Abstract At micro/nano scale, the stiffening effect and flexoelectric effect of strain gradient play important roles in the electromechanical coupling response of piezoelectric micro-components. In this paper, the large deflection bending problem of circular piezoelectric micro-actuator is studied based on the extended linear dielectric theory. In addition to the piezoelectric effect, the flexoelectric effect, the stiffening effect of strain gradient and the high-order electric field effect of polarization gradient are introduced. According to the variational principle, a size-dependent model of circular piezoelectric micro-actuator is established to investigate its electromechanical coupling response. The contributions of piezoelectric effect and flexoelectric effect on large deflection behaviors of piezoelectric micro-actuator are revealed. It is hoped that the research results will be helpful to further understand the electromechanical coupling properties of piezoelectric micro-components and improve the control precision of piezoelectric micro-actuator.

In recent years, the frequent occurrence of eutrophication problems in water bodies has been caused by changes in the climate environment and overexploitation of natural resources by humans. Chlorophyll-a, as a key indicator for water body assessment, plays an important role in eutrophication research and has a profound impact on the global biogeochemical cycle of the climate process. Studies have shown that temperature can directly or indirectly affect the concentration of chlorophyll-a by influencing the growth of algae and water quality indicators in water bodies. Considering the temperature factor in the inversion of chlorophyll-a concentration is a novel research approach. Based on the influence of temperature on chlorophyll-a concentration, we propose the idea of inverting global ocean chlorophyll-a concentration based on temperature zoning. Using monthly average remote sensing reflectance data from VIIRS (Visible and Infrared Imaging Radiometer Suite), combined with the results of temperature zoning, the OC3V(SST) model was constructed to invert the monthly average chlorophyll-a concentration in the global ocean in October 2018. The OC3V(SST) model has been validated by applying it to the remaining 11 months of January, April, July, and October in 2017, 2018, and 2019, as well as the entire 31-day dataset of October 2018. The results indicate that temperature zonation can significantly improve the inversion accuracy of chlorophyll-a and further explore the spatial distribution patterns of global chlorophyll-a concentrations across various temperature ranges based on monthly averages from the global ocean. Additionally, the study investigates the continuity issues of various models and the correlation between temperature and chlorophyll-a.

We aimed to determine the molecular characteristics of carbapenem-resistant Pseudomonas aeruginosa strains 18081308 and 18083286, which were isolated from the urine and the sputum of two Chinese patients, respectively. Additionally, we conducted a comparative analysis between Tn6411 carrying blaIMP-1 in strain 18083286 and transposons from the same family available in GenBank. Bacterial genome sequencing was carried out on strains 18081308 and 18083286 to obtain their whole genome sequence. Average nucleotide identity (ANI) was used for their precise species identification. Serotyping and multilocus sequence typing were performed. Furthermore, the acquired drug resistance genes of these strains were identified. The carbapenem-resistant P. aeruginosa strains isolated in the present study were of sequence type ST865 and serotype O6. They all carried the same resistance genes (aacC2, tmrB, and blaIMP-1). Tn6411, a Tn7-like transposon carrying blaIMP-1, was found in strain 18083286 by single molecule real time (SMRT) sequencing. We also identified the presence of this transposon sequence in other chromosomes of P. aeruginosa and plasmids carried by Acinetobacter spp. in GenBank, indicating the necessity for heightening attention to the potential transferability of this transposon.

(1) Background: Antibiotic resistance in bacteria is an urgent global threat to public health. Migratory birds can acquire antibiotic-resistant and pathogenic bacteria from the environment or through contact with each other and spread them over long distances. The objectives of this study were to explore the relationship between migratory birds and the transmission of drug-resistant pathogenic Escherichia coli. (2) Methods: Faeces and swab samples from migratory birds were collected for isolating E. coli on the Inner Mongolia Plateau of northern China from 2018 to 2023. The resistant phenotypes and spectra of isolates were determined using a BD Phoenix 100 System. Conjugation assays were performed on extended-spectrum β-lactamase (ESBL)-producing strains, and the genomes of multidrug-resistant (MDR) and ESBL-producing isolates were sequenced and analysed. (3) Results: Overall, 179 isolates were antibiotic-resistant, with 49.7% MDR and 14.0% ESBL. Plasmids were successfully transferred from 32% of ESBL-producing strains. Genome sequencing analysis of 91 MDR E. coli strains identified 57 acquired resistance genes of 13 classes, and extraintestinal pathogenic E. coli and avian pathogenic E. coli accounted for 26.4% and 9.9%, respectively. There were 52 serotypes and 54 sequence types (STs), including ST48 (4.4%), ST69 (4.4%), ST131 (2.2%) and ST10 (2.2%). The international high-risk clonal strains ST131 and ST10 primarily carried blaCTX-M-27 and blaTEM-176. (4) Conclusions: There is a high prevalence of multidrug-resistant virulent E. coli in migratory birds on the Inner Mongolian Plateau. This indicates a risk of intercontinental transmission from migratory birds to livestock and humans.

