A dual Z-scheme NiO/NiFe2O4/Fe2O3 photocatalyst is prepared via incomplete solid state chemical combustion reaction of Ni(OH)2 and Fe(OH)3. The formed perfect interfaces between NiO and NiFe2O4 and between NiFe2O4 and Fe2O3 facilitate the transfers of photo-induced electrons. The photocatalytic degradation of methylene blue and simultaneous production of hydrogen was performed to evaluate the activity of the prepared samples. The dual Z-scheme NiO/NiFe2O4/Fe2O3 (600–2) photocatalyst obtained by heat treatment of Ni(OH)2 and Fe(OH)3 at 600 °C for 2.0 h shows an excellent photocatalytic performance. Additionally, the influences of simulated sunlight irradiation time and methylene blue concentration on the photocatalytic reactions are investigated. Besides, the reusability of sample is assessed via four cycle experiments. Further, a possible mechanism on the photocatalytic reaction is proposed. Maybe, this work would provide an ingenious idea for the construction of dual Z-scheme photocatalyst and the exploration for photocatalytic degradation of organic pollutants with simultaneous hydrogen production.

The metabolic product of type I collagen synthesis, intact procollagen type I N propeptide (intact PINP), is a potential marker of bone formation and osteoporosis, which is not affected by kidney function. We sought to establish a chemiluminescent immunoassay method for the detection of serum intact PINP with previously prepared paired monoclonal antibodies and to evaluate the diagnostic value of the assay in osteoporosis. Using the capture molecule and monoclonal antibody as detection molecule, a diagnostic reagent was developed to detect intact PINP in serum with magnetic nanosphere carriers by the chemiluminescence method, and its analytical performance in the laboratory was evaluated. Serum intact PINP was measured in 142 healthy people and 115 osteoporosis patients. Results were matched with results of a similar test kit, Roche total PINP Elecsys Chemiluminescent Immunoassay Assay. Compared with the performance of the Roche PINP assay product, our method had higher sensitivity (0.02 ng/mL), wider linear range (0.02-1500 ng/mL), and anti-interference. Serum intact PINP values in osteoporosis patients were significantly higher than in healthy subjects (p < 0.001). Our method had good consistency compared with the Roche PINP assay (r = 0.9794). This chemiluminescence method for detecting serum intact PINP (CLIA-intact PINP) with magnetic nanosphere carrier technology meets the requirements of a clinical testing reagent and is expected to have clinical application after further evaluation and can compete with expensive imported kits on the market.

The objective of this work is to develop and verify the analytical performance of a chemiluminescence immunoassay for the specific sequence β-carboxy-terminal cross-linking telopeptide of type I collagen (β-CTX) in human serum. Two specific monoclonal antibodies (mAb-8A03 and mAb-3D12) with high affinity for β-CTX were selected, and, under optimized conditions, a chemiluminescence immunoassay method (CLIA) for β-CTX was established. The CLIA of β-CTX detected β-CTX in a wide range of 2.0-6000 ng/L. The recovery rate in serum is 95-105%, the specificity is high, and the cross-reaction rate with common easily interfering substances is low (not more than 0.01%). The CLIA correlates well with Roche electrochemiluminescence immunoassay (ECLIA), with a correlation coefficient of 0.9551, which fully meets the requirements of clinical analysis. The developed β-CTX CLIA kit has high sensitivity and good stability. It has the same performance as the commercial Roche ECLIA kit and can be applied clinically.

