Abstract Aim The purpose of this study was to interpret real-world clinical data to analyze the surgical safety and survival outcomes of patients with initial unresectable hepatocellular carcinoma (uHCC) after conversion therapy. Methods A retrospective analysis was performed on 2984 hepatocellular carcinoma (HCC) patients hospitalized in Shandong Cancer Hospital Affiliated to Shandong First Medical University from June 1st, 2019 to June 1st, 2023. Clinicopathological features, response to systemic and/or loco-regional treatments, surgical resection rate after conversion therapy, surgical safety, and postoperative recurrence were analyzed. Results A total of 38 patients were successfully converted to obtain surgical resection. 35 patients underwent radical resection. A high objective response rate (ORR) (52.6% under RECIST v1.1 and 78.9% under mRECIST criteria) was observed in patients under conversion therapy, and the disease control rate (DCR) was 100%. Pathologic complete response (pCR) was 42.9%. Treatment-related adverse events (TRAEs) of any grade were observed in 37 patients (97.4%). Safety of conversion or direct surgery continues to improve. The median follow-up time was 19.3 months. The 1-year Disease-free survival (DFS) rate of patients with direct surgery and patients with conversion surgery were 91.4% and 86.8%, respectively. Conclusions With conversion therapy, a small percentage (1.81%) of uHCC patients are likely to be converted to radical resection. Local combined systemic therapy is a relatively safe and effective conversion therapy, and the safety of surgery is gradually improved after successful conversion. Preliminary follow-up data showed satisfactory survival benefits for patients undergoing conversion surgery. Trial registration This was a retrospective study and it did not interfere with treatment decisions.

Men are particularly sensitive to penis size, especially those with a deformed or injured penis. This can lead to a strong desire for penis enlargement surgery. Given the ethical sensitivities of the penis, penile implants need to be developed with both efficacy and safety. In this study, a polyvinyl alcohol (PVA) patch for penile enlargement prepared via cyclic freeze‒thaw cycles and alkaline treatment. The PVA hydrogels treated with 5 M NaOH had the best mechanical properties and stability. A negative Poisson's ratio structure is incorporated into the design of the enlargement patch, which allows it to conform well to the deformation of the penis. In rabbit models, the enlarged patches can effectively enlarge the penis without degradation or fibrosis while maintaining long-term stability in vivo. This innovation not only provides a safe option for patients in need of penile enlargement but also promises to make a broader contribution to the field of dynamic tissue repair.

Abstract Postoperative neurological dysfunction (PND) is one of the most common complications after a total aortic arch replacement (TAAR). Electrical impedance tomography (EIT) monitoring of cerebral hypoxia injury during TAAR is a promising technique for preventing the occurrence of PND. This study aimed to explore the feasibility of electrical impedance tomography (EIT) for warning of potential brain injury during total aortic arch replacement (TAAR) through building the correlation between EIT extracted parameters and variation of neurological biomarkers in serum. Patients with Stanford type A aortic dissection and requiring TAAR who were admitted between December 2021 to March 2022 were included. A 16-electrode EIT system was adopted to monitor each patient’s cerebral impedance intraoperatively. Five parameters of EIT signals regarding to the hypothermic circulatory arrest (HCA) period were extracted. Meanwhile, concentration of four neurological biomarkers in serum were measured regarding to time before and right after surgery, 12 h, 24 h and 48 h after surgery. The correlation between EIT parameters and variation of serum biomarkers were analyzed. A total of 57 TAAR patients were recruited. The correlation between EIT parameters and variation of biomarkers were stronger for patients with postoperative neurological dysfunction (PND(+)) than those without postoperative neurological dysfunction (PND(−)) in general. Since the need for vascular networks to supply oxygen and nutrients while expelling metabolic waste, most cells can only survive within 200 ​μm of blood vessels; thus, the construction of well-developed blood vessel networks is essential for the manufacture of artificial tissues and organs. Three-dimensional (denoted as 3D) printing is a scalable, reproducible and high-precision manufacturing technology. In the past several years, there have been many breakthroughs in building various vascularized tissues, greatly promoting the development of biological tissue engineering. This paper highlights the latest progress of 3D printed vascularized tissues and organs, including the heart, liver, lung, kidney, and penis. We also discuss the application status and potential of the above printed tissues, and prospect the further requirement of 3D printing technology for manufacturing clinically useable vascularized tissues.

