Abstract

Tetranychus cinnabarinus (Boisduval), a worldwide acarine pest, has garnered significant attention in recent studies, particularly regarding the role of ecdysteroids. Ecdysteroids (moulting hormone) played key roles in regulating the important physiological processes of arthropods, such as ecdysis, metamorphosis reproduction and immunity. However, the underlying mechanisms of ecdysteroid biosynthesis and signalling have not been elucidated in T. cinnabarinus. Therefore, in this study, we utilized RACE technology in conjunction with RT‐PCR to obtain the complete sequence of three Halloween genes disembodied (dib), shadow (sad), shade (shd) and ecdysone receptor (EcR). Moreover, we analysed the expression level of these target genes in T. cinnabarinus, which was found to be expressed at all developmental stages. When using sublethal concentration of chromafenozide for bioassay on nymph of T. cinnabarinus, as the concentration increased, the ecdysis rate, mortality and ecdysteroid content were increased, and the moulting duration was decreased. We also characterized the expression on nymph of T. cinnabarinus after treatment with sublethal concentrations of chromafenozide, which were up‐regulated. Based on these findings, we concluded that the treatment of sublethal chromafenozide enhanced Halloween gene and nuclear receptor gene expression, aided in the investigation of the molecular regulatory mechanism for moulting of T. cinnabarinus, and provided a theoretical basis for new agents. 14. Fatty Acid Elongase Gene PcELO7is Essential for Lipid Accumulation and Fecundity of Panonychus citri(Acari: Tetranychidae)

Abstract

RNA interference (RNAi) has been proposed as a promising strategy for sustainable and ecofriendly pest control. The insect cuticle lipids were deposited on the body surface and functioned as a defense against chemical xenobiotics. They consisted of aliphatic compounds, including free fatty acids (FFAs). However, elongase of very long chain fatty acids (ELOs) is essential for FFA biosynthesis; the function of ELO is still unknown in many arthropods, including Panonychus citri(P. citri). In this study, three ELOs were cloned. Developmental-specific mRNA expression results revealed that three PcELOswere highly expressed in egg and adult females. Whereas PcELO7was dominantly expressed in adult females. Under spirobudiclofen stress, ELOs mRNA expression had different changes, and PcELO7was down-regulated. The silencing of PcELO7resulted in a dramatic reduction of oviposition and hatchability. Significant reduction of FFA contents was also examined within PcELO7-repressed P. citri. In addition, we found that PcELO7mRNA levels were related to fecundity and could affect triacylglycerol (TG) contents. The findings demonstrated that the introduction of dsPcELO7via oral feeding induced the RNA interference-mediated silencing of a special target gene and could result in mortality and reproduction. In conclusion, PcELO7is a special RNAi target for P. citricontrol, and its lethal mechanism might be disturbing lipids biosynthesis.

41. Transcriptome analysis of Tetranychus cinnabarinus responses to exposure of an insecticide (diflubenzuron).

Abstract

Diflubenzuron, a benzoylphenylurea insecticide that interferes with chitin biosynthesis, causes arthropods to moult abnormally and die. However, its mechanism of action in Tetranychus cinnabarinus is still unclear. In order to explore the effects of different sublethal concentrations of diflubenzuron on T. cinnabarinus, we conducted a high-throughput RNA-seq technology to identify the variations in transcriptomic profile of T. cinnabarinus larvae. The results revealed that 470 and 49 differentially expressed genes were identified in LC50- and LC70-treated groups, comparing with the control. We also identified and analyzed the detoxification enzymes involved in the transcritome of T. cinnabarinus, including 34 cytochrome P450 genes, 17 glutathione-s-transferase genes (GSTs), 12 acetylcholinesterase genes (AChEs) and 9 ABC transporter genes. In addition, differentially expressed genes analysis showed that the gene expression levels of detoxification enzymes were generally enhanced. At the same time, seven and 11 genes were involved in chitin synthesis and degradation ways, respectively. The expression level of most genes involved in chitin synthesis and degradation pathway were generally up-regulated after exposure to sublethal concentrations of diflubenzuron. Moreover, for transcriptome validation, the mRNA expression results of ten specially expressed genes by quantitative real-time PCR demonstrated that these gene expression trends were consistent with that of the transcriptome data. Together, all these results suggested that sublethal concentrations of diflubenzuron exposure affected gene expression of major detoxification enzymes and chitin metabolism genes in T. cinnabarinus. These findings may be helpful to further understand the possible molecular mechanism of benzoylphenylurea insecticides in T. cinnabarinus, as well as in other spider mites. [ABSTRACT FROM AUTHOR]

Abstract

As an important predatory mite, Stratiolaelaps scimitus (Womersley) is commercially produced to control pests of edible fungi and greenhouse. In this study, the complete mitochondrial (mt) genome of S. scimitus was sequenced by Illumina HiSeq, which comparatively analysed all reported mt genome rearrangement of mesostigmatid species. The mt genome of S. scimitus was a typical circular molecule of 16,468 bp (GenBank: MK912042) and contained the complete set of 37 genes as other metazoan mitochondrial genomes. The AT-and GC-skew on the majority strand was −0.061 and 0.365, respectively, which were reversed compared to those found in most metazoan mitochondrial genomes. Like other mesostigmatid mites, many gene rearrangements occurred in the mt genome of S. scimitus. The six mesostigmatid families containing rearranged mt genomes were characterized by unique gene order, except Diplogyniidae and Parasitidae. Shared gene clusters were found in four mesostigmatid families (Laelapidae, Blattisociidae, Phytoseiidae, and Macrochelidae), indicating that they are synapomorphic for these families.
