Environmental factors (such as temperature and water) and their coupling effects will change the unloading creep properties of surrounding rock after tunnel excavation, threatening the stability of surrounding rock and the safety maintenance of construction sites. To analyze the effects of temperature, seepage flow, and their coupling on the creep characteristics of sandstone unloading, the triaxial unloading creep test was carried out on the sandstone specimen (φ50 mm×100 mm) by using the self-designed temperature control system and the improved pore water conduction specimen sealing device. The results show that during the creep time of 300 min after unloading the confining stress, the axial compression and radial expansion creep speed increase gradually and then slow down. The strain is mainly radial creep. The decrease in temperature leads to the increase in axial creep deformation and the decrease in radial creep deformation of the dry and saturated specimens. With the increase in seepage water pressure, the axial strain reduces, and the seepage water pressure suppresses the axial creep deformation, while the radial strain increases significantly, and the expansion phenomenon is obvious. Seepage water pressure promotes radial creep deformation. With the increase of seepage water pressure, the axial unloading creep rate decreases and the radial unloading creep rate increases. Under the coupling effect of temperature and seepage, the creep rate curve of unloading is divided into the attenuation stage and the stable stage, and the deformation in the attenuation stage is dominant. The radial creep rate is higher than the axial creep rate, and the fluctuation amplitude of the radial creep rate is small, which reaches stability fast. This study can provide scientific information for the stability evaluation of surrounding rock in deep underground engineering.

IntroductionPorcine Parvovirus (PPV) is a significant pathogen in the pig industry, with eight genotypes, including PPV7, identified since its emergence in 2016. Co-infections with viruses such as Porcine Circovirus 2 (PCV2) and Porcine Reproductive and Respiratory Syndrome Virus (PRRSV) pose serious risks to swine health. Thus, there is an urgent need for rapid, sensitive, and specific detection methods suitable for use in field settings or laboratories with limited resources.MethodsWe developed a CRISPR/Cas12a-based assay combined with recombinase polymerase amplification (RPA) for the rapid detection of PPV7. Specific RPA primers and five CRISPR RNAs (crRNAs) were designed to target a highly conserved region within the NS1 gene of PPV7. Optimization of crRNA and single-stranded DNA (ssDNA) concentrations was performed to enhance the assay’s performance.ResultsCrRNA optimization identified crRNA-05 as the optimal candidate for Cas12a-based detection of PPV7, as all synthesized crRNAs demonstrated similar performance. The optimal crRNA concentration was determined to be 200 nM, yielding consistent results across tested concentrations. For ssDNA optimization, the strongest fluorescence signal was achieved with 500 nM of the FAM-BHQ ssDNA receptor. The assay showed a minimal detection limit of 100copies/μl for PPV7, confirmed through fluorescence and lateral flow detection methods. Specificity testing indicated that only PPV7 DNA samples returned positive results, confirming the assay’s accuracy. In tests of 50 lung tissue samples from diseased pigs, the RPA-Cas12a assay identified 29 positive samples (58%), surpassing the 22 positive samples (44%) detected by conventional PCR. This highlights the RPA-Cas12a method’s enhanced detection capability and its potential utility in clinical surveillance and management of PPV7 in swine populations.DiscussionThe RPA-Cas12a assay effectively detects PPV7 in clinical samples, enhancing disease surveillance and control in pigs. Its adaptability to resource-limited settings significantly improves PPV7 management and prevention strategies, thereby supporting the overall health and development of the pig industry.

Abstract Palladium‐catalyzed Suzuki–Miyaura (SM) coupling is a valuable method for forming C─C bonds, including those between aryl moieties. However, achieving atroposelective synthesis of axially chiral styrenes via SM coupling remains challenging. In this study, a palladium‐catalyzed atroposelective Suzuki–Miyaura coupling between gem‐diborylalkenes and aryl halides is presented. Using the monophosphine ligand Me‐BI‐DIME (L2), a range of axially chiral tetra‐substituted acyclic styrenes with high yields and excellent enantioselectivities are successfully synthesized. Control experiments reveal that the gem‐diboryl group significantly influences the product enantioselectivities and the coupling prefers to occur at sites with lower steric hindrance. Additionally, the alkenyl boronate group in the products proves versatile, allowing for various transformations while maintaining high optical purities.

Bistatic radar exhibits spatial isomerism and diverse configurations, leading to unique clutter characteristics distinct from those of monostatic radar. The clutter rank serves as a pivotal indicator of clutter characteristics, enabling the quantification of clutter severity. Space-time adaptive processing (STAP) is a critical technique to detect moving targets, and clutter rank determines the number of independent and identically distributed (IID) training samples and the degree of freedom (DOF) for effective suppression of clutter that STAP requires. Therefore, the accurate estimation of clutter rank for bistatic radar can provide a crucial indicator for designing and constructing STAP processors, thereby facilitating fast and efficient clutter suppression in bistatic radar systems. This study is based on the idea that clutter rank is the number of prolate spheroidal wave function (PSWF) orthogonal bases utilized for approximating the clutter signal. Firstly, the challenge of utilizing PSWF orthogonal bases for approximating the clutter signal in bistatic radar is elucidated. This pertains to the fact that, unlike monostatic radar clutter, bistatic radar clutter is not capable of being expressed as a single-frequency signal. The clutter rank estimation for bistatic radar is thus derived as the frequency bandwidth estimation. Secondly, to achieve this estimation, the frequency distribution of each individual scattering unit is investigated, thereby determining their extending frequency broadening (EFB) as compared to that of single-frequency. Subsequently, the integral average of EFB across the entire range bin is computed, ultimately enabling the acquisition of bistatic radar’s frequency bandwidth. Finally, the estimation method is extended to non-side-looking mode and limited observation areas with pattern modulation. Simulation experiments confirm that our proposed method provides accurate clutter rank estimations, surpassing 99% proportions of large eigenvalues across various bistatic configurations, observation modes, and areas.

The reservoirs of ultra-deep and low-permeability sandstones typically exhibit characteristics of lithological tightness and poor physical properties. Fractures control the oil and gas content as well as the productivity of such reservoirs. However, the distribution of fractures is complex, exhibiting strong heterogeneity. Therefore, a systematic study on reservoir fracture modeling can provide geological foundations for the development of such reservoirs. Due to the considerable burial depth of these reservoirs, conventional methods relying solely on seismic information have limited reliability, and the established discrete network models of fractures are often less dependable. In this paper, taking the X gas reservoir in a basin in western China as an example, we discuss a fracture modeling method based on the integration of geological information to enhance the efficiency and accuracy of fracture modeling. The modeling method primarily involves the use of deterministic methods to obtain large-scale fractures, while random simulation is employed for small and medium-scale fractures. The fracture development control factors and seismic attribute information are integrated using permanence of ratios (PR) model to establish a fracture development probability field model. Subsequently, the geometric parameters of fractures and the fracture density model are used as input parameters to generate a discrete network model of small and medium-scale fractures using a object-based modeling method. Finally, based on the fracture equivalent property model and verified through geological understanding, analysis of production dynamics, and numerical simulation of the gas reservoir, it is demonstrated that the fracture model established using the proposed method aligns with geological understanding and exhibits high reliability.

