Recent investigations have revealed that puerarin (PU) alleviates cadmium (Cd)-caused hepatic damage via inhibiting oxidative stress. Mitochondria are dynamic organelles and play a critical part in regulating the occurrence of oxidative stress, but the role of mitochondria in the protection of PU against hepatocellular damage caused by Cd exposure remains unknown. Thus, this study was aimed to clarify this issue using mouse hepatocyte AML-12 cell line. Transmission electron microscopy analysis firstly showed that PU prevents Cd-induced mitochondrial ultrastructure damage. Mitochondrial network image analysis by confocal microscopy revealed that PU exerts the protection against Cd-induced cytotoxicity via restoring mitochondrial network fragmentation. Also, mitochondrial dynamic protein expression profiles showed that enhanced fission protein levels and inhibited fusion protein levels in Cd-treated cells were significantly reversed by PU, suggesting the protective effect of PU against Cd-induced mitochondrial fission. Moreover, changes of intracellular ATP level and protein levels of key regulators involving in mitochondrial biogenesis indicated that Sirtuin-1(Sirt1) pathway may be involved in the protection of Cd-impaired mitochondrial function by PU. Next, Sirt1 protein levels in treated cells were effectively regulated by genetic knockdown or chemical agonist SRT1720. Accordingly, alleviation of Cd-induced mitochondrial fission assays and cell viability by PU was markedly regulated by SRT1720 or Sirt1 knockdown, suggesting the indispensable role of Sirt1 in this process. Collectively, these findings highlight that PU prevents Cd-induced mitochondrial fission to alleviate cytotoxicity via Sirt1-dependent pathway, which provide novel evidences to fully understand the hepatoprotective action of PU against heavy metal toxicity.

Threonine aspartase 1 (TASP1) was reported to function in the development of cancer. However, the regulatory mechanism of TASP1 in gastric cancer (GC) remains unclear. In this study, we determined the expression of TASP1 in tissues of GC patients, GC cells by qRT-PCR, and western blot and assessed the relationship between TASP1 and GC cell proliferation and migration via CCK-8 and transwell assay. It was found that the expression of TASP1 in GC tissues or GC cell lines was significantly higher than that in normal adjacent tissues or normal cells. The proliferation and migration of GC cells were inhibited upon TASP1 knockdown. Mechanism investigation revealed that TASP1 promoted GC cell proliferation and migration through upregulating the p-AKT/AKT expression. TASP1 induced GC cell migration via the epithelial -mesenchymal transition (EMT) pathway. In conclusion, TASP1 promotes GC progression through the EMT and AKT/p-AKT pathway, and it may serve as a new potential biomarker and therapeutic target for GC.

Cadmium (Cd) is a common heavy metal contamination that is highly toxic to liver. Puerarin (PU), a potent free radical scavenger, has been shown to exert cytoprotective effect in numerous pathological processes. However, whether PU affords protection against Cd-induced hepatotoxicity remains unclear to be known. Here, we aimed to investigate the protective effect of PU on Cd-induced hepatotoxicity in an immortalized mouse hepatocyte line, AML-12. First, Cd-induced cytotoxicity in AML-12 cells was obviously ameliorated by PU treatment. Also, Cd-induced apoptotic cell death was markedly alleviated by PU treatment, evidenced by two methods. Simultaneously, Cd-elevated malondialdehyde and reactive oxygen species levels were significantly reduced by PU administration, demonstrating the antioxidant effect of PU against Cd exposure. Moreover, Cd-induced blockage of autophagic flux in AML-12 cells was obviously restored by PU treatment, evidenced by immunoblot analysis of autophagy marker proteins and tandem fluorescent-tagged LC3 method. Resultantly, Cd-induced autophagosome accumulation was significantly alleviated by PU treatment. In conclusion, these observations demonstrate that PU treatment alleviates Cd-induced hepatic cell damage by inhibiting apoptosis and restoring autophagy activity, which is intimately related with its antioxidant activity.

Two new termite-pathogenic species, Ophiocordyceps globiperitheciata and O. longistipes, are described from Yunnan Province, China. Six-locus (ITS, nrSSU, nrLSU, tef-1α, rpb1 and rpb2) phylogenetic analyses in combination with morphological observations were employed to characterize these two species. Phylogenetically, O. globiperitheciata is most closely related to Hirsutella cryptosclerotium and O. communis, whereas O. longistipes shares a sister relationship with O. fusiformis. However, O. globiperitheciata differs from H. cryptosclerotium by parasitizing Blattodea and producing clavate, unbifurcated stromata. Ophiocordyceps globiperitheciata is distinguished from O. communis by multiple stromata, shorter asci and ascospores. Ophiocordyceps longistipes differs from O. fusiformis in producing larger stromata, perithecia, asci and ascospores, as well as smaller citriform or oval conidia. Morphological descriptions of the two new species and a dichotomous key to the 19 termite-pathogenic Ophiocordyceps species are presented.

