Endocrine-disrupting chemicals (EDCs) are now ubiquitously distributed in the environment. Tetrabromobisphenol A bis(2-hydroxyethyl) ether (TBBPA-DHEE) pollution in environment media poses a significant threat to humans and aquatic organisms as a result of its potential neurotoxicity and endocrine-disrupting effect. The endocrine-disrupting effects of TBBPA-DHEE on aquatic organisms, however, have received limited attention. In this study, the neurotoxicity and reproductive endocrine-disruptive effect of TBBPA-DHEE was evaluated by observing the neurobehavioral changes, vitellogenin (VTG), testosterone, 17β-estradiol and gene expression levels in adult male and female zebrafish exposed to TBBPA-DHEE (0.05, 0.2 and 0.3 mg/L) for 100 days. Furthermore, transcriptomic analysis was conducted to unravel other potential neuroendocrine-disrupting mechanism. Our result showed TBBPA-DHEE significantly (p 0.05) altered the locomotor behavior and motor coordination abilities in both sexes. Steroid hormone and VTG levels were also altered indicating the neuroendocrine-disrupting effect of TBBPA-DHEE on the hypothalamic-pituitary-gonadal-axis. A total of 1568 genes were upregulated and 542 genes downregulated in males, whereas, 1265 upregulated and 535 downregulated genes were observed in females. The KEGG enrichment analysis showed that cell cycle and p55 signaling pathways were significantly enriched due to TBBPA-DHEE exposure. These pathways and its component genes are potential target of EDCs. The significant upregulation of genes in these pathways could partly explain the neuroendocrine disrupting effect of TBBPA-DHEE. The observed toxic effects of TBBPA-DHEE observed in this study is confirmation of the endocrine-disrupting toxicity of this chemical which would be valuable in biosafety evaluation and biomonitoring of TBBPA-DHEE for public health purposes.

Background The ultrasensitive monitoring strategy of zearalenone (ZEN) is essential and desirable for food safety and human health. In the present study, a coupling of gold nanoparticles-DNA barcode and direct competitive immunoassay-based real-time polymerase chain reaction signal amplification (RT-IPCR) for ZEN close to the sensitivity of PCR-like levels is described and evaluated. Results The RT-IPCR benefited from the use of a DNA barcode and RT-PCR detection strategy, thus resulting in ultrasensitive and simple detection for ZEN. Under the optimal RT-IPCR, the linear range of detection was from 0.5 to 1000 pg mL-1 and the limit of detection was 0.5 pg mL-1 , which was 400-fold lower than the enzyme-linked immunosorbent assay. The detection procedure was simplified and the detection time was shortened. The specificity, accuracy and precision of the RT-IPCR confirmed a high performance. ZEN-positive contamination levels were from 0.056 to 152.12 ng g-1 by the RT-IPCR, which was demonstrated to be highly reliable by liquid chromatography-tandem mass spectrometry. Conclusion The proposed RT-IPCR could be used as an alternative for detecting ZEN with satisfactory ultrasensitivity, simplicity, low cost and high-throughput. The present study could provide a strategy for the ultrasensitive detection of the small molecule with a simple and practical approach, which has significant appeal and application prospects.

In the present study, the neurotoxicity and mechanisms of di-(2-ethylhexyl) phthalate (DEHP) exposure on pubertal normal (P-normal) and pubertal type 2 diabetes mellitus (P-T2DM) mice were investigated by typical neurobehavioral methods and transcriptome analysis. Pubertal male ICR mice were orally exposed to DEHP (0.18, 1.8, 18 and 180 mg/kg/d) for 3 weeks. In Open field test, DEHP significantly increased the time in central area staying and decreased the total distance and clockwise (CW) rotation of P-normal and P-T2DM mice. Morris water maze showed that DEHP significantly increased the latency in locating platform and decreased the original platform quadrant and residence time in target quadrant of P-normal and P-T2DM mice. Transcriptome analysis results revealed the effects of DEHP exposure on neural signaling pathway including biogenic amines neurotransmitters, nerve receptors, neurobiological processes, etc. Enzyme-linked immunosorbent assay (ELISA) and western blotting results showed that DEHP significantly decreased the contents of 5-HT, cAMP, GABA and Ca2+, the levels of CREB, phosphorylation of PKA, ERK1/2 and CREB, increased the levels of CaM and phosphorylation of CaMKII in P-normal and P-T2DM mice. Factorial analysis results showed that P-T2DM mice were more sensitive than those of P-normal mice. The potential neurotoxicity mechanism of DEHP may be synergistically mediated by the cAMP-PKA-ERK1/2-CREB signaling and the Ca2+ signaling pathway.

