This study focused on improving the flavor quality of seasonings, and enzymatic hydrolysis of soybean protein isolate (SPI) seasoning via traditional technology may lead to undesirable flavors. Herein, we aimed to develop a new type of SPI seasoning through microbial fermentation to improve its flavor quality. The effect of Corynebacterium glutamicum fermentation on the flavoring compounds of seasonings in SPI enzymatic hydrolysate was examined. Sensory evaluation showed that the SPI seasoning had mainly aromatic and roasted flavor, and the response signals of S18 (aromatic compounds), S24 (alcohols and aldehydes), and S25 (esters and ketones) sensors of the electronic nose differed significantly. Overall, 91 volatile compounds were identified via gas chromatography–mass spectrometry. SPI seasonings contained a higher number of alcohols, ketones, aromatics, and heterocyclic compounds than traditional seasonings, which had stronger cheese, fatty, and roasted aromas. According to the relative odor activity value (ROAV) analysis, n-pentylpyrzine, 2,6-dimethylpyrazine, and tetramethylpyrazine are the key flavoring compounds (ROAV ≥ 1) of SPI seasoning, which may impart a unique roasted and meaty aroma. Therefore, the fermentation of SPI enzymatic hydrolysate with C. glutamicum may improve the flavor quality of its products, providing a new method for the development and production of new seasoning products. Tomato is perishable and requires preservation to extend its shelf life. In this study, we conducted selection processes to identify essential oils that can help to avoid spoilage and deterioration during tomato storage and extend the shelf life. Thereafter, we determined the phosphatase activity assay, potassium ion concentration, and electron microscopy to study the antibacterial mechanism of essential oil. We found that Enterobacter ludwigii W01 was the dominant spoilage bacterium in tomatoes with cracked and curled skin. We selected oregano essential oil from 12 essential oils (oregano essential oil, lemon essential oil, osmanthus essential oil, cypress wood essential oil, tea tree essential oil, licorice essential oil, Baili essential oil, white camphor essential oil, Shancang seed essential oil, rosemary essential oil, rose essential oil, and cinnamon essential oil) which could significantly inhibit the activity of E. ludwigii W01. However, the diameter of the inhibition zone for Wh, Te, Cy, Li, Rm, Le, and Os is 0 mm, the diameter of the inhibition zone for Ba, Sh, and Ro was less than 1.0 mm, whereas the diameter of the inhibition zone for Ci and Or was greater than 2.0 mm. The diameter of the suppression circle for Ci and Or was greater than 2.0 mm, while Ci was lesser than Or. Oregano essential oil can damage the cell wall of E. ludwigii W01, leading to the leakage of the alkaline phosphatase stored between the cell wall and the cell membrane which can increase the alkaline phosphatase activity in the bacterial solution. Meanwhile, the addition of oregano essential oil significantly altered the cellular morphology of E. ludwigii W01. Spraying the surface of fresh tomato fruits with 1 MIC (0.125%) of oregano essential oil prolonged the storage time to 15 days, without significant changes in its sensory attributes. Objective: Several laboratory and imaging assays are required to diagnose multiple myeloma (MM). Serum and urine immunofixation electrophoresis are two key assays to diagnose MM, while they have not been extensively utilized in Chinese hospitals. Serum light chain (sLC), β2 microglobulin (β2-MG), lactic dehydrogenase (LDH), and immunoglobulin (Ig) are routinely measured in the majority of Chinese hospitals. Imbalance of sLC ratio (involved light chain/uninvolved light chain) is frequently observed in MM patients. This study aimed to evaluate the screening value of sLC ratio, β2-MG, LDH, and Ig in MM patients using receiver operating characteristic (ROC) curves. Methods: Data of 303 suspected MM patients, who were admitted to the Taizhou Central Hospital between March 2015 and July 2021, were retrospectively analyzed. In total, 69 patients (MM arm) met the International Myeloma Working Group (IMWG) updated criteria for the diagnosis of MM, while 