Bees play a crucial role in terrestrial ecosystems. However, beekeepers are unable to monitor the state of beehives (bees and environment) all the time, which often results in bees escaping or even dying. Currently, some researchers provided the scheme of intelligent beehive monitoring system equipped with the Internet of Things (IoT), There remain two challenges: accurately monitor the environmental status around the hive and accurately track and monitor bees in real time. With the development of the IoT and computer vision algorithms, we hope to provide an automated and efficient system to meet the above challenges. In this paper, we proposed a hive monitoring system, and build a visualization module in the cloud to monitor the activity of bee colonies and the environmental dynamic changes. (1) We proposed a multi-bee tracking algorithm to solve the problem of monitoring bees at the door of the hive; (2) we constructed a dataset containing various complex scenes, named BEE22, for training and testing the performance of our algorithm; (3) we designed a bee counting rule, based on results of multi-bee tracking algorithm, to reasonably count the bees entering or leaving the beehive; (4) we have deployed multiple sensors around(center, margin, door, and environment) the hive to accurately reflect the changes in the environment around the hive. Experimental results demonstrate the effectiveness and excellence of our system. In particular, the tracking performance of the multi-bee tracking algorithm reaches 83.5 % ± 0.7 % Multiple Object Tracking Accuracy (MOTA) and 77.3 %±0.2 % Multiple Object Tracking Precision (MOTP), speeds up to 16 frames per second, compared with other algorithms, MOTA and Identity F1 Score (IDF1) are improved by 5.4 % and 8.2 % respectively. Moreover, our counting algorithm also achieved excellent results, with root mean square error (RMSE) of 1.3 ± 0.1, 0.2 ± 0.0, and 1.6 ± 0.1 in counting the number of bees current, entry, and out scene in an episode, respectively. After that, the system will be deployed and monitored for a long time in the actual scenario, it was found that the activity of bees decreased significantly under heavy rainfall conditions. Additionally, the activity of the bee colony will also increase accordingly, when the amplitude is 500 dB to 2000 dB, the temperature of the center of the beehive is 25 °C to 37 °C, and the humidity is 48 % to 67 %. In summary, our system can provide valuable information for bee farmers to make control decisions on hives.

In recent years, milk powder adulteration has emerged as a matter of great concern. In this study, a rapid, accurate, and efficient detection method based on surface-enhanced Raman spectroscopy (SERS) combined with principal component analysis (PCA) was established to detect milk powder adulteration. The “coffee ring” effect-based gold nanoparticles (Au NPs) as the SERS-enhancing substrate were coupled with a portable Raman spectrometer, which enabled the differentiation of various brands of milk powder and the detection of melamine in milk powder. The substrate exhibited good SERS enhancement ability with an enhancement factor of 104. Furthermore, a strong linear correlation with a correlation coefficient (R2) of 0.9903 was observed between the melamine Raman intensity and concentration from 0.5 to 5.0 mg/kg. The calculated limit of detection of melamine (LOD) was 0.15 mg/kg, while the limit of quantitation (LOQ) was 0.5 mg/kg. In addition, when the method was applied to the detection of melamine in milk powder samples, this method achieved the recovery rates of melamine in milk powder samples ranged from 92.83% to 98.86% with relative standard deviations between 0.84% and 1.14%. In summary, the established method offers the advantages of cost-effectiveness, less sample requirement, and shorter detection time, meeting the needs for milk powder classification and rapid melamine detection.

BackgroundIn the face of increasing antifungal resistance among Candida albicans biofilms, this study explores the efficacy of a combined treatment using Kangbainian lotion (KBN) and miconazole nitrate (MN) to address this challenge.MethodsUsing UPLC-Q-TOF/MS Analysis for Identification of Active Compounds in KBN Lotion; FICI for synergy evaluation, XTT and ROS assays for biofilm viability and oxidative stress, fluorescence and confocal laser scanning microscopy (CLSM) for structural and viability analysis, and real-time fluorescence for gene expression.ConclusionOur study indicates that the combined application of KBN and MN somewhat impacts the structural integrity of Candida albicans biofilms and affects the expression of several key genes involved in biofilm formation, including ALS1, ALS3, HWP1, HSP90, and CSH1. These preliminary findings suggest that there may be a synergistic effect between KBN and MN, potentially influencing not only the structural aspects of fungal biofilms but also involving the modulation of genetic pathways during their formation.

Abstract To investigate the clinical characteristics of Guillain–Barré syndrome (GBS) in patients with primary Sjögren’s syndrome (SS). Records of patients with positive anti-SSA antibodies hospitalized in the Beijing Tiantan Hospital between December 2011 and May 2020 were retrieved. Patients who fulfilled the criteria for diagnosis of GBS and primary SS were included, and their clinical data were analyzed. Among the 785 patients with positive anti-SSA, 52 patients were identified in this study. They were 27 males and 25 females with median age of 59 years old. Besides anti-SSA antibodies, multiple autoantibodies were detected in these patients including antinuclear antibody, anti-Ro52, anti-mitochondrial M2, anti-thyroid peroxidase and anti-thyroglobulin autoantibodies. Preceding infection was reported in 42 patients. Hyporeflexia/areflexia and limbs weakness were the most common manifestation and 35 patients presented cranial nerve injuries. GBS disability score of 3, 4 and 5 was scaled in 28 (53.8%), 15 (28.8%) and 3 (5.8%) patients respectively. Forty-six patients received intravenous immunoglobulin (IVIG) monotherapy, 5 patients were treated by IVIG plus glucocorticoids, and 51 patients improved during hospitalization. The frequency of male gender among the patients with both GBS and primary SS suggests an independent onset of GBS and the co-existence of these autoimmune diseases in patients with multiple autoantibodies. Majority of patients with GBS and primary SS experience benign disease course.