Gut microbes thrive by utilising host energy and, in return, provide valuable benefits, akin to a symbiotic relationship. Here, metagenomic sequencing was performed to characterise and compare the community composition, diversity and antibiotic resistance of the gut microbiota of Relict gull (Larus relictus) and Anatidae species. Alpha diversity analysis revealed that the intestinal microbial richness of L. relictus was significantly lower than that of Anatidae, with distinct differences observed in microbial composition. Notably, the intestines of L. relictus harboured more pathogenic bacteria such as clostridium, which may contribute to the decline in their population and endangered status. A total of 117 strains of Escherichia coli were isolated, with 90.60% exhibiting full susceptibility to 21 antibiotics, while 25.3% exhibited significant biofilm formation. Comprehensive Antibiotic Resistance Database data indicated that glycopeptide resistance genes were the most prevalent type carried by migratory birds, alongside quinolone, tetracycline and lincosamide resistance genes. The abundance of resistance genes carried by migratory birds decreased over time. This metagenomic analysis provides valuable insights into the intestinal microbial composition of these wild bird species, offering important guidance for their conservation efforts, particularly for L. relictus, and contributing to our understanding of pathogen spread and antibiotic-resistant bacteria.

Cadmium (Cd)-induced oxidative stress detrimentally affects hyperaccumulator growth, thereby diminishing the efficacy of phytoremediation technology aimed at Cd pollution abatement. In the domain of plant antioxidant mechanisms, the role of glutathione peroxidase (GPX) in conferring Cd tolerance to tobacco (Nicotiana tabacum) remained unclear. Our investigation employed genome-wide analysis to identify 14 NtGPX genes in tobacco, revealing their organization into seven subgroups characterized by analogous conserved domain patterns. Notably, qPCR analysis highlighted NtGPX8a as markedly responsive to Cd2+ stress. Subsequent exploration through yeast two-hybridization unveiled NtGPX8a’s utilization of thioredoxins AtTrxZ and AtTrxm2 as electron donors, and without interaction with AtTrx5. Introduction of NtGPX8a into Escherichia coli significantly ameliorated Cd-induced adverse effects on bacterial growth. Transgenic tobacco overexpressing NtGPX8a demonstrated significantly augmented activities of GPX, SOD, POD, and CAT under Cd2+ stress compared to the wild type (WT). Conversely, these transgenic plants exhibited markedly reduced levels of MDA, H2O2, and proline. Intriguingly, the expression of NtGPX8a in both E. coli and transgenic tobacco led to increased Cd accumulation, confirming its dual role in enhancing Cd tolerance and accumulation. Consequently, NtGPX8a emerges as a promising candidate gene for engineering transgenic hyperaccumulators endowed with robust tolerance for Cd-contaminated phytoremediation.

BackgroundWohlfahrtiimonas chitiniclastica is an emerging fly-borne zoonotic pathogen, which causes infections in immunocompromised patients and some animals. Herein, we reported a W. chitiniclastica BM-Y from a dead zebra in China.MethodsThe complete genome sequencing of BM-Y showed that this isolate carried one chromosome and one novel type of blaVEB–1-carrying plasmid. Detailed genetic dissection was applied to this plasmid to display the genetic environment of blaVEB–1.ResultsThree novel insertion sequence (IS) elements, namely ISWoch1, ISWoch2, and ISWoch3, were found in this plasmid. aadB, aacA1, and gcuG were located downstream of blaVEB–1, composing a gene cassette array blaVEB–1–aadB–aacA1–gcuG bracketed by an intact ISWoch1 and a truncated one, which was named the blaVEB–1 region. The 5′-RACE experiments revealed that the transcription start site of the blaVEB–1 region was located in the intact ISWoch1 and this IS provided a strong promoter for the blaVEB–1 region.ConclusionThe spread of the blaVEB–1-carrying plasmid might enhance the ability of W. chitiniclastica to survive under drug selection pressure and aggravate the difficulty in treating infections caused by blaVEB–1-carrying W. chitiniclastica. To the best of our knowledge, this is the first report of the genetic characterization of a novel blaVEB–1-carrying plasmid with new ISs from W. chitiniclastica.

Alongside widely accepted geometric information, airborne laser bathymetry (ALB) typically captures the temporal profile sample (waveforms) of both the emitted laser pulses and their echoes. These waveforms provide radiometric properties (backscattering intensity) of sensed targets and assist with precise strip registration, classification of fine ground cover (sediment), and advanced geometric modelling. However, effective intensity data extraction is essential because the intensity provided by the LAS file is mixed with invalid signals, multiple echo intensity, and intermittent intensity variations caused by improper interference of automatic gain control, making it unusable for direct analysis. To address this issue, a flexible index sharing mechanism between waveform, coordinate, and intensity data is constructed for interference intensity tracking. A novel waveform classification-based approach is proposed to efficiently extract ALB intensity by dividing waveform data into six categories using morphological differences and topographic data of waveforms. To ensure accurate analysis, duplicate and invalid waveforms are eliminated, leaving only genuine intensity readings. Additionally, a triple spline fit is employed to restore oversaturated waveform segments that were previously suppressed due to exceeding the device's maximum measurable limit. To address the problem of mixing multiple return intensities, waveforms are decomposed using various decomposition models. This approach ensures that different waveform categories retain their respective varying return intensities. The approach is then tested on an ALB dataset collected using Optech Aquarius around Yuanzhi Island in the South China Sea. The results demonstrate a considerable advancement in data quality when compared to LAS file intensity products with a reduction in maximum deviation of 883 [digital number, DN], standard deviation of 350 [DN], and mean absolute error of 310 [DN].