AIM: To analyze the efficacy of multimedia visual physiological stimulation combined with stereoscopic system training in the treatment of ametropic amblyopia in children.METHODS: A total of 102 patients(102 eyes)with ametropic amblyopia admitted to our hospital from June 2021 to June 2022 were randomly divided into two groups. The control group consisted of 51 patients(51 eyes)who received multimedia visual physiological stimulation on the basis of conventional treatment, while the observation group consisted of 51 patients(51 eyes)who received stereoscopic training on the basis of the control group. The binocular fusion function, binocular stereopsis and binocular disparity of the two groups of children were evaluated at 3mo after treatment, and the clinical efficacy was assessed based on the improvement of vision.RESULTS: After 3mo of treatment, the normal rates of binocular fusion function, binocular stereopsis, and binocular disparity in the observation group were significantly higher than those in the control group(all P

Abstract Objective To examine the psychometric properties of the Chinese version of the Clinically Useful Depression Outcome Scale (CUDOS) in the Chinese patients with major depressive disorder (MDD) using Rasch analysis. Methods The sample consisted of 283 patients with MDD (69% females). The Rasch model was applied to examine the overall fit of the Chinese version of CUDOS and the fit of the 18 items. Dimensionality, item-model fit, differential item functioning (DIF), reliability, ordering of response category and targeting were tested to examine the psychometric properties of the Chinese version of CUDOS. Results Rasch analysis demonstrated the unidimensionality of the Chinese version of CUDOS. Of the 18 items, three items (item 4, item 5, item 6) showed misfit in the model. After merging item 4 into item 3 and item 6 into item 5, the overall model fit improved. The person separation index (PSI) was 3.0 and the person reliability coefficient was 0.90. No evidence of significant DIF was found when associated with gender and age. No disordered category and threshold of the rating response were observed, which meant the response category setting was reasonable. The mean ability of person was − 0.53. Conclusion The results suggested that the Chinese version of CUDOS has acceptable psychometric properties. In order to improve the quality and applicability of the Chinese version of CUDOS, the merging of item 4 into item 3 and item 6 into item 5 are suggested.

The coarse-grained heat-affected zone specimens of X80 pipeline steel were produced by welding thermal simulation under different heat inputs of 10, 30, and 55 kJ/cm to study the effects of heat input on microstructure evolution and corrosion characterization. The corrosion resistance of coarse-grained heat-affected zones was poorer than that of base metal due to less homogenous in the former. For 10 kJ/cm coarse-grained heat-affected zone, the corrosion resistance was poorer than the others due to the more adsorption hydrogen around the needle-like martensite/austenite constituents and greater galvanic driving force between the needle-like martensite/austenite constituents and ferrite. In carbonate/bicarbonate solution, better corrosion resistance for coarse-grained heat-affected zones was obtained when the heat input is 30 kJ/cm, which can be attributed to the severe coarse martensite/austenite constituents for 55 kJ/cm coarse-grained heat-affected zone. In the H2S environment, the better corrosion resistance for coarse-grained heat-affected zone was obtained when the heat input is 55 kJ/cm, which can be attributed to the protective effect of corrosion products. In addition, the high content of M/A constituents for 30 kJ/cm CGHAZ was good for hydrogen adsorption, which was adverse to the corrosion resistance in acid environments.

Resistance traits of plants can be activated both at the damaged site and undamaged parts. Systemic resistance induced by local exogenous abscisic acid (ABA) application alleviated negative effect of low water availability on growth performance of clonal plant. However, timing of systemic resistance was poorly understood. Timing of systemic resistance refers to its activation and decay time within clonal network. Clonal fragment of Centella asiatica with four successive ramets (including first-oldest, second-older, third-old and fourth-young ramets) subjected to low water availability (20% soil moisture content) was used to explore effects of local exogenous ABA application on the timing of resistance activation and decay. Systemic resistance activated by local exogenous ABA application after 4 days remained at least 28 days. Compared with control, biomass accumulation of whole clonal fragment, root biomass and ratio of belowground to aboveground biomass significantly increased by local exogenous ABA application after 28 days. It is suggested that rapid activation and delay of resistance response induced by local exogenous ABA application within clonal network may improve fitness of clonal plant subjected to abiotic stress.

