Abstract Gene‐editing technology has become a transformative tool for the precise manipulation of biological genomes and holds great significance in the field of animal disease‐resistant breeding. Mastitis, a prevalent disease in animal husbandry, imposes a substantial economic burden on the global dairy industry. In this study, a regulatory sequence gene editing breeding strategy for the successful creation of a gene‐edited dairy (GED) goats with enhanced mastitis resistance using the ISDra2‐TnpB system and dairy goats as the model animal is proposed. This included the targeted integration of an innate inflammatory regulatory sequence (IRS) into the promoter region of the lysozyme (LYZ) gene. Upon Escherichia Coli (E. coli) mammary gland infection, GED goats exhibited increased LYZ expression, showing robust anti‐mastitis capabilities, mitigating PANoptosis activation, and alleviating blood‐milk‐barrier (BMB) damage. Notably, LYZ is highly expressed only in E. coli infection. This study marks the advent of anti‐mastitis gene‐edited animals with exogenous‐free gene expression and demonstrates the feasibility of the gene‐editing strategy proposed in this study. In addition, it provides a novel gene‐editing blueprint for developing disease‐resistant strains, focusing on disease specificity and biosafety while providing a research basis for the widespread application of the ISDra2‐TnpB system.

BackgroundCold environments reduce work efficiency and increase the risk of frostbite. ObjectiveTo investigate the effects on cold protection and manual performance of rescue gloves in cold environments, and to preliminarily verify the feasibility of related quantitative characterization methods. MethodsAn experiment was conducted in a large and a small artificial climate chamber. 12 male right-handed university students were recruited. The experiment was divided into two parts, with 0-10 min as the baseline phase and 10-50 min as the test phase. Subjects sat quietly in a 16 ℃ large chamber, and hands were exposed to a small chamber at 0 ℃ either barely (control group) or with rescue gloves (experimental group). Skin temperatures of the five fingers (thumb, index finger, middle finger, ring finger, and little finger), as well as the dorsal and palm of the hands were measured. In addition, grip strength, manual dexterity, maximum finger flexion, and tactile sensitivity of the hands were measured as the indicators of manual performance. ResultsIn the control group, cold-induced vasodilation occurred when the skin temperature of the five fingers were lowered to about 10 ℃. The maximum rate of change in skin temperature of the control group was in the following order: thumb＞index finger＞middle finger＞little finger＞ring finger＞dorsal＞palm, 165.7 ℃·h−1 in the thumb, 80.0 and 89.9 ℃·h−1 in the palm and dorsal respectively. In addition, the minimum skin temperature of the thumb was 6.6 ℃. Compared with the control group: the rate of change of skin temperature in the experimentalgroup decreased by about 50%; the grip strength decreased by 15% after wearing the rescue gloves (P0.05). ConclusionThis study proved that the rescue gloves improved the thermal comfort of hands while better balancing the effect of cold exposure on manual performance. The quantitative characterization methods can evaluate the protective performance and manual performance of the rescue gloves, which provides a reliable theoretical foundation and reference basis for further experiments.

Store separation is one of the essential issues for weapons in warfare missions, and matters to the carrier safety. In this paper, based on the CFD methods including polyhedral hybrid mesh, overset mesh and 6-DOF equations of rigid body motion, the simulation of store separation of a calibration model was carried out. The effect of initial ejection forces on the separation safety was studied, and the separation characteristics in both the longitudinal and lateral motions were analyzed, which can be used as a reference for engineering applications.

Abstract The Gabija system is a newly discovered bacterial immune system that consists of GajA and GajB. Here we report the cryo-EM structure of the Gabija complex from Bacillus cereus VD045 at 3.6 Å, which provides the direct evidence of interactions between GajA and GajB. The Gabija complex is an octameric ring structure with four GajA and four GajB. GajA is an OLD nucleases family protein, while GajB belongs to the SF1 helicases. The Gabija complex has sequence-specific DNA nuclease activity and prefers circular rather than linear DNA as substrate, its activity is more sensitive to concentrations change of nucleotides compared to GajA alone. Our data suggest a mechanism of Gabija immunity: the nuclease activity of Gabija complex is inhibited under physiological conditions, while it is activated by depletion of NTP and dNTP upon the replication and transcription of invading phages and cleave the circular DNA to prevent phage DNA replication.

