Abstract Background CD56 has been observed in malignant tumours exhibiting neuronal or neuroendocrine differentiation, such as breast cancer, small-cell lung cancer, and neuroblastoma. Abnormal glycosylation modifications are thought to play a role in regulating tumour cell proliferation, migration, and invasion. Nevertheless, the exact roles and molecular mechanisms of CD56 and polysialylated CD56 (PSA-CD56) in the development and progression of clear cell renal cell carcinoma (ccRCC) remain elusive. Here we unveil the biological significance of CD56 and PSA-CD56 in ccRCC. Methods In this study, we employed various techniques, including immunohistochemistry (IHC), RT-qPCR, and western blot, to examine the mRNA and protein expression levels in both human ccRCC tissue and cell lines. Lentivirus infection and CRISPR/Cas9 system were utilized to generate overexpression and knockout cell lines of CD56. Additionally, we conducted several functional assays, such as CCK-8, colony formation, cell scratch, and transwell assays to evaluate cell growth, proliferation, migration, and invasion. Furthermore, we established a xenograft tumor model to investigate the role of CD56 in ccRCC in vivo. To gain further insights into the molecular mechanisms associated with CD56, we employed the Hedgehog inhibitor JK184 and the β-catenin inhibitor Prodigiosin. Results CD56 was significantly overexpressed in both human ccRCC tissues and renal cancer cell lines compared to adjacent normal tissues and normal renal epithelial cells. In vitro and in vivo experiments revealed that the knockout of CD56 inhibited the proliferation, migration, and invasion capabilities of ccRCC cells, whereas the overexpression of PSA-CD56 promoted these capacities. Finally, PSA-CD56 overexpression was found to activate both the Hedgehog and Wnt/β-catenin signaling pathways. Conclusion Our findings demonstrate that the oncogenic function of CD56 polysialylation plays a vital role in the tumorigenesis and progression of ccRCC, implying that targeting PSA-CD56 might be a feasible treatment target for ccRCC.

Plot text is very valuable supporting information in movie recommendations. It has several characteristics: 1) It is rich in content. Each movie often has a document of more than 200 words to describe it, which can give the movie a rich semantic meaning. 2) Objectivity. Plot texts are different from review information. A movie may have thousands of reviews with mixed and conflicting opinions. However, a film has only one plot text, which is fair in tone and does not take a position. Despite its appealing properties and potential for accurate movie portrayal, the lack of a building block for effectively mining plot semantics has led to the marginalization of plot text in the design of movie recommendation algorithms. Therefore, in this paper, we explore the application of the Transformer, currently the best natural language processing module, to learning movie plot texts to help achieve more accurate rating prediction. We propose the "Plot-Aware Transformer" model (PAT) to model the process of "user-movie" rating interaction. We test the PAT model on several movie datasets and demonstrated that the model is competitive. In all tasks, PAT achieves state-of-the-art performance compared to baseline experiments.

Primary cardiac myxofibrosarcoma is a rare form of cardiac malignant tumors. MFS usually involves the left atrium and presents as a unicentric or multicentric tumor mass. We reported on a 37-year-old female who presented with chest tightness and dyspnea for a month, dry cough, and occasionally having blood streak sputum for half a month. Echocardiography, cardiac computed tomography and cardiac positron emission tomography revealed multiple tumors in the heart. The right ventricle and right pulmonary artery were involved, with occlusion of the right pulmonary artery. Cardiac tumors were surgically resected and were consistent with low-grade MFS. No recurrence or metastasis occurred at 20 months of follow-up.

In this paper, we use a graph capsule network to capture the spatial dependence of air quality data and meteorological data among cities, then use an LSTM network to model the temporal dependence of air pollution levels in specific cities and finally implement PM2.5 concentration prediction. We propose a graph-capsule-LSTM model based on a graph-capsule network and an LSTM network. The model uses a graph capsule network to model the neighboring feature information of the target city and then combines the local data of the target city to form the final feature vector. The feature mapping on the time axis is then used to obtain the temporal feature sequences of the target nodes, which are fed into the LSTM network for learning and prediction. Experiments show that the method achieves better results than the latest baseline model in the PM2.5 prediction task. While demonstrating that the capsule network outperforms the convolutional network, it also shows that this capsule network is very competent for the task of PM2.5 prediction.

Context Puerarin (Pue) and tanshinone IIA (Tan IIA) are often used in combination in the treatment of cerebrovascular diseases.Objective To investigate the neuroprotective effect and synergic mechanism of Pue-Tan IIA on the treatment of ischaemic stroke (IS).Materials and methods IS was induced in rats by middle cerebral artery occlusion (MCAO). Rats were intraperitoneally injected with Pue (36 mg/kg), Tan IIA (7.2 mg/kg), or Pue-Tan IIA (36 and 7.2 mg/kg) for five times [30 min before ischaemia, immediately after reperfusion (0 h), 24, 48, and 72 h after reperfusion]. After administration, neurological function assessment and histological changes in the brain were performed. S-100β and NSE levels were measured to determine the severity of brain injury. Oxidative stress parameters and inflammatory mediators were measured. Abstract Background Hand knob stroke is a rare clinical disorder frequently misdiagnosed as peripheral neuropathy. The purpose of this study is to recognize this particular type of stroke by analyzing clinical features, etiology, and prognosis. Methods We enrolled 19 patients with acute hand knob stroke in the Department of Neurology of the Beijing Geriatric Hospital from January 2018 to January 2022, and the clinical and imaging data of the patients during hospitalization and follow-up were collected and summarized. Results Acute hand knob stroke accounted for 0.9% of all acute stroke, and ischemic stroke (17 cases, 89.5%) was more than hemorrhagic stroke (2 cases, 10.5%). All patients presented sudden contralateral hand paresis, 12 (63.2%) of them had only isolated hand paralysis, and the location of the lesion corresponded to different finger weakness. The cause of hand knob hemorrhage was hypertension, while the causes of hand knob infarction were mainly small-vessel occlusion (SVO) (35.3%) and large-artery atherosclerosis (LAA) (35.3%), and the rare causes include carotid artery dissection and carotid body tumor. After a median follow-up 13.5 months, the prognosis of 94.7% patients was good, and one patient (5.3%) had recurrent stroke. Conclusions Hand knob stroke is a rare stroke with a good prognosis and a low stroke recurrence rate. Ischemic stroke is the predominant type and the main clinical manifestation is hand paresis. The cause of hand knob hemorrhage is hypertensive, while SVO and LAA are the main causes of hand knob infarction, but there are some rare etiologies.

