Package delivery via ridesharing provides appealing benefits of lower delivery cost and efficient vehicle usage. Most existing ridesharing systems operate the matching of ridesharing in a centralized manner, which may result in the single point of failure once the controller breaks down or is under attack. To tackle such problems, our goal in this paper is to develop a blockchain-based package delivery ridesharing system, where decentralization is adopted to remove intermediaries and direct transactions between the providers and the requestors are allowed. To complete the matching process under decentralized structure, an Event-Triggered Distributed Deep Reinforcement Learning (ETDDRL) algorithm is proposed to generate/update the real-time ridesharing orders for the new coming ridesharing requests from a local view. Simulation results reveal the vast potential of the ETDDRL matching algorithm under the blockchain framework for the promotion of the ridesharing profits. Finally, we develop an application for Android-based terminals to verify the ETDDRL matching algorithm.

Traditional villages possess distinct regional cultural traits and serve as a concrete manifestation and inheritance of traditional culture. In today’s era, where the preservation of traditional villages is emphasized, the study of Korean traditional villages in Northeast China, with their ethnic culture and unique spatial patterns, becomes significantly important. In this paper, we adopt the spatial gene perspective and establish a framework for analyzing spatial genes in traditional villages. Based on frequency analysis of qualitative data obtained from NVivo 14, 16 spatial gene entries were identified. Village spatial gene entries were quantified using ArcGIS, FRAGSTATS and other software. Additionally, 47 spatial gene fragments were extracted using the natural breaks method. Spatial gene sequences were mined using Spearman correlation analysis, and spatial gene maps were created. The study results indicate that: (1) the primary genes of traditional Korean villages include close to mountain, close to water, globular boundary, low integration road, etc. (2) There are significant correlations between spatial genes such as distance from mountains, mountain coverage and road integration, water coverage and boundary shape index. (3) Plain, mountain, and hill-type villages differ in characteristics such as road linear density, water coverage, and courtyard type.

For engineering structures with saline soil as a filling material, such as channel slope, road subgrade, etc., the rich soluble salt in the soil is an important potential factor affecting their safety performance. This study examines the Atterberg limits, shear strength, and compressibility of carbonate saline soil samples with different NaHCO3 contents in Northeast China. The mechanism underlying the influence of salt content on soil macroscopic properties was investigated based on a volumetric flask test, a mercury intrusion porosimetry (MIP) test, and a scanning electron microscopic (SEM) test. The results demonstrated that when NaHCO3 contents were lower than the threshold value of 1.5%, the bound water film adsorbed on the surface of clay particles thickened continuously, and correspondingly, the Atterberg limits and plasticity index increased rapidly as the increase of sodium ion content. Meanwhile, the bonding force between particles was weakened, the dispersion of large aggregates was enhanced, and the soil structure became looser. Macroscopically, the compressibility increased and the shear strength (mainly cohesion) decreased by 28.64%. However, when the NaHCO3 content exceeded the threshold value of 1.5%, the salt gradually approached solubility and filled the pores between particles in the form of crystals, resulting in a decrease in soil porosity. The cementation effect generated by salt crystals increased the bonding force between soil particles, leading to a decrease in plasticity index and an improvement in soil mechanical properties. Moreover, this work provides valuable suggestions and theoretical guidance for the scientific utilization of carbonate saline soil in backfill engineering projects.

Tillering is an important agronomic trait of rice (Oryza sativa) that affects the number of effective panicles, thereby affecting yields. The phytohormone auxin plays a key role in tillering. Here we identified the high tillering and semi-dwarf 1 (htsd1) mutant with auxin-deficiency root characteristics, such as shortened lateral roots, reduced lateral root density, and enlarged root angles. htsd1 showed reduced sensitivity to auxin, but the external application of indole-3-acetic acid (IAA) inhibited its tillering. We identified the mutated gene in htsd1 as AUXIN1 (OsAUX1, LOC_Os01g63770), which encodes an auxin influx transporter. The promoter sequence of OsAUX1 contains many SQUAMOSA PROMOTER BINDING PROTEIN-LIKE (SPL) binding sites, and we demonstrated that SPL7 binds to the OsAUX1 promoter. TEOSINTE BRANCHED1 (OsTB1), a key gene that negatively regulates tillering, was significantly downregulated in htsd1. Tillering was enhanced in the OsTB1 knockout mutant, and the external application of IAA inhibited tiller elongation in this mutant. Overexpressing OsTB1 restored the multi-tiller phenotype of htsd1. These results suggest that SPL7 directly binds to the OsAUX1 promoter and regulates tillering in rice by altering OsTB1 expression to modulate auxin signaling.

Abstract This study aimed to develop and validate a nomogram model that includes clinical and laboratory indicators to predict the risk of metabolic-associated fatty liver disease (MAFLD) in young Chinese individuals. This study retrospectively analyzed a cohort of young population who underwent health examination from November 2018 to December 2021 at The Affiliated Hospital of Southwest Medical University in Luzhou City, Sichuan Province, China. We extracted the clinical and laboratory data of 43,040 subjects and randomized participants into the training and validation groups (7:3). Univariate logistic regression analysis, the least absolute shrinkage and selection operator regression, and multivariate logistic regression models identified significant variables independently associated with MAFLD. The predictive accuracy of the model was analyzed in the training and validation sets using area under the receiver operating characteristic (AUROC), calibration curves, and decision curve analysis. In this study, we identified nine predictors from 31 variables, including age, gender, body mass index, waist-to-hip ratio, alanine aminotransferase, low-density lipoprotein cholesterol, high-density lipoprotein cholesterol, uric acid, and smoking. The AUROC for the subjects in the training and validation groups was 0.874 and 0.875, respectively. The calibration curves show excellent accuracy of the nomogram. This nomogram which was based on demographic characteristics, lifestyle habits, anthropometrics, and laboratory data can visually and individually predict the risk of developing MAFLD. This nomogram is a quick and effective screening tool for assessing the risk of MAFLD in younger populations and identifying individuals at high risk of MAFLD, thereby contributing to the improvement of MAFLD management.

ABSTRACT This study aimed to characterize carbapenem-resistant Acinetobacter baumannii (CRAB) isolates from Jiangxi patients using whole-genome sequencing (WGS). We subjected 100 clinical CRAB strains isolated from the three local largest teaching hospitals to WGS and antimicrobial susceptibility testing. Molecular epidemiology was investigated using multilocus sequence typing, core genome multilocus typing, core genome single-nucleotide polymorphism phylogeny, and pulsed-field gel electrophoresis. The most prevalent acquired carbapenemase was blaOXA-23, predominant in all isolates (100%). Isolates belonging to the dominating international clone IC2 accounted for 92% of all isolates. International IC11 (ST164Pas/ST1418Ox) clone was found in an additional 8% (eight isolates), with seven isolates (87.5%) carrying an acquired additional blaNDM-1 carbapenemase. The oxa23-associated Tn2009, either alone or in a tandem repeat structure containing four copies of blaOXA-23, was discovered in 62% (57 isolates) of IC2. The oxa23-associated Tn2006 was identified in 38% (35 isolates) of IC2 and all IC11 isolates. A putative conjugative RP-T1 (formerly RepAci6) plasmid with blaOXA-23 in Tn2006 within AbaR4, designated pSRM1.1, was found in IC2 A. baumannii strain SRM1. The blaNDM-1 gene found in seven IC11 isolates was located on a novel Tn6924-like transposon, a first-time report in IC11. These findings underscore the significant importance of real-time surveillance to prevent the further spread of CRAB.IMPORTANCECarbapenem-resistant Acinetobacter baumannii (CRAB) is notorious for causing difficult-to-treat infections. To elucidate the molecular and clinical epidemiology of CRAB in Jiangxi, clinical CRAB isolates were collected and underwent whole-genome sequencing and antibiotic susceptibility phenotyping. Key findings included the predominance of OXA-23-producing IC2 A. baumannii, marked by the emergence of OXA-23 and NDM-1-producing IC11 strains.