The thermo-rheological structure of the lithosphere is an effective method to reveal the thermal state within the lithosphere. Studies of the thermo-rheological structure of the lithosphere in geothermal anomaly areas can effectively constrain the contribution of heat sources. The southeastern China is an important distribution region for geothermal resources, with a large number of natural hot springs emerging on the land surface. Boreholes in this region have identified high geothermal gradients at depth. However, the mechanism of the heat sources is still controversial, and whether the hot dry rock resources exists is not clear. In this study, we take the Huangshadong geothermal field in Huizhou of Guangdong as the research target. We analyze the temperature distribution and rheological strength of the lithosphere, discuss the heat sources of the Huangshadong geothermal field, examine the thermal influence of shallow hydrothermal systems, and predict the prospects of dry hot rock resources. The results show that the heat flux under the influence of hydrothermal systems in the Huangshadong geothermal field is 130.3 mW/m2, and the crustal heat flow is similar to the mantle heat flow, showing a warm-crust-warm-mantle lithospheric thermal structure. In addition, the structural heat flux reaches 30.5–60.3 mW/m2. The rheological structure of the lithosphere shows that the middle crust has a ductile rheological layer, the upper crust and the lower crust are mainly controlled by brittle failure, and the lower crust and the lithosphere mantle show coupling in the rheological structure, which indicates a relatively stable crustal bottom boundary. The heat sources of the Huangshadong geothermal field is dominated by the tectonic heat source, and the mantle heat source and radiogenic heat production are the main heat source components. The main parts of tectonic heat source include the heat accumulation in regional deep faults and cyclic heat transfer in hydrothermal systems, both of which may carry heat to the surface through “relay” heat transfer. The proportion of heat accumulation of regional deep faults in the tectonic heat source is the key factor affecting the prospects of dry hot rock resources.

The Nanping geothermal field in Hainan is situated within the Wuzhi Mountain fold belt of the South China fold system based on its geotectonic units. Although there is abundant surface heat detected and widespread distribution of Late Mesozoic granite in the area, the geological background of geothermal resources remains unclear. In this article, we collected core samples from boreholes within the Nanping geothermal field to conduct testing and analysis on rock thermal conductivity and heat-production rate. By combining these results with temperature logging data, we discuss a method for diterming the heat flow background of convective geothermal system. Furthermore, the study analyzed the geothermal flux and deep thermal structure of the research area. The results demonstrate that the average radioactive heat production rate of the Baocheng rock mass in the study area is 3.16 μW/m³, primarily attributed to the decay heat of Th and U, while the heat contribution of K is negligible. The thermal conductivity values of the rocks are relatively high, ranging from 2.29 to 3.75 W/(mK), slightly exceeding the average thermal conductivity of the upper crust. The study area represents a typical convective geothermal field influenced by groundwater convection, exhibiting a high geothermal temperature gradient. Using the groundwater-correction method, the geothermal flux in the study area is calculated to be 89–108.27 mW/m², of which the thermal conduction component is 73.17 mW/m² and the convective component is 15.83–35.1 mW/m². Among these components, heat generated from radioactive decay of crustal radioactive elements contributes 35.44 mW/m² to thermal conduction, while deep mantle conduction accounts for a heat flux is 37.73 mW/m², with a ratio of 1:1.07 between them. The difference between crustal and mantle heat fluxes is minimal in this region, indicating an approximation towards a “crust-mantle heat source balance zone”. Furthermore, the thickness of the “hot” lithosphere in the study area ranges from 42 to 46 km, indicating significant characteristics of extension-thinning.

