Abstract The primary cause of viral encephalitis (VE) is invasion of the central nervous system (CNS) by the virus, which leads to neuroinflammation and poses a significant threat to global public health. Microglia, as CNS-resident macrophages, play a crucial role in neuroinflammation and are often identified as the preferred target for the prevention or treatment of VE. In this study, we used pseudorabies virus (PRV)-induced VE in mice and pigs as a model to investigate the regulation of microglial responses during viral encephalitis and explored the mechanism of microglial activation. Cellular experiments revealed that microglial activation was accompanied by cell migration, characteristic morphological changes, phagocytosis, inflammatory cytokine production, and antigen presentation. Transcriptome analysis revealed that genes related to inflammation in PRV-infected BV2 cells were significantly enriched. The expression of the NOD1 gene in BV2 cells was significantly increased during PRV infection, after which NOD1 in BV2 cells was silenced by siRNA and overexpressed via a plasmid. NOD1 was found to be involved in the secretion of cytokines in BV2 cells by regulating the MAPK/NF-κB signalling pathway. Mouse and pig experiments have shown that NOD1 is involved in the secretion of cytokines by microglia by regulating the MAPK/NF-κB signalling pathway during PRV infection.

Avian leukosis virus (ALV) is an avian oncogenic retrovirus that can impair immunological function, stunt growth and decrease egg production in avian flocks. The capsid protein (P27) is an attractive candidate for ALV diagnostics. In the present study, a new hybridoma cell (1F8) stably secreting an anti-P27 monoclonal antibody (mAb) was developed. The mAb exhibited a high affinity constant (Ka) of 8.65 × 106.0 L/mol, and it could be used for the detection of ALV-A/B/J/K strains. Moreover, a total of eight truncated recombinant proteins and five synthetic polypeptides were utilized for the identification of the B-cell epitopes present on P27. The results revealed that 218IIKYVLDRQK227 was the minimal epitope recognized by 1F8, which had never been reported before. Additionally, the epitopes could strongly react with different ALV subgroup’s specific positive serum and had a complete homology among all the ALV subgroups strains. Finally, a new sandwich ELISA method was created for the detection of ALV antigens, demonstrating increased sensitivity compared to a commercially available ELISA kit. These results offer essential knowledge for further characterizing the antigenic composition of ALV P27 and will facilitate the development of diagnostic reagents for ALV.

We report the experimental observation of soliton molecules in a dispersion-managed thulium-doped (Tm-doped) fiber laser utilizing a black phosphorus saturable absorber (BP-SA). A variety of soliton molecules can be detected with the increase of pump power and the adjustment of cavity polarization state. Additionally, we use numerical simulation to investigate the soliton dynamics and further explore soliton molecules with variable and controllable properties. Stable separation-changeable soliton molecule has been numerically observed through changing small signal gain G in the cavity. These results enhance research into multi-soliton complexes as well as its practical application.

The wettability and high-temperature mechanical properties of porous BN/Si3N4 ceramics brazed with SiTi22 (wt. %) filler were studied. It is manifested that SiTi22 filler presents remarkable wetting and spreading capabilities on the porous BN/Si3N4 ceramic surface. An interfacial reaction layer is generated at the interface, and the thickness of the reaction layer initially grows and subsequently remains constant with the escalation of temperature. Carbon coating modification is beneficial to the wettability and high-temperature mechanical properties of porous BN/Si3N4 ceramics. The wetting driving force is mainly controlled by the interfacial reaction at the three-phase line of the wetting front. The mechanical properties of the carbon-coated joints are significantly enhanced in comparison with uncoated joints. The joint strength attains a maximum value of roughly 73 MPa in the shear test implemented at 800 °C. The strength of the joint is significantly enhanced mainly due to the TiN0.7C0.3 particles that consume energy by changing the crack propagation direction, and the SiC nanowires strengthen the connection by bridging.

Abstract It is not clear as to whether weight bearing and ambulation may affect bone growth. Our goal was to study the role of mechanical loading (one of the components of ambulation) on endochondral ossification and longitudinal bone growth. Thus, we applied cyclical, biologically relevant strains for a prolonged time period (4 weeks) to one tibia of juvenile mice, while using the contralateral one as an internal control. By the end of the 4‐week loading period, the mean tibial growth of the loaded tibiae was significantly greater than that of the unloaded tibiae. The mean height and the mean area of the loaded tibial growth plates were greater than those of the unloaded tibiae. In addition, in female mice we found a greater expression of PTHrP in the loaded tibial growth plates than in the unloaded ones. Lastly, microCT analysis revealed no difference between loaded and unloaded tibiae with respect to the fraction of bone volume relative to the total volume of the region of interest or the tibial trabecular bone volume. Thus, our findings suggest that intermittent compressive forces applied on tibiae at mild–moderate strain magnitude induce a significant and persistent longitudinal bone growth. PTHrP expressed in the growth plate appears to be one growth factor responsible for stimulating endochondral ossification and bone growth in female mice.

Salmonella, a prevalent foodborne pathogen, poses a significant social and economic strain on both food safety and public health. The application of phages in the control of foodborne pathogens represents an emerging research area. In this study, Salmonella pullorum phage vB_SpuM_X5 (phage X5) was isolated from chicken farm sewage samples. The results revealed that phage X5 is a novel Myoviridae phage. Phage X5 has adequate temperature tolerance (28 °C–60 °C), pH stability (4–12), and a broad host range of Salmonella bacteria (87.50% of tested strains). The addition of phage X5 (MOI of 100 and 1000) to milk inoculated with Salmonella reduced the number of Salmonella by 0.72 to 0.93 log10 CFU/mL and 0.66 to 1.06 log10 CFU/mL at 4 °C and 25 °C, respectively. The addition of phage X5 (MOI of 100 and 1000) to chicken breast inoculated with Salmonella reduced bacterial numbers by 1.13 to 2.42 log10 CFU/mL and 0.81 to 1.25 log10 CFU/mL at 4 °C and 25 °C, respectively. Phage X5 has bactericidal activity against Salmonella and can be used as a potential biological bacteriostatic agent to remove mature biofilms of Salmonella or for the prevention and control of Salmonella.