PurposeThe purpose of this study was to evaluate the ability of global inhomogeneity index (GI) and left-right asymmetry index (AI) based on electrical impedance tomography (EIT) to be used in assessing cerebral perfusion heterogeneity. The diagnostic value of these two indices in identifying abnormalities in the degree of cerebral perfusion heterogeneity was also explored.MethodsIn this study, Transcranial Doppler (TCD) was used as a control, and unilateral carotid artery was compressed to change the degree of heterogeneity of cerebral perfusion in 15 healthy volunteers. The control group consisted of an additional 15 volunteers without any intervention. EIT perfusion images were obtained by calculating the impedance difference between at the beginning and end of cerebral vasodilation. Subsequently, GI and AI were calculated based on the pixel values of intracranial regions.ResultsThe GI and AI values in the non-carotid artery compression (NCAC) group were significantly lower than those in the unilateral carotid artery compression (UCAC) group (P < 0.001), whereas there was no significant difference between the left carotid artery compression (LCAC) and right carotid artery compression (RCAC) groups. ROC analysis showed that the area under the curve (AUC), specificity and sensitivity of GI in distinguishing between NCAC and UCAC were 0.94, 0.90 and 0.87, respectively. The AUC, specificity and sensitivity of AI in distinguishing between NCAC and UCAC were 0.86, 0.87 and 0.73, respectively.ConclusionThe results demonstrated that the GI and AI effectively quantify the distribution of intracranial perfusion, demonstrating excellent validity and interindividual comparability, and the ability to detect abnormal cerebral perfusion heterogeneity.

Highlights An innovative class of versatile form-stable composite phase change materials (CPCMs) was fruitfully exploited, featuring MXene/phytic acid hybrid depositing on non-carbonized wood as a robust support. The wood-based CPCMs showcase enhanced thermal conductivity of 0.82 W m−1 K−1 (4.6 times than polyethylene glycol) as well as high latent heat of 135.5 kJ kg−1 (91.5% encapsulation) with thermal durability and stability throughout at least 200 heating and cooling cycles. The wood-based CPCMs have good solar-thermal-electricity conversion, flame-retardant, and electromagnetic shielding properties.

Highlights The SiO2 nanofiber membranes and MXene@c-MWCNT6:4 as one unit layer (SMC1) were bonded together with 5 wt% PVA solution. When the structural unit is increased to three layers, the resulting SMC3 has an average electromagnetic interference SET of 55.4 dB and a low thermal conductivity of 0.062 W m−1 K−1. SMCx exhibit stable electromagnetic interference shielding and excellent thermal insulation even in extreme heat and cold environment.

Abstract Notable-HCC (NCT05185531) is a phase 1b trial, aiming to evaluate the safety and preliminary effectiveness of neoadjuvant PD-1 blockade plus stereotactic body radiotherapy (SBRT) in early-stage resectable hepatocellular carcinoma (HCC). Twenty patients with HCC of BCLC stage 0-A received 3 $$\times $$ × Gy SBRT and two cycles of tislelizumab, an anti-PD-1 monoclonal antibody before the curative HCC resection. Primary endpoints were the surgery delay, radiographic and pathological tumor response after the neoadjuvant therapy, safety and tolerability. During the neoadjuvant therapy, treatment-related adverse events (TRAEs) of grade 1-2 occurred in all 20 patients (100%), eight patients (40%) had grade 3 TRAEs, no grade 4 to 5 TRAE occurred, and all resolved without corticosteroids treatment. Per mRECIST, the objective response rate was 63.2% (12/19), with 3 complete response; the disease control rate was 100%. Two (10.5%) patients achieved complete pathological response. No surgery delay occurred. The neoadjuvant therapy did not increase the surgical difficulty or the incidence of complications. Secondary endpoints of disease-free survival and overall survival were not mature at the time of the analysis. Our pilot trial shows that neoadjuvant therapy with anti-PD-1 + SBRT is safe and promotes tumor responses in early-stage resectable HCC.