Amitriptyline (AMI), the most commonly prescribed tricyclic antidepressant, is widely detected in water environments. Exposure to AMI may lead to diverse adverse effects on aquatic organisms, but little is known about the effect of short-term exposure to AMI on the gut microbiota of aquatic organisms and their recovery characteristics. In the present study, adult zebrafish (Danio rerio) were exposed to AMI (0, 2.5, 10, and 40 μg/L) for seven days, and then allowed to recover in AMI-free culture water for 21 days. The exposure caused gut damages in all the AMI treated groups of zebrafish, which became more severe after recovery compared to the control group. AMI exposure also disturbed the microbiota of zebrafish guts and rearing water even after the 21-day recovery period. Furthermore, AMI exposure affected microbes involved in the substance and energy metabolic functions in zebrafish guts and tended to increase the abundance of microbial genera associated with opportunistic pathogens. In addition, the microbial predicted metabolic functions in AMI-exposed guts of zebrafish were significantly altered after the 21-day recovery period, explaining the persistent effects of short-term exposure to AMI. The results of this study suggest that acute exposure to AMI may have persistent impacts on the gut histomorphology and the gut microbiota in aquatic organisms.

Tributyltin (TBT) and polychlorinated biphenyls (PCBs) are typical organic pollutants that co-exist in the aquatic environment. Despite increasing awareness of their combined impacts, knowledge about their potential interactive effects remains limited. In this study, we investigated the effects of TBT-PCBs mixtures on the survival, development, and hatching of embryos of Japanese medaka (Oryzias latipes). Our goal was to assess the interactive effects of TBT and PCBs on early-life-stage fish. We used in ovo nanoinjection to simulate the maternal transfer process and to control exposure doses at 0, 7.5, 15, 30 ng/g-egg (for both toxins). A factorial design (4 × 4) corresponding to the above doses was used to define the formulation of mixtures. Results showed that injection of mixtures of TBT and PCBs into fertilized eggs caused significantly elevated rates of mortality, yolk-sac shrinkage, swim-up failure, and deformities of medaka embryos/larvae. TBT and PCBs interacted synergistically to induce the above developmental abnormalities. In contrast, exposure to only TBT increased the hatching times of embryos, whereas exposure to only PCBs decreased the hatching times. The dose ratios, dose levels, and interactive effect of TBT and PCBs in a mixture played significant roles in determining the final impact on time-to-hatching. Our results suggested that in ovo exposure to a mixture of TBT and PCBs may interact synergistically to induce developmental abnormalities in fishes.

Microplastic (MP) pollution and the related impacts on aquatic species have drawn worldwide attention. However, knowledge of the kinetic profiles of MPs in fish remains fragmentary. In this study, we conducted exposure and depuration tests of the following fluorescent-labeled MPs: polyethylene (PE; sphere with 200 or 20 µm diameter) and polystyrene (PS; sphere with 20 or 2 µm diameter) using juvenile Japanese medaka (Oryzias latipes). The distribution and concentration of MPs in medaka were directly determined in-situ after tissue transparency. During the 14-day exposure, MPs was mainly detected in the gastrointestinal tract, while some MPs at the size of ≤ 20 µm were located in the area of the gills and head. The bioconcentration factor (BCF; L/kg) for MPs in medaka was estimated as 74.4 (200 µm PE), 25.7 (20 µm PE), 16.8 (20 µm PS), and 139.9 (2 µm PS). Within the first five days of depuration, MPs were exponentially eliminated from the fish body, but 2 µm PS-MPs could be still detected in the gastrointestinal tract at the end of the 10-day depuration phase. Our results suggest that MPs 2 µm in diameter may pose ecological risks to aquatic species due to their relatively higher BCF and the potential for long-term persistence in the body.