Abstract Background Despite widespread acknowledgment of the impact of stressful life events on suicide risk, the understanding of the psychological mechanisms underlying the relationship between stressful life events and suicide risk in major depressive disorder (MDD) remain unclear. This study aim to examine whether the distress tolerance mediates the relationship between the stressful life events and suicide risk in patients with MDD. Methods A cross-sectional study was carried out among 125 Chinese patients with MDD, mean age was 27.05 (SD=0.68) and 68.8% were females. The 17-item Hamilton Depression Rating scale (HAMD-17), the validated Chinese version of the Mini International Neuropsychiatric Interview (MINI) suicide module, Life Events Scale (LES) and Distress Tolerance Scale (DTS) were utilized to evaluate depressive symptoms, stressful life events, levels of distress tolerance, and suicide risk, respectively. Mediation analyses was used to test the mediation effect of distress tolerance on the relationship between stressful life events and suicide risk. Context Shen-Shi-Jiang-Zhuo formula (SSJZF) exhibits a definite curative effect in the clinical treatment of non-alcoholic fatty liver disease (NAFLD).Objective To explore the therapeutic effect and mechanism of SSJZF on NAFLD.Materials and methods Sprague Dawley rats were randomly divided into control, NAFLD, positive drug (12 mg/kg/day), SSJZF high-dose (200 mg/kg/day), SSJZF middle-dose (100 mg/kg/day), and SSJZF low-dose (50 mg/kg/day) groups. After daily intragastric administration of NAFLD rats for 8 weeks, lipid metabolism and hepatic fibrosis were evaluated by biochemical indices and histopathology. Then we uncovered the main active compounds and mechanism of SSJZF against NAFLD by integrating RNA-sequencing and network pharmacology, and PI3K/AKT pathway activity was verified by western blot.Results High dose SSJZF had the best inhibitory effect on hepatic lipid accumulation and fibrosis in rats with NAFLD, which significantly down-regulated total triglycerides (58%), cholesterol (62%), aspartate aminotransferase (57%), alanine aminotransferase (41%) andγ-glutamyl transpeptidase (36%), as well as the expression of ACC (5.3-fold), FAS (12.1-fold), SREBP1C (2.3-fold), and CD36 (4.4-fold), and significantly reduced collagen deposition (67%). Then we identified 23 compounds of SSJZF that acted on 25 key therapeutic targets of NAFLD by integrating RNA-sequencing and network pharmacology. Finally, we also confirmed that high dose SSJZF increased p-PI3K/PI3K (1.6-fold) and p-AKT/AKT (1.6-fold) in NAFLD rats.Discussion and conclusion We found for first time that SSJZF improved NAFLD in rats by activating the PI3K/Akt pathway. These findings provide scientific support for SSJZF in the clinical treatment of NAFLD and contribute to the development of new NAFLD drugs.

Abstract Background Due to the outbreak and rapid spread of coronavirus disease 2019 (COVID-19), more than 160 million patients have become convalescents worldwide to date. Significant alterations have occurred in the gut and oral microbiome and metabonomics of patients with COVID-19. However, it is unknown whether their characteristics return to normal after the 1-year recovery. Methods We recruited 35 confirmed patients to provide specimens at discharge and one year later, as well as 160 healthy controls. A total of 497 samples were prospectively collected, including 219 tongue-coating, 129 stool and 149 plasma samples. Tongue-coating and stool samples were subjected to 16S rRNA sequencing, and plasma samples were subjected to untargeted metabolomics testing. Results The oral and gut microbiome and metabolomics characteristics of the 1-year convalescents were restored to a large extent but did not completely return to normal. In the recovery process, the microbial diversity gradually increased. Butyric acid-producing microbes and Bifidobacterium gradually increased, whereas lipopolysaccharide-producing microbes gradually decreased. In addition, sphingosine-1-phosphate, which is closely related to the inflammatory factor storm of COVID-19, increased significantly during the recovery process. Moreover, the predictive models established based on the microbiome and metabolites of patients at the time of discharge reached high efficacy in predicting their neutralizing antibody levels one year later. Conclusions This study is the first to characterize the oral and gut microbiome and metabonomics in 1-year convalescents of COVID-19. The key microbiome and metabolites in the process of recovery were identified, and provided new treatment ideas for accelerating recovery. And the predictive models based on the microbiome and metabolomics afford new insights for predicting the recovery situation which benefited affected individuals and healthcare.

