Bile acids (BAs) are natural metabolites in mammals and have the potential to function as drugs against viral infection. However, the limited understanding of chenodeoxycholic acid (CDCA) receptors and downstream signaling, along with its lower suppression efficiency in inhibiting virus infection limits its clinical application. In this study, we demonstrate that farnesoid X receptor (FXR), the receptor of CDCA, negatively regulates interferon signaling, thereby contributing to the reduced effectiveness of CDCA against virus replication. FXR deficiency or pharmacological inhibition enhances interferon signaling activation to suppress virus infection. Mechanistically, FXR impairs the DNA binding and transcriptional abilities of activated interferon regulatory factor 3 (IRF3) through interaction. Reduced IRF3 transcriptional activity by FXR–IRF3 interaction significantly undermines the expression of Interferon Beta 1 (IFNB1) and the antiviral response of cells, especially upon the CDCA treatment. In FXR-deficient cells, or when combined with Z-guggulsterone (GUGG) treatment, CDCA exhibits a more potent ability to restrict virus infection. Thus, these findings suggest that FXR serves as a limiting factor for CDCA in inhibiting virus replication, which can be attributed to the “signaling-brake” roles of FXR in interferon signaling. Targeting FXR inhibition represents a promising pharmaceutical strategy for the clinical application of BAs metabolites as antiviral drugs.

Heat moisture treatment (HMT) is a widely used method for starch modification due to its safe, green, environmentally friendly, and efficient characteristics. In this study, quinoa starch was modified by HMT with varying moisture contents (20%, 25%, and 30%) at a temperature of 120 °C for 10 hours, and its rheological properties, in vitro digestive properties, and structure were further evaluated. The steady rheology results indicated that all the samples were shear-thinning non-Newtonian fluids. After undergoing HMT, the apparent viscosity of quinoa starch decreased significantly. Additionally, the samples' apparent viscosity decreased as the moisture content of HMT increased. The results of frequency sweep showed that the storage modulus G' of quinoa starch decreased with the increase of moisture content of HMT, while the loss modulus G'', and loss factor increased with the increase of moisture content of HMT. The thixotropic results showed that the thixotropic ring area of quinoa starch decreased with the increase of the moisture content of HMT. Abstract Background Diabetes mellitus is generally accompanied by dyslipidaemia, but inconsistent relationships between lipid profiles and diabetes are noted. Moreover, genetic variations in insertion/deletion (I/D) polymorphisms at angiotensin-converting enzyme gene (ACE) and T/C polymorphisms in the angiotensin type 1 receptor gene (AGTR1) are related to diabetes and lipid levels, but the associations are controversial. Thus, the current research aimed to explore the effects of ACE I/D, AGTR1 rs5182 and diabetes mellitus on serum lipid profiles in 385 Chinese participants with an average age of 75.01 years. Methods The ACE I/D variant was identified using the polymerase chain reaction (PCR) method, whereas the AGTR1 rs5182 polymorphism was identified using the PCR-based restriction fragment length polymorphism (PCR-RFLP) method and verified with DNA sequencing. Total cholesterol (TC), triglyceride (TG), apolipoprotein A (ApoA), apolipoprotein B (ApoB), high-density lipoprotein cholesterol (HDL-C) and low-density lipoprotein cholesterol (LDL-C) levels were measured using routine methods, and the lipid ratios were calculated. Results ACE I/D, but not AGTR1 rs5182, was a predictor of TG/HDL-C for the whole study population. Both ACE I/D and AGTR1 rs5182 were predictors of HDL-C and LDL-C levels in females but not in males. Moreover, in females, diabetes mellitus and ACE I/D were identified as predictors of TG and TG/HDL-C, whereas AGTR1 rs5182 and diabetes mellitus were predictors of TG/HDL-C. Moreover, diabetes mellitus and the combination of ACE I/D and AGTR1 rs5182 variations were predictors of TG and TG/HDL-C exclusively in females. Conclusions The results demonstrated the potential for gender-dependent interactions of ACE I/D, AGTR1 rs5182, and diabetes on lipid profiles. These findings may serve as an additional explanation for the inconsistent changes of blood lipids in individuals with diabetes mellitus, thereby offering a novel perspective for the clinical management of blood lipid levels in diabetic patients.

Abstract Artificial photosynthesis using carbon nitride (g-C3N4) holds a great promise for sustainable and cost-effective H2O2 production, but the high carrier recombination rate impedes its efficiency. To tackle this challenge, we propose an innovative method involving multispecies iodine mediators (I−/I3 −) intercalation through a pre-photo-oxidation process using potassium iodide (suspected deteriorated “KI”) within the g-C3N4 framework. Moreover, we introduce an external electric field by incorporating cationic methyl viologen ions to establish an auxiliary electron transfer channel. Such a unique design drastically improves the separation of photo-generated carriers, achieving an impressive H2O2 production rate of 46.40 mmol g−1 h−1 under visible light irradiation, surpassing the most visible-light H2O2-producing systems. Combining various advanced characterization techniques elucidates the inner photocatalytic mechanism, and the application potential of this photocatalytic system is validated with various simulation scenarios. This work presents a significative strategy for preparing and applying highly efficient g-C3N4-based catalysts in photochemical H2O2 production.