Cetylpyridinium chloride (CPC) is a cationic surfactant that has been widely used as an antibacterial ingredient in pharmaceutical and personal care products. Due to its high residue in surface waters, there is increasing concern over the potential risk of CPC to aquatic ecosystems. However, knowledge of its impacts on fish is still limited. Therefore, this study exposed juvenile and adult zebrafish to CPC (0, 10, and 40 μg/L) for four days. Subsequently, changes in their behavioral traits and brain levels of several neurotransmitters were investigated. The behavioral assay showed that CPC exposure significantly decreased the locomotor activity and social interaction of zebrafish at both life stages, and juveniles were more sensitive to CPC exposure than adults. In the control groups, the brain neurotransmitters concentrations increased with age in zebrafish. However, CPC exposure tended to increase the brain neurotransmitter levels of juveniles but decreased their levels in adults. Correlation analysis revealed that the brain monoamine neurotransmitters and their turnover might play important roles in the life stage-dependent behavioral response to CPC. In particular, the DOPAC/DA ratio was significantly associated with CPC-induced hypoactivity and reduced social interactions in juveniles but not adults. Our findings demonstrated that CPC exposure could cause abnormal behavior in juvenile and adult zebrafish and disturb their brain neurotransmitters, even at environmentally relevant concentrations, and thus highlighted the necessity for further assessing its potential risks to aquatic ecosystems.

The combined effect of microplastics and pharmaceuticals on aquatic organisms is an issue of concern. In this laboratory study, we evaluated the combined effect of polystyrene microplastics (2-μm diameter) and diazepam on the social behavior of medaka (Oryzias latipes) by using the shoaling behavior test with five treatment groups: solvent control, polystyrene microplastics exposure (0.04 mg/L), low-concentration diazepam exposure (0.03 mg/L), high-concentration diazepam exposure (0.3 mg/L), and polystyrene microplastics and low-concentration diazepam co-exposure. After 7 days of exposure, the shoal-leaving behavior of the high-concentration diazepam exposure group (8.9 ± 8.3 counts/medaka) and the co-exposure group (6.8 ± 6.7 counts/medaka) was significantly greater than that in the solvent control group (1.8 ± 2.6 counts/medaka). Even after 5 days of recovery, medaka in the co-exposure group left the shoal more often (7.3 ± 5.0 counts/medaka) than those in the solvent control group (2.6 ± 2.6 counts/medaka), whereas the shoal-leaving behavior in other exposure groups, except for the high-concentration diazepam exposure group, was restored. Our findings show that the combined effects of diazepam and polystyrene microplastics suppressed medaka social behavior, suggesting that the presence of microplastics can enhance the adverse effects of pollutants on the social behavior of aquatic organisms.