The Huangshadong geothermal field (HGF), situated in the contact zone between Mesozoic granites and NE-striking dominant faults in South China, has great geothermal potential. Petrogenesis of reservoir rock plays an important role in understandings its genetic mechanism and assessing geothermal potential. However, due to the lack of rock sample at depth collected from the geothermal reservoir, the petrogenesis of granites in the geothermal reservoirs of the HGF, remains an enigma. This study elucidated the petrogenetic characteristics of these granites sampled directly from geothermal reservoir at the depth of ∼3,000 km and their geothermal implications through zircon U-Pb dating, geochemical analysis, and Hf isotopic analysis. The zircon U-Pb ages indicate that the magmatism evolution of HGF contains three eras, namely, Cretaceous (135 ± 4 to 143.6 ± 2.8 Ma), Jurassic (152.7 ± 2.7 to 176.7 ± 1.8 Ma), and Permian granites (251 ± 9.1 to 251 ± 5 Ma) from the youngest to oldest. The reservoir granites were emplaced during the latest stage of Cretaceous intrusion, as indicated by the zircon U-Pb ages (135 ± 4 Ma and 135.3 ± 2.4 Ma) of rock samples from the deep part of well HR-1. These Cretaceous rocks are highly fractionated I-type granites, featuring high SiO2, K2O, and Na2O contents, high Rb/Sr ratios, low Zr/Hf, Nb/Ta, and Th/U ratios, and A/CNK values of 1.05–1.13. Compared to other Cretaceous granites outcropping on the margin of the HGF, these granites have undergone the strongest fractional differentiation. The Cretaceous granites in the HGF are high-heat-producing rocks (>5 μW/m3), with an average heat production rate of 6.63 μW/m3. Notably, the Cretaceous reservoir granites (as reservoir rocks) serve as an important heat source for the formation of geothermal resources in the HGF. In addition, the zircon Hf isotopic composition indicates that the reservoir Cretaceous granites originated from Meso-to Paleo-Proterozoic lower crustal materials (TDM2: 1,385 to 1907 Ma).

In order to evaluate the prospects of drift step recovery diode (DSRD) based on Silicon Carbide (SiC) in pulsed power field, the characteristics of DSRD based on SiC and Silicon (Si) are compared. The simulation model of the circuit based on Sentaurus TCAD is established and a trigger circuit using the saturated transformer is adopted. The simulation results show that under the same conditions, SiC DSRD has advantages in the aspect of rise time and it is less affected by pedestal effect, which is detrimental to the pulse quality and produces energy loss. The advantage of Si DSRD is that the amplitude of the output pulse voltage is high. By testing the voltage and the rise time of the output pulse, the influence of different factors including turns ratio and input voltage are discussed. For single chip of SiC DSRD with 5 kV blocking voltage, the turns ratio of transformer should be 1:5 to ensure that the parasitic inductance of transformer is low and the input voltage should be more than 500 V to guarantee that the magnetic core can be saturated. For Si DSRD stacked in series, the rise time and the width of output pulse is relatively stable and have little correlation with the changes of the input voltage, the forward pumping time and the circuit parameters. The experiment proves that SiC DSRD and Si DSRD have the advantages of the rise time and the output pulse voltage, respectively, which is consistent with the simulation results.

The combination of levulinic acid (LVA) and sodium dodecyl sulfate (SDS) in recent years has shown a considerable potential to use as an antimicrobial intervention. The objectives of this study were to evaluate the antimicrobial efficacy of the combination against Staphylococcus aureus in both planktonic and biofilm states and to investigate the transcriptional changes in S. aureus biofilms coincubated with sublethal concentrations of LVA and/or SDS. The minimum inhibitory concentrations (MICs) of LVA and SDS determined by the microdilution method were 3.125 and 0.039 mg/mL, respectively. An additive bacteriostatic interaction (fractional inhibitory concentration index = 1) between the two compounds was observed by the checkerboard assay, whereas a synergistic bactericidal activity was displayed by the time-kill assay. The biomass and viable cells in the biofilms were reduced by both antimicrobials either alone or in combination in a dose-dependent manner. Transcriptomics indicated that more differentially expressed (DE) genes were observed in the biofilm treated with SDS (103 up- and 205 downregulated DE genes) and LVA + SDS (187 up and 162 down) than that coincubated with LVA (34 up and 32 down). The SDS and LVA + SDS treatments mainly affected the expression of genes responsible for cell surface proteins, virulence factors, adhesins, and capsular polysaccharides. Both the antibiofilm assay and the transcriptomics indicated that SDS, not LVA, was the major chemical contributing to the antibacterial efficacy of the combination. This study reveals the behavioral responses and protective mechanisms of S. aureus to LVA and SDS applied individually or in combination.