Heavy metals may coexist with pesticides in farmland through wastewater irrigation, application of pesticides and chemical fertilizers, or unappropriated waste disposal. Heavy metals are toxic to soil microorganism, which may influence the environmental behavior of pesticides subsequently. In this study, the influence of Cd2+ and Pb2+ on the degradation of α-cypermethrin and its metabolites, 3-phenoxphenoxybenzoic acid (3-PBA) and 3-(2’,2’-dichlorovinyl)-2,2-dimethylcyclopropane carboxylic acid (DCCA), were investigated through soil incubation experiment. It was found heavy metals like Cd2+ and Pb2+ will inhibit the degradation of α-cypermethrin, especially at high concentrations. Pb2+ has a stronger inhibitory effect on the degradation of α-cypermethrin than Cd2+ in the same concentration. With the presence of 10 mg/kg Pb2+, the half-life of α-cypermethrin increased from 41.1 to 99.9 days, even the half-life was 129.3 days with 50 mg/kg of Pb2+. Besides, heavy metals influenced the chiral selective degradation of α-cypermethrin. The enantiomer fraction was near 0.5 when 10 mg/kg of heavy metals existed. Furthermore, the adverse effects of heavy metals on soil urease, catalase, and sucrase activity were assayed. In tested concentrations (10 and 50 mg kg-1), the heavy metals result in strong inhibition of the activity of the enzymes present on soil, jeopardizing the biodegradation by the microbiome and which may inhibit the degradation of α-cypermethrin.

The miR let-7b-5p (a kind of microRNAs) has many pathophysiological regulation effects, including human hepatocellular carcinoma (HCC) pathogenesis. This study investigated whether nanoparticle-mediated miR let-7b-5p could jointly enhance the therapeutic effect of sorafenib on HCC by inhibiting the proliferation of HCC cells, inducing apoptosis, and reversing drug resistance. We evaluated the level of miR let-7b-5p in sorafenib-resistant HepG2 cells (HepG2R) and HepG2 HCC cells by qRT-PCR and analyzed the biological effects of hepatocellular carcinoma treated with sorafenib with miR let-7b-5p, and further studied the toxicity of nanoparticles (Ab-miR-NPs) that deliver miR let-7b-5p mimics and target GPC3 on the surface of hepatocellular carcinoma cells. Results showed that, in HepG2 cells, the expression level of miR let-7b-5p was significantly higher than that in HepG2R cells. Targeted nanoparticle Ab-miR-NPs mediated the delivery of miR let-7b-5p to the HCC cytoplasm and released miRNA after being broken down, down-regulating the expression of IGF1R and inhibiting AKT/mTOR and Ras/Raf signal transmission. Ab-miR-NPs not only enhanced the proliferation of sorafenib in cultured HepG2R cells and induced cell apoptosis efficiency, but they also improved the anti-tumor activity in the mouse models. These results indicated that GPC3 antibody-modified PLGA-PLL (polylactic acid-glycolic acetic copolymer grafted hyper-branched polylysine) loaded miR let-7b-5p polymer nanoparticles combined with sorafenib may be a new treatment strategy for HCC resistant to sorafenib.

Hepatocellular carcinoma (HCC) is the sixth most common cancer in the global. It could be a potential resistant mechanism that paclitaxel activated the PI3K/mTOR and MAPK pathway in HCC cells. So inhibiting the abnormal activation of two pathway, which may a promising targeted therapy strategy. MTT and clone formation assays were used to measure cells proliferation activity; cell cycle distribution and apoptosis were measured by flow cytometry; western blot assessed phosphorylation level of proteins associated with cell cycle, apoptosis, PI3K/mTOR and MAPK pathway; Student t test or one-way ANOVA to measure significant differences between the means.BEZ235 combined with HPG-PCL-PTX blocked the activation of PI3K/mTOR and MAPK pathway in HCC cells, effectively inhibited cells proliferation and arrested the cell cycle at the G2/M phase. The most important is that the combination of two drugs caused mitochondrial membrane potential change, induced cytochrome C release and bind to APAF-1 to initiate the caspase-9 apoptosis program, promoted HCC cell apoptosis. HPG-PCL-PTX/BEZ235 enhanced antitumor effectiveness in HepG2R cell xenograft mouse models. In vivo study suggested that BEZ235 combined with HPG-PCL-PTX enhanced effects of HPG-PCL-PTX through inhibited the PI3K/Akt/mTOR and MAPK pathway in HCC cells, supressed cells proliferation, arrested the cell cycle in a G2/M phase and promoted apoptosis. Antitumor effects of BEZ235 and HPG-PCL-PTX were also confirmed in mice bearing HepG2R tumors. Taken together, the results of this study describe a promising strategy using PTX and BEZ235 in a nanoparticle formulation for treatment of PTX-resistant HCC.

