Background: Ticks are widely distributed throughout China and are the second most prevalent pathogen vectors in the world, following only mosquitoes. Tick bites can lead to Lyme disease, forest encephalitis, and other illnesses that may result in death under severe circumstances. Materials and methods: Ticks collected from March 2021 to May 2023 were pooled and used in metatranscriptomic analyses to gain insight into the diversity and distribution of tick-borne viruses in Inner Mongolia. Next-generation sequencing (NGS) outcomes were validated, and viral prevalence across distinct tick species was determined through the application of polymerase chain reaction (PCR) paired with Sanger sequencing., Results: A total of 20 RNA viruses belonging to at least 8 families, including Chuviridae, Flaviviridae, Solemoviridae, Nairoviridae, Partitiviridae, Phenuiviridae, Rhabdoviridae, and Totiviridae, and to unclassified families were identified by NGS. Five of the identified RNA viruses (Nuomin virus, Yezo virus, tick-borne encephalitis virus, Alongshan virus, and Beiji nairovirus) are considered human pathogens. A potential human pathogen, Mukawa virus, was also among the identified viruses. Ixodes persulcatus carried a significantly greater number of viral species than did Dermacentor nuttalli, Hyalomma marginatum, and Haemaphysalis concinna. The prevalence of coinfection with multiple viruses differed in I. persulcatus from Hinggan League and Hulun Buir, and Beiji nairovirus was the codominant virus species., Conclusions: There is a remarkable diversity of RNA viruses harboured by ticks in Inner Mongolia, with variations observed in the distribution of these tick-borne viruses across different regions and tick hosts., Competing Interests: The authors have declared that no competing interests exist., (Copyright: © 2024 Su et al. Ticks can carry multiple pathogens, and Inner Mongolia's animal husbandry provides excellent environmental conditions for ticks. This study characterized the microbiome of ticks from different geographical locations in Inner Mongolia; 905 Dermacentor nuttalli and 36 Ixodes persulcatus were collected from sheep in three main pasture areas and from bushes within the forested area. Mixed DNA samples were prepared from three specimens from each region and tick species. Microbial diversity was analyzed by 16S rRNA sequencing, and α and β diversity were determined. The predominant bacterial genera were Rickettsia (54.60%), including Rickettsiales bacterium Ac37b (19.33%) and other Rickettsia (35.27%), Arsenophonus (11.21%), Candidatus Lariskella (10.84%), and Acinetobacter (7.17%). Rickettsia bellii was identified in I. persulcatus, while Rickettsiales bacterium Ac37b was found in D. nuttalli from Ordos and Chifeng. Potential Rickettsia and Anaplasma coinfections were observed in the Ordos region. Introduction: Menstrual discomfort affects women's quality of life, which is an important public health issue. Evidence confirming the link between passive smoking and menstrual discomfort is limited. Therefore, the aim of this study is to investigate the aforementioned topic on the basis of a cross-sectional study of 2,571 non-smoking Chinese nurses., Methods: Demographic information and passive smoking were assessed using a self-administered questionnaire. Menstrual discomfort was characterized as dysmenorrhea, illness or weakness, bed rest, and restlessness during menstruation, which was assessed using a modified version of the Cornell Medical Index-Health Questionnaire. Multivariate-adjusted odds ratio (OR) and 95% confidence intervals (CIs) were estimated using the logistic regression model., Results: A total of 1:195 nurses (46.48%) were exposed to passive smoking. Compared with non-passive smoking nurses, passive smoking nurses were more likely to have menstrual discomfort symptoms (72.38 vs. 64.39%), especially symptoms of dysmenorrhea (49.54 vs. 42.08%), illnesses or weakness (48.28 vs. 42.08%), and restlessness during menstruation (53.05 vs. 46.22%). Exposure to passive smoking was significantly associated with menstrual discomfort (OR = 1.41, 95%CI: 1.19-1.67), especially symptoms of dysmenorrhea (OR = 1.32, 95%CI: 1.13-1.56), illness or weakness (OR = 1.24, 95%CI: 1.06-1.46), and restlessness (OR = 1.26, 95%CI: 1.08-1.48) during menstruation. This study aimed to investigate the association between passive smoking and physical and psychological health in Chinese nurses. Participants of this cross-sectional study comprised 2,484 non-smoking nurses. Passive smoking and demographic information were assessed using a self-administered questionnaire. Physical, psychological, and overall health status of nurses were measured using the Cornell Medical Index (CMI) health questionnaire. Multivariate-adjusted odds ratio (OR) and 95% confidence interval (CI) for nurses' health were estimated by exposure to passive smoking using unconditional logistic regression models. A total of 1,219 nurses (49.07%) were exposed to passive smoking. Of these, 609 (24.52%), 160 (6.44%), and 587 (23.63%) nurses had poorer physical, mental, and overall health, respectively. After adjusting for other confounding factors, compared with the non-passive smoking group, passive smoking was associated with poor physical (OR = 1.51, 95% CI: 1.25-1.83), mental (OR = 1.48, 95% CI: 1.07-2.07), and overall (OR = 1.58, 95% CI: 1.30-1.93) health of nurses, respectively. We also carried out subgroup analyses stratified by age, department, and professional title, which showed that most findings supported the main results. Laser powder bed fusion (LPBF) is being increasingly used in the fabrication of complex-shaped structure parts with high precision. It is easy to form martensitic microstructure in Ti-6Al-4V alloy during manufacturing. Pre-heating the powder bed can enhance the thermal field produced by cyclic laser heating during LPBF, which can tailor the microstructure and further improve the mechanical properties. In the present study, all the Ti-6Al-4V alloy samples manufactured by LPBF at different powder bed temperatures exhibit a near-full densification state, with the densification ratio of above 99.4%. When the powder bed temperature is lower than 400 °C, the specimens are composed of a single α' martensite. As the temperature elevates to higher than 400 °C, the α and β phase precipitate at the α' martensite boundaries by the diffusion and redistribution of V element. In addition, the α/α' lath coarsening is presented with the increasing powder bed temperature. The specimens manufactured at the temperature lower than 400 °C exhibit high strength but bad ductility. Moreover, the ultimate tensile strength and yield strength reduce slightly, whereas the ductility is improved dramatically with the increasing temperature, when it is higher than 400 °C.