Photodetectors based on anisotropic materials are attracting lots of attention. This research systematically investigates the anisotropic photoresponse of photodetectors based on the two-dimensional WSe2/MoSe2 lateral heterostructure through quantum transport simulations. The transport direction is crucial for photocurrent. In the armchair direction, the photocurrent is sensitive to ultraviolet light, while in the zigzag direction, cyan light is significant. The photocurrent magnitude in the zigzag direction is 79 times larger than that in the armchair direction. The extinction ratio (ER) also exhibits strong anisotropy between the two directions. Infrared and visible light have more influence on the ER in the armchair direction, and ultraviolet light is crucial in the zigzag direction. The research provides a promising candidate for directional sensitivity photodetectors.

Abstract Background The role of the dysfunction of left atrium in the occurrence and development of cardiovascular disease has been gradually recognized. We aim to compare the impact on left atrial (LA) function between patients with hypertrophic cardiomyopathy (HCM) and hypertension (HTN) without LA enlargement using cardiovascular magnetic resonance feature tracking (CMR-FT), and if possible, explore the capability of LA function for providing clinical implication and predicting clinical adverse events in the early stage of cardiovascular disease. Methods Consecutive 60 HCM patients and 60 HTN patients with normal LA size among 1413 patients who underwent CMR were retrospectively analyzed as well as 60 controls. Left atrial and ventricular functions were quantified by volumetric and CMR-FT derived strain analysis from long and short left ventricular view cines. The primary endpoint was a composite of all-cause death, stroke, new-onset or worsening heart failure to hospitalization, and paroxysmal or persistent atrial fibrillation. Results Compared to the controls, both HTN and HCM participants had impaired LA reservoir function (εs) and conduit function (εe) with the different stage of LA booster pump dysfunction (εa). LA strain was more sensitive than LV longitudinal strain (GLS) for evaluate primary endpoint (εs: 33.9% ± 7.5 vs. 41.2% ± 14.3, p = 0.02; εe: 13.6% ± 6.2 vs. 17.4% ± 10.4, p = 0.03; εa: 20.2% ± 6.0 vs. 23.7% ± 8.8, p = 0.07; GLS: -19.4% ± 6.4 vs. -20.0% ± 6.8, p = 0.70, respectively). Summary: Background: The pathophysiology and subsequent myocardial dysfunction of heart failure with preserved ejection fraction (HFpEF) with comorbid obesity has not been extensively described. This study aimed to investigate the clinical features and cardiovascular magnetic resonance (CMR) derived myocardial strain and tissue characteristics in patients with HFpEF and comorbid obesity phenotype. Methods: In this prospective cohort study, we included consecutive patients admitted to Fuwai hospital in China who underwent CMR. Patients with HFpEF or obesity were diagnosed with demographic data, clinical presentation, laboratory test, and echocardiography or CMR imaging. The key exclusion criteria were cardiomyopathy, primary valvular heart disease, and significant coronary artery disease. Participant data were obtained from the electronic medical records database or inquiry. Comparisons of clinical features and CMR derived structural and functional parameters amongst different groups were made using one-way analysis of variance, or χ2 tests, and post hoc Bonferroni analysis where appropriate. Findings: Between January 1, 2019 and July 31, 2021, 280 participants (108 patients with HFpEF and obesity, 50 patients with HFpEF and normal weight, 72 patients with obesity, and 50 healthy controls) were enrolled. Compared with patients with HFpEF and normal weight, patients with HFpEF and obesity were younger males, and had higher plasma volume, uric acid and hemoglobin levels, yet less often atrial fibrillation, and lower NT-proBNP levels, and had higher left ventricular mass index, end-diastole/systole volume index, lower left atrial volume index, and worse myocardial strains (all p ≤ 0.05), but no remarkable difference in late gadolinium enhancement (LGE) presence and extracellular volume fraction (ECV). After adjusting for age, atrial fibrillation, and coronary artery disease, only global longitudinal strain (GLS, p = 0.031) and early-diastolic global longitudinal strain rate (eGLSR, p = 0.043) were considerably worse in patients with HFpEF and obesity versus patients with HFpEF and normal weight. Furthermore, early-diastolic strain rates showed no linear association with ECV in patients with HFpEF and obesity. Moreover, GLS demonstrated the highest diagnostic ability when compared with traditional CMR structural parameters and ECV to diagnose patients with HFpEF and obesity in the setting of obesity. Interpretation: Higher systemic inflammation, and worse GLS and eGLSR may be the distinct features of obesity-related HFpEF phenotype; strains and ECV may represent different mechanisms of HFpEF with obesity, deserving further study. Litter decomposition is an important source of carbon accumulation in the permafrost peatlands. Climate warming has led to shrub expansions and accelerated litter mixing with soils and fluctuations in the water table. However, little is known about how changes in the position of the litter will affect litter decomposition under climate warming. To reveal the mechanisms of response of the location of litter in the soil and climate warming to litter decomposition in permafrost peatlands. Here, we selected the evergreen shrub, Chamaedaphne calyculata, and the deciduous shrub, Vaccinium uliginosum, from the permafrost peatlands of the Greater Hing’an Mountains, China. The leaf litter was placed on the soil surface (no-mixing) and mixed with the soil (soil-litter mixing), and then it was incubated for 124 days at 15°C (control) and 20°C (warming). Our results showed that warming significantly increased the CO2 emission rates of C. calyculata and V. uliginosum by 19.9 and 17.4%, respectively. When compared to no-mixing, the CO2 emission rates were reduced (not significantly) by 1.5 (C. calyculata) and increased 13.6% (V. uliginosum) with soil-litter mixing. Interestingly, soil-litter mixing suppressed the positive effect of warming on the CO2 emission rates relative to no-mixing, and the suppressing effects in the V. uliginosum subplot were stronger than those in the C. calyculata subplot. Specifically, warming significantly increased the CO2 emissions of C. calyculata by 27.4% under no-mixing but the increase decreased to 13.1% under soil-litter mixing. Similarly, warming induced significant increases in the CO2 emissions of V. uliginosum, with an increase of 38.8% under no-mixing but non-significant increases (1.9%) were observed under soil-litter mixing. The combination of the enzyme activities of β-1,4-glucosidase, β-1,4-xylosidase and β-D-1,4-cellobiosidase and laccase and phenolics explained more than 60.0% of the variability in the CO2 emissions of C. calyculata and V. uliginosum, respectively. Our study highlights the importance of litter positions in mediating the responses of litter decomposition to climate warming and shrub expansions in the northern peatlands.