In cold mountain environments, rock glaciers and talus represent common periglacial landforms. Accurate monitoring of their activity is crucial for understanding alpine kinematics. Presently, spaceborne SAR satellites monitor periglacial landform deformations mainly utilizing medium-resolution data. However, capturing surface structure deformations accurately and comprehending the movement mechanisms remain challenging. This study employs high-resolution GF-7 optical stereo images and Radarsat-2 SAR data, introducing a multibaseline persistent scatterer and distributed scatterer combined multitemporal InSAR (MT-InSAR) method to identify rock glaciers and talus landforms, as well as to analyze their deformations. Initially, rock glaciers and talus are outlined using GF-7 optical images, and digital surface models are extracted. The developed MT-InSAR method then detects the line of sight and slope-parallel deformations of these landforms. Radarsat-2 monitoring reveals that 47.5% of identified rock glaciers are classified as active, while talus deformations are less active compared to rock glaciers. By utilizing high-resolution optical and SAR satellite data, we first documented the intricate deformation features within standard rock glaciers, such as the front, lateral margins, and optionally ridge-and-furrow deformations, as well as their interaction with the surrounding terrain topography. This discovery offers evidence of gravity-driven forces impacting rock glacier movement. We have also conducted a deformation analysis of the talus, obtaining spatial deformation characteristics of typical talus. It was observed that the deformation of the talus is jointly influenced by the conditions of surface debris cover and the slope of the terrain. This study highlights the value of high-resolution optical and SAR satellites in studying periglacial geomorphology dynamics.

Rock glaciers are typical periglacial landforms with tongue or lobate morphological shapes and characterized by the distinct front, lateral margins, and often by ridge-and-furrow surface topography textures as well as kinematic characteristics, widely distributed in alpine environments. Multitemporal Synthetic aperture radar interferometry (MT-InSAR), is a remote sensing technique with demonstrated effectiveness for detecting landform kinematics. However, its application to rock glaciers is challenged by temporal decorrelation and atmospheric phase noises due to complex topography and snow cover. We designed a quadtree segmentation and parallel computing-based MT-InSAR method to improve the quality and efficiency of deformation measurement of rock glaciers. We applied the method to a rock glacier inventory of the Nyainqêntanglha Range, China, derived from high-resolution Gaofen-2 images, to quantify the activity rate of each rock glacier. Results showed that 32.1% (6,389) of the identified rock glaciers exhibited slope-parallel deformation rates exceeding 100 mm/y. The activities of the rock glaciers exhibited strong correlations with their distance to glaciers, precipitation, freeze–thaw magnitude, and permafrost occurrence probability. The results demonstrate the effectiveness of the developed segmentation-parallel MT-InSAR method for monitoring rock glacier deformation over a large region.

The leaf scorching trait at flowering is a crucial thermosensitive phenotype in maize under high temperature stress (HS), yet the genetic basis of this trait remains poorly understood. In this study, we genotyped a 254 RIL-F2:8 population, derived from the leaf scorch-free parental inbred line Abe2 and the leaf scorching maternal inbred line B73, using the specific-locus amplified fragment sequencing (SLAF-seq) method. A total of 10,112 polymorphic SLAF markers were developed, and a high-density genetic map with a total length of 1,475.88 cM was constructed. The average sequencing depth of the parents was 55.23X, and that of the progeny was 12.53X. Then, we identified a total of 16 QTLs associated with thermotolerant traits at flowering, of which four QTLs of leaf scorching damage (LS) were distributed on chromosomes 1 (qLS1), 2 (qLS2.1, qLS2.2) and 3 (qLS3), which could explain 19.73% of phenotypic variation. Combining one qLS1 locus with QTL-seq results led to the identification of 6 candidate genes. Expression experiments and sequence variation indicated that Zm00001d033328, encoding N-acetyl-gamma-glutamyl-phosphate reductase, was the most likely candidate gene controlling thermotolerant traits at flowering. In summary, the high-density genetic map and genetic basis of thermotolerant traits lay a critical foundation for mapping other complex traits and identifying the genes associated with thermotolerant traits in maize.

In this paper, a multi-region dynamic economic dispatch model with power balance constraint is constructed to seek the actual reserve demand and reasonable reserve allocation method of the system by relying on the dynamic response process of the units to the power balance of the interconnected system. For this kind of dynamic optimization problem with differential equation constraints, a discrete-optimization rolling solution strategy based on scale-optimization is proposed. First, in the discrete part of the dispatching model, basing on the correlation between the continuous quantities discrete scale and the active real-time balancing effect of the dispatching scheme, a continuous quantities discrete scale optimization strategy is proposed to maintain the safe and economic operation of the system; then, for the convergence problem of the solution of the dispatching model after discretization, this paper introduces the idea of decentralized coordination optimization and the operator splitting quadratic programming (OSQP), and through the limited information exchange between the regional grid and the coordinator, the consistent coordination of consistent variables is achieved. The simulation results verify the effectiveness of the model and the solution strategy.

The increasing incidence of retrogressive thaw slumps (RTSs) in permafrost regions underscores the need for detailed spatial and temporal analysis using InSAR technology to monitor and predict dynamic changes in the process of RTSs. Nevertheless, current InSAR deformation forecasting methods employing deep learning strategies such as the traditional long short-term memory (LSTM) and recent transformer models encounter difficulties in effectively capturing temporal features. Moreover, they are limited in their ability to directly integrate spatial information. In this paper, an innovative deep learning approach named Spacetimeformer is proposed for predicting medium- and short-term InSAR deformation of RTSs in the Chumar River area. This method employs a transformer architecture with a spatiotemporal attention mechanism, which enhances the long-term prediction capabilities of time series models and dynamic spatial modeling. It is applicable to multivariate InSAR spatiotemporal deformation prediction problems. The findings include a list of 72 RTSs compiled based on derived InSAR deformation maps and Sentinel-2 optical images, of which 64 have an average deformation rate exceeding 10 mm/year, indicating signs of permafrost degradation. The density distribution of the displacement maps predicted by the Spacetimeformer model aligned well with the InSAR deformation maps obtained from the small baseline subset (SBAS) method, with the overall prediction deviation controlled within 20 mm. In addition, the point-scale prediction results were compared with LSTM and transformer models. This study indicates that the Spacetimeformer network achieved good results in predicting the deformation of RTSs, with a root mean square error of 1.249 mm. The Spacetimeformer method for deformation prediction with the spacetime mechanism presented in this study can serve as a general framework for multivariate deformation prediction based on InSAR results. It can also quantitatively assess the spatial deformation characteristics and deformation trends of RTSs.

To address traditional impedance control methods' difficulty with obtaining stable forces during robot-skin contact, a force control based on the Gaussian mixture model/Gaussian mixture regression (GMM/GMR) algorithm fusing different compensation strategies is proposed. The contact relationship between a robot end effector and human skin is established through an impedance control model. To allow the robot to adapt to flexible skin environments, reinforcement learning algorithms and a strategy based on the skin mechanics model compensate for the impedance control strategy. Two different environment dynamics models for reinforcement learning that can be trained offline are proposed to quickly obtain reinforcement learning strategies. Three different compensation strategies are fused based on the GMM/GMR algorithm, exploiting the online calculation of physical models and offline strategies of reinforcement learning, which can improve the robustness and versatility of the algorithm when adapting to different skin environments. The experimental results show that the contact force obtained by the robot force control based on the GMM/GMR algorithm fusing different compensation strategies is relatively stable. It has better versatility than impedance control, and the force error is within ~±0.2 N.