Abstract Proteins containing a ubiquitin regulatory X (UBX) domain are cofactors of Cell Division Cycle 48 (CDC48) and function in protein quality control. However, whether and how UBX-containing proteins participate in host–microbe interactions remain unclear. Here we show that MoNLE1, an effector from the fungal pathogen Magnaporthe oryzae, is a core virulence factor that suppresses rice immunity by specifically interfering with OsPUX8B.2. The UBX domain of OsPUX8B.2 is required for its binding to OsATG8 and OsCDC48-6 and controls its 26 S proteasome–dependent stability. OsPUX8B.2 and OsCDC48-6 positively regulate plant immunity against blast fungus, while the high-temperature tolerance heat-shock protein OsBHT, a putative cytoplasmic substrate of OsPUX8B.2–OsCDC48-6, negatively regulates defense against blast infection. MoNLE1 promotes the nuclear migration and degradation of OsPUX8B.2 and disturbs its association with OsBHT. Given the high conservation of MoNLE1 among fungal isolates, plants with broad and durable blast resistance might be generated by engineering intracellular proteins resistant to MoNLE1.

Abstract As testicular mesenchymal stromal cells, stem Leydig cells (SLCs) show great promise in the treatment of male hypogonadism. The therapeutic functions of mesenchymal stromal cells are largely determined by their reciprocal regulation by immune responses. However, the immunoregulatory properties of SLCs remain unclear. Here, we observe that SLCs transplantation restore male fertility and testosterone production in an ischemia‒reperfusion injury mouse model. SLCs prevent inflammatory cascades through mitochondrial transfer to macrophages. Reactive oxygen species (ROS) released from activated macrophages inducing mitochondrial transfer from SLCs to macrophages in a transient receptor potential cation channel subfamily member 7 (TRPM7)-mediated manner. Notably, knockdown of TRPM7 in transplanted SLCs compromised therapeutic outcomes in both testicular ischemia‒reperfusion and testicular aging mouse models. These findings reveal a new mechanism of SLCs transplantation that may contribute to preserve testis function in male patients with hypogonadism related to immune disorders.

Objective: Understansding the changing patterns of in vivo electrical properties for the target tissue is crucial for the accurate temperature monitoring and the treatment efficacy in thermal therapy. Our research aims to investigate the changing patterns and the reversibility of in vivo electrical properties for both healthy livers and liver tumors in a mouse model over a frequency range of 1 Hz to 1 MHz at temperatures between 30 °C to 90 °C.Methods and materials: The mice were anesthetized and the target organ was exposed. An 808-nm near-infrared laser was employed as the heating source to heat the organ in vivo. The four-needle electrode, connected to an impedance analyzer, was utilized to obtain the impedance at varying temperatures, which were monitored by a thermocouple.Results: The findings indicated a gradual decline in impedance with an increase in temperature. Furthermore, the impedance was normalized to that at 30 °C, and the real part of the normalized impedance was defined as the k-values, which range from 0 to 1. The results demonstrated a linear correlation between k-values and temperatures (R2 > 0.9 for livers and R2 > 0.8 for tumors). Abstract The potential of three‐dimensional (3D) printing technology in the fabrication of advanced polymer composites is becoming increasingly evident. This review discusses the latest research developments and applications of 3D printing in polymer composites. First, it focuses on the optimization of 3D printing technology, that is, by upgrading the equipment or components or adjusting the printing parameters, to make them more adaptable to the processing characteristics of polymer composites and to improve the comprehensive performance of the products. Second, it focuses on the 3D printable novel consumables for polymer composites, which mainly include the new printing filaments, printing inks, photosensitive resins, and printing powders, introducing the unique properties of the new consumables and different ways to apply them to 3D printing. Finally, the applications of 3D printing technology in the preparation of functional polymer composites (such as thermal conductivity, electromagnetic interference shielding, biomedicine, self‐healing, and environmental responsiveness) are explored, with a focus on the distribution of the functional fillers and the influence of the topological shapes on the properties and functional characteristics of the 3D printed products. The aim of this review is to deepen the understanding of the convergence between 3D printing technology and polymer composites and to anticipate future trends and applications.