The alterations of plant composition and diversity pose a threat to the stability of the carbon pool in boreal peatland under climate change. We collected the samples of three plant functional types (deciduous shrubs, evergreen shrubs, and sedge) in seven permafrost peatlands of the Great Hing’an Mountains, China, and measured the properties of total carbon (TC), nitrogen (TN), and phosphorus (TP), their stoichiometric ratios (C:N, C:P, and N:P), and the stable isotope values (δ13C and δ15N) of six tissues (ranging from leaves to roots). For TC, TN, and TP, the contents had an average of 470.69 ± 1.56, 8.03 ± 0.23, and 1.71 ± 0.61 mg·g−1, respectively. TC contents of sedge were lower than those of shrubs for the whole plant. The allocations of N and P to shrub leaves were higher than to stems and roots. There was a similar trend of TN and TP contents, and stoichiometric ratios from leaves to roots between deciduous shrubs and evergreen shrubs. Shrubs and sedge have similar C: N in leaves and fine roots, while leaves of sedge C:P and N:P ratios were higher than shrubs, mainly showed that sedge is N and P co-limitation and shrubs are N limitation. The values of δ13C and δ15N were significantly higher in leaves and roots of sedge than those of shrubs, which means shrubs have higher nutrient acquisition strategies. These results support the shrubs are expanding in the boreal peatland under climate warming through nutrient competition. TC contents of all deciduous shrubs and sedge tissues were positively linear correlated to MAT and the values of δ13C and δ15N in sedge had significant relationships with MAT and MAP. Our results imply warming can increase plant photosynthesis in boreal peatland, and sedge was more sensitive to climate change. These findings would be helpful to understanding the responses of different plant tissues to climate changes in permafrost peatland.

Abstract Background As the paucity of data focusing on evaluating cardiac structure and function in patients with or without gene mutation, this study was sought to investigate the correlation between genotype and cardiac magnetic resonance (CMR) phenotype in patients with left ventricular non-compaction cardiomyopathy (LVNC) and to explore prognostic relevance in this cohort if possible. Hydrogen production using photocatalyst for water splitting is considered to be one of the most promising methods to solve the energy challenge. The feasibility of CdS/SiI2 heterostructure as a photocatalyst is systematically investigated in geometrical, electronic, optical and transport properties based on the density functional theory (DFT) calculations. The calculated results demonstrate different configurations of CdS/SiI2 heterostructure have a typical type-II direct bandgap with bandgap values ranging from 2.08 to 2.21 eV, which are closer to the ideal bandgap for water-splitting. Not only the band alignment of type-II heterostructures, but also the built-in electric field generated between the two monolayers can effectively drive carriers spatial separation, thus reducing the carrier recombination rate. At the same time, the light absorption of the heterostructures has been substantially enhanced. The biaxial strain can modulate the bandgap values and the band alignment to match different application requirements, and it can also modulate the light absorption intensity and range. At last, the calculated carrier mobility of the heterostructure is up to 103 cm2 V−1 s−1. Such ultrahigh carrier mobility implies a further reduction in the carrier recombination rate and an excellent performance in photocatalyst. To sum up, these calculated results all indicate that the CdS/SiI2 heterostructure with low carrier recombination and ideal bandgap is a particularly promising photocatalyst.

