nanofibril gel emulsions, the GA−MG dual-structured gel emulsions showed significantly improved mechanical performance as well as good thermoresponsive behavior. Moreover, these stable GA−MG gel emulsions can be used as food-grade delivery vehicles for encapsulating and protecting hydrophobic and hydrophilic bioactive cargoes. They also have great potential as novel and efficient aroma delivery systems showing highly controlled volatile release. The dual-structured emulsion strategy is expected to broaden the applications of natural saponin GA-based gel emulsions in the food, pharmaceutical, and personal care industries.

Ionic migration degrades not only the characteristics of halide perovskite solar cells, but also those of perovskite X-ray detectors. Here Yang et al. employ heteroepitaxial BiOBr to passivate Cs2AgBiBr6 double perovskite, which suppresses ionic migration and obtain high performance X-ray detectors.

Summary: Resin embedding is widely used to dissect the fine structure of bio-tissue with electron and optical microscopy. However, it is difficult to embed large-volume tissues with resin. Here, we modified the formula of LR-White resin to prevent the sample cracking during polymerization process and applied this method to the intact brains of mouse, ferret, and macaque. Meanwhile, we increased the fluorescence preservation rate for green fluorescent protein (GFP) from 73 ± 4.0% to 126 ± 3.0% and tdTomato from 60 ± 3.3% to 117 ± 2.8%. Combined with the whole-brain imaging system, we acquired the cytoarchitectonic information and the circuit information such as individual axon and boutons which were labeled with multiple fluorescent proteins. This method shows great potential in the study of continuous fine microstructure information in large-volume tissues from different species, which can facilitate the neuroscience research and help the understanding of the structure-function relationship in complex bio-tissues.

The naturally occurring saponins exhibit remarkable interfacial activity and also possess many biological activities linking to human health benefits, which make them particularly attractive as bifunctional building blocks for formulation of colloidal multiphase food systems. This review focuses on two commonly used food-grade saponins, Quillaja saponins (QS) and glycyrrhizic acid (GA), with the aim of clarifying the relationship between the structural features of saponin molecules and their subsequent self-assembly and interfacial properties. The recent applications of these two saponins in various colloidal multiphase systems, including liquid emulsions, gel emulsions, aqueous foams and complex emulsion foams, are then discussed. A particular emphasis is on the unique use of GA and GA nanofibrils as sole stabilizers for fabricating various multiphase food systems with many advanced qualities including simplicity, ultrastability, stimulability, structural viscoelasticity and processability. These natural saponin and saponin-based colloids are expected to be used as sustainable, plant-based ingredients for designing future foods, cosmetics and pharmaceuticals.

7S protein is one of four major storage proteins in soybean. The present study compared the relative cholesterol-lowering activity of the 7S protein with that of total soybean protein isolate (SPI). Forty-eight hamsters were divided into six groups fed non-cholesterol diet or one of five high-cholesterol diets containing 24.2% casein, 12.1–24.2% SPI, 12.1–24.2% 7S protein, respectively. Results showed the 7S protein reduced plasma total cholesterol (TC) by 20–41% while SPI reduced it by 9–16%. 7S protein increased the fecal excretion of neutral sterols by 43–116%, while SPI only had 18–68% increase. 7S protein increased the excretion of total bile acids by 208–367%, while SPI only increased it by 41–86%. 7S protein could not only down regulate gene expression of HMG-CoA reductase but also up regulate gene expression of CYP7A1. It was concluded that dietary 7S protein was more hypocholesterolemic than SPI, mediated by increasing the sterol excretion and decreasing the cholesterol synthesis.

Paraffin embedding is widely used in microscopic imaging for preparing biological specimens. However, owing to significant fluorescence quenching during the embedding process, it is not compatible with fluorescent-labeling techniques, such as transgenic and viral labeling using green fluorescent protein (GFP). Here, we investigate the quenching mechanism and optimize the embedding process to improve the preservation of fluorescence intensity. The results show that dehydration is the main reason for fluorescence quenching during paraffin embedding, caused by the full denaturation of GFP molecules in ethyl alcohol. To evaluate fluorescent and morphological preservation, we modified the embedding process using tertiary butanol (TBA) instead of ethyl alcohol. Fluorescence intensity following TBA dehydration increased 12.08-fold of that observed in the traditional method. We obtained uniform fluorescence maintenance throughout the whole mouse brain, while the continuous apical dendrites, spines, and axon terminals were shown evenly within the cortex, hippocampus, and the amygdala. Moreover, we embedded a whole rat brain labeled with AAV in the prelimbic cortex (Prl). With the axon terminals in different areas, such as the caudate putamen, thalamus, and pyramidal tract, the results showed a continuous tract of Prl neurons throughout the whole brain. This method was also suitable for tdTomota labeled samples. These findings indicate that this modified embedding method could be compatible with GFP and provides a potential turning point for applications in the fluorescent labeling of samples.