Thermochemical conversions are pathways for biomass utilization to produce various value-added energy and chemical products. For the development of novel thermochemical conversion technologies, an accurate understanding of the reaction performance and kinetics is essential. Given the diversity of the thermal analysis techniques, it is necessary to understand the features and limitations of the reactors, ensuring that the selected thermal analysis reactor meets the specific need for reaction characterization. This paper provides a critical overview of the thermal analysis reactors based on the following perspectives: 1) gas flow conditions in the reactor, 2) particle's external and internal heat and mass transfer limitations, 3) heating rate, 4) temperature distribution, 5) nascent char production and reaction, 6) liquid feeding and atomization, 7) simultaneous sampling and analyzing of bed materials, and 8) reacting atmosphere change. Finally, prospects and future research directions in the development of analysis techniques are proposed.

Abstract Reconstructing ancient transportation networks is critical to studying past human mobility patterns. China’s Haidai region was a thriving political and economic hub during the Bronze and Early Iron Ages. We used GIS spatial analysis techniques to build a “Settlement Interaction Model” based on archaeological data from the Haidai region during the Bronze and Early Iron Age (Shang Dynasty, Western Zhou Dynasty, Spring & Autumn Period, and Warring States period). The eight-level road network maps with traffic attributes were distinguished based on topography and settlement size. The total lengths of the road networks were estimated to be 19,112 km in the Shang Dynasty, 35,269 km in the Western Zhou Dynasty, 51,555 km in the Spring & Autumn Period, and 77,456 km in the Warring States Period, with the average road flows of 6.6, 31.7, 42.8, and 75.5, respectively. Abstract The development of unconventional long-wavelength fluorescent polymer hydrogels without using polycyclic aromatic hydrocarbons or extended π-conjugation is a fundamental challenge in luminescent materials owing to a lack of understanding regarding the spatial interactions induced inherent clustering-triggered emission under water-rich conditions. Inspired by the color change of protein astaxanthin as a result of heat-induced denaturation, we propose a thermodynamically driven strategy to develop red fluorescence (~610 nm) by boiling multiple hydrogen-bonded poly(N-acryloylsemicarbazide) hydrogels in a water bath. We reveal that thermodynamically driven conformational changes of polymer chains from isolated hydrogen bonding donor-acceptor structures to through-space interaction structures induce intrinsic fluorescence shifts from blue to red during clustering-triggered emission. The proposed multiple hydrogen-bonding supramolecular hydrogel shows good fluorescence stability, mechanical robustness, and 3D printability for customizable shaping. We provide a viable method to prepare nonconventional long-wavelength fluorescent hydrogels towards soft fluorescent devices without initially introducing any fluorescent components.

Silicon (Si) is a promising substitute for graphite anode due to the high theoretical specific capacity (4200 mAh g−1). However, too large volume change exists during the lithiation/delithiation process. Composite anode, prepared by mixing Si with graphite, can realize higher specific capacity than graphite and much better cycle performance than Si anode. However, the capacity decay caused by pulverization of Si particles is still a great challenge. Here, a cross-linkable binder rich in nitrile, carboxyl and hydroxyl groups is designed for composite silicon-graphite (Si-C) anode. The nitrile and hydroxyl groups can be in situ cross-linked in the batteries through Ritter reaction. The cross-linked binder has excellent resilience and good adhesion to the active materials and current collector. The cycle performance of the cell with cross-linked binder is much better than the counterpart. Scanning electron microscopy results of the cycled Si-C anode show that the cross-linked binder can suppress the volume expansion and pulverization. Moreover, the investigation with X-ray photoelectronic spectrum and density function theory calculation demonstrate that the decomposition of ester solvent and LiPF6 on Si anode has been mitigated and more stable SEI film is formed on the Si-C anode. Our strategy of in situ cross-linking binder in the batteries has provided a feasible way for designing the next generation of silicon-based anodes with higher specific capacity and longer cycling life.