Ponds are abundant in the boreal peatland landscape, which are potential hotspots for greenhouse gas (GHG) emissions. However, compared to large lakes, ponds are difficult to identify by satellite, and they have not been adequately studied. Here, we observed methane (CH4), carbon dioxide (CO2), and nitrous oxide (N2O) fluxes in the growing season at three sites along the water table gradient from the pelagic zone, littoral zone and bog across a shallow pond in a boreal peatland landscape in Northeastern China. The results showed that the littoral zone, dominated by herb Carex, was the hotspot for CH4 emissions. CH4 fluxes in littoral zone averaged 78.98 ± 19.00 mg m−2 h−1. The adjacent bog was a weak source of CH4 emissions, with an average flux of 0.07 ± 0.05 mg m−2 h−1. Within the pond, CH4 was mainly emitted through ebullition, accounting for 88.56% of the total CH4 fluxes, and the ebullition fluxes were negatively correlated with dissolved oxygen (DO). CO2 fluxes were highest in the pelagic zone, with an average of 419.76 ± 47.25 mg m−2 h−1. Wind and strong sediment respiration were key factors that led to the high fluxes. The observed three sites were all atmospheric N2O sinks ranging from −0.92 to −10.90 μg m−2 h−1. This study highlights the spatial variation in greenhouse gas fluxes from the pond and its adjacent bog, ignoring the ecotone area may underestimate CH4 fluxes. Although ponds are a hotspot for CH4 and CO2 emissions, they can also be a sink for N2O, which provides a reference for the quantification of global pond GHG fluxes. Therefore, finer-scale in situ observations are necessary to better understand the feedback of permafrost peatland ponds to global warming.

The geometrical, electronic, and optical properties of the PtS2/MoTe2 heterostructure are systematically investigated based on the density functional theory (DFT) calculations. The indirect bandgap (0.93 eV) PtS2/MoTe2 heterostructure shows high dynamical and mechanical stability at room temperature. Type-II band alignment of the system facilitates the separation of photogenerated electrons and holes at the interface. Biaxial strain and external electric field (Efield) can not only induce the band alignment of the PtS2/MoTe2 heterostructure transition from type-II to type-I, but also can realize semiconductor–metal transition. The PtS2/MoTe2 heterostructure can occur redshift with a peak appearing near infrared region and its power conversion efficiency (PCE) reaches 13.60%. These properties are beneficial for application in infrared detector and solar cells. In addition, biaxial strain enhances the light adsorption intensity of the PtS2/MoTe2 heterostructure and tensile strain expands the light adsorption range. The adsorption intensity increases with the Efield increasing, implying the Efield can effectively modulate the optical properties. Our results indicate that the PtS2/MoTe2 heterostructure has a bright prospect in the application of solar cell, infrared detector, optoelectronic and nanoelectronic devices, as well as provide a useful theoretical direction for the fabrication of PtS2/MoTe2 heterostructure.

Background. The most common site of prostate cancer metastasis is bone tissue with many recent studies having conducted genomic and clinical research regarding bone metastatic prostate cancer. However, further work is needed to better define those patients that are at an elevated risk of such metastasis. Methods. SEER and TCGA databases were searched to develop a nomogram for predicting prostate cancer bone metastasis. Results. Herein, we leveraged the Surveillance, Epidemiology, and End Results (SEER) database to construct a predictive nomogram capable of readily and accurately predicted the odds of bone metastasis in prostate cancer patients. This nomogram was utilized to assign patients with prostate cancer included in The Cancer Genome Atlas (TCGA) to cohorts at a high or low risk of bone metastasis (HRBM and LRBM, respectively). Comparisons of these LRBM and HRBM cohorts revealed marked differences in mutational landscapes between these patient cohorts, with increased frequencies of gene fusions, somatic copy number variations (CNVs), and single nucleotide variations (SNVs), particularly in the P53 gene, being evident in the HRBM cohort. We additionally identified lncRNAs, miRNAs, and mRNAs that were differentially expressed between these two patient cohorts and used them to construct a competing endogenous RNA (ceRNA) network. Moreover, three weighted gene co-expression network analysis (WGCNA) modules were constructed from the results of these analyses, with KIF14, MYH7, and COL10A1 being identified as hub genes within these modules. We further found immune response activity levels in the HRBM cohort to be elevated relative to that in the LRBM cohort, with single sample gene enrichment analysis (ssGSEA) scores for the immune checkpoint signature being increased in HRBM patient samples relative to those from LRBM patients. Conclusion. We successfully developed a nomogram capable of readily detecting patients with prostate cancer at an elevated risk of bone metastasis.

Background: Cardiac magnetic resonance (CMR) has been shown to improve the diagnosis of myocarditis, but no systematic comparison of this technique is currently available. The purpose of this study was to compare the 2009 and 2018 Lake Louise Criteria (LLC) for the diagnosis of acute myocarditis using 3.0 T MRI with endomyocardial biopsy (EMB) as a reference and to provide the cutoff values for multiparametric CMR techniques.Methods: A total of 73 patients (32 ± 14 years, 71.2% men) with clinically suspected myocarditis undergoing EMB and CMR with 3.0 T were enrolled in the study. Patients were divided into two groups according to EMB results (EMB-positive and -negative groups). The CMR protocol consisted of cine-SSFP, T2 STIR, T2 mapping, early and late gadolinium enhancement (EGE, LGE), and pre- and post-contrast T1 mapping. Their potential diagnostic ability was assessed with receiver operating characteristic curves.Results: The myocardial T1 and T2 relaxation times were significantly higher in the EMB-positive group than in the EMB-negative group. Optimal cutoff values were 1,228 ms for T1 relaxation times and 58.5 ms for T2 relaxation times with sensitivities of 86.0 and 83.7% and specificities of 93.3 and 93.3%, respectively. The 2018 LLC had a better diagnostic performance than the 2009 LLC in terms of sensitivity, specificity, positive predictive value, and negative predictive value. T1 mapping + T2 mapping had the largest area under the curve (0.95) compared to other single or combined parameters (2018 LLC: 0.91; 2009 LLC: 0.76; T2 ratio: 0.71; EGEr: 0.67; LGE: 0.73; ). The diagnostic accuracy for the 2018 LLC was the highest (91.8%), followed by T1 mapping (89.0%) and T2 mapping (87.7%).Conclusion: Emerging technologies such as T1/ T2 mapping have significantly improved the diagnostic performance of CMR for the diagnosis of acute myocarditis. The 2018 LLC provided the overall best diagnostic performance in acute myocarditis compared to other single standard CMR parameters or combined parameters. There was no significant gain when 2018LLC is combined with the EGE sequence.