In order to ensure the safety of traffic, asphalt pavement is commonly required to utilize aggregates with excellent anti-abrasion property. This results in the lack of high-quality aggregates. The incorporation of solid waste in the aggregates is regarded as a high potential alternative for solving this problem. Since its material properties, such as rough surface, high Polished Stone Value (PSV) and the excellent adhesion property of asphalt, Basic Oxygen Furnace (BOF) slag can effectively improve the skid resistance of asphalt mixtures. First, the material properties of BOF slag are reviewed in this study. Then, the skid resistance of asphalt mixtures and aggregates are commendably evaluated by the Polished Stone Value test, Wehner/Schulze Tester, Aachen Polishing Machine, British Pendulum Test and Sand Patch test. The physical and mechanical properties of BOF slag play a key role in asphalt mixtures. This review found that the skid resistance mechanism of the BOF slag asphalt mixture is governed by factors such as BOF slag properties, incorporation methods and gradation types. Finally, the economic and environmental benefits of BOF slag asphalt mixtures were discussed. In addition, the function of gas catalysis and the melting of ice and snow can be added to the BOF slag asphalt mixture for a cleaner development in engineering. Furthermore, the existing problems, research directions and corresponding measures in this field are directed towards more durable and functional asphalt pavement construction.

Understanding amazingly complex brain functions and pathologies requires a complete cerebral vascular atlas in stereotaxic coordinates. Making a precise atlas for cerebral arteries and veins has been a century-old objective in neuroscience and neuropathology. Using micro-optical sectioning tomography (MOST) with a modified Nissl staining method, we acquired five mouse brain data sets containing arteries, veins, and microvessels. Based on the brain-wide vascular spatial structures and brain regions indicated by cytoarchitecture in one and the same mouse brain, we reconstructed and annotated the vascular system atlas of both arteries and veins of the whole mouse brain for the first time. The distributing patterns of the vascular system within the brain regions were acquired and our results show that the patterns of individual vessels are different from each other. Reconstruction and statistical analysis of the microvascular network, including derivation of quantitative vascular densities, indicate significant differences mainly in vessels with diameters less than 8 μm and large than 20 μm across different brain regions. Our precise cerebral vascular atlas provides an important resource and approach for quantitative studies of brain functions and diseases.

Three-dimensional imaging of whole mammalian brains at single-neuron resolution has generated terabyte (TB)- and even petabyte (PB)-sized datasets. Due to their size, processing these massive image datasets can be hindered by the computer hardware and software typically found in biological laboratories. To fill this gap, we have developed an efficient platform named TDat, which adopts a novel data reformatting strategy by reading cuboid data and employing parallel computing. In data reformatting, TDat is more efficient than any other software. In data accessing, we adopted parallelization to fully explore the capability for data transmission in computers. We applied TDat in large-volume data rigid registration and neuron tracing in whole-brain data with single-neuron resolution, which has never been demonstrated in other studies. We also showed its compatibility with various computing platforms, image processing software and imaging systems.

With the development of hybrid imaging scanners, micro-CT is widely used in locating abnormalities, studying drug metabolism, and providing structural priors to aid image reconstruction in functional imaging. Due to the low contrast of soft tissues, segmentation of soft tissue organs from mouse micro-CT images is a challenging problem. In this paper, we propose a mouse segmentation scheme based on dynamic contrast enhanced micro-CT images. With a homemade fast scanning micro-CT scanner, dynamic contrast enhanced images were acquired before and after injection of non-ionic iodinated contrast agents (iohexol). Then the feature vector of each voxel was extracted from the signal intensities at different time points. Based on these features, the heart, liver, spleen, lung, and kidney could be classified into different categories and extracted from separate categories by morphological processing. The bone structure was segmented using a thresholding method. Our method was validated on seven BALB/c mice using two different classifiers: a support vector machine classifier with a radial basis function kernel and a random forest classifier. The results were compared to manual segmentation, and the performance was assessed using the Dice similarity coefficient, false positive ratio, and false negative ratio. The results showed high accuracy with the Dice similarity coefficient ranging from 0.709 ± 0.078 for the spleen to 0.929 ± 0.006 for the kidney.