The development of enantioselective alkyl-alkyl cross-couplings with coinstantaneous formation of a stereogenic center without the use of sensitive organometallic species is attractive yet challenging. Herein, we report the intermolecular regio- and enantioselective formal hydrofunctionalizations of acrylamides, forging a stereogenic center α-position to the newly formed C sp3 -C sp3 bond for the first time. The use of a newly developed chiral ligand enables the electronically-reversed formal hydrofunctionalizations, including hydroalkylation, hydrobenzylation, and hydropropargylation, offering an efficient way to access diverse enantioenriched amides with a tertiary α-stereogenic carbon center which is facile to racemize. This operationally simple protocol allows for the anti-Markovnikov enantioselective hydroalkylation, and unprecedented hydrobenzylation, hydropropargylation under mild conditions with excellent functional group compatibility, delivering a wide range of amides with excellent levels of enantioselectivity., (© 2020 Wiley-VCH GmbH.)

A three-dimensional (3D) flower-shaped Li 4 Ti 5 O 12 -graphene (Gr) hybrid micro/nanostructures and pine needles derived carbon nanopores (PNDCN) has been prepared by using the effective hydrothermal process. Due to the unique micro/nanostructures which can provide abundant surface active sites, the obtained 3D Li 4 Ti 5 O 12 -Gr displays a high specific capacitance of 706.52 F g -1 at 1 A g -1 . The prepared PNDCN also exhibits high specific capacitance of 314.50 F g -1 at 1 A g -1 benefiting from its interconnected honeycomb-like hierarchical and open structure, which facilitates the diffusion and reaction of electrolyte ions and enables an isotropic charging/discharging process. An asymmetric supercapacitor utilizing Li 4 Ti 5 O 12 -Gr as positive electrode and PNDCN as negative electrode has been fabricated, it delivers a high energy density of 35.06 Wh kg -1 at power density of 800.08 W kg -1 and outstanding cycling stability with 90.18% capacitance retention after 2000 cycles. Various V 2 O 5 three-dimensional nanostructures are synthesized using a facile template-free hydrothermal method and evaluated for use as supercapacitor electrode materials. As a result, the yolk-shell structure assembled from ultrathin nanosheets shows the best electrochemical performance, with a specific capacitance of 704.17 F g -1 at 1.0 A g -1 and a high capacity retention of 89% over 4000 cycles at 3.0 A g -1 . In addition, a continuous three-dimensional porous coralline-shaped carbon is synthesized from osmanthus and has a large Brunauer-Emmett-Teller surface area of 2840.88 m 2 g -1 . Then, an asymmetric supercapacitor is developed using the as-prepared yolk-shell V 2 O 5 as a positive electrode and the osmanthus derived coralline-shaped carbon as a negative electrode. This exhibits an energy density of 29.49 W h kg -1 at a power density of 800 W kg -1 with a good cycling performance that retains 90.6% of its initial capacity after 2000 cycles at 3.0 A g -1 . Furthermore, two cells in series can easily brightly light up a light-emitting diode (3 V), further demonstrating the great potential of the prepared materials for high-performance supercapacitor devices.

An ultrasensitive electrochemical biosensor for microRNA (miRNA) is developed based on tungsten oxide-graphene composites coupling with catalyzed hairpin assembly target recycling and enzyme signal amplification. WO3-Gr is prepared by a simple hydrothermal method and then coupled with gold nanoparticles to act as a sensing platform. The thiol-terminated capture probe H1 is immobilized on electrode through Au-S interaction. In the presence of target miRNA, H1 opens its hairpin structure by hybridization with target miRNA. This hybridization can be displaced from the structure by another stable biotinylated hairpin DNA (H2), and target miRNA is released back to the sample solution for next cycle. Thus, a large amount of H1-H2 duplex is produced after the cyclic process. At this point, a lot of signal indicators streptavidin-conjugated alkaline phosphatase (SA-ALP) are immobilized on the electrode by the specific binding of avidin-biotin. Then, thousands of ascorbic acid, which is the enzymatic product of ALP, induces the electrochemical-chemical-chemical redox cycling to produce a strongly electrochemical response in the presence of ferrocene methanol and tris (2-carboxyethyl) phosphine. Under the optimal experimental conditions, the established biosensor can detect target miRNA down to 0.05fM (S/N=3) with a linear range from 0.1fM to 100pM, and discriminate target miRNA from mismatched miRNA with a high selectivity., (Copyright © 2016 Elsevier B.V. Tungsten oxide (WO 3 ), which was originally poor in capacitive performance, is made into an excellent electrode material for supercapacitors by dispersing it on graphene (Gr). The obtained Gr-WO 3 hybrids are characterized by X-ray diffraction, Raman spectroscopy, high-resolution transmission electron microscopy and scanning electron microscopy techniques, and evaluated as electrode materials for high-performance supercapacitors by cyclic voltammetry, galvanostatic charge-discharge curves and electrochemical impedance spectroscopy. A great improvement in specific capacitance is achieved with the present hybrids, from 255 F g -1 for WO 3 nanoparticles to 580 F g -1 for Gr-WO 3 hybrids (scanned at 1 A g -1 in 2 M KOH over a potential window of 0 to 0.45 V). The Gr-WO 3 hybrid exhibits an excellent high rate capability and good cycling stability with more than 92% capacitance retention over 1000 cycles at a current density of 5 A g -1 . The enhancement in supercapacitor performance of Gr-WO 3 is not only attributed to its unique nanostructure with large specific surface area, but also its excellent electro-conductivity, which facilitates efficient charge transport and promotes electrolyte diffusion. As a whole, this work indicates that Gr-WO 3 hybrids are a promising electrode material for high-performance supercapacitors.

