Abstract A novel method is proposed for rail fastener detection based on millimetre‐wave (mmWave) radar, mmWave radar fastener counter (MMW‐FC), which can accurately detect and record the fasteners in real‐time as the train traverses its route. Under circumstances where GNSS signals remain unavailable for prolonged durations, precise train localisation can be accomplished by correlating the number of fasteners derived from this method with the corresponding track map. Initially, MMW‐FC utilises fast Fourier transform and adaptive beamforming to focus the energy reflected from fasteners. Subsequently, it applies an adaptive template‐matching algorithm to detect each fastener. Furthermore, by leveraging known fastener spacing and the average time for trains to pass adjacent fasteners, the Kalman filter can execute precise speed tracking, used as a speed reference when adjusting the matching template adaptively. The experimental results indicate that the proposed method can precisely count the fasteners the train encounters in diverse road and speed conditions. The fastener counter maintains the Counting Error less than 0.067%, the speed error stays below 1.8 km/h, and the maximum values of the mean absolute error and root mean square error for speed are 0.7337 and 0.9584 km/h, respectively.

Abstract Objectives To develop an objective method based on texture analysis on MRI for diagnosis of congenital muscular torticollis (CMT). Material and methods The T1- and T2-weighted imaging, Q-dixon, and T1-mapping MRI data of 38 children with CMT were retrospectively analyzed. The region of interest (ROI) was manually drawn at the level of the largest cross-sectional area of the SCM on the affected side. MaZda software was used to obtain the texture features of the T2WI sequences of the ROI in healthy and affected SCM. A radiomics diagnostic model based on muscle texture features was constructed using logistic regression analysis. Fatty infiltration grade was calculated by hematoxylin and eosin staining, and fibrosis ratio by Masson staining. Correlation between the MRI parameters and pathological indicators was analyzed. The diffusion of chloride ions in reinforced concrete under the seawater environment will affect the physical or mechanical properties and durability. Sulfate ions also influence the diffusion of chloride ions by generating expansion products through physical and chemical reactions. Further research on the effects of chloride binding and sulfate ion attack on chloride ion diffusion will deepen the understanding of concrete failure mechanism. In this paper, a quaternary blend based on calcium sulfoaluminate (CSA) cement was prepared and laboratory immersion tests were conducted in artificial seawater with varying salt concentrations. Initially, chloride ion concentration measurements and XRD tests were conducted at different corrosion ages and salt concentrations. Subsequently, the chloride binding properties of CSA-based materials was quantified through the analysis of chloride ion concentration and the phases of hydration products. Meanwhile, the corrosive effect of sulfate ions on chloride ion diffusion was determined corresponds to the content of expansive hydration products. Furthermore, numerical simulations were employed to evaluate the impact of sulfate ion corrosion, chloride binding, and their combined effects on chloride ion diffusion, and the results were compared with free chloride ion concentration (Cf). The results indicate that the total chloride ion concentration (Ct) exhibits a linear relationship with increasing salt concentrations of seawater in CSA-based materials. Meanwhile, the bound chloride ion concentration (Cb) follows a logarithmic growth pattern. The chloride binding rate experiences a linear decline with the increase of chloride ion concentration in the artificial seawater. Additionally, the relationship between Cf and Cb or Ct can be described using different isothermal adsorption models for the CSA-based materials. Among these models, the Langmuir model is suitable for characterizing the relationship between Cf and Cb, while the Freundlich model is suitable for describing the relationship between Cf and Ct. Furthermore, it is noteworthy that sulfate ion attack and chloride binding exert nearly equivalent effects on the diffusion of chloride ions in CSA-based materials at short corrosion age and low chloride ion concentration. However, the corrosive impact of sulfate ions gradually evolves into the dominant factor in the progression of corrosion over time and the increase in seawater salt concentration.

Abstract Background Fractures of hands and feet are common in children, but relevant epidemiological studies are currently lacking. We aim to study the epidemiological characteristics of hand and foot fractures and growth plate injuries in children and provide a theoretical basis for their prevention, diagnosis, and treatment. Methods We retrospectively analyzed the data of children with hand and foot fractures who were hospitalized at Shenzhen Children’s Hospital between July 2015 and December 2020. Data on demographic characteristics, fracture site, treatment method, etiology of injury, and accompanying injuries were collected. The children were divided into four age groups: infants, preschool children, school children, and adolescents. The fracture sites were classified as first-level (the first–fifth finger/toe, metacarpal, metatarsal, carpal, and tarsal) and second-level (the first–fifth: proximal phalanx, middle phalanx, distal phalanx, metacarpal, and metatarsal) sites. The changing trends in fracture locations and injury causes among children in each age group were analyzed. Results Overall, 1301 children (1561 fractures; 835 boys and 466 girls) were included. The largest number of fractures occurred in preschool children (n = 549, 42.20%), with the distal phalanx of the third finger being the most common site (n = 73, 15.57%). The number of fractures in adolescents was the lowest (n = 158, 12.14%), and the most common fracture site was the proximal phalanx of the fifth finger (n = 45, 29.61%). Of the 1561 fractures, 1143 occurred in the hands and 418 in the feet. The most and least common first-level fracture sites among hand fractures were the fifth (n = 300, 26.25%) and first (n = 138, 12.07%) fingers, respectively. The most and least common first-level foot fracture locations were the first (n = 83, 19.86%) and fourth (n = 26, 6.22%) toes, respectively. The most common first-level and second level etiologies were life related injuries (n = 1128, 86.70%) and clipping injuries (n = 428, 32.90%), respectively. The incidence of sports injuries gradually increased with age, accounting for the highest proportion in adolescents (26.58%). Hand and foot fractures had many accompanying injuries, with the top three being nail bed injuries (570 cases, 36.52%), growth plate injuries (296 cases, 18.96%), and distal severed fracture (167 cases, 10.70%). Among the 296 growth plate injuries, 246 occurred on the hands and 50 on the feet. Conclusions In contrast to previous epidemiological studies on pediatric hand and foot fractures, we mapped the locations of these fractures, including proximal, shaft, distal, and epiphyseal plate injuries. We analyzed the changing trends in fracture sites and injury etiologies with age. Hand and foot fractures have many accompanying injuries that require attention during diagnosis and treatment. Doctors should formulate accident protection measures for children of different ages, strengthen safety education, and reduce the occurrence of accidental injuries.