Safety monitoring of hydraulic structures is a critical task in the field of hydraulic engineering construction. This study developed a method for preprocessing and classifying monitoring data for the identification of gross errors in hydraulic structures. By utilizing linear regression and wavelet analysis techniques, it effectively differentiated various waveform characteristics in data sets, such as Sinusoidal Wave Cyclical, Triangular Wave Cyclical, Seasonal Cyclical, and Weakly Cyclical growth types. In the experiments for gross error identification, the 3σ algorithm, K-medoids algorithm, and Isolation Forest algorithm were applied to test the data. The results showed that the K-medoids algorithm excelled in processing Sinusoidal Wave Cyclical Data Sets; the 3σ algorithm adapted better to Triangular Wave Cyclical Data Sets; the Isolation Forest algorithm performed well in handling data sets with significant anomalies or atypical fluctuations and excelled in scenarios with strong seasonality and large data fluctuations; and for complex Weakly Cyclical Growth Data Sets, all three algorithms were less effective, indicating the potential need for more advanced analysis methods or a combination of multiple techniques. Testing on actual engineering data further confirmed the importance of using specific gross error identification techniques for special data types after data set pre-classification, providing a more effective technical solution for the safety monitoring of hydraulic structures.

High yield is a major objective for peanut (Arachis hypogaea L.) breeding worldwide. However, fewer yield-related quantitative trait loci (QTL) have been reported in peanut than in other staple food crops such as rice (Oryza sativa), wheat (Triticum aestivum), and maize (Zea mays). This study aimed to identify stable major-effect QTL associated with pod yield per plant, hundred-pod weight for double-seeded pods, hundred-seed weight, shelling percentage, and pod number per plant, allowing us to predict candidate genes by means of transcriptome and genome sequencing. To this end, we used a population of recombinant inbred lines comprising 192 F9:11 families derived from a JH6 × KX01-6 cross to construct a high-resolution genetic map (1705.7 cM) consisting of 2273 polymorphic SNPs, with 0.75 cM (on average) between adjacent SNPs. We identified two high-confidence, yield-related QTL, qHYF_A08 and qHYF_B06, explaining 5.78%–31.40% of phenotypic variation and with LOD values of 5.10–24.48, in six environments. qHYF_A08 mainly explained the variation in shelling percentage, whereas qHYF_B06 explained variation in hundred-pod weight and hundred-seed weight and accounted for 8.77%–31.40% of the variation in effective pod number per plant, pod number per plant, and shelling percentage. We narrowed down qHYF_B06 to an 890-kb interval using an advanced mapping population. Transcriptome and genome analyses revealed that only Arahy.129FS0 and Arahy.3R9A5K in the candidate mapping interval were differentially expressed between JH6 and KX01-6, with substantial structural variations in their promoter and coding regions. Genotypes of 208 peanut accessions determined using a diagnostic CAPS marker suggested that the two haplotypes of Arahy.3R9A5K were highly associated with hundred-seed weight and hundred-pod weight; this diagnostic CAPs marker could therefore be useful for selecting high-yielding lines during peanut breeding. Overall, our results provide valuable information for cloning alleles with favorable effects on peanut yield.

Abstract. Cellular immune responses as well as generalized and periarticular bone loss are the key pathogenic features of rheumatoid arthritis (RA). Under the pathological conditions of RA, dysregulated inflammation and immune processes tightly interact with skeletal system, resulting in pathological bone damage via inhibition of bone formation or induction of bone resorption. Single-cell omics technologies are revolutionary tools in the field of modern biological research.They enable the display of the state and function of cells in various environments from a single-cell resolution, thus making it conducive to identify the dysregulated molecular mechanisms of bone destruction in RA as well as the discovery of potential therapeutic targets and biomarkers. Here, we summarize the latest findings of single-cell omics technologies in osteoimmunology research in RA. These results suggest that single-cell omics have made significant contributions to transcriptomics and dynamics of specific cells involved in bone remodeling, providing a new direction for our understanding of cellular heterogeneity in the study of osteoimmunology in RA.

Abstract Pseudorabies virus (PRV) can infect multiple hosts and lead to fatal encephalitis. There is a significant increase in the number of microglia in the brain of animals infected with PRV. However, whether and how microglia contribute to central nervous system damage in PRV infection remain unknown. In the present study, we elucidated that PRV infection can cause more severe inflammatory cell infiltration, thicker and more numerous vessel sleeve walls, and more severe inflammatory responses in the brains of natural hosts (pigs) than in those of nonnatural hosts (mice). In a mice infection model, activated microglia restricted viral replication in the early stage of infection. Acute neuroinflammation caused by microglia hyperactivation at late-stage of infection. Furthermore, in vitro experiments revealed that microglia restricted viral replication and decreased viral infectivity. This may be associated with the phagocytic ability of microglia because we observed a significant increase in the expression of the membrane receptor TREM2 in microglia, which is closely related to phagocytosis, we observed that depletion of microglia exacerbated neurological symptoms, blood–brain barrier breakdown, and peripheral lymphocyte infiltration. Taken together, we revealed the dual role of microglia in protecting the host and neurons from PRV infection.

Ischemic stroke is a common cerebrovascular disease with high mortality, high morbidity, and high disability. Cerebral ischemia/reperfusion injury seriously affects the quality of life of patients. Luteolin-7-O-β-d-glucuronide (LGU) is a major active flavonoid compound extracted from Ixeris sonchifolia (Bge.) Hance, a Chinese medicinal herb mainly used for the treatment of coronary heart disease, angina pectoris, cerebral infarction, etc. In the present study, the protective effect of LGU on cerebral ischemia/reperfusion injury was investigated in an oxygen–glucose deprivation/reoxygenation (OGD/R) neuronal model and a transient middle cerebral artery occlusion (tMCAO) rat model. In in vitro experiments, LGU was found to improve the OGD/R-induced decrease in neuronal viability effectively by the MTT assay. In in vivo experiments, neurological deficit scores, infarction volume rates, and brain water content rates were improved after a single intravenous administration of LGU. These findings suggest that LGU has significant protective effects on cerebral ischemia/reperfusion injury in vitro and in vivo. To further explore the potential mechanism of LGU on cerebral ischemia/reperfusion injury, we performed a series of tests. The results showed that a single administration of LGU decreased the content of EB and S100B and ameliorated the abnormal expression of tight junction proteins ZO-1 and occludin and metalloproteinase MMP-9 in the ischemic cerebral cortex of the tMCAO 24-h injury model. In addition, LGU also improved the tight junction structure between endothelial cells and the degree of basement membrane degradation and reduced the content of TNF-α and IL-1β in the brain tissue. Thereby, LGU attenuated cerebral ischemia/reperfusion injury by improving the permeability of the blood–brain barrier. The present study provides new insights into the therapeutic potential of LGU in cerebral ischemia.