We investigated the pollution levels of 6 heavy metals and 29 dioxins (polychlorinated dibenzo-p-dioxins (PCDDs), polychlorinated dibenzofurans (PCDFs), and dioxin-like polychlorinated biphenyls (DL-PCBs)) in intertidal and supratidal zones by using wharf roaches (Ligia spp.) collected from 12 sampling sites on the coast of Northeast Japan from November 2011 to June 2012. The total concentrations of heavy metals ranged from 177 to 377 µg/g-dry weight (dw), and the predominant metals were copper, zinc, and aluminum. The order of the detected level of heavy metals was zinc &gt, aluminum &gt, copper &gt, cadmium &gt, lead &gt, chromium, and this trend was similar to a previous report. The total toxic equivalent (TEQ) value of the PCDD/Fs ranged from less than the limit of detection (&lt, LOD) to 2.33 pg-TEQ/g-dw, and the predominant congener was octachlorodibenzodioxin (&lt, LOD to 110 pg/g-dw). Compared with PCDD/Fs, DL-PCBs were detected at a predominantly higher level (total TEQ value: 0.64–27.79 pg-TEQ/g-dw). Detected levels of dioxins, especially DL-PCBs in the wharf roach, were like those in the bivalves. These results indicate that the wharf roach could reflect heavy metals and dioxin pollution in the supratidal zones and is a suitable environmental indicator for these environmental pollutants. This is the first study to investigate heavy metals, PCDD/Fs, and DL-PCBs pollution in coastal isopods in Japan.

Peroxiredoxin (Prx) is a relatively recently discovered antioxidant enzyme family that scavenges peroxides and is known to be present in organisms from biological taxa ranging from bacteria to multicellular eukaryotes, including photosynthetic organisms. Although there have been many studies of the Prx family in higher plants, green algae, and cyanobacteria, few studies have concerned raphidophytes and dinoflagellates, which are among the eukaryotic algae that cause harmful algal blooms (HABs). In our proteomic study using 2-D electrophoresis, we found a highly expressed 2-Cys peroxiredoxin (2-CysPrx) in the raphidophyte Chattonella marina var. antiqua, a species that induces mass mortality of aquacultured fish. The abundance of the C. marina 2-CysPrx enzyme was highest in the exponential growth phase, during which photosynthetic activity was high, and it then decreased by about a factor of two during the late stationary growth phase. This pattern suggested that 2-CysPrx is a key enzyme involved in the maintenance of high photosynthesis activity. In addition, the fact that the depression of photosynthesis by excessively high irradiance was more severe in the 2-CysPrx low-expression strain (wild type) than in the normal-expression strain (wild type) of C. marina suggested that 2-CysPrx played a critical role in protecting the cell from oxidative stress caused by exposure to excessively high irradiance. In the field of HAB research, estimates of growth potential have been desired to predict the population dynamics of HABs for mitigating damage to fisheries. Therefore, omics approaches have recently begun to be applied to elucidate the physiology of the growth of HAB species. In this review, we describe the progress we have made using a molecular physiological approach to identify the roles of 2-CysPrx and other antioxidant enzymes in mitigating environmental stress associated with strong light and high temperatures and resultant oxidative stress. We also describe results of a survey of expressed Prx genes and their growth-phase-dependent behavior in C. marina using RNA-seq analysis. Finally, we speculate about the function of these genes and the ecological significance of 2-CysPrx, such as its involvement in circadian rhythms and the toxicity of C. marina to fish.

The widely used antifoulant tributyltin chloride (TBT) is highly toxic to aquatic organisms. In the present study, four-week-old Japanese medaka (Oryzias latipes) juveniles were orally exposed to TBT at 1 and 10 ng/g bw/d for 1, 2, and 4 weeks, respectively. Half of the tested medaka juveniles showed bone morphology alterations in both 1 and 10 ng/g bw/d TBT 4-week exposure groups. Nile Red (NR) staining showed that the juveniles exposed to 1 ng/g bw/d TBT for 2 and 4 weeks had significantly enlarged adipocyte areas. The mRNA-Seq analysis indicated that 1 ng/g bw/d TBT exposure for 2 weeks affected bone morphology through developmental processes. The GO and KEGG analyses suggested that the adipogenic effect of TBT observed in this study may be induced by metabolic processes, oxidative phosphorylation, and fatty acid degradation and metabolism pathways. Therefore, both morphological observation and mRNA-Seq analysis showed obesogenic effects and developmental toxicity of TBT to juvenile Japanese medaka.