Regenerative endodontic therapy is a promising approach to restore the vitality of necrotic teeth, however, pulp regeneration in mature permanent teeth remains a substantial challenge due to insufficient developmental signals. The dentin is embryologically and histologically similar to the pulp, which contains a cocktail of pulp-specific structural proteins and growth factors, thus we proposed an optimizing strategy to obtain dentin matrix extracted proteins (DMEP) and engineered a DMEP functionalized double network hydrogel, whose physicochemical property was tunable by adjusting polymer concentrations to synchronize with regenerated tissues. In vitro models showed that the biomimetic hydrogel with sustained release of DMEP provided a beneficial microenvironment for the encapsulation, propagation and migration of human dental pulp stem cells (hDPSCs). The odontogenic and angiogenic differentiation of hDPSCs were enhanced as well. To elicit the mechanism hidden in the microenvironment to guide cell fate, RNA sequencing was performed and 109 differential expression of genes were identified, the majority of which enriched in cell metabolism, cell differentiation and intercellular communications. The involvement of ERK, p38 and JNK MAPK signaling pathways in the process was confirmed. Of note, in vivo models showed that the injectable and in situ photo-crosslinkable hydrogel was user-friendly for root canal systems and was capable of inducing the regeneration of highly organized and vascularized pulp-like tissues in root segments that subcutaneously implanted into nude mice. Taken together, this study reported a facile and efficient way to fabricate a cell delivery hydrogel with pulp-specific developmental cues, which exhibited promising application and translation potential in future regenerative endodontic fields.

3-D brain electrical impedance tomography (EIT) holds great promise for real-time noninvasive imaging of various brain injuries. However, a reference method for selecting high-performance electrode configurations has not been proposed. In this article, the optimization of electrode layout, stimulation and measurement protocols, and the number of electrodes are sequentially performed. The signal quality and image reconstruction performance of simulated perturbations in four cortical regions are evaluated with various levels of noise taken into consideration. The results showed that, considering cost and convenience, the best number of electrodes is 20, which should be placed in the suboccipital and central vertex regions as needed. Electrodes with large spacing at different heights are mainly the driving electrodes, and the potential is collected in the appropriate adjacent channels. These principles are expected to provide general guidance for the electrode configuration methods of 3-D brain EIT in clinical applications.

BackgroundIn intracranial pathologic conditions of intracranial pressure (ICP) disturbance or hemodynamic instability, maintaining appropriate ICP may reduce the risk of ischemic brain injury. The change of ICP is often accompanied by the change of intracranial blood status. As a non-invasive functional imaging technique, the sensitivity of electrical impedance tomography (EIT) to cerebral hemodynamic changes has been preliminarily confirmed. However, no team has conducted a feasibility study on the dynamic detection of ICP by EIT technology from the perspective of non-invasive whole-brain blood perfusion monitoring. In this study, human brain EIT image sequence was obtained by in vivo measurement, from which a variety of indicators that can reflect the tidal changes of the whole brain impedance were extracted, in order to establish a new method for non-invasive monitoring of ICP changes from the level of cerebral blood perfusion monitoring.MethodsValsalva maneuver (VM) was used to temporarily change the cerebral blood perfusion status of volunteers. The electrical impedance information of the brain during this process was continuously monitored by EIT device and real-time imaging was performed, and the hemodynamic indexes of bilateral middle cerebral arteries were monitored by transcranial Doppler (TCD). The changes in monitoring information obtained by the two techniques were compared and observed.ResultsThe EIT imaging results indicated that the image sequence showed obvious tidal changes with the heart beating. The microenvironment of the wound bed is essential in the regulation of wound repair. In this regard, strategies that provide a repairing favorable microenvironment may effectively improve healing outcomes. Herein, we attempted to use electrical stimulation (ES) to boost the paracrine function of adipose-derived stem cells from rats (rASCs). By examining the concentrations of two important growth factors, VEGF and PDGF-AA, in the cell culture supernatant, we found that ES, especially 5 μA ES, stimulated rASCs to produce more paracrine factors (5 μA-PFs). Further studies showed that ES may modulate the paracrine properties of rASCs by upregulating the levels of TRPV2 and TRPV3, thereby inducing intracellular Ca2+ influx. To deliver the PFs to the wound to effectively improve the wound microenvironment, we prepared a heparinized PGA host-guest hydrogel (PGA-Hp hydrogel). Moreover, PGA-Hp hydrogel loaded with 5 μA-PFs effectively accelerated the repair process of the full-thickness wound model in rats. Our findings revealed the effects of ES on the paracrine properties of rASCs and highlighted the potential application of heparinized PGA host-guest hydrogels loaded with PFs derived from electrically stimulated rASCs in wound repair.