Amyloid-β (Aβ), the major component of neuritic plaques in Alzheimer's disease (AD), is derived from sequential proteolytic cleavage of amyloid protein precursor (APP) by secretases. In this study, we found that cystatin C (CysC), a natural cysteine protease inhibitor, is able to reduce Aβ40 secretion in human brain microvascular endothelial cells (HBMEC). The CysC-induced Aβ40 reduction was caused by degradation of β-secretase BACE1 through the ubiquitin/proteasome pathway. In contrast, we found that CysC promoted secretion of soluble APPα indicating the activated non-amyloidogenic processing of APP in HBMEC. Further results revealed that α-secretase ADAM10, which was transcriptionally upregulated in response to CysC, was required for the CysC-induced sAPPα secretion. Knockdown of SIRT1 abolished CysC-triggered ADAM10 upregulation and sAPPα production. Taken together, our results demonstrated that exogenously applied CysC can direct amyloidogenic APP processing to non-amyloidgenic pathway in brain endothelial cells, mediated by proteasomal degradation of BACE1 and SIRT1-mediated ADAM10 upregulation. Our study unveils previously unrecognized protective role of CysC in APP processing.

The objective of this study was to examine the role and possible mechanisms of toll-like receptor 2 (TLR2) in high-mobility group box chromosomal protein 1 (HMGB1)-induced mouse mesangial cell (MMC) proliferation and glomeruli proliferation of MRL/Fas(lpr) mice. First, the expression of proliferating cell nuclear antigen (PCNA), TLR2 and Forkhead box protein O1 (FoxO1) messenger RNA (mRNA) and protein in the glomeruli of MRL/Fas(lpr) mice was quantified, and the correlation with cell proliferation of glomeruli was analyzed. Then, lipopolysaccharide (LPS), TLR2 neutralization antibody, and small hairpin TLR2 (shTLR2) were used to confirm the role of TLR2 in HMGB1-induced MMC proliferation. Furthermore, wild-type FoxO1 (WT-FoxO1) vector was used to investigate the effect of FoxO1 pathway on HMGB1-induced MMC proliferation. Finally, electroporation was used to knockdown TLR2 in the glomeruli of MRL/Fas(lpr) mice, and renal function, FoxO1, and PCNA expression were detected. The results showed that the TLR2 expression was upregulated and FoxO1 expression was decreased in the glomeruli of MRL/Fas(lpr) mice, and these effects were significantly correlated with cell proliferation of the glomeruli. In vitro, the TLR2 neutralization antibody and the WT-FoxO1 vector, both reduced the MMC proliferation levels induced by HMGB1. The TLR2 neutralization antibody also blocked the HMGB1-dependent activation of the FoxO1 pathway and cell proliferation. In addition, transfection with shTLR2 decreased the proliferation levels and PCNA expression induced by HMGB1. In vivo, treatment with shTLR2 significantly reduced the PCNA expression in the glomeruli of MRL/Fas(lpr) mice and improved renal function. In addition, treatment with shTLR2 or blocking of TLR2 also reduced the translocation of FoxO1. Thus, TLR2 plays a critical role in HMGB1-induced glomeruli cell proliferation through the FoxO1 signaling pathway in lupus nephritis.

Context: The elevated low-density-lipoprotein cholesterol (LDL-C) in menopausal women is associated with higher risks of cardiovascular diseases., Objective: The aim of this study is to investigate the influence and mechanism by which high postmenopausal FSH levels affect lipid profiles., Methods: The serum FSH and lipid levels were examined in 400 Chinese postmenopausal women. The FSH receptor (FSHR) expression was identified in liver and HepG2 cells by PCR and Western blotting. The effects of FSH on lipid metabolism were confirmed in an ovariectomized mouse model by using GnRH agonist with or without additional FSH to mimic different FSH status. LDL receptor (LDLR), a necessary factor for clearance of LDL-C through endocytosis, was examined by PCR and Western blotting., Results: The postmenopausal women with higher serum FSH (≥78.3 IU/L at baseline) had higher serum total cholesterol and LDL-C levels than those women with FSH levels of 40-78.3 IU/L (P < .01). The improvements of total cholesterol and LDL-C levels were more significant in higher FSH women group after treatment with hormone replacement therapy. It was only in the women whose FSH levels were reduced more than 30% after hormone replacement therapy who showed significant improvement of lipid levels. Ovariectomized mice had high serum FSH and lipids levels and reduced hepatic LDLR expression. In HepG2 cells, FSH inhibited the LDLR in a dose- and time-dependent manner, and the FSHR knockdown with specific siRNA reversed the lower LDLR induced by FSH., Conclusions: FSH may interact with its receptors in hepatocytes and reduce LDLR levels, which subsequently attenuates the endocytosis of LDL-C, resulting in an elevated circulating LDL-C level.