Wetland restoration significantly affects ecosystem carbon and nitrogen cycles by changing soil physicochemical properties and plant properties. To reveal the effect of wetland restoration on soil organic carbon (SOC) and total nitrogen (TN) storage, we measured the SOC and TN content and calculated their storage in a restoration chronosequence (1, 4, 8, 13, 17, 27 years) and compared them with those in a soybean field and a natural marsh, all in Sanjiang Plain, northeast China. Our results showed that the SOC and TN content and storage in the investigated sites increased with restoration time. However, the restoration rate was faster in the first 8 years but slowed down after 8 years of restoration. The SOC and TN content and storage decreased with soil depths in the eight sites. After 27 years of restoration, the carbon and nitrogen storage in the restored site was 2.8 and 1.9 times that in the soybean field, respectively. However, the SOC and TN storage in the 27-year restored site was 45.3% and 35.1% lower than that in the natural marsh, respectively. This study assessed the responses of soil microbial properties to natural restoration and investigated the relationships between soil microbial properties and soil physico-chemical and plant properties. We selected a restoration chronosequence (1, 4, 8, 13, 17, 27 years) after farmland abandonment, a soybean field, and a natural marsh in Sanjiang Plain, northeast China. For each site, we analyzed the soil microbial biomass carbon and nitrogen (MBC and MBN), four enzymes (β-glucosidase, invertase, catalase, urease) activities, soil physico-chemical properties at 0–50 cm depths, and plant properties (biomass, height, and coverage). The MBC and MBN contents increased with restoration time, but MBN content slowed down after 8 years of restoration. After 27 years of restoration, the soil MBC and MBN contents were 15.7 and 3.2 times of those in the soybean field, but the largest contents of MBC and MBN in the restored sites were 7.78%, 27.76% lower than those in natural marshes, respectively. Moreover, soil enzyme activities and the geometric mean of enzymatic activities (GME) also increased with restoration but slowed down after 13 years of restoration. After 27 years of restoration, the GME was 2.9 times than that in the soybean field, but the largest GME in the restored sites was 31.15% lower than that in the natural marsh. MBC and MBN contents, soil enzyme activities, and GME had significant relationships with soil C:N ratio, organic carbon, nutrients (total nitrogen, available nitrogen, total phosphorus), bulk density, moisture content, pH, plant properties, (i.e. biomass, height, and coverage) (p < 0.01). Redundancy analysis revealed that soil C:N ratio, pH, moisture content, total nitrogen and phosphorus were main factors affecting MBC and MBN contents and enzyme activities. In conclusion, soil microbial properties can respond positively to the natural restoration process of the reclaimed marshes and were significantly correlated with specific parameters of soil physico-chemical and plant properties.

Renal cell carcinoma (RCC) is one of the most prevalent malignant tumors of the urinary system, accounting for around 2% of all cancer diagnoses and deaths worldwide. Clear cell RCC (ccRCC) is the most prevalent and aggressive histology with an unfavorable prognosis and inadequate treatment. Patients’ progression-free survival is considerably improved by surgery; however, 30% of patients develop metastases following surgery. Identifying novel targets and molecular markers for RCC prognostic detection is crucial for more accurate clinical diagnosis and therapy. Glycosylation is a critical post-translational modification (PMT) for cancer cell growth, migration, and invasion, involving the transfer of glycosyl moieties to specific amino acid residues in proteins to form glycosidic bonds through the activity of glycosyltransferases. Most cancers, including RCC, undergo glycosylation changes such as branching, sialylation, and fucosylation. In this review, we discuss the latest findings on the significance of aberrant glycans in the initiation, development, and progression of RCC. The potential biomarkers of altered glycans for the diagnosis and their implications in RCC have been further highlighted.

Permafrost peatland is a unique ecosystem that represents a huge carbon terrestrial pool. Soil quality has a relatively high level of variation at small scale in this ecosystem and is closely related to the carbon cycle. To quantify this variability, we analyzed total organic carbon, total nitrogen, total phosphorus, nutrient, and element (sodium, magnesium, potassium, and calcium), and microbial respiration activities (microbial biomass carbon, soil basal respiration, metabolic, and microbial quotients) in the humus layer to deeper soil layer of 6 plots at small scale (meters). For all samples, the coefficients of variation (CV) values of total carbon concentrations were lowest and these values of sodium, magnesium, potassium, and calcium concentrations were higher than those of total carbon, nitrogen, and phosphorus concentrations. The largest variations of total phosphorus, ash, and Na were in the 10–20 cm layer with soil depths. The litter decomposition and water table may cause this variation at small-scale. The CV values of microbial respiration activities were largest compared with soil properties. There were different correlations between basal respiration and soil properties among the plots. Our results showed that soil properties and microbial respiration activities in permafrost peatland exhibits considerable variability at small scale. This variability indicates that sampling location and number are very important in peatland studies if we want to accurately estimate the biogeochemistry in a peatland.

Renal cell carcinoma is a highly heterogeneous cancer group, and the complex microenvironment of the tumor provides appropriate immune evasion opportunities. The molecular mechanism of immune escape in renal cell carcinoma is currently a hot issue, focusing primarily on the major complex of histocompatibility, immunosuppressive cells, their secreted immunosuppressive cytokines, and apoptosis molecule signal transduction. Immunotherapy is the best treatment option for patients with metastatic or advanced renal cell carcinoma and combination immunotherapy based on a variety of principles has shown promising prospects. Comprehensive and in-depth knowledge of the molecular mechanism of immune escape in renal cell carcinoma is of vital importance for the clinical implementation of effective therapies. The goal of this review is to address research into the mechanisms of immune escape in renal cell carcinoma and the use of the latest immunotherapy. In addition, we are all looking forward to the latest frontiers of experimental combination immunotherapy.