Glass fiber reinforce polymer (GFRP) bars have better durability than traditional steel bars, but there are few studies on their long-term performance in actual service environments especially the marine environment. To investigate the durability of GFRP bars used in cross-sea bridges, this research delves into the performance of 132 such bars, spanning diameters of 10, 12, 16, and 25 mm, in an environment that closely mimics real-world condition. The GFRP bars, encapsulated in concrete, were submerged in seawater solution, maintained at either room temperature or 60°C, for periods spanning from 0 to 183 days. Then the tensile strength and elastic modulus degradation of GFRP bars were investigated by tensile tests. Scanning electron microscope (SEM) was also used to analyze the micro-degradation mechanism of GFRP bars. In addition, this paper compares the four existing models for predicting long-term mechanical properties of GFRP bars with the experimental data, so as to determine the formulas and methods to be used in the design. The result shows that the elastic modulus of GFRP bars after 183 days of immersion in concrete pore solution was almost unchanged while the tensile strength decreased significantly. Among them, the tensile strength of 10 mm diameter GFRP bars decreased by 41.52 %, after 183 days of immersion at high temperature (60°C). The SEM method revealed that the main cause of such phenomena was the weakening of the bond between the fibers and matrix, which led to a decrease in the interface property. Finally, by fitting and comparing the relationship between the test data and the existing prediction models for the long-term mechanical properties of GFRP bars, the residual tensile strength of GFRP bars used in the actual project of Yanzhou Bridge for 100 years was predicted according to the optimal prediction model, i.e. the Serb model, and the results showed that the residual strength of GFRP bars is able to meet the current standard requirement.

The stimulator of interferon genes (STING), an integral adaptor protein in the DNA-sensing pathway, plays a pivotal role in the innate immune response against infections. Additionally, it presents a valuable therapeutic target for infectious diseases and cancer. We observed that fangchinoline (Fan), a bis-benzylisoquinoline alkaloid (BBA), effectively impedes the replication of vesicular stomatitis virus (VSV), encephalomyocarditis virus (EMCV), influenza A virus (H1N1), and herpes simplex virus-1 (HSV-1) in vitro. Fan treatment significantly reduced the viral load, attenuated tissue inflammation, and improved survival in a viral sepsis mouse model. Mechanistically, Fan activates the antiviral response in a STING-dependent manner, leading to increased expression of interferon (IFN) and interferon-stimulated genes (ISGs) for potent antiviral effects in vivo and in vitro. Notably, Fan interacts with STING, preventing its degradation and thereby extending the activation of IFN-based antiviral responses. Collectively, our findings highlight the potential of Fan, which elicits antiviral immunity by suppressing STING degradation, as a promising candidate for antiviral therapy.

The Pt1-MoS2-def sensors exhibit high SO2 responses and extremely low limit of detection (3.14% to 500 ppb SO2) at room temperature. The Pt1-MoS2-def sensors array can realize real-time monitoring of SO2 for plant growth.

Two monosaccharides, glucose and xylose, were used as glycosyl donors to investigate the effects of reaction time, reaction temperature, protein-to-sugar ratio and protein concentration on the wet glycosylation reaction of soybean protein isolate. The allergenicity of SPI-glucose derivative (SPI-glucose) and SPI-xylose derivative (SPI-xylose) was analyzed. SPI-glucose and SPI-xylose were used to produce the hypo-allergenic Chiba tofu. The results showed that xylose was more likely to glycosylate with SPI than glucose, but it was also more likely to produce melanoidin. The highest degree of glycosylation reaction was observed at the protein concentration of 25 mg/mL. The degree of glycosylation reaction was positively correlated with the reaction temperature, reaction time, and the amount of sugar added. The results of SDS-PAGE and fourier transform infrared spectroscopy showed that glycosylation caused covalent binding of SPI and sugar molecules, and the free amino group content of SPI was reduced. The degree of glycosylation reaction was negatively correlated with the allergenicity of SPI, and the allergenicity of SPI-xylose could be reduced by up to 50%. Chiba tofu made from SPI-xylose using glutamine aminotransferase (TGase) had good elasticity and chewiness.

Abstract Rolling bearings working in oscillating applications often suffer from false brinelling and fretting corrosion that limit their service life. At present, the study of oscillation-induced failures is primarily dominated by a large number of sliding and bench tests. However, most of the previous work are not able to fully describe the two closely related but different failure mechanisms. In this study, the origin and evolution of the two failures are thoroughly investigated under strictly controlled oscillating rolling conditions. The results show that failure evolves from false brinelling to fretting corrosion under dry friction conditions regardless of the oscillation amplitude. This process is accompanied by a large area peeling and serious oxidation, and the oxygen content increases from 2.50 wt% to 21.22 wt%. Oxidation is a crucial factor in the evolution of false brinelling into fretting corrosion. However, the distribution of microwear and oxygen content depend on the oscillation amplitude. Under lubrication conditions, the friction coefficient (COF) is smaller in the initial stage and led to local slip or transient instability of rolling elements. Therefore, the size of the wear marks and COF increased and appeared to be larger than those under a dry friction condition. However, grease can separate the contact surfaces, only surface deformation owing to false brinelling and slight damage at the roughness level occurred. The evolution of false brinelling and fretting corrosion is also closely related to residual stress. False brinelling often occurrs with residual stress accomulation, meanwhile, the residual stress increases from 120 to 300 MPa. When peeling occurs, residual stress is released. The failure pattern transformed from false brinelling to fretting corrosion and is accompanied by oxidation. So far, none of investigation is able to show satisfactory evolution of oscillating-induced failures, this study may contribute to more scientific understanding of rolling bearings against long-run reciprocating oscillating wear.