Background: Neuroblastoma is an embryonic neoplasm originating from the neural crest with cellular heterogeneity as one of its oncobiological characteristics. This study was undertaken to determine whether human neuroblastoma contains stem cell-like cells., Methods: Twenty patients with neuroblastoma who have been treated in our hospital since January 2005 were divided into pre-operative chemotherapy (10 patients) and non-chemotherapy (10) groups. Tumor specimens of the patients were taken and paraffin sections were made. The expressions of stem cell markers CD133, ABCG2, CD117 and nestin were immunohistochemically detected in the specimens. Neuroblastoma cells were stained with Hoechst 33342 and PI. The side population (SP) cells were analyzed by the fluorescence-activated cell sorter. The disparity drug resistance to cisplatin (DDP) of SP and non-SP cells was measured with MTT colorimetric assay. The oncogenicity of SP and non-SP cells was identified in nude mice., Results: There was no significant difference in the expression intensity of CD117 and nestin between the two groups of specimens (P>0.05). There was a significant difference between the two groups in terms of the expression intensity of CD133 and ABCG2 (P<0.05). The SP cells accounted for 0.2%-1.3% of the total human neuroblastoma cells and were decreased to 0.1%-0.5% after verapamil treatment. The SP and non-SP cells showed disparity in cell growth experiment and drug resistance to DDP. Oncogenicity experiment revealed that nude mice could erupt tumor by an injection of l×10(6) SHSY5Y and WIV SP cells. However, the nude mice could not form tumor by an injection of l×10(6) non-SP cells., Conclusion: Neuroblastoma might contain cancer stem cell-like cells.

A mouse model was used to compare the number and function of mitochondria in oocytes and embryos obtained by superovulation and in a natural cycle (control group). The superovulation group had a higher number of total oocytes, MII oocytes, embryos with two pronuclei, 2-cell embryos and blastocysts than the control group (P<0.05 for all). The superovulation group had high proportion of MII oocytes with low number of mitochondrial (mt) DNA copies. The average number of mtDNA copies, ATP level and mitochondrial membrane potential (

α-Fe2O3 nanoparticles are uniformly coated on the surface of α-MoO3 nanorods through a two-step hydrothermal synthesis method. As the anode of a lithium-ion battery, α-Fe2O3@α-MoO3 core-shell nanorods exhibit extremely high lithium-storage performance. At a rate of 0.1 C (10 h per half cycle), the reversible capacity of α-Fe2O3@α-MoO3 core-shell nanorods is 1481 mA h g(-1) and a value of 1281 mA h g(-1) is retained after 50 cycles, which is much higher than that retained by bare α-MoO3 and α-Fe2O3 and higher than traditional theoretical results. Such a good performance can be attributed to the synergistic effect between α-Fe2O3 and α-MoO3 , the small size effect, one-dimensional nanostructures, short paths for lithium diffusion, and interface spaces. Soybean is an important crop not only for human consumption but also for its addition of nitrogen to the soil during crop rotation. China has the largest collection of cultivated soybeans (Glycine max) and wild soybeans (Glycine soja) all over the world. The platform of soybean core, mini core and integrated applied core collections has been developed in the past decade based on systematic researches which included the sampling strategies, statistical methods, phenotypic data and SSR markers. Meanwhile, intergrated applied core collections including accessions with single or integrated favorite traits are being developed in order to meet the demand of soybean breeding. These kinds of core collections provide powerful materials for evaluation of germplasm, identification of trait-specific accessions, gene discovery, allele mining, genomic study, maker development, and molecular breeding. Some successful cases have proved the usefulness and efficiency of this platform. The platform is helpful for enhancing utilization of soybean genetic resources in sustainable crop improvement for food security. The efficient utilization of this platform in the future is relying on accurate phenotyping methods, abundant functional markers, high-throughput genotyping platforms, and effective breeding programs.

Objective: To analyze the endemic situation of malaria in Shanxian County, Shandong Province, China in the last 10 years, so as to understand the epidemiological characteristics of the disease and provide the evidence for the establishment of control strategy to eliminate malaria., Methods: The data related to the endemic situation of malaria in Shanxian County from 2002 to 2011 were collected and analyzed., Results: From 2002 to 2011, 125 cases of malaria were reported, and the annual incidences were in the range of 0.18/100,000-2.00/100,000. Totally 60.80% of the cases focused on 3 townships, namely Fugang, Huanggang and Caozhuang. All the 125 cases were new vivax malaria cases, among them, 121 were locally infected, and the other 4 were imported. The epidemic season was from June to October, and the peak time was from July to September. There were more male cases than female cases. The ages of the cases were from 1 to 83 years old, and the incidence in those aged from 46-60 years old was higher., Conclusions: The malaria incidence in Shanxian County is on the rise, and the 3 townships in the south are the key areas for control. The control measures including the surveillance of floating population and mosquitoes, and malaria control health education should be strengthened.