BackgroundIncidence of antimicrobial-resistant infections has increased dramatically worldwide in the past few decades. To combat this alarming problem, strategies preventing colonization with resistant pathogens have been proposed. Fecal microbiota transplantation(FMT)may help in decolonizing multidrug-resistant organisms(MRO) and in preventing recurrent infections. In this study, efficacy of FMT against multidrug resistance was assessed.MethodsThree patients that suffered from multidrug resistance complicated with diarrhea were enrolled from January 2019 to September 2020. All had severe pulmonary infections. Standard FMT was given, and whole metagenome sequencing of stool was conducted before and after FMT.ResultsAll patients accepted FMT, and 12 samples were harvested. All treatments were successful, and there were no adverse events. All patients showed a decrease in species of intestinal flora, but FMT could recover some of the diversity. Normal people had the lowest abundance of antimicrobial-resistant genes (ARGs), but the patients had the highest abundance before FMT. FMT decreased ARGs to some extent. Nineteen ARG subtypes were significantly different between the normal group and the patients.ConclusionsPatients with MRO infections had increased abundance of ARGs and low bacterial diversity. FMT eradicated ARGs and restored microbial diversity, and there were no serious adverse events.Trial registration: retrospectively registered.

Antibiotics could enter farmlands through sewage irrigation or manure application, causing combined pollution with pesticides. Antibiotics may affect the environmental fate of pesticides and even increase their bioavailability. In this study, the influence of monensin on the degradation, toxicity, and availability of atrazine in soil-earthworm microcosms was investigated. Monensin inhibited the degradation of atrazine, changed the metabolite patterns in soil, and increased the bioavailability of atrazine in earthworms. Atrazine and monensin had a significant synergistic effect on earthworms in the acute toxic test. In long-term toxicity tests, co-exposure of atrazine and monensin also led to worse effects on earthworms including oxidative stress, energy metabolism disruption, and cocoon production compared to single exposure. The expression of tight junction proteins was down-regulated significantly by monensin, indicating that the intestinal barrier of earthworms was weakened, possibly causing the increased bioavailability of atrazine. The expressions of heat shock protein 70 (Hsp70) and reproductive and ontogenetic factors (ANN, TCTP) were all downregulated in binary exposure, indicating that the resilience and cocoon production of earthworms were further weakened under combined pollution. Monensin disturbed the energy metabolism and weakened the intestinal barrier of earthworms. These results showed that monensin increased the risks of atrazine in agricultural areas.

With diminishing pristine water, wastewater-affected waters that contain complex anthropogenic compounds are becoming important sources of drinking water and the compounds will inevitably react with disinfectants to form disinfection byproducts (DBPs). Secondary amines such as diphenylamine (DPA) analogues are considered as potential precursors of N-nitrosamines. In this study, an in situ 14N/15N-labeling and screening workflow was used to systematically investigate the formation of nitrogenous DBPs (N-DBPs) and putative reaction pathways. Twenty-four pairs of N-DBPs were generated and identified from chloramination of DPA through two main pathways, in which chloramines reacted with the amino and phenyl functional groups to form N-nitrosodiphenylamine and monochlorinated 5,10-dihydro-phenazine (Cl-DiH-Phe), respectively. Cl-DiH-Phe could further produce phenazine and the coupling products with another DPA molecule. Selective N-DBP formation was pH and dose-dependent, and the same reactions were observed for additional two aromatic DPA analogues. Effects of alkyl substituents on the formation pathways were investigated using a series of dialkyl and N-alkyl aromatic analogues. Only the amino pathway to form nitrosamines was noticed for dialkyl amines, nevertheless, both the main reactions occurred for N-alkyl aromatic amines. These findings suggested that the reaction with chloramines through a phenyl pathway was likely to be crucial for novel nitrogenous heterocyclic byproducts.

