Abstract

Introduction: Palatal displacement of maxillary anterior teeth is common in clinical practice. Previous studies have reported that the labial bone around palatally‐displaced incisors is thinner than that around normally‐placed teeth. Therefore, it is necessary to elucidate alveolar bone changes after alignment to guide orthodontic treatment. In this study, we investigated the alveolar bone changes around palatally‐displaced maxillary lateral incisors before and after treatment, and the effects of extraction and age using cone‐beam computed tomography. Methods: In this retrospective study, 55 patients with unilateral palatally‐displaced maxillary lateral incisors were included. Three‐dimensional alveolar bone changes were measured at three levels (25%, 50% and 75% of the root length) using cone‐beam computed tomography. Group comparisons were made between displaced and control teeth, extraction and non‐extraction groups, and adult and minor groups. Results: After orthodontic treatment, labiopalatal and palatal alveolar bone widths decreased at all measured levels. Labial alveolar bone width increased significantly at P25, but decreased at P75. Concavity decreased, while tooth‐axis angle, tooth length, B‐CEJ and P‐CEJ increased. Changes in LB and LP at P75, B‐CEJ and P‐CEJ were statistically significant. After treatment, the tooth‐axis angle on the PD side increased by 9.46°. The change in tooth‐axis angle on the PD side was significantly smaller, and LB and LP decreased more at P75, in the extraction group. Conclusions: Compared to the control teeth, alveolar bone thickness and height for the displaced teeth decreased more significantly after treatment. Abstract

Abstract Background Studies focusing on accuracy of intraoral digital models in the palatal region are scarce. The present study aimed to investigate the influence of different scanning sequences on palatal trueness and to assess deviation and distribution character of trueness in palate. Methods Overall, 35 participants accepted three types of procedures to acquire upper digital models. Indirect models digitalised from plaster models were considered as the reference. Two direct digital models were acquired using TRIOS 3 POD intraoral scanners, namely Groups Tr1 and Tr2, wherein intraoral scanning differed in terms of palatal scanning sequences. Based on a modified dental-level superimposition method, 3D measurements of trueness in palate and palatal vault region (PVR) for palatal stable regional superimposition in Groups Tr1 and Tr2, respectively, were performed. Absolute deviations were measured for trueness, while signed deviations were analysed for shape distortion. Colour-coded maps were used for quantitative analysis of deviation distribution pattern. Paired t test was used to analyse differences in palatal trueness between different scanning sequences. One-way repeated-measures analysis of variance and Bonferroni test were used to compare trueness measurements among different superimposition methods. Intraclass correlation coefficient (ICC) was used to verify reproducibility of the proposed method. Results Palatal trueness in Group Tr1 (118.59 ± 37.67 μm) was slightly less accurate than that (108.25 ± 33.83 μm) in Group Tr2 (p = 0.012 0.90. Conclusions Scanning sequences can affect palatal trueness. Palatal scanning should be initiated at the palatal side of the posterior teeth where the initial scan begins. For 3D PVR superimposition, distal boundary of the selected region should be adjusted mesially whilst referring to intraoral digital models. Abstract

Many SARS-CoV-2 neutralizing antibodies (nAbs) lose potency against variants of concern. In this study, we developed 2 strategies to produce mutation-resistant antibodies. First, a yeast library expressing mutant receptor binding domains (RBDs) of the spike protein was utilized to screen for potent nAbs that are least susceptible to viral escape. Among the candidate antibodies, P5-22 displayed ultrahigh potency for virus neutralization as well as an outstanding mutation resistance profile. Additionally, P14-44 and P15-16 were recognized as mutation-resistant antibodies with broad betacoronavirus neutralization properties. P15-16 has only 1 binding hotspot, which is K378 in the RBD of SARS-CoV-2. The crystal structure of the P5-22, P14-44, and RBD ternary complex clarified the unique mechanisms that underlie the excellent mutation resistance profiles of these antibodies. Secondly, polymeric IgG enhanced antibody avidity by eliminating P5-22's only hotspot, residue F486 in the RBD, thereby potently blocking cell entry by mutant viruses.