High reversible lithium storage capacity is obtained from novel SnO2/ZnWO4 core-shell nanorods. At C/20 (20 h per half cycle) rate, the reversible capacity of SnO2/ZnWO4 core-shell nanorods is as high as 1000 mA h g(-1), much higher than that of pure ZnWO4, SnO2, or the traditional theoretical result of the simple mixture. Such performance can be attributed to the synergistic effect between the nanostructured SnO2 and ZnWO4. The distinct electrochemical activity of ZnWO4 nanorods probably activates the irreversible capacity of the SnO2 nanoparticles. The objective is to investigate the effect of high mobility group box-1 (HMGB1) on lipid deposition in γ-interferon (IFN-γ)-stimulated mouse mesangial cell line (MMC) and to determine whether the Janus kinase 2 and signal transducer and activator of transcription 1 (JAK2/STAT1) signaling pathway plays an important role in this process. We employed a control group, an IFN-γ stimulation group, and an IFN-γ + AG490 (JAK2 inhibitor) group. RNA interference aimed at sterol regulatory element-binding protein-1 (SREBP-1) or HMGB1 was used to investigate the effect of these proteins on IFN-γ-induced lipid deposition. Western blotting was used to detect phospho (p)-JAK2, JAK2, p-STAT1, STAT1, SREBP-1, fatty acid synthase (FAS), and HMGB1 protein expression. RT-PCR was used to detect SREBP-1, FAS, and HMGB1 mRNA. Oil Red O staining and the triglyceride assay were used to detect lipid deposition and triglyceride content. Results were as follows: 1) IFN-γ increased MMC cell lipid deposition, triglyceride content, and p-JAK2, p-STAT1, SREBP-1, and FAS expression; 2) SREBP-1 inhibition prevented FAS upregulation and attenuated IFN-γ-induced MMC cell lipid deposition and triglyceride content; 3) HMGB1 upregulated SREBP-1 and FAS mRNA and protein levels, which increased lipid deposition in MMC cells. Small interfering RNA-mediated inhibition of HMGB1 decreased SREBP-1 and FAS expression and lipid accumulation; 4) AG490 decreased upregulation of HMGB1 and p-JAK2/p-STAT1, as well as IFN-γ-induced lipogenesis. In conclusion, the JAK2/STAT1 pathway mediates IFN-γ-induced lipogenesis in MMC cells through regulation of HMGB1/SREBP-1/FAS.

A rapid and sensitive flow-injection chemiluminescence (CL) method for the determination of glipizide was developed on the basis of finding that glipizide can enhance the CL intensity of the luminol-K 3 Fe(CN) 6 system. In optimum condition, the increased CL intensity was directly proportional to the concentration of glipizide in the range from 4.0×10 -8  g/mL to 1.0×10 -6  g/mL and the detection limit was 1.0×10 -8  g/mL glipizide. The relative standard deviation (RSD) of the developed method was 2.1% with 11 repeated measurements of 1.0×10 -7  g/mL glipizide. The developed method has been successfully applied to the analysis of glipizide in its pharmaceutical preparations.

Genotyping that differentiates between strains of Mycobacterium tuberculosis based on variations in the genome of the organism is a useful tool for molecular epidemiological studies of tuberculosis. In the present study, we identified a combination of five mycobacterial interspersed repetitive unit (MIRU) loci and three variable numbers of tandem repeats (VNTR) loci of M. tuberculosis that showed a discriminatory power comparable to that of the 15 to 24 loci-based typing proposed for international standardisation in analysing 210 paediatric isolates obtained from Chongqing, China.

Reconstructing the evolutionary history of a set of species is an elementary problem in biology, and methods for solving this problem are evaluated based on two characteristics: accuracy and efficiency. Neighbor-joining reconstructs phylogenetic trees by iteratively picking a pair of nodes to merge as a new node until only one node remains; due to its good accuracy and speed, it has been embraced by the phylogeny research community. With the advent of large amounts of data, improved fast and precise methods for reconstructing evolutionary trees have become necessary. We improved the neighbor-joining algorithm by iteratively picking two pairs of nodes and merging as two new nodes, until only one node remains. We found that another pair of true neighbors could be chosen to merge as a new node besides the pair of true neighbors chosen by the criterion of the neighbor-joining method, in each iteration of the clustering procedure for the purely additive tree. These new neighbors will be selected by another iteration of the neighbor-joining method, so that they provide an improved neighbor-joining algorithm, by iteratively picking two pairs of nodes to merge as two new nodes until only one node remains, constructing the same phylogenetic tree as the neighbor-joining algorithm for the same input data. By combining the improved neighbor-joining algorithm with styles upper bound computation optimization of RapidNJ and external storage of ERapidNJ methods, a new method of reconstructing phylogenetic trees, FastJoin, was proposed. Experiments with sets of data showed that this new neighbor-joining algorithm yields a significant speed-up compared to classic neighbor-joining, showing empirically that FastJoin is superior to almost all other neighbor-joining implementations.

WO(3) nanorods are uniformly coated with SnO(2) nanoparticles via a facile wet-chemical route. The reversible capacity of SnO(2)/WO(3) core-shell nanorods is 845.9 mA h g(-1), higher than that of bare WO(3) nanorods, SnO(2) nanostructures, and traditional theoretical results. Such behavior can be attributed to a novel mechanism by which nanostructured metallic tungsten makes extra Li(2)O (from SnO(2)) reversibly convert to Li(+). This mechanism is confirmed by x-ray diffraction results. Our results open a way for enhancing the reversible capacity of alloy-type metal oxide anode materials.

Extremely high sensitivity and low working temperature of gas sensors are realized from SnO(2)/α-MoO(3) heterostructure nanobelts. Their sensitivity against 500 ppm ethanol is up to 67.76 at the working temperature of 300 °C, which is higher than that of bare α-MoO(3) and SnO(2) nanostructures. Also the working temperature can be lowered down to 120 °C. Such behaviors are attributed to the variation of the junction barrier at the SnO(2)/α-MoO(3) interface. The present results imply that heterostructured 1D nanomaterials may yield gas sensors with improved characteristics, and can be applied to a wide range of gas sensors.