Postoperative infections following implant‐related spinal surgery are severe and disastrous complications for both orthopaedic surgeons and patients worldwide. They can cause neurological damage, disability, and death. To better understand the mechanism of these destructive complications and intervene in the process, further research is needed. Therefore, there is an urgent need for efficient, accurate, and easily available animal models to study the pathogenesis of spinal infections and develop new and effective anti‐bacterial methods. In this paper, we provide a general review of the commonly used animal models of postoperative implant‐related spinal infections, describe their advantages and disadvantages, and highlight the significance of correctly choosing the model according to the infection aspect under investigation. These models are valuable tools contributing to the better understanding of postoperative spinal infections and will continue to facilitate the invention of novel preventative and treatment strategies for patients with postoperative spinal infections. However, although they are valid and reproducible in some respects, the current animal models present certain limitations. Future ideal spinal infection animal models may assess the bacterial load of the same animal in real‐time in vivo, and better mimic the human anatomy as well as surgical techniques. Strains other than Staphylococcus aureus account for a large proportion of postoperative spinal infections, and thus, the establishment of models to evaluate other types of microbial infections is expected in the future. Furthermore, novel transgenic models established on advancements in genome editing are also likely to be developed in the future.

IntroductionNortheast China has always been an area with severe brucellosis prevalence. This study will identify Brucella in Northeast China and test its resistance to antibiotics, in order to clarify its resistance mechanism. Brucella is a widespread and highly pathogenic bacteria that poses serious threats to public health and animal husbandry.MethodsIn this study, 61 Brucella isolates were identified by abortus-melitensis-ovis-suis polymerase chain reaction (AMOS-PCR) for biotypes and epidemic potential was clarified by multi-locus sequence analysis. Whole-genome sequencing (WGS) was performed and the antibiotic susceptibility of the Brucella strains against 13 antibiotics was detected with the use of E-test strips.ResultsThe results showed that all of the isolates were Brucella melitensis ST8, group CC4 with little genetic variation and obvious geographical characteristics. All 61 Brucella isolates were sensitive to doxycycline, tetracycline, minocycline, levofloxacin, ciprofloxacin, gentamicin, and streptomycin, while 24.6%, 86.9%, 65.6%, 27.9%, 3.3%, and 1.6% were resistant to rifampin, azithromycin, cefepime, cefoperazone/sulbactam, cefotaxime, and meperidine/sulfamethoxazole, respectively. This is the first report of cephalosporin-resistant B. melitensis in China. The WGS results indicated that about 60% of the antibiotic resistance genes were associated with efflux pumps (mainly the resistance nodulation division family).DiscussionBrucellosis is usually treated with antibiotics for several months, which can easily lead to the emergence of antibiotic resistance. To ensure the effectiveness and safety of antibiotics for treatment of brucellosis, continuous surveillance of antibiotic susceptibility is especially important.

Carbapenem-resistant Klebsiella pneumoniae (CRKP) infection is a serious problem in hospitals worldwide. We monitored a tertiary hospital in Changchun, Jilin Province, China, and found that CRKP was the major species among the carbapenem-resistant isolates in sewage. Subsequently, we evaluated the drug susceptibility, resistance genes, virulence genes, outer pore membrane protein-related genes (OmpK35 & OmpK 36), multi-locus sequence typing and replicons, biofilm formation capabilities, and resistance to chlorine-containing disinfectants among KP isolates. Identification of drug sensitivity, multiple resistance profiles were observed including 77 (82.80%) multidrug resistant (MDR), 16 (17.20%) extensive drug resistant (XDR). Some antibiotic resistance genes were detected, the most prevalent carbapenemase gene was blaKPC, and 16 resistance genes were associated with other antibiotics. In addition, 3 (3.23%) CRKP isolates demonstrated loss of OmpK-35 and 2 (2.15%) demonstrated loss of OmpK-36. In the detection of multi-locus sequence typing (MLST), 11 ST11 isolates carried virulence genes. The most common replicon type was IncFII. Biofilm-forming capabilities were demonstrated by 68.8% of the isolates, all of which were resistant to chlorine-containing disinfectants. The results of the study showed that antibiotic-resistant isolates, especially CRKP, could resist disinfectants in hospital wastewater, and improper treatment of hospital wastewater may lead to the spread of drug-resistant bacteria and their genes. Thus, these bacteria must be eliminated before being discharged into the municipal sewage system.

To study antimicrobial resistance and virulence genes of Klebsiella pneumoniae and Raoultella strains isolated from captive giant pandas. Non-duplicate fecal samples were collected from 128 giant pandas during 2017-2019. All isolated microbial strains were tested for antimicrobial drug susceptibility using BD verification panels. Four extended-spectrum β-lactamase resistance genes, nine virulence genes and six capsular serotype genes were detected using PCR. 42 K. pneumoniae and nine Raoultella strains were isolated from different giant pandas. Antibiotic resistance rates were 1.9%-23.5%, except for ampicillin, and 7.8% of the isolates were multidrug-resistant to 7-10 antibiotic classes. This is the first time that a multidrug-resistant R. ornithinolytica strain has been isolated from captive giant pandas. The blaTEM, blaCTX-M, blaSHV and blaDHA genes were detected in four MDR ESBL- K. pneumoniae strains. The rmpA, iutA, ybtS, iroN and iroB genes were positively detected in 11.7% of the isolates. Capsular serotype (K2, K5, K54 and K57) genes were all detected in four K. pneumoniae strains, and one was identified as hypervirulent. This study showed that MDR ESBL- K. pneumoniae, hypervirulent K. pneumoniae, MDR R. ornithinolytica and the colistin-resistant strain may pose risks to captive giant pandas and their keepers, and that the diversity of antibiotic resistance and virulence genes in Klebsiella and Raoultella should be monitored regularly.