234 patients were non-MM (non-MM arm). All patients' sLC, β2-MG, LDH, and Ig were measured using commercially available kits according to the manufacturer's instructions. The ROC curve analysis was employed to assess the screening value of sLC ratio, β2-MG, LDH, creatinine (Cr) and Ig. The statistical analysis was carried out by SPSS 26.0 (IBM, Armonk, NY, USA) and MedCalc 19.0.4 (Ostend, Belgium) software. Results: There was no significant difference between the MM and non-MM arms in terms of gender, age and Cr. The median sLC ratio in the MM arm was 11.5333, which was significantly higher than that of 1.9293 in the non-MM arm (P<0.001). The area under the curve (AUC) of sLC ratio was 0.875, which indicated a robust screening value. The optimal sensitivity and specificity were 81.16% and 94.87% respectively, when the sLC ratio was set as 3.2121. The serum levels of β2-MG and Ig were higher in the MM arm than those in the non-MM arm (P<0.001). The AUC values of β2-MG, LDH, and Ig were 0.843 (P<0.001), 0.547 (P = 0.2627), and 0.723 (P<0.001), respectively. The optimal cutoff values of β2-MG, LDH, and Ig were 1.95 mg/L, 220 U/L, and 46.4 g/L respectively, in the context of screening value. The triple combination of sLC ratio (3.2121), β2-MG (1.95 mg/L), and Ig (46.4 g/L) yielded a higher screening value compared with that of sLC ratio alone (AUC, 0.952; P<0.0001). The triple combination had a sensitivity of 94.20% and a specificity of 86.75%. The addition of LDH to the triple combination and formation of quadruple combination did not optimize the screening value, with AUC, sensitivity, and specificity of 0.952, 94.20%, and 85.47%, respectively. Conclusion: The triple combination strategy (sLC ratio, 3.2121; β2-MG, 1.95 mg/L; Ig, 46.4 g/L) is accompanied by remarkable sensitivity and specificity for screening MM in Chinese hospitals. During microbial industrial production, microorganisms often face diverse stressors, including organic solvents, high salinity, and high sugar levels. Enhancing microorganism tolerance to such stresses is crucial for producing high-value-added products. Previous studies on the mechanisms of 2-phenylethanol (2-PE) tolerance in Saccharomyces cerevisiae revealed a potential connection between the sugar transporter-like protein (Stl1) mutation (F427L) and increased tolerance to high sugar and salt stress, suggesting a broader role in multistress tolerance. Herein, we showed that the Stl1 F427L mutant strain (STL) exhibits significantly improved multistress tolerance in the presence of glycerol. Molecular dynamics simulations indicated that Stl1 F427L may enhance glycerol molecular binding, resulting in a significant increase in the intracellular glycerol content of the mutant strain STL. Additionally, under multistress conditions, pyruvate and ergosterol levels and catalase (CAT) and superoxide dismutase (SOD) activities were significantly increased in the mutant strain STL compared with the control strain 5D. This resulted in a notable increase in cell membrane toughness and a decrease in intracellular reactive oxygen species levels. These findings highlight the mechanism by which Stl1 F427L enhances S. cerevisiae tolerance to multistress. Importantly, they provide novel insights into and methodologies for improving the resilience of industrial microorganisms., Importance: Stl1 F427L exhibits improved strain tolerance to multistress when adding glycerol, may enhance glycerol molecular binding, and can make a significant increase in intracellular glycerol content. Granulocytic sarcoma (GS), also known as extramedullary myeloid tumor, is a rare malignant neoplasm composed of immature myeloid cells. Although it is most commonly associated with acute myeloid leukemia (AML), a subset of GS cases can occur prior to the development of AML. GS can present in a variety of extramedullary locations, including bone, skin, lymph nodes, and the female reproductive system. We report the case of a 45-year-old Asian female patient who presented to our hospital in July 2015 with a three-month history of left-sided lumbago. A contrast-enhanced computed tomography (CT) scan of the abdomen and pelvis revealed a retroperitoneal mass and an enlarged uterine