Supervised machine learning (SML) has become a crucial tool for estimating water quality parameters (WQPs) from satellite images. Its effectiveness relies heavily on synchronised in-situ datasets covering diverse water bodies. However, collecting such datasets is time-consuming, resulting in temporal gaps between sampling and imaging. In addition, the in situ dataset may exhibit an imbalance. These imperfections could introduce uncertainties to SML-derived models, compromising the accuracy of the WQP estimates. Using in situ data collected automatically every four hours, the estimation of both optically active parameters (OAPs) and non-optically active parameters (nOAPs) in the Middle Reaches of the Yangtze River (MRYR) serves as an example to illustrate the importance of this challenge in freshwater remote sensing. Additionally, the investigation was extended to estimate OAPs and nOAPs in lakes of Wuhan through manual sampling measurements, thereby bridging theoretical insights with real-world applications. Employing four ML algorithms, the SML-based models for each WQP were calibrated using in situ datasets with different spatio-temporal distributions. The results demonstrated that precision decreased with increasing time gaps, whereas most nOAPs (COD, TP, TN, pH, and DO) showed greater robustness to the time gap than the OAPs (turbidity, Secchi depth, Chl-a, and algae density). In sports, athletes’ initial training is crucial for their whole career. Initial training at the relevant board athletes are professionally trained for national and international competitions. However, the selection of potential athletes for traditional training is very hectic and uncertain due to the involvement of various factors. Thus, we need an intelligent approach to efficiently deal with the uncertain factors in selecting and recommending athletes for training camp. A multi-attribute group decision-making (MAGDM) approach offers a structured framework for evaluating and selecting athletes, considering various attributes that reflect stakeholder preferences and needs. In MAGDM, the human point of view is crucial. Several frameworks deal with the extraction of information in the decision-making process. The interval-valued picture fuzzy rough set (IVPFRS) contributes significantly to lower the uncertainty in the data derived from real-world scenarios. In this study, new aggregation operators (AOs) based on the Schweizer-Sklar t-norm (SSTN) and the Schweizer-Sklar t-conorm (SSTC), i.e., interval-valued picture fuzzy rough weighted averaging (IVPFRSSWA) and interval-valued picture fuzzy rough weighted geometric (IVPFRSSWG), are developed to aggregate the data in the form of the interval-valued picture fuzzy rough values (IVPFRVs). After their fundamental qualities are examined, the created AOs are used to solve the MAGDM problem. The results vary based on the values of the involved parameters in SSTN and SSTC. The findings obtained also contrast with those of other known AOs. Additionally, a graphic representation of each observation and result is provided.

Digital technology has brought drastic changes to the design methods, values, and design tools of neighbourhood public spaces, thereby changing the behavioural patterns of people in neighbourhood public spaces. Therefore, people’s requirements for urban public space have changed and are characterised by high efficiency, high precision, humanization, and high aesthetics. Scholars at home and abroad have conducted many studies and practises on the application of digital technology to neighbourhood design, but there is a lack of systematic research practises on the use and analysis of multi-faceted data. This paper selects the Xiaomi Beijing Changping II design project, simulates and deduces the physical environment of its location, simulates the results of the natural environment by using the data of sunshine, wind, and water catchment in the physical environment, and then simulates the crowd’s action paths by combining kinetic algorithms and ant algorithms to optimise the design methods, processes, and results of the neighbourhood public space on the basis of this research. The research team designed five groups of programmes for the project based on different design methods and processes, and conducted a comparative study of the five groups of programmes through the hierarchical analysis method in conjunction with the fuzzy comprehensive evaluation method, as well as discussing them in conjunction with the actual bidding results of the project and the scoring conclusions of the industry experts. The results of the study show that the design scheme for neighbourhood public space based on physical environment simulation and crowd simulation is better able to take advantage of the analytical and predictive advantages of the technology and unite with the designer’s aesthetic interests, balancing the data objectivity and aesthetic subjectivity in the design process. The method is more likely to achieve a design solution that combines systematicity, foresight, rationality, and aesthetics, and provides an empirical case for the application of data simulation in public space, aiming to improve the rationality of public space design and solve the data-objective problems faced by the design of public space at this stage.