High-resolution remote sensing images have the advantage of timeliness, and they can display feature information in more detail. Deep learning embodies its unique characteristics in land cover classification, target recognition, and other fields, which can automatically learn the in-depth feature information of images and make accurate classification decisions. However, when deep learning models extract high-dimensional abstract feature information, they often ignore and lose part of the underlying features essential for classification accuracy. This article proposes a dual-channel fully convolutional network (D-FCN), whose two channels, respectively, take image data and low-level features such as color, texture, and shape as the different input data to combine the underlying features with high-dimensional abstract features. To reduce the complexity of the model, we add a large number of skip connections between the model and make full use of the advantages of weight sharing and local connections to connect spatial context information. We used multifeature information as the model input and compared and analyzed the impact of different features on the land cover classification accuracy, and finally obtained the most suitable combination of multifeature information. In addition, we provide a small-scale land cover classification dataset with labels to verify the applicability and transferability of the D-FCN, and use the optimal combination of multifeature information to conduct comparative experiments on the small-scale dataset. The experimental results show that D-FCN has outstanding applicability and transferability. Compared with other state-of-the-art models, D-FCN has a more challenging performance and greatly reduces model complexity.

Accurate bathymetric data in shallow water is of increasing importance for navigation safety, coastal management, and marine transportation. Satellite-derived bathymetry (SDB) is widely accepted as an effective alternative to conventional acoustic measurements in coastal areas, providing high spatial and temporal resolution combined with extensive repetitive coverage. Many previous empirical SDB approaches are unsuitable for precision bathymetry mapping in various scenarios, due to the assumption of homogeneous bottom over the whole region, as well as the neglect of various interfering factors (e.g., turbidity) causing radiation attenuation. Therefore, this study proposes a bottom-type adaption-based SDB approach (BA-SDB). Under the consideration of multiple factors including suspended particulates and phytoplankton, it uses a particle swarm optimization improved LightGBM algorithm (PSO-LightGBM) to derive depth of each pre-segmented bottom type. Based on multispectral images of high spatial resolution and in situ observations of airborne laser bathymetry and multi-beam echo sounder, the proposed approach is applied in shallow water around Yuanzhi Island, and achieves the highest accuracy with an RMSE value of 0.85 m compared to log-ratio, multi-band, and classical machine learning methods. The results of this study show that the introduction of water-environment parameters improves the performance of the machine learning model for bathymetric mapping.

Abstract Flexoelectricity will dominate the electromechanical coupling of intelligent components in MEMS/NEMS due to its size-dependency. This paper focuses on investigating the flexoelectric responses of intelligent components of the circular plate type, which are commonly used in MEMS/NEMS. Utilizing Hamilton’s principle, the nonlinear flexoelectric circular plate model is presented by combining von Kármán plate theory and flexoelectric theory. The equilibrium equations and all boundary conditions are obtained and then discretized. The nonlinear static bending of the simply supported axisymmetric flexoelectric circular plate is investigated by combining DQM and iteration method. The distributions of dimensionless bending deflection and electric potential are analyzed under different loads. Moreover, the nonlinear free vibration behaviors are also investigated by combining the Galerkin method and Lindstedt–Poincaré Method. The flexoelectric effect and stiffening effect of strain gradient are revealed. This paper will be helpful to promote the application of flexoelectric intelligent components of the circular plate type, which are encountered commonly in engineering.

ObjectiveTo precisely determine the species of a carbapenem-resistant Pseudomonas strain 1809276 isolated from the urine of a Chinese patient and analyze its integrative and conjugative element (ICE) 1276 formation mechanism.MethodsSingle-molecule real-time (SMRT) sequencing was carried out on strain 18091276 to obtain the complete chromosome and plasmid (pCN1276) sequences, and average nucleotide identity (ANI) was used for precise species identification. The ICEs in GenBank with the same integrase structure as ICE 1276 were aligned. At the same time, the transfer ability of blaIMP−1 and the antibiotic sensitivity of Pseudomonas juntendi 18091276 were tested.ResultsThis bacterium was P. juntendi, and its drug resistance mechanism is the capture of the accA4' gene cassette by the Tn402-like type 1 integron (IntI1-blaIMP−1) to form In1886 before its capture by the ΔTn4662a-carrying ICE 1276. The acquisition of blaIMP−1 confers carbapenem resistance to P. juntendi 18091276.ConclusionThe formation of blaIMP−1-carrying ICE 1276, its further integration into the chromosomes, and transposition and recombination of other elements promote bacterial gene accumulation and transmission.

