26. Identification of key immune genes of osteoporosis based on bioinformatics and machine learning

Abstract

IntroductionImmunity is involved in a variety of bone metabolic processes, especially osteoporosis. The aim of this study is to explore new bone immune-related markers by bioinformatics method and evaluate their ability to predict osteoporosis.MethodsThe mRNA expression profiles were obtained from GSE7158 in Gene expression Omnibus (GEO), and immune-related genes were obtained from ImmPort database (https://www.immport.org/shared/). immune genes related to bone mineral density(BMD) were screened out for differential analysis. protein-protein interaction (PPIs) networks were used to analyze the interrelationships between different immune-related genes (DIRGs). Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses of DIRGs function were performed. A least absolute shrinkage and selection operation (LASSO) regression model and multiple Support Vector Machine-Recursive Feature Elimination (mSVM-RFE) model were constructed to identify the candidate genes for osteoporosis prediction The receiver operator characteristic (ROC) curves were used to validate the performances of predictive models and candidate genes in GEO database (GSE7158,GSE13850).Through the RT - qPCR verify the key genes differentially expressed in peripheral blood mononuclear cells Finally, we constructed a nomogram model for predicting osteoporosis based on five immune-related genes. CIBERSORT algorithm was used to calculate the relative proportion of 22 immune cells.ResultsA total of 1158 DEGs and 66 DIRGs were identified between high-BMD and low-BMD women. These DIRGs were mainly enriched in cytokine−mediated signaling pathway, positive regulation of response to external stimulus and the cellular components of genes are mostly localized to external side of plasma membrane. And the KEGG enrichment analysis were mainly involved in Cytokine−cytokine receptor interaction, PI3K−Akt signaling pathway, Neuroactive ligand−receptor interaction,Natural killer cell mediated cytotoxicity. Then five key genes (CCR5, IAPP, IFNA4, IGHV3-73 and PTGER1) were identified and used as features to construct a predictive prognostic model for osteoporosis using the GSE7158 dataset.ConclusionImmunity plays an important role in the development of osteoporosis.CCR5, IAPP, IFNA4, IGHV3-73 and PTGER1were play an important role in the occurrences and diagnosis of OP.

Abstract

Aiming to address the problem of regional dynamic target search under weak communication conditions, this paper proposes a UUV cluster search method based on cumulative probability optimization. First, by estimating the probability distribution of the initial target location, an initial probability map is established. Then, based on the Bayesian model and Markov decision model, the target probability distribution is periodically updated, and based on the cumulative detection probability optimal principle of the UUV cluster, the UUV cluster is guided to search the region with high detection probability preferentially. Finally, we implement the simulation experiment and compare with the random search method. The results verify that the proposed method has higher search efficiency in the cases of without prior information and with prior information. [ABSTRACT FROM AUTHOR]

Abstract

Biochar, an eco-friendly, porous carbon-rich material, is widely studied for immobilizing heavy metals in contaminated environments. This study prepared tobacco stalks, a typical agricultural waste, into biochar (TSB) modified by hydroxyapatite (HAP) at co-pyrolysis temperatures of 350 °C and 550 °C to explore its Cd(II) adsorption behavior and relevant mechanisms. XRD, SEM–EDS, FTIR, and BET analyses revealed that HAP successfully incorporated onto TSB, enriching the surface oxygen-containing functional groups (P–O and carboxyl), and contributing to the enhancement of the specific surface area from 2.52 (TSB350) and 3.63 m2/g (TSB550) to 14.07 (HAP–TSB350) and 18.36 m2/g (HAP–TSB550). The kinetics of Cd(II) adsorption onto TSB and HAP–TSB is well described by the pseudo-second-order model. Isotherm results revealed that the maximum adsorption capacities of Cd(II) on HAP–TSB350 and HAP–TSB550 were approximately 13.17 and 14.50 mg/g, 2.67 and 9.24 times those of TSB350 and TSB550, respectively. The Cd(II) adsorption amounts on TSBs and HAP–TSBs increased significantly with increasing pH, especially in HAP–TSB550. Ionic strength effects and XPS analysis showed that Cd(II) adsorption onto HAP–TSBs occurred mainly via electrostatic interaction, cation exchange with Ca2+, complexation with P–O and –COOH, and surface precipitation. These findings will provide a modification strategy for the reutilization of tobacco agricultural waste in the remediation of heavy metal contaminated areas. [ABSTRACT FROM AUTHOR]

48. Rational design of animal-derived biochar composite for peroxymonosulfate activation: Understanding the mechanism of singlet oxygen-mediated degradation of sulfamethoxazole

Abstract

Animal-derived biochar are identified as a promising candidate for peroxymonosulfate (PMS) activation due to the abundant aromatics and oxygen-containing functional groups. The current investigation focuses on pig carcass-derived biochar (800-BA-PBC) by ball milling-assisted alkali activation. The results showed that 800-BA-PBC could effectively activate PMS and degraded 94.2% sulfamethoxazole (SMX, 10 mg/L) within 40 min. The reaction rate constant was found to be 47 times higher than that observed with PBC. The enhanced catalytic activity is mainly attributed to the increase in specific surface area, the increase content of oxygen-containing groups on the surface, and the formation of graphitic nitrogen. The quenching tests and electron paramagnetic resonance (EPR) analysis demonstrated that 1O2 is the main active species in the degradation of SMX. Moreover, the 800-BA-PBC + PMS system can maintain excellent degradation rate under different water quality, wide pH range, and the presence of different anions. The degradation pathways of SMX in the optimal system are also evaluated through intermediate identification and DFT calculation. These results indicate that the catalytic system has high anti-interference ability and practical application potential. This investigation provides new insight into the rational design of animal-derived biochar and develops a low-cost technology for the treatment of antibiotic containing wastewater. © 2023 Elsevier Ltd