Abstract The effects of injection pressure and ambient pressure on biodiesel spray characteristics were studied by coupling the in‐nozzle flow, and a constant‐volume combustion bomb experimental platform was built to verify the accuracy of the coupled numerical model. Spray tip penetration (STP), spray velocity, and droplet size/number distribution of biodiesel were calculated at different injection pressures (200, −250, −300, and −350 MPa) and environmental pressures (0.1, −0.5, −1.0, and −2.0). The sensitivity to injection pressures and environmental pressures was analyzed with particle concentration as the characteristic parameter. With the increase of injection pressure, the STP of biodiesel increased by 5%, which resulted in a faster decrease in the increasing rate of STP (V). Under the same conditions, with the increase of injection pressure, the axial and radial velocities of fuel droplets gradually rose, and the high‐speed area was enlarged, while the number of droplets and Sauter mean diameter (SMD) space generally decreased. Compared with the injection pressure, the environmental pressure significantly inhibited the STP and velocity of biodiesel, and the increasing rate of STP (V) decreased gradually and the number of droplet particles and SMD rose with the increase of the environmental pressure.

Abstract To provide guidance for plague surveillance and a reliable basis for plague prevention and control, we analyzed the habitat characteristics of Himalayan marmots, developed Himalayan marmot information collection system V3.0 based on global navigation satellite system (GNSS), remote sensing, and geographic information system (“3S”) technology, and drew a predictive spatial distribution map of Himalayan marmots in Qinghai Province. Field survey data of 352 marmot plague sites in Qinghai Province were collected in 2014, and the data from 80 sample sites were included. The Himalayan marmot habitat characteristics were analyzed based on “3S” technology using five environment variables (elevation, slope, aspect, vegetation cover, and grass type) and the geographical coordinates. Himalayan marmot information collection system V3.0, which has been approved by the National Copyright Administration of the People’s Republic of China (No.00764743), was used to draw a predictive spatial distribution map of Himalayan marmots in Qinghai province. Moreover, from 2015 to 2017, positioning data of the plague-foci and plague-free areas in Qinghai Province were collected using GNSS receptor for field validations to verify the accuracy of the marmot predictive spatial distribution map. Elevation, slope, vegetation cover, and grassland type were identified as important environmental factors that determine the spatial distribution of Himalayan marmots. The suitable range of environmental features was 3400–4600 m elevation, 5°–20° slope, 0.60–1.00 vegetation cover, and alpine meadows. The Himalayan marmot predictive spatial distribution map in Qinghai Province based on “3S” technology and marmot information collection system V3.0 had a spatial resolution of 30 m. Field validation in areas of Qinghai Province revealed a prediction accuracy and mean absolute error of 0.8669 and 0.1331, respectively, which indicated excellent prediction accuracy. This study greatly improved the work efficiency of plague surveillance and effectively reduced the work intensity of researchers. Application of “3S” technology and marmot information collection system V3.0 has improved the data collection efficiency, provided new technical means for plague investigation and research, and provided a reference for development of plague surveillance programs. The research results will play a positive role in promoting the improvement and perfection of plague prevention and control strategies in Qinghai province and even in China.

For most patients with advanced thymic epithelial tumors (TETs), a complete resection is a strong indicator of a better prognosis. But sometimes, primary surgery is unsatisfactory, and preoperative therapy is needed to facilitate complete resection. Neoadjuvant chemotherapy is the most used form of preoperative therapy. But studies on neoadjuvant chemotherapy have included mainly patients with thymoma; its efficacy in patients with thymic carcinoma is less known. Neoadjuvant chemoradiation has also been explored in a few studies. Novel therapies such as immunotherapy and targeted therapy have shown efficacy in patients with recurrent/metastatic TETs as a second-line option; their role as preoperative therapy is still under investigation. In this review, we discuss the existing evidence on preoperative therapy and the insight it provides for current clinical practice and future studies.

Chronic thromboembolic pulmonary hypertension (CTEPH) is characterized by thrombotic obstruction of the pulmonary arteries, and right ventricular (RV) dysfunction is a major cause of death. Cardiac magnetic resonance (CMR) is the gold standard for assessing heart wall deformation; therefore, we aimed to determine the prognostic value of CMR strain in patients with CTEPH. Strain derived by CMR was measured at the time of diagnosis in 45 patients with CTEPH, and the relationship between RV strain and prognosis was determined through follow-up. The value of RV strain in the prognostic model was compared with that of pulmonary arterial hypertension (PAH) risk stratification. The RV global peak longitudinal strain (GLS) and global peak circumferential strain (GCS) in CTEPH patients were lower than the normal references of RV strain in the control group. The penetration of distributed energy resources (DERs) and energy-intensive resources is gradually increasing in active distribution networks (ADNs), which leads to frequent and severe voltage violation problems. As a densely distributed flexible resource in the future distribution network, 5G base station (BS) backup battery is used to regulate the voltage profile of ADN in this paper. First, the dispatchable potential of 5G BS backup batteries is analyzed. Considering the spatial-temporal characteristics of electric load for 5G BS, the dispatchable capacity of backup batteries at different time intervals is evaluated based on historical heat map data. Then, a voltage profile optimization model for ADN is established, consisting of 5G BS backup batteries and other voltage regulation resources. In this model, the charging/discharging behavior of backup batteries is based on its evaluation result of dispatchable capacity. Finally, the range of charging/discharging cost coefficients of 5G BS that benefits ADN and 5G operators are analyzed respectively. Further, an incentive policy for 5G operators is proposed. Under this policy, the charging/discharging cost coefficients of 5G BS can achieve a win-win situation for ADN and 5G operators. As an emerging flexible resource in ADN, the effectiveness and economy of 5G BS backup batteries participating in voltage profile optimization are verified in a test distribution network.

Multi-microgrids (MMGs) suffer from power shortages due to the loss of utility grid support when an unintentional transition occurs. This imposes a transient shock on the system voltage and frequency. To maintain the frequency stability and power balance of an islanded MMG, this paper presents an underfrequency load shedding (UFLS) strategy with adaptive variation. A comprehensive load evaluation method based on a composite least squares support vector machine (CLS-SVM) is proposed to ensure uninterrupted power for critical loads. This method considers the comprehensive evaluation influence factors (CEIFs) of loads. Then, a least squares support vector machine (LS-SVM) provides the load shedding determination factors, transforming the problem of determining critical loads into a 0-1 planning problem. A method with adaptive variation is proposed to solve the UFLS model. The effectiveness of the proposed strategy is verified for an MMG model based on a modified IEEE 33-bus system. The test results show that: 1) the average accuracy of the proposed method is 21.05% higher than that based on LS-SVM; 2) compared with UFLS strategies based on the load level alone and on an intelligent algorithm, the frequency fluctuation range of the proposed strategy is 12.50% and 19.23% lower, respectively, and the frequency recovery time is 3.90% and 5.73% shorter, respectively; 3) compared with PSO, GOA and GA, the standard deviation of the iterative mean of the proposed algorithm decreases by 36.22%, 53.42%, and 34.00%, respectively. The proposed strategy can reduce the frequency fluctuation range and frequency recovery time while maintaining strong adaptability.

Abstract Introduction Excessiv generalization of fear contributes to the development and maintenance of pain. Prior research has demonstrated the importance of perception in fear generalization and found that individuals in painful conditions exhibited perceptual bias. However, the extent to which perceptual bias in pain affects the generalization of pain‐related fear and its underlying neural activity remains unclear. Methods Here, we tested whether perceptual bias in experimental pain individuals led to the overgeneralization of pain‐related fear by recording behavioral and neural responses. To this end, we established an experimental pain model by spraying capsaicin on the surface of the seventh cervical vertebra of the participant. A total of 23 experimental pain participants and 23 matched nonpain controls learned fear conditioning and then completed the fear generalization paradigm combined with the perceptual categorization task. Results We found that the novel and safety cues were more likely to be identified as threat cues in the experimental group, resulting in higher US expectancy ratings compared to the control group. The event‐related potential results showed that the experimental group exhibited earlier N1 latency and smaller P1 and late positive potential amplitudes than those in the control group. Conclusion Our findings suggest that the experimental pain individuals exhibited an excessive generalization of fear affected by perceptual bias and reduced their attentional allocation to pain‐related fear stimuli.