Mineral nutrition plays an important role in crop growth, yield and quality. MiR156 is a regulatory hub for growth and development. To date, the understanding of miR156-mediated mineral homeostasis is limited. In this study, we overexpressed Nta-miR156a in the tobacco cultivar TN90 and analyzed the effects of miR156 on mineral element homeostasis in tobacco by comparative transcriptome analysis. The results showed that the overexpression of miR156a caused significant morphological changes in transgenic tobacco. Chlorophyll and three anti-resistance markers, proline, total phenolics, and total flavonoids, were altered due to increased miR156 expression levels. Interestingly, the distribution of Cu, Mn, Zn, and Fe in different tissues of transgenic tobacco was disordered compared with that of the wild type. Comparative transcriptome analysis showed that the overexpression of miR156 resulted in 2656 significantly differentially expressed genes. The expression levels of several metal-transport-related genes, such as NtABC, NtZIP, NtHMA, and NtCAX, were significantly increased or decreased in transgenic tobacco. These results suggest that miR156 plays an essential role in regulating mineral homeostasis. Our study provides a new perspective for the further study of mineral nutrient homeostasis in plants.

Superoxide dismutases (SODs) play an important role in protecting plants against ROS toxicity induced by biotic and abiotic stress. Recent studies have shown that the SOD gene family is involved in plant growth and development; however, knowledge of the SOD gene family in tobacco is still limited. In the present study, the SOD gene family was systematically characterized in the tobacco genome. Based on the conserved motif and phylogenetic tree, 15 NtSOD genes were identified and classified into three subgroups, including 5 NtCSDs, 7 NtFSDs and 3 NtMSDs. The predicted results of the transport peptide or signal peptide were consistent with their subcellular localization. Most NtSOD genes showed relatively well-maintained exon-intron and motif structures in the same subgroup. An analysis of cis-acting elements in SOD gene promoters showed that NtSOD expression was regulated by plant hormones, defense and stress responses, and light. In addition, multiple transcription factors and miRNAs are predicted to be involved in the regulation of NtSOD gene expression. The qPCR results indicated specific spatial and temporal expression patterns of the NtSOD gene family in different tissues and developmental stages, and this gene family played an important role in protecting against heavy metal stress. The results of functional complementation tests in the yeast mutant suggested that NtCSD1a, NtFSD1e and NtMSD1b scavenge ROS produced by heavy metal stress. This study represents the first genome-wide analysis of the NtSOD gene family, which lays a foundation for a better understanding of the function of the NtSOD gene family and improving the tolerance of plants to heavy metal toxicity.

This study aimed to evaluate antimicrobial resistance, virulence, and the genetic diversity of Aeromonas isolated from migratory birds from Guangxi Province, Guangdong Province, Ningxia Hui Autonomous Region, Jiangxi Province, and Inner Mongolia in China. A total of 810 samples were collected, including fresh feces, cloacal swabs, and throat swabs. The collected samples were processed and subjected to bacteriological examination. The resistance to 21 antibiotics was evaluated. A phylogenetic tree was constructed using concatenated gltA-groL-gyrB-metG-PPSA-recA sequences. Eight putative virulence factors were identified by PCR and sequencing, and a biofilm formation assay was performed using a modified microtiter plate method. In total, 176 Aeromonas isolates were isolated including A. sobria, A. hydrophila, A. veronii, and A. caviae. All isolates showed variable resistance against all 16 tested antibiotic discs, and only one antibiotic had no reference standard. Six kinds of virulence gene markers were discovered, and the detection rates were 46.0% (hlyA), 76.1% (aerA), 52.3% (alt), 4.5% (ast), 54.0% (fla), and 64.2% (lip). These strains were able to form biofilms with distinct magnitudes; 102 were weakly adherent, 14 were moderately adherent, 60 were non-adherent, and none were strongly adherent. Our results suggest that migratory birds carry highly virulent and multidrug-resistant Aeromonas and spread them around the world through migration, which is a potential threat to public health.

Because of its light weight, high strength, designable, composite materials are more and more widely used in the field of aerospace, especially in the field of general aviation. In this paper, the structural design and analysis of the composite wing (include wing beam, rib, skin, leading edge and trailing edge) of a four-seater electric aircraft were carried out. The finite element numerical analysis model of the composite wing was established, and the deformation and strain distribution of the composite wing under the limited load required by airworthiness regulations were calculated. Finally, the static test of the whole composite wing was carried out to verify the accuracy of the finite element results. The finite element analysis and testing results showed that the structural design of the composite wing of the electric aircraft meets the structural strength requirements specified in the airworthiness standard (part 23 of China civil aviation regulations: air-worthiness requirements for normal, practical, special effects and commuting aircraft: CCAR 23-R3)

(1) Background: there has been a growing concern about pet-spread bacterial zoonosis in recent years. This study aimed to investigate the trend in drug-resistance of canine Escherichia coli isolates in northeast China between 2012–2021 and the differences in drug-resistance of E. coli of different origins in 2021. (2) Methods: E. coli were isolated from feces or anal swab samples from dogs and cats, and their antibiotic susceptibility profiles and phylogenetic grouping were identified. PCR was applied on the extended spectrum β-lactamase (ESBL) E. coli for antibiotic resistance genes. (3) Results: five hundred and fifty-four E. coli isolates were detected in 869 samples (63.75%). The multidrug resistance (MDR) rates of E. coli in pet dogs showed a decreasing trend, but working dogs showed the opposite trend. Resistance genes blaCTX-M and blaCTX-M+TEM were dominant among the ESBL producers (n = 219). The consistency between the resistance phenotypes and genes was high except for fluoroquinolone-resistant ESBL E. coli. All ESBL E. coli-carrying blaNDM were isolated from working dogs, and one of the strains carried mcr-1 and blaNDM-4. Phylogroup B2 was the dominant group in pet cats, and more than half of the isolates from companion cats were ESBL E. coli. (4) Conclusions: the measures taken to reduce resistance in China were beginning to bear fruit. Companion cats may be more susceptible to colonization by ESBL E. coli. The problem of resistant bacteria in working dogs and pet cats warrants concern.