Deep learning has become a recent, modern technique for big data processing, with promising results and large potential. For recommender systems, user and item information can be used as input vectors to perform prediction tasks. However, augmenting the number of layers to improve feature extraction will increase the computational complexity considerably and may not achieve the desired results. This paper proposes a method called attention convolution collaborative filtering (Att-ConvCF), which integrates an attention mechanism with a collaborative filtering model to improve the effectiveness of the feature extraction by reassigning the weights of feature vectors. Descriptive documents for the items are used to enrich the background information through a convolutional neural network. Finally, extensive experiments with real-world datasets were performed, and the results showed that Att-ConvCF could effectively extract the feature values of the data and significantly outperform the existing recommendation models.

ST3Gal IV is one of the principal sialyltransferases responsible for the biosynthesis of α2, 3-sialic acid to the termini N-glycans or O-glycans of glycoproteins and glycolipids. It has been reported that ST3Gal IV expression is associated with gastric carcinoma, pancreatic adenocarcinoma and breast cancer. While the expression and functions of ST3Gal IV in cervical cancer are still poorly understood. In this study, we found that ST3Gal IV was downregulated in human cervical cancer tissues compared to normal cervix tissues, and ST3Gal IV expression was negatively associated with the pathological grade of cervical cancer. ST3Gal IV upregulation inhibited the growth and proliferation of cervical cancer HeLa and SiHa cells in vitro and in vivo. Furthermore, ST3Gal IV overexpression enhanced the expression of several Notch pathway components such as Jagged1, Notch1, Hes1 and Hey1, while cell cycle protein expression like Cyclin D1, Cyclin E1, CDK2 and CDK4 were decreased. These results indicate that expression of ST3Gal IV is reduced in cervical cancer and plays a negative role in cell proliferation via Notch/p21/CDKs signaling pathway. Thus, sialyltransferase ST3Gal IV might be a target for the diagnosis and therapy of cervical cancer.

All current recommendation algorithms, when modeling user–item interactions, basically use dot product. This dot product calculation is derived from matrix factorization. We argue that an inherent drawback of matrix factorization is that latent semantic vectors of users or items sometimes do not satisfy triangular inequalities, which may affect the performance of the recommendation. Recently, metric factorization was proposed to replace matrix factorization and has achieved some improvements in terms of recommendation accuracy. However, similar to matrix factorization, metric factorization still uses a simple, linear fashion. In this paper, we explore leveraging nonlinear deep neural networks to realize Euclidean distance interaction between users and items. We propose a generic Neural Metric Factorization Framework (NMetricF), which learns representations for users and items by incorporating Euclidean metric factorization into deep neural networks. Extensive experiments on six real-world datasets show that, compared to the previous recommendation algorithms based purely on rating data, NMetricF achieves the best performance.

Bladder cancer is one of the most common malignant tumors of the urogenital system with high morbidity and mortality worldwide. Early diagnosis and personalized treatment are the keys to successful bladder cancer treatment. Due to high postoperative recurrence rates and poor prognosis, it is urgent to find suitable therapeutic targets and biomarkers. Glycans are one of the four biological macromolecules in the cells of an organism, along with proteins, nucleic acids, and lipids. Glycans play important roles in nascent peptide chain folding, protein processing, and translation, cell-to-cell adhesion, receptor-ligand recognition, and binding and cell signaling. Glycans are mainly divided into N-glycans, O-glycans, proteoglycans, and glycosphingolipids. The focus of this review is the discussion of glycans related to bladder cancer. Additionally, this review also addresses the clinical value of glycans in the diagnosis and treatment of bladder cancer. Abnormal glycans are likely to be potential biomarkers for bladder cancer.

Rosa roxburghii tratt (RRT), widely distributed in the southwest of China, is favored by consumers for its good taste and healthy functions. In this study, thirty-seven compounds of Rosa roxburghii Tratt (RRT) were identified and quantified by gas chromatography–olfactometry (G–O) and gas chromatography–mass spectrometry (GC–MS) analysis. Furthermore, ethyl 2-methylpropanoate, ethyl butanoate, ethyl 2-methylbutyrate, and ethyl hexanoate were present with much higher odor activity values (OAVs) than other compounds. The key notes were confirmed by omission tests. Possible interaction among key notes was investigated through odor intensity determination and sensory analysis. It showed fruity and woody notes had synergistic effects. Full factorial design was used to evaluate the notes contribution to the whole odor. One important finding is the major effect of order interactions, fruity note (X1) and woody note (X4) especially, emphasizing the existence of complex interactions occurring between odor notes. The interaction X1X4 was further investigated. The woody note has a positive effect when the fruity note is also in the mixture but tends to show a negative effect otherwise.

In patients undergoing coronary artery bypass grafting (CABG), ischemia/reperfusion injury (IRI) is the main contributor to organ dysfunction. Aging-induced vascular damage may be further aggravated during CABG. Favorable effects of conditioned medium (CM) from mesenchymal stem cells (MSCs) have been suggested against IRI. We hypothesized that adding CM to saline protects vascular grafts from IRI in rats. We found that CM contains 28 factors involved in apoptosis, inflammation, and oxidative stress. Thoracic aortic rings from 15-month-old rats were explanted and immediately mounted in organ bath chambers (aged group) or underwent 24 h of cold ischemic preservation in saline-supplemented either with vehicle (aged-IR group) or CM (aged-IR+CM group), prior to mounting. Three-month-old rats were used as referent young animals. Aging was associated with an increase in intima-to-media thickness, an increase in collagen content, higher caspase-12 mRNA levels, and immunoreactivity compared to young rats. Impaired endothelium-dependent vasorelaxation to acetylcholine in the aged-IR group compared to the aged-aorta was improved by CM (aged 61 ± 2% vs. aged-IR 38 ± 2% vs. aged-IR+CM 50 ± 3%, p < 0.05). In the aged-IR group, the already high mRNA levels of caspase-12 were decreased by CM. CM alleviates endothelial dysfunction following IRI in 15-month-old rats. The protective effect may be related to the inhibition of caspase-12 expression.