Abstract Introduction Real-world studies on neoadjuvant dual anti-HER2 therapy combined with chemotherapy for breast cancer (BC) are scarce in China. This study aimed to evaluate the efficacy and safety of neoadjuvant dual anti-HER2 therapy combined with chemotherapy in a real-world setting. Moreover, differences in estrogen receptor (ER), progesterone receptor (PR), human epidermal growth factor receptor 2 (HER2), and proliferation cell nuclear antigen (Ki-67) expression pre- and post-neoadjuvant therapy were analyzed. Methods Clinical and pathological data of patients with HER2-positive BC who received neoadjuvant dual anti-HER2 therapy combined with chemotherapy at Liaoning Cancer Hospital & Institute, China, between September 2021 and September 2023, were retrospectively reviewed. Results Among 179 included patients, a pathologic complete response (pCR) was achieved in 109 patients (60.9%). The univariate analysis results indicated that the hormone receptor (HR) status (P = 0.013), HER2 status (P = 0.003), and cycles of targeted treatment (P = 0.035) were significantly correlated with pCR. Subsequent multivariable analysis showed that HR negative and HER2 status 3 + were independent predictive factors of pCR. Anemia was the most common adverse event (62.0%), and the most common grade 3–4 adverse event was neutropenia (6.1%). The differences in HER2 (34.5%) and Ki-67 (92.7%) expression between core needle biopsy and the residual tumor after neoadjuvant therapy were statistically significant, whereas the differences were insignificant in terms of ER or PR status. Conclusions The combination of neoadjuvant trastuzumab and pertuzumab with chemotherapy showed good efficiency, and the toxic side effects were tolerable in patients with BC. In cases where pCR was not achieved after neoadjuvant therapy, downregulation of HER2 and Ki-67 expressions was observed.

Introduction: Upper urinary tract stones combined with parenchymal infiltrative renal pelvic cancer are challenging to detect on imaging and to evaluate the differential diagnosis. Case Presentation: The symptoms and diagnoses in three cases of parenchymal infiltrative renal pelvic cancer and upper urinary tract stones that occurred between June 2019 and June 2022 were reviewed. Primary symptoms of lumbar discomfort and hematuria were evident in all 3 patients. Preoperative computed tomography (CT) abdominal imaging revealed that all three cases had hydronephrosis along with renal stones, while the other two cases only had localized hypoenhancement of the renal parenchyma, which was only thought to be limited inflammatory changes in the renal cortex as a result of the combination of renal pelvis infection. After percutaneous nephrolithotomy or ureteroscopic lithotripsy, a combined renal pelvis tumor was discovered in all of these instances. Radical tumor surgery was later performed. One patient who had several tumor metastases passed away 6 months after surgery. A case with multiple metastases was discovered 15 months after surgery and survived with the help of the current chemotherapy. A case with a bladder tumor recurrence was discovered 16 months after surgery and had transurethral bladder tumor electrosurgery and routine bladder perfusion chemotherapy. Conclusion: Upper urinary tract stones and parenchymal infiltrative pyel carcinoma have atypical imaging, easily confused with infectious diseases. CT or computed tomography urography (CTU) must be considered by urologists. Patients who have a CT with local renal parenchyma density should be suspected of having parenchymal invasive renal pelvis carcinoma; a needle biopsy ought to be performed; and repeat biopsies may be performed if necessary. High-risk individuals need multiple, sufficient biopsies as needed and a comprehensive intraoperative assessment of the renal pelvic mucosa.

This work reports an exceptional reversed yield strength asymmetry at room temperature for a rare-earth free magnesium alloy containing a mass of fine dispersed quasicrystal (I-phase) precipitates. Although exhibiting traditional basal texture, it owns an exceptional CYS/TYS as high as ∼1.17. Electron back-scattered diffraction (EBSD) and transmission electron microscopy (TEM) examinations indicate pyramidal and prismatic dislocations plus tensile twinning being activated after immediate yielding in compression while basal and non-basal dislocations in tension. I-phase particles transferred the concentrated stress by self-twinning to provide the driving force for tensile twin initiating in neighboring grains, thereby significantly increasing the critical resolved shear stress of tensile twinning to possibly the level of pyramidal slip, finally leading to the dominance of pyramidal slip plus tensile twinning in texture grains. This results in a higher contribution on yield strength by ∼55 MPa in compression than in tension, which reasonably agrees with the experimental yield strength difference (∼38 MPa). It can be concluded that I-phase particles influence deformation modes in tension and in compression, finally result in reversed yield strength asymmetry.