Amino acid permeases (AAPs) transporters are crucial for the long-distance transport of amino acids in plants, from source to sink. While Arabidopsis and rice have been extensively studied, research on foxtail millet is limited. This study identified two transcripts of SiAAP9, both of which were induced by NO3− and showed similar expression patterns. The overexpression of SiAAP9L and SiAAP9S in Arabidopsis inhibited plant growth and seed size, although SiAAP9 was found to transport more amino acids into seeds. Furthermore, SiAAP9-OX transgenic Arabidopsis showed increased tolerance to high concentrations of glutamate (Glu) and histidine (His). The high overexpression level of SiAAP9 suggested its protein was not only located on the plasma membrane but potentially on other organelles, as well. Interestingly, sequence deletion reduced SiAAP9’s sensitivity to Brefeldin A (BFA), and SiAAP9 had ectopic localization on the endoplasmic reticulum (ER). Protoplast amino acid uptake experiments indicated that SiAAP9 enhanced Glu transport into foxtail millet cells. Overall, the two transcripts of SiAAP9 have similar functions, but SiAAP9L shows a higher colocalization with BFA compartments compared to SiAAP9S. Our research identifies a potential candidate gene for enhancing the nutritional quality of foxtail millet through breeding.

Ear photosynthesis plays a key role in wheat photosynthesis during the grain filling stage, particularly under drought stress. Thus, dissecting the responsibilities of the glume and awn in photosynthetic carbon fixation and assimilates transportation during the grain filling stage in spikes is imperative. In this study, the detachment of the glume (DG) and awn (DA) of a wheat variety (Pubing143) was used to estimate their influences on ear photosynthesis and dry matter distribution. Radioactive carbon-14 (14C) isotope was detected by a multifunctional liquid scintillation counting system. The accumulation of 14C assimilates and their contributions to grain weight were then calculated. Under well-watered conditions, ear photosynthesis was reduced by 16.8 % and 46.2 % 25 d after anthesis (DAA) in the de-glumed control (DGC) and de-awned control (DAC) treatments, respectively, compared with the intact ear control (IEC). Under drought stress, ear photosynthesis was reduced by 46 % and 74.2 % at 25 DAA after removing the glume and awn, respectively. Under normal conditions, the number of 14C assimilates of DGC, and DAC was reduced by 14.6 % and 20.9 % in grains at 25 DAA, respectively, compared with the IEC. Compared with IED, the 14C assimilates of DGD, and DAD declined by 17.2 % and 27 %, respectively, in grains at 25 DAA under drought conditions. Under well-watered conditions, the grain weight per pot was reduced by 11.2 % and 25.4 % in the de-glumed control (DGC) and de-awned control (DAC) treatments, respectively, compared with the intact ear control (IEC). The grain weight per pot was further reduced after removing the glume and awn (16 % and 32.2 %, respectively) under drought stress. Furthermore, the awn contribution to grain weight was twice that of the glume. Our results suggest that the glume and awn of ears play prominent roles during grain filling in wheat, especially under drought stress, and that the awn is more crucial than the glume.

Abstract Chimeric antigen receptor T (CAR-T) cell targeting CD19 antigen has achieved exhilarative clinical efficacy in B-cell malignancies. However, challenges still remain for the currently approved anti-CD19 CAR-T therapies, including high recurrence rates, side effects and resistance. Herein, we aim to explore combinatorial therapy by use of anti-CD19 CAR-T immunotherapy and gallic acid (GA, an immunomodulatory natural product) for improving treatment efficacy. We assessed the combinatorial effect of anti-CD19 CAR-T immunotherapy with GA in cell models and a tumor-bearing mice model. Then, the underlying mechanism of GA on CAR-T cells were investigated by integrating network pharmacology, RNA-seq analysis and experimental validation. Furthermore, the potential direct targets of GA on CAR-T cells were explored by integrating molecular docking analysis with surface plasmon resonance (SPR) assay. The results showed that GA significantly enhanced the anti-tumor effects, cytokine production as well as the expansion of anti-CD19 CAR-T cells, which may be mainly through the activation of IL4/JAK3-STAT3 signaling pathway. Furthermore, GA may directly target and activate STAT3, which may, at least in part, contribute to STAT3 activation. Overall, the findings reported here suggested that the combination of anti-CD19 CAR-T immunotherapy with GA would be a promising approach to increase the anti-lymphoma efficacy.

The role of human activity in shaping the terrestrial environment has been a core scientific issue of interest across various disciplines. However, it remains unclear whether there are significant differences in the patterns of the anthropogenic impact on the terrestrial environment in terms of spatial and temporal dimensions, and we are yet to identify the underlying factors that have driven it. Here, we present an analysis of sporopollen and geochemical proxies from a section of the Anjiangbei site (AJB) on the Yunnan Plateau, spanning the Ming–Qing period, in order to explore the spatio-temporal variation in the anthropogenic impact on the terrestrial environment in the Lake Dian basin. Integrating the reported multidisciplinary evidence, we aim to reveal the influencing factors of anthropogenic impact. Our results show that there were remarkable differences in anthropogenic impact on the terrestrial environment in the Lake Dian basin between the Late Bronze Age and the Ming–Qing period. Changes in crop vegetation and the forest were all affected by human activity in the Lake Dian basin during the two periods, and were more evident during the Ming–Qing period. The heavy metal pollution in the soil was obvious during the Ming–Qing period. The increase in the intensity of human activity, especially the rise in population, could be attributed to changes in the hydrological environment in the Lake Dian basin during the Late Bronze Age and to geopolitical change during the Ming–Qing period. This study reveals the different patterns in human impact on the terrestrial environment in the Lake Dian basin during the Late Bronze Age and the Ming–Qing period, providing new evidence to enable a deeper understanding of past human–environment interactions on the Yunnan Plateau.

Foxtail millet (Setaria italica L.) is an important crop grown worldwide as a food and fodder crop owing to its potential nutritional and feed values. High-efficiency herbicide varieties is crucial to achieving efficient weeding and ensuring successful foxtail millet production. Herbicides affect several morphological and physiological indicators of foxtail millet. In this study we aimed to evaluate the damage caused by herbicides, improve their effectiveness, and select indicators that accurately reflect herbicide resistance in foxtail millet. Jingu 21, which has the largest planting area in Shanxi province and even the whole of China, is selected as the experimental material to be sown in the field in 2022. A total of 31 herbicides were applied, and 21 traits, including morphological, physiological, and yield-component traits, were measured to assess millet resistance. Principal component analysis was employed to transform these 21 conventional traits into seven independent and comprehensive indexes. These indexes were screened using regression analysis, resulting in the selection of the following indicators: the surface area of the second leaf from the top, peroxidase activity, catalase activity, malondialdehyde content, chlorophyll (a + b), grain weight per ear, and yield. Through membership function and cluster analyses, the resistance of Jingu 21 to 31 herbicides was divided into five categories: extremely weakly resistant, weakly resistant, moderately resistant, strongly resistant, and extremely strongly resistant. Jingu 21 exhibited extremely strong resistance to lactofen, butachlor, and anilofos. After an investigation into the effectiveness of herbicides, it was found that eight herbicides had good effects.