The ventilation/perfusion lung scan is recommended to exclude chronic thromboembolic pulmonary hypertension in the diagnostic algorithm of pulmonary hypertension, but its role in pulmonary arterial hypertension (PAH) has not been well explored. We characterized the lung perfusion pattern assessed by lung perfusion scintigraphy in idiopathic PAH (IPAH) patients and evaluate the potential prognostic significance of the patchy pattern perfusion defect. A total of 318 patients with IPAH confirmed by right heart catheterization who performed lung perfusion scintigraphy were included. On lung perfusion scintigraphy, 134 patients had normal lung perfusion and 184 patients showed patchy perfusion defects. In comparison to patients with normal lung perfusion, patients with patchy perfusion defects experienced significantly higher mean pulmonary arterial pressure (58.0 ± 15.4 mmHg vs. 54.1 ± 16.2 mmHg, P = 0.027) and total pulmonary resistance (1192.6 ± 533.7 dyn·s·cm −5 vs. 1067.2 ± 549.3 dyn·s·cm −5 , P = 0.042). During a median follow-up period of 884.0 days, 53 patients reached the primary endpoint of all-cause mortality. On univariate Cox analysis, the patchy pattern of perfusion defect was significantly associated with the all-cause mortality (hazard ratio [HR] = 2.47, 95% confidence interval [CI] = 1.32–4.63, P = 0.005). Patients with patchy perfusion defects had a worse outcome (log-rank = 8.605, P = 0.003). On multivariate analysis, the patchy pattern remained as a significant independent predictor of the endpoint (HR = 2.30, 95% CI = 1.22–4.31, P = 0.010). IPAH patients presented with heterogeneity in lung perfusion and the patchy pattern of lung perfusion defect commonly existed. Patients with patchy pattern identified by lung perfusion scintigraphy were associated with more severe disease and worse outcome.

Permafrost thawing may lead to the release of carbon and nitrogen in high-latitude regions of the Northern Hemisphere, mainly in the form of greenhouse gases. Our research aims to reveal the effects of permafrost thawing on CH4 and N2O emissions from peatlands in Xiaoxing’an Mountains, Northeast China. During four growing seasons (2011–2014), in situ CH4 and N2O emissions were monitored from peatland under permafrost no-thawing, mild-thawing, and severe-thawing conditions in the middle of the Xiaoxing’an Mountains by a static-chamber method. Average CH4 emissions in the severe-thawing site were 55-fold higher than those in the no-thawing site. The seasonal variation of CH4 emission became more aggravated with the intensification of permafrost thawing, in which the emission peaks became larger and the absorption decreased to zero. The increased CH4 emissions were caused by the expansion of the thawing layer and the subsequent increases in soil temperature, water table, and shifts of plant communities. However, N2O emissions did not change with thawing. Permafrost thawing increased CH4 emissions but did not impact N2O emissions in peatlands in the Xiaoxing’an Mountains. Increased CH4 emissions from peatlands in this region may amplify global warming.

The key point of our work was evaluating the impact of terpene alcohols on the aroma expression of terpenes recombination in Chrysanthemum essential oils. Using pure commercial products, various aromatic recombinations were prepared, consisting of terpenes recombination and six terpene alcohols, all the concentrations found in Chrysanthemum essential oils. There were five groups of terpene alcohols mixtures performed very interesting with the addition or omission tests. The “olfactory threshold„ of the terpenes recombination had a notable decrease when adding isoborneol, d-Fenchyl alcohol respectively through the Feller’s additive model analysis. Furthermore, the descriptive test indicated that the addition of terpene alcohols mixture had the different effect on fruity, floral, woody, green, and herbal aroma intensity. Specifically, when isoborneol was added to the terpenes recombination in squalane solution, it was revealed that isoborneol had a synergy impact on herbal and green notes of the terpenes recombination and masked the fruity note.

The incidence of obesity is increasing worldwide. It was reported that endoplasmic reticulum stress (ERS) could inhibit insulin receptor signaling by activating c-Jun N-terminal kinase (JNK) in the liver. However, the relationship between ERS and insulin receptor signaling in the brain during obesity remains unclear. The aim of the current study was to assess whether ERS alters insulin receptor signaling through the hyper-activation of JNK in the hippocampus and frontal cortex in the brains of obese rats. Obesity was induced using a high fat diet (HFD). The Morris water maze test was then performed to evaluate decreases in cognitive function, and western blot was used to verify whether abnormal insulin receptor signaling was induced by ERS in HFD rats exhibiting cognitive decline. In addition, to determine whether ERS activated JNK and consequently impaired insulin receptor signaling, SH-SY5Y cells were treated with the JNK inhibitor, SP600125, followed by tunicamycin or thapsigargin, and primary rat hippocampal and cortical neurons were transfected with siRNA against IRE1α and JNK. We found that the expression of phosphorylation of PKR-like kinase (PERK), phosphorylation of α subunit of translation initiation factor 2 (eIF2α), and phosphorylation of inositol-requiring kinase-1α (IRE-1α) were increased in the brains of rats with HFD when compared with control rats. The level of serine phosphorylation of insulin receptor substrate-1 (IRS-1) was also increased, while protein kinase B (PKB/Akt) was reduced. ERS was also found to inhibit insulin receptor signaling via the activation of JNK in SH-SY5Y cells, primary rat hippocampal, and cortical neurons. These results indicate that ERS was increased, thereby resulting in impaired insulin receptor signaling in the hippocampus and frontal cortex of obese rats.