Background Gefitinib is a first-generation epidermal growth factor receptor-tyrosine kinase inhibitor (EGFR-TKI) that has become first-line treatment for patients with mutant EGFR non-small cell lung cancer (NSCLC). Despite its anti-tumor activity, the benefit of gefitinib in patients with wild-type EGFR NSCLC is debated. This work aimed to evaluate the effects of gefitinib on cisplatin-resistant wild-type EGFR NSCLC cells in in vitro and in vivo animal xenografts. Methods We established a cisplatin-resistant wild-type EGFR NSCLC cell line, H358R, to evaluate the cells’ sensitivity to gefitinib compared with that of parental cell line H358. We first tested the p-EGFR of gefitinib’s target in H358R and H358 cell line by western blot and immunofluorescence. 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT), clone formation assay, flow cytometry and annexin V-fluorescein/propidium iodide staining were used to investigate cellular proliferation and apoptosis of H358R/H358 cells treated with gefitinib, and the anti-tumor effect was evaluated in female BALB/c nude mice models of xenografts in vivo. Results EGFR and the downstream node molecules ERK and AKT were significantly more phosphorylated in H358R than in the parental cells and were inhibited by gefitinib. In H358R cells, gefitinib increased the inhibition of cell survival/proliferation, and the promotion of apoptosis in vitro. The increased anti-tumor effect was present also in H358R xenografts in vivo. Conclusions Abnormal activation of EGFR in H358R cells results in enhanced sensitivity to gefitinib. The improved efficacy of gefitinib on cisplatin-resistant wild-type EGFR NSCLC cells suggests that gefitinib as sequential therapy for patients with cisplatin-resistant wild-type EGFR NSCLC should be considered.

The usual first-line strategy of wild-type EGFR (wtEGFR) non-small cell lung cancer (NSCLC) remains cisplatin-based chemotherapy. However, cisplatin often loses effectiveness because most tumors acquire drug resistance over time. As EGFR is the most important pro-survival/proliferation signal receptor in NSCLC cells, we aimed at investigating whether cisplatin resistance is related to EGFR activation and further evaluating the combined effects of cisplatin/gefitinib (EGFR-tyrosine kinase inhibitor, EGFR-TKI) on cisplatin-resistant wtEGFR NSCLC cells. EGFR activation was analysed in parental and cisplatin-resistant wtEGFR NSCLC cell lines (H358 and H358R, A549 and A549R). Cellular proliferation and apoptosis of H358R/A549R cells treated with cisplatin or gefitinib, alone or in combination were investigated, and the related effector protein was detected by western blot analysis. Anti-tumor effect of two drugs combined was evaluated in animal models of H358R xenografts in vivo. EGFR was significantly phosphorylated in cisplatin-resistant wtEGFR NSCLC cells H358R and A549R than their parental cells. In H358R and A549R cells, anti-proliferative ability of gefitinib was further improved, and gefitinib combined with cisplatin enhanced inhibition of cellular survive/proliferation, and promotion of apoptosis in vitro. The combined effects were also associated with the inhibition of EGFR downstream effector proteins. Similarly, in vivo, gefitinib and cisplatin in combination significantly inhibited tumor growth of H358R xenografts. Abnormal activation of EGFR may induce wtEGFR NSCLC cell resistance to cisplatin. The combined effects of cisplatin/gefitinib suggest that gefitinib, as a combination therapy for patients with cisplatin-resistant wtEGFR NSCLC should be considered.