Vibrio cholerae serogroup O1 (V. cholerae O1) is closely associated with cholera epidemics and has two main immunologically distinguishable serotypes, Ogawa and Inaba. Isolates serotype as Ogawa if the O-antigen polysaccharide (O-PS) is methylated or as Inaba if the O-PS is not methylated. This methylation is mediated by a methyltransferase encoded by the rfbT gene, and the mutation and low expression of rfbT results in serotype switch from Ogawa to Inaba. Previously, we have shown that cAMP receptor protein (CRP) activates rfbT. In this study, we demonstrated that histone-like nucleoid structuring protein (H-NS) is directly involved in the transcriptional repression of rfbT. This finding is supported by the analyses of rfbT mRNA level, rfbT-lux reporter fusions, electrophoretic mobility shift assay (EMSA), and DNase I footprinting assay. The rfbT mRNA abundances were significantly increased by deleting hns rather than fis which also preferentially associates with AT-rich sequences. A single-copy chromosomal complement of hns partly restored the down-regulation of rfbT. Analysis of rfbT-lux reporter fusions validated the transcriptional repression of hns. Subsequent EMSA and DNase I footprinting assay confirmed the direct binding of H-NS to rfbT promoter and mapped the exact binding site which was further verified by site-directed mutagenesis and promoter functional analysis. Furthermore, we found that in hns deletion mutant, CRP is no longer required for transcriptionally activating rfbT, suggesting that CRP functions as a dedicated transcription factor to relieve H-NS repression at rfbT. Together, this study expanded our understanding of the genetic regulatory mechanism of serotype conversion by global regulators in V. cholerae O1.

The deformation and failure of the surrounding rock during roadway excavation often determine the choice of supporting methods. To study the deformation behavior and failure modes of the surrounding rock after excavation under unloading stress, structural model tests were carried out with a novel experimental device. The present structural model test using partial hollow thick-walled cylinder cement mortar specimen φ200 mm × 280 mm with a horizontal central circular hole of 60 mm diameter and the hollow height of 160 mm was conducted to investigate the deformation, failure characteristics, and AE response of a whole testing process from excavation to postunloading state. Experimental results revealed that the amount of deformation behind the surface is significantly higher than that in front of the surface, and the radial strain increases with the increase of the distance from the surface within the range affected by unloading. Furthermore, the unloading rate has a little effect on the radial deformation of the surrounding rock in front of the surface, but has a substantial effect on the radial deformation behind the excavation surface. The peak value of the strain rate at the unloading rate of 2 MPa/s is much higher than that at the unloading rate of 0.1 MPa/s. According to AE results and the failure of opening the boundary, the increase of unloading rate triggers and exacerbates the damage of the specimen under high in situ stress conditions. The surrounding rock expanded to the inner hollow, accompanied by large dilation and volume changes, and it resulted in the shrinkage of the hole diameter. A large number of rock slices are generated at the opening curved free surface and then fell off, whose morphology is similar to the rock blocks that fell off after caving failure and rock burst in situ field. The results show that the system can accurately simulate the mechanical response and acoustic emission response of the excavated surrounding rock, which provides a new experimental method for further study of the unloading response of the surrounding rock.

In recent years, significant progress has been made in image super-resolution (SR) methods based on convolutional neural networks. However, most of them do not fully utilize multi-scale feature correspondence in the image SR process, resulting in blurred and artifact detail restoration, especially for SR tasks with larger scaling factors (i.e. $\times 4$ and $\times 8$ ). A multi-scale feature-enhanced SR network (MFENet) is proposed to solve the problems mentioned above. Specifically, the super-resolution method is enhanced by utilizing multi-scale feature correspondence in two ways. On the one hand, we propose an effective multi-scale non-local attention module and it can fully exploit a large number of low-level features at different scales of a single low-resolution (LR) feature map by exhaustively evaluating the correlations in local feature, in-scale non-local feature, and cross-scale non-local feature. On the other hand, the mapping from high-resolution (HR) to LR images is explored by a feedback branch we introduced, that provides an additional constraint for images reconstruction. It can penalize incorrect SR predictions to achieve better SR performance. Experimental results on 5 benchmark datasets show that our method outperforms state-of-the-art methods in terms of both subjective visual quality and objective quantitative metrics.

Cholera, caused by pathogenic Vibrio cholerae, poses a significant public health risk through water and food transmission. Biofilm-associated V. cholerae plays a crucial role in seasonal cholera outbreaks as both a reservoir in aquatic environments and a direct source of human infection. Although VP3, a lytic phage, shows promise in eliminating planktonic V. cholerae from the aquatic environment, its effectiveness against biofilm-associated V. cholerae is limited. To address this limitation, our proposed approach aims to enhance the efficacy of VP3 in eliminating biofilm-associated V. cholerae by augmenting the availability of phage receptors on the surface of Vibrio cholerae. TolC is a receptor of VP3 and a salt efflux pump present in many bacteria. In this study, we employed NaCl as an enhancer to stimulate TolC expression and observed a significant enhancement of TolC expression in both planktonic and biofilm cells of V. cholerae. This enhancement led to improved adsorption of VP3. Importantly, our findings provide strong evidence that high salt concentrations combined with VP3 significantly improve the elimination of biofilm-associated V. cholerae. This approach offers a potential strategy to eliminate biofilm-formation bacteria by enhancing phage–host interaction.

Low-temperature stress limits the growth and development of foxtail millet. Freezing stress caused by sudden temperature drops, such as late-spring coldness, often occurs in the seedling stage of foxtail millet. However, the ability and coping strategies of foxtail millet to cope with such stress are not clear. In the present study, we analyzed the self-regulatory mechanisms of freezing stress in foxtail millet. We conducted a physiological study on foxtail millet leaves at −4 °C for seven different durations (0, 2, 4, 6, 8, 10, and 12 h). Longer freezing time increased cell-membrane damage, relative conductance, and malondialdehyde content. This led to osmotic stress in the leaves, which triggered an increase in free proline, soluble sugar, and soluble protein contents. The increases in these substances helped to reduce the damage caused by stress. The activities of superoxide dismutase, peroxidase, and catalase increased reactive oxygen species (ROS) content. The optimal time point for the response to freezing stress was 8 h after exposure. The transcriptome analysis of samples held for 8 h at −4 °C revealed 6862 differentially expressed genes (DEGs), among which the majority are implicated in various pathways, including the starch and sucrose metabolic pathways, antioxidant enzyme pathways, brassinolide (BR) signaling pathway, and transcription factors, according to Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment. We investigated possible crosstalk between BR signals and other pathways and found that BR signaling molecules were induced in response to freezing stress. The beta-amylase (BAM) starch hydrolase signal was enhanced by the BR signal, resulting in the accelerated degradation of starch and the formation of sugars, which served as emerging ROS scavengers and osmoregulators to resist freezing stress. In conclusion, crosstalk between BR signal transduction, and both starch and sucrose metabolism under freezing stress provides a new perspective for improving freezing resistance in foxtail millet.