Tree species classification is crucial for forest resource investigation and management. Remote sensing images can provide monitoring information on the spatial distribution of tree species and multi-feature fusion can improve the classification accuracy of tree species. However, different features will play their own unique role. Therefore, considering various related factors about the growth of tree species such as spectrum information, texture structure, vegetation phenology, and topography environment, we fused multi-feature and multi-temporal Sentinel-2 data, which combines spectral features with three other types of features. We combined different feature-combinations with the random forest method to classify Changbai Mountain tree species. Results indicate that topographic features participate in tree species classification with higher accuracy and more efficiency than phenological features and texture features, and the elevation factor possesses the highest importance through the Mean Decrease in Gini (MDG) method. Finally, we estimated the area of the target tree species and analyzed the spatial distribution characteristics by overlay analysis of the Classification 3 result and topographic features (elevation, slope, and aspect). Our findings emphasize that topographic factors have a great influence on the distribution of forest resources and provide the basis for forest resource investigation.

In 2007, China discovered a hydrothermal anomaly in the Longqi hydrothermal area of the Southwest Indian Ridge. It was the first seabed hydrothermal area discovered in the ultraslow spreading ocean ridge in the world. Understanding the types of seabed sediments in this area is critical for studying the typical topography and geological characteristics of deep-sea seabed hydrothermal areas. The traditional classification of deep-seabed sediments adopts box sampling or gravity column sampling and identifies the types of seabed sediments through laboratory analysis. However, this classification method has some shortcomings, such as the presence of discrete sampling data points and the failure of full-coverage detection. The geological sampling in deep-sea areas is particularly inefficient. Hence, in this study, the EM122 multibeam sonar data collected in the Longqi hydrothermal area, Southwest Indian Ridge, in April 2019 are used to analyze multibeam backscatter intensity. Considering various errors in the complex deep-sea environment, obtaining backscatter intensity data can truly reflect seabed sediment types. Through unsupervised and supervised classification, the seabed sediment classification in the Longqi hydrothermal area was studied. The results showed that the accuracy of supervised classification is higher than that of unsupervised classification. Thus, unsupervised classification is primarily used for roughly classifying sediment types without on-site geological sampling. Combining the genetic algorithm (GA) and the support vector machine (SVM) neural network, deep-sea sediment types, such as deep-sea clay and calcareous ooze, can be identified rapidly and efficiently. Based on comparative analysis results, the classification accuracy of the GA-SVM neural network is higher than that of the SVM neural network, and it can be effectively applied to the high-precision classification and recognition of deep-sea sediments. In this paper, we demonstrate the fine-scale morphology and surface sediment structure characteristics of the deep-sea seafloor by finely processing high-precision deep-sea multibeam backscatter intensity data. This research can provide accurate seabed topography and sediment data for the exploration of deep-sea hydrothermal resources and the assessment of benthic habitats in deep-sea hydrothermal areas.

Migratory birds are recently recognized as Vibrio disease vectors, but may be widespread transporters of Vibrio strains. We isolated Vibrio cholerae (V. cholerae) and Vibrio metschnikovii (V. metschnikovii) strains from migratory bird epidemic samples from 2017 to 2018 and isolated V. metschnikovii from migratory bird feces in 2019 from bird samples taken from the Inner Mongolia autonomous region of China. To investigate the evolution of these two Vibrio species, we sequenced the genomes of 40 V. cholerae strains and 34 V. metschnikovii strains isolated from the bird samples and compared these genomes with reference strain genomes. The pan-genome of all V. cholerae and V. metschnikovii genomes was large, with strains exhibiting considerable individual differences. A total of 2,130 and 1,352 core genes were identified in the V. cholerae and V. metschnikovii genomes, respectively, while dispensable genes accounted for 16,180 and 9,178 of all genes for the two strains, respectively. All V. cholerae strains isolated from the migratory birds that encoded T6SS and hlyA were non-O1/O139 serotypes without the ability to produce CTX. These strains also lacked the ability to produce the TCP fimbriae nor the extracellular matrix protein RbmA and could not metabolize trimetlylamine oxide (TMAO). Thus, these characteristics render them unlikely to be pandemic-inducing strains. However, a V. metschnikovii isolate encoding the complete T6SS system was isolated for the first time. These data provide new molecular insights into the diversity of V. cholerae and V. metschnikovii isolates recovered from migratory birds.