ABSTRACT The role of novel β-lactam/β-lactamase inhibitor combinations in ceftazidime-nonsusceptible (CAZ-NS) and imipenem-nonsusceptible (IPM-NS) Pseudomonas aeruginosa has not been fully elucidated. This study evaluated the in vitro activity of novel β-lactam/β-lactamase inhibitor combinations against Pseudomonas aeruginosa clinical isolates, determined how avibactam restored ceftazidime activity, and compared the activity of ceftazidime-avibactam (CZA) and imipenem-relebactam (IMR) against KPC-producing P. aeruginosa. Similar high susceptibility rates for CZA, IMR, and ceftolozane-tazobactam (88.9% to 89.8%) were found for 596 P. aeruginosa clinical isolates from 11 hospitals in China, and a higher susceptibility rate to ceftazidime than imipenem was observed (73.5% versus 63.1%). For CAZ-NS and IPM-NS isolates, susceptibility rates for CZA, ceftolozane-tazobactam, and IMR were 61.5% (75/122), 54.9% (67/122), and 51.6% (63/122), respectively. For CAZ-NS, IPM-NS but CZA-susceptible isolates, 34.7% (26/75) harbored acquired β-lactamases with KPC-2 predominant (n = 19), and 45.3% (34/75) presented overexpression of chromosomal β-lactamase ampC. Among 22 isolates carrying KPC-2 carbapenemase alone, susceptibility rates to CZA and IMR were 86.4% (19/22) and 9.1% (2/22), respectively. Notably, 95% (19/20) of IMR-nonsusceptible isolates had an inactivating mutation of oprD gene. In conclusion, CZA, ceftolozane-tazobactam, and IMR exhibit high activity against P. aeruginosa, and CZA is more active than IMR against CAZ-NS and IPM-NS isolates as well as KPC-producing P. aeruginosa. Avibactam overcomes ceftazidime resistance engendered by KPC-2 enzyme and overexpressed AmpC. IMPORTANCE The emergence of antimicrobial resistance poses a particular challenge globally, and the concept of P. aeruginosa with “difficult-to-treat” resistance (DTR-P. aeruginosa) was proposed. Here, P. aeruginosa clinical isolates were highly susceptible to three β-lactamase inhibitor combinations, CZA, IMR, and ceftolozane-tazobactam. The combination of KPC-2 enzyme and nonfunctional porin OprD contributed to IMR resistance in P. aeruginosa, and CZA was more active than IMR in fighting against KPC-2-producing P. aeruginosa. CZA also showed good activity against CAZ-NS and IPM-NS P. aeruginosa, primarily by inhibiting KPC-2 enzyme and overproduced AmpC, supporting the clinical use of CZA in the treatment of infections caused by DTR-P. aeruginosa.

Objective: The current study aimed to investigate the prognostic value of albumin-bilirubin (ALBI) score in predicting clinical outcomes of pancreatic cancer patients after pancreatoduodenectomy with liver metastasis following radiofrequency ablation.Methods: This retrospective study included 90 pancreatic cancer patients after pancreatoduodenectomy with liver metastasis from January 2012 to December 2018. In this study, the Chi-square or Fisher’s exact tests, the receiver operating characteristic (ROC) curve, Kaplan-Meier method and Log-rank test, univariate and multivariate Cox proportional hazard regression analyses, nomogram, calibration curves and decision curve analysis were used for all statistical analysis.Results: We analyzed the optimal cut-off value of ALBI by ROC curve, and the optimal cut-off value was −2.60. According to ALBI score, these patients were divided into two groups: low ALBI group (n = 33) and high ALBI group (n = 57). Patients with low ALBI score was significantly related to longer progression free survival (PFS) (p = 0.0002, HR: 3.039, 95% CI: 1.772–5.210) and overall survival (OS) (p = 0.0005, HR: 2.697, 95% CI: 1.539–4.720). The 1-, 3-, and 5-year PFS and OS rates in low ALBI group were higher than those in high ALBI group. ALBI was a potential independent prognostic factor for pancreatic cancer patients after pancreatoduodenectomy with liver metastasis following radiofrequency ablation. Moreover, the nomogram was used to predict the 1-, 3-, and 5-year survival probabilities of PFS and OS. The calibration curve shown that the prediction line matched the reference line well for postoperative 3-year PFS and OS. The DCA shown that nomogram model was better than the only ALBI, and indicated the ability for clinical decision-making, especially in 1-year PFS, and 3-, 5-year OS.Conclusion: ALBI is a potential independent factor for PFS and OS, and can predict the prognosis of pancreatic cancer patients after pancreatoduodenectomy with liver metastasis following radiofrequency ablation.

Plague caused by Yersinia pestis remains a public health threat worldwide. Because multidrug-resistant Y. pestis strains have been found in both humans and animals, phage therapy has attracted increasing attention as an alternative strategy against plague. However, phage resistance is a potential drawback of phage therapies, and the mechanism of phage resistance in Y. pestis is yet to be investigated. In this study, we obtained a bacteriophage-resistant strain of Y. pestis (S56) by continuously challenging Y. pestis 614F with the bacteriophage Yep-phi. Genome analysis identified three mutations in strain S56: waaA* (9-bp in-frame deletion 249GTCATCGTG257), cmk* (10-bp frameshift deletion 15CCGGTGATAA24), and ail* (1-bp frameshift deletion A538). WaaA (3-deoxy-D-manno-octulosonic acid transferase) is a key enzyme in lipopolysaccharide biosynthesis. The waaA* mutation leads to decreased phage adsorption because of the failure to synthesize the lipopolysaccharide core. The mutation in cmk (encoding cytidine monophosphate kinase) increased phage resistance, independent of phage adsorption, and caused in vitro growth defects in Y. pestis. The mutation in ail inhibited phage adsorption while restoring the growth of the waaA null mutant and accelerating the growth of the cmk null mutant. Our results confirmed that mutations in the WaaA–Cmk–Ail cascade in Y. pestis contribute to resistance against bacteriophage. Our findings help in understanding the interactions between Y. pestis and its phages.

SHLD1, as a component of the Shieldin (SHLD) complex, mediates DNA repair via non-homologous end-joining (NHEJ), an essential process during lymphocyte development. Here the authors show that SHLD1 is actually dispensable for lymphocyte development and V(D)J recombination, but is essential for class-switching recombination in activated B cells.

PurposeTo evaluate whether postoperative circulating tumor DNA (ctDNA) in plasma of patients with non-small cell lung cancer (NSCLC) can be used as a biomarker for early detection of molecular residual disease (MRD) and prediction of postoperative recurrence.MethodsThis study subjects were evaluated patients with surgical resected non-small cell lung cancer. All eligible patients underwent radical surgery operation followed by adjuvant therapy. Tumor tissue samples collected during operation were used to detect tumor mutation genes, and blood samples collected from peripheral veins after operation were used to collect ctDNA. Molecular residue disease (MRD) positive was defined as at least 1 true shared mutation identified in both the tumor sample and a plasma sample from the same patient was.ResultsPositive postoperatively ctDNA was associated with lower recurrence-free survival (RFS).The presence of MRD was a strong predictor of disease recurrence. The relative contribution of ctDNA-based MRD to the prediction of RFS is higher than all other clinicopathological variables, even higher than traditional TNM staging. In addition, MRD-positive patients who received adjuvant therapy had improved RFS compared to those who did not, the RFS of MRD-negative patients receiving adjuvant therapy was lower than that of patients not receiving adjuvant therapy.ConclusionsPost-operative ctDNA analysis is an effective method for recurrence risk stratification of NSCLC, which is beneficial to the management of patients with NSCLC.

