6. Analysis and mechanism of biochar on soil microbial community structure of tobacco bacterial wilt

Abstract

As tobacco bacterial wilt is a bacterial disease that is increasingly harmful to tobacco plants′ growth, it is of great significance to explore the changes in the soil microbial community structure and its biological regulation mechanism after tobacco plant suffers from bacterial wilt. Field experiments were conducted to investigate the differences of bacterial community structure among four treatments: ZCTR(conventional fertilization, rhizosphere soil of healthy tobacco plants), ZCSW(application of 1.2 t·hm-2 biochar, rhizosphere soil of healthy tobacco plants), QKTR(conventional fertilization, rhizosphere soil of bacterial wilt tobacco plants), and QKSW(application of 1.2 t·hm-2 biochar, rhizosphere soil of bacterial wilt tobacco plants). After the biochar application, the bacteria′s α diversity increased, albeit without a significant difference. The bacterial community richness and structure changed. Under the same fertilization conditions, the bacterial α diversity and community abundance of bacterial wilt tobacco plants in the rhizosphere soil were less than those of healthy soil. The bacterial α diversity and community richness of the rhizosphere soil of diseased tobacco plants without biochar were higher than that of healthy tobacco plants, the abundance of Gemmatimonadetes and Acidobacteria increased by 0.26 and 1.27 percent points, while the abundance of Proteobacteria and Actinobacteria decreased by 1.08 and 0.14 percent points. Compared with no biochar treatment, the plant height, leaf length, leaf width and stem girth of the biochar application treatment increased by 11.83%, 16.88%, 6.70% and 10.80%, respectively, and the disease index decreased. The incidence rate of the bacterial wilt was reduced by 9.1 percent points with the biochar application treatment compared with the no biochar treatment. Studies have shown that biochar application can alleviate the decline in bacterial abundance caused by bacterial wilt, reduce the relative abundance of pathogenic bacteria, and drive the increase in the relative abundance of functional growth-promoting bacteria(Gemmatimonadetes). This reduces the incidence of bacterial wilt and preliminarily clarifies the biochar′s mechanism on the micro-ecological regulation of bacterial wilt in tobacco-growing soil.

16. Correlation analysis between soil and flue-cured tobacco composition and quality.

Abstract

In order to study the effect of soil composition on flue-cured tobacco composition and quality in tobacco planting area, Nanping K326 tobacco was selected to measure the contents of basic components in soil and tobacco, and the correlation between contents and smoking quality was further analyzed. The results show that the contents of available nitrogen and phosphorus in soil are positively correlated with the content of nicotine in C3f flue-cured tobacco ( r> 0. 64) . The contents of total nitrogen, total sugar and reducing sugar are positively correlated with soil organic matter ( r> 0. 72). The C3F volatility is positively correlated with soil available potassium ( r > 0. 96 ) and total nitrogen and potassium ( r > 0. 65 ) . There is a positive correlation between volatility of B2f flue-cured tobacco and total nitrogen ( r> 0. 84) and sugar ( r> 0. 74) . The results indicate that soil nutrients affect the content of chemical composition in flue-cured tobacco, as well as tobacco quality and class. The volatility of flue-cured tobacco relates to the contents and their proportion of total nitrogen, potassium, nicotine and total sugar. [ABSTRACT FROM AUTHOR]

18. Microstructural Modeling and Simulation of a Carbon Black-Based Conductive Polymer-A Template for the Virtual Design of a Composite Material.

Abstract

Carbon black is the most frequently applied conductive additive in rubber and polymer composites. In this work, we show how a carbon black microstructure in a polymer matrix can be conclusively modeled based on carbon black aggregation as well as an agglomeration mechanism using a state-of-the-art mathematical model. This novel and flexible microstructural modeling method enables us to virtually investigate the morphology of conductive additives within a polymer matrix and can be adapted to many conductive polymer combinations used for different applications. Furthermore, we calculate the electrical conductivity of the composite using a finite volume-based as well as a discrete element-based simulation technique and validate the results with experimental data. Utilizing a novel discrete element method (DEM) modeling technique, we were able to improve calculation times by a factor of 12.2 compared to finite volume method (FVM) simulations while maintaining high accuracy.