Furfural is an essential compound that contributes to the distinctive flavor of sauce-flavored Baijiu. However, traditional detection methods are hindered by lengthy and complex sample preparation procedures, as well as the need for expensive equipment. Therefore, there is an urgent need for a new approach that allows rapid detection. In this study, we developed a novel surface-enhanced Raman spectroscopy (SERS) substrate by constructing MXene (Ti3C2TX) @Ag nanoparticles (Ag NPs) through an electrostatic attraction method. The MXene (Ti3C2TX) @Ag NPs were successfully fabricated, with adsorbed NaCl-treated Ag NPs uniformly absorbed on the surface of MXene (Ti3C2TX), creating high-density distributed SERS “hot spots”. The prepared substrate demonstrated excellent sensitivity, uniformity, repeatability, and long-term stability, with a low detectable concentration of 10−9 M for R6G (Rhodamine 6G) and an enhancement factor of up to 7.08 × 105. When applied for the in situ SERS detection of furfural in Baijiu, the detection limit was as low as 0.5 mg/L. Overall, the proposed method offers rapid, low-cost, and sensitive quantitative analysis, which is significant not only for detecting furfural in Baijiu but also for identifying hazardous substances and distinguishing between authentic and counterfeit Baijiu products.

Carotenoids belonging to the class of tetraterpenoids have extensive applications in medicine, food, nutrition, cosmetics, and feed. Among them, lutein and zeaxanthin can prevent macular degeneration in the elderly, which is very important for protecting vision. Here, we introduce the first metabolomic analysis of Sphingopyxis sp. USTB-05, aiming to shed light on the biosynthesis of carotenoids. Sphingopyxis sp. USTB-05 has the complete methylerythritol 4-phosphate (MEP) pathway and carotenoid biosynthesis pathway, especially involved in the bioconversion of zeaxanthin, violaxanthin, and astaxanthin. Metabolomic profiling identified seven carotenes and six xanthophylls synthesized by Sphingopyxis sp. USTB-05. Zeaxanthin, in particular, was found to be the most abundant, with a content of 37.1 µg/g dry cells. Collectively, the results presented herein greatly enhance our understanding of Sphingopyxis sp. USTB-05 in carotenoids biosynthesis, and thus further accelerate its fundamental molecular investigations and biotechnological applications.

Microcystins (MCs) are the most widespread cyanobacterial toxins in eutrophic water body. As high toxic intermediate metabolites, linearized MCs are further catalyzed by linearized microcystinase (MlrB) of Sphingopyxis sp. USTB-05. Here MlrB structure was studied by comprizing with a model representative of the penicillin-recognizing enzyme family via homology modeling. The key active sites of MlrB were predicted by molecular docking, and further verified by site-directed mutagenesis. A comprehensive enzymatic mechanism for linearized MCs biodegradation by MlrB was proposed: S77 transferred a proton to H307 to promote a nucleophilic attack on the peptide bond (Ala-Leu in MC-LR or Ala-Arg in MC-RR) of linearized MCs to form the amide intermediate. Then water was involved to break the peptide bond and produced the tetrapeptide as product. Meanwhile, four amino acid residues (K80, Y171, N173 and D245) acted synergistically to stabilize the substrate and intermediate transition states. This study firstly revealed the enzymatic mechanism of MlrB for biodegrading linearized MCs with both computer simulation and experimental verification.

Abstract In biological systems, molecular assembly primarily relies on the assistance of molecular chaperones. Inspired by nature, strategies like ‘chaperone‐assisted assembly’ and ‘catalyzed assembly’ have been proposed for the sophisticated control of molecular assembly. Nonetheless, significant challenges remain in the rational design of such systems, calling for a deep understanding of underlying principles. Herein, we demonstrate an artificial chaperone serves a dual role, that is catalyst in low dosages and inhibitor in high dosages, in regulating the supramolecular polymerization of peptides. Low dosages of carboxymethyl cellulose, as the chaperones, catalyze the assembly of Aβ16‐22 peptides into fibrils through multi‐step phase separation, while high dosages trap the peptides into coacervate intermediates and therefore inhibit the fibrillation. Consequently, the quantity of chaperones does not follow the intuition that ‘more is better’ for catalyzing assembly but instead has an optimal molar ratio. Investigation reveals that the interplay and evolution of electrostatic and hydrophobic interactions are the keys to achieving these processes. This study provides insights into the multifaceted roles artificial chaperones may play in a dosage‐dependent manner and enriches the toolkit for efficient and controllable construction of complex assembly systems.

The rapid development of science and technology, as well as the trend toward intelligence, has resulted in a surge in the demand for excellent energy storage devices for smart devices. At present, the most widely used lithium ion batteries have limited further development because of their high cost and various disadvantages of electrolytes (flammable and toxic). In spite of the low costs and safety of aqueous zinc-ion batteries, the high radius of the zinc ions and strong electrostatic interactions make cathode materials without excellent performance difficult to find. Various oxidation states, structural diversity, excellent multiplicity performance and high capacity have attracted researchers to vanadium-based oxides in recent years. However, vanadium-based oxides have disadvantages such as poor structural stability and low electrical conductivity, so this paper summarizes the progress of research on vanadium oxide cathode materials such as V2O5, VO2, V2O3, etc., and provides an outlook on future development directions.