The Jing-Mi Diversion Canal is a large-scale water diversion project in Beijing. Routine monitoring is crucial for the reliability and stability of urban water supply. Compared with traditional monitoring methods, interferometric synthetic aperture radar (InSAR) has the advantages of large scale and high accuracy. Based on the small baseline subset InSAR, 187 ascending and 102 descending SAR images obtained from Sentinel-1 were used to detect the deformation along the diversion canal from 2017 to 2023. The results show that there was a sinking trend along the diversion canal. The subsidence was serious in the first half of the canal, and continued to sink from 2019 to 2020. The subsidence was alleviated in 2023. Combined with leveling measurements, the InSAR deformation monitoring results of important pumping station buildings were verified. The measurement accuracy of InSAR can reach the millimeter level. We extracted the groundwater level time series and subsidence for risky canal segments. Through pixel-by-pixel comparison, it was found that fluctuations in groundwater level would have some impact on surface deformation. Severe local subsidence or uplift deformation occasionally occurred. To ensure the safety of water diversion, the monitoring and maintenance of relevant pump station buildings in risky areas should be increased in the future.

Ulcerative colitis (UC) is one of the primary inflammatory bowel diseases (IBDs) and causes a serious threat to human public health around the world. Currently, there are no proven safe and effective treatment options to treat UC. Fraxetin (Fxt) is a widely recognized antioxidant and anti-inflammatory legume derived from ash bark. In the present study, we investigated the protective effect and mechanism of Fxt on UC. Our results showed that Fxt significantly attenuated the body weight, colon length reduction, tissue damage, and disease activity index induced by dextran sodium sulphate (DSS). Moreover, the DSS-induced activation of the NF-κB pathway and NLRP3 inflammasomes was inhibited, and the inflammatory response was reduced. Fxt restored gut barrier function by increasing the number of goblet cells and the levels of tight junction proteins (ZO-1 and occludin). In addition, Fxt can alter the intestinal microbiota by enhancing the diversity of the microbiota, increasing the relative abundance of beneficial bacteria and inhibiting the growth of harmful bacteria. These results revealed that Fxt alleviates DSS-induced colitis by modulating the inflammatory response, enhancing epithelial barrier integrity and regulating the gut microbiota. This study may provide a scientific basis for the potential therapeutic effect of Fxt in the prevention of colitis and other related diseases.

Luteolin-7-O-β-d-glucuronide (LGU) is a major active flavonoid glycoside compound that is extracted from Ixeris sonchifolia (Bge.) Hance, and it is a Chinese medicinal herb mainly used for the treatment of coronary heart disease, angina pectoris, cerebral infarction, etc. In the present study, the neuroprotective effect of LGU was investigated in an oxygen glucose deprivation (OGD) model and a middle cerebral artery occlusion (MCAO) rat model. In vitro, LGU was found to effectively improve the OGD-induced decrease in neuronal viability and increase in neuronal death by a 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay and a lactate dehydrogenase (LDH) leakage rate assay, respectively. LGU was also found to inhibit OGD-induced intracellular Ca2+ overload, adenosine triphosphate (ATP) depletion, and mitochondrial membrane potential (MMP) decrease. By Western blotting analysis, LGU significantly inhibited the OGD-induced increase in expressions of receptor-interacting serine/threonine-protein kinase 3 (RIP3) and mixed lineage kinase domain-like protein (MLKL). Moreover, molecular docking analysis showed that LGU might bind to RIP3 more stably and firmly than the RIP3 inhibitor GSK872. Immunofluorescence combined with confocal laser analyses disclosed that LGU inhibited the aggregation of MLKL to the nucleus. Our results suggest that LGU ameliorates OGD-induced rat primary cortical neuronal injury via the regulation of the RIP3/MLKL signaling pathway in vitro. In vivo, LGU was proven, for the first time, to protect the cerebral ischemia in a rat middle cerebral artery occlusion (MCAO) model, as shown by improved neurological deficit scores, infarction volume rate, and brain water content rate. The present study provides new insights into the therapeutic potential of LGU in cerebral ischemia.

Introduction: This paper proposes a power system fault prediction method that utilizes a GA-CNN-BiGRU model. The model combines a genetic algorithm (GA), a convolutional neural network (CNN), and a bi-directional gated recurrent unit network Bidirectional Gated Recurrent Unit to accurately predict and analyze power system faults.Methods: The proposed model employs a genetic algorithm for structural search and parameter tuning, optimizing the model structure. The CNN is used for feature extraction, while the bi-directional gated recurrent unit network is used for sequence modeling. This approach captures the correlations and dependencies in time series data and effectively improves the prediction accuracy and generalization ability of the model.Results and Discussion: Experimental validation shows that the proposed method outperforms traditional and other deep learning-based methods on multiple data sets in terms of prediction accuracy and generalization ability. The method can effectively predict and analyze power system faults, providing crucial support and aid for the operation and management of power systems.

Objective: PD-L1 and PD-L2 expression levels determine immune evasion and the therapeutic efficacy of immune checkpoint blockade. The factors that drive inducible PD-L1 expression have been extensively studied, but mechanisms that result in constitutive PD-L1 expression in cancer cells are largely unknown. Methods: DNA elements were deleted in cells by CRISPR/Cas9-mediated knockout. Protein function was inhibited by chemical inhibitors. Protein levels were examined by Western blot, mRNA levels were examined by real-time RT-PCR, and surface protein expression was determined by cellular immunofluorescence and flow cytometry. Immune evasion was examined by in vitro T cell-mediated killing. Results: We determined the core regions (chr9: 5, 496, 378–5, 499, 663) of a previously identified PD-L1L2-super-enhancer (SE). Through systematic analysis, we found that the E26 transformation-specific (ETS) variant transcription factor (ETV4) bound to this core DNA region but not to DNA surrounding PD-L1L2SE. Genetic knockout of ETV4 dramatically reduced the expressions of both PD-L1 and PD-L2. ETV4 transcription was dependent on ERK activation, and BRAF/TAK1-induced ERK activation was dependent on extracellular signaling from αvβ3 integrin, which profoundly affected ETV4 transcription and PD-L1/L2 expression. Genetic silencing or pharmacological inhibition of components of the PD-L1L2-SE-associated pathway rendered cancer cells susceptible to T cell-mediated killing. Conclusions: We identified a pathway originating from the extracellular matrix that signaled via integrin/BRAF/TAK1/ERK/ETV4 to PD-L1L2-SE to induce PD-L1-mediated immune evasion. These results provided new insights into PD-L1L2-SE activation and pathways associated with immune checkpoint regulation in cancer.