Dibutyl phthalate (DBP) and diisobutyl phthalate (DiBP) have been reported to exhibit reproductive toxicity in vertebrates. However, the combined effect of DBP and DiBP on offspring of exposed parents remains unclear, especially for aquatic organisms such as fish. The aims of this study were to assess the effects of parental co-exposure to DBP and DiBP on early development of zebrafish offspring, and to explore the potential molecular mechanisms involved. The early developmental indicators and transcriptomic profiles of F1 larvae were examined after parental exposure to DBP, DiBP and their mixtures (Mix) for 30 days. Results showed that parental exposure to DBP and DiBP, alone or in combination, resulted in increased hatchability at 48 hpf and heart rate at 96 hpf, and increased the prevalence of malformations and mortality in F1 larvae. Generalized linear model (GLM) suggested an antagonistic interactive effect between DBP and DiBP on mortality and malformations of F1 larvae. The transcriptomic analysis revealed that the molecular mechanisms of parental co-exposure were different from those of either chemical alone. Disruption of molecular functions involved unfolded protein binding, E-box binding and photoreceptor activity in F1 larvae. These findings provide initial insights in the potential mechanism of action of parental co-exposure to DBP and DiBP.

The vulnerability to environmental insults is heightened at early stages of development. However, the neurotoxic potential of bisphenol A (BPA) and bisphenol S (BPS) at developmental windows remains unclear. To investigate the mechanisms mediating the developmental neurotoxicity, zebrafish embryos were treated with 0.01, 0.03, 0.01, 0.3, 1 μM BPA/BPS. Also, we used Tg(HuC:GFP) zebrafish to investigate whether BPA/BPS could induce neuron development. The reduction in body length, and increased heart rate were significant in 0.3 and 1 μM BPA/BPS groups. The green fluorescence protein (GFP) intensity increased at 72 hpf and 120 hpf in Tg(HuC:GFP) larvae which was consistent with the increased mRNA expression of elval3 following BPS treatments, an indication of the plausible effect of BPS on embryonic neuron development. Additionally, BPA/BPS treatments elicited hyperactivity and reduced static time in zebrafish larvae, suggesting behavioral alterations. Moreover, qRT-PCR results showed that BPA and BPS could interfere with the normal expression of development-related genes vegfa, wnt8a, and mstn1 at the developmental stages. The expression of neurodevelopment-related genes (ngn1, elavl3, gfap, α1-tubulin, mbp, and gap43) were significantly upregulated in BPA and BPS treatments, except for the remarkable downregulation of mbp and gfap elicited by BPA at 48 (0.03 μM) and 120 hpf (0.3 μM) respectively; ngn1 at 48 hpf for 0.1 μM BPS. Overall, our results highlighted that embryonic exposure to low concentrations of BPA/BPS could be deleterious to the central nervous system development and elicit behavioral abnormalities in zebrafish at developmental stages.

By combining the photopolymerization method with surface imprinting techniques, a magnetic photocatalyst modified with a POPD–CdS heterojunction embedded imprinted layer (a POPD–CdS heterojunction embedded magnetic imprinted photocatalyst) was successfully prepared. And ciprofloxacin (CIP) can be recognized and degraded by the POPD–CdS heterojunction embedded magnetic imprinted photocatalyst. The coefficient of selectivity of the POPD–CdS heterojunction embedded magnetic imprinted photocatalyst relative to CdS, POPD–CdS and a non-imprinted photocatalyst were 1.88, 1.98 and 2.12, respectively. Moreover, the secondary pollution resulting from CdS photo-corrosion can also be effectively suppressed by the surface imprinted layer. More importantly, the POPD–CdS heterojunction, as a functional monomer, was embedded into the surface imprinted layer. This method not only ensured sufficient contact between POPD and CdS, but also effectively solved the problem that the photocatalytic activity decreased drastically because of coverage by the surface imprinted layer. Therefore, this work offers an innovative concept and technical approach for the application of the selective removal of specific target pollutants in complex water environments.