Abstract Background Porcine reproductive and respiratory syndrome virus (PRRSV) infection causes severe inflammatory response, respiratory disease and sow reproductive failure. Quercetin is among the widely occurring polypheno found abundantly in nature. Quercetin has anti‐inflammatory, anti‐oxidative and anti‐viral properties. Objectives This study aimed to explore the effect and mechanism of quercetin on PRRSV‐induced inflammation in MARC‐145 cells. Methods Observing the cytopathic effect and measurements of inflammatory markers in MARC‐145 cells collectively demonstrate that quercetin elicits a curative effect on PRRSV‐induced inflammation. Liquid chromatography–mass spectrometry was further used for a non‐targeted metabolic analysis of the role of quercetin in the metabolic regulation of PRRSV inflammation in MARC‐145 cells. Results It was shown that quercetin attenuated PRRSV‐induced cytopathy in MARC‐145 cells. Quercetin treatment inhibited PRRSV replication in MARC‐145 cells in a dose‐dependent manner. We also found that quercetin inhibited PRRSV‐induced mRNA expression and secretion levels of tumour necrosis factor‐α, interleukin 1β and interleukin 6. Metabolomics analysis revealed that quercetin ameliorated PRRSV‐induced inflammation. Pathway analysis results revealed that PRRSV‐induced pathways including arachidonic acid metabolism, linoleic acid, glycerophospholipid and alanine, aspartate and glutamate metabolism were suppressed by quercetin. Moreover, we confirmed that quercetin inhibited the activation of NF‐κB/p65 pathway, probably by attenuating PLA2, ALOX and COX mRNA expression. Conclusions These results provide a crucial insight into the molecular mechanism of quercetin in alleviating PRRSV‐induced inflammation.

Abstract Somatic mutations related to clonal hematopoiesis of indeterminate potential (CHIP) are risk factors for stroke. The impact of DNMT3A, the most mutated gene in CHIP, on clinical functional outcomes of acute ischemic stroke (AIS) remains unclear. In a well‐characterized cohort of 8524 ischemic stroke patients, we demonstrated that DNMT3A‐driven CHIP was significantly associated with neurological disability in these patients. With a stroke mouse model of transient middle cerebral artery occlusion (tMCAO), we demonstrated that DNMT3A protein levels in the brain penumbra increased. The DNMT3A inhibitor RG108 administration amplified neutrophil proliferation in the blood, promoted neutrophil infiltration into the brain penumbra, and exaggerated proinflammatory activation in tMCAO male mice. DNMT3A inhibition also significantly increased infarct volume and worsened neurobehavioral function in tMCAO male mice. In conclusion, DNMT3A somatic mutations are associated with worsened neurological disability in some patients with AIS, potentially through increased neutrophil proliferation and infiltration in the ischemic brain region. These findings suggest a possible mechanism for proinflammatory activation and tissue damage in the affected brain tissue, highlighting the need for further research in this area.

Ecosystem surface plays an important role in maintaining ecological balance and supporting economic development. Accurate and effective identification of ecosystem surface with different ecological uses allows for better categorization and management and contributes to the orderly restoration of ecosystems. In this study, the Human Footprint Index (HFI) is constructed by comprehensively considering the factors of biodiversity, human activity impacts, and geometric features, and combined with the multi-factor knowledge constraints such as changes in land cover. We carry out accurate and effective identification and analysis of ecosystem surfaces, using Changzhou City’s tree cover as a test area. The results show that: (1) The degree of tree cover affected by human activities in Changzhou City varies greatly, and Gross Domestic Product (GDP) and elevation are the main factors affecting the level of the HFI. (2) The distribution of native ecosystem surface and artificial ecosystem surface are uneven, with more than 95% of native ecosystem surfaces distributed in the peripheral areas of the city with higher elevations and farther away from the city center. Artificial ecosystem surfaces are more likely to be found in areas with high GDP and frequent human activities, mainly in urban centers with a large proportion of buildings and along roads. (3) The overall identification accuracy of the ecosystem surface is 83.20%, which is higher in the urban center area with a large proportion of buildings and slightly lower in the area with a large proportion of agricultural production land. The results of the study provide a method for the objective, accurate and effective identification of ecosystem surface, which can improve the enthusiasm of local governments in fulfilling their ecological protection duties and contribute to the efficient achievement of the goal of ecological civilization construction.

Abstract Background Clonal hematopoiesis of indeterminate potential (CHIP) is a phenomenon that characterizes individuals with somatic mutations that are related to hematologic malignancy but without hematologic abnormalities. Presence of CHIP is associated with the atherosclerotic cardiovascular disease through the activation of the interleukin 6 (IL-6) pathway; however, its role on unfavorable functional outcomes in different etiologies of ischemic stroke remains unclear. We aimed to investigate the association between CHIP-related gene mutations and unfavorable functional outcomes of ischemic stroke with different etiologies. Methods We prospectively studied a cohort of 3396 stroke patients with identified etiologies, and identified CHIP and the presence of the IL6R variant (IL6R p.Asp358Ala) by whole-genome sequencing. The IL6R p.Asp358Ala coding mutation was used as a genetic inhibition for IL-6 signaling. The primary outcome was unfavorable functional outcome [(Modified Rankin Scale), mRS 2–6] at 3 months. Results Of the 3396 patients, 110 (3.2%) were CHIP carriers and the median age was 62 years (IQR, 54.0–69.0). The CHIP increased the risk of unfavorable functional outcome among patients with hyper-inflammation status of high-sensitivity C-reactive protein (hsCRP) > median levels in patients with large-artery atherosclerosis (LAA) (OR 2.45, 95% CI 1.00–5.98, p = 0.049, p interaction = 0.01). Presence of IL6R variant (IL6R p.Asp358Ala) could attenuate the risk of unfavorable functional outcome only in patients with CHIP (OR 0.30, 95%CI 0.12–0.76, p = 0.01, p interaction = 0.02), and especially in LAA patients with CHIP (OR 0.1, 95%CI 0.02–0.42, p = 0.002; p interaction = 0.001). Conclusion CHIP is associated with unfavorable functional outcomes in patients with LAA stroke and hyper-inflammation. Genetic IL-6 signaling inhibition might attenuate the risk of unfavorable functional outcomes in CHIP carriers, especially in LAA stroke patients.