Background: The combination of BEZ235 with sorafenib (SFB) enhances anti-hepatocellular carcinoma (HCC) efficacy of the two agents. However, pharmacokinetic profiles in vivo and different endocytosis abilities of these two drugs hinder their therapeutic application.Research design and methods: In this work, we developed d-α-tocopheryl polyethylene glycol 1000 succinate - polycaprolactone polymer nanoparticles (NPs) for co-delivery of SFB and BEZ235 (SFB/BEZ235-NPs). Explored the anti-proliferative and pro-apoptotic effects of SFB/BEZ235-NPs through in vitro and in vivo experiments.Results: Stabilized SFB/BEZ235-NPs were prepared with optimized drug ratio, yielding high encapsulation efficiency, low polydispersity, and enhanced cellular internalization in HepG2 cells. Synergistic cytotoxicity and pro-apoptotic ability were documented. In vivo pharmacokinetic results revealed extended circulation and bioavailability of SFB/BEZ235-NPs compared with those of free drugs. SFB/BEZ235-NPs enhanced antitumor effectiveness in SFB-resistant HCC xenograft mouse models.Conclusion: Taken together, the results of this study describe a promising strategy using SFB and BEZ235 in a nanoparticle formulation for treatment of SFB-resistant HCC.

Dietary intake is an essential way for pesticides to enter the human body. The effects of dietary pattern on the risks of pesticides and what diet can reduce the damage are largely unknown. Here, it is found that Mediterranean diet and Vegetarian diet could alleviate insulin resistance and obesity induced by chlorpyrifos, while Western diet could aggravate that. Gut microbiota and chlorpyrifos bioavailability mediated by the diets were involved in these effects. Both the dietary pattern and chlorpyrifos could change the composition of gut microbiota. Chlorpyrifos caused gut dysbacteriosis which was an important reason for the induced metabolic syndrome. Mediterranean diet and Vegetarian diet could maintain gut microbiota homeostasis and increase intestinal bacteria producing short-chain fatty acids, repair the gut microbiota and intestinal barrier damaged by chlorpyrifos. High dietary fat intake increased the bioavailability of chlorpyrifos, which aggravated the gut dysbacteriosis and destruction of intestinal integrity. Thus, the amount of endotoxin entering the blood increased and caused low-grade inflammation, which was also an important pathway of metabolic syndrome. The results suggested that although it was almost impossible to avoid the exposure to pesticides in modern life, healthy diets could regulate beneficial gut microbiota and alleviate the risk of pesticide exposure.

Pesticides are widely used and frequently detected in the environment. The evaluation on the toxic effects of the co-exposure of two or more pesticides or related metabolites could reflect the real situation of the exposing risks. In this study, zebrafish was used as a model to investigate the potential toxic interactions of chlorpyrifos and 1,1-dichloro-2,2-bis(p-chlorophenyl)ethylene (p,p'-DDE) on the survival rate, oxidative stress response and neurotoxicity, as well as their bioaccumulation and distribution in tissues. Farnesoid X receptor (FXR) is a bile acid-related nuclear receptor and is considered a promising target to treat several liver disorders. Cilofexor is a selective FXR agonist and has already entered phase III trials in primary sclerosing cholangitis (PSC) patients. Pruritis caused by cilofexor treatment is dose dependent. The binding characteristics of cilofexor with FXR and its pruritogenic mechanism remain unclear. In our research, the affinity of cilofexor bound to FXR was detected using an isothermal titration calorimetry (ITC) assay. The binding mechanism between cilofexor and FXR-LBD is explained by the cocrystal structure of the FXR/cilofexor complex. Structural models indicate the possibility that cilofexor activates Mas-related G protein-coupled receptor X4 (MRGPRX4) or G protein-coupled bile acid receptor 1 (GPBAR1), leading to pruritus. In summary, our analyses provide a molecular mechanism of cilofexor binding to FXR and provide a possible explanation for the dose-dependent pruritis of cilofexor.