Molecular recognition was utilized to fabricate bioinspired calcium phosphate (Ca-P) coating on bioabsorbable magnesium alloys through small biomolecules such as Vitamin C (VC). Ca-P and VC hybrid coating (Ca-PVC) was successfully fabricated on AZ31 Mg alloy. The surface morphology and chemical composition of the coatings were investigated using SEM, XRD, and FTIR together with XPS. The results showed that the Ca-PVC coating was composed of bamboo leaf-like Ca-P particles with a thickness of about three times that of the Ca-P coating. The surface roughness of the Ca-PVC coating (1.12 ± 0.12 μm) was lower than that (3.14 ± 1.93 μm) of Ca-P coating, suggesting the formation of refined Ca-P particles resulting from the VC addition. The corrosion resistance of the coated samples was characterized via electrochemical polarization, impedance spectroscopy, and immersion hydrogen evolution tests. The cell toxicity of the coated samples was evaluated utilizing mouse MC3T3-E1 pre-osteoblasts. The charge transfer resistance (Rct) of the Ca-PVC coated alloy increased as compared to the bare and Ca-P coated alloy samples. The Ca-PVC coated alloy exhibited minimal corrosion current density (1.36 × 10−6 A cm−2), which is one order of magnitude lower in comparison to that of the Ca-P coated alloy. These results confirm that VC addition greatly enhanced the coating resistance on AZ31 Mg alloy. It was also noticed that the Ca-PVC coated samples rapidly induced the formation of apatite after immersion in Hank's solution. VC was mainly transformed to L-Threonic acid, which facilitated the nucleation process of the Ca-PVC coating and significantly increased the thickness, density, and bonding strength of the coating. With enhanced corrosion resistance property and excellent biocompatibility, Ca-PVC coating has great potential for application in biodegradable Mg-based alloys.

Objective To compare the disease burden of gastric cancer among Chinese residents in 1990 and 2017 for providing references to decision making on gastric cancer prevention and health management. Methods Using the data of 1990 and 2017 from the Global Burden of Diseases, Injuries, and Risk Factors Study (GBD) 2017, we analyzed the changes in years of life lost (YLL), years living with disability (YLD) and disability-adjusted life years (DALY) due to gastric cancer in Chinese population between 1990 and 2017. Results In contrast with those in 1990, higher gastric cancer-related YLL (7 648 000 person-years [PYs] vs. 7 340 000 PYs), YLD ( 167 000 PYs vs. 74 000 PYs), standardized YLD rate (8.5/100 000 vs. 8.4/100 000) and DALY (7 815 000 PYs vs. 7 415000 PYs) were observed in 2017 in Chinese population, with the increasing percentage of 4.2%, 125.7%, 1.2%, and 5.4%; but the standardized YLL rate declined from 779/100 000 to 383.2/100 000 and the standardized DALY rate declined from 787.3/100 000 to 391.7/100 000. There were gender, age, and region differences in the changes between 1990 and 2017 for disease burden due to gastric cancer in the population; in the males, the YLD increased by 152.1%, higher than 76.9% in the females; while the standardized YLL and DALY rate decreased by 46.4% and 45.8% in the males, both lower than in female population (58.5% and 58.0%); the YLL, standardized YLD rate, and DALY of the males increased by 13.7%, 11.6%, and 15.1% but those of the females decreased by 12.7%, 19.0%, and 11.9%, respectively; the DALY decreased in people aged < 50 and 55 – 59 years but increased in the people of other age groups, with the highest increase of 133.5% in the people ≥ 80 years; the standardized DALY rate decreased among all age groups and the decrease percentage (75.7%) was the highest for the people aged 20 – 24 years; the highest increase of DALY (86.2%) was observed in the population of Shaanxi province whereas the greatest decrease of DALY (25.1%) was in the population of Tibet Autonomous Region; the standardized DALY rate decreased in all 33 provincial-level administrative divisions, with the greatest decrease (66.7%) in Beijing and the least decrease (14.7%) in Shaanxi province. Conclusion The disease burden of gastric cancer in Chinese population decreased in 2017 compared to that in 1990 but the decrease differed by gender and region and the disease burden is still at a relatively high level.

Background and objectivesThe Movement Disorder Society’s Unified Parkinson’s Disease Rating Scale Part III (MDS-UPDRS III) is mostly common used for assessing the motor symptoms of Parkinson’s disease (PD). In remote circumstances, vision-based techniques have many strengths over wearable sensors. However, rigidity (item 3.3) and postural stability (item 3.12) in the MDS-UPDRS III cannot be assessed remotely since participants need to be touched by a trained examiner during testing. We developed the four scoring models of rigidity of the neck, rigidity of the lower extremities, rigidity of the upper extremities, and postural stability based on features extracted from other available and touchless motions.MethodsThe red, green, and blue (RGB) computer vision algorithm and machine learning were combined with other available motions from the MDS-UPDRS III evaluation. A total of 104 patients with PD were split into a train set (89 individuals) and a test set (15 individuals). The light gradient boosting machine (LightGBM) multiclassification model was trained. Weighted kappa (k), absolute accuracy (ACC ± 0), and Spearman’s correlation coefficient (rho) were used to evaluate the performance of model.ResultsFor model of rigidity of the upper extremities, k = 0.58 (moderate), ACC ± 0 = 0.73, and rho = 0.64 (moderate). For model of rigidity of the lower extremities, k = 0.66 (substantial), ACC ± 0 = 0.70, and rho = 0.76 (strong). For model of rigidity of the neck, k = 0.60 (moderate), ACC ± 0 = 0.73, and rho = 0.60 (moderate). For model of postural stability, k = 0.66 (substantial), ACC ± 0 = 0.73, and rho = 0.68 (moderate).ConclusionOur study can be meaningful for remote assessments, especially when people have to maintain social distance, e.g., in situations such as the coronavirus disease-2019 (COVID-19) pandemic.