Green synthesis of metal nanoparticles (NPs) has received extensive attention over other conventional approaches due to their non-toxic nature and more biocompatibility. Herein we report gold and silver NPs (AuNPs@AV and AgNPs@AV) prepared by employing a green approach using crude extract of Aconitum violaceum Jacquem. ex Stapf. Abstract Taking abundant and sustainable solar energy as the only energy source, solar‐powered interface evaporation has been regarded as a promising method to alleviate the pressure of freshwater shortage. However, the uptake of clean water from brine is constantly accompanied by evaporation of water and condensation of vapor, which inevitably generates salt solid, preventing further continuous and stable evaporation. The most direct method is to fabricate a photothermal material with salt self‐resistance by using the reflux of salt ions. Here, a superhydrophilic interconnected biomass carbon absorber (SBCA) is prepared by freeze‐drying and carbonization, realizing strong liquid pumping, and self‐blocking salt. In combination with superior broadband light absorption (94.91%), high porosity (95.9%), superhydrophilicity, and excellent thermal localization, an evaporation device with excellent evaporation rate (2.45 kg m−2 h−1 under 1 kW m−2) is successfully proposed. In the meantime, the porous skeleton and rapid water transport can enhance the diffusion of salt ions and slow down the rate of salt deposition. As a result, no salt deposition is found on the SBCA surface after continuous irradiation at 1 kW m−2 for 15 h. The design can provide a convenient and low‐cost efficient strategy for solar steam generators to address clean water acquisition.

BackgroundFAS-associated death structural domain (FADD) proteins are important proteins that regulate apoptosis and are also involved in many nonapoptotic pathways in tumors. However, how dysregulated FADD affects the development of lung adenocarcinoma (LUAD) remains unknown.MethodTranscriptome profiles and corresponding clinical information of LUAD patients were convened from different databases, and the results were validated by qRT−PCR and cell counting kit-8 using LUAD cell lines. Potential associations between FADD and tumor malignancy, the immune microenvironment, genomic stability, and treatment sensitivity in LUAD patients were revealed by systematic bioinformatics analysis.ResultsFADD was significantly overexpressed in LUAD, and patients with higher expression levels of FADD had a worse prognosis and more advanced tumor stage. Functional analysis revealed that elevated expression of FADD was associated with cell cycle dysregulation, angiogenesis, and metabolic disturbances. In addition, overexpression of FADD was associated with a higher infiltration of suppressive immune cells. From a single-cell perspective, cells with lower FADD expression are more active in immune-related pathways. FADD was associated with more genomic mutations, especially TP53. Patients with high FADD expression are more likely to benefit from conventional chemotherapy, while those with low FADD expression are more suitable for immunotherapy.ConclusionsUpregulated FADD is associated with worse prognosis, immune exhaustion, and tumor malignancy in LUAD patients. In addition, FADD can be an efficient indicator for assessing sensitivity to chemotherapy and immunotherapy. Therefore, FADD has the potential to serve as a new target for precision medicine and targeted therapy for LUAD.

The porous TiCO ceramic was synthesized through a one-step sintering method, utilizing phenolic resin, TiO2 powder, and KCl foaming agent as raw materials. Ni(NO3)2·6H2O was incorporated as a catalyst to facilitate the carbothermal reaction between the pyrolytic carbon and TiO2 powder. The influence of Ni(NO3)2·6H2O catalyst content (0, 5, 10 wt.% of the TiO2 powder) on the microstructure, compressive strength, and thermal conductivity of the resultant porous TiCO ceramic was examined. X-ray diffraction and X-ray photoelectron spectroscopy results confirmed the formation of TiC and TiO in all samples, with an increase in the peak of TiC and a decrease in that of TiO as the Ni(NO3)2·6H2O content increased from 0% to 10%. Scanning electron microscopy results demonstrated a morphological change in the pore wall, transforming from a honeycomb-like porous structure composed of well-dispersed carbon and TiC-TiO particles to rod-shaped TiC whiskers, interconnected with each other as the catalyst content increased from 0% to 10%. Mercury intrusion porosimetry results proved a dual modal pore-size distribution of the samples, comprising nano-scale pores and micro-scale pores. The micro-scale pore size of the samples minorly changed, while the nano-scale pore size escalated from 52 nm to 138 nm as the catalyst content increased from 0 to 10%. The morphology of the pore wall and nano-scale pore size primarily influenced the compressive strength and thermal conductivity of the samples by affecting the load-bearing capability and solid heat-transfer conduction path, respectively.

Abstract We first present a new definition for some form of exponential stability of solutions, including H-exponential stability, H-exponentially asymptotic stability, H-uniformly exponential stability, and H-uniformly exponentially asymptotic stability for a class of set dynamic equations on time scales. Employing Lyapunov-like functions on time scales, we provide the sufficient conditions for the exponential stability of the trivial solution for such set dynamic equations.

Abstract Traumatic brain injury (TBI) contributes to the key causative elements of neurological deficits. However, no effective therapeutics have been developed yet. In our previous work, extracellular vesicles (EVs) secreted by stem cells from human exfoliated deciduous teeth (SHED) offered new insights as potential strategies for functional recovery of TBI. The current study aims to elucidate the mechanism of action, providing novel therapeutic targets for future clinical interventions. With the miRNA array performed and Real-time PCR validated, we revealed the crucial function of miR-330-5p transferred by SHED-derived EVs (SHED-EVs) in regulating microglia, the critical immune modulator in central nervous system. MiR-330-5p targeted Ehmt2 and mediated the transcription of CXCL14 to promote M2 microglia polarization and inhibit M1 polarization. Identified in our in vivo data, SHED-EVs and their effector miR-330-5p alleviated the secretion of inflammatory cytokines and resumed the motor functional recovery of TBI rats. In summary, by transferring miR-330-5p, SHED-EVs favored anti-inflammatory microglia polarization through Ehmt2 mediated CXCL14 transcription in treating traumatic brain injury.

