Background/Aims: To investigate the differential expression of RING finger (RNF) proteins in Barrett esophagus (BE) and esophageal adenocarcinoma (EAC). Methods: The differential expression of RNFs in normal esophagus (NE), BE, and EAC was screened using microarray assay. Real-time quantitative polymerase chain reaction (PCR), tissue microarray assay, and Western blot analysis were independently performed to detect the mRNA and protein expression of screened RNFs. Results: The expression of nine RNFs in the BE or EAC was 2-fold higher than those in NE. Among these proteins, the RNF32 and RNF121 expression in BE was 20.3-fold and 16.4-fold higher, respectively, than that in NE, and the expression of RNF24, RNF130, RNF141, RNF139, RNF11, RNF14, and RNF159 was upregulated more than 2-fold compared with NE. The expression of nine RNFs was not only upregulated in the EAC but was also positively related to the RNF expression in BE. The PCR results also indicated increased expression of these RNFs in BE and EAC compared to NE. Furthermore, the mRNA expression of all RNFs, except for RNF141 in EAC, was dramatically higher than those in the BE. Similar results were also obtained from the Western blot analysis. Introduction: The aim of this study was to investigate the role of Mad1/c-Myc in telomerase regulation in gastric cancer cells in order to gain insight into telomerase activity and to evaluate PinX1 as a putative inhibitor of gastric cancer.Methods: PinX1 and PinX1siRNA eukaryotic expression vectors were constructed by recombinant technology and transfected into gastric carcinoma cells using Lipofectamine 2000. Telomerase activity was measured by the telomeric repeat amplification protocol. Apoptosis of gastric cancer cells was analyzed by flow cytometry and transmission electron microscopy. Reverse transcription-polymerase chain reaction and Western blotting were used to assess the expression levels of PinX1 and Mad1/c-Myc.Results: We found that PinX1-negative gastric cancer cells showed significantly higher telomerase activity than did the PinX1-postive cells. PinX1-transfection reduced telomerase activity in PinX1-negative gastric cancer cells and exhibited an upregulation of Mad1 and downregulation of c-Myc expression. Pinx1 RNAi treatment led to downregulation of Mad1 and upregulation of c-Myc.Conclusion: Suppression of telomerase activity mediated by PinX1 is involved in the Mad1/c-Myc pathway. Background: Barrett's esophagus (BE), a squamous-to-columnar metaplasia, may originate from growth-promoting mutations in metaplastic stem cells. Nucleostemin is a protein highly expressed in undifferentiated embryonic stem cells. The objectives of this study were to explore the potential role of nucleostemin in the pathogenesis of BE METHODS: The expression profiles of 30,968 genes were compared between BE and normal esophageal tissues (n = 6 in each group) by using oligo microarray. Three siRNA plasmid expression vectors against nucleostemin, pRNAi-1, pRNAi-2, and pRNAi-3, were constructed and transfected into HT29 cells. In addition, HT29 cells were exposed to 100-1,000 microM chenodeoxycholic acid (CDC), a bile acid, for 2, 12, and 24 h, and then messenger RNA and protein expressions of nucleostemin and CDX2 were determined by reverse-transcriptase polymerase chain reaction and Western blotting.Results: Four hundred and twenty-six differentially expressed genes were detected in BE; 142 were upregulated and 284 downregulated. Nucleostemin was downregulated while CDX2 was upregulated. In vitro, all the recombinant plasmids inhibited the nucleostemin expression in transfected HT29 cells, with pRNAi-1 being the most effective. CDX2 expression was significantly increased in pRNAi-1-transfected HT29 cells, compared with that in the empty plasmid (pRNAT-U6.1/Neo) transfected or untransfected HT29 cells. In addition, CDX2 expression was increased whereas nucleostemin expression was decreased in a dose- and time-dependent manner in HT29 cells treated with CDC.Conclusion: These findings suggest that the inhibition of nucleostemin expression in "esophageal stem cells" in response to bile acid exposure may be involved in the pathogenesis of BE through upregulating CDX2 expression. Gene chip methods are applied to the study of gene expression. The differentially expressed genes in different specimens may be detected with parallel analysis by gene chip, which has greatly improved the traditional experiments in that only a single or several gene expressions need to be observed for each test, thereby speeding up the identification of differentially expressed genes and the construction of differential expression profiles. Many studies have applied this technology for Barrett's metaplasia and adenocarcinoma, and identified a number of candidate genes useful as biomarkers in cancer staging, prediction of recurrence, prognosis and treatment selection. This review described the gene expression profile and molecular changes related to Barrett's metaplasia and adenocarcinoma of the esophagus, with emphasis on its prognostic value and possibilities for targeted therapy in a clinical setting. The article presents a case study of a 59-year-old male patient who has a medical history of melena and dizziness without prior history of dysphagia. The patient has underwent an endoscopy which shows ulcers on polyps which are potentially bleeding and could result to pharyngeal anesthesia. The article discusses that most patients with fibrovascular polyps of the esophagus and hypopharynx underwent surgical treatment through an endoscopic polypectomy.

Phloem-feeding insects feed on plant phloem using their stylets. While ingesting phloem sap, these insects secrete saliva to circumvent plant defenses. Previous studies have shown that, to facilitate their feeding, many phloem-feeding insects can elicit the salicylic acid- (SA-) signaling pathway and thus suppress effective jasmonic acid defenses. However, the molecular basis for the regulation of the plant's defense by phloem-feeding insects remains largely unknown. Here, we show that Bt56, a whitefly-secreted low molecular weight salivary protein, is highly expressed in the whitefly primary salivary gland and is delivered into host plants during feeding. Overexpression of the Bt56 gene in planta promotes susceptibility of tobacco to the whitefly and elicits the SA-signaling pathway. In contrast, silencing the whitefly Bt56 gene significantly decreases whitefly performance on host plants and interrupts whitefly phloem feeding with whiteflies losing the ability to activate the SA pathway. Proteinprotein interaction assays show that the Bt56 protein directly interacts with a tobacco KNOTTED 1-like homeobox transcription factor that decreases whitefly performance and suppresses whiteflyinduced SA accumulation. The Bt56 orthologous genes are highly conserved but differentially expressed in different species of whiteflies. In conclusion, Bt56 is a key salivary effector that promotes whitefly performance by eliciting salicylic acid-signaling pathway. [ABSTRACT FROM AUTHOR]

As Artificial Intelligence (AI) is rapidly integrated into existing technologies which has brought forth the fourth industrial and learning revolution, designing curriculum for AI education has become an important strategic development for education authorities throughout the world. Framed broadly from the Theory of Planned Behavior, this study examined a structural equational model to establish the interrelationships of students’ perceived usefulness, attitude towards using AI, subjective norms to learn about AI, basic literacy about AI and their behavioral intention to learn about AI. In addition, it examines the moderation effects of readiness, social good, and optimism on the research model. The findings confirm the hypothesized model. In addition, various moderation effects were found among students’ perception of readiness, social good, and optimism for AI. The implications of the study point towards the need to consider these factors in designing AI curriculum to foster students’ behavioral intention to learn AI. [ABSTRACT FROM AUTHOR]