Background Noise and drug-induced hearing loss (HL) is becoming more and more serious, but the integration and analysis based on transcriptomics and proteomics are lacking. On the one hand, this study aims to integrate existing public transcriptomic data on noise and gentamicin-induced HL. On the other hand, the study aims to establish the gentamicin and noise-induced HL model of guinea pigs, then to perform the transcriptomic and proteomic analyses. Through comprehensive analysis of the above data, we aim to screen, predict, and preliminarily verify biomarkers closely related to HL.

Abstract Background Congenital muscular torticollis (CMT) is a common musculoskeletal disease affecting infants and young children. If CMT is not treated correctly and timely, it can lead to limited head and neck movements, head and neck deviation, and abnormal posture. In order to improve patients' symptoms and alleviate the negative impact of the disease on their lives, we are committed to exploring the treatment of CMT. Methods The general clinical and ultrasonographic data of 2599 children with CMT who received standardized treatment at Shenzhen Children’s Hospital from 2004 to 2020 were retrospectively reviewed. According to given treatment, children with CMT were divided into the physiotherapy group, physiotherapy combined with glucocorticoid treatment group, and surgical treatment group. We divided children with CMT into local mass, uniform thickening, and atrophy according to ultrasound features. General clinical information, treatment, and ultrasound examination data in each group were compared. Additionally, electronic medical records of 2344 patients admitted due to CMT in 17 tertiary children’s hospitals of China’s Futang Research Center of Pediatric Development (FRCPD) from 2015 to 2019 were retrospectively analyzed. Data on sex, age, year of admission and discharge, and treatment costs during hospitalization were extracted from the first medical record pages according to the ICD codes. The data were assessed for normality using the Kolmogorov–Smirnov test. Depending on the data distribution, they were analyzed using parametric tests, such as the t-test, or non-parametric tests. AIM: To investigate the effect of modified Chufeng Yisun Decoction on ocular surface inflammation after pterygium surgery.METHODS: A total of 60 patients(60 eyes)with primary pterygium who underwent pterygium surgery were randomly divided into control group and study group, with 30 cases(30 eyes)in each group. In the control group, patients were treated with pranoprofen eye drops, tobramycin dexamethasone eye drops, and deproteinized calf blood extract eye gel after the surgery. To uncover the role of satellite cells (SCs) in paravertebral muscle development and aging, we constructed a single-nucleus transcriptomic atlas of mouse paravertebral muscle across seven timepoints spanning the embryo (day 16.5) to old (month 24) stages. Eight cell types, including SCs, fast muscle cells, and slow muscle cells, were identified. An energy metabolism-related gene set, TCA CYCLE IN SENESCENCE, was enriched in SCs. Forty-two skeletal muscle disease-related genes were highly expressed in SCs and exhibited similar expression patterns. Among them, Pdha1 was the core gene in the TCA CYCLE IN SENESCENCE; Pgam2, Sod1, and Suclg1 are transcription factors closely associated with skeletal muscle energy metabolism. Transcription factor enrichment analysis of the 42 genes revealed that Myod1 and Mef2a were also highly expressed in SCs, which regulated Pdha1 expression and were associated with skeletal muscle development. These findings hint that energy metabolism may be pivotal in SCs development and aging. Three ligand-receptor pairs of extracellular matrix (ECM)-receptor interactions, Lamc1-Dag1, Lama2-Dag1, and Hspg2-Dag1, may play a vital role in SCs interactions with slow/fast muscle cells and SCs self-renewal. Finally, we built the first database of a skeletal muscle single-cell transcriptome, the Musculoskeletal Cell Atlas (http://www.mskca.tech), which lists 630,040 skeletal muscle cells and provides interactive visualization, a useful resource for revealing skeletal muscle cellular heterogeneity during development and aging. Our study could provide new targets and ideas for developing drugs to inhibit skeletal muscle aging and treat skeletal muscle diseases.

IntroductionThe ecological and environmental pollution problem at the source of agriculture cannot be ignored, and the manifestation of the fertilizer reduction effect of pro-environmentally agricultural technologies (PEATs) will help motivate farmers to adopt technology, thereby promoting sustainable agricultural development.MethodsFrom the dual perspectives of farmers’ perception and behavior effects, this paper uses 607 survey data of Chinese farmers, and an endogenous switching regression model is employed to identify the influencing factors of farmers’ adoption of PEATs and manifest its fertilizer reduction effect.Results and discussionThe results of the perception survey show that the farmers’ recognition of the fertilizer reduction effect of PEATs is not high, and the technical effect needs to be further demonstrated. Moreover, the estimated results suggest that PEATs can significantly reduce the fertilizer application of farmers. Specifically, if farmers who have adopted PEATs do not adopt them, they will apply more chemical fertilizers, the farmers who have not adopted PEATs will use less chemical fertilizer if they do. Overall, the main influencing factors for farmers adopting PEATs include education level, government officials, cultivated land area, soil fertility, information access channels, and the distance of home-agricultural technology station. This study aims to provide empirical evidence for the formulation of strategies and plans to promote sustainable agricultural development.

