17. Causal relationship between circulating vitamin C and 25-hydroxyvitamin D concentrations and common mental disorders - a Mendelian randomization study

Abstract

Mental disorders seriously affect people's health and social stability. This Mendelian randomization (MR) study was designed to investigate the causal relationship between circulating vitamin C (VC) or 25-hydroxyvitamin D (25(OH)D) levels and mental disorders. The data used for the MR analysis were derived from the summary genome-wide association studies (GWAS) database for VC and 25(OH)D and from the FinnGen consortium for fourteen mental disorders. Based on the inverse variance weighted (IVW) method, we found a potential causal association between circulating VC and anxiety disorders (IVW: OR=1.139, 95 %CI: 1.0231.269, P= 0.018). However, no causal association was found between VC or 25(OH)D and other mental disorders (P> 0.05). In the reverse MR analysis, individuals with Alzheimer's disease was causally associated with higher concentrations of circulating VC(P= 0.012), while individuals with anxiety disorders had a negative association between the concentrations of 25(OH)D (P= 0.012). However, the current evidence does not support a causal relationship between VC or 25(OH)D and other mental disorders. In addition, there was no causal association between circulating VC and 25(OH)D (P> 0.05). Future studies are needed to confirm these findings and to elucidate the mechanisms of potential causality.

Abstract

The normal growth and development of skeletal muscle is essential for the health of the body. The regulation of skeletal muscle by intestinal microorganisms and their metabolites has been continuously demonstrated. Acetate is the predominant short-chain fatty acids synthesized by gut microbiota through the fermentation of dietary fiber; however, the underlying molecular mechanisms governing the interaction between acetate and skeletal muscle during the rapid growth stage remains to be further elucidated. Herein, specific pathogen-free (SPF) mice, germ-free (GF) mice, and germ-free mice supplemented with sodium acetate (GS) were used to evaluate the effects of acetate on the skeletal muscle growth and development of young mice with gut microbiota deficiency. We found that the concentration of serum acetate, body mass gain, succinate dehydrogenase activity, and expression of the myogenesis maker gene of skeletal muscle in the GS group were higher than those in the GF group, following sodium acetate supplementation. Furthermore, the transcriptome analysis revealed that acetate activated the biological processes that regulate skeletal muscle growth and development in the GF group, which are otherwise inhibited due to a gut microbiota deficiency. The in vitro experiment showed that acetate up-regulated Gm16062 to promote skeletal muscle cell differentiation. Overall, our findings proved that acetate promotes skeletal muscle growth and development in young mice via increasing Gm16062 expression. [ABSTRACT FROM AUTHOR]

Abstract

The evidence regarding the beneficial effects of probiotics/synbiotic supplementation have been revealed by several meta-analyses, however some of these studies have fielded inconsistent results and a conclusion has yet to be reached. Therefore, the aim of present umbrella meta-analyses was to assess relevant evidence and elucidate the efficacy of probiotics/synbiotic supplementation in glycemic control. A comprehensive search in four databases (Cochrane library, PubMed, Web of science and Scopus) was performed to collect relevant studies up to August 2022, the pooled effects were measured with the use of random/fix-effect model depends on the heterogeneity. A total of 47 eligible meta-analyses involving 47,720 participants were identified to evaluate the pooled effects. The overall results showed that probiotics/synbiotic supplementation delivered significant decreases in fast plasma glucose (ES = −0.408, 95% CI: −0.518, −0.298; P < 0.001; I2 = 82.996, P < 0.001), fast plasma insulin (ES = −1.165, 95% CI: −1.454, −0.876; P < 0.001; I2 = 89.629, P < 0.001), homeostasis model assessment of insulin resistance (ES = −0.539, 95% CI: −0.624, −0.454; P < 0.001; I2 = 56.716, P < 0.001), and glycosylated hemoglobin (ES = −0.186, 95% CI: −0.270, −0.102; P < 0.001; I2 = 59.647, P = 0.001). Subgroup analysis showed that patients with impaired glucose homeostasis might benefit the most from probiotics/synbiotic supplementation. 31. Analysis and Experiment of Vibration Isolation Performance of a Magnetic Levitation Vibration Isolator with Rectangular Permanent Magnets

Abstract

Purpose: Magnetic levitation vibration isolator (MLVI) is an efficient method to suppress the external oscillations transmitted to some small instruments in the laboratories. Methods: This paper first presents a basic configuration of a kind of MLVI with four sets of the repulsive rectangular permanent magnets. Then, the parametric studies are carried out to discuss how the structure and air gap parameters influence the magnetic force, stiffness, and natural frequency of the MLVI. Moreover, as an MLVI case study, an operating point is set and the size dimensions of the MLVI are determined based on the parametric studies. The transmissibility of the MLVI is calculated by the dynamic equation at the operating point. Results: An experimental table is constructed for the analytical validation. The experimental results proved the validity of the theoretical analysis and showed that there are good consistencies between the experiment results and the theoretical calculation, which obtains that the natural frequency of the MLVI is 4.75 Hz and the initial cut-off frequency of the MLVI is approximately 6 Hz at the operating point. When the frequency of the external oscillations reaches 16 Hz, the attenuation amplitude becomes -?20 dB. Conclusion: All of these show ability of the proposed MLVI to attain low-frequency vibration isolation, and provide a new type of basic passive MLVI to exploit an active MLVI for practical applications.

Abstract

In this work, a poly(vinylidenefluoride-co-trifluoroethylene) (PVTF) coating on a titanium (Ti) substrate was prepared, and Ti metal surfaces were treated by physical or chemical methods to achieve a high bonding strength with PVTF. Scanning electron microscopy (SEM), atomic force microscope (AFM), and static water contact angles (WCA) were used to characterize the Ti metal surfaces. Further, mechanical stretching testing was employed to measure the bonding strength of PVTF coatings. The possible mechanism for the improved bonding strength could be the higher OH concentrations on Ti metal surfaces, which could lead to the formation of chemical bonds with the F atom of PVTF chains. Finally, a CCK-8 analysis of bone marrow mesenchymal stem cells (BMSCs) cultured on the PVTF coatings confirmed that the physical and chemical treatments had no significant differences in biocompatibility.