Abstract The myodural bridge (MDB) is a dense connective tissue bridge connecting the suboccipital muscles to the spinal dura mater, and it has been proven to be a normal common existing structure in humans and mammals. Some scholars believe that the suboccipital muscles can serve as a dynamic cerebrospinal fluid (CSF) pump via the MDB, and they found head rotations promote the CSF flow in human body, which provided evidence for this hypothesis. Head movement is a complex motion, but the effects of other forms of head movement on CSF circulation are less known. The present study explored the effects of head-nodding on CSF circulation. The CSF flow of 60 healthy volunteers was analyzed via cine phase-contrast magnetic resonance imaging at the level of the occipitocervical junction before and after one-minute-head-nodding period. Furthermore, the CSF pressures of 100 volunteers were measured via lumbar puncture before and after 5 times head-nodding during their anesthetizing for surgical preparation. As a result, it was found that the maximum and average CSF flow rates at the level of the upper border of atlas during ventricular diastole were significantly decreased from 1.965 ± 0.531 to 1.839 ± 0.460 ml/s and from 0.702 ± 0.253 to 0.606 ± 0.228 ml/s respectively. In the meantime, the changes in the ratio of cranial and caudal orientation of the net flow volume were found differed significantly after the one-minute-head-nodding period (p = 0.017). And on the other hand, the CSF pressures at the L3–L4 level were markedly increased 116.03 ± 26.13 to 124.64 ± 26.18 mmH2O. In conclusion, the head-nodding has obvious effects on CSF circulation and head movement is one of the important drivers of cerebrospinal fluid circulation. We propose that the suboccipital muscles, participating in various head movements, might pull the dura sac via the myodural bridge, and thus, head movement provides power for the CSF circulation.

Heavy metal air pollution poses a serious threat to human health and the environment in Chinese tourist cities. In this study, we investigated the temporal and spatial variations of atmospheric heavy metal pollution using moss bags in Xichang, a tourist destination in Southwest China. The biomonitoring investigation used an indigenous moss (Taxiphyllum taxirameum) transplanted into bags. Moss bags were exposed to 22 sites including industrial, agricultural, urban/residential, tourist, and high-traffic sites, across four different seasons in 2019–2020. The results showed that T. taxirameum was a good biomonitor of air pollution in Xichang. Among the 22 sample points, air pollution was the worst along the G102 motorway. Heavy metal emissions varied in different regions and directions. Temporal changes significantly influenced the heavy metals accumulated in moss bags, with low deposition of most elements observed at nearly all sampling sites in summer. Different seasons and regions were important factors affecting atmospheric heavy metal pollution. Based on the correlation analysis and the positive matric factorization model, the results revealed that heavy metals in moss bags in Xichang were mainly derived from anthropogenic sources and atmospheric deposition. Overall, this research provides an important reference for air pollution monitoring in urban areas.

The light-small multi-rotor unmanned aerial vehicle (UAV) carrying with a mini synthetic aperture radar (SAR) payload has provided an advanced information acquisition technology for the target detecting and imaging fields. In this paper, a W-band compact SAR system is proposed, which has been successfully deployed on the unmanned aerial vehicle (UAV) platform demonstrating state-of-the-art imaging resolution of about 4.5 cm. In order to achieve remarkable compactness, light weight and powersaving feature for the proposed system, the techniques of the three-dimensional (3-D) integration are adopted, where a whole-system size of only 67 × 60×50 mm3 and the weight of 400 g are obtained. This proposed system achieves higher SAR-imaging resolution thanks to the contributions of many efforts, including the technique of high-linearity waveform generation, the substrate integrated waveguide (SIW) antenna with high isolation by the electromagnetic band-gap (EBG) mechanism, and a novel motion compensation method for the imaging formation. To validate this proposed idea, a UAV SAR prototype operating at W-band is designed and fabricated. The measurements show great signal-to-noise ratio imaging results with excellent focusing effect. The proposed SAR system is promisingly an ideal candidate for the target detecting/imaging applications deployed on the UAV platform.