Carbonyl compounds are an important class of by-products that are generated in disinfection reactions. These chemicals are ingredients contributing to toxicology in the drinking water system, the compositions and structures of which are worthy of attention. In this study, a chemical derivatization method based on simultaneous light/heavy isotope labeling was established for general recognition of carbonyl compounds and carbonyl disinfection by-products (DBPs) as per the humic substance reference standard (Suwannee river fulvic acid II, SRFA) before and after ozonation, chlorination, and chloramination. Decomposition of macromolecular components into polar carbonyl species was observed to be the most prominent pathway in ozone treatment due to the efficient reactivity of ozone with phenols and alkoxy aromatic rings. As a result, alteration of molecular characteristics was noticed. For instance, ozone-induced carbonyl DBPs in the highly oxygenated compound classes (0.67 ≤ O/C ≤ 1.2, 0.6 < H/C ≤ 1.5) possessed higher O/C but contained less oxygen numbers and carbon numbers. Cl/Br-carbonyl-DBPs were identified after chlorination and chloramination, and I-carbonyl-DBPs were found in ozone and chloramine treatments. Several major halogenated carbonyl homologues were further recognized, including halogenated 4-oxobutenoic acid analogues, halogenated 2,5-dioxohex-3-enoic acid analogues, and halogenated 4-cyclopentene-1,3-diones analogues. These findings illustrate the presence of abundant carbonyl DBPs in water disinfection, and hence their impacts on human health deserve further investigation.

Chlordane-related compounds are ubiquitously detected in the environment and can transfer and accumulate to animals through food chain to cause adverse effects. In this study, the dynamic distribution and the enantiomeric profile of chlordane-related compounds in laying hens over time were investigated. The effect of these compounds on immune-associated cells in the intestinal tract and histopathology in some tissues were also evaluated after long-term exposure. The chlordane-related compounds preferentially accumulated in fat, followed by the intestines, ovum, and egg yolk during long-term exposure. The metabolites heptachlor epoxide and oxychlordane were mainly formed in the liver of hens by epoxidation or hydroxylation. The high accumulation ratios of trans-nonachlor and MC5 were found in ovum and egg yolk after long-term exposure, implying a greater risk to the hens' offspring. Chlordane-related compounds may cause abnormal lipometabolism and glycometabolism in liver of hens. Additionally, (-)-Cis-chlordane was dominant in all tissues of laying hens and its dominance increased over time. Conversely, (+)-isomer of metabolite oxychlordane was overwhelmingly dominant during the experiment. These findings about enantioselectivity, metabolic processes and toxicological effects are crucial in understanding the exposure risk of chlordane-related compounds.

Labels or tapes are widely used on fresh fruits and vegetables, which may contain phthalates (PAEs). There are few studies on the contamination pathway of PAEs from labels or tapes to food stuff. In this study, the concentrations of eleven PAEs in adhesive labels, tapes, labeled fruits and vegetables on the market were investigated. The eleven PAEs were detected with the total concentration of 7.44–30.51 mg/m2 in labels and tapes. Diethyl phthalate (DEP), di-n-butyl phthalate (DIBP), dimethyl phthalate (DMP), Bis (2-ethylhexyl) phthalate (DEHP) and di-n-butyl phthalate (DBP) had the highest detection frequency in adhesive material samples. The concentrations of PAEs in labeled fruits were higher than that in unlabeled fruits, especially in the peel, indicating PAEs could transfer from labels or tapes to fruits and vegetables. Furthermore, the migration behaviors of PAEs from labels or tapes to apples, avocados and celery were investigated. It was found that the PAEs could penetrate to apple and avocado pulp through the peel, resulting in the residue of the PAEs in the whole fruit. Unlike apple peel, the thick avocado peel was more difficultly penetrated by the PAEs. Due to the high lipid content, the PAEs distributed more evenly in avocado pulp than in apple pulp. The migrations up to a maximum of 4.16 mg/kg were found for butyl benzyl phthalate (BBP) in avocado peel and up to a maximum of 0.188 mg/kg in avocado pulp. The average migration of the PAEs in celery ranged from 0.3 to 26.1 μg/kg in three days and the low migration might result from the rough surface and less contacting area. These findings suggest that the use of labels or tapes in direct contact may increase the risk of PAEs exposure to humans through fruits and vegetables.