A smart grid is a new type of power system based on modern information technology, which utilises advanced communication, computing and control technologies and employs advanced sensors, measurement, communication and control devices that can monitor the status and operation of various devices in the power system in real-time and optimise the dispatch of the power system through intelligent algorithms to achieve efficient operation of the power system. However, due to its complexity and uncertainty, how to effectively perform real-time prediction is an important challenge. This paper proposes a smart grid real-time prediction model based on the attention mechanism of convolutional neural network (CNN) combined with bi-directional long and short-term memory BiLSTM.The model has stronger spatiotemporal feature extraction capability, more accurate prediction capability and better adaptability than ARMA and decision trees. The traditional prediction models ARMA and decision tree can often only use simple statistical methods for prediction, which cannot meet the requirements of high accuracy and efficiency of real-time load prediction, so the CNN-BiLSTM model based on Bayesian optimisation has the following advantages and is more suitable for smart grid real-time load prediction compared with ARMA and decision tree. CNN is a hierarchical neural network structure containing several layers such as a convolutional layer, pooling layer and fully connected layer. The convolutional layer is mainly used for extracting features from data such as images, the pooling layer is used for the dimensionality reduction of features, and the fully connected layer is used for classification and recognition. The core of CNN is the convolutional operation, a locally weighted summation operation on the input data that can effectively extract features from the data. In the convolution operation, different features can be extracted by setting different convolution kernels to achieve feature extraction and classification of data. BiLSTM can capture semantic dependencies in both directions. The BiLSTM structure consists of two LSTM layers that process the input sequence in the forward and backward directions to combine the information in both directions to obtain more comprehensive contextual information. BiLSTM can access both the front and back inputs at each time step to obtain more accurate prediction results. It effectively prevents gradient explosion and gradient disappearance while better capturing longer-distance dependencies. The CNN-BiLSTM extracts features of the data and then optimises them by Bayes. By collecting real-time data from the power system, including power, load, weather and other factors, our model uses the features of CNN-BiLSTM to deeply learn real-time load data from smart grids and extract key features to achieve future load prediction. Meanwhile, the Bayesian optimisation algorithm based on the model can optimise the model’s hyperparameters, thus improving the model’s prediction performance. The model can achieve accurate prediction of a real-time power system load, provide an important reference for the dispatch and operation of the power system, and help optimise the operation efficiency and energy utilisation efficiency of the power system.

Soil organic carbon (SOC) and soil inorganic carbon (SIC) has important effects on soil physical, chemical and biological properties. They play an important role in coordinating the relationship between soil water and air, increasing soil water holding capacity and improving plant productivity. In this study, a boosted regression trees (BRT) model was developed to map the spatial distribution carbon stocks in the semi-arid region of Northeast China in 1990 and 2015. During the two periods, 10-fold cross-validation technology was used to test the performance and uncertainty of BRT model. In order to construct the model, 9 environmental variables (derived from climate, topography and biology) and 173 (1990) and 223 (2015) topsoil (0–30 cm) samples were used. The comparison between estimated and observed data shows that the RMSE of SOC and SIC stocks were 0.53 kgm−2 and 0.19 kgm−2 in 1990, and 0.65 kgm−2 and 0.20 kgm−2 in 2015, respectively. Elevation, normalized difference vegetation index, mean annual precipitation and Landsat band 3 were identifies as critical environmental factors for simulating the spatial distribution of SOC, accounting for 76.6 % and 70.3 % of the total relative importance in 1990 and 2015, respectively. Mean annual precipitation, mean annual temperature and topographic wetness index were the critical environmental factors for simulating the spatial variation of SIC during the two periods. Land use change also played an important role in the spatial variability of SOC and SIC stocks. In the past 25 years, soil carbon stocks decreased from 6.2 kg m−2 in 1990 to 5.9 kg m−2 in 2015. The spatial distribution pattern of SOC was high in northeastern area and low in southwestern area during the two periods, while the spatial distribution pattern of SIC was opposite to that of SOC stocks. The mapped soil carbon stock distribution is fundamental to future study of soil carbon cycle and regional carbon balance in semi-arid regions.

Abstract Silicon is expected to become the ideal anode material for the next generation of high energy density lithium battery because of its high theoretical capacity (4200 mAh g−1). However, for silicon electrodes, the initial coulombic efficiency (ICE) is low and the volume of the electrode changes by over 300% after lithiation. The capacity of the silicon electrode decreases rapidly during cycling, hindering the practical application. In this work, a slidable and highly ionic conductive flexible polymer binder with a specific single‐ion structure (abbreviated as SSIP) is presented in which polyrotaxane acts as a dynamic crosslinker. The ionic conducting network is expected to reduce the overall resistance, improve ICE and stabilize the electrode interface. Furthermore, the introduction of slidable polyrotaxane increases the reversible dynamics of the binder and improves the long‐term cycling stability and rate performance. The silicon anode based on SSIP provides a discharge capacity of ≈1650 mAh g−1 after 400 cycles at 0.5C with a high ICE of upto 92.0%. Additionally, the electrode still exhibits a high ICE of 87.5% with an ultra‐high Si loading of 3.84 mg cm−2 and maintains a satisfying areal capacity of 5.9 mAh cm−2 after 50 cycles, exhibiting the potential application of SSIP in silicon‐based anodes.

Forest soil carbon is a major carbon pool of terrestrial ecosystems, and accurate estimation of soil organic carbon (SOC) stocks in forest ecosystems is rather challenging. This study compared the prediction performance of three empirical model approaches namely, regression kriging (RK), multiple stepwise regression (MSR), random forest (RF), and boosted regression trees (BRT) to predict SOC stocks in Northeast China for 1990 and 2015. Furthermore, the spatial variation of SOC stocks and the main controlling environmental factors during the past 25 years were identified. A total of 82 (in 1990) and 157 (in 2015) topsoil (0–20 ​cm) samples with 12 environmental factors (soil property, climate, topography and biology) were selected for model construction. Randomly selected 80% of the soil sample data were used to train the models and the other 20% data for model verification using mean absolute error, root mean square error, coefficient of determination and Lin's consistency correlation coefficient indices. We found BRT model as the best prediction model and it could explain 67% and 60% spatial variation of SOC stocks, in 1990, and 2015, respectively. Predicted maps of all models in both periods showed similar spatial distribution characteristics, with the lower SOC in northeast and higher SOC in southwest. Mean annual temperature and elevation were the key environmental factors influencing the spatial variation of SOC stock in both periods. SOC stocks were mainly stored under Cambosols, Gleyosols and Isohumosols, accounting for 95.6% (1990) and 95.9% (2015). Overall, SOC stocks increased by 471 ​Tg ​C during the past 25 years. Our study found that the BRT model employing common environmental factors was the most robust method for forest topsoil SOC stocks inventories. The spatial resolution of BRT model enabled us to pinpoint in which areas of Northeast China that new forest tree planting would be most effective for enhancing forest C stocks. Overall, our approach is likely to be useful in forestry management and ecological restoration at and beyond the regional scale.