Elongases of very-long-chain fatty acids (Elovls) are critical rate-limiting enzymes that are involved in LC-PUFA biosynthesis through catalyzing the two-carbon elongation of a pre-existing fatty acyl chain. Thus far, several Elovls have been extensively studied in teleost. However, the functional and physiological roles of Elovls in chondrichthyans have rarely been reported. In this study, we identified and characterized elovl2 from the endangered Chinese sturgeon (Acipenser sinensis) by whole genome scanning. The results show that the coding sequence of elovl2 was 894 bp in length, for a putative protein of 297 amnio acids. Comparative genomic analyses indicated that Chinese sturgeon elovl2 was evolutionarily conserved. Functional characterization in yeast demonstrated that the Chinese sturgeon Elovl2 could efficiently elongate C20 (ARA and EPA) and C22 (22:4n-6 and 22:5n-3) substrates, confirming its critical roles in LC-PUFA biosynthesis. Spatial and temporal expression analyses showed high elovl2 mRNA levels were detected in the liver and brain and showed an increase trend both in embryonic and post-hatching stages. Interestingly, diets with vegetable oils as lipid sources could significantly induce the high expression of elovl2 in Chinese sturgeon, implying that the endogenous LC-PUFA biosynthesis pathway was stimulated by lack of LC-PUFA in their diets. Our findings will enhance our understanding about the evolutionary and functional roles of elovl2 and provide novel insights into the LC-PUFA biosynthesis mechanism in vertebrates.

The maize anthracnose stalk rot and leaf blight diseases caused by the fungal pathogen Colletotrichum graminicola is emerging as an important threat to corn production worldwide. In this work, we provide an improved genome assembly of a C. graminicola strain (TZ-3) by using the PacBio Sequel II and Illumina high-throughput sequencing technologies. The genome of TZ-3 consists of 36 contigs with a length of 59.3 Mb. After correction and evaluation with the Illumina sequencing data and BUSCO, this genome showed a high assembly quality and integrity. Gene annotation of this genome predicted 11,911 protein-coding genes, among which 983 secreted protein-coding genes and 332 effector genes were predicted. Compared with previous genomes of C. graminicola strains, TZ-3 genome is superior in nearly all parameters. The genome assembly and annotation will enhance our knowledge of the genetic makeup of the pathogen and molecular mechanisms underlying its pathogenicity and will provide valuable insights into genome variation across different regions. Ideal repair of intestinal injury requires a combination of leakage-free sealing and postoperative antiadhesion. However, neither conventional hand-sewn closures nor existing bioglues/patches can achieve such a combination. To this end, we develop a sandwiched patch composed of an inner adhesive and an outer antiadhesive layer that are topologically linked together through a reinforced interlayer. The inner adhesive layer tightly and instantly adheres to the wound sites via -NHS chemistry; the outer antiadhesive layer can inhibit cell and protein fouling based on the zwitterion structure; and the interlayer enhances the bulk resilience of the patch under excessive deformation. This complementary trilayer patch (TLP) possesses a unique combination of instant wet adhesion, high mechanical strength, and biological inertness. Both rat and pig models demonstrate that the sandwiched TLP can effectively seal intestinal injuries and inhibit undesired postoperative tissue adhesion. The study provides valuable insight into the design of multifunctional bioadhesives to enhance the treatment efficacy of intestinal injuries.