Cardio-cerebrovascular disease, related to high mortality and morbidity worldwide, is a type of cardiovascular or cerebrovascular dysfunction involved in various processes. Therefore, it is imperative to conduct additional research into the pathogenesis and new therapeutic targets of cardiovascular and cerebrovascular disorders. Long non-coding RNAs (lncRNAs) have multiple functions and are involved in nearly all cellular biological processes, including translation, transcription, signal transduction, and cell cycle control. LncR-Meg3 is one of them and is becoming increasingly popular. By binding proteins or directly or competitively binding miRNAs, LncR-Meg3 is involved in apoptosis, inflammation, oxidative stress, endoplasmic reticulum stress, epithelial-mesenchymal transition, and other processes. Recent research has shown that LncR-Meg3 is associated with acute myocardial infarction and can be used to diagnose this condition. This article examines the current state of knowledge regarding the expression and regulatory function of LncR-Meg3 in relation to cardiovascular and cerebrovascular diseases. The abnormal expression of LncR-Meg3 can influence neuronal cell death, inflammation, apoptosis, smooth muscle cell proliferation, etc., thereby aggravating or promoting the disease. In addition, we review the bioactive components that target lncR-Meg3 and propose some potential delivery vectors. A comprehensive and in-depth analysis of LncR-Meg3’s role in cardiovascular disease suggests that targeting LncR-Meg3 may be an alternative therapy in the near future, providing new options for slowing the progression of cardiovascular disease.

High serum uric acid levels, known as hyperuricemia (HUA), are associated with an increased risk of developing gout, chronic kidney disease, cardiovascular disease, diabetes, and other metabolic syndromes. In this study, a promising bacterial strain capable of biodegrading uric acid (UA) was successfully isolated from Baijiu cellar mud using UA as the sole carbon and energy source. The bacterial strain was identified as Bacillus paramycoides-YC02 through 16S rDNA sequence analysis. Under optimal culture conditions at an initial pH of 7.0 and 38 °C, YC02 completely biodegraded an initial UA concentration of 500 mg/L within 48 h. Furthermore, cell-free extracts of YC02 were found to catalyze and remove UA. These results demonstrate the strong biodegradation ability of YC02 toward UA. To gain further insight into the mechanisms underlying UA biodegradation by YC02, the draft genome of YC02 was sequenced using Illumina HiSeq. Subsequent analysis revealed the presence of gene1779 and gene2008, which encode for riboflavin kinase, flavin mononucleotide adenylyl transferase, and flavin adenine dinucleotide (FAD)-dependent urate hydroxylase. This annotation was based on GO or the KEEG database. These enzymes play a crucial role in the metabolism pathway, converting vitamin B2 to FAD and subsequently converting UA to 5-hydroxyisourate (HIU) with the assistance of FAD. Notably, HIU undergoes a slow non-enzymatic breakdown into 2-oxo-4-hydroxy-4-carboxy-5-ureidoimidazoline (OHCU) and (S)-allantoin. The findings of this study provide valuable insights into the metabolism pathway of UA biodegradation by B. paramycoides-YC02 and offer a potential avenue for the development of bacterioactive drugs against HUA and gout.

Abstract Background Sepsis may be accompanied by acute respiratory distress syndrome (ARDS) in patients admitted to intensive care units (ICUs). It is essential to identify prognostic biomarkers in patients with sepsis and ARDS. Objective Determine whether changes in the level of serum fibroblast growth factor 21 (FGF21) can predict the 28-day mortality of ICU patients with sepsis and ARDS. Methods Consecutive sepsis patients were divided into two groups (Sepsis + ARDS and Sepsis-only), and the Sepsis + ARDS group was further classified as survivors or non-survivors. Demographic data and comorbidities were recorded. The Sequential Organ Failure Assessment (SOFA) score and serum levels of cytokines and other biomarkers were recorded 3 times after admission. Abstract The activation of DNA‐dependent kinase (DNA‐PKcs) upon DNA damage contains a cascade of reactions, covering acetylation by TIP60, binding with Ku70/80, and autophosphorylation. However, how cells regulate TIP60‐mediated acetylation of DNA‐PKcs and the following DNA‐PKcs activation upon DNA damage remains obscure. This present study reported that TIP60 is hyper‐SUMOylated in normal conditions, but upon irradiation‐induced DNA damage, small ubiquitin‐like modifier (SUMO)‐specific protease 3 (SENP3)‐mediated deSUMOylation of TIP60 promoted its interaction with DNA‐PKcs to form the TIP60‐DNA‐PKcs complex. We show that TIP60 SUMOylation is reduced quickly in response to DNA damage and the deSUMOylation of TIP60 by SENP3 is required for DNA‐PKcs acetylation and its autophosphorylation. Comet and γH2AX immunofluorescence assay showed that knockdown of SENP3 impaired DNA damage repair. Using the NHEJ report system, we found that knockdown of SENP3 affected the efficiency of NHEJ. Further exploration using clonogenic survival assay, cell viability assay and cytoflow assay suggested that leaking SENP3 increased the sensitivity of tumour cells to serval DNA damage treatment. Overall, our findings revealed a previously unidentified role of SENP3 in regulating DNA‐PKcs activity and DNA damage repair.