We report, for the first time, the generation of a 1173 nm acousto-optic Q-switched self-Raman vortex laser with an axicon-based annular pump system. A 20 mm long Nd:GdVO4 crystal was used as the self-Raman crystal. Both the fundamental field and the first-Stokes field were investigated using the respective output couplers. In comparison with both vortex fields, a noticeable beam cleaning-up effect and pulse compression were observed from the 1063 nm fundamental field to the 1173 nm first-Stokes field. A Stokes field carrying a unitary topological charge was achieved. Finally, the average output power of the first-Stokes vortex emission reached 454 mW under an incident pump power of 19.5 W, corresponding to a pulse width of 45.7 ns. It was beneficial to apply a high peak power from the Q-switched laser and self-Raman conversion to expand the applications of the vortex laser beam.

Diode end-pumped Nd,La:CaNb2O6 self-Raman laser with acousto-optic Q-switching was successfully demonstrated for the first Stokes wave generation at 1,174 nm. A 1.0 at.% Nd3+ and 1.0 at.% La3+-doped CaNb2O6 crystal in dimensions 3 × 3 × 14.3 mm3 was used as the self-Raman laser crystal. Doping 1 at.% La3+ ions into this crystal could subdue the fluorescence quenching caused by cross-relaxation between Nd3+ ions and finally improve the laser output performance. Under the incident pump power of 9.9 W, the first Stokes wave at 1,174 nm with a maximum output power up to 928 mW was obtained, with the diode to Stokes conversion efficiency of about 9.4%. The results show that the Nd,La:CaNb2O6 is also a promising self-Raman crystal for efficient fundamental and Raman laser operation.

Avian leukosis (AL), caused by avian leukosis virus (ALV), is a contagious tumor disease that results in significant economic losses for the poultry industry. Currently, ALV-A, B, J, and K subgroups are the most common in commercial poultry and cause possible coinfections. Therefore, close monitoring is necessary to avoid greater economic losses. In this study, a novel multiplex quantitative polymerase chain reaction (qPCR) assay was developed to detect ALV-A, ALV-B, ALV-J, and ALV-K with limits of detection of 40, 11, 13.7, and 96 copies/µL, respectively, and no cross-reactivity with other ALV subtypes and avian pathogens. We detected 852 cell cultures inoculated with clinical samples using this method, showing good consistency with conventional PCR and ELISA. The most prevalent ALV strain in Hubei Province, China, was still ALV-J (11.74%). Although single infections with ALV-A, ALV-B, and ALV-K were not found, coinfections with different subgroup strains were identified: 0.7% for ALV-A/J, 0.35% for ALV-B/J, 0.25% for ALV-J/K, and 0.12% for ALV-A/B/K and ALV-A/B/J. Therefore, our novel multiplex qPCR may be a useful tool for molecular epidemiology, clinical detection of ALV, and ALV eradication programs.

Abstract Hydroxyapatite (HA) bioceramic scaffolds were fabricated by using digital light processing (DLP) based additive manufacturing. Key issues on the HA bioceramic scaffolds, including dispersion, DLP fabrication, sintering, mechanical properties, and biocompatibility were discussed in detail. Firstly, the effects of dispersant dosage, solid loading, and sintering temperature were studied. The optimal dispersant dosage, solid loading, and sintering temperature were 2 wt%, 50 vol%, and 1250 °C, respectively. Then, the mechanical properties and biocompatibility of the HA bioceramic scaffolds were investigated. The DLP-prepared porous HA bioceramic scaffold was found to exhibit excellent mechanical properties and degradation behavior. From this study, DLP technique shows good potential for manufacturing HA bioceramic scaffolds.

Geographical indications of agricultural products are characterized by high quality and regional attributes, while they are more likely to be counterfeited by similar products from nearby regions. Accurate discrimination of origin on small geographical scales is extremely important for geographical indications of agricultural products to avoid food fraud. In this study, a widely targeted metabolomics based on ultra-high-performance liquid chromatography–tandem mass spectrometry combined with multivariate statistical analysis was used to distinguish the geographical origin of Pinggu Peach of Beijing and its two surrounding areas in Heibei province (China). Orthogonal partial least squares-discriminant analysis (OPLS-DA) based on 159 identified metabolites showed significant separation from Pinggu and the other adjacent regions. The number of the most important discriminant variables (VIP value >1) was up to 62, which contributed to the differentiation model. The results demonstrated that the metabolic fingerprinting combined with OPLS-DA could be successfully implemented to differentiate the geographical origin of peach from small-scale origins, thus providing technical support to further ensure the authenticity of geographical indication products. The greenness of the developed method was assessed using the Analytical GREEnness Metric Approach and Software (ARGEE) tool. It was a relatively green analytical method with room for improvement.

Enterococcus faecalis is a potential animal and human pathogen. Improper use of antibiotics encourages resistance. Bacteriophages and their derivatives are promising for treating drug-resistant bacterial infections. In this study, phylogenetic and electron microscopy analyses of phage vB_EfaS_WH1 (WH1) isolated from chicken feces revealed it to be a novel phage in the family Siphoviridae. WH1 showed good pH stability (4–11), temperature tolerance (4–60 °C), and broad E. faecalis host range (60% of isolates). Genome sequencing revealed a 56,357 bp double-stranded DNA genome with a G+C content of 39.21%. WH1 effectively destroyed E. faecalis EF01 biofilms, even at low concentrations. When WH1 was applied at 1 × 105 to 1 × 109 PFU/g to chicken breast samples stored at 4 °C, surface growing E. faecalis were appreciably eradicated after 24 h. The phage WH1 showed good antibacterial activity, which could be used as a potential biocontrol agent to reduce the formation of E. faecalis biofilm, and could also be used as an alternative for the control of E. faecalis in chicken products.

Newborn piglets are prone to diarrhea after weaning as a result of changes in their environment and feed. Enterotoxigenic Escherichia coli (ETEC) K88 strain is a typical pathogen that causes diarrhea in such stage of piglets. Hermetia illucens larvae are widely used in livestock and poultry production because of their high nutritional value and immunoregulatory effects. This study aimed to evaluate the protective effects of H. illucens feed in protecting against ETEC induced diarrhea in piglets and to unravel the mechanisms of immune modulation and intestinal barrier maintenance. The results showed that after ETEC infection, citric acid in the serum of the groups fed on H. illucens larvae increased significantly, which stimulated macrophages to secrete cytokines that promote B lymphocyte differentiation, ultimately increasing the production of IgA and IgG in serum. Concomitantly, citric acid also had a positive effect on the intestinal barrier damaged due to ETEC infection by inhibiting the production of inflammatory cytokines, reducing the Bcl-2/Bax ratio, and promoting the expression of tight junction proteins. Correlation analysis showed that the increase of citric acid levels might be related to Massilia. Thus, citric acid derived from H. illucens larvae can improve the immune performance of weaned piglets and reduce ETEC-induced damage to the intestinal barrier in weaned piglets.