Abstract Background Evaluation of new treatment policies is often costly and challenging in complex conditions, such as hepatitis C virus (HCV) treatment, or in limited-resource settings. We sought to identify hypothetical policies for HCV treatment that could best balance the prevention of cirrhosis while preserving resources (financial or otherwise). Methods The cohort consisted of 3792 HCV-infected patients without a history of cirrhosis or hepatocellular carcinoma at baseline from the national Veterans Health Administration from 2015 to 2019. To estimate the efficacy of hypothetical treatment policies, we utilized historical data and reinforcement learning to allow for greater flexibility when constructing new HCV treatment strategies. We tested and compared four new treatment policies: a simple stepwise policy based on Aspartate Aminotransferase to Platelet Ratio Index (APRI), a logistic regression based on APRI, a logistic regression on multiple longitudinal and demographic indicators that were prespecified for clinical significance, and a treatment policy based on a risk model developed for HCV infection. Results The risk-based hypothetical treatment policy achieved the lowest overall risk with a score of 0.016 (90% CI 0.016, 0.019) while treating the most high-risk (346.4 ± 1.4) and the fewest low-risk (361.0 ± 20.1) patients. Compared to hypothetical treatment policies that treated approximately the same number of patients (1843.7 vs. 1914.4 patients), the risk-based policy had more untreated time per patient (7968.4 vs. 7742.9 patient visits), signaling cost reduction for the healthcare system. Conclusions Off-policy evaluation strategies are useful to evaluate hypothetical treatment policies without implementation. If a quality risk model is available, risk-based treatment strategies can reduce overall risk and prioritize patients while reducing healthcare system costs.

In order to accurately obtain photometric information of high concentration SO42− and other substances in the process industry, the spectroscopy behavior of SO42−, S2−, Ni2+ and Cu2+ in air and nitrogen atmosphere was compared based on the UV-visible spectrophotometer with a nitrogen replacing the oxygen. Different from Ni2+ and Cu2+, the accuracy of SO42− and S2− in the ultraviolet region was effectively improved by using a nitrogen atmosphere (P detection results were regressed within the limited standard range, RE < 5%). The nitrogen atmosphere suppressed the additional light attenuation caused by its absorption of ultraviolet rays by isolating oxygen and was also reflected in the decrease in the degree of red shift of the characteristic wavelength for SO42− with increasing concentration. Therefore, the detection results of SO42− showed an effective improvement in sensitivity. Nevertheless, according to the complementary experimental results and theoretical calculations, in addition to oxygen absorption, the low detection accuracy of SO42− high concentration is also attributed to the reduction of the energy required for electronic excitation per unit group caused by the interaction between SO42− groups, resulting in a deviation of the C-A curve from linearity at high concentrations. The influence of this intermolecular force on the detection results is far more important than oxygen absorption. The research can provide reliable theoretical guidance and technical support for the pollution-free direct measurement of high-concentration solutions in the process industry and promote the sustainable development of the process industry.

Dalinor Lake, the second-largest endorheic salt lake in Inner Mongolia, has shown a shrinking trend given the lack of a significant decrease in precipitation (PRE). Based on high-spatial-resolution datasets, we employed a linear regression model, Theil–Sen median trend analysis, the Mann–Kendall test, and a land use transfer matrix to identify the spatio-temporal distribution and trends of PRE and actual evapotranspiration (AET) at the watershed scale during 2001–2019; then, the water deficit (WD) caused by land use changes in different surface lithology zones was analyzed. The results showed that the annual PRE and WD of the Dalinor Lake watershed showed insignificant upward trends, while the annual AET showed a significant upward trend. Spatially, about 89% of the watershed showed a significant upward trend for AET, while 12% showed a weak significant upward trend for PRE. The WDs of the aeolian sand zone and the sand, gravel, and silt accumulation zone were most heavily affected by the new increased land use from 2001 to 2019, accounting for 43.14% and 25.56% of the total WD of the watershed, respectively. Specifically, the WD of the aeolian sand zone caused by the new increased grassland and farmland in 2019 accounted for 41.92% and 18.52% of the total WD of the zone, respectively. The WD of the sand, gravel, and silt accumulation zone caused by the new increased grassland and farmland in 2019 accounted for 37.07% and 35.59% of the total WD of the zone, respectively. The WD caused by the new increased land use was increased by 7.78 million m3 in 2019 compared with the corresponding land use type in 2001, which would decrease the water yield. It is necessary to strengthen the protection of regional forest ecosystems in the granite and terrigenous clastic rock zone; standardize pasture management and reduce farmland reclamation in the sand, gravel, and silt accumulation zone, the aeolian sand zone, and the basalt platform zone; and reduce unnecessary impervious land construction in the aeolian sand zone.

Early reperfusion into the myocardium after ischemia causes myocardial ischemia–reperfusion (I/R) injury and ferroptosis was involved. Ischemia activates the expression of a series of oxidative stress genes and their downstream regulatory genes, including ferroptosis-related genes such as nuclear factor E2-related factor 2 (Nrf2), glutathione peroxidase 4 (GPX4), and SLC7A11. This study adopted primary cardiomyocytes and I/R in rats to evaluate the ferroptosis and changing of Nrf2-SLC7A11/heme oxygenase-1 (HO-1) in vitro and in vivo. Online analysis tools were used to predict the possible target Kelch-like ECH-associated protein 1 (Keap1) of miR-432-5p. The mimic of miR-432-5p plasmid was constructed to verify the effect of miR-432-5p on ferroptosis. We found that hypoxia/reoxygenation (H/R) in cardiomyocytes and I/R in rats induced lipid peroxidation and ferroptosis in cardiomyocytes. The activation of the Nrf2-SLC7A11/HO-1 pathway protects cardiomyocytes from ferroptosis. Downregulation of miR-432-5p has been confirmed in H/R cardiomyocytes (in vitro) and cardiomyocytes in myocardial infarction rats (in vivo). Upregulation of miR-432-5p inhibited ferroptosis of cardiomyocytes induced by RAS-selective lethal 3 (RSL3), an inhibitor of GPX4 and ferroptosis inducer through decreasing the binding protein of Nrf2, Keap1, which was confirmed by bioinformatics and mutation assay. Knockdown Nrf2 attenuates the protection effect of miR-432-5p on H/R cardiomyocytes. Intravenous delivery of liposome carriers of miR-432-5p remarkably ameliorated cardiomyocyte impairment in the I/R animal model. In conclusion, miR-432-5p inhibits the ferroptosis in cardiomyocytes induced by H/R by activating Nrf2/SLC7A11 axis by degrading Keap1 and is a potential drug target for clinical myocardial infarction treatment.

The demand for lithium resources is increasing sharply with the rapid development of electric vehicles. It is of great economic significance to expand the geological resources of lithium and improve the utilization rate of lithium-containing salt lakes. In this paper, the hydrochemical types of the lithium-containing salt lakes in the Tibet Plateau were classified according to a large amount of hydrochemical data obtained from a recent investigation on the Tibet Plateau. In addition, the lithium extraction methods used in the salt lakes within each hydrochemical type area were analyzed and summarized, which provided a reference for the selection of lithium extraction processes in the same hydrochemical type of lithium-containing salt lakes in the future. The binding energies of Li(Ι) and anions in salt lakes with different hydrochemical types were calculated by density functional theory, which provides the theoretical basis for selecting the best lithium extraction technology in different salt lakes. We emphasize that the process with the combined characteristics of high efficiency, economy and environmental protection should be selected according to the hydrochemical type of different salt lakes. In the future, different salt lakes should focus on direct lithium extraction technology from the original brine.