BACKGROUND: Although many studies have confirmed a relationship between microRNAs (miRNAs) and cholangiocarcinoma (CCA), the real-time dynamics of miRNA function have not been examined. METHODS: miRNA reporter constructs were generated using a recombinant adeno-associated virus vector, which contained complementary sequences for six miRNAs (miR-200a, miR-200b, miR-21, miR-146a, miR-155, and miR-221), along with two independent expression cassettes encoding the fluorescent reporter genes Fluc and Gluc. The spatio-temporal function of each miRNA was monitored both in CCA and control tissues. RESULTS: All miRNAs participated in CCA development, with distinct patterns of expression over time. The activity of miR-21 was significantly lower in female T3N0M0 CCA tissue relative to controls at three time points, yet was higher in two male T3N1M0 CCA tissues. The difference in miR-200b function between two male T3N1M0 CCA tissues and their corresponding controls peaked at 24 h, while function in a female T3N0M0 CCA was detected only at 72 h. The four remaining miRNAs (miR-200a, miR146a, miR-155, and miR-221) displayed patient-specific activity patterns in both CCA and control tissues. CONCLUSION: Significant variability was observed in the temporal function of all six miRNAs, which may play an important role in the development of CCA.

Multiple organ systems, including the brain, which undergoes changes that may increase the risk of cognitive decline, are adversely affected by diabetes mellitus (DM). Here, we demonstrate that type 2 diabetes mellitus (T2DM) db/db mice exhibited hippocampus-dependent memory impairment, which might associate with a reduction in dendritic spine density in the pyramidal neurons of brain, Aβ1-42 deposition in the prefrontal cortex (PFC) and hippocampus, and a decreased expression of neurostructural proteins including microtubule-associated protein (MAP2), a marker of dendrites, and postsynaptic density 95 (PSD95), a marker of excitatory synapses. To investigate the effects of the ZiBuPiYin recipe (ZBPYR), a traditional Chinese medicine recipe, on diabetes-related cognitive decline (DACD), db/db mice received daily administration of ZBPYR over an experimental period of 6 weeks. We then confirmed that ZBPYR rescued learning and memory performance impairments, reversed dendritic spine loss, reduced Aβ1-42 deposition and restored the expression levels of MAP2 and PSD95. The present study also revealed that ZBPYR strengthened brain leptin and insulin signaling and inhibited GSK3β overactivity, which may be the potential mechanism or underlying targets of ZBPYR. These findings conclude that ZBPYR prevents DACD, most likely by improving dendritic spine density and attenuating brain leptin and insulin signaling pathway injury. Our findings provide further evidence for the effects of ZBPYR on DACD.

Ultraviolet-B (UV-B) stress appears to be more striking than other research works because of the thin ozone layer. The protective influence of an exogenous nitric oxide donor and sodium nitroprusside (SNP) on the growth properties of Larix gmelinii seedlings was investigated under ultraviolet-B radiation conditions. The results indicated that 0.1 mM SNP could effectively alleviate the damage caused by ultraviolet-B radiation, and improved the seedling growth properties, the relative water content, and photosynthetic pigment content in leaves. Additionally, the photosynthetic capacity and antioxidant enzyme activity were increased during the exposure. On the contrary, the damage caused by active oxygen was decreased in SNP-treated seedling leaves. The damage caused by ultraviolet-B radiation was slightly reduced after treating with 0.01 mM SNP. Nevertheless, treatment with 0.5 mM SNP had a negative effect under ultraviolet-B radiation. Furthermore, supplementing NO (nitric oxide) improved the photosynthetic capacity and antioxidant enzyme activity and alleviated the damage of caused by active oxygen. The best effective concentration of SNP was 0.1 mM. Therefore, a suitable amount of exogenous NO can protect the Larix gmelinii seedlings and increase their tolerance to ultraviolet-B radiation.

The permafrost carbon–climate feedback is one of the major mechanisms in controlling the climate–ecosystem interactions in northern high latitudes. Of this feedback, methane (CH _4 ) emission from natural wetlands is critically important due to its high warming potential. The freeze–thaw transition has been confirmed to play an important role in annual CH _4 budget, yet the magnitude of this effect is uncertain. An intensive field campaign was carried out in the Sanjiang Plain, Northeast China to estimate the CH _4 emission in the spring freeze–thaw transition period. The observation concluded that a large CH _4 source was caused by spring thaw; the maximum hourly emission rate was 48.6 g C m ^−2 h ^−1 , more than three orders of the regularly observed CH _4 emission rate in the growing season. In some sporadically observed ‘hot spots’, the spring thawing effect contributed to a large CH _4 source of 31.3± 10.1 g C m ^−2 , which is approximately 80% of the previously calculated annual CH _4 emission in the same study area. If our results are typical for natural wetlands in the Northern Hemisphere permafrost region, we estimate a global CH _4 source strength of 0.5–1.0 Tg C (1 Tg =10 ^12 g) caused by spring thaw in the Northern Hemisphere permafrost region in the year 2011. Combining with available satellite and flask data, a regional extrapolation reaches a temporal pattern of CH _4 emission during 2003–2009 which is consistent with recently observed changes in atmospheric CH _4 concentration in the high latitudes. This suggests that the CH _4 emission upon spring thaw in the high latitudes might be enhanced by the projected climate warming. These findings indicate that the spring thawing effect is an important mechanism in the permafrost carbon–climate feedback and needs to be incorporated in Earth system models.