Objective: To evaluate the application of spacer oligonucleotide typing (Spoligotyping) and mycobacterial interspersed repetitive unit-variable-number tandem repeat (MIRU-VNTR) analysis in the molecular-epidemiological study of tuberculosis and to discuss the characteristics of pediatric Mycobacterium (M.) tuberculosis strains in Chongqing., Methods: M. tuberculosis strains isolated and typed by Spoligotyping and MIRU-VNTR respectively, from the children patients in Chongqing and to compare the results from both methods, epidemiologically., Results: By means of Spoligotyping, 210 clinical isolates were divided into 2 gene groups, displaying 44 genotypes. Among them, the biggest group was M. tuberculosis Beijing family, including 130 strains (61.90%), using the Spoligotyping. From the results of MIRU-VNTR, 24 loci showed different polymorphism and the HGI of different loci set (12 old loci, 15 basic loci and 24-loci set) increased accordingly. The subtle difference in HGI was originated from one locus ETR-B, which was included in the 24-locus system. The diversity of each loci and MIRU-VNTR set for non-Beijing genotype strains was higher than that of the Beijing genotype strains., Conclusion: In this study, it was preliminarily confirmed the existence of high polymorphism of M. tuberculosis while the Beijing Family was the main genotype and main prevalent strain in children of Chongqing area. Spoligotyping prior to 15-locus with ETR-B combination seemed more suitable for the massive epidemiological investigation of pediatric tuberculosis patients.

Pd-ZnO nanoflowers with high uniformity were prepared via a novel one-step hydrothermal route. High sensitivity, fast response, high selectivity and low work temperature are obtained from Pd-ZnO nanoflower sensors. The sensitivity upon exposure to 300 ppm ethanol is up to 168 at 300  °C and maintains 2.6 at 120  °C. Such behaviors can be attributed to Schottky contact at the Pd/ZnO interface and catalytic activity of Pd nanoparticles. The present results open a way for uniform surface modification of one-dimensional nanostructures with Pd nanoparticles and further enhancing their gas sensing performance.

Aim: To investigate the association of the cyclooxygenases-2 (COX-2) polymorphisms and susceptibility to gastric cancer (GC) by means of meta-analysis., Methods: Publications addressing the association between polymorphisms of COX-2 and susceptibility to GC were selected from the MEDLINE, EMBASE and CBMdisc databases. Data was extracted from the studies by 2 independent reviewers. The meta-analyses were performed by RevMan 5.0.23. From these data, odds ratio (OR) with 95% confidence interval (CI) was calculated., Results: Ten studies were retrieved reporting a total of 11 COX-2 polymorphisms. Carriers of -765C, -1195A, -1290G, *2430T alleles and *429TT genotype revealed increased risk for GC (OR = 1.71, 95% CI: 1.01-2.90, P = 0.05; OR = 1.58, 95% CI: 1.05-2.38, P = 0.03; OR = 1.55, 95% CI: 1.01-2.39, P = 0.05; OR = 2.62, 95% CI: 1.20-5.73, P = 0.02 and OR = 0.74, 95% CI: 0.59-0.95, P = 0.02, respectively)., Conclusion: The -765C, -1195A, -1290G, *2430T alleles and *429TT genotype of COX-2 polymorphisms were determined a significant association with susceptibility to GC.

Abnormal gas sensing characteristics are observed at low temperature in uniformly loaded Pt@SnO(2) nanorod gas sensors. The sensors operated at 200 degrees C exhibit opposite variations of resistances, and the change of resistance decreases with increasing ethanol concentration. In contrast, the sensors operated at 300 degrees C show regular behavior and the sensitivity is extremely high. Such behaviors are ascribed to Pt-catalyzed morphological changes of ionsorbed oxygen at low temperature. The present results are the bases for further investigating the effect of ionsorbed oxygen morphologies on gas sensing.

A combined biological (augmented membrane bioreactor (MBR)) and photochemical (photocatalysis and ozonation) treatment has been proposed for bromoamine acid (BAA) removal in dyeing wastewater. It was demonstrated that the color and chemical oxygen demand removal in the sequential augmented MBR was about 90% and 50%, respectively. By ribosomal intergenic spacer analysis, it was found that the introduced strain QYY was maintained as the predominant species and the diversity of the system was relatively low throughout the operation. Photocatalysis and ozonation processes were efficient to treat the effluents from MBR with high color and total organic carbon removal more than 90% within 120 min. Therefore, the hybrid treatment system is a possible way to achieve the complete mineralization of BAA.

Objective: The aims of this study were to evaluate whether the presence of -2518A/G polymorphism in the distal regulatory region of the monocyte chemotactic protein-1 (MCP-1) was associated with tuberculosis (TB) in Chongqing Han population and to find whether it has a significant impact on the pediatric patient., Method: One hundred children [ < or = 15 years old, mean age (7.3+/-4.6) years, 53 male, 47 female] and one hundred adults [51 male, 49 female, age (44.6+/-13.5) years with TB] and 200 healthy controls of comparable age were screened for genotype by PCR-sequence-specific primer (SSP) method. MCP-1 levels in the sera were detected by ELISA., Result: (1) TB patients and controls showed different single nucleotide polymorphism (SNP) distribution patterns (58%, 36%). MCP-1 alleles -2518G was associated with increased TB susceptibility (P<0.01). (2) The -2518 GG genotypes was associated with increased TB susceptibility (32% in TB patients and 13% in non-TB controls respectively, P<0.01). (3) The odds of developing TB in genotypes GG were higher than those in homozygous AA, and the risk was higher in children than in adult (7.0-fold in children and 5.1-fold in adults, respectively). (4) Cases of homozygous GG had the highest plasma levels of MCP-1, which increased the likelihood of developing TB. Furthermore, higher levels were observed in children than in adults., Conclusion: These findings suggest that persons bearing the MCP-1 genotype GG produce high concentrations of MCP-1, which increases the risk of active TB infection in Chongqing Han people. These findings are more significant in child patients than in adult patients with TB.