Modeling the interactive behavior of human drivers is essential for achieving safe and fully autonomous vehicles. Unfortunately, most decision-making systems employed in current autonomous vehicles rely on complex deep neural network models that function as black boxes with opaque reasoning that hampers human interpretation. Drawing upon the needs theories endorsed by psychologists and driving-related psychological research, we summarize five fundamental driving needs underlying the driver’s behavior: safety, dominance, achievement, order, and relatedness. Leveraging the behavior selection module from general cognitive architectures, we propose a decision-making model explicitly tailored for autonomous vehicles, comprising three distinct modules: needs assessment, motivation generation, and behavior selection. We conducted experiments to evaluate the proposed model using a self-developed 2D simulator based on Unity. The results intuitively visualized the motivation and behavior of self-driving vehicles. This model demonstrates remarkable proficiency in handling routine tasks, such as independent and complete driving tasks, intersection navigation, and maneuvering among multiple vehicles.

The study aims to explore an effective approach to improve the sensory quality and consumer satisfaction of cookies in the food industry. L. reuteri and L. rhamnosus were chosen to ferment egg yolk and their effects on dough properties and physicochemical properties, flavor, texture, color, and sensory acceptability of cookies were studied. Results show that the utilization of fermented egg yolk significantly decreased baking loss and increased spread factor of cookies. GC–MS analysis indicates different Lactobacillus species enhanced cookie flavor through unique mechanisms. Texture analysis shows cookies prepared with L. rhamnosus-fermented egg yolk had significantly lower hardness (1807.12 g) than control cookies (2028.34 g). Sensory evaluation reveals the L. reuteri-fermented egg yolk significantly improved the overall acceptability of cookies by enhancing appearance, flavor, and mouthfeel scores. These findings have practical implications for food manufacturers seeking to enhance their product's quality and appeal, thereby gaining a competitive edge in the market.

Abstract Chromobox protein homolog 2 (CBX2) exerts a multifaceted impact on the progression of aggressive cancers. The proteasome-dependent pathway is crucial for modulating CBX2 regulation, while the specific regulatory roles and mechanisms of deubiquitinating enzymes targeting CBX2 remain poorly understood. Mass spectrometry analysis identified ubiquitin-specific peptidase 27X (USP27X) as a deubiquitinating enzyme that targets CBX2. Overexpression of USP27X significantly enhances CBX2 levels by promoting deubiquitination, while deficiency of USP27X leads to CBX2 degradation, thereby inhibiting tumorigenesis. Furthermore, it has been revealed that glycogen synthase kinase 3 beta (GSK3β) can directly bind to and phosphorylate USP27X, thereby enhancing the interaction between USP27X and CBX2 and leading to further stabilization of the CBX2 protein. Clinically, the co-expression of high levels of USP27X and CBX2 in breast cancer tissues is indicative of a poor prognosis for patients with this disease. These findings collectively underscore the critical regulatory role played by USP27X in modulating CBX2, thereby establishing the GSK3β-USP27X-CBX2 axis as a pivotal driver of malignant progression in breast cancer.

Abstract Background Platycodon grandiflorus (Jacq.) A. DC is a famous traditional Chinese medicine in China and an authentic medicine in Inner Mongolia. It has been traditionally used as an expectorant in cough and also has anti-inflammatory and other pharmacological effects. As a homologous plant of medicine and food, P. grandiflorus is widely planted in Northeast China. Soil salinity isa limiting factor for its cultivation. In this study, we comprehensively described the physiological characteristics of P. grandiflorus and combined transcriptomics and metabolomics to study the response of roots of P. grandiflorus to salt stress. Results Overall, 8,988 differentially expressed genes were activated and significantly altered the metabolic processes. In total, 428 differentially abundant metabolites were affected by salt stress. After moderate and severe salt stress, most of the differentially abundant metabolites were enriched in the L-phenylalanine metabolic pathway. Through the comprehensive analysis of the interaction between key genes and metabolites, the main pathways such as lignin compound biosynthesis and triterpene saponin biosynthesis were completed. The relative content of compounds related to lignin biosynthesis, such as caffeic acid, coniferin, and syringing, increased under salt stress, and the related genes such as PAL, C4H, and the key enzyme gene UGT72E2 were activated to adapt to the salt stress. Platycodon saponin is one of the major triterpene saponins in P. grandiflorus, and Platycodin D is its most abundant major bioactive component. Under severe salt stress, Platycodin D level increased by nearly 1.77-fold compared with the control group. Most of the genes involved insynthetic pathway of Platycodin D, such as HMGCR, GGPS, SE, and LUP, were upregulated under salt stress. Conclusion Salt stress led to a decrease in the biomass and affected the activities of antioxidant enzymes and contents of osmotic regulators in the plant. These results provided not only novel insights into the underlying mechanisms of response of P. grandiflorus to salt stress but also a foundation for future studies on the function of genes related to salt tolerance in the triterpenoid saponin biosynthesis pathway.