High-temperature hydrothermal activity areas in western Sichuan Province, China are ideal objects for studying deep Earth science, extreme ecological environments, and comprehensive geothermal utilization. To understand the geochemical characteristics of rare Earth elements (REEs) in the Chaluo hot springs in western Sichuan Province, the authors analyzed the composition and fractionation of REEs in the hot springs through hydrochemical analysis, REE tests, and North American Shale Composite-normalized REE patterns. Moreover, the composition and complex species of REEs in the geothermal water in the Chaluo area were determined through calculation and simulation analysis using the Visual MINTEQ 3.0 software. The results are as follows. In terms of hydrochemical type, all geothermal water in the Chaluo area is of the Na-HCO3 type. The cations in the geothermal water are mainly controlled by water-rock interactions and evaporation, the anions are determined by water-rock interactions, and the hydrochemical processes are primarily controlled by the dissolution of silicate minerals. The total REE content of the geothermal water in the Chaluo hot springs is 0.306 ± 0.103 ug/L. It is low compared to the Kangding area and is primarily affected by the reductive dissolution of Fe oxides/hydroxides, followed by pH. The geothermal water in the Chaluo area is rich in light rare Earth elements (LREEs) because of the presence of Fe oxides. It shows positive Eu and Ce anomalies due to the combined effects of the dissolution of Eh and Mn oxides and surface water. Furthermore, the positive Eu anomalies are also caused by the water-rock interactions between the Qugasi Formation and deep geothermal water. Similar to alkaline water bodies, the complex species of REEs in the geothermal water mainly include Ln(CO3)2−, LnCO3+, and LnOH2+, which is caused by the stability constants of complexation reactions.

Nonalcoholic fatty liver disease (NAFLD) has been demonstrated to be closely associated with intestinal barrier functions. The aim of this study is to investigate the effects of nuciferine on preventing hepatic steatosis with relation to intestinal barrier functions in HFD-fed rats. The results showed nuciferine mitigated body weight gain, hyperlipemia and hepatic steatosis with regulating lipid metabolic gene expressions. Meanwhile, nuciferine decreased LPS-induced inflammation through inhibiting TLR4/MyD88/NF-κB pathway and reinforced intestinal barriers through increasing occludin and zonula occluden-1 expression, goblet cells, mucin2 (MUC2) expression. Furthermore, we found that nuciferine changed the relative abundance of mucus-related microbiota (Akkmensia muciniphila, Ruminococcaceae) and LPS-producing microbiota (Desulfovibrionaceae), increased the concentration of fecal acetic acid and propionic acid accompanied by decreased fecal pH. Together, anti-NAFLD effects of nuciferine are related to improving multiple intestinal barrier functions. These findings imply that nuciferine could be a promising agent for NAFLD with a target towards gut-liver axis.

The aim of this study was to elucidate the effect of pH on bacterial resistance mechanisms to copper (Cu) stress by genomic and transcriptomic analysis. Klebsiella michiganensis cells were exposed to 0.5 mM CuCl2 at pH 4 and 5. Lower pH (pH 5) induced Cu precipitation in the medium. Transcriptomic analyses indicated that two groups of genes related to quorum sensing (QS) systems (lsrABCDFGKR) and type II secretion systems (T2SS) (gspCDEFGHIJKLM) were significantly up-regulated at pH 4 only. These results suggest that T2SS may be induced and controlled by QS, thereby contributing to the formation of extracellular polymeric substances (EPS) and the secretion of proteins to prevent Cu ions from entering cells. Six Cu resistance genes (cusABF, copA, cueO, and gene05308) were more significantly up-regulated at pH 4 than at pH 5. In addition, the relative expression (log2|FC=) of the sulfur assimilation genes cysHJIK was relatively higher at pH 4 than at pH 5, while the gene encoding organic sulfur metabolism, tauB, was also significantly up-regulated at only pH 4. These results indicate that the Cu efflux system can remove intracellular Cu ions from cells, and that the sulfur assimilation system is related to the detoxification of Cu ions. Furthermore, increased free Cu ions at lower pH (4) could induce communication signals among cells, thereby stimulating the response of T2SS-related genes in K. michiganensis to tolerate Cu stress. Consequently, the resistance of K. michiganensis to Cu stress is a multisystem collaborative process composed of intracellular and extracellular components.

Macrolides are a significant family of natural products with diverse structures and bioactivities. Considerable effort has been made in recent decades to isolate additional macrolides and characterize their chemical and bioactive properties. The majority of macrolides are obtained from marine organisms, including sponges, marine microorganisms and zooplankton, cnidarians, mollusks, red algae, bryozoans, and tunicates. Sponges, fungi and dinoflagellates are the main producers of macrolides. Marine macrolides possess a wide range of bioactive properties including cytotoxic, antibacterial, antifungal, antimitotic, antiviral, and other activities. Cytotoxicity is their most significant property, highlighting that marine macrolides still encompass many potential antitumor drug leads. This extensive review details the chemical and biological diversity of 505 macrolides derived from marine organisms which have been reported from 1990 to 2020.