In view of the multimetal barrier environment of nuclear power plant, by considering the factors such as transmission power, transmission position, and multipath interference, based on the simulation of metal pipes and equipment, this paper carries out the barrier area channel modeling in logarithmic fading mode and makes quantitative analysis on the channel transmission, path loss, channel power characteristics, and so on under the metal barrier environment. Based on the channel modeling, this paper optimizes the coverage of the network in the obstacle area by using the improved teaching and learning group intelligent algorithm. The simulation results show that the improved teaching and learning algorithm can optimize the network coverage of the obstacle area well, and under the four obstacle modules, 14 nodes can cover the whole area by more than 99%. This provides a solution to the problem of network coverage in the practical application of wireless sensor networks.

Purpose: To investigate the anti-thrombosis and anti-tumor effect of the water extract of the caulis of Sargentodoxa cuneata (Oliv.) Rehd. et Wils. (WCSW) in rat and mouse models. Methods: WCSW extract was prepared and the main constituents were determined by high pressure liquid chromatography (HPLC). The acute toxicity of the extract was determined in mice. Platelet aggregation in rat platelet-rich plasma (PRP) was examined to evaluate the effect of the extract on platelet function. Thereafter, the cytotoxic activity of WCSW on HL60, A549, S180 and H22 cells was determined by 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. In vivo antitumor effect of WCSW was further evaluated on H22 cells transplanted in mice, while the expression of caspase-3, caspase-9, Bcl-2 and Bax proteins were assayed by Western blot analysis. Results: Protocatechuic acid, rhodiola glucoside and chlorogenic acid were identified as the main constituents of WCSW. Platelet aggregation was significantly inhibited by treatment with the extract at concentrations of 1, 5 and 10 mg/mL. WCSW also showed significant inhibitory effect on HL60, A549, S180 and H22 cells in vitro with half maximal inhibitory concentration (IC50 value of 321.9, 285.0, 130.3 and 76.1 μg/mL, respectively. Furthermore, WCSW exhibited obvious anti-tumor effect on H22 transplanted tumor in vivo. After treatment with WCSW, caspase-3, caspase-9 and Bax were significantly (p < 0.05) up-regulated, whereas Bcl-2 was significantly (p < 0.05) down-regulated in the tumor tissues. Conclusion: WCSW possesses significant antithrombosis and anti-tumor effect, and therefore, has the potentials to be developed into effective drugs for clinical treatment of cancer and thrombosis diseases. AIM: To study the optical coherence tomography(OCT)treatment of optic fiber thickness in patients with high myopia combined with glaucoma.METHODS: Totally 160 eyes of 80 patients with high myopia and glaucoma who were treated in our hospital during 2017-01/12 were selected as the observation group, and 60 healthy volunteers(120 eyes)who underwent physical examination in our hospital during the same period were selected as the control group. OCT was used to analyze the thickness of optic nerve fibers in both groups.RESULTS: Subjects compared with control group, the observation group of patients with nasal side, temporal, under the nose, nasal, under the temporal and the week on the average thickness of the optic nerve fiber layer is reduced, and the temporal side nerve fiber layer thickness increased(PPCONCLUSION: In patients with high myopia combined with glaucoma, the optic nerve fibers in all quadrants except the temporal side become thinner.

Glycerol is a new green organic solvent. It not only acts as a phase transfer catalyst but also as a recyclable solvent by significantly enhancing the intramolecular cyclization. A series of 4-aryl-7,7 -dimethyl-5-oxo-3,4,5,6,7,8-hexahydrocoumarin derivatives were synthesized by one-pot multicomponent reaction of Meldrum’s acid with benzaldehyde, and cyclohexanedione in glycerol. Compared to classic methods, this method has the advantages of milder reaction conditions, good yields, easy processing and environmental benignity.

Abstract Many studies have established the high diagnostic accuracy of shear wave elastography (SWE) for the detection of prostate cancer (PCa); however, its utility remains a subject of debate. This meta-analysis sought to appraise the overall accuracy of SWE for the detection of PCa. A literature search of the PubMed, Embase, Cochrane Library, Web of Science and CNKI (China National Knowledge Infrastructure) databases was conducted. In all of the included studies, the diagnostic accuracy of SWE was compared with that of histopathology, which was used as a standard. Data were pooled, and the sensitivity, specificity, area under the curve (AUC), positive likelihood ratio (PLR), negative likelihood ratio (NLR), and diagnostic odds ratio (DOR) were calculated to estimate the accuracy of SWE. The pooled sensitivity and specificity for the diagnosis of PCa by SWE were 0.844 (95% confidence interval: 0.696–0.927) and 0.860 (0.792–0.908), respectively. The AUC was 0.91 (0.89–0.94), the PLR was 6.017 (3.674–9.853), and the NLR was 0.182 (0.085–0.389). The DOR was 33.069 (10.222–106.982). Thus, SWE exhibited high accuracy for the detection of PCa using histopathology as a diagnostic standard. Moreover, SWE may reduce the number of core biopsies needed.