The cytochrome P450 monooxygenases (CYP450) are the largest enzyme family in plant metabolism and widely involved in the biosynthesis of primary and secondary metabolites. Foxtail millet (Setaria italica (L.) P. Beauv) can respond to abiotic stress through a highly complex polygene regulatory network, in which the SiCYP450 family is also involved. Although the CYP450 superfamily has been systematically studied in a few species, the research on the CYP450 superfamily in foxtail millet has not been completed. In this study, three hundred and thirty-one SiCYP450 genes were identified in the foxtail millet genome by bioinformatics methods, which were divided into four groups, including forty-six subgroups. One hundred and sixteen genes were distributed in thirty-three tandem duplicated gene clusters. Chromosome mapping showed that SiCYP450 was distributed on seven chromosomes. In the SiCYP450 family of foxtail millet, 20 conserved motifs were identified. Cis-acting elements in the promoter region of SiCYP450 genes showed that hormone response elements were found in all SiCYP450 genes. Of the three hundred and thirty-one SiCYP450 genes, nine genes were colinear with the Arabidopsis thaliana genes. Two hundred SiCYP450 genes were colinear with the Setaria viridis genes, including two hundred and forty-five gene duplication events. The expression profiles of SiCYP450 genes in different organs and developmental stages showed that SiCYP450 was preferentially expressed in specific tissues, and many tissue-specific genes were identified, such as SiCYP75B6, SiCYP96A7, SiCYP71A55, SiCYP71A61, and SiCYP71A62 in the root, SiCYP78A1 and SiCYP94D9 in leaves, and SiCYP78A6 in the ear. The RT-PCR data showed that SiCYP450 could respond to abiotic stresses, ABA, and herbicides in foxtail millet. Among them, the expression levels of SiCYP709B4, SiCYP71A11, SiCYP71A14, SiCYP78A1, SiCYP94C3, and SiCYP94C4 were significantly increased under the treatment of mesotrione, florasulam, nicosulfuron, fluroxypyr, and sethoxydim, indicating that the same gene might respond to multiple herbicides. The results of this study will help reveal the biological functions of the SiCYP450 family in development regulation and stress response and provide a basis for molecular breeding of foxtail millet.

Vibrio fluvialis is a halophilic Gram-negative bacterium regarded as an emerging unusual enteric pathogen of increasing public health concern. Our previous work has identified two type VI secretion systems (T6SSs) in V. fluvialis, VflT6SS1, and VflT6SS2, and the latter is functional in mediating interbacterial competitiveness. However, its antibacterial effectors remain to be clarified. In this work, we focused on a new potential effector/immunity pair TssI2/TsiI2. Bioinformatics analysis revealed that the C-terminal domain of TssI2 belongs to a widespread family of pesticin, and its antibacterial toxicity and corresponding protection by TsiI2 were proved via bacterial killing assays, and their action sites were localized to the periplasm of bacterial cells. The interaction of TssI2 and TsiI2 was demonstrated by the bacterial adenylate cyclase two-hybrid, protein pull-down and isothermal titration calorimetry assays. Site-directed mutagenesis demonstrated that, in addition to Glu-844, Thr-863, and Asp-869, which correspond to three reported residues in pesticin of Yersinia pestis, additional residues including Phe-837, Gly-845, Tyr-851, Gly-867, Gln-963, Trp-975, and Arg-1000 were also proved to be crucial to the bactericidal activity of TssI2. Muramidase/lysozyme-related peptidoglycan (PG) hydrolase activities of TssI2 and its variants were validated with permeabilized Escherichia coli cells and purified PG substrate. Based on sequence homologies at C-terminals in various V. fluvialis isolates, TssI2 was subdivided into five clusters (12–22% identity among them), and the antibacterial activities of representative effectors from other four Clusters were also confirmed through periplasmic over-expression in E. coli host. Two selected cognate immunities were proved to confer protection against the toxicities of their effectors. Additionally, TsiI2, which belongs to Cluster I, exhibited cross-protection to effector from Cluster V. Together, current findings expand our knowledge of the diversity and consistency of evolved VgrG effectors in V. fluvialis and on how VflT6SS2 mediates a competitive advantage to gain a better survival.

Psoriasis is a chronic, inflammatory skin disease, frequently associated with dyslipidemia. Lipid disturbance in psoriasis affects both circulatory system and cutaneous tissue. Epidermal Langerhans cells (LCs) are tissue-resident DCs that maintain skin immune surveillance and mediate various cutaneous disorders, including psoriasis. However, the role of LCs in psoriasis development and their lipid metabolic alternation remains unclear. Here, we demonstrate that epidermal LCs of psoriasis patients enlarge with longer dendrites and possess elevated IL-23p19 mRNA and a higher level of neutral lipids when compared with normal LCs of healthy individuals. Accordantly, epidermal LCs from imiquimod-induced psoriasis-like dermatitis in mice display overmaturation, enhanced phagocytosis, and excessive secretion of IL-23. Remarkably, these altered immune properties in lesional LCs are tightly correlated with elevated neutral lipid levels. Moreover, the increased lipid content of psoriatic LCs might result from impaired autophagy of lipids. Bulk RNA-Seq analysis identifies dysregulated genes involved in lipid metabolism, autophagy, and immunofunctions in murine LCs. Overall, our data suggest that dysregulated lipid metabolism influences LC immunofunction, which contributes to the development of psoriasis, and therapeutic manipulation of this metabolic process might provide an effective measurement for psoriasis.

Since the 1990s, grammatical complexity has received substantial research attention in applied linguistics (Bulté and Housen, 2014). The representation of grammatical complexity has expanded in L2 writing with the application of diverse measures in empirical studies in the recent three decades (1991–2020). In response to this situation, we found it important to revisit grammatical complexity, and an exploratory factor analysis was applied to explore latent dimensions (i.e., factors) of grammatical complexity in L2 writing. We analyzed Lu’s (2011) 14 grammatical complexity measures in the L2 corpus of the British Academic Written English Corpus. We then proposed a four-factor model with “clausal subordination,” “phrasal construction”, “global length unit” and “others.” The four factors generally align with the types of grammatical complexity proposed in Lu (2011), but differences on six measures are also found. Noteworthy points were discussed to interpret the reasons behind the differences. Research implications are provided to show further research directions.

Despite its simplicity and high accuracy, the real-time efficiency of OTSU segmentation is not high. In order to promote the real-timeliness of image segmentation, this paper introduces the fruitfly optimization algorithm (FOA) to OTSU segmentation, creating an FOA-OTSU segmentation algorithm. In the proposed algorithm, the optimal threshold for segmentation is searched for by the FOA. The classic Lena picture, Flower picture and Cameraman picture were used for simulation experiment, and the results were evaluated by signal-to-noise ratio (SNR) and peak signal-to-noise ratio (PSNR). The results show that the segmentation time is reduced by about 50.0% on the premise of the PSNR and SNR evaluation criteria and the segmentation effect basically unchanged. The simulations indicate that our method converges faster and consumes fewer time than traditional OTSU algorithm, without sacrificing the segmentation accuracy. The research provides a desirable tool with high real-time performance of rapid image segmentation.

The urban heat island (UHI) effect in cities and its driving factors have long been investigated. 3-D buildings are key components of urban structures and have notable effect on the UHI effect. However, due to the incomplete 3-D building information in urban database, only a few studies investigated the impact of 3-D building morphology factors on the land surface temperature (LST). In this study, a total set of 14 2-D and 3-D building morphology factors were selected to investigate the correlation between UHI and building morphology across three megacities in China (Beijing, Wuhan, Shanghai) with Landsat-8 LST scenarios in summer and winter of 2018 and 2019, respectively. Both Spearman correlation and partial correlation analysis were applied at block scale after urban functional zone (UFZ) mapping. A number of significant observations were noted based on the multispatial and multitemporal experimental results:1) both 2-D and 3-D building morphology factors influence LST, among them building coverage ratio, building ground area divided by facade area (GA2FA), and sky view factor yield stronger correlations with LST; 2) UFZ mapping scheme and partial correlation method help with controlling the anthropogenic heat release, greenspace, and water bodies coverage variables when targeting at the influence of building morphology factors on LST. This study provides insights for building morphology design in future urban planning and management.