Discontinuation of amitriptyline (AMI) has been demonstrated to induce long-term withdrawal syndromes in mammals. However, no studies have focused on the persistent impacts of short-term AMI exposure on teleosts. Here, following exposure to AMI (2.5 and 40 μg/L) for 7 days (E7), zebrafish were transferred into AMI-free water to recover for 21 days (R21). The behavior, brain neurotransmitters, and brain transcriptional profiles were investigated on E7 and R21. AMI exposure induced persistent hypoactivity (2.5 and 40 μg/L) and abnormal schooling behavior (40 μg/L). AMI also induced long-term impacts on the brain serotonin (5-HT), 5-hydroxyindoleacetic acid, norepinephrine, and acetylcholine levels, several of which showed significant correlations with the locomotor activity or schooling behavior. Transcriptional analysis revealed persistent dysregulation in the pathways involved in the circadian rhythm, glycan biosynthesis and metabolism, and axon guidance in brain samples. Twelve genes were predicted as key driver genes in response to AMI exposure, and their significantly differential expression may direct changes across the related molecular networks. Moreover, upregulated brain 5-HT may serve as the central modulator of the persistent AMI pathogenesis in zebrafish. Considering AMI residues in natural waters may temporarily exceed μg/L, corresponding persistent adverse effects on teleosts should not be ignored.

The present study investigated how principal environmental factors such as temperature and light intensity change the toxicological properties of thiobencarb (TB) herbicide to the green alga, Raphidocelis subcapitata. At first, we investigated the inhibitory effect of TB (0, 15.6, 31.2, 62.4, and 125 μg L-1) on growth of R. subcapitata at five temperatures (10, 15, 20, 25, or 30 °C) for 144 h exposure and calculated 72- and 144-h effective concentration values (EC10, 20, and EC50) for growth rate. All EC values significantly decreased with an increasing temperature. The maximum quantum yield of photosystem II in R. subcapitata exposed to 125 μg L-1 of TB was also significantly inhibited with increased temperature. These physiological effects could explain the lower EC values at high temperatures. Then, single and interactive effects of TB, temperature, and light intensity on growth rate were investigated by three-way of analysis of variance. As a result, single and interactive effects were detected in all explanatory variables. These results suggest that temperature and light intensity change the acute toxicity parameter in R. subcapitata exposed to TB and must be considered in evaluating the risk of TB.

Tetrabromobisphenol S (TBBPS), an emerging brominated flame retardant (BFR) has been widely detected in the environment, and may potentially pose environmental risks. However, data on the occurrence and toxic effects of TBBPS are limited. Circadian rhythms govern multiple behavioral and physiological processes, and their disruption is closely associated with various pathological conditions. Little is known about the potential for TBBPS to perturb circadian rhythm networks or circadian-driven locomotor behavior. In the present study, behavior assays and gene expression analysis based on circadian rhythm pathways were designed to investigate the potential circadian rhythm impairments and subsequent adverse effects caused in 120 h post-fertilization (hpf) zebrafish larvae by TBBPS. The development of embryos was inhibited by TBBPS exposure even at concentrations below the maximal non-lethal concentration (MNLC, 3.47 mg/L). Our results indicated remarkable alterations in the expression of several key circadian rhythm genes due to TBBPS exposure. Compared to control, the expression of per1a, per1b, per3, cry2, and csnk1da was increased, while the expression of clocka, clockb, cry4, cry1ba, arntl1a, and cank1db was decreased. Significant alterations of the circadian rhythm network could be observed in the zebrafish embryos. TBBPS exposure also significantly affected the behavioral responses of larvae. Our findings suggest the circadian rhythm network could be a potential target of TBBPS. Further study is needed to explore whether the transcriptional alterations in circadian rhythm translate into physiological effects.