The changes of volatile composition and other quality traits of blueberry during postharvest storage were investigated. Blueberries were packaged in vented clam-shell containers, and stored at 0 °C for 0, 15 and 60 days, followed by storage at room temperature (25 °C) for up to 8 days for quality evaluation. The firmness, pH, and total soluble solids increased by 8.42%, 8.92% and 42.9%, respectively, after 60 days of storage at 0 °C. Titratable acidity decreased 18.1% after 60 days of storage at 0 °C. The volatile change was monitored using headspace–solid-phase microextraction–gas chromatography–quadrupole time-of-flight–mass spectrometry (HS-SPME-TOF-MS) and off-odor was evaluated by sensory panel. Volatile compounds generally showed a downward trend during cold storage. However, the subsequent shelf life was the most remarkable period of volatile change, and was represented by the strong fluctuation of ethyl acetate and the rapid decrease of terpenoids. Extending storage from 15 to 60 days under cold condition still resulted in an acceptable odor. However, subsequent storage at higher temperature resulted in a quick deterioration in sensory acceptability. The results proved that cold storage was a reliable way to maintain the quality of blueberry, and flavor deterioration during subsequent shelf life was more fatal to the blueberry flavor.

The energy reserves of hot dry rock resources are huge, thus a model to predict engineering production for efficient and stable development and utilization is sought. Based on the geological characteristics of dry rock resources in Guide Basin, Qinghai Province, China, the fully coupled wellbore–reservoir simulator—T2Well—is used to model a production system using water as a heat transfer medium and simulate the system’s operation to analyze the influence of different injection rates on heat extraction. In later production stages, output temperature and reservoir pressure decrease by 10–30°C and 0.5–30 MPa, depending on injection rate; this occurs earlier and to a greater extent at higher injection rates; thermal breakthrough also occurs earlier (7–10 years). The heat extraction rate is 1–20 MW and the cumulative heat extracted is 2.1–24.2 × 10 5 J. Lower injection rates result in relatively low heat extraction rates. For maximum economic benefit, an injection rate of 50–75 kg/s is ideal.

Type I DNA methyltransferases contain one specificity subunit (HsdS) and two modification subunits (HsdM). The electron microscopy model of M.EcoKI-M₂S₁ methyltransferase shows a reasonable closed state of this clamp-like enzyme, but the structure of the open state is still unclear. The 1.95 Å crystal structure of the specificity subunit from Thermoanaerobacter tengcongensis (TTE-HsdS) shows an unreported open form inter-domain orientation of this subunit. Based on the crystal structure of TTE-HsdS and the closed state model of M.EcoKI-M₂S₁, we constructed a potential open state model of type I methyltransferase. Mutational studies indicated that two α-helices (aa30-59 and aa466-495) of the TTE-HsdM subunit are important inter-subunit interaction sites in the TTE-M₂S₁ complex. DNA binding assays also highlighted the importance of the C-terminal region of TTE-HsdM for DNA binding by the TTE-M₂S₁ complex. On the basis of structural analysis, biochemical experiments and previous studies, we propose a dynamic opening and closing mechanism for type I methyltransferase.

Objective To explore the predictive value of Neutrophil-to-lymphocyte ratio (NLR) and fibrinogen-to-albumin ratio (FAR) for elderly patients with acute exacerbation of chronic obstructive pulmonary disease (AECOPD) complicated with pulmonary hypertension. Methods A total of 50 elderly AECOPD patients with pulmonary hypertension who were hospitalized in the First Hospital of Shanxi Medical University from January to June 2021 were selected as the combined group, and 61 elderly AECOPD patients without pulmonary hypertension during the same period were selected as the control group. The clinical data and laboratory parameters of the patients at admission were collected, the risk factors of pulmonary hypertension in elderly patients with AECOPD were analyzed by multivariate logistic regression, and the predictive value of NLR and FAR for pulmonary hypertension in elderly patients with AECOPD was evaluated by ROC curve. Results There were significant differences in age, body mass index, length of hospital stay, AECOPD severity grade, use of ventilator, intravenous hormone therapy, antibiotic course, NLR, FAR, white blood cell count, neutrophil count, hemoglobin, fibrinogen, D-dimer, procalcitonin, B-type natriuretic peptide, partial pressure of carbon dioxide, right atrial diameter, right ventricular diameter, lymphocyte count, eosinophil count, albumin, oxygenation index, left atrial diameter and left ventricular end-diastolic diameter between the two groups (P