Abstract Background Patients with hepatitis C virus (HCV) frequently remain at risk for cirrhosis after sustained virologic response (SVR). Existing cirrhosis predictive models for HCV do not account for dynamic antiviral treatment status and are limited by fixed laboratory covariates and short follow up time. Advanced fibrosis assessment modalities, such as transient elastography, remain inaccessible in many settings. Improved cirrhosis predictive models are needed. Methods We developed a laboratory-based model to predict progression of liver disease after SVR. This prediction model used a time-varying covariates Cox model adapted to utilize longitudinal laboratory data and to account for antiretroviral treatment. Individuals were included if they had a history of detectable HCV RNA and at least 2 AST-to-platelet ratio index (APRI) scores available in the national Veterans Health Administration from 2000 to 2015, Observation time extended through January 2019. We excluded individuals with preexisting cirrhosis. Covariates included baseline patient characteristics and 16 time-varying laboratory predictors. SVR, defined as permanently undetectable HCV RNA after antiviral treatment, was modeled as a step function of time. Cirrhosis development was defined as two consecutive APRI scores > 2. We predicted cirrhosis development at 1-, 3-, and 5-years follow-up. Results In a national sample of HCV patients (n = 182,772) with a mean follow-up of 6.32 years, 42% (n = 76,854) achieved SVR before 2016 and 16.2% (n = 29,566) subsequently developed cirrhosis. The model demonstrated good discrimination for predicting cirrhosis across all combinations of laboratory data windows and cirrhosis prediction intervals. AUROCs ranged from 0.781 to 0.815, with moderate sensitivity 0.703–0.749 and specificity 0.723–0.767. Conclusion A novel adaptation of time-varying covariates Cox modeling technique using longitudinal laboratory values and dynamic antiviral treatment status accurately predicts cirrhosis development at 1-, 3-, and 5-years among patients with HCV, with and without SVR. It improves upon earlier cirrhosis predictive models and has many potential population-based applications, especially in settings without transient elastography available.

With the rapid development of wearable electronic products and smart devices, there is an urgent need to develop efficient electromagnetic interference (EMI)shielding materials to eliminate the resulting radiation pollution. Herein, hydrophobic and multi-responsive conductive composite film with core-shell structure and sandwich structure are constructed via simple in-situ polymerization and dip coating methods. The thin silicone coating can enhance the bonding fastness of the substrate and the conductive functional layer and endow the composite film excellent waterproof performance. The composite film with a thickness of 45 μm exhibits outstanding electrical conductivity of 62.15 S/cm and EMI shielding effectiveness (EMI SE) of 37.71 dB. Surprisingly, the composite film also exhibits good waterproof performance and excellent electro-optical thermal response, and has good stability in complex environments. This work proves the advantages of constructing multiple conductive interfaces inside the nanofiber film, and provides a new preparation strategy for efficient multifunctional shielding materials, which is expected to be widely used in the next generation of wearable electronic devices and wearable smart clothing.

Eosinophilic asthma is the predominant phenotype of asthma, and although these patients are sensitive to glucocorticoid therapy, they also experience many side effects. Lonicerin is a kind of bioflavonoid isolated from the Chinese herb Lonicera japonica Thunb, which has anti-inflammatory and immunomodulatory effects. The aim of this study was to elucidate the effects of lonicerin on eosinophilic asthma and its potential mechanisms. Here, we established a house dust mite (house dust mite)-induced eosinophilic asthma model in BALB/c mouse, and evaluated the effects of lonicerin on it. Our results showed that lonicerin significantly reduced airway hyperresponsiveness the number of inflammatory cells (especially eosinophils) and the elevation of interleukin (IL)-4, IL-5, IL-13 and eotaxin in bronchoalveolar lavage fluid (BALF) supernatants of mice. Additionally, lonicerin also eminently blunted inflammatory infiltration and mucus secretion, as well as mRNA levels of Mucin 5AC (MUC5AC) in lung tissue. Furthermore, results of network pharmacology and molecular docking revealed that Src kinase and epidermal growth factor receptor may be the potential targets responsible for the effects of lonicerin. Finally, in vivo experiments confirmed that lonicerin inhibited activation of the Src/EGFR pathway by decreasing their phosphorylation. Taken together, the present study demonstrated that lonicerin could suppress HDM-induced eosinophilic asthma in mice through inhibiting the activation of Src/EGFR pathway, which also provides a basis for further research as a new potentially therapeutic agent for eosinophilic asthma and its underlying mechanisms in the future.

In a study funded by the U.S. Department of Energy (DOE) National Energy Technology Laboratory (NETL), the University of North Dakota (UND) Energy and Environmental Research Center (EERC) performed a first-of-its-kind round-robin interlaboratory study (RRIS) to determine lab-to-lab and method-to-method variability in analyzing the rare earth element (REE) content of domestic resources. Analyses of REEs on eleven different materials were accomplished by the laboratories using four different procedures: ASTM D6357 (Procedure A), ASTM D4503 (Procedure B), an alternate in-house procedure for digestion and REE analysis (Procedure C), and neutron activation analysis (NAA). The results of the RRIS suggest that NAA is the most accurate and reliable method for many of the REEs in these types of materials; however, the method is limited in that it is able to determining only ten of the sixteen REEs. Five of the seven labs reporting data for Procedure A and three of the five labs reporting for Procedure B showed excellent performance in terms of repeatability, reproducibility, agreement with NAA, and SRM recoveries based on AOAC International guidelines on method performance. This indicates that when strictly followed these methods are suitable for REE determination in most materials, although are subject to the overall capabilities and experience of individual laboratories.

Mg-3Sn-1Mn-xLa alloy bars were prepared using backward extrusion, and the effects of the La content on the microstructures and mechanical properties of the alloy were systematically studied using an optical microscope (OM), X-ray diffraction (XRD), scanning electron microscope (SEM), transmission electron microscope (TEM), and tensile tests. The results of this research show that the Mg2Sn phases were mainly formed at the α-Mg grain boundaries and within the grains in the Mg-3Sn-1Mn alloy. After adding a certain amount of La, the plate-shaped MgSnLa compounds consisting of Mg17La2, Mg2Sn, and La5Sn3 gradually disappeared in the α-Mg matrix and grain boundaries. With an increase in La content, the Mg2Sn phase in the crystal was gradually refined and spheroidized. When the content of La reached 1.5%, the tensile strength of the alloy reached 300 Mpa and the elongation reached 12.6%, i.e., 25% and 85% increases, respectively, compared to the Mg-3Sn-1Mn alloy. The plate-shaped compound of Mg-3Sn-1Mn-1.5La had an average length of 3000 ± 50 nm, while the width was 350 ± 10 nm. Meanwhile, the extruded alloy’s grain size was significantly refined, and there were many small cleavage steps and dimples in the fracture surface of the alloy. When the La content reached 2%, the alloy performance showed a downward trend due to the coarsening of the grains. The formed plate-shaped MgSnLa compounds and Mg2Sn phases were consistent with the α-Mg matrix. They effectively pinned the dislocations and grain boundaries, which is the main reason for strengthening the mechanical properties of extrusion alloys.

Abstract Background Acinetobacter baumannii is a common nosocomial pathogen that poses a huge threat to global health. Owing to the severity of A. baumannii infections, it became necessary to investigate the epidemiological characteristics of A. baumannii in Chinese hospitals and find the reasons for the high antibiotic resistance rate and mortality. This study aimed to investigate the epidemiologic and genetic characteristics of A. baumannii isolated from patients with hospital acquired pneumonia (HAP), bloodstream infection (BSI) and urinary tract infection (UTI) in China and uncover potential mechanisms for multi-drug resistance and virulence characteristics of A. baumannii isolates. Results All isolates were classified into two primary clades in core gene-based phylogenetic relationship. Clonal complex 208 (CC208) mainly consisted of ST195 (32 %) and ST208 (24.6 %). CC208 and non-CC208 isolates had carbapenem resistance rates of 96.2 and 9.1 %, respectively. Core genes were enriched in ‘Amino acid transport and metabolism’, ‘Translation’, ‘Energy production and conversion’, ‘Transcription’, ‘Inorganic ion transport and metabolism’ and ‘Cell wall/membrane/envelope synthesis’. Most isolates possessed virulence factors related to polysaccharide biosynthesis, capsular polysaccharide synthesis and motility. Eleven isolates belong to ST369 or ST191 (oxford scheme) all had the virulence factor cap8E and it had a higher positive rate in UTI (35.3 %) than in BSI (18.9 %) and HAP (12.9 %). ABGRI1 antibiotic resistance islands were responsible for streptomycin, tetracycline and sulfonate resistance. The bla OXA−23 gene was the most probable cause for carbapenem resistance, although the bla OXA−66 gene with nonsynonymous SNPs (F82L, I129L) was not. Conclusions A. baumannii is a genomically variable pathogen that has the potential to cause a range of infectious diseases. There is high proportion of carbapenem resistance in isolates from all three infection sites (HAP, BSI and UTI), which can be attributed to the bla OXA−23 gene. CC208 is the predominant clone in bla OXA−23 -carrying A. baumannii that should be monitored. Virulence factors involving bacteria motility and polysaccharide biosynthesis which are widespread in clinical A. baumannii strains deserve our attention.