Z-scheme nitrogen-deficient graphitic carbon nitride/ferric oxide (GCNx/FO) photocatalysts were obtained by a one-step calcination method. The number of nitrogen defects was tuned by the application of KOH, while the ferric oxide phase and magnetism was controlled by the usage of ferrous oxalate. Interconversion between the hematite (α) and maghemite (γ) phases of Fe2O3 was analyzed by X-ray diffraction, and magnetism of the GCNx-0.1/FO products provided via vibrating sample magnetometer test. The introduction of nitrogen defects narrowed the bandgap and extended the spectral range of light absorption while improving the ability to capture and activate N2 effectively. Surface defects were employed as active sites, and the ammonia generation concentration reached approximately 69.7 μmol/L/h by N2 photofixation under visible light. Due to their ease of synthesis and tuning, high photocatalytic activity, convenient and rapid collection for reuse, and excellent stability, GCNx/FO composited photocatalysts possess vast potential for use in clean ammonia production.

We investigated polycyclic aromatic hydrocarbon (PAH) pollution of the intertidal and supratidal zone using the wharf roach (Ligia sp.) as a novel environmental indicator. PAH and alkylated PAH (alkPAH) concentrations in wharf roaches collected from nine sampling sites on the central and northern coast of Japan in November 2011 were analyzed. Total alkPAH concentrations were higher than those of PAHs at all sampling sites. Seven sampling sites had relatively low total concentrations of PAHs and alkPAHs (57.7–130 and 276–770 ng/g-dry, respectively); they included sites in Miyagi Prefecture (St. 5 and 9) that were seriously damaged by the Great East Japan Earthquake and by the following Tsunami on 11 March 2011. Two sampling sites (St. 2: Kominato, Aomori Prefecture; St. 3: Usushiri, Hokkaido) had higher concentrations of total PAHs and alkPAHs (PAHs, 1025 and 591 ng/g-dry, respectively; alkPAHs, 1490 and 1160 ng/g-dry, respectively). Diagnostic ratio calculation showed that St. 2 and 3 had PAHs of pyrolytic origin, possibly from vehicle exhaust gas or industrial fuel combustion, or both. The concentrations of these pollutants in the wharf roach were similar to those in bivalves and therefore likely reflected PAH pollution at the collection sites. The wharf roach may be a suitable candidate indicator of PAH pollution, not only in the intertidal zone inhabited by the currently used bivalve indicators, but also in the supratidal zone.

Although most exposures to chlorpyrifos (CPF) in natural flowing waters are brief and episodic, there have been a few reports of the persistence of abnormal fish behaviors caused by such acute exposure. The present study focused on the behavioral and biochemical responses of Japanese medaka (Oryzias latipes) to acute, sublethal exposure to CPF, as well as the persistence of the effects during a 3-week recovery test in CPF-free water. The medaka became hyperactive and exhibited an elevated anxiety state after a 4-day exposure to 0.024mg/L of CPF, but they recovered from these abnormal behavioral responses within 7days of recovery treatment. In contrast, persistent impacts on some startle responses to a sudden stimulation (induced by a ball drop) were observed in medaka exposed to CPF. The reaction latency did not change immediately after the 4-day exposure, but was significantly prolonged by as much as 21days after the termination of exposure. The post-stimulus swimming distance within 5s significantly decreased on the day immediately after the 4-day exposure, but it significantly increased after 7days of recovery treatment. The activity of acetylcholinesterase (AChE) in the brains of medaka was significantly inhibited on the day immediately after the 4-day exposure, but it returned to 80% and 110% of that in control fish on days 7 and 21 of the recovery period, respectively. However, AChE activities in the eyes of exposed medaka were persistently inhibited and declined to 33%, 71%, and 72% of that in control fish on days 0 (immediately after the 4-day exposure), 7, and 21 of recovery, respectively. Correlation analysis suggested that the changes of AChE activities in the brains of medaka may underlie some of the observed acute behavioral changes, and the changes of AChE activities in the eyes may contribute to the persistence of the abnormalities in the reaction latency of the startle response. Our findings suggest that medaka need a long time to recover from acute, sublethal exposure to CPF, and the persistence of the behavioral abnormalities might affect their fitness in natural habitats.