Abstract Zika virus (ZIKV) has elicited global concern due to its unique biological features, unusual transmission routes, and unexpected clinical outcomes. Although ZIKV transmission through anal intercourse has been reported in humans, it remains unclear if ZIKV is detectable in the stool, if it can infect the host through the anal canal mucosa, and what the pathogenesis of such a route of infection might be in the mouse model. Herein, we demonstrate that ZIKV RNA can be recovered from stools in multiple mouse models, as well as from the stool of a ZIKV patient. Remarkably, intra-anal (i.a.) inoculation with ZIKV leads to efficient infection in both Ifnar1 −/− and immunocompetent mice, characterized by extensive viral replication in the blood and multiple organs, including the brain, small intestine, testes, and rectum, as well as robust humoral and innate immune responses. Moreover, i.a. inoculation of ZIKV in pregnant mice resulted in transplacental infection and delayed fetal development. Overall, our results identify the anorectal mucosa as a potential site of ZIKV infection in mice, reveal the associated pathogenesis of i.a. infection, and highlight the complexity of ZIKV transmission through anal intercourse.

Migratory birds play an important role in the spread of multidrug-resistant (MDR) bacteria. To investigate the prevalence of MDR Escherichia coli in migratory birds in China and potential relationships with the environment, a total of 1387 samples (fecal samples, cloacal swabs, or throat swabs) were collected from migratory birds from three different river basins in China. The collected samples were processed and subjected to bacteriological examinations. Antimicrobial susceptibility testing of the recovered isolates was performed using the E-test for the detection of minimum inhibitory concentrations (MICs). Some antibiotic resistance genes were detected and the PCR products were confirmed by sequencing. In total, 478 (34.7%) E. coli isolates were recovered. The results showed that the drug-resistant E. coli isolates were highly resistant to β-lactams (43.7%) and tetracycline (22.6%), and 73 (15.3%) were MDR, including eight that were extended spectrum β-lactamase-positive. The retrieved strains harbored the blaCTX-M, blaTEM-1, tet(A), tet(B), tet(M), sul1, sul2, sul3, cmlA, floR, and intI1 genes with a prevalence of 5.9%, 36.4%, 80.5%, 11.9%, 6.8%, 6.8%, 47.5%, 12.7%, 50.8%, 37.3%, and 61.0%, respectively. The drug resistance rate of the isolates from southern China was higher than those from northern China. The E. coli samples collected for migratory birds in the Pearl River Basin had the highest proportion (46.7%) MDR isolates. Furthermore, MDR bacteria carried by migratory birds were closely related to the antibiotic content in the basin, which confirms that MDR bacteria carried by migratory birds are likely acquired from the environment. This study also confirmed that migratory birds are potential transmitters of MDR bacteria, demonstrating the need to reduce the use and emission of antibiotics and further in-depth studies on the mechanisms underlying drug resistance of bacteria.

Extracting road information from high-resolution remote sensing images (HRI) can provide crucial geographic information for many applications. With the improvement of remote sensing image resolution, the image data contain more abundant feature information. However, this phenomenon also enhances the spatial heterogeneity between different types of roads, making it difficult to accurately discern the road and non-road regions using only spectral characteristics. To remedy the above issues, a novel residual attention and local context-aware network (RALC-Net) is proposed for extracting a complete and continuous road network from HRI. RALC-Net utilizes a dual-encoder structure to improve the feature extraction capability of the network, whose two different branches take different feature information as input data. Specifically, we construct the residual attention module using the residual connection that can integrate spatial context information and the attention mechanism, highlighting local semantics to extract local feature information of roads. The residual attention module combines the characteristics of both the residual connection and the attention mechanism to retain complete road edge information, highlight essential semantics, and enhance the generalization capability of the network model. In addition, the multi-scale dilated convolution module is used to extract multi-scale spatial receptive fields to improve the model’s performance further. We perform experiments to verify the performance of each component of RALC-Net through the ablation study. By combining low-level features with high-level semantics, we extract road information and make comparisons with other state-of-the-art models. The experimental results show that the proposed RALC-Net has excellent feature representation ability and robust generalizability, and can extract complete road information from a complex environment.

Given the recent Zika virus (ZIKV) epidemic, development of an effective vaccine is of high importance. Here, the authors use a licensed live-attenuated flavivirus vaccine backbone to develop a ZIKV vaccine and determine immunogenicity, safety and protection profiles in different animal models.

Zika virus (ZIKV) has become a global public health emergency due to its rapidly expanding range and its ability to cause severe congenital defects such as microcephaly. However, there are no FDA-approved therapies or vaccines against ZIKV infection. Through our screening of viral entry inhibitors, we found that chloroquine (CQ), a commonly used antimalarial and a FDA-approved drug that has also been repurposed against other pathogens, could significantly inhibit ZIKV infection in vitro, by blocking virus internalization. We also demonstrated that CQ attenuates ZIKV-associated morbidity and mortality in mice. Finally, we proved that CQ protects fetal mice from microcephaly caused by ZIKV infection. Our methodology of focusing on previously identified antivirals in screens for effectiveness against ZIKV proved to be a rapid and efficient means of discovering new ZIKV therapeutics. Selecting drugs that were previously FDA-approved, such as CQ, also improves the likelihood that they may more quickly reach stages of clinical testing and use by the public.