Glutathione S-transferases (GSTs) are a critical superfamily of multifunctional enzymes in plants. As a ligand or binding protein, GSTs regulate plant growth and development and detoxification. Foxtail millet (Setaria italica (L.) P. Beauv) could respond to abiotic stresses through a highly complex multi-gene regulatory network in which the GST family is also involved. However, GST genes have been scarcely studied in foxtail millet. Genome-wide identification and expression characteristics analysis of the foxtail millet GST gene family were conducted by biological information technology. The results showed that 73 GST genes (SiGSTs) were identified in the foxtail millet genome and were divided into seven classes. The chromosome localization results showed uneven distribution of GSTs on the seven chromosomes. There were 30 tandem duplication gene pairs belonging to 11 clusters. Only one pair of SiGSTU1 and SiGSTU23 were identified as fragment duplication genes. A total of ten conserved motifs were identified in the GST family of foxtail millet. The gene structure of SiGSTs is relatively conservative, but the number and length of exons of each gene are still different. The cis-acting elements in the promoter region of 73 SiGST genes showed that 94.5% of SiGST genes possessed defense and stress-responsive elements. The expression profiles of 37 SiGST genes covering 21 tissues suggested that most SiGST genes were expressed in multiple organs and were highly expressed in roots and leaves. By qPCR analysis, we found that 21 SiGST genes were responsive to abiotic stresses and abscisic acid (ABA). Taken together, this study provides a theoretical basis for identifying foxtail millet GST family information and improving their responses to different stresses.

Thermoelectric (TE) technology, which can convert scrap heat into electricity, has attracted considerable attention. However, broader applications of TE are hindered by lacking high-performance thermoelectric materials, which can be effectively progressed by regulating the carrier concentration. In this work, a series of PbSe(NaCl)x (x = 3, 3.5, 4, 4.5) samples were synthesized through the NaCl salt-assisted approach with Na+ and Cl− doped into their lattice. Both theoretical and experimental results demonstrate that manipulating the carrier concentration by adjusting the content of NaCl is conducive to upgrading the electrical transport properties of the materials. The carrier concentration elevated from 2.71 × 1019 cm−3 to 4.16 × 1019 cm−3, and the materials demonstrated a maximum power factor of 2.9 × 10−3 W m−1 K−2. Combined with an ultralow lattice thermal conductivity of 0.7 W m−1 K−1, a high thermoelectric figure of merit (ZT) with 1.26 at 690 K was attained in PbSe(NaCl)4.5. This study provides a guideline for chemical doping to improve the thermoelectric properties of PbSe further and promote its applications.

To accelerate the industrialization of bicomponent fibers, fiber-based flexible devices, and other technical fibers and to protect the property rights of inventors, it is necessary to develop fast, economical, and easy-to-test methods to provide some guidance for formulating relevant testing standards. A quantitative method based on cross-sectional in-situ observation and image processing was developed in this study. First, the cross-sections of the fibers were rapidly prepared by the non-embedding method. Then, transmission and reflection metallographic microscopes were used for in-situ observation and to capture the cross-section images of fibers. This in-situ observation allows for the rapid identification of the type and spatial distribution structure of the bicomponent fiber. Finally, the mass percentage content of each component was calculated rapidly by AI software according to its density, cross-section area, and total test samples of each component. By comparing the ultra-depth of field microscope, differential scanning calorimetry (DSC), and chemical dissolution method, the quantitative analysis was fast, accurate, economical, simple to operate, energy-saving, and environmentally friendly. This method will be widely used in the intelligent qualitative identification and quantitative analysis of bicomponent fibers, fiber-based flexible devices, and blended textiles.

Recent deep learning has shown great potential in super-resolution (SR) tasks. However, most deep learning-based SR networks are optimized via pixel-level loss (i.e., L1, L2, and MSE), which forces the networks to output the average of all possible predictions, leading to blurred details. Especially in SR tasks with large scaling factors (i.e., ×4, ×8), the limitation is further aggravated. To alleviate this limitation, we propose a Gradient-Prior-based Super-Resolution network (GPSR). Specifically, a detail-preserving Gradient Guidance Strategy is proposed to fully exploit the gradient prior to guide the SR process from two aspects. On the one hand, an additional gradient branch is introduced into GPSR to provide the critical structural information. On the other hand, a compact gradient-guided loss is proposed to strengthen the constraints on the spatial structure and to prevent the blind restoration of high-frequency details. Moreover, two residual spatial attention adaptive aggregation modules are proposed and incorporated into the SR branch and the gradient branch, respectively, to fully exploit the crucial intermediate features to enhance the feature representation ability. Comprehensive experimental results demonstrate that the proposed GPSR outperforms state-of-the-art methods regarding both subjective visual quality and objective quantitative metrics in SR tasks with large scaling factors (i.e., ×4 and ×8).

Chimeric antigen receptor (CAR) T cells targeting CD19 antigen have produced remarkable clinical outcomes for cancer patients. However, identifying measures to enhance effector function remains one of the most challenging issues in CD19-targeted immunotherapy. Here, we report a novel approach in which a microRNA (miRNA) or short-hairpin RNA (shRNA) cassette was integrated into CAR-expressing retroviral vectors. Using this system, we generated anti-CD19 CAR-T cells co-expressing miR155 or LSD1 shRNA and found that anti-CD19 CAR-T cells with miR155 upregulation or LSD1 downregulation exhibited increased anti-tumor functions in vitro and in vivo. Transcriptional profiling analysis by RNA sequencing revealed the targets of miR155 and LSD1 in anti-CD19 CAR-T cells. Our experiments indicated that introduction of miRNA or shRNA expression into anti-CD19 CAR T-cells might be an effective strategy to improve the anti-tumor effects of CAR-T cell therapy.

This paper addresses the channel impairment to enhance the system performance of visible light communication (VLC). Inspired by the model-solving procedure in the conventional equalizer, the channel impairment compensation is formulated as a spatial memory pattern prediction problem, then we propose efficient deep-learning (DL)-based nonlinear post-equalization, combining the Volterra-aided convolutional neural network (CNN) and long-short term memory (LSTM) neural network, to mitigate the system nonlinearity and then recover the original transmitted signal from the distorted one at the receiver end. The Volterra structure is employed to construct a spatial pattern that can be easily interpreted by the proposed scheme. Then, we take advantage of the CNN to extract the implicit feature of channel impairments and utilize the LSTM to predict the memory sequence. Results demonstrate that the proposed scheme can provide a fairly fast convergence during the training stage and can effectively mitigate the overall nonlinearity of the system at testing. Furthermore, it can recover the original signal accurately and exhibits an excellent bit error rate performance as compared with the conventional equalizer, demonstrating the prospect and validity of this methodology for channel impairment compensation.

Multiple myeloma (MM) is a tumor type characterized by the unregulated proliferation of clonal plasma cells in the bone marrow. Immunotherapy based on chimeric antigen receptor T cell (CAR-T) therapy has achieved exciting success in the treatment of hematological malignant tumors. CD38 is highly and evenly expressed in MM and is an attractive target for MM treatment. Here, we successfully constructed two novel second-generation CAR-T cells targeting CD38 by retroviral vector transduction. CD38 CAR-T cells could be activated effectively after stimulation with CD38-positive tumor cells and secrete cytokines such as IFN-γ and TNF-α to promote tumor cell apoptosis in in vitro experiments. Real-time fluorescence monitoring experiments, luciferase detection experiments and flow cytometry experiments revealed the efficient and specific killing abilities of CD38 CAR-T cells against CD38-positive tumor cells. The proliferation ability of CD38 CAR-T cells in vitro was higher than that of untransduced T cells. Further antitumor experiments in vivo showed that CD38 CAR-T cells could be quickly activated to secrete IFN-γ and eliminate tumors. Thus, novel CD38-targeted second-generation CAR-T cells have efficient and specific antitumor activity and may become a novel therapy for the clinical treatment of MM.