A high-quality protein substitute, Hermetia illucens (black soldier fly) larvae powder, is rich in protein and often used in animal feed. This study aimed to investigate the feasibility and optimal ratio of replacing fish meal with H. illucens larvae in weaned piglets and to demonstrate the effects on piglets' growth performance, intestinal microflora and immune performance. Forty-eight female weaned piglets were randomly classified into three groups. Each group consisted of eight pens (replicates), with two piglets per pen. Three groups containing different proportions of H. illucens larvae (0, 4, and 8%) were referred to as C, HI4, and HI8. We first designed a 28-day feeding experiment to detect growth performance; after that, the piglets were induced with oral gavage of enterotoxigenic Escherichia coli K88 (ETEC K88) and recording diarrhea on day 29 of the experiment. Samples were taken on the 32nd day to detect the effect of H. illucens larvae on the immune performance of the weaned piglets. H. illucens larvae replacement did not cause any obvious change in the growth performance nether in HI4 nor in HI8 of weaned piglets with 28 d feeding stage. H. illucens larvae could improve the intestinal health of weaned piglets by increasing the content of Lactobacillus and reducing the content of Streptococcus. Compared with C+K88 group, the diarrhea rate was attenuated for the H. illucens supplemented group. The integrity of ileum villi in HI4+K88 and HI8+K88 groups was better than that in C+K88 group, and the villi in C+K88 group were severely damaged. The expression of IL-10, Occludin and Claudin-3 in the intestinal mucosa of the HI4+K88 group and HI8+K88 group were significantly increased (P < 0.05), and the expression of TNF-α was significantly decreased (P < 0.05) compared with the C+K88 group. The results of immunoblotting also validated that the same ETEC K88 treatment of weaned piglets enhanced the expression of tight junction protein in the intestinal mucosa of the H. illucens addition group. ETEC-induced diarrhea will be reduced by the diet of weaned piglets containing H. illucens larvae, ameliorating the immune performance of piglets. Our results indicates that the optimal dosage of H. illucens replacement in weaned piglets is 4%.

The nondestructive assessment of nitrogen status in strawberry was performed to analyze its growth and provide guidance for precise management of N fertilizer using Fourier transform near-infrared reflectance (FT-NIR) spectroscopy with leaf spectral reflectance. The leaf soil plant analysis development (SPAD) value is considered as an indicator that indirectly reflects the nitrogen status in strawberry. However, the variation of cultivars leads to differences in the cell structure and light scattering and/or reflection effects of strawberry leaves during strawberry N status assessment, which means, leaf SPAD threshold value and the specific N demand will vary with different cultivars. As a result, accurate measurement of SPAD values and N status in strawberries with multiple cultivars is still challenging. In this study, the individual-cultivar, hybrid-cultivar, and multi-cultivar models were developed for the determination of SPAD values, and the performance of the models in lessening the impact of cultivar variation was studied and compared. The individual-cultivar model was constructed on the basis of a single cultivar of strawberry leaf. The hybrid-cultivar model was developed by merging the spectrum reflectance data and SPAD values of all studied leaf samples, and a multi-cultivar model was built in combination with cultivar identification, individual-cultivar models, and model search strategy. The results indicated that the multi-cultivar model was superior to the other two models for SPAD value estimation of strawberry leaves from different cultivars, with the overall Rp and RMSEP values of 0.966 and 0.468, respectively. We demonstrate that the leaf N content of strawberry is profoundly affected by cultivar variation, and establishing a multi-cultivar model is useful in monitoring the nitrogen status and guiding N fertilization recommendations for different strawberry cultivars.

An effective and trustworthy traceability system contributes to improving food quality and safety and responds to consumers’ demand for food provenance information. Safe meat and its products are crucial to consumers and society. Livestock feeding regime and geographical origin are closely related to the properties and the safety of animal origin food, but the information is often invisible to consumers, which makes is easier to use fraudulent practices throughout the whole supply chain. Technologies and their proper use in traceability systems are important for the safety of animal origin foods. An essential component in an integrated traceability chain includes individual animal identification and trace back of related meat products. In this review, we examine the technologies for individual animal identification, including the radio frequency identification system and DNA fingerprinting. For meat products, traceability technologies focus on the chemical components fingerprinting, including measurement of stable isotope ratios, mineral element tracing and organic component fingerprinting. Also, future trends in food traceability systems need to be improved to promote the establishment of more efficient and trustworthy traceability systems to ensure food safety and quality up to standard.

Several studies have reported an intricate link between the G protein-coupled receptor 109A (GPR109A) and intestinal health. Upon activation, induced by butyric acid and β-hydroxybutyric acid, GPR109A regulates the expression of tight junction proteins, exerts anti-inflammatory effects, and maintains the integrity of the intestinal barrier. However, its function and the mechanism of action in combating the infection caused by exogenous pathogenic microorganisms remain unclear. This study established an animal model of infection by oral enterotoxigenic Escherichia coli (ETEC) gavage to examine the underlying mechanism(s) and protective effects of GPR109A on the intestinal tract. Experimental GPR109A–/–and GPR109A+/+ mice were orally administered with 1 × 109 colony-forming units (CFUs) of ETEC, and changes in body weight were then observed. The colonization and translocation of ETEC in the intestine were detected by the plate counting method. The expression of tight junction proteins and the levels of inflammatory factors and secretory IgA (SIgA) in the intestine were detected by quantitative real-time polymerase chain reaction (q-PCR), western blotting, enzyme-linked immunosorbent assay (ELISA), and immunohistochemistry. The results demonstrated that GPR109A–/–mice were more susceptible to ETEC infection, showing more severe inflammatory reactions and intestinal damage. Moreover, the secretion of IgA in the intestinal tract of GPR109A+/+ mice was significantly increased after ETEC infection, whereas the IgA levels in GPR109A–/–mice did not change significantly. We added 5 g/L sodium butyrate to the drinking water of all mice. The GPR109A+/+ mice were protected against ETEC infection and no effect was observed in GPR109A–/–mice. Similarly, sodium butyrate increased the SIgA content in the gut of the GPR109A+/+ mice and no effect was observed in GPR109A–/–mice. In conclusion, activated GPR109A is effective against the colonization and translocation of ETEC in the gut and maintains the integrity of the intestinal barrier, possibly by promoting the secretion of intestinal IgA.