Aconitine is one of the main bioactive and toxic ingredients of Aconitum species. Increasingly, aconitine has been reported to induce neurotoxicity. However, whether aconitine has effects on the dopaminergic nervous system remains unclear. In this study, zebrafish embryos at 6-days postfertilization were exposed to aconitine at doses of 0.5, 1, and 2 μM for 24 h, and SH-SY5Y cells were treated with 50, 100, and 200 μM of aconitine for 24 h. Results demonstrated that aconitine treatment induced deformities and enhanced the swimming behavior of zebrafish larvaes. Aconitine exposure suppressed cell proliferation and increased the number of reactive oxygen species and apoptosis in zebrafish larvaes and SH-SY5Y cells. Aconitine altered the levels of dopamine and its metabolites by regulating the expression of genes and proteins related to dopamine synthesis, storage, degradation, and reuptake in vivo and in vitro. Moreover, aconitine activated the AC/cAMP/PKA pathway by activating the dopamine D1 receptor (D1R) and inhibiting the dopamine D2 receptor (D2R) to disturb intracellular calcium homeostasis, eventually leading to the damage of nerve cells. Furthermore, the D1R antagonist SCH23390 and D2R agonist sumanirole pretreatment effectively attenuated the excitatory state of larvaes. Sumanirole and PKA antagonist H-89 pretreatment effectively decreased intracellular Ca2+ accumulation induced by aconitine in vivo. SCH23390 and sumanirole also reduced aconitine-induced cytotoxicity by inhibiting the AC/cAMP/PKA pathway in vitro. These results suggested that dopamine homeostasis imbalance and dopamine receptors (DRs)-mediated AC/cAMP/PKA pathway activation might be vital mechanisms underlying aconitine-induced neurological injury.

Linker histone H1.2, which belongs to the linker histone family H1, plays a crucial role in the maintenance of the stable higher-order structures of chromatin and nucleosomes. As a critical part of chromatin structure, H1.2 has an important function in regulating chromatin dynamics and participates in multiple other cellular processes as well. Recent work has also shown that linker histone H1.2 regulates the transcription levels of certain target genes and affects different processes as well, such as cancer cell growth and migration, DNA duplication and DNA repair. The present work briefly summarizes the current knowledge of linker histone H1.2 modifications. Further, we also discuss the roles of linker histone H1.2 in the maintenance of genome stability, apoptosis, cell cycle regulation, and its association with disease.

Cancer driver gene is a type of gene with abnormal alterations that initiate or promote tumorigenesis. Driver genes can be used to reveal the fundamental pathological mechanisms of tumorigenesis. These genes may have pathological changes at different omics levels. Thus, identifying cancer driver genes involving two or more omics levels is essential. In this study, a computational investigation was conducted on lung cancer driver genes. Four omics levels, namely, epigenomics, genomics, transcriptomics, and post-transcriptomics, were involved. From the driver genes at each level, the Laplacian heat diffusion algorithm was executed on a protein–protein interaction network for discovering latent driver genes at this level. A following screen procedure was performed to extract essential driver genes, which contained three tests: permutation, association, and function tests, which can exclude false-positive genes and screen essential ones. Finally, the intersection operation was performed to obtain novel driver genes involving two omic levels. The analyses on obtained genes indicated that they were associated with fundamental pathological mechanisms of lung cancer at two corresponding omics levels.

Understanding nutrient quantity and quality in forest soils is important for sustainable management of forest resources and maintaining forest ecosystem services. In this study, six soil nutrient indicators, including soil organic matter (SOM), total nitrogen (TN), total phosphorus (TP), available nitrogen (AN), available phosphorus (AP), and available potassium (AK) were measured in five different aged stands of Chinese fir forests in subtropical China. A structural equation model (SEM) was developed based on these soil nutrients indicators in order to better evaluate the soil fertility index (SFI) in these studied forests. The results show that soil nutrient contents changed with the soil depth in different age groups. The SOM decreased in a specific order: over mature > mature > near mature > middle > young stands. The TN content of the soil gradually decreased with increased soil depth throughout all age groups. The SEM indicated that the TN had the highest weight of 0.4154, while the TP had the lowest weight at 0.1991 for estimating the SFI. The weights of other indicators (AN, SOM, AP, and AK) ranged 0.2138–0.3855 in our study. The established SEM satisfied the fitness reference values and was able to accurately describe the forest soil nutrient status through the SFI. The overall SFI values were significantly higher in over mature stands than in young-aged stands and in topsoil than in deeper soil in all examined forests. Soil TN, AP, and AK were the most important nutrient indicators to the evaluation of the SFI in the study sites. The results confirmed that the SEM was suitable to estimate the weights of the SFI and better describe the soil nutrient status in forests. Our research provides an innovative approach to assess a soil nutrient status and soil fertility and provides a scientific basis for accurate implementation of soil nutrient assessment in forest ecosystems.