Abstract Retinal ischemia‒reperfusion (I/R) injury can cause significant damage to human retinal neurons, greatly compromising their functions. Existing interventions have been proven to have little effect. Ferroptosis is a newly discovered type of programmed cell death that has been found to be involved in the process of ischemia‒reperfusion in multiple organs throughout the body. Studies have shown that it is also present in retinal ischemia‒reperfusion injury. A rat model of retinal ischemia‒reperfusion injury was constructed and treated with deferoxamine. In this study, we found the accumulation of Fe2+, reactive oxygen species (ROS), malondialdehyde (MDA), and the consumption of glutathione (GSH) via ELISA testing; increased expression of transferrin; and decreased expression of ferritin, SLC7A11, and GPX4 via Western blotting (WB) and real-time PCR testing. Structural signs of ferroptosis (mitochondrial shrinkage) were observed across multiple cell types, including retinal ganglion cells (RGCs), photoreceptor cells, and pigment epithelial cells. Changes in visual function were detected by F-VEP and ERG. The results showed that iron and oxidative stress were increased in the retinal ischemia‒reperfusion injury model, resulting in ferroptosis and tissue damage. Deferoxamine protects the structural and functional soundness of the retina by inhibiting ferroptosis through the simultaneous inhibition of hemochromatosis, the initiation of transferrin, and the degradation of ferritin and activating the antioxidant capacity of the System Xc-GSH-GPX4 pathway.

A hydrophobic and ultralight cellulose aerogel (CA) was reinforced by polyethyleneimine (PEI) and functionalized by methyltrimethoxysilane (MTMS). Adding PEI improved the mechanical strength and the elastic resilience of the resulting material due to the flexibility enhancement of the cellulose chains, which prevented the collapse of the pore structure and contributed to the uniform pore size distribution. The hydrophobic property of the aerogels with the functionalization of MTMS was improved, which can prevent the pore structure from collapsing due to the absorption of water. The maximum compression modulus of aerogel reached 1.1 MPa at the strain of 80%, and its hydrophobic water contact angle was up to 112°. The hydrophobic composite aerogels exhibited ultrahigh efficiency in sound absorption across a wide frequency range from 500 to 6300 Hz, and their average absorption coefficient was greater than 0.74. The light weight, high porosity, and environmentally friendly aerogels presented in this work are promising for efficient sound absorption. They have potential applications in noise pollution treatment.

Abstract Immune cell infiltration in response to myocyte death regulates extracellular matrix remodeling and scar formation after myocardial infarction (MI). Caspase-recruitment domain family member 9 (CARD9) acts as an adapter that mediates the transduction of pro-inflammatory signaling cascades in innate immunity; however, its role in cardiac injury and repair post-MI remains unclear. We found that Card9 was one of the most upregulated Card genes in the ischemic myocardium of mice. CARD9 expression increased considerably 1 day post-MI and declined by day 7 post-MI. Moreover, CARD9 was mainly expressed in F4/80-positive macrophages. Card9 knockout (KO) led to left ventricular function improvement and infarct scar size reduction in mice 28 days post-MI. Additionally, Card9 KO suppressed cardiomyocyte apoptosis in the border region and attenuated matrix metalloproteinase (MMP) expression. RNA sequencing revealed that Card9 KO significantly suppressed lipocalin 2 (Lcn2) expression post-MI. Both LCN2 and the receptor solute carrier family 22 member 17 (SL22A17) were detected in macrophages. Subsequently, we demonstrated that Card9 overexpression increased LCN2 expression, while Card9 KO inhibited necrotic cell-induced LCN2 upregulation in macrophages, likely through NF-κB. Lcn2 KO showed beneficial effects post-MI, and recombinant LCN2 diminished the protective effects of Card9 KO in vivo. Lcn2 KO reduced MMP9 post-MI, and Lcn2 overexpression increased Mmp9 expression in macrophages. Slc22a17 knockdown in macrophages reduced MMP9 release with recombinant LCN2 treatment. In conclusion, our results demonstrate that macrophage CARD9 mediates the deterioration of cardiac function and adverse remodeling post-MI via LCN2.