BackgroundThe synergistic effectiveness of combining immune checkpoint inhibitors with targeted therapies has shown promise in improving the conversion rate for unresectable hepatocellular carcinoma (HCC) patients to a potentially resectable status. However, the efficacy of this approach in the context of HCC with extrahepatic metastasis remains to be conclusively determined.Case presentationWe report a rare case of advanced HCC with extrahepatic metastasis who achieved long-term survival by a combination of systemic therapy (sintilimab and sorafenib) followed by laparoscopic hepatectomy. A 63-year-old man presented at our hospital with discomfort on the right side of his waist. An enlarged right hepatic lobe mass was subsequently revealed by CT scan. The patient’s medical history, including a prior infection with hepatitis B virus, cirrhosis of the liver and an alpha-fetoprotein (AFP) level measuring 41.28 ng/ml substantiated the clinical diagnosis of HCC. On October 30th, 2019, the patient received 200 mg sintilimab intravenously (q3w) plus 200–400 mg BID sorafenib orally, along with antiviral therapy. After six cycles, his disease achieved partial response (PR). On April 26th, 2021, He underwent a laparoscopic hepatectomy. The patient achieved a sustained period of no evidence of disease for 2.5 years and with drug-free survival for 2 years after the resection. His current overall survival is estimated at approximately 4 years.ConclusionsThis case highlights the potential of combining sintilimab and sorafenib in transforming HCC with extrahepatic metastasis into a condition amenable to surgical resection, suggesting that this treatment approach, followed by surgery, may lead to complete remission.

Cognitive decline has been linked to periodontitis through an undetermined pathophysiological mechanism. This study aimed to explore the mechanism underlying periodontitis-related cognitive decline and identify therapeutic strategies for this condition. Using single-nucleus RNA sequencing we found that changes in astrocyte number, gene expression, and cell‒cell communication were associated with cognitive decline in mice with periodontitis. In addition, activation of the NOD-like receptor family pyrin domain containing 3 (NLRP3) inflammasome was observed to decrease the phagocytic capability of macrophages and reprogram macrophages to a more proinflammatory state in the gingiva, thus aggravating periodontitis. To further investigate this finding, lipid-based nanoparticles carrying NLRP3 siRNA (NPsiNLRP3) were used to inhibit overactivation of the NLRP3 inflammasome in gingival macrophages, restoring the oral microbiome and reducing periodontal inflammation. Furthermore, gingival injection of NPsiNLRP3 reduced the number of Serpina3nhigh astrocytes in the hippocampus and prevented cognitive decline. This study provides a functional basis for the mechanism by which the destruction of periodontal tissues can worsen cognitive decline and identifies nanoparticle-mediated restoration of gingival macrophage function as a novel treatment for periodontitis-related cognitive decline.