Soil organic carbon (SOC) and soil total nitrogen (STN) are major soil indicators for soil quality and fertility. Accurate mapping SOC and STN in soils would help both managed and natural soils and ecosystem management. This study developed an improved similarity-based approach (ISA) to predicting and mapping topsoil (0–20 cm soil depth) SOC and STN in a coastal region of northeastern China. Six environmental variables including elevation, slope gradient, topographic wetness index, the mean annual temperature, the mean annual temperature, and normalized difference vegetation index were used as predictors. Soil survey data in 2012 was designed based on the clustering of the study area into six climatic vegetation landscape units. In each landscape unit, 20–25 sampling points were determined at different landform positions considering local climate, soil type, elevation and other environmental factors, and finally 126 sampling points were obtained. Soil sampling from the depth of 0–20 cm were used for model prediction and validation. The ISA model performance was compared with the geographically weighted regression (GWR), regression kriging (RK), boosted regression trees (BRT) considering mean absolute prediction error (MAE), root mean square error (RMSE), coefficient of determination (R2), and maximum relative difference (RD) indices. We found that the ISA method performed best with the highest R2 and lowest MAE, RMSE compared to GWR, RK, and BRT methods. The ISA method could explain 76% and 83% of the total SOC and STN variability, respectively, 12–40% higher than other models in the study area. Elevation had the largest influence on SOC and STN distribution. We conclude that the developed ISA model is robust and effective in mapping SOC and STN, particularly in the areas with complex vegetation-landscape when limited samples are available. The method needs to be tested for other regions in our future research.

Arable land ecosystems are among the most important terrestrial systems. The issues of carbon sequestration and emission reductions in arable land ecosystems have received extensive attention. Countries around the world have actively issued policies to manage arable land ecosystems. At present, more than 100 countries have made carbon neutralization target commitments. Various arable land management measures and arable land planting strategies have important impacts on the carbon storage of arable land ecosystems. Research on arable land carbon is of great significance to global climate change. This study attempts to investigate the problems and deficiencies in the current research by summarizing a number of studies, including the main methods for the quantitative research of carbon sources and sinks as well as the influencing factors in these ecosystems. In this study, it is found that due to the differences of climate patterns, soil properties and management practices in arable land ecosystems, the factors affecting carbon sources and sinks are of great heterogeneity and complexity. Generally, variations in natural factors affect the carbon balance in different regions, while human management measures, such as irrigation, fertilization and the degree of agricultural mechanization, are the leading factors causing changes to carbon sources and sinks in these ecosystems. In addition, there are still great uncertainties in the evaluation of carbon sources and sinks in these ecosystems caused by different estimation models and methods. Therefore, emphasis should be placed on model parameter acquisition and method optimization in the future. This review provides a scientific basis for understanding carbon sources and sinks in arable land ecosystems, enhancing their carbon sink capacity and guiding low-carbon agriculture on arable land.

Summary: PD-L1 and PD-L2 are important targets for immune checkpoint blockade, but how tumor cells achieve their expression remains to be addressed. Here, we find that PD-L1 and PD-L2 are co-expressed in cancer cell lines and tissues across different cancer types. In breast cancer, MDA-MB-231 and SUM-159 cells show high expression of both PD-L1 and PD-L2. The expression of both PD-L1 and PD-L2 is greatly reduced upon treatment of inhibitors of super-enhancers. Bioinformatic analysis identifies a potential super-enhancer (PD-L1L2-SE) that is located between the CD274 and CD273 genes. Genetic deletion of PD-L1L2-SE profoundly reduces the expression of PD-L1 and PD-L2. PD-L1L2-SE-deficient cancer cells fail to generate immune evasion and are sensitive to T cell-mediated killing. Notably, epigenetic activation of such a region (PD-L1L2-SE) is correlated with PD-L1 and PD-L2. Taken together, we identify a super-enhancer (PD-L1L2-SE) that is responsible for the overexpression of PD-L1 and PD-L2 as well as immune evasion in cancer. : It is largely unknown how cancer cells achieve the expression of the twin co-inhibitory ligands, PD-L1 and PD-L2. Xu et al. report a super-enhancer called PD-L1L2-SE located between the genes encoding PD-L1 and PD-L2 that can induce immune evasion through synchronously initiating the expression of PD-L1 and PD-L2. Based on the high sensitivity and stable fluorescence of CdTe quantum dots (QDs) in conjunction with a specific DNA aptamer, the authors describe an aptamer-based fluorescence assay for the determination of Salmonella Typhimurium. The fluorescence detection and quantification of S. Typhimurium is based on a magnetic separation system, a combination of aptamer-coated Fe3O4 magnetic particles (Apt-MNPs) and QD-labeled ssDNA2 (complementary strand of the aptamer). Apt-MNPs are employed for the specific capture of S. Typhimurium. CdTe QD-labeled ssDNA2 was used as a signaling probe. Simply, the as-prepared CdTe QD-labeled ssDNA2 was first incubated with the Apt-MNPs to form the aptamer-ssDNA2 duplex. After the addition of S. Typhimurium, they could specifically bind the DNA aptamer, leading to cleavage of the aptamer-ssDNA2 duplex, accompanied by the release of CdTe QD-labeled DNA. Thus, an increased fluorescence signal can be achieved after magnetic removal of the Apt-MNPs. The fluorescence of CdTe QDs (λexc/em = 327/612 nm) increases linearly in the concentration range of 10 to 1010 cfu•mL-1, and the limit of detection is determined to be 1 cfu•mL-1. The detection process can be performed within 2 h and is successfully applied to the analysis of spiked food samples with good recoveries from 90% to 105%.

Forest ecosystems play an important role in regional carbon and nitrogen cycling. Accurate and effective monitoring of their soil organic carbon (SOC) and soil total nitrogen (STN) stocks provides important information for soil quality assessment, sustainable forestry management and climate change policy making. In this study, a geographical weighted regression (GWR) model, a multiple stepwise regression (MLSR) model, and a boosted regression trees (BRT) model were compared to obtain the best prediction of SOC and STN stocks of the forest ecosystems in northeastern China. Five-hundred and thirteen topsoil (0–30 cm) samples (10.32 kg m−2 (±0.53) for SOC, 1.21 kg m−2 (±0.32) for STN), and 9 remotely-sensed environmental variables were collected and used for the model development and verification. By comparing with independent verification data, the best model (BRT) achieved R2 = 0.56 and root mean square error (RMSE) = 00.85 kg m−2 for SOC stocks, R2 = 0.51 and RMSE = 0.22 kg m−2 for STN stocks. Of all the remotely-sensed environment variables, soil adjusted vegetation index (SAVI) and normalized difference vegetation index (NDVI) are of the highest relative importance in predicting SOC and STN stocks. The spatial distribution of the predicted SOC and STN stocks gradually decreased from northeast to southwest. This study provides an attempt to rapidly predict SOC and STN stocks in the dense vegetation covered area. The results can help evaluate soil quality and facilitate land policy and regulation making by the government in the region.