Halobenzoquinones (HBQs) are ubiquitous disinfection by-products (DBPs) in chlorinated drinking water with various health risks including reproductive toxicity, while the potential mechanisms are still unclear. Although green tea exhibits common detoxifying properties, its ability to mitigate the toxicity of HBQs still needs to be further deepened and explored. This study attempted to investigate the possible mechanism of the most common HBQ, 2,6-dichloro-1,4-benzoquinone (2,6-DCBQ) induced reproductive toxicity and elucidate the protective effect of green tea using a series of liquid chromatography-tandem mass spectrometry (LC-MS) approaches. Firstly, in vivo experiments showed that 2,6-DCBQ could induce testicular damage in male rats via significantly decreasing sperm-associated Leydig cells and seminiferous tubules. Then, in vitro incubation of 2,6-DCBQ with amino acids suggested that 2,6-DCBQ could bind to proteins via residues of cysteine or lysine and provided five additional modification patterns. Following, proteomics analysis revealed that at least 42 proteins were modified by 2,6-DCBQ, which were mainly enriched in the reproductive system. These results highlighted the significance of covalent protein modification in 2,6-DCBQ reproductive toxicity. Fortunately, we found that catechins (a class of major components of green tea) could competitively bind to 2,6-DCBQ in vivo and in vitro, reducing the amount and type of 2,6-DCBQ-protein adducts, thereby attenuating the reproductive system damage caused by 2,6-DCBQ. This study provides new insights into 2,6-DCBQ-induced reproductive system damage and reveals a new mechanism of green tea detoxification. Moreover, these findings offer potential strategies for alleviating the harmful impacts of environmental toxicants on human health.

Abstract Background Purple non-heading Chinese cabbage [Brassica campestris (syn. Brassica rapa) ssp. chinensis] has become popular because of its richness in anthocyanin. However, anthocyanin only accumulates in the upper epidermis of leaves. Further studies are needed to investigate the molecular mechanisms underlying the specific accumulation of it. Results In this study, we used the laser capture frozen section method (LCM) to divide purple (ZBC) and green (LBC) non-heading Chinese cabbage leaves into upper and lower epidermis parts (Pup represents the purple upper epidermis, Plow represents the purple lower epidermis, Gup represents the green upper epidermis, Glow represents the green lower epidermis). Through transcriptome sequencing, we found that the DIHYDROFLAVONOL 4-REDUCTASE-encoding gene BcDFR, is strongly expressed in Pup but hardly in others (Plow, Gup, Glow). Further, a deletion and insertion in the promoter of BcDFR in LBC were found, which may interfere with BcDFR expression. Subsequent analysis of gene structure and conserved structural domains showed that BcDFR is highly conserved in Brassica species. The predicted protein-protein interaction network of BcDFR suggests that it interacts with almost all functional proteins in the anthocyanin biosynthesis pathway. Finally, the results of the tobacco transient expression also demonstrated that BcDFR promotes the synthesis and accumulation of anthocyanin. Conclusions BcDFR is specifically highly expressed on the upper epidermis of purple non-heading Chinese cabbage leaves and regulates anthocyanin biosynthesis and accumulation. Our study provides new insights into the functional analysis and transcriptional regulatory network of anthocyanin-related genes in purple non-heading Chinese cabbage.

As autonomous driving technology advances, connected and autonomous vehicles (CAVs) will coexist with human-driven vehicles (HDVs) for an extended period. The deployment of CAVs will alter traffic flow characteristics, making it crucial to investigate their impacts on mixed traffic. This study develops a hybrid control framework that integrates a platoon control strategy based on the “catch-up” mechanism with lane management for CAVs. The impacts of the proposed hybrid control framework on mixed traffic flow are evaluated through a series of macroscopic simulations, focusing on fundamental diagrams, traffic oscillations, and safety. The results illustrate a notable increase in road capacity with the rising market penetration rate (MPR) of CAVs, with significant improvements under the hybrid control framework, particularly at high MPRs. Additionally, traffic oscillations are mitigated, reducing shockwave propagation and enhancing efficiency under the hybrid control framework. Four surrogate safety measures, namely time to collision (TTC), criticality index function (CIF), deceleration rate to avoid a crash (DRAC), and total exposure time (TET), are utilized to evaluate traffic safety. The results indicate that collision risk is significantly reduced at high MPRs. The findings of this study provide valuable insights into the deployment of CAVs, using control strategies to improve mixed traffic flow operations.