cervix with an accompanying 57×52 mm mass. A cervical biopsy confirmed the diagnosis of GS. Immunohistochemical (IHC) analysis of the biopsy showed that the neoplastic cells were positive for CD34, CD15, CD33, CD43, lysozyme, and myeloperoxidase. The patient was subsequently treated with an idarubicin and cytarabine-based regimen for four cycles. A follow-up CT scan of the abdomen and pelvis demonstrated a significant reduction in the size of the previous lesions. Unfortunately, the patient passed away in April 2016 due to a cerebral hemorrhage. Introduction: Outcome-based education (OBE) leads to revolutionary change in medical education, but each medical school is acknowledged to develop suited framework based on specific positioning, educational philosophy and expected outcomes., Methods: Based on the graduate outcomes of basic medical education in China released by Working Committee for the Accreditation of Medical Education (WCAME) which officially recognized by World Federation for Medical Education (WFME), Taizhou University re-documented the overall organization of the curriculum and classified the courses as "Crucial, Supporting and Associated (CSA)" categories to connect the graduate outcomes and course objectives., Results: We redefined the standard for graduates in Taizhou University Medical School including 34 items in four domains and redesigned the curriculum with 76 courses classified as CSA. Empirical data reveals a substantial improvement in students' performance on Standardized Competence Test for Clinical Medicine Undergraduates in China (SCTCMU) by implementing the CSA system between 2022 and 2023. Purpose: To detect JAK2 p.V617F and measure allele burden in peripheral blood (PB) and bone marrow (BM) aspirates in patients with suspected myeloproliferative neoplasms (MPNs)., Methods: Patients with suspected MPNs were prospectively enrolled between August 2017 and May 2019, and their PB and BM were collected during the same period. Quantitative fluorescence polymerase chain reaction (PCR) was used to detect the copy number of JAK2 wild type and the V617F mutant; the JAK2 V617F proportion was also calculated. The JAK2 p.V617F proportion in PB was compared to that in BM by Chi-square test., Results: Among 54 patients with suspected MPNs, 43 of them were eligible for analysis. The JAK2 p.V617F in PB had the same sensitivity and specificity as BM (all P>0.05). Background: To assess the value of plasma vitamin D level and nomogram model in predicting the prognosis of patients with small cell lung cancer (SCLC) treated with platinum plus etoposide (PPE) as first-line chemotherapy., Methods: In this retrospective study, we included 178 patients with SCLC. The data of 25(OH)D level, basic clinical information, overall survival (OS) and progression-free survival (PFS) were collected. Moreover, a nomogram was constructed to predict the prognosis of the patients., Results: The median OS value for patients with 25(OH)D < 10 ng/mL was 12.5 months. The median PFS value was 6.6 months. Sex, smoking status, clinical stage, and plasma vitamin D were independent prognostic predictors. Besides, the decision curve analysis and receiver operating characteristic curve indicated that the nomogram prediction models showed positive clinical benefit., Conclusions: The plasma vitamin D level is of great significance in prognosis of patients with SCLC. Objective: We developed a new culture-complex auto-inducing media (CAI) for heterogenous protein expression in Escherichia coli., Methods: To test expression efficiency in the CAI, we constructed seven different plasmids named p-1, p-2, p-3, p-4, p-5, p-6 and p-7. These plasmids were transformed into E. coli BL21, then expressed in both Luria-Bertani madia LB and CAI. To improve the expression level even more, we analyzed the composition of the CAI and optimized the culture., Results: The expression levels of seven fusion proteins in CAI were four times higher than those in Luria-Bertani. Through a series of changes we formed a new optimized culture (CAI-4)., Conclusion: Comparing to the expression levels of these fusion proteins (P-1, P-2, P-3) in CAI, the expression levels of fusion proteins in CAI-4 increased 2-fold.