Chinese materia medica (CMM) has been applied for the prevention and treatment of diseases for thousands of years. However, arrhythmia, myocardial ischemia, heart failure, and other cardiac adverse reactions during CMM application were gradually reported. CMM-induced cardiotoxicity has aroused widespread attention. Our review aimed to summarize the risk compounds, preclinical toxicity evaluation, and potential mechanisms of CMM-induced cardiotoxicity. All relevant articles published on the PubMed, Embase, and China National Knowledge Infrastructure (CNKI) databases for the latest twenty years were searched and manually extracted. The risk substances of CMM-induced cardiotoxicity are relatively complex. A single CMM usually contains various risk compounds, and the same risk substance may exist in various CMM. The active and risk substances in CMM may be transformed into each other under different conditions, such as drug dosage, medication methods, and body status. Generally, the risk compounds of CMM-induced cardiotoxicity can be classified into alkaloids, terpenoids, steroids, heavy metals, organic acids, toxic proteins, and peptides. Traditional evaluation methods of chemical drug-induced cardiotoxicity primarily include cardiac function monitoring, endomyocardial biopsy, myocardial zymogram, and biomarker determination. In the preclinical stage, CMM-induced cardiotoxicity should be systematically evaluated at the overall, tissue, cellular, and molecular levels, including cardiac function, histopathology, cytology, myocardial zymogram, and biomarkers. Thanks to the development of systematic biology, the higher specificity and sensitivity of biomarkers, such as genes, proteins, and metabolic small molecules, are gradually applied for evaluating CMM-induced cardiotoxicity. Previous studies on the mechanisms of CMM-induced cardiotoxicity focused on a single drug, monomer or components of CMM. The interaction among ion homeostasis (sodium, potassium, and calcium ions), oxidative damage, mitochondrial injury, apoptosis and autophagy, and metabolic disturbance is involved in CMM-induced cardiotoxicity. Clarification on the risk compounds, preclinical toxicity evaluation, and potential mechanisms of CMM-induced cardiotoxicity must be beneficial to guide new CMM development and post-marketed CMM reevaluation.

A simple and efficient synthetic route was developed to obtain composites by mechanically ball-milling Li2ZrO3 (LZO) with Li3V2(PO4)3/C (LVPC). LZO improves the ionic conductivity of LVPC, compensates for the transfer of Li+ between the LVPC and the electrolyte, reduces the impedance of LVPC, and protects LVPC from side reactions caused by direct contact with the electrolyte. The composite consisting of 4.0 wt% of LZO exhibited a considerable electrochemical performance. The 1st discharge capacity reached 192 mAh g−1 at 30 mA g−1 between 3.0 and 4.8 V, and remained at 153 mAh g−1 after 50 cycles. The EIS analysis showed that the charge transfer and diffusion of Li+ in the LZO-LVPC composites were more favorable than those of pristine LVPC. The synthetic method proposed in this work is simple and efficient, which is convenient for large-scale production.

Compared with other materials, monoclinic Li1+xV3O8 as the promising cathode includes many strong points, such as high theoretical capacity, abundant raw materials, safety characteristic and low environmental impact. However, the primary problems facing the development of Li1+xV3O8 for energy storage devices are poor cycling performance, low rate capability and short service life. Recently, many researches have taken various measures to overcome these shortcomings. In this mini-review, we summarized the recent developments of Li1+xV3O8 and focus on the improving of Li1+xV3O8 by ions-doping, which is deemed to a significant manner to improve the electrochemical performances of Li1+xV3O8.

RNAs can mimic the information storage, replication and catalysis functions of DNAs and proteins, providing physical evidence for the widely accepted RNA world hypothesis that RNA alone constitutes earlier life forms. However, the currently found molecular chaperones for correcting RNA folding are proteins, which poses a question on whether RNA itself can avoid misfolding in the absence of proteins. Here, we report two examples that RNA/ssDNA strands can serve as molecular chaperones to assist the folding of RNA/ssDNA structures with complex intramolecular interactions. We found that these RNA chaperones can disrupt the misfolded double-hairpin structures, assisting them to refold into native single-hairpin structures, which is a typical catalytical assembly called catassembly. The refolding rate was increased by at least three orders of magnitude. The identification that RNA can function as molecular chaperone for RNA folding may expand our understanding on RNA molecular chaperone and provides clues to rational design of catassembler.

The sergeants-and-soldiers effect, in which a few chiral units induce chirality in a large number of achiral molecules, is difficult to quantify at the molecular level. Here, the authors devise an elegant strategy—combining theory and a system of pure organic polyhedra with chiral and achiral vertices—to understand the mechanism of chiral amplification in discrete molecular assemblies.

A class of linear impulsive fuzzy dynamic equations on time scales is considered by using the generalized differentiability concept on time scales. Some novel criteria and general forms of solutions are established for such models whose significance lies in proposing the possibility to get unifying forms of solutions for discrete and continuous dynamical systems under uncertainty and to unify corresponding problems in the framework of fuzzy dynamic equations on time scales. Finally, some examples show the applicability of our results.