ObjectiveThe purposes of this study are to explore (1) whether comorbid depressive symptoms in patients with chronic back pain (CBP) affect the pain matrix. And (2) whether the interaction of depression and CBP exacerbates impaired brain function.MethodsThirty-two patients with CBP without comorbid depressive symptoms and thirty patients with CBP with comorbid depressive symptoms were recruited. All subjects underwent functional magnetic resonance imaging (fMRI) scans. The graph theory analysis, mediation analysis, and functional connectivity (FC) analysis were included in this study. All subjects received the detection of clinical depressive symptoms and pain-related manifestations.ResultCompared with the CBP group, subjects in the CBP with comorbid depressive symptoms (CBP-D) group had significantly increased FC in the left medial prefrontal cortex and several parietal cortical regions. The results of the graph theory analyses showed that the area under the curve of small-world property (t = −2.175, p = 0.034), gamma (t = −2.332, p = 0.023), and local efficiency (t = −2.461, p = 0.017) in the CBP-D group were significantly lower. The nodal efficiency in the ventral posterior insula (VPI) (t = −3.581, p = 0.0007), and the network efficiency values (t = −2.758, p = 0.008) in the pain matrix were significantly lower in the CBP-D group. Both the topological properties and the FC values of these brain regions were significantly correlated with self-rating depression scale (SDS) scores (all FDR corrected) but not with pain intensity. Further mediation analyses demonstrated that pain intensity had a mediating effect on the relationship between SDS scores and Pain Disability Index scores. Likewise, the SDS scores mediated the relationship between pain intensity and PDI scores.ConclusionOur study found that comorbid depressive symptoms can aggravate the impairment of pain matrix function of CBP, but this impairment cannot directly lead to the increase of pain intensity, which may be because some brain regions of the pain matrix are the common neural basis of depression and CBP.

Objective: Automatic detection of auditory stimuli, represented by the mismatch negativity (MMN), facilitates rapid processing of salient stimuli in the environment. The amplitude of MMN declines with ageing. However, whether automatic detection of auditory stimuli is affected by visually perceived negative emotions with normal ageing remains unclear. We aimed to evaluate how fearful facial expressions affect the MMN amplitude under ageing.Methods: We used a modified oddball paradigm to analyze the amplitude of N100 (N1) and MMN in 22 young adults and 21 middle-aged adults.Results: We found that the amplitude of N1 elicited by standard tones was smaller under fearful facial expressions than neutral facial expressions and was more negative for young adults than middle-aged adults. The MMN amplitude under fearful facial expressions was greater than neutral facial expressions, but the amplitude in middle-aged adults was smaller than in young adults.Conclusion: Visually perceived negative emotion promotes the extraction of auditory features. Additionally, it enhances the effect of auditory change detection in middle-aged adults but fails to compensate for this decline with normal ageing.Significance: The study may help to understand how visually perceived emotion affects the early stage of auditory information processing from an event process perspective.

Klebsiella pneumoniae is a nosocomial opportunistic pathogen that can cause pneumonia, liver abscesses, and infections of the bloodstream. The resistance and pathogenicity of K. pneumoniae pose major challenges to clinical practice. However, the ecology and pathogenic mechanisms of K. pneumoniae have not been fully elucidated. Among these mechanisms, the secretion systems encoded by strains of the bacteria confer adaptive advantages depending on the niche occupied. The type VI secretion system (T6SS) is a multi-protein complex that delivers effector proteins to the extracellular environment or directly to eukaryotic or prokaryotic cells. T6SSs are widely distributed in Gram-negative bacteria and play an important role in bacterial virulence and the interactions between bacteria and other microorganisms or the environment. This study aimed to enhance the understanding of the characteristics of T6SSs in K. pneumoniae through an in-depth comparative genomic analysis of the T6SS in 241 sequenced strains of K. pneumoniae. We identified the T6SS loci, the synteny of the loci in different species, as well as the effectors and core T6SS-related genes in K. pneumoniae. The presence of a T6SS was a common occurrence in K. pneumoniae, and two T6SS clusters are the most prevalent. The variable region downstream of the gene vgrG usually encodes effector proteins. Conserved domain analysis indicated that the identified putative effectors in K. pneumoniae had the functions of lipase, ribonuclease, deoxyribonuclease, and polysaccharide hydrolase. However, some effectors did not contain predicted functional domains, and their specific functions have yet to be elucidated. This in silico study represents a detailed analysis of T6SS-associated genes in K. pneumoniae and provides a foundation for future studies on the mechanism(s) of T6SSs, especially effectors, which may generate new insights into pathogenicity and lead to the identification of proteins with novel antimicrobial properties.

The diversification of B-cell receptor (BCR), as well as its secreted product, antibody, is a hallmark of adaptive immunity, which has more specific roles in fighting against pathogens. The antibody diversification is from recombination-activating gene (RAG)-initiated V(D)J recombination, activation-induced cytidine deaminase (AID)-initiated class switch recombination (CSR), and V(D)J exon somatic hypermutation (SHM). The proper repair of RAG- and AID-initiated DNA lesions and double-strand breaks (DSBs) is required for promoting antibody diversification, suppressing genomic instability, and oncogenic translocations. DNA damage response (DDR) factors and DSB end-joining factors are recruited to the RAG- and AID-initiated DNA lesions and DSBs to coordinately resolve them for generating productive recombination products during antibody diversification. Recently, cohesin-mediated loop extrusion is proposed to be the underlying mechanism of V(D)J recombination and CSR, which plays essential roles in promoting the orientation-biased deletional end-joining . Here, we will discuss the mechanism of DNA damage repair in antibody diversification.

Immunoglobulin class switch recombination (CSR) plays an important role in humoral imm\une responses by changing the effector functions of antibodies. CSR occurs between highly repetitive switch (S) sequences located upstream of immunoglobulin constant gene exons. Switch sequences differ in size, the nature of their repeats, and the density of the motifs targeted by the activation-induced cytidine deaminase (AID), the enzyme that initiates CSR. CSR involves double-strand breaks (DSBs) at the universal Sµ donor region and one of the acceptor S regions. The DSBs ends are fused by the classical non-homologous end-joining (C-NHEJ) and the alternative-NHEJ (A-NHEJ) pathways. Of the two pathways, the A-NHEJ displays a bias towards longer junctional micro-homologies (MHs). The Sµ region displays features that distinguish it from other S regions, but the molecular basis of Sµ specificity is ill-understood. We used a mouse line in which the downstream Sγ3 region was put under the control of the Eµ enhancer, which regulates Sµ, and analyzed its recombination activity by CSR-HTGTS. Here, we show that provision of Eµ enhancer to Sγ3 is sufficient to confer the recombinational features of Sµ to Sγ3, including efficient AID recruitment, enhanced internal deletions and robust donor function in CSR. Moreover, junctions involving Sγ3 display a bias for longer MH irrespective of sequence homology with switch acceptor sites. The data suggest that the propensity for increased MH usage is an intrinsic property of Sγ3 sequence, and that the tandem repeats of the donor site influence the choice of the A-NHEJ.