ObjectiveCurrently, sentinel lymph node biopsy (SLNB) is increasingly used in endometrial cancer, but the rate of missed metastatic lymph nodes compared to systemic lymph node dissection has been a concern. We conducted a systematic review and meta-analysis to evaluate the false negative rate (FNR) of SLNB in patients with endometrial cancer and to explore the risk factors associated with this FNR.Data sourcesThree databases (PubMed, Embase, Web of Science) were searched from initial database build to January 2023 by two independent reviewers.Research eligibility criteriaStudies were included if they included 10 or more women diagnosed with International Federation of Gynecology and Obstetrics (FIGO) stage I or higher endometrial cancer, the study technique used sentinel lymph node localization biopsy, and the reported outcome metrics included false negative and/or FNR.Study appraisal and synthesis methodsTwo authors independently reviewed the abstracts and full articles. The FNR and factors associated with FNR were synthesized through random-effects meta-analyses and meta-regression.The resultsWe identified 62 eligible studies. The overall FNR for the 62 articles was 4% (95% CL 3-5).There was no significant difference in the FNR in patients with high-risk endometrial cancer compared to patients with low-risk endometrial cancer. There was no difference in the FNR for whether frozen sections were used intraoperatively. The type of dye used intraoperatively (indocyanine green/blue dye) were not significantly associated with the false negative rate. Cervical injection reduced the FNR compared with alternative injection techniques. Indocyanine green reduced the FNR compared with alternative Tc-99m. Postoperative pathologic ultrastaging reduced the FNR.ConclusionsAlternative injection techniques (other than the cervix), Tc-99m dye tracer, and the absence of postoperative pathologic ultrastaging are risk factors for a high FNR in endometrial cancer patients who undergo SLNB; therefore, we should be vigilant for missed diagnosis of metastatic lymph nodes after SLNB in such populations.Systematic review registrationhttp://www.crd.york.ac.uk/PROSPERO/, identifier CRD42023433637.

The high temperature of modern electronic devices poses great challenges to the safety and reliability of applications. Reasonable and effective control of device operating temperature for enhancing device performance and promoting the development of the electronics industry has a positive effect. Here, an innovative and efficient passive cooling technology is proposed to reduce the device temperature using polymerized metal fin and smart hydrogel to form smart fin. The smart fin carries away the waste heat generated by the device through evaporation and regeneration is accomplished by capturing water molecules from the environment. The smart fin is demonstrated to low down the temperature of a heater with heat flux of 2900 W/m2 by 48.3 °C and restores to its initial state after 160 min. At a threshold temperature of 60 °C, it promoted the maximum usage power by 49.4%. The evaporation and regeneration capacity of the smart fin can be controlled by ambient temperature, relative humidity, and smart hydrogel thickness. Specifically, high temperatures promote water evaporation and absorption, high humidity suppresses evaporation but favors absorption, and excessive thickness impedes water transport. This thermal management strategy with simple structure, low-cost, and easy-preparation is expected to become a universal technology in the electronics industry.

Developmental dysplasia of the hip (DDH) is the most common hip deformity in pediatric orthopedics. One of the common pathological changes in DDH is the thickening and hypertrophy of the ligamentum teres. However, the underlying pathogenic mechanism responsible for these changes remains unclear. This study represents the first time that the heterogeneity of cell subsets in the abnormal ligamentum teres of patients with DDH has been resolved at the single-cell and spatial levels by snRNA-Seq and MiP-Seq. Through gene set enrichment and intercellular communication network analyses, we found that receptor-like cells and ligament stem cells may play an essential role in the pathological changes resulting in ligamentum teres thickening and hypertrophy. Eight ligand–receptor pairs related to the ECM–receptor pathway were observed to be closely associated with DDH. Further, using the Monocle R package, we predicted a differentiation trajectory of pericytes into two branches, leading to junctional ligament stem cells or fibroblasts. The expression of extracellular matrix-related genes along pseudotemporal trajectories was also investigated. Using MiP-Seq, we determined the expression distribution of marker genes specific to different cell types within the ligamentum teres, as well as differentially expressed DDH-associated genes at the spatial level.

Glycosylation is a crucial post-translational modification process where sugar molecules (glycans) are covalently linked to proteins, lipids, or other biomolecules. In this highly regulated and complex process, a series of enzymes are involved in adding, modifying, or removing sugar residues. This process plays a pivotal role in various biological functions, influencing the structure, stability, and functionality of the modified molecules. Glycosylation is essential in numerous biological processes, including cell adhesion, signal transduction, immune response, and biomolecular recognition. Dysregulation of glycosylation is associated with various diseases. Glycation, a post-translational modification characterized by the non-enzymatic attachment of sugar molecules to proteins, has also emerged as a crucial factor in various diseases. This review comprehensively explores the multifaceted role of glycation in disease pathogenesis, with a specific focus on its implications in osteoarthritis (OA). Glycosylation and glycation alterations wield a profound influence on OA pathogenesis, intertwining with disease onset and progression. Diverse studies underscore the multifaceted role of aberrant glycosylation in OA, particularly emphasizing its intricate relationship with joint tissue degradation and inflammatory cascades. Distinct glycosylation patterns, including N-glycans and O-glycans, showcase correlations with inflammatory cytokines, matrix metalloproteinases, and cellular senescence pathways, amplifying the degenerative processes within cartilage. Furthermore, the impact of advanced glycation end-products (AGEs) formation in OA pathophysiology unveils critical insights into glycosylation-driven chondrocyte behavior and extracellular matrix remodeling. These findings illuminate potential therapeutic targets and diagnostic markers, signaling a promising avenue for targeted interventions in OA management. In this comprehensive review, we aim to thoroughly examine the significant impact of glycosylation or AGEs in OA and explore its varied effects on other related conditions, such as liver-related diseases, immune system disorders, and cancers, among others. By emphasizing glycosylation's role beyond OA and its implications in other diseases, we uncover insights that extend beyond the immediate focus on OA, potentially revealing novel perspectives for diagnosing and treating OA.