Abstract Different dimensions of carbon materials with various features have captured numerous interests due to their applications on the tremendous fields. Restricted by the raw materials and devices, the controlling of their morphology is a major challenge. Utilizing the catalytic features of the intermediates from the low‐cost salts and polymerization of 0D carbon quantum dots (CQDs), 0D CQDs are expected to self‐assemble into 1/2/3D carbon structures with the assistance of temperature‐induced intermediates (e.g., ZnO, Ni, and Cu) from the salts (ZnCl2, NiCl2, and CuCl). The formation mechanisms are illustrated as follows: 1) the “orient induction” to evoke “vine style” growth mechanism of ZnO; 2) the “dissolution–precipitation” of Ni; and 3) the “surface adsorption self‐limited” of Cu. Subsequently, the degree of graphitization, interlayer distance, and special surface area are investigated in detail. 1D structure from 700 °C as anode displays a high Na‐storage capacity of 301.2 mAh g−1 at 0.1 A g−1 after 200 cycles and 107 mAh g−1 at 5.0 A g−1 after 5000 cycles. Quantitative kinetics analysis confirms the fundamentals of the enhanced rate capacity and the potential region of Na‐insertion/extraction. This elaborate work opens up an avenue toward the design of carbon with multidimensions and in‐depth understanding of their sodium‐storage features.

A facile method has been used to synthesize NaV3O8 powders with oxalic acid as the complexing agent. This soft-synthesis technique can effectively reduce the calcination temperature for synthesizing NaV3O8 powders to 300 °C, which is much lower than that in the solid-state synthesis. The thus-synthesized NaV3O8 powders are characterized by XRD, SEM and galvanostatic charge-discharge test. Among the as-prepared powders, the NaV3O8 powder obtained at 350 °C exhibits morphology of porous particles. Liquid chromatography, coupled with tandem mass spectrometry, has become one of the most popular methods for the analysis of post‐transcriptionally modified transfer ribonucleic acids (tRNAs). Given that the information collected using this platform is entirely determined by the mass of the analyte, it has proven to be the gold standard for accurately assigning nucleobases to the sequence. For the past few decades many labs have worked to improve the analysis, contiguous to instrumentation manufacturers developing faster and more sensitive instruments. With biological discoveries relating to ribonucleic acid happening more frequently, mass spectrometry has been invaluable in helping to understand what is happening at the molecular level. Here we present a brief overview of the methods that have been developed and refined for the analysis of modified tRNAs by liquid chromatography tandem mass spectrometry.

Li3V2(PO4)3−xBrx/carbon (x = 0.08, 0.14, 0.20, and 0.26) composites as cathode materials for lithium-ion batteries were prepared through partially substituting PO43− with Br−, via a rheological phase reaction method. The crystal structure and morphology of the as-prepared composites were characterized by X-ray diffraction (XRD) and scanning electron microscopy (SEM), and electrochemical properties were evaluated by charge/discharge cycling and electrochemical impedance spectroscopy (EIS). XRD results reveal that the Li3V2(PO4)3−xBrx/carbon composites with solid solution phase are well crystallized and have the same monoclinic structure as the pristine Li3V2(PO4)3/carbon composite. It is indicated by SEM images that the Li3V2(PO4)3−xBrx/carbon composites possess large and irregular particles, with an increasing Br− content. Among the Li3V2(PO4)3−xBrx/carbon composites, the Li3V2(PO4)2.86Br0.14/carbon composite shows the highest initial discharge capacity of 178.33 mAh·g−1 at the current rate of 30 mA·g−1 in the voltage range of 4.8–3.0 V, and the discharge capacity of 139.66 mAh·g−1 remains after 100 charge/discharge cycles. Even if operated at the current rate of 90 mA·g−1, Li3V2(PO4)2.86Br0.14/carbon composite still releases the initial discharge capacity of 156.57 mAh·g−1, and the discharge capacity of 123.3 mAh·g−1 can be maintained after the same number of cycles, which is beyond the discharge capacity and cycleability of the pristine Li3V2(PO4)3/carbon composite. EIS results imply that the Li3V2(PO4)2.86Br0.14/carbon composite demonstrates a decreased charge transfer resistance and preserves a good interfacial compatibility between solid electrode and electrolyte solution, compared with the pristine Li3V2(PO4)3/carbon composite upon cycling.

Asset management of distribution systems is an important issue for smart grid. Maintenance scheduling, as an important part of asset management, affects the reliability of distribution equipment and power supply. This research focuses on long-term distribution system maintenance scheduling aided by available operation information, which is a prominent advantage of smart grid over conventional distribution systems. In this paper, the historical and future operation information in smart grid is taken into account through a decoupled time-varying reliability model of equipment. Based on distribution system reliability assessment, a maintenance scheduling model is proposed to determine the optimal implementation time of maintenance activities to minimize distribution systems’ total cost, while satisfying reliability requirements. A combined algorithm that consists of particle swarm optimization and tabu search is designed and applied to the optimization problem. Numerical result verifies that the proposed method can schedule long-term maintenance of distribution systems in smart grid economically and effectively.