Post-collisional potassic-ultrapotassic volcanic rocks are widely developed in the Lhasa block, Qinghai-Tibet region. In this paper, it was observed for the first time that dilute alkali elements–Li, Rb, and Cs–exhibit supernormal enrichment in the research data of numerous potassic-ultrapotassic volcanic rocks in the Lhasa block. At the same time, there are few studies on the genesis of enrichment. Currently, 568 sets of published volcanic rock data and 8 sets of measured data in the Lhasa block are sorted in detail, and the genesis of dilute alkali element enrichment is explained by means of geochemical research methods. It is believed that the high degree of magmatic fractionation of potassic-ultrapotassic volcanic rocks in the Lhasa block is the main reason for the abnormal enrichment of dilute alkali elements such as Li, Rb, and Cs. The abnormal enrichment area is mainly located in the central and western parts of the Lhasa block with an age range of 25–13 Ma. The discrimination range of Zr/Hf and Nb/Ta with a high degree of differentiation of potassic-ultrapotassic volcanic rocks in the Lhasa block is divided by analogy with the research results of highly fractionated granites, which provides a reference for the study of the supernormal enrichment of dilute alkali elements in potassic-ultrapotassic volcanic rocks in this area.

IMP-45, a variant of IMP-9, is one of the dominant metallo-β-lactamases (MBLs) in clinical carbapenem-resistant Pseudomonas aeruginosa (CRPA) isolates in China. The aim of this study was to investigate the distribution and mechanism of dissemination of blaIMP-45. MBL genes were detected by PCR in 173 non-duplicate CRPA isolates collected from Hospital HS in Shanghai and 605 P. aeruginosa isolates from a multicenter surveillance of blaIMP-45 in China. In total, 17 IMP-45-producers (14 from Hospital HS and 3 from other hospitals) were identified. Molecular typing identified an outbreak of 11 IMP-45-producing ST508 CRPA in the ICU of Hospital HS. Conjugation assays and whole genome sequencing were conducted among IMP-45-producers. Genomic comparison revealed that 16 blaIMP-45-carrying plasmids (9 from this study and 7 from GenBank) shared a similar backbone with IncP-2 blaIMP-9-carrying plasmid pOZ176 but lacked repA-oriV-parAB region. repA2 gene was presented in pOZ176, blaIMP-45-carrying plasmids (17 from this study and 7 from GenBank) and 15 megaplasmids from GenBank. Phylogenetic analysis of repA2 showed that most blaIMP-45-carrying plasmids were clustered into a sublineage separate from the one containing pOZ176. This IncP-2 plasmid sublineage contributed to the dissemination of blaIMP-45 among genetically diverse P. aeruginosa and recruited multiple resistance genes during its evolution.

Plague, caused by Yersinia pestis, is a natural focus infectious disease. In China, plague is classified as category A, with the highest risk and hazard among the infectious diseases. Qinghai used to be considered as one of the most serious areas of plague in China. In recent years, thank to the measures in eight aspects summarized as the “Qinghai model” which were adopted to prevent and control the human plague in Qinghai, Qinghai has not experienced any plague case reported for eight years. In early 2020, coronavirus disease 2019 (COVID-19) outbroke in China. The Qinghai model on plague was employed to deal with the COVID-19 emergency in Qinghai Province. The Qinghai Center for Disease Control and Prevention (Qinghai CDC) and hospitals, along with the departments of public security, animal husbandry and other departments, quickly tracked and treated the patients with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and started surveillance programs on close contacts timely. At present, the cure rate of patients has reached 100%, and close contacts have been effectively quarantined and tested to avoid the spread of COVID-19. The findings from the study suggest that the prevention and control measures undertaken in Qinghai Province might be effective in dealing with the category A infectious diseases such as COVID-19 and other diseases.

The Gudui geothermal field records the highest temperature at equivalent borehole depths among the mainland hydrothermal systems in mainland China. Located about 150 ​km southeast of Lhasa City, the capital of Tibet, the Gudui geothermal field belongs to the Sangri–Cuona rift belt, also known as the Sangri–Cuona geothermal belt, and is representative of the non-volcanic geothermal systems in the Himalayas. In this study, oxygen-18 and deuterium isotope compositions as well as 87Sr/86Sr ratios of water samples collected from the Gudui geothermal field were characterized to understand the origin and mixing processes of the geothermal fluids at Gudui. Hydrogen and oxygen isotope plots show both, deep and shallow reservoirs in the Gudui geothermal field. Deep geothermal fluids are the mixing product of magmatic and infiltrating snow-melt water. Calculations show that the magma fluid component of the deep geothermal fluids account for about 21.10%–24.04%; magma fluids may also be a contributing source of lithium. The linear relationship of the 87Sr/86Sr isotopic ratio versus the 1/Sr plot indicates that shallow geothermal fluids form from the mixing of deep geothermal fluids with cold groundwater. Using a binary mixing model with deep geothermal fluid and cold groundwater as two end-members, the mixing ratios of the latter in most surface hot springs samples were calculated to be between 5% and 10%. Combined with basic geological characteristics, hydrogen and oxygen isotope characteristics, strontium concentration, 87Sr/86Sr ratios, and the binary mixing model, we infer the 6th-Class Reservoirs Evolution Conceptual Model (6-CRECM) for the Gudui geothermal system. This model represents an idealized summary of the characteristics of the Gudui geothermal field based on our comprehensive understanding of the origin and mixing processes of the geothermal fluid in Gudui. This study may aid in identifying the geothermal and geochemical origin of the Gudui high-temperature hydrothermal systems in remote Tibet of China, whose potential for geothermal development and utilization is enormous and untapped.

Objectives: The rise of carbapenem resistance in Acinetobacter baumannii represents a challenge for the therapeutic management of infections. The present study aimed to investigate the sequence types (STs) and carbapenem resistance in A. baumannii strains collected from various clinical specimens from patients admitted to five tertiary-care hospitals in Pakistan. Methods: A total of 156 A. baumannii clinical strains were analysed for antimicrobial susceptibility, followed by genetic screening for carbapenem resistance determinants. All of the strains were typed by multilocus sequence typing (MLST) according to the Pasteur scheme. Results: Of the 156 A. baumannii isolates, 139 (89.1%) were carbapenem-resistant, of which 136 carried blaOXA-23-like genes. Interestingly, the most commonly identified ST was ST589 (n = 52), classified as clonal complex 1 (CC1). ST2 was the second most common (n = 38), corresponding to CC2/92 (Pasteur/Oxford scheme), which was distributed in all five hospitals. Conclusion: Diverse clones of carbapenem-resistant A. baumannii, including previously reported STs as well as new STs, carrying blaOXA-23 are distributed in Pakistan. This is the first study to describe the molecular epidemiology of widely disseminated A. baumannii isolates in Pakistan. The findings will help to improve our knowledge of the predominant STs and will be valuable for a deeper understanding of resistance mechanisms among various STs.

Abstract Background Alveolar echinococcosis (AE) is a zoonotic parasitic disease caused by Echinococcus multilocularis larval tapeworm infections in humans that severely impairs the health of affected patients in the northern hemisphere. Methods The expression levels of 20 cytokines associated with AE infection were measured by enzyme-linked immunosorbent assay, and the correlations between these cytokines were analysed in the R programming language. Results Serum cytokine levels differed among individuals in both the AE patient and healthy control groups. The results of the correlations among the cytokines showed obvious differences between the two groups. In the AE patients group, Th1 and Th2 cytokines formed a more complicated network than that in the healthy control group. Conclusions The altered correlations between Th1 and Th2 cytokines may be closely associated with AE infection, which may provide a new explanation for the essential differences between AE patients and healthy individuals.

Wireless networks have been generating a plethora of unstructured and highly-correlated big data with hidden anomalies. The anomalies may bring inaccurate predictions of network behaviors, which further lead to inefficient system designs such as proactive caching placement. Current Random Matrix Theory (RMT) approaches are unable to detect hidden anomalies with a satisfying tolerance of data correlation. We present a novel data Decomposition aided Random Matrix Theory (DC-RMT) framework, which enables a real-time anomaly detection of large scale multi-dimensional and highly-correlated data. The detection results show that the proposed DC-RMT methodology can detect anomalies with an accuracy of 28 times better than RMT applied without data decomposition. The prediction results present a 6 times higher accuracy than data with anomaly, which will facilitate the identification of regions of interests, and contribute to the improvement of resource allocation efficiency and user QoE.