Background/purpose: Cancer is an important part of the global burden of childhood diseases. Head and neck carcinoma in children is rare and related research is limited. This study aimed to investigate the clinicopathological features of childhood head and neck carcinoma. Materials and methods: Forty-two cases of childhood head and neck carcinoma treated in our institution were reviewed and analyzed. Results: Median age overall was 11 years. Twenty-three patients (54.8%) were male and 19 (45.2%) were female. Parotid gland location was most common (54.8%). Mucoepidermoid carcinoma and squamous cell carcinoma were the most common histological types (57.1% and 11.9%, respectively). Two patients had a history of bone marrow transplantation and two had a history of odontogenic keratocyst. The recurrence rate after treatment was 8.6%. Conclusion: Early diagnosis and treatment and close follow-up of childhood head and neck carcinoma are warranted to prevent recurrence and improve clinical outcome.

The minimum ignition equivalence ratio of the strut stabilizer is an important parameter in the design of integrated afterburners. The ignition location significantly affects the ignition equivalence ratio and flame propagation, and therefore, it should be deeply studied. The ignition equivalence ratio and flame propagation at different axial ignition locations downstream of the strut stabilizer are studied in this paper. When the ignition distance is approximately the bluff body trailing edge width, a lower ignition equivalence ratio is required for ignition, and the flame propagates faster through the entire combustion chamber. For different ignition locations, the generated flame kernel at different locations all first propagates to the shear layer. Subsequently, the unilateral flame rapidly extends, ultimately igniting the entire combustion chamber. The flame propagation trajectory depends on the ignition location controlled by the non-reacting flow field and the distribution of kerosene concentration. The flame propagation trajectory mainly includes three paths: (1) the flame kernel is directly downstream the shear layer when the ignition location is close to the tail edge of the stabilizer, (2) the flame propagates upstream into the shear layer in a U-shape when the ignition location is far from the stabilizer but still in the recirculation zone, and (3) the flame propagates upstream into the recirculation zone and shear layer in a U-shape when the ignition location is outside the recirculation zone. In addition, the time for flame propagation to the shear layer is directly related to the ignition performance when the ignition location is within the recirculation zone. If the flame reaches the shear layer in a longer time, there will be more energy loss during the flame propagation process, and the ignition performance will deteriorate. The speed of the flame-trailing edge extension is directly related to the ignition fuel-air ratio, and the downstream extension of the flame is mainly affected by the turbulence velocity in the shear layer.

In this study, the texture formation mechanism of a TA15 titanium alloy under different plane strain compression conditions was investigated by analyzing the slipping, dynamic recrystallization (DRX) and phase transformation behaviors. The results indicated that the basal texture component basically appears under all conditions, since the dominant basal slip makes the C-axis of the α grain rotate to the normal direction (ND, i.e., compression direction), but it has a different degree of deflection. With an increase in deformation amount, temperature or strain rate, {0001} poles first approach the ND and then deviate from it. Such deviation is mainly caused by a change in slip behaviors and phase transformation. At a smaller deformation amount and higher strain rate, inhomogeneous deformation easily causes a basal slip preferentially arising from the grain with a soft orientation, resulting in a weak basal texture component. A greater deformation amount can increase the principal strain ratio, thereby promoting other slip systems to be activated, and a lower temperature can increase the critical shear stress of the basal slip, further causing a dispersive orientation under these conditions. At a higher temperature and a lower strain rate, apparent phase transformation will induce the occurrence of lamellar α whose orientation obeys the Burgers orientation of the β phase, thereby disturbing and weakening the deformation texture. As for DRX, continuous-type (CDRX) is most common under most conditions, whereas CDRX grains have a similar orientation to deformed grains, so DRX has little effect on overall texture. Moreover, the microhardness of samples is basically inversely proportional to the grain size, and it can be significantly improved as lamellar α occurs. In addition, deformed samples with a weaker texture present a higher microhardness due to the smaller Schmidt factors of the activated prism slip at ambient loading.