A novel ionic liquid (IL) based on catalytic functional metal rhenium, [Smim][ReO4] (1-heptyl-3-methyl-imidazolium perrhenate) was synthesized and characterized. Density and surface tension values of the IL were determined at different temperatures, and the volume and surface properties were calculated and discussed, respectively. Furthermore, the synthesized ionic liquid [Smim][ReO4] was used as a green solvent and catalyst for homogeneous catalyzed epoxidation of olefin with urea hydrogen peroxide (UHP) oxidant. The effect of factors of catalyst, oxidant, reaction time, and reaction temperature was discussed. The conversion of cyclohexene and cyclooctene is over 99% at optimum conditions. The IL [Smim][ReO4] as catalyst and solvent are characterized by high efficiency, long service life and recoverability, which is a better green homogeneous catalyst for epoxidation of olefins. Easy separation, recycle, nontoxicity and homogeneous catalysis are the main advantages of perrhenate ionic liquids over other heterogeneous catalysts containing organic solvents.

Ionic liquids (ILs) as novel functional desulfurization materials have attracted increasing attentions. Metal-based ionic liquids (MILs) are classified into three types of metal chloride ILs, metal oxide ILs, and metal complex ILs based on the definition and basic structure of MILs in this critical review. On the basis of the properties of ILs such as structure designability, super dissolution performance, good thermal and chemical stability, nonflammability, and wide electrochemical window, MILs exhibit unique advantages on hydrophobicity, oxidation performance, and Bronsted-Lewis acidity. Therefore, MILs possess both the absorption and oxidation centers for the intramolecular adsorption and oxidation to improve the oxidative desulfurization (ODS) process. During the novel nonaqueous wet oxidative desulfurization process (Nasil), H2S can be oxidized into elemental sulfur with hydrophobic MILs, which can be regenerated by oxygen for recycle, to solve the problems of low sulfur capacity, low sulfur quality, and severe secondary pollution in the aqueous Lo-Cat wet oxidative desulfurization process. Another outstanding feature of MILs in ODS is biomimetic catalysis, which has the function of activating molecular oxygen and improving the oxidation performance. Metal oxide ILs and metal complex ILs are used in combination with hydrogen peroxide or oxygen with the existing water to generate a Fenton-like reaction to convert hydrophobic organic sulfur or SO2 into hydrophilic sulfoxide/sulfone or sulfur acid, respectively. However, the corrosion of Cl- to the equipment and emulsification phenomenon in the extraction process of sulfoxide/sulfone separation still need further study. Furthermore, the promising strategies to construct highly efficient and green desulfurization processes for large-scale applications are provided.

Acquired resistance of hepatocellular carcinoma (HCC) to sorafenib (SFB) is the main reason for the failure of SFB treatment of the cancer. Abnormal activation of the PI3K/AKT/mTOR pathway is important in the acquired resistance of SFB. Therefore, we investigated whether BEZ235 (BEZ) could reverse acquired sorafenib resistance by targeting the PI3K/mTOR pathway. A sorafenib-resistant HCC cell line Huh7R was established. MTT assay, clone formation assay, flow cytometry, and immunofluorescence were used to analyze the effects of BEZ235 alone or combined with sorafenib on cell proliferation, cell cycle, apoptosis, and autophagy of Huh7 and Huh7R cells. The antitumor effect was evaluated in animal models of Huh7R xenografts in vivo. Western blot was used to detect protein levels of the PI3K/AKT/mTOR pathway and related effector molecules. In vitro results showed that the Huh7R had a stronger proliferation ability and antiapoptosis effect than did Huh7, and sorafenib had no inhibitory effect on Huh7R. SFB + BEZ inhibited the activation of the PI3K/AKT/mTOR pathway caused by sorafenib. Moreover, SFB + BEZ inhibited the proliferation and cloning ability, blocked the cell cycle in the G0/G1 phase, and promoted apoptosis in the two cell lines. The autophagy level in Huh7R cells was higher than in Huh7 cells, and BEZ or SFB + BEZ further promoted autophagy in the two cell lines. In vivo, SFB + BEZ inhibited tumor growth by inducing apoptosis and autophagy. We concluded that BEZ235 enhanced the sensitivity of sorafenib through suppressing the PI3K/AKT/mTOR pathway and inducing autophagy. These observations may provide the experimental basis for sorafenib combined with BEZ235 in trial treatment of HCC.