Heavy metal pollution is a serious threat to human health and the ecological environment, but adsorption technology based on nano adsorbents can effectively treat the crisis. However, due to the nanoscale effect, nano adsorbents have some crucial shortcomings, such as recycling difficulty and the loss of nanoparticles, which seriously limit their application. The feasible assembly of nano adsorbents is an accessible technology in urgent need of a breakthrough. In this study, three-dimensional (3D) adsorbent (MF/Ti3C2Tx/PmPD) with excellent performance and favorable recyclability was prepared by interfacial polymerization with melamine foam (MF) as the framework, two-dimensional (2D) titanium carbide (Ti3C2Tx) as the bridge and Poly (m-Phenylenediamine) (PmPD) as the active nano component. The morphology, structure, mechanical property of MF/Ti3C2Tx/PmPD and reference MF/PmPD were investigated through a scanning electron microscope (SEM), Fourier transformed infrared spectra (FT-IR), Raman scattering spectra and a pressure-stress test, respectively. Owning to the regulation of Ti3C2Tx on the morphology and structure of PmPD, MF/Ti3C2Tx/PmPD showed excellent adsorption capacity (352.15 mg/g) and favorable cycling performance. R–P and pseudo-second-order kinetics models could well describe the adsorption phenomenon, indicating that the adsorption process involved a composite process of single-layer and multi-layer adsorption and was dominated by chemical adsorption. In this research, the preparation mechanism of MF/Ti3C2Tx/PmPD and the adsorption process of Cr(VI) were systematically investigated, which provided a feasible approach for the feasible assembly and application of nano adsorbents in the environmental field.

To study shifts in the intestinal microbiota during estrus synchronization in ruminants, we characterized the intestinal microbiota in grazing Simmental cows and the possible mechanism that mediates this shift. Fourteen postpartum Simmental beef cows were synchronized beginning on day 0 (D0) with a controlled internal release device (CIDR), and cloprostenol was injected on D9 when the CIDR was withdrawn. Synchronization ended with timed artificial insemination on D12. Serum and rectal samples harvested on D0, D9, and D12 were analyzed to assess the reproductive hormones and microbiota. Reproductive hormones in the serum of the host were measured using enzyme-linked immunosorbent assay. The microbiota was characterized using 16S rRNA sequencing of the V3–V4 hypervariable region, alpha diversity and beta diversity analyses (principal coordinate analysis, PCoA), cladogram of the linear discriminant analysis effect size (LEfSe) analysis, and microbiota function analysis. Levels of the reproductive hormones, except gonadotropin-releasing hormone (p > 0.05), shifted among D0, D9, and D12 (p < 0.05). Decreased community diversity (Chao1 and ACE) was observed on D12 compared with D0 (p < 0.05). The beta diversity (PCoA) of the microbiota shifted markedly among D0, D9, and D12 (p < 0.05). The LEfSe analysis revealed shifts in the intestinal microbiota communities among D0, D9, and D12 (p < 0.05 and LDA cutoff >3.0). The KEGG pathway analysis showed that carbohydrate metabolism, genetic information and processing, the excretory system, cellular processes and signaling, immune system diseases, and the metabolism were altered (p < 0.05). Reproductive hormones (especially estradiol) were correlated with the alpha diversity indices, beta diversity indices, and an abundance of biomarkers of the shifting intestinal microbiota (p < 0.05). In conclusion, the structure, composition, and function of the intestinal microbiota were shifted during estrus synchronization in a grazing Simmental cow model, and these shifts were mediated by reproductive hormones.

The study of human dietary structures is an effective means of elucidating the subsistence patterns of our prehistoric ancestors and can highlight the processes through which humans interacted with the environment. We conducted stable isotope and archeobotanical analyses of human, animal, and plant remains at the Jiangxifen site, dated to ~900–400 BC, to explore human paleodiets and determine the environmental adaptation strategies adopted by humans in the middle valley of the Jinshajiang River in Yunnan Province. Humans predominantly consumed C4 foods (e.g., millet) and C4 food-fed animal protein sources, with smaller contributions from C3 food plants (e.g., rice) and C3 food-fed animal protein sources. We argued that the local dry-hot environment was the reason for the accessibility of C4 plants in the studied area. A comparison of our results with previously published archeobotanical data and isotopic evidence from human bones in other Bronze Age sites in Yunnan Province revealed high spatial variability in diets of human and subsistence strategies during this period. These differences were caused by the highly varying living environment of each region, which was related to fragmentation resulting from the geomorphological features of Yunnan Province.

Psoriasis is an immune-mediated chronic inflammatory skin disease primarily mediated by the activation of interleukin (IL)-17-producing T cells. Traditional Chinese Medicine (TCM) represents one of the most effective complementary and alternative medicine (CAM) agents for psoriasis, which provides treasured sources for the development of anti-psoriasis medications. Xiao-Yin-Fang (XYF) is an empirically developed TCM formula that has been used to treat psoriasis patients in Shanghai Changhai Hospital for over three decades. Imiquimod (IMQ)-induced psoriasis-like dermatitis mouse model was utilized to investigate the therapeutic effects of XYF by the assessment of disease severity and skin thickness. Flow cytometric assay was performed to explore the influence of XYF on skin-related immunocytes, primarily T cells. And, RNA sequencing analysis was employed to determine the alternation in gene expression upon XYF therapy. We discovered that XYF alleviated psoriasis-like skin inflammation mainly through suppressing dermal and draining lymph-node IL-17-producing γδT (γδT17) cell polarization. Moreover, XYF therapy ameliorated the relapse of psoriasis-like dermatitis and prohibited dermal γδT cell reactivation. Transcriptional analysis suggested that XYF might regulate various inflammatory signaling pathways and metabolic processes. In conclusion, our results clarified the therapeutic efficacy and inner mechanism of XYF therapy in psoriasis, which might promote its clinical application in psoriasis patients and facilitate the development of novel anti-psoriasis drugs based on the bioactive components of XYF.

An effective classifier combining convolutional neural network and regularized extreme learning machine (called as CNN-RELM) is presented in this paper. Firstly, CNN-RELM trains the convolutional neural network (CNN) using the gradient descent method until the learning target accuracy reaches. Then the fully connected layer of CNN is replaced by regularized extreme learning machine (RELM) optimized by genetic algorithm and the rest layers of the CNN remain unchanged. The experiments on different face databases are given to evaluate the performance of CNN-RELM. The experimental results show that CNN-RELM is a feasible classifier and it outperforms CNN and RELM. Due to the uniting of CNN and RELM, CNN-RELM have the advantages of CNN and RELM and it is easier to learn and faster in testing.

Building an artificial double helix is a compelling challenge, and most strategies rely on the intertwining of two helical strands. Here, in a very different approach, the authors construct a supramolecular double helix from multiple synthetic small molecules chained together by double crossed halogen bonds.