Abstract Background Jiangtang decoction (JTD) is a China patented drug which contains Euphorbia humifusa Willd, Salvia miltiorrhiza Bunge, Astragalus mongholicus Bunge, Anemarrhena asphodeloides Bunge, and Coptis chinensis Franch. For decades, it has also been used clinically to treat diabetic nephropathy (DN) effectively; however, the associated mechanisms remain unknown. Thus, the present study aimed to examine the protective efficacy of JTD in DN and elucidate the underlying molecular mechanisms. Methods A diabetic model using KK-Ay mice received a daily administration of JTD for 12 weeks. Body weight, blood glucose, triglycerides (TGs), total cholesterol (TC), urea nitrogen (UN), creatinine (Cr), and microalbumin/urine creatinine (MA/UCREA) was measured every 4 weeks. Furthermore, on the day of the sacrifice, blood, urine, and kidneys were collected to assess renal function according to general parameters. Pathological staining was performed to evaluate the protective renal effect of JTD. In addition, the levels of inflammatory cytokines (tumor necrosis factor-α [TNF-α], interleukin [IL]-6 and intercellular adhesion molecule [ICAM]-1), insulin receptor substrate [IRS]-1, advanced glycation end products [AGEs], and receptor of glycation end products [RAGE] were assessed. Finally, the phosphoinositide 3-kinase (PI3K)/protein kinase B (Akt) signaling pathway and involvement of nuclear factor-κB (NF-κB) was further analyzed. Results After 12 weeks of metformin and JTD administration, the mice exhibited a significant amelioration in glucose and lipid metabolism dysfunction, reduced morphological changes in the renal tissue, decreased urinary albumin excretion, and normalized creatinine clearance. JTD treatment also reduced the accumulation of AGEs and RAGE, up-regulated IRS-1, and increased the phosphorylation of both PI3K (p85) and Akt, indicating that the activation of the PI3K/Akt signaling pathway was involved. Additionally, JTD administration reduced the elevated levels of renal inflammatory mediators and decreased the phosphorylation of NF-κB p65. Conclusions These results demonstrate that JTD might reduce inflammation in DN through the PI3K/Akt and NF-κB signaling pathways.

In this study, we evaluated the potential of peripheral blood mononuclear cells (PBMC) derived long non-coding RNAs (lncRNAs) as biomarkers for acute myocardial infarction (AMI). To assess the value of PBMCs-derived lncRNAs levels in predicting clinical outcomes in AMI. We measured the PBMC-derived levels of 10 individual lncRNAs which are known to be relevant to cardiovascular disease in PBMCs from 132 AMI patients and 104 healthy participants using quantitative RT-PCR. Abstracts: Objective: To summarize the use rate, safety, efficacy of antithrombotics in stroke/transient ischemic attack (TIA) prevention, and reasons for not using dabigatran etexilate (DE) in Shanghai, China. Methods: Non-valvular atrial fibrillation (NVAF)-associated stroke patients were prospectively registered as an electronic database. Use rate of antithrombotics and reasons for not using DE were extracted during follow-up. Patients' baseline characteristics, recurrent ischemic stroke/TIA events and bleeding complications were analyzed. Patients: From April 2012 to August 2014, 110 inpatients with NVAF-associated stroke were studied in our hospital. NVAF was diagnosed by 12-lead electrocardiogram, 24 h Holter and echocardiography. Results: Before introduction of DE (April 2013), use rates of warfarin and antiplatelets were 28.9% (11/38) and 60.5% (23/38) respectively; after that, use rates of warfarin, DE, and antiplatelets were 20.8% (15/72), 12.5% (9/72), and 43.1% (31/72). The DE did not improve use of anticoagulants (P = 0.639). There were 19 (17.3%) recurrent ischemic stroke events up to October 2015; two (9.5%) in the non-user group, 10 (18.5%) in the antiplatelet group, and seven (20.0%) in the anticoagulants group (P = 0.570). Furthermore, recurrence rates were similar between the DE group (20.0%) and the Warfarin group (20.0%, P = 1.000). The most common reason for not using DE was financial concerns (61.0%), followed by inconvenience to purchase (14.0%) and hemorrhage concerns (11.0%). Two patients using warfarin found fecal occult blood so they stopped warfarin and began to use antiplatelet drugs. No bleeding event occurred in the other groups. Only one patient had side effects (dyspepsia and gastroesophageal reflux) from DE. Conclusion: The use rate of either DE or warfarin in Shanghai was low; DE had not improved anticoagulation therapy for NVAF patients in Shanghai mainly because DE had not been covered by health insurance. Type 2 Diabetes mellitus (T2DM) is closely correlated with cognitive impairment and neurodegenerative disease. Bushen Huoxue (BSHX) is a compound Chinese medicine used clinically to treat diabetes-induced cognitive impairment. However, its underlying mechanisms remain unclear. In the present study, KKAy mice, a genetic model of type 2 diabetes with obesity and insulin resistant hyperglycemia, received a daily administration of BSHX for 12 weeks. Blood glucose was measured every 4 weeks. After 12 weeks, BSHX treatment significantly ameliorated the T2DM related insults, including the increased blood glucose, the impaired spatial memory, decreased cerebral blood flow (CBF), occurrence of albumin leakage, leukocyte adhesion and opening capillary rarefaction. Meanwhile, the downregulation of the tight junction proteins (TJ) claudin-5, occludin, zonula occluden-1 (ZO-1) and JAM-1 between endothelial cells, amyloid-β (Aβ) accumulation in hippocampus, increased AGEs and RAGE, and expression of RhoA/ROCK/moesin signaling pathway and phosphorylation of Src kinase in KKAy mice were significantly protected by BSHX treatment. These results indicate that the protective effect of BSHX on T2DM-induced cognitive impairment involves regulation of RhoA/ROCK1/moesin signaling pathway and phosphorylation of Src kinase.