BackgroundPersistent infection with high-risk human papillomavirus (HR-HPV) can lead to cervical intraepithelial neoplasia and cancer. At present, there is no medication that specifically targets HR-HPV infection.ObjectiveThis study aimed to evaluate the effectiveness of different interventions in promoting HR-HPV regression using a MeSH meta-analysis method.MethodsA search for randomized controlled trials (RCTs) reporting different interventions for the treatment of HR-HPV infection included PubMed, Web of Science, Embase and Cochrane Library from the inception of the databases to March 8, 2023. Two researchers independently screened the articles, extracted data, and evaluated the quality. The literature that met the inclusion criteria was selected, the quality and risk of bias of the included studies were assessed according to the Cochrane 5.1 manual, and NMA was performed using Stata 16.0. The area under the cumulative ranking probability graph (SUCRA) represented the probability that each treatment would be the best intervention.ResultsNine studies involving 961 patients and 7 treatment options were included in the analysis. The results of the network meta-analysis indicated the following rank order in terms of promoting HR-HPV conversion: Anti-HPV biological dressing > vaginal gel > imiquimod > REBACIN® > interferon > probiotics > observation/placebo > Polyphenon E.ConclusionAnti-HPV biological dressing treatment was found to be significantly effective in promoting HR-HPV conversion. BackgroundPancreatic adenocarcinoma carries a grim prognosis, and there are few recognized effective second-line treatment strategies. We attempted to evaluate the efficacy and safety of a combination of S-1, sintilimab, and anlotinib as a second-line treatment in pancreatic cancer patients with liver metastasis.MethodsPancreatic cancer patients with liver metastases were recruited. S-1 was administered orally at 25 mg/m2 bid, anlotinib was administered orally at 12 mg qd from day 1 to day 14, and sintilimab was administered intravenously at 200 mg on day 1. This method was repeated every 21 days, and the therapeutic effect was evaluated every 3 cycles. The primary outcome was the objective response rate (ORR).ResultsOverall, 23 patients were enrolled in this study of whom 19 patients had objective efficacy evaluation. The ORR was 10.5% (95% CI 0.4%–25.7%) in the evaluable population. The progression-free survival (PFS) was 3.53 (95% CI 2.50–7.50) months, and the overall survival (mOS) was 8.53 (95% CI 4.97–14.20) months. Grade 3 adverse events were 26.1%, and no grade 4 or above adverse events occurred. High-throughput sequencing was performed on the tumor tissues of 16 patients; patients with HRD-H (n = 10) had shorter PFS than those with HRD-L (n = 6) (2.43 vs. 5.45 months; P = 0.043), but there was no significant difference in OS between the two groups (4.43 vs. 9.35 months; P = 0.11).ConclusionsThis study suggests the advantage of S-1 combined with sintilimab and anlotinib in extending OS as a second-line therapy in pancreatic cancer patients with liver metastasis.Clinical Trial Registration: ChiCTR2000030659

Effects of 3 wt% RE (rare earth) addition on microstructures and mechanical properties of a high-pressure die casting Mg–8Zn–6Al (ZA86) alloy before and after aging treatment was thoroughly studied in this work. Microstructural characterizations present that RE addition has indiscernible influence on the well-defined dendrite structure and grain size, but would additionally introduce Al-RE intermetallic phases, which were identified as Al3RE and Al11RE. Simultaneously, two groups of Mg–Al–Zn ternary phases were detected in alloys with/without RE addition, while their volume fraction was clearly reduced by RE addition. After aging at 90 °C for 320 h, a large density of precipitates were observed particularly near grain/cell boundaries, and were identified as MgZn2, Mg4Zn7, and I-phase. Although RE addition seems to have no influence on the components of precipitates, it obviously improves their volume fraction and slightly increases their size. Finally, the yield strength of the ZA86 alloy after aging was improved by ∼33 MPa by RE addition. Therefore, the die cat ZA86 alloy exhibits high-strength and certain precipitation strengthening ability, and RE addition can significantly enhance the precipitation strengthening, accomplishing the highest yield strength in the reported HPDC magnesium alloys, being ∼267 MPa.

The development of [(bpy)Cu(O2CCF2SO2F)2] represents an advancement in the field of organic synthesis, potentially serving as a valuable addition to the arsenal of existing trifluoromethylating agents.