Abstract Aim To understand the proportion of uHCC (unresectable hepatocellular carcinoma) patients who achieve successful conversion resection in a high-volume setting with state of the art treatment options. Methods We retrospectively reviewed all HCC patients hospitalized to our center from June 1st, 2019 to June 1st, 2022. Conversion rate, clinicopathological features, response to systemic and/or loco-regional therapy and surgical outcomes were analyzed. Results A total of 1,904 HCC patients were identified, with 1672 patients receiving anti-HCC treatment. 328 patients were considered up-front resectable. Of the remaining 1344 uHCC patients, 311 received loco-regional treatment, 224 received systemic treatment, and the remainder (809) received combination systemic plus loco-regional treatment. Following treatment, one patient from the systemic group and 25 patients from the combination group were considered to have resectable disease. A high objective response rate (ORR) was observed in these converted patients (42.3% under RECIST v1.1 and 76.9% under mRECIST criteria). The disease control rate (DCR) reached 100%. 23 patients underwent curative hepatectomy. Major post-operative morbidity was equivalent in the both groups (P=0.76). Pathologic complete response (pCR) was 39.1%. During conversion treatment, grade 3 or higher treatment-related adverse events (TRAEs) were observed in 50% of patients. The median follow-up time was 12.9 months (range, 3.9~40.6) from index diagnosis and 11.4 months (range, 0.9~26.9) from resection. Three patients experienced disease recurrence following conversion surgery. Conclusions By intensive treatment, a small sub-group of uHCC patients (2%) may potentially be converted to curative resection. Loco-regional combined with systemic modality was relative safe and effective in the conversion therapy. Short-term outcomes are encouraging, but long-term follow-up in a larger patient population are required to fully understand the utility of this approach.

While hydrogels find applications in wearable sensors and electronic skins, they are prone to fatigue fractures upon deformation cycling. Xiong et al. report the synthesis of conductive polymerizable rotaxane hydrogels, exhibiting large fatigue resistance, for 3D printable flexible sensors.

The classical 3D-printed scaffolds have attracted enormous interests in bone regeneration due to the customized structural and mechanical adaptability to bone defects. However, the pristine scaffolds still suffer from the absence of dynamic and bioactive microenvironment that is analogous to natural extracellular matrix (ECM) to regulate cell behaviour and promote tissue regeneration. To address this challenge, we develop a black phosphorus nanosheets-enabled dynamic DNA hydrogel to integrate with 3D-printed scaffold to build a bioactive gel-scaffold construct to achieve enhanced angiogenesis and bone regeneration. The black phosphorus nanosheets reinforce the mechanical strength of dynamic self-healable hydrogel and endow the gel-scaffold construct with preserved protein binding to achieve sustainable delivery of growth factor. We further explore the effects of this activated construct on both human umbilical vein endothelial cells (HUVECs) and mesenchymal stem cells (MSCs) as well as in a critical-sized rat cranial defect model. The results confirm that the gel-scaffold construct is able to promote the growth of mature blood vessels as well as induce osteogenesis to promote new bone formation, indicating that the strategy of nano-enabled dynamic hydrogel integrated with 3D-printed scaffold holds great promise for bone tissue engineering.

ObjectiveThe purpose of this paper is to compare the differences in the features of multifrequency electrical impedance tomography (MFEIT) images of human heads between healthy subjects and patients with brain diseases and to explore the possibility of applying MFEIT to intracranial abnormality detection.MethodsSixteen healthy volunteers and 8 patients with brain diseases were recruited as subjects, and the cerebral MFEIT data of 9 frequencies in the range of 21 kHz - 100 kHz of all subjects were acquired with an MFEIT system. MFEIT image sequences were obtained according to certain imaging algorithms, and the area ratio of the ROI (AR_ROI) and the mean value of the reconstructed resistivity change of the ROI (MVRRC_ROI) on both the left and right sides of these images were extracted. The geometric asymmetry index (GAI) and intensity asymmetry index (IAI) were further proposed to characterize the symmetry of MFEIT images based on the extracted indices and to statistically compare and analyze the differences between the two groups of subjects on MFEIT images.ResultsThere were no significant differences in either the AR_ROI or the MVRRC_ROI between the two sides of the brains of healthy volunteers (p > 0.05); some of the MFEIT images mainly in the range of 30 kHz – 60 kHz of patients with brain diseases showed stronger resistivity distributions (larger area or stronger signal) that were approximately symmetric with the location of the lesions. However, statistical analysis showed that the AR_ROI and the MVRRC_ROI on the healthy sides of MFEIT images of patients with unilateral brain disease were not significantly different from those on the affected side (p > 0.05). Endowing adhesives with both dry and wet adhesion to surfaces from hard substrates to soft biological tissues is challenging. Herein, a branched supramolecular polymer is introduced in dry‐ and wet‐crosslinking forms to adaptively achieve dry and wet interfacial adhesion. The branched supramolecular polymer (PAMU) with multiple pendant functional moieties is prepared by one‐pot reversible addition‐fragmentation chain transfer polymerization. The PAMU polymer solution can firmly adhere to glass sheets after the water evaporates. The aqueous solution of PAMU polymer with residual double bonds can be further polymerized to form a wet crosslinking network. The hydrogen bonding moieties endow the supramolecular network with self‐healing properties, while the in situ polymerization of the PAMU polymer leads to stable adhesion to wet tissues. This study provides a promising approach to develop supramolecular adhesives for versatile adhesion applications.