The reconstruction methods of a rational secret sharing based on the Markov decision was studied.Firstly,a rational secret sharing system model was proposed using the Markov decision process,which included the players set,the states set,the risk preference function,the state transfer function,the return function,etc.The risk preference function was introduced in order to depict the state set and the state transfer function in this model.Secondly,a rational secret sharing scheme was constructed based on the proposed system model,which was able to solve the secret reconstruction problems according to the Markov strategy.Finally,the functional relations of among the discount factor,the return func-tion and the risk preference function was proposed in this scheme.The analysis results show that the proposed model and scheme are rationality and validity.

Initially, to determine whether the magnetic nanoparticle Fe3O4 and Au can facilitate the anticancer drug to reverse the resistance of cancer cells, we have explored the cytotoxic effect of daunomycin (DNR) with and without the magnetic nano-Fe3O4 or nano-Au on K562 and K562/A02 cells by MTT assay. Besides, the intracellular DNR concentration and apoptosis of the K562/A02 cells was further investigated by flow cytometry and confocal fluorescence microscopic studies. The MDR1 gene expression of the K562/A02 cells was also studied by RT-PCR method. Our results indicate that 5.0 × 10−7M nano-Fe3O4 or 2.0 × 10−8M nano-Au is biocompatible and can apparently raise the intracellular DNR accumulation of the K562/A02 cells and increase the apoptosis of tumor cells. Moreover, our observations illustrate that although these two kinds of nanoparticles themselves could not lower the MDR1 gene expression of the K562/A02 cells, yet they could degrade the MDR1 gene level when combining with anticancer drug DNR. Concentrations and bioconcentration potential of nine elements (Ca, Cu, Fe, K, Mg, Mn, Na, P, and Zn) in ten species of wild edible Boletus and the corresponding underlying soils were analyzed. The analyses were performed using inductively coupled plasma atomic emission spectrophotometer. Boletus showed relative abundant contents of P, K, Fe, Mg, Ca, and Na and less of Zn, Cu, and Mn. Caps compared to stalks were enriched in P, K, Cu, Mg, and Zn, while stalks were enriched in Mn. The elements such as P and K were accumulated (BCF>1), while Ca, Fe, Mg, Mn, and Na were excluded (BCF

ICP-AES and microwave assisted digestion were applied to determine P, Mg, Ca, Zn, Na, Cu, Ba, Ni, V, Cd, Sr, Co, and Li in the caps and stipes of Boletus edulis collected from six spatially distant sites in Yunnan province, China. Fruiting bodies of King Bolete are abundant in P, Mg, Ca, Zn, Cu, and Na, followed by Ba, Cd, Ni, V, Li, Sr, and Co. Contents of P, Mg, Zn, and Cu are more abundant in caps than in stipes of King Bolete. However, elements such as Na, Ba, Cd, Ni, V, Li, Sr, and Co prefer to accumulate in stipes of mushrooms from Yaoan, Chuxiong. The results of this study indicate that spatial variations of elements between caps and stipes are mainly related to different bedrock soil geochemistry and enrichment capability for various elements.