Abstract Background In this study, to provide a theoretical basis for understanding the clinical characteristics of epiphyseal fractures in children and improving their management, we explored and analyzed the proportions of different types of epiphyseal fractures in children and evaluated the causes of injury and epidemiological characteristics. Methods We retrospectively analyzed children younger than 18 years with fresh epiphyseal fractures who were admitted to our hospital from July 2015 to February 2020. Demographic information, injury mechanisms, fracture characteristics, fracture classification and surgical information were collected. Results A total of 1124 pediatric patients (1147 epiphyseal fractures), including 789 boys and 335 girls, were included in this study. Epiphyseal fractures were classified as Salter-Harris type II (1002 cases), type IV (105 cases), type III (25 cases), Salter-Harris type I (14 cases), and Salter-Harris type V (1 case). The number of fracture sites peaked in the adolescent group (440 cases). The most three common sites of epiphyseal fractures were the distal radius (460 cases) in which Salter-Harris type II fractures were the most common (454 cases) and Salter-Harris type I (3 cases), Salter-Harris type IV (2 cases), Salter-Harris type III was the least common (1 case). Followed by phalanges of fingers (233 cases) in which Salter-Harris type II fractures were the most common (224 cases) and Salter-Harris type IV (4 cases), Salter-Harris type I (3 cases), Salter-Harris type III fractures were the least common (2 cases). Distal humerus (146 cases) in which Salter-Harris type II fractures were the most common (95 cases), followed by Salter-Harris type IV (49 cases), Salter-Harris type I fractures were the least common (2 cases). The most three important causes of fractures were falls (720 patients), car accident injuries (68 patients), and basketball falls (43 patients). Among the 1124 children with epiphyseal fractures, 1058 were treated mainly by surgery and the ratio of open and closed reduction was 1:5.3. Eighty-eight patients showed an interval > 72 h between the injury and the hospital visit. Among these 88 patients, the most common fracture type was distal radial epiphyseal fracture (32 cases), and all fractures were of Salter-Harris type II. Conclusions The epidemiological characteristics of epiphyseal fractures in children indicate the need to strengthen health and safety education and protective measures to prevent the occurrence of these fractures in children. In addition, emergency surgeons and orthopedic surgeons in general hospitals should strengthen their basic knowledge of diagnosing and treating epiphyseal injuries in children to reduce missed diagnoses, misdiagnoses or malpractice.

Abstract Background Serum uric acid (SUA), an end-product of purine catabolism diffused in the blood, is positively associated with the risk of type 2 diabetes mellitus (T2DM). However, in the T2DM population, the association of SUA fluctuation ( $$\Delta$$ Δ SUA) with the functional outcome of ischemic stroke (IS) is still unclear. Accordingly, this study aimed to assess the correlation between $$\Delta$$ Δ SUA and short-term IS functional outcomes in T2DM patients. Methods All T2DM patients diagnosed with IS in the China National Stroke Registry III were included. $$\Delta$$ Δ SUA, which was defined as the difference between the SUA levels at baseline and 3 months after symptom onset, was classified into two groups, i.e., elevated $$\Delta$$ Δ SUA ( $$\Delta$$ Δ SUA > 0) and reduced $$\Delta$$ Δ SUA ( $$\Delta$$ Δ SUA $$\le$$ ≤ 0). The outcomes measured using the Modified Rankin Scale (mRS) were scored from 0 to 6, and poor functional outcome was defined as an mRS score of 3–6 at 3 months after IS. Results Among the 1255 participants (mean age: 61.6 ± 9.8 years), 64.9% were men. Patients with elevated $$\Delta$$ Δ SUA had a lower incidence of poor functional outcomes at 3 months. Compared with reduced $$\Delta$$ Δ SUA, elevated $$\Delta$$ Δ SUA at 0–50 μmol/L (odds ratio [OR] = 0.46, 95% confidence interval [CI] = 0.28–0.78, p = 0.004) and 50–100 μmol/L (OR = 0.40, 95% CI = 0.21–0.77, p = 0.006) was significantly correlated with a reduced risk of poor functional outcomes at 3 months. Conclusion This study showed that a moderate increase in $$\Delta$$ Δ SUA in the range of 0–100 μmol/L at 3 months after IS might be beneficial in T2DM adults and more studies are warranted to confirm this.

Abstract Background In large-scale high-throughput sequencing projects and biobank construction, sample tagging is essential to prevent sample mix-ups. Despite the availability of fingerprint panels for DNA data, little research has been conducted on sample tagging of whole genome bisulfite sequencing (WGBS) data. This study aims to construct a pipeline and identify applicable fingerprint panels to address this problem. Results Using autosome-wide A/T polymorphic single nucleotide variants (SNVs) obtained from whole genome sequencing (WGS) and WGBS of individuals from the Third China National Stroke Registry, we designed a fingerprint panel and constructed an optimized pipeline for tagging WGBS data. This pipeline used Bis-SNP to call genotypes from the WGBS data, and optimized genotype comparison by eliminating wildtype homozygous and missing genotypes, and retaining variants with identical genomic coordinates and reference/alternative alleles. WGS-based and WGBS-based genotypes called from identical or different samples were extensively compared using hap.py. In the first batch of 94 samples, the genotype consistency rates were between 71.01%-84.23% and 51.43%-60.50% for the matched and mismatched WGS and WGBS data using the autosome-wide A/T polymorphic SNV panel. This capability to tag WGBS data was validated among the second batch of 240 samples, with genotype consistency rates ranging from 70.61%-84.65% to 49.58%-61.42% for the matched and mismatched data, respectively. We also determined that the number of genetic variants required to correctly tag WGBS data was on the order of thousands through testing six fingerprint panels with different orders for the number of variants. Additionally, we affirmed this result with two self-designed panels of 1351 and 1278 SNVs, respectively. Furthermore, this study confirmed that using the number of genetic variants with identical coordinates and ref/alt alleles, or identical genotypes could not correctly tag WGBS data. Conclusion This study proposed an optimized pipeline, applicable fingerprint panels, and a lower boundary for the number of fingerprint genetic variants needed for correct sample tagging of WGBS data, which are valuable for tagging WGBS data and integrating multi-omics data for biobanks.