BackgroundDeer tuberculosis is a chronic zoonotic infectious disease, despite the existence of socio-economic and zoonotic risk factors, but at present, there has been no systematic review of deer tuberculosis prevalence in mainland China. The aim of this meta-analysis was to estimate the overall prevalence of deer TB in mainland China and to assess possible associations between potential risk factors and the prevalence of deer tuberculosis.MethodologyThis study was searched in six databases in Chinese and English, respectively (1981 to December 2023). Four authors independently reviewed the titles and abstracts of all retrieved articles to establish the inclusion exclusion criteria. Using the meta-analysis package estimated the combined effects. Cochran’s Q-statistic was used to analyze heterogeneity. Funnel plots (symmetry) and used the Egger’s test identifying publication bias. Trim-and-fill analysis methods were used for validation and sensitivity analysis. we also performed subgroup and meta-regression analyses.ResultsIn this study, we obtained 4,400 studies, 20 cross-sectional studies were screened and conducted a systematic review and meta-analysis. Results show: The overall prevalence of tuberculosis in deer in mainland China was 16.1% (95% confidence interval (CI):10.5 24.6; (Deer tuberculosis infected 5,367 out of 22,215 deer in mainland China) 5,367/22215; 1981 to 2023). The prevalence in Central China was the highest 17.5% (95% CI:14.0–21.9; 63/362), and among provinces, the prevalence in Heilongjiang was the highest at 26.5% (95% CI:13.2–53.0; 1557/4291). Elaphurus davidianus was the most commonly infected species, with a prevalence of 35.3% (95% CI:18.5–67.2; 6/17). We also assessed the association between geographic risk factors and the incidence of deer tuberculosis.ConclusionDeer tuberculosis is still present in some areas of China. Assessing the association between risk factors and the prevalence of deer tuberculosis showed that reasonable and scientific-based breeding methods, a suitable breeding environment, and rapid and accurate detection methods could effectively reduce the prevalence of deer tuberculosis. In addition, in the management and operation of the breeding base, improving the scientific feed nutrition standards and establishing comprehensive standards for disease prevention, immunization, quarantine, treatment, and disinfection according to the breeding varieties and scale, are suggested as ways to reduce the prevalence of deer tuberculosis.

IntroductionMassive tree mortality events in western Canada due to widespread infestation by mountain pine beetle (MPB) are expected to impact local-to-regional evapotranspiration (ET) dynamics during and after a disturbance. How ecosystem-level ET and its components may vary with canopy-tree mortality (treefall) and subsequent understory recovery remains unclear.MethodsWe used 10 years of continuous eddy-covariance and remote-sensing data (2007–2016) and machine-learning models based on random forest and xgboost to determine forest- and climate-driven effects at temporal scales appropriate for a lodgepole pine-dominated stand following a major, five-year MPB disturbance initiated in the summer of 2006.ResultsTotal annual ET over the 10 years ranged from 207.2 to 384.6 mm, with annual plant transpiration (T) contributing to 57 ± 5.4% (mean ± standard deviation) of annual ET. Annual ET initially declined (2007–2011) and then increased (2011–2016), with ET and T/ET increasing at statistically non-significant rates of approximately 3.2 and 1.2% per year from 2007 to 2016. Air temperature (Ta) and vapor pressure deficit (VPD) were the most important predictors of seasonal variation in ET and T/ET during the 10-year period, with high Ta, VPD, and photosynthetically active radiation (PAR) causing ET and T/ET to increase. Annual ET increased with both increasing spring Ta and decreasing VPD. Annual T/ET was shown to increase with increasing VPD and decrease with increasing volumetric soil water content at a 5-cm depth (VWC5). Enhanced vegetation index (EVI, an indicator of canopy greenness) lagged T and overstory tree mortality, whereas previous- and current-year values of EVI were shown to be poor predictors of annual ET and T/ET.Discussion and conclusionsThese findings suggest that the promotion of climate factors on forest ecosystem-level water vapor fluxes may offset reductions promoted by MPB outbreaks. Climate processes affected water vapor fluxes more than biotic factors, like stand greenness, highlighting the need to include climate-regulatory mechanisms in predictive models of ET dynamics during and subsequent to stand disturbance. Climate and forest-greenness effects on water vapor fluxes need to be explored at even longer time scales, e.g., at decadal scales, to capture long-drawn-out trends associated with stand disturbance and its subsequent recovery.

Ventricular septal rupture (VSR) is a catastrophic mechanical complication of acute myocardial infarction (AMI) that can result in acute heart failure. Delaying operative intervention frequently leads to cardiogenic shock and multi-organ failure. Here we report a case of massive anterior MI complicated with VSR that was discovered through cardiac Doppler ultrasound and suspected multiple organ hemorrhage. The patient showed signs of rapid cardiogenic shock and eventually died. The morphological changes of VSR and MI were identified during necropsy, and microscopic examinations of the heart, brain, and kidney revealed multiple organ hemorrhage. This autopsy case suggested that the complication of VSR caused by AMI results in a reduction of oxygen and nutrient content of the circulating blood throughout the body and, eventually, functional failure of multiple organs. We provide clinical and pathological evidence elucidating changes in multiple organs under the severe condition of post-infarction VSR and demonstrate the consequences of a lack of immediate surgery and sufficient medical intervention for a patient suffering from AMI with VSR.