We experimentally demonstrate an integrated strictly non-blocking silicon 4 × 4 optical switch chip that can be operated in both thermo-optic (TO) and electro-optic (EO) switching modes. It is based on the double-layer network (DLN) architecture and consists of twelve 2 × 2 Mach-Zehnder interferometer (MZI) switch elements. TO phase shifters based on TiN microheaters and EO phase shifters based on p-i-n diodes are embedded in both waveguide arms of the MZI elements. The power consumption for TO and EO switching is 34 mW/π and 7 mW/π, respectively. The on-chip insertion losses are 1.74 ± 0.59 dB and 3.79 dB ± 1.32 dB for TO and EO switching, respectively. Due to the merits of the DLN architecture and the optimized performance of the switch elements, the chip possesses low crosstalk of -29.1 dB and -19.4 dB for TO and EO switching, respectively. Quadrature phase-shift keying (QPSK) optical signals with a data rate of 64 Gb/s are transmitted through the switch with no observable deteriorations. Such an optical switch is a promising candidate for both optical circuit switching and optical packet switching for a variety of applications.

Real-time and accurate vehicle tracking by Cameras and Surveillance can provide strong support for the acquisition and application of important traffic parameters, which is the basis of the traffic condition evaluation and the reasonable traffic command and dispatch. To deal with difficult problems of vehicle tracking research in a complex environments, such as occlusion, sudden illumination change, similar target interference and real-time tracking, measures are taken as follows. Firstly, the existing color local entropy particle filter tracking method is improved. The symmetry of information entropy is used to overcome the tracking failure caused by large-area occlusion. Secondly, the SIFT feature tracking method is improved to enhance real-time performance and robustness. Thirdly, two tracking methods were combined according to their characteristics, aiming at effectively improving the quasi-determination and real-time performance of vehicle tracking. Fourthly, Kalman filter was used to predict the motion state of vehicles. According to the SIFT characteristics and license plate information of vehicles, the exact position of the lost target vehicles is quickly located. It has been verified by experiments that our method has effectively improved the accuracy and real-time performance of vehicle tracking in complex situations.

BCMA-targeting chimeric antigen receptor (CAR)-T cell therapy has shown remarkable clinical efficacy against multiple myeloma, yet antigen escape and tumor relapse still occur after the use of these therapies. Designing CAR-T therapies that targets multiple antigens simultaneously seems a feasible way to avoid antigen escape, and it has been extensively studied elsewhere. Here, we report novel BCMA-OR-CD38 Tan CAR T cells that can trigger robust cytotoxicity against target cells expressing either BCMA or CD38. We demonstrate that, in in vitro studies, these BCMA-OR-CD38 Tan CAR T cells exhibit similar CAR expression, superior cytotoxicity and antigen-stimulated T cell proliferation as compared to single-targeted CAR T cells or CD38-OR-BCMA Tan CAR T cells. Importantly, these BCMA-OR-CD38 Tan CAR-T cells can achieve complete tumor clearance in myeloma-bearing mice with no relapse observed through the course of these experiments. Finally, this BCMA-OR-CD38 Tan CAR was fully compatible with existing clinical grade T cell manufacturing procedures and can be implemented using current clinical protocols. Taken together, our results present an effective solution to the challenge of antigen escape in BCMA CAR T-cell therapies.

Objective This study aims to identify effective gene networks and biomarkers to predict response and prognosis for HER2-negative breast cancer patients who received sequential taxane-anthracycline neoadjuvant chemotherapy. Materials and Methods Transcriptome data of training dataset including 310 HER2-negative breast cancer who received taxane-anthracycline treatment and an independent validation set with 198 samples were analyzed by weighted gene co-expression network analysis (WGCNA) approach in R language. Gene ontology (GO) terms and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways analysis were performed for the selected genes. Module-clinical trait relationships were analyzed to explore the genes and pathways that associated with clinicopathological parameters. Log-rank tests and COX regression were used to identify the prognosis-related genes. Results We found a significant correlation of an expression module with distant relapse–free survival (HR = 0.213, 95% CI [0.131–0.347], P = 4.80E−9). This blue module contained genes enriched in biological process of hormone levels regulation, reproductive system, response to estradiol, cell growth and mammary gland development as well as pathways including estrogen, apelin, cAMP, the PPAR signaling pathway and fatty acid metabolism. From this module, we further screened and validated six hub genes (CA12, FOXA1, MLPH, XBP1, GATA3 and MAGED2), the expression of which were significantly associated with both better chemotherapeutic response and favorable survival for BC patients. Conclusion We used WGCNA approach to reveal a gene network that regulate HER2-negative breast cancer treatment with taxane-anthracycline neoadjuvant chemotherapy, which enriched in pathways of estrogen signaling, apelin signaling, cAMP signaling, the PPAR signaling pathway and fatty acid metabolism. In addition, genes of CA12, FOXA1, MLPH, XBP1, GATA3 and MAGED2 might serve as novel biomarkers predicting chemotherapeutic response and prognosis for HER2-negative breast cancer.

The development of hair follicle in cashmere goats shows significant periodic change, as with mice and humans. However, for cashmere goat with double-coat, the periodic change may be due to other regulatory molecules and signal pathways. To understand the mechanism of periodic development of hair follicle, we performed a weighted gene coexpression network analysis (WGCNA) to mine key genes and establish an interaction network by utilizing the NCBI public dataset. Ten coexpression modules, including 7689 protein-coding genes, were constructed by WGCNA, six of which are considered to be significantly related to the development of the hair follicle cycle. A functional enrichment analysis for each model showed that they are closely related to ECM- receptor interaction, focal adhesion, PI3K-Akt signaling pathway, estrogen signaling pathway, and so on. Combined with the analysis of differential expressed genes, 12 hub genes from coexpression modules were selected as candidate markers, i.e., COL1A1, C1QTNF6, COL1A2, AQP3, KRTAP3-1, KRTAP11-1, FA2H, NDUFS5, DERL2, MRPL14, ANTKMT and XAB2, which might be applied to improve cashmere production.

Individual tree segmentation is essential for many applications in city management and urban ecology. Light Detection and Ranging (LiDAR) system acquires accurate point clouds in a fast and environmentally-friendly manner, which enables single tree detection. However, the large number of object categories and occlusion from nearby objects in complex environment pose great challenges in urban tree inventory, resulting in omission or commission errors. Therefore, this paper addresses these challenges and increases the accuracy of individual tree segmentation by proposing an automated method for instance recognition urban roadside trees. The proposed algorithm was implemented of unmanned aerial vehicles laser scanning (UAV-LS) data. First, an improved filtering algorithm was developed to identify ground and non-ground points. Second, we extracted tree-like objects via labeling on non-ground points using a deep learning model with a few smaller modifications. Unlike only concentrating on the global features in previous method, the proposed method revises a pointwise semantic learning network to capture both the global and local information at multiple scales, significantly avoiding the information loss in local neighborhoods and reducing useless convolutional computations. Afterwards, the semantic representation is fed into a graph-structured optimization model, which obtains globally optimal classification results by constructing a weighted indirect graph and solving the optimization problem with graph-cuts. The segmented tree points were extracted and consolidated through a series of operations, and they were finally recognized by combining graph embedding learning with a structure-aware loss function and a supervoxel-based normalized cut segmentation method. Experimental results on two public datasets demonstrated that our framework achieved better performance in terms of classification accuracy and recognition ratio of tree.

