5. Correlations of analgesic dosage of morphine with SLC6A4 gene polymorphisms in patients with lung cancer.

Abstract

OBJECTIVE: To investigate the correlations of analgesic dosage of morphine with solute carrier family 6 member 4 (SLC6A4) gene polymorphisms in patients with lung cancer. PATIENTS AND METHODS: A total of 200 lung cancer patients without cancer pain were selected as painless group, and another 200 lung cancer patients with cancer pain as cancer pain group. Visual Analogue Scale (VAS) was applied to grade the pain, the patients in cancer pain group were treated with morphine, and the dosage of morphine within 24 h was recorded. Then, the genomic deoxyribonucleic acid (DNA) was extracted from the peripheral blood of the research subjects, and the polymorphisms rs1042173 and rs7224199 of SLC6A4 gene were detected. RESULTS: There was a difference in the genotype distribution of SLC6A4 gene rs7224199 between painless group and cancer pain group (p=0.004), and the frequency of GG genotype was remarkably higher in cancer pain group [75 (0.375)]. The frequency of heterozygous model AC of rs1042173 and recessive model GT + TT of rs7224199 in cancer pain group was evidently lower than that in painless group (p=0.048, p=0.043). Besides, the lung cancer patients in cancer pain group had markedly lower frequency of AG haplotype (p=0.000), but notably higher frequency of AT (p=0.000) and CG (p=0.000) haplotypes of SLC6A4 gene rs1042173 and rs7224199 than those in painless group. No significant differences in genotypes of SLC6A4 gene rs1042173 (p=0.241) and rs7224199 (p=0.316) were detected among the degrees of cancer pain in cancer pain group. The analgesic dosage of morphine for the lung cancer patients was prominently correlated with the genotypes of SLC6A4 gene rs1042173 in cancer pain group. Moreover, in cancer pain group, there were significant differences in the dosage within 24 h (p=0.025), at 24 h after weight correction (p=0.001) and at 24 h after correction of weight and body surface area (p=0.000) among the genotypes, and the morphine dosage for the patients with CC genotype was significantly lower. Furthermore, the morphine dosage within 24 h (p=0.047), at 24 h after weight correction (p=0.042) and at 24 h after correction of weight and body surface area (p=0.031) were distinctly associated with the haplotypes of SLC6A4 gene in cancer pain group, of which the patients with CT haplotype were administered with a remarkably lower morphine dosage. CONCLUSIONS: The morphine dosage for analgesia has significant correlations with SLC6A4 gene polymorphisms in patients with lung cancer. [ABSTRACT FROM AUTHOR]

Abstract

Purpose: The purpose of this study was to determine the prevalence of human papillomavirus (HPV) in the population in Kunming, Yunnan, China, before and after the introduction of HPV preventive vaccines., Materials and Methods: In total, 28,959 patients were enrolled in this study between 1 January 2016 and 31 August 2019. HPVs were genotyped using a flow-through hybridisation technique, and differences in HPV infection rates before and after the introduction of an HPV vaccine were determined., Results: The prevalence of HPV before and after the introduction of HPV vaccines was 17.74% and 17.11%, respectively. This difference was not statistically significant (χ 2 = 1.920, P > 0.05). The HPV infection rates showed a bimodal U-shaped curve for all age groups. The most common genotypes of HPV detected were HPV52, HPV16, HPV58 and HPV39., Conclusion: Although the overall HPV infection rate in Kunming did not change significantly after the introduction of HPV vaccines, differences in HPV infection rates and multi-typic HPV infection rates were evident in certain age groups., Competing Interests: None

Abstract

To better understand the molecular mechanisms of multiple-allele-inherited male sterility in Chinese cabbage (Brassica rapa ssp pekinensis), differentially expressed genes in fertile and sterile plants must be isolated. We used cDNA-AFLP analysis to isolate differentially expressed genes in fertile and sterile buds of the two-type line, AB01. Sixteen high-quality sequences were generated, 11 of which were up-regulated in fertile buds, and five of which were up-regulated in sterile buds. Based on BLAST screening and functional annotation, these genes have homology with genes encoding known flower- or bud-specific proteins, metabolism-related proteins and cell-structure proteins. In addition, the full-length cDNA sequences of the actin gene were cloned from the cabbage plants by RACE and used as an internal standard for semi-quantitative reverse transcription-PCR. Expression of three flower- or bud-specific differentially expressed transcript-derived fragments in fertile and sterile buds was examined using RT-PCR; the expression patterns of these genes were similar to the patterns observed in the cDNA-AFLP analysis.

9. Blood lead and cadmium in a general population in Jinan City, China.

Abstract

In 1991, blood samples were obtained from 150 adult Jinan citizens (74 men and 76 women at the ages of 20 to 57 years) who had no known occupational exposure to heavy metals. Age, sex, two social habits of smoking and drinking (in terms of daily consumption) and negative occupational history were examined in a medical interview. The samples were analyzed for lead (Pb-B) and cadmium (Cd-B) with a flame atomic absorption spectrometer. The geometric mean (GM) Pb-B and Cd-B were 92.3 and 0.94 micrograms/l, respectively, among 39 nonsmoking men, whereas the counterpart values were 123.4 micrograms/l and 2.61 micrograms/l among 35 smoking men (mean consumption; > 15 cigarettes/day); the difference was significant both for Pb-B and Cd-B. Comparison between 39 male and 76 female nonsmokers showed that Pb-B was significantly higher in men (92.3 micrograms/l) than in women (71.6 micrograms/l, whereas the difference in Cd-B (0.94 micrograms/l) for men versus 0.83 micrograms/l for women) was insignificant. When the women were classified by decade of age and Cd-B were compared, there was a trend of age-dependent increase in Cd-B from 0.60 micrograms/l in 20s to 1.24 micrograms/l in 40s, followed by no further increase at higher ages. Age-dependent changes were not remarkable in Pb-B in women, or Cd-B and Pb-B in men. No significant time-dependent changes were observed when the present results were compared with the results from two similar studies conducted in 1983 and 1985, respectively.