Background: Sorafenib, an oral multi-kinase inhibitor of rapidly accelerated fibrosarcoma; vascular endothelial growth factor receptor-2/3, platelet-derived growth factor receptor, c-Kit, and Flt-3 signaling, is approved for treatment of advanced hepatocellular carcinoma (HCC). However, the benefit of sorafenib is often diminished because of acquired resistance through the reactivation of ERK signaling in sorafenib-resistant HCC cells. In this work, we investigated whether adding LY3214996, a selective ERK1/2 inhibitor, to sorafenib would increase the anti-tumor effectiveness of sorafenib to HCC cells. Methods: The Huh7 cell line was used as a cell model for treatment with sorafenib, LY3214996, and their combination. Phosphorylation of the key kinases in the Ras/Raf/MAPK and PI3K/Akt pathways, protein expression of the cell cycle, and apoptosis migration were assessed with western blot. MTT and colony-formation assays were used to evaluate cell proliferation. Wound-healing assay was used to assess cell migration. Cell cycle and apoptosis analyses were conducted with flow cytometry. Results: LY3214996 decreased phosphorylation of the Ras/Raf/MAPK and PI3K/Akt pathways, including p-c-Raf, p-P90RSK, p-S6K and p-eIF4EBP1 activated by sorafenib, despite increased p-ERK1/2 levels. LY3214996 increased the anti-proliferation, anti-migration, cell-cycle progression, and pro-apoptotic effects of sorafenib on Huh7R cells. Conclusions: Reactivation of ERK1/2 appears to be a molecular mechanism of acquired resistance of HCC to sorafenib. LY3214996 combined with sorafenib enhanced the anti-tumor effects of sorafenib in HCC. These findings form a theoretical basis for trial of LY3214996 combined with sorafenib as second-line treatment of sorafenib-resistant in advanced HCC.

The construction of highly efficient and multifunctional composite photocatalysts has attracted extensive attention in the field of environmental governance. In this study, an efficient coated ZH-scheme (SnS2@CdS)/TiO2(0 0 1) photocatalyst is resoundingly prepared by hydrothermal, isoelectric point and continuous ion layer adsorption reaction methods and a sequence of characterizations are carried out. The photocatalytic activity of (SnS2@CdS)/TiO2(0 0 1) photocatalyst is evaluated via degradation of phenol with simultaneous conversion of Cr(VI) under sunlight irradiation. Furthermore, the effects of initial concentrations of phenol and Cr(VI), variety of photocatalyst, solution pH, (SnS2@CdS)/TiO2(0 0 1) photocatalyst dosage and recycling times on phenol degradation and Cr(VI) conversion are investigated. The crucial active species of phenol degradation are explored by capture experiments. When the coated ZH-scheme (SnS2@CdS)/TiO2(0 0 1) photocatalyst dosage is 1.0 g/L, phenol and Cr(VI) concentrations are 20 mg/L and 40 mg/L, respectively and solution pH is 4.0, the phenol degradation rate and the Cr(VI) conversion rate could reach 85.52 % and 95.69 %, respectively. Additionally, the possible photocatalytic mechanism of phenol degradation with synchronous Cr(VI) conversion caused by (SnS2@CdS)/TiO2(0 0 1) photocatalyst is proposed. It can be hoped that this work will provide a feasible way to treat high concentrations of organic pollutants and simultaneously convert heavy metal ions.