WRKY transcription factors play central roles in developmental processes and stress responses of wheat. Most WRKY proteins of the same group (Group III) have a similar function in abiotic stress responses in plants. TaWRKY46, a member of Group III, was up-regulated by PEG treatment. TaWRKY46-GFP fusion proteins localize to the nucleus in wheat mesophyll protoplasts. Overexpression of TaWRKY46 enhanced osmotic stress tolerance in transgenic Arabidopsis thaliana plants, which was mainly demonstrated by transgenic Arabidopsis plants forming higher germination rate and longer root length on 1/2 Murashige and Skoog (MS) medium containing mannitol. Furthermore, the expression of several stress-related genes (P5CS1, RD29B, DREB2A, ABF3, CBF2, and CBF3) was significantly increased in TaWRKY46-overexpressing transgenic Arabidopsis plants after mannitol treatment. Taken together, these findings proposed that TaWRKY46 possesses vital functions in improving drought tolerance through ABA-dependent and ABA-independent pathways when plants are exposed to adverse osmotic conditions. TaWRKY46 can be taken as a candidate gene for transgenic breeding against osmotic stress in wheat. It can further complement and improve the information of the WRKY family members of Group III.

A fully hydraulic variable valve system based on spark-ignition engine BJ486EQ is introduced in this article, which can provide fully variable valve lifts, valve timing, and valve opening duration. A simulation model of fully hydraulic variable valve system is established, which is verified through experimental intake valve lift and hydraulic pressure. It is found that pressure fluctuation in hydraulic system of fully hydraulic variable valve system becomes greater with the increase in engine speed, which even leads to “lift distortion.” The cam profile of fully hydraulic variable valve system and the structure of the throttling valve which is used to reduce valve seating speed are optimized by simulation. The lift distortion and rebound do not occur in the optimized fully hydraulic variable valve system, which is indicated by the results of simulation and experiments when the engine speed is 5000 r/min, and valve seating characteristics become stable and soft.

Ascorbate-glutathione (ASA-GSH) cycle is a major pathway of H2O2 scavenging and an effective mechanism of detoxification in plants. The differences in photosynthesis, chlorophyll content (Chl), relative water content (RWC), antioxidants and antioxidative enzyme activities involved in ASA-GSH metabolism were measured between the flag leaves and spike bracts (glumes and lemmas) during grain filling under drought stress. The expression of APX1, GRC1, DHAR, MDHAR, GPX1, and GS3 in ASA-GSH cycle was also measured. Compared with the flag leaves, the spike bracts exhibited stable net photosynthetic rate (PN) and chlorophyll content (Chl), a lower accumulation of reactive oxygen species (ROS), and more enhanced percentages of antioxidant enzyme activities and key enzymes gene transcription levels involved in ASA-GSH metabolism during the grain-filling stage under drought conditions. This could be the reasonable explanation for the more stable photosynthetic capacity in spikes, and the glumes and lemmas senesced later than the flag leaves at the late grain-filling stage. Also, the function of ASA-GSH cycle could not be ignored in alleviating oxidative damage by scavenging more excess ROS in spikes under drought stress.

Background/Aims: Although early studies show that Mdm2 is the primary E3 ubiquitin ligase for the p53 tumor suppressor, an increasing amount of data suggests that p53 ubiquitination and degradation are more complex than once thought. Here, we investigated the role of RNF125, a non-Mdm2 ubiquitin-protein ligase, in the regulation of p53. Methods and Results: RNF125 physically interacted with p53 in exogenous/endogenous co-immunoprecipitation (IP) and GST-pull down assay, and a C72/75A mutation of RNF125 did not interfere with this interaction. Expression of RNF125 decreased the level of p53 in a dose-dependent manner, whereas knockdown of RNF125 by RNA interference increased the level of p53. As shown by Western blotting and ubiquitin assay, RNF125 ubiquitinated p53 and targeted it for proteasome degradation. Furthermore, RNF125 repressed p53 functions including p53-dependent transactivation and growth inhibition. Conclusion: Our data suggest that RNF125 negatively regulates p53 function through physical interaction and ubiquitin-mediated proteasome degradation.

Background/Aims: Recent reports showed that proteasome subunit alpha type-7 (PSMA7) was overexpressed in colorectal cancer. To investigate the mechanism of PSMA7 in promotion of colorectal cancer, we screened for its interaction partners. Methods and Results: This study found that PSMA7 associated with nucleotide-binding oligomerization domain-containing protein 1 (NOD1) by yeast two-hybrid screening, co-immunoprecipitation (IP), and GST-pull down assay. As shown by Western blotting and ubiquitin assay, PSMA7 downregulated the expression of NOD1 in a proteasome-dependent manner. Overexpression of PSMA7 in HCT116 cells resulted in an inhibition of NOD1-mediated apoptosis and NF-κB activation, whereas knockdown of PSMA7 by RNA interference enhanced NOD1 activity. Conclusion: Our data suggest that PSMA7 is a negative regulator of the NOD1, and may promote tumor growth by its inhibitory role on NOD1.

Carcinoembryonic antigen (CEA) is a broad-spectrum tumor marker used in clinical applications. The primarily clinical method for measuring CEA is based on chemiluminescence in serum during enzyme-linked immunosorbent assays (ELISA) in 96-well plates. However, this multi-step process requires large and expensive instruments, and takes a long time. In this study, a high-throughput centrifugal microfluidic device was developed for detecting CEA in serum without the need for cumbersome washing steps normally used in immunoreactions. This centrifugal microdevice contains 14 identical pencil-like units, and the CEA molecules are separated from the bulk serum for subsequent immunofluorescence detection using density gradient centrifugation in each unit simultaneously. To determine the optimal conditions for CEA detection in serum, the effects of the density of the medium, rotation speed, and spin duration were investigated. The measured values from 34 clinical serum samples using this high-throughput centrifugal microfluidic device showed good agreement with the known values (average relative error = 9.22%). These results indicate that the high-throughput centrifugal microfluidic device could provide an alternative approach for replacing the classical method for CEA detection in clinical serum samples.

A novel way to measure humidity through testing the emissivity of an area radiant source is presented in this paper. The method can be applied in the environment at near room temperature (5~95 °C) across the relative humidity (RH) range of 20~90% RH. The source, with a grooved radiant surface, works in the far infrared wavelength band of 8~12 μm. The Monte-Carlo model for thermal radiation was set up to analyze the V-grooved radiant surface. Heat pipe technology is used to maintain an isothermal radiant surface. The fuzzy-PID control method was adopted to solve the problems of intense heat inertia and being easily interfered by the environment. This enabled the system to be used robustly across a large temperature range with high precision. The experimental results tested with a scanning radiant thermometer showed that the radiant source can provide a uniform thermal radiation capable of satisfying the requirements of humidity testing. The calibration method for the radiant source for humidity was explored, which is available for testing humidity.

As one of the most important applications in peer-to-peer (P2P) networks, the video-on-demand (VoD) system freely supports video cassette recorder (VCR) operation for users. However, the users may experience significant playback delay after frequent VCR operations in the VoD system, which will affect the quality of experience (QoE) of the users. Hence, selecting an appropriate data-prefetching strategy to support better VCR operation is an important approach to improve the QoE. This paper proposes a data-prefetching strategy (DSA) to determine the most suitable anchor interval by considering the playback delay and positioning satisfaction. According to the DSA, we use the multiple-attribute decision-making (MADM) theory to model the selection of intervals of prefetching data blocks (i.e., anchor interval) and the technique for ordering preference by similarity to an ideal solution (TOPSIS) algorithm to solve the MADM. The simulation results show that the DSA strategy obtains higher positioning satisfaction than the existing schemes, which is approximately 60% higher than the anchor points, popular parts of video, and user interests (API)-based method. Moreover, with the increase in network bandwidth, the DSA strategy can minimize the playback delay after VCR operation using relative few extra bandwidths.