Wrought magnesium alloys with high-strength and high-ductility property have been attracting more and more interest in automotive and aerospace industries, and the conventional Mg-Zn based alloys are widely accepted as the representative one. Development of Mg-Zn based alloys with rare earth (RE) element additions and with modified preparation processes domains the evolution of their potential structural applications. Nonetheless, their highest tensile yield strength (TYS) is still in the order of 400 MPa, clearly lower than that of the Mg-RE based alloys, in the order of 500 MPa, much less compared with the high-strength aluminum alloys and titanium alloys. According to many previous investigations, two relatively efficient methods to improve mechanical properties of ZK based alloys were revealed as: alloying by RE and optimizing wrought processes. However, comprehensive combinative investigation was hitherto not conducted to date. Thus, it is imperative in the next work to further improve the yield strength to the order of 500 MPa or much higher, and simultaneously modify the tension-compression strength asymmetry of the ZK series alloys. Maybe, developing novel wrought processes for ZK60 (+ RE) alloys or exploring appropriate pre-dealing treatments before or after the wrought processes might be an important approach to accomplish a much higher level of balance of high-strength and high-ductility.

The interfacial bonding capacity between cement emulsified asphalt composite binder (CEACB) and reclaimed asphalt pavement (RAP) plays a critical role in improving the pavement performance of cold recycled asphalt emulsion mixtures (CRAEMs). This study aims to investigate the formation and development of the interfacial bonding capacity between CEACB and RAP. First, the dynamic wettability and the spreading behaviors of CEACB on RAP surfaces were explored according to the surface free energy theory. Second, digital image processing (DIP) technology was employed to recognize interfacial failure patterns. Lastly, the influence of internal and external factors on the interfacial bonding capacity between CEACB and RAP during the curing process was analyzed via grey relational analysis (GRA). The results indicate that a moderate cement content with a mass ratio of asphalt to cement equivalent to 1.0 can significantly enhance the wettability of CEACB on RAP surfaces. By appropriately prolonging the curing time and controlling the curing temperature, it is possible to increase the bonding strength between CEACB and RAP. Additionally, a strong correlation exists between initial wettability and ultimate bonding capacity during the bonding strength curing process. The good wettability that developed in the initial stage of interfacial strength formation relates to the decreased spalling rate of CEACB on the RAP surface. This study is not only devoted to understanding the mechanisms that can enhance CRAEM performance but also provides important guidance for practical engineering applications of cold recycled asphalt pavements.

We propose and demonstrate a curvature sensor based on long-period fiber grating (LPFG) with asymmetric index modulation. The LPFG is fabricated in single-mode fiber with femtosecond laser micromachining. The grating structure is not introduced in the central fiber core, but is located off-axis with a distance of a few micrometers. Experimental results indicate that the offset distance has direct influence on the grating spectra. By utilizing such an asymmetric structure, two-dimensional vector curvature sensing can be realized. For an LPFG with an offset distance of 6 μm, the curvature sensitivity is around 29 nm/m−1 in the 0° and 180° direction and about 20 nm/m−1 in the 90° and 270° direction. The difference in curvature sensitivity in different bending directions makes the sensor capable of distinguishing the curvature orientation. The temperature response of the sensor is also experimentally investigated, and results indicate that the sensor has a very low temperature cross-sensitivity of 0.003 m−1/°C. The characteristics of high curvature sensitivity, two-dimensional bending direction identification, and compact structure make the device an ideal candidate to be applied in the field of power grid health monitoring and intelligent robotics.

The reservoir space of tight reservoir with low-permeability is controlled by micro and nano scale pores, making the influence of capillary force significantly enhanced. Therefore, understanding the microscopic charging characteristics of oil and gas is the basis for analyzing the migration and accumulation of reservoirs. In this paper, the self-developed on-line three-dimensional microscopic imaging system for core fluid displacement is used to observe the oil charging process of two tight reservoir samples, and core-level and pore-level quantitative analysis methods for oil content characteristics are proposed. Taking the on-line nuclear magnetic resonance testing with the same process of displacement as a contrast, it is revealed that the average difference of on-line 2D DR (Digital Radiography) images at different times can be used to evaluate the overall oil content change of the sample. The calculation method of pore level fluid saturation based on high-precision pore network extraction algorithm realizes the quantitative evaluation of oil charging degree of the CT resolved pores and pore throats. The combination of multi-level data and different methods can meet the different needs of different researches on dynamic feature capture, pore resolution and imaging field of vision. The analysis results show that the oil saturation of two rock samples from different tight reservoirs in the Ordos Basin increases rapidly at the beginning and slows down later with oil injection increasing. At the same injection flow rate, the oil saturation of the sample with higher permeability increases faster at the initial oil charging stage, making its final oil saturation higher. With the increase of oil injection, the oil saturation of macropores in the sample with higher permeability continuously increases, while that of macropores in the sample with lower permeability shows a U-shaped change, showing the characte-ristics of repeated occupation of pores by oil and water.

Bio3D is a family of R packages for the analysis of biomolecular sequence, structure, and dynamics. Major functionality includes biomolecular database searching and retrieval, sequence and structure conservation analysis, ensemble normal mode analysis, protein structure and correlation network analysis, principal component, and related multivariate analysis methods. Here, we review recent package developments, including a new underlying segregation into separate packages for distinct analysis, and introduce a new method for structure analysis named ensemble difference distance matrix analysis (eDDM). The eDDM approach calculates and compares atomic distance matrices across large sets of homologous atomic structures to help identify the residue wise determinants underlying specific functional processes. An eDDM workflow is detailed along with an example application to a large protein family. As a new member of the Bio3D family, the Bio3D-eddm package supports both experimental and theoretical simulation-generated structures, is integrated with other methods for dissecting sequence-structure-function relationships, and can be used in a highly automated and reproducible manner.