Ochratoxin A (OTA) is a mycotoxin commonly found in several food commodities worldwide with potential nephrotoxic, hepatotoxic and carcinogenic effects. We previously showed for the first time that OTA treatment enhanced glycolysis in human gastric epithelium (GES-1) cells in vitro. Here, we found that OTA exposure activated inflammatory responses, evidenced by increasing of NF-κB signaling pathway-related protein (p-p65 and p-IκBα) expressions and elevating of inflammatory cytokine (IL-1β and IL-6) mRNA expressions in GES-1 cells. To elucidate the role of glycolysis in inflammatory effects triggered by OTA, we pretreated GES-1 cells with glycolysis inhibitor (2-deoxy-D-glucose, 2-DG) before OTA exposure. The result showed that 2-DG reduced the protein expressions of p-p65 and p-IκBα and alleviated the mRNA expressions of inflammatory cytokines in OTA-treated GES-1 cells. Furthermore, OTA activated the mTOR/HIF-1α pathway by increasing the protein expressions of p-mTOR, p-eIF4E and HIF-1α, and inhibition of mTOR with rapamycin or silencing HIF-1α with siRNA significantly attenuated OTA-enhanced glycolysis by reducing glycolysis related genes and thereby decreasing inflammatory effects of GES-1 cells. These results demonstrate that OTA activates inflammatory responses in GES-1 cells and this is controlled by mTOR/HIF-1α pathway-mediated glycolysis enhancement. Our findings provide a novel mechanistic view into OTA-induced gastric cytotoxicity.

AbstractPurpose Due to the poor and unpredictable prognosis of breast cancer (BC) patients with bone metastasis, it is necessary to find convenient and available prognostic predictors. This study aimed to recognize the clinical and prognostic factors related to clinical laboratory examination and to construct a prognostic nomogram for BC bone metastasis.Methods We retrospectively analyzed 32 candidate indicators from clinical features and laboratory examination data of 276 BC patients with bone metastasis. Univariate and multivariate regression analyses were performed to identify significant prognostic factors related to BC with bone metastasis. Nomogram was constructed and estimated by receiver operating characteristic (ROC) curves, calibration curves, and decision curve analysis.Results Patients were randomly grouped into training (n = 197) and validation cohorts (n = 79). In training cohort, the multivariate regression analysis revealed that age, other organ metastasis sites, serum level of lactate dehydrogenase, globulin, white blood cell count, mean corpuscular volume, mean corpuscular hemoglobin, and monocyte ratio were independent prognostic factors for BC with bone metastasis. The prognostic nomogram in training cohort exhibited areas under the ROC curve (AUCs) of 0.797, 0.782, and 0.794, respectively, for predicting 1-, 3-, and 5-year overall survival. In validation cohort, the nomogram still showed acceptable discrimination ability (AUCs: 0.723, 0.742, and 0.704) and calibration.Conclusion This study constructed a novel prognostic nomogram for BC patients with bone metastasis. It could serve as a potential tool of survival assessment to help individual treatment decision-making for clinicians.

Abstract Background Neutrophils consume a large amount of energy when performing their functions. Compared with other white blood cells, neutrophils contain few mitochondria and mainly rely on glycolysis and gluconeogenesis to produce ATP. The inflammatory site is hypoxic and nutrient poor. Our aim is to study the role of abnormal adenosine metabolism of neutrophils in the asthmatic airway inflammation microenvironment. Method In this study, an asthma model was established by intratracheal instillation of Aspergillus fumigatus extract in Ecto-5'-Nucleotidase (CD73) gene–knockout and wild-type mice. Multiple analyses from bronchoalveolar lavage fluid (BALF) were used to determine the levels of cytokines and chemokines. Immunohistochemistry was used to detect subcutaneous fibrosis and inflammatory cell infiltration. Finally, adenosine 5’-(α, β-methylene) diphosphate (APCP), a CD73 inhibitor, was pumped subcutaneously before Aspergillus attack to observe the infiltration of inflammatory cells and subcutaneous fibrosis to clarify its therapeutic effect. Result PAS staining showed that CD73 knockout inhibited pulmonary epithelial cell proliferation and bronchial fibrosis induced by Aspergillus extract. The genetic knockdownof CD73 significantly reduced the production of Th2 cytokines, interleukin (IL)-4, IL-6, IL-13, chemokine (C–C motif) ligand 5 (CCL5), eosinophil chemokine, neutrophil IL-17, and granulocyte colony-stimulating factor (G-CSF). In addition, exogenous adenosine supplementation increased airway inflammation. Finally, the CD73 inhibitor APCP was administered to reduce inflammation and subcutaneous fibrosis. Conclusion Elevated adenosine metabolism plays an inflammatory role in asthma, and CD73 could be a potential therapeutic target for asthma.