Accurately mapping the spatial distribution information of soil organic carbon (SOC) stocks is a key premise for soil resource management and environment protection. Rapid development of satellite remote sensing provides a great opportunity for monitoring SOC stocks at a large scale. In this study, based on 12 environmental variables of multispectral remote sensing, topography and climate and 236 soil sampling data, three different boosted regression tree (BRT) models were compared to obtain the most accurate map of SOC stocks covering the forest area of Lvshun District in the Northeast China. Four validation indexes, including mean absolute error (MAE), root mean square error (RMSE), coefficient of determination (R2), and Lin’s concordance correlation coefficient (LCCC) were calculated to evaluate the performance of the three models. The results showed that the full variable model performed the best, except the model using multispectral remote sensing variables. In the full variable model, the regional SOC stocks are primarily determined by multispectral remote sensing variables, followed by topographic and climatic variables, with the relative importance of variables in the model being 63%, 28%, and 9%, respectively. The average prediction results of full variables model and only multispectral remote sensing variables model were 8.99 and 9.32 kg m−2, respectively. Our results indicated that there is a strong dependence of SOC stocks on multispectral remote sensing data when forest ecosystems have dense natural vegetation. Our study suggests that the multispectral remote sensing variables should be used to map SOC stocks of forest ecosystems in our study region.

Water-saving ground cover rice production systems (GCRPS) are gaining popularity in many parts of the world. We aimed to describe the characteristics of root growth, morphology, distribution, and water uptake for a GCRPS.A traditional paddy rice production system (TPRPS) was compared with GCRPS in greenhouse and field experiments. In the greenhouse, GCRPS where root zone average soil water content was kept near saturation (GCRPSsat), field capacity (GCRPSfwc) and 80% field capacity (GCRPS80%), were evaluated. In a two-year field experiment, GCRPSsat and GCRPS80% were applied.Similar results were found in greenhouse and field experiments. Before mid-tillering the upper soil temperature was higher for GCRPS, leading to enhanced root dry weight, length, surface area, specific root length, and smaller diameter of roots but lower water uptake rate per root length compared to TPRPS. In subsequent growth stages, the reduced soil water content under GCRPS caused that the preponderance of root growth under GCRPSsat disappeared in comparison to TPRPS. Under other GCRPS treatments (GCRPSfwc and GCRPS80%), significant limitation on root growth, bigger root diameter and higher water uptake rate per root length were found.Discrepancies in soil water and temperature between TPRPS and GCRPS caused adjustments to root growth, morphology, distribution and function. Even though drought stress was inevitable after mid-tillering under GCRPS, especially GCRPS80%, similar or even enhanced root water uptake capacity in comparison to TPRPS might promote allocation of photosynthetic products to shoots and increase water productivity.

BACKGROUND: This study was designed to examine a novel role of COX-2/PGE2 signaling as a regulator of PTPRJ expression in endothelial cells. METHODS: A bioinformatics analysis of a whole genome array was carried out to search for regulators of PTPRJ expression in endothelial cells. PTPRJ expression was also measured in endothelial cells derived from a balloon injury-induced neointimal hyperplasia model in male New Zealand Rabbits. Changes in PTPRJ expression in HUVEC cells was examined by RT-PCR and western blotting after transfection of COX-2 plasmids or treatment with varying concentrations of a COX-2 inhibitor. RESULTS: A significant correlation was identified between COX-2 and PTPRJ in GSE39264 (Pearson correlation coefficient = -0.87; n = 22; P < 0.01, two-tailed). PTPRJ expression was reduced during the progression of neointimal hyperplasia after balloon injury, which correlated with an increase in COX-2 expression. In HUVECs, after transfection with 1 µg/ml, 0.5 µg/ml, or 0.25 µg/ml COX-2 plasmids, PTPRJ protein expression was reduced to 0.60- (± 0.08), 0.75- (± 0.09), and 0.88- (± 0.04) fold, respectively, while mRNA expression was reduced to 0.15- (± 0.03), 0.26- (± 0.05), and 0.47- (± 0.09) fold, respectively. After treatment of HUVECs with 10 µmol/L or 20 µmol/L celecoxib, the reduction in PTPRJ expression induced by COX-2 over-expression was not only rescued but in fact increased by 2.05-fold (± 0.28) and 3.34-fold (± 0.37), respectively, compared with control. CONCLUSIONS: Our results suggest that COX-2/PGE2 signaling may function as a negative regulator of PTPRJ expression in endothelial cells both in vivo and vitro.

Shipping in China plays a global role, and has led worldwide maritime transportation for the last decade. However, without taking national or local port boundaries into account, it is impossible to determine the responsibility that each local authority has on emission controls, nor compare them with land-based emissions to determine the priority for controlling these emissions. In this study, we provide national- to port-level inventories for China. The results show that in 2013, the total emissions of CO, non-methane volatile organic compounds (NMVOCs), nitrogen oxides (NO _x ), particulate matter (PM), SO _2 and CO _2 were 0.0741 ± 0.0004 Tg∙yr ^−1 , 0.0691 ± 0.0004 Tg∙yr ^−1 , 1.91 ± 0.01 Tg∙yr ^−1 , 0.164 ± 0.001 Tg∙yr ^−1 , 1.30 ± 0.01 Tg∙yr ^−1 and 86.3 ± 0.3 Tg∙yr ^−1 in China, respectively. By providing high-resolution spatial distribution maps of these emissions, we identify three hotspots, centered on the Bohai Rim Area, the Yangtze River Delta and Pearl River Delta. These three hotspots account for 8% of the ocean area evaluated in this study, but contribute around 37% of total shipping emissions. Compared with on-road mobile source emissions, NO _x and PM emissions from ships are equivalent to about 34% and 29% of the total mobile vehicle emissions in China. Moreover, this study provides detailed emission inventories for 24 ports in the country, which also greatly contributes to our understanding of global shipping emissions, given that eight of these ports rank within the top twenty of the port league table. Several ports in China suffer emissions 12–147 times higher than those at Los Angeles port. The ports of Ningbo-Zhou Shan, Shanghai, Hong Kong and Dalian dominate the port-level inventories, with individual emissions accounting for 28%–31%, 10%–14%, 10%–12% and 8%–14% of total emissions, respectively.