ABSTRACT: The aim of this study was to elucidate the different effects and difference mechanism of gelling properties among egg white (EW) treated with different heating intensities and the composite addition of rhamnolipid and soybean lecithin. Particle size analyzer, potentiometric analyzer, surface hydrophobicity method, and Fourier transform infrared spectroscopy techniques were used to determine the physicochemical properties and molecular structure, respectively. Low-field nuclear magnetic resonance, magnetic resonance imaging, texture profile analysis, and scanning electron microscopy techniques were used to analyze the gelling properties and gel structure, respectively. And we illuminate the different mechanisms in the gelling properties of the EW with various treatments and key internal factors that play important roles in improving gelling properties by establishing the link between the gelling properties and relevant characteristics by mixed effects model and visual network analysis. The results indicate raising the content of rhamnolipid decreased the migration of immobilized water in the EW gel and the free water content. At the heating intensities of 55 °C/3.5, 65 °C/2.5, and 67 °C/1.5 min, with an increase in rhamnolipid, the gel's cohesiveness, gumminess, and chewiness gradually increased. The mixed effects model indicated that heating intensities and composite ratios have a 2-way interaction on zeta potential, the relaxation time of bound water (T21), the content of bound water (P21), the content of immobilized water (P22), and fractal dimension (df) attributes (P < 0.05). The visual network analysis showed that the protein solubility, the relaxation time of immobilized water (T22), surface hydrophobicity, zeta potential, average particle size (d43) and the relaxation time of free water (T23) are critical contributors to the different gelling properties of EW subjected to various treatments and the improvement of gelling properties. This study will provide theoretical guidance for the development of egg white products and the expansion of egg white's application scope in the egg product processing industry.

Abstract The interplay between bacteria and their host influences the homeostasis of the human immune microenvironment, and this reciprocal interaction also affects the process of tissue damage repair. A variety of immunomodulatory commensal bacteria reside in the body, capable of delivering membrane vesicles (MVs) to host cells to regulate the local immune microenvironment. This research revealed, for the initial time, the significant enhancement of mucosal and cutaneous wound healing by MVs secreted by the human commensal Lactobacillus reuteri (RMVs) through modulation of the inflammatory environment in wound tissue. Local administration of RMVs reduces the proportion of pro‐inflammatory macrophages in inflamed tissues and mitigates the level of local inflammation, thereby facilitating the healing of oral mucosa and cutaneous wounds. The elevated oxidative stress levels in activated pro‐inflammatory macrophages can be modulated by RMVs, resulting in phenotypic transformation of macrophages. Furthermore, 3‐hydroxypropionaldehyde present in RMVs can decrease the mitochondrial permeability of macrophages and stabilize the mitochondrial membrane potential, thereby promoting the conversion of macrophages to an anti‐inflammatory phenotype. This study pioneers the significance of commensal bacterial MVs in tissue injury repair and presents a novel concept for the repair of tissue damage.

Egg yolk peptides were prepared by enzymatic hydrolysis of defatted egg yolk powder. The cryoprotective effects of these peptides were assessed by examining the functional, structural and physicochemical properties of egg yolk. Results indicated that egg yolk peptides could reduce viscosity and prevent the accumulation of egg yolk proteins during freezing, thereby mitigating the deterioration of its functional characteristics. The peptide hydrolyzed for 3 h demonstrated the lowest fluidity in thawed egg yolk. Additionally, Fourier transform infrared (FTIR) spectra and intrinsic fluorescence spectra revealed that egg yolk peptides could reduce damage to the secondary and tertiary structures of proteins, enhancing the stability of the protein network. Changes in the content of secondary structures showed that egg yolk peptides increased β-turns, decreased β-sheets, and maintained overall structural stability and strength. This study provides new insights for egg yolk peptides to inhibit frozen egg yolk gelation.