The performances and microbial population changes in laboratory-scale membrane bioreactor (MBR) augmented with Sphingomonas xenophaga QYY were investigated in the present study. It was demonstrated that after 30 days acclimation, the non-augmented MBR system were able to degrade bromoamine acid (BAA) well. However, the efficiency of the system decreased with BAA concentration increasing. While the augmented MBR showed higher capability, in which the color and COD removal were more than 90% and 50%, respectively. By ribosomal intergenic spacer analysis (RISA), it was found that BAA-utilizing populations gradually increased to become the dominant species in the non-augmented MBR. However, the augmented MBR possessed relatively stable treatment abilities, in which the introduced strain QYY could be persistent and co-exist well with the indigenous populations.

Aim: To explore whether cyclooxygenase 2 (COX-2) -765G>C polymorphism is associated with susceptibility of colorectal cancer (CRC) and to evaluate the risk of colorectal cancer in relation to environmental exposures and polymorphism., Methods: We conducted a case-control study of 137 patients with colorectal cancer and 199 cancer-free controls in northeast China. Multivariate logistic regression analysis was performed to calculate the adjusted odds ratio (OR) and 95% confidence interval (95% CI)., Results: The -765G>C polymorphism was not independently associated with CRC risk. However, risk associated with the polymorphism differed by smoking and body mass index (BMI). Smoking and BMI associated risks were stronger among those with -765GG genotype, showing that smokers had a 2.682-fold greater risk of CRC than nonsmokers (51/43 vs 68/126, P = 0.006). Compared to those with a normal body mass index (BMI 18.5-22.9), those with overweight (BMI 23-24.9) had a 3.909-fold higher risk of CRC (OR = 3.909, 95% CI = 2.081-7.344; P < 0.001), while those with obesity (BMI > 25) had a 2.031- fold higher risk of CRC (OR = 1.107, 95% CI = 1.107-3.726; P = 0.022)., Conclusion: Although COX-2 -765G>C polymorphism is not associated with an increased risk of CRC, -765GG genotype appears to be related to an increased risk in the presence of smoking and higher BMI.

Aim: To explore the role of the matrix metalloproteinase-9 (MMP-9) polymorphism in colorectal cancer (CRC) in a northeast Chinese population., Methods: Genotyping of MMP-9-1562C>T and 279R>Q polymorphisms was carried out on blood samples from 137 colorectal cancer patients and 199 controls using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Multivariate logistic regression models were used to calculate adjusted odds ratios (OR) and 95% confidence intervals (95% CI)., Results: The distribution of MMP-9 -1562C>T and 279 R>Q genotype was not significantly associated with the risk of CRC. However, the risk of llymph node metastasis of CRC was increased in patients with the -1562T allele (OR = 2.601; 95% CI = 1.160-5.835; P = 0.022). The frequency of MMP-9 279RR + RQ genotype was higher than the QQ genotype among CRC patients younger than sixty years old (OR = 0.102; 95% CI = 0.013-0.812; P = 0.012)., Conclusion: Our results indicated that the MMP-9-1562C>T polymorphism affects lymph node metastasis of CRC. In addition, the MMP-9 279R allele may lead to a younger age of onset of colorectal cancer.

Objective: To investigate the inhibitory effect of siRNA on heparanase expression and invasion ability gastric cancer cells., Methods: A heparanase mRNA-targeting double-stranded siRNA was designed with the bioinformatics technology. Human gastric cancer cells of the line SGC7901 were cultured and transfected with the siRNA of the concentrations of 5, 10, 20, and 40 nmol/L respectively. Forty-eight hours later RT-PCR and Western blotting were applied to detect the mRNA and protein expression of heparanase. Millicell chamber assay was performed to detect the invasion ability of the SGC7901 cells. Blank control group and negative control group were set., Results: The mRNA expression level of the cells transfected with the siRNA of the concentrations 20 nmol/L and 40 nmol/L were 0.207 +/- 0.095 and 0.200 +/- 0.085 respectively, both significantly lower than that of the control group (0.60 +/- 0.09, both P < 0.05). Western blotting showed that the protein expression of heparanase of the different siRNA subgroups were all decreased dose-dependently; and no heparanase band was seen in the 40 nmol/L subgroup. The invasion rate of the siRNA group was significantly lower than that of the control group with a mean inhibition rate of (61 +/- 36)%., Conclusion: RNAi inhibits the expression of heparanase and the invasion ability of human gastric cancer cells. Heparanase may be a new target in treatment of gastric cancer's metastasis.

We show that the combination of X-ray reflectivity, tryptophan fluorescence spectrum, and fluorescence quenching by bromine provides a useful tool to probe the location of lysozyme in lipid bilayers. To this end, we prepare lamellar complexes composed of phospholipids and lysozyme on solid surfaces using a solution-casting method. The proteins lie spontaneously between adjacent bilayers in the complexes. The results indicate that lysozyme may penetrate into the lipid bilayers. But the penetration depth is very shallow, and the tryptophan residues do not penetrate beyond the interface between the hydrocardon core and the headgroup region of the lipid bilayer. The penetration becomes saturated when more proteins are incorporated into the lamellar complex. The excess proteins stay in the interlamellar aqueous layers.

Controllable depositing of relatively thick inorganic sublayers into organic templates to fabricate organic-inorganic superlattices is of great importance. We report a novel approach to fabricating phospholipid/Ni(OH)(2) superlattices by electrochemical deposition of the inorganic component into solid-supported multilamellar templates. The well-ordered and highly oriented multilamellar templates are produced by spreading small drops of lipid solution on silicon surfaces and letting the solvent evaporate slowly. The templates which are used as working electrodes preserve the lamellar structure in the electrolyte solution. The resulting superlattices are highly oriented. The thickness of the nickel hydroxide is controlled by the concentration of nickel ions in the electrolyte bath. The electron density profiles derived from the X-ray diffraction data reveal that the thickness of the nickel hydroxide sublayers increases from 15 to 27 A as the concentration of nickel nitrate increases from 0.005 mol/L to 0.08 mol/L. We expect that the new method can be extended to depositing a variety of inorganic components including metals, oxides, and semiconductors.