Cold recycled mixture (CRM) has been widely used around the world mainly because of its good ability to resist reflection cracking. In this study, mixed-mode cracking tests were carried out by the designed rotary test device to evaluate the cracking resistance of CRM. Through the finite element method, the heterogeneous model of CRM based on its meso-structure was established. The cracking process of CRM was simulated using the extended finite element method, and the influence of different notch lengths on its anti-cracking performance was studied. The results show that the mixed-mode fracture test method can effectively evaluate the cracking resistance of CRM by the proposed fracture parameters. The virtual tests under three of five kinds of mixed-cracking modes have good simulation to capture the cracking behavior of CRM. The effect of notch length on the initial crack angle and the crack propagation process of the CRM is mainly related to the distribution characteristics of its meso-structure. With the increase of the proportion of Mode II cracking, the crack development path gradually deviates, and the failure elements gradually increase. At any mixed-mode level, there is an obvious linear relationship between the peak load, fracture energy, and the notch length.

We determined the genotypes of seven Bacillus anthracis strains that were recovered from nine anthrax outbreaks in North-East China from 2010 to 2014, and two approved vaccine strains that are currently in use in China. The causes of these cases were partly due to local farmers being unaware of the presence of anthrax, and butchers with open wounds having direct contact with anthrax-contaminated meat products. The genotype of five of the seven recovered strains was A.Br.001/002 sub-lineage, which was concordant with previously published research. The remaining two cases belongs to the A.Br.Ames sub-lineage. Both of these strains displayed an identical SNR pattern, which was the first time that this genotype was identified in North-East China. Strengthening education in remote villages of rural China is an important activity aimed at fostering attempts to prevent and control anthrax. The genotype of the vaccine strain Anthrax Spore Vaccine No.II was A.Br.008/009 and A.Br.001/002 for the vaccine strain Anthrax Spore Vaccine Non-capsulated. Further studies of their characteristics are clearly warranted.

The presence of multidrug-resistant bacterial pathogens in the environment poses a serious threat to public health. The opportunistic Acinetobacter spp. are among the most prevalent causes of nosocomial infections. Here, we performed complete genome sequencing of the Acinetobacter calcoaceticus strain XM1570, which was originally cultivated from the sputum of a patient diagnosed with pneumonia in Xiamen in 2010. We identified carbapenem resistance associated gene bla(NDM-1) located on a 47.3-kb plasmid. Three methods--natural reproduction, sodium dodecyl sulfate treatment and nalidixic acid treatment--were used to eliminate the bla(NDM-1)-encoding plasmid, which achieved elimination rates of 3.32% (10/301), 83.78% (278/332), and 84.17% (298/354), respectively. Plasmid elimination dramatically increased antibiotic sensitivity, reducing the minimum bacteriostatic concentration of meropenem from 256 µg/ml in the clinical strain to 0.125 µg/ml in the plasmid-eliminated strain. Conjugation transfer assays showed that the bla(NDM-1)-containing plasmid could be transferred into Escherichia coli DH5α:pBR322 in vitro as well as in vivo in mice. The bla(NDM-1) genetic environment was in accordance with that of other bla(NDM-1) genes identified from India, Japan, and Hong-Kong. The multilocus sequence type of the isolate was identified as ST-70. Two novel genes encoding intrinsic OXA and ADC were identified and named as OXA-417 and ADC-72. The finding of bla(NDM-1) in species like A. calcoaceticus demonstrates the wide spread of this gene in gram-negative bacteria which is possible by conjugative plasmid transfer. The results of this study may help in the development of a treatment strategy for controlling NDM-1 bacterial infection and transmission.

This paper describes the development of a compact and lightweight joint torque sensor for space manipulators. Space manipulators with torque sensors can not only apply force control approach to more precise and more dexterous space missions, but also monitor the occurrence of unpredicted events as accidental impacts with objects they have to manipulate. At present, most of the compact torque sensors, designed without consideration of space environment, cannot be directly used in space applications. In this paper, we propose a compact and lightweight design for the joint torque sensor based on strain gauge, for the reason of good physical, chemical, and mechanical stabilities under high temperature environment. In addition, to reduce the interferences by axial forces generated by assembling condition and joint motors, the structural optimization for the sensing element is carried out. The proposed design is simulated by FEM software ANSYS, and it shows successful measurements of the torque with a load capacity of 10 Nm, which is sufficient for the torque generated in robot joints. The effect of axial loading is also analyzed by ANSYS. The designed sensor is manufactured by duralumin alloy. The calibration for the sensor is carried out, and several experiments are conducted to ensure its feasibility with the space manipulator.

British neo-Marxism is a novel theory that emerged and developed in the UK during the period from 1950s to 1980s. It encompasses issues of history, culture, politics, society, technology, and outer space as it continues to broaden alternate critical research approaches. It carries on the intellectual tradition of British Marxism and is guided by the guiding role of Marx's thought. British neo-Marxism has contributed to the formation of multiple neo-Marxist thinking paradigms with unique British characteristics, and its theatrical implications concern the new historicism focused on cultural criticism, the neo-rationalist thinking of changing social orders, and the neo-humanistic thinking aiming to fulfill the reform of social institutions. Its innovation in the systemization of thinking paradigms is doubtless an effect of the paradigm shift in the way of thinking of humankind to change the social order. Our study of British neo-Marxism is of important theoretical significance and practical value for both the enhancement of research on Marxist philosophy, and the innovation in the Marxist thinking paradigm.