We investigated the gene structure and predicted amino acid sequence of the antioxidant enzyme 2-Cys peroxiredoxin (2-Cys Prx) in the raphidophyte Chattonella marina, which is a harmful algal bloom (HAB) species. The open reading frame of 2-Cys Prx was 585 bp long and encoded a protein consisting of 195 amino acids. The putative amino acid sequence contained two cysteine residues located at the 49th and 170th amino acid positions from the N-terminal methionine residue. The sequence also possessed 2-Cys Prx characteristic motifs, F (FFYPLDFTFVCPTEI) and EVCP. The position of the 2-Cys Prx gene relative to several others (ycf59 – 2-CysPrx – rpl35 – rpl20) was the same as that found in the chloroplast genome in the raphidophyte Heterosigma akashiwo. The present research investigated the effect of diuron on sinking rate and the physiochemical changes in two marine diatoms, Thalassiosira pseudonana (single-celled species) and Skeletonema marinoi-dohrnii complex (chain-forming species). The results revealed that the sinking rate of both diatoms exposed to diuron at a level of 50% effective concentration for growth (EC50) decreased significantly compared with the control. Photosynthetic performance (Fv/Fm and PIABS) of both diatoms also decreased significantly with diuron exposure. The number of cells per chain in S. marinoi-dohrnii decreased significantly with diuron treatment, but T. pseudonana cell diameter remained stable. Neutral lipid concentration per cell was significantly higher compared with control at 72 h in both diatom species exposed to EC50 level diuron. And water-soluble protein concentration per cell at 72 h was lower than control in the T. pseudonana EC50 group only. These biochemical changes may decrease specific gravity of cells and seems to cause a decreased sinking rate in diatoms. The positive significant correlation between the numbers of cells per chain and sinking rate in S. marinoi-dohrnii indicated that chain length is also an important factor in sinking rate regulation for chain-forming diatoms. Thus, our present study suggested that suppression of photosynthetic performance and the resultant physiochemical changes induced the decreased sinking rate that may inhibit the normal survival strategy (avoidance from the surface layer where strong light either causes photo-inhibition or interrupts resting cell formation). Therefore, the use of antifouling agents should be considered for the sustainable marine environment.

Tributyltin-binding protein type 2 (TBT-bp2), a homolog of α1-acid glycoprotein, may contribute to both accumulation and detoxification of TBT in fish. In this study, we conducted acute TBT exposure tests using both wide-type (WT) and TBT-bp2-/- (KO) strains of medaka and compared their responses in survival time and accumulation of TBT. Deficiency of TBT-bp2 significantly accelerated the time to death of medaka and decreased the LC50 of TBT, indicating that the KO-strain is more sensitive to TBT. No significant difference in the intrinsic TBT concentration in surviving fish was observed between the two strains. However, the intrinsic TBT concentration in dead KO-strain was significantly lower than that in WT-strain. These findings provide direct evidence, supporting the hypothesis that TBT-bp2 plays a critical role in the detoxification of TBT in fish.

This study was performed to elucidate the uptake and bioaccumulation of polystyrene microplastics (PS-MPs) in Japanese medaka (freshwater fish) and Java medaka (marine fish), and to assess its impacts on the survival, reproduction, and gene expression of Japanese medaka. Both species were exposed to 2-μm fluorescent PS-MPs (107 beads/L) for 3 weeks. The bioaccumulation factor of PS-MPs for Java medaka was calculated at about 4 × 102, higher than that for Japanese medaka (about 1 × 102). The exposure had no significant effects on the survival and reproduction of Japanese medaka. The mRNA sequencing analysis showed that the expression of a few genes involved in the cell adhesion, xenobiotic metabolic process, brain development, and other functions in medaka intestines significantly changed after exposure. These results suggest that virgin PS-MPs can potentially accumulate in medaka intestines, but has limited toxicity to Japanese medaka at the concentration up to 107 beads/L.