Objective: To investigate the incidence and the relevant high-risk factors of congenital hearing loss for infants from well-baby nursery and newborn intensive care unit. To explore the scientific and feasible mode of early diagnosis, following-up and early intervention., Methods: Two-stage universal newborn hearing screening ( UNHS ) were performed by using transient otoacoustic emission ( TEOAE ). Auditory brainstem responses ( ABRs) and 40 Hz auditory event related potentials (40 Hz-AERPs) was used as diagnostic tests for those failed re-screening at three-month old. Two kinds of infants who had abnormal ABRs or high-risk factors of hearing loss received following-up and routine audiological evaluation from 6 months to 3 years of age. Each infant with congenital hearing loss received personalized intervention., Results: The incidence of congenital hearing loss among infants who received UNHS was 5.73 per thousand. The incidence of congenital hearing loss was 20.02 per thousand in NICU and 3.67 in WBN., Conclusions: Whether passed the UNHS or not , infants from NICU should receive ABR and 40 HzAERP tests. The following-up population includes infants with abnormal ABRs or high-risk factors of hearing loss. Following-up and early intervention should be personalized.

Objective: To study the alveolar ultrastructural changes and the interaction between Pneumocystis carinii (PC) and alveoli in Pneumocystis carinii pneumonia (PCP) patients after renal transplantation., Methods: Twenty-seven patients with suspected PCP after renal transplantation were examinated by bronchoscopy with bronchoalveolar lavage (BAL) and transbronchial lung biopsy (TBLB). The BAL fluid was centrifuged and the sediments were stained for PC. Cases for which electron microscope showed alveolar tissue in TBLB specimen were included. Then the clinical features, PC, alveolar epithelial damage, exudate in alveolar space, inflammatory cell infiltration, and fibrous tissue in the interstitial space were analyzed and evaluated., Results: Twenty-three cases were studied. The mean time from renal transplantation to onset of illness was 5.6 months, and that from onset of illness to hospitalization was 5.5 days. Clinical features included fever, dyspnea, unproductive cough, and scanty chest signs, to hypoxemic respiratory failure. Chest CT showed diffuse lung interstitial changes in 22 of the 23 cases, 9 with consolidation. After treatment with SMZco, the fever resolved in 1 - 5 days, and the general state of the patients became better, and 19 patients were dischaged within 1 month. PC in BAL fluid was found by special staining in 18 patients, while PC was found by electron microscope in 14 patients. In most cases PC was few in the lung tissue, but in 3 cases abundant PC filled the alveolar space. PC was seen in two forms, the cyst and the trophozoite. Electron and light microscopes showed alveolar exudate, inflammation in interstitium and alveolar space, interstitial fibrosis, and alveolar epithelial damage in all patients., Conclusion: In PCP patients after renal transplantation there was marked alveolar damage, which was the major pathological change in the lung.

Objective: To explore the role of hypercoagulation in the metastasis of carcinoma., Methods: The effect of Tetramethylpyrazine (TTMP) on platelet functions among the 25 advanced cases of lung carcinoma, and 26 matched control subjects were investigated in the study. Their ages varied from 31-86 years (mean 58.2) in lung carcinoma group (13 male, 12 female) and 36 to 61 (mean 52.9) in the control group (16 male, 10 female). The pathologic types were as follows: 7 cases of squamous cell cancer, 12 adenocarcinoma, 2 small cell carcinoma and 4 undistinguished type. The TNM stage revealed 14 cases in stage IIIa, 3 in stage IIIb and 8 in stage IV. The site of metastasis included mediastinal lymph node, pleura, supraclavicular lymph node, brain, spine, costa, skin and pericardium. The levels of plasma TXB2, 6-keto-PGF1 alpha, VIII:C, vWF, AT-III:a, AT-III:Ag, Fg and blood PAdT, PAgT were measured before and after the intravenous infusion of 80 mg TTMP in patients with lung carcinoma., Results: The levels of TXB2, 6-keto-PGF1 alpha, VIII:C, vWF and Fg in lung carcinoma group were significantly elevated, while the levels of PAdT was greatly decreased, compared with the control group, no significant differences in levels of PAgT, AT-III:a and AT-III:Ag were found between the two groups. After the infusion of TTMP the levels of PAdT, PagT, VIII:C, dWF and Fg were decreased significantly, while TXB2, 6-keto-PGF1 alpha, AT-III:a and AT-III:Ag remained unchanged., Conclusions: TTMP inhibits the adhesion and aggregatory functions of blood platelet and the activity of coagulation factors. It might be one of the mechanisms of TTMP's antimetastasis of lung carcinoma.

During 5 consecutive years of 100 mm photofluorography, 21 cases of primary lung cancer were discovered among 211,881 checked subjects in 54 factories in the suburbs of Beijing. Detection rate was 9.9/100,000. In the male smokers 40 years old or over, who comprised 13.7% of all examined with a detection rate of 55.0/100,000. Among the 21 cases, peripheral cancer was 61.9%, in which 38.4% were stage I 69.2% were treated by surgical resection, much higher than that of the central type. It may be concluded that photofluorography in mass screening has a certain value in early detection of the peripheral lung cancer and the male smokers 40 years or over should be examined as a priority group.