As an ancient and endangered species unique to the Yangtze River in China, the wild population of the Dabry’s sturgeon has become scarce. Due to the long time till the first sexual maturity of Dabry’s sturgeon, the population of artificially bred Dabry’s sturgeon recovered slowly. As a member of the tachykinin family, TAC1 has been reported to have a variety of functions in mammals such as pain control, smooth muscle contraction and reproductive cycle regulation, but the function of Tac1 in fish has been rarely reported. In this study, we synthesized two tac1 gene products, Substance P (SP) and neurokinin A (NKA), and further verified the effect of two tac1 gene products on the secretion of related hormones in the pituitary of Dabry’s Sturgeon by intraperitoneal injection and co-culture of primary cells. Expression studies revealed that the newly cloned tac1 were mainly distributed in the hypothalamus and pituitary tissue of the brain. In prepubertal Dabry’s sturgeon, this study showed that the two gonadotropins’ mRNA levels in pituitary tissue can be significantly increased by SP and NKA through intraperitoneal injection, and the LH protein level in serum was also increased. Further study showed that both NKA and SP could promote the two gonadotropins’ mRNA expression in pituitary cells of Dabry’s sturgeon. In addition, we explored the optimal dose and time of SP and NKA on pituitary cells is 24 h and over 10 nM. These results, as a whole, suggested that tac1 gene products play an important role in gonadotropin release and gonadal development in prepubertal Dabry’s sturgeon.

Corticotropin-releasing hormone (CRH) is known for its crucial role in the stress response system, which could induce pituitary adrenocorticotropic hormone (ACTH) secretion to promote glucocorticoid release in the adrenal gland. However, little is known about other pituitary actions of CRH in teleosts. Somatolactin is a fish-specific hormone released from the neurointermediate lobe (NIL) of the posterior pituitary. A previous study has reported that ACTH was also located in the pituitary NIL region. Interestingly, our present study found that CRH could significantly induce two somatolactin isoforms’ (SLα and SLβ) secretion and synthesis in primary cultured grass carp pituitary cells. Pharmacological analysis further demonstrated that CRH-induced pituitary somatolactin expression was mediated by the AC/cAMP/PKA, PLC/IP3/PKC, and Ca2+/CaM/CaMK-II pathways. Finally, transcriptomic analysis showed that both SLα and SLβ should play an important role in the regulation of lipid metabolism in primary cultured hepatocytes. These results indicate that CRH is a novel stimulator of somatolactins in teleost pituitary cells, and somatolactins may participate in the stress response by regulating energy metabolism.

Human reproductive organs are of vital importance to the life of an individual and the reproduction of human populations. So far, traditional methods have a limited effect in recovering the function and fertility of reproductive organs and tissues. Thus, aim to replace and facilitate the regrowth of damaged or diseased tissue, various biomaterials are developed to offer hope to overcome these difficulties and help gain further research progress in reproductive tissue engineering. In this review, we focus on the biomaterials and their four main applications in reproductive tissue engineering: in vitro generation and culture of reproductive cells; development of reproductive organoids and models; in vivo transplantation of reproductive cells or tissues; and regeneration of reproductive tissue. In reproductive tissue engineering, designing biomaterials for different applications with different mechanical properties, structure, function, and microenvironment is challenging and important, and deserves more attention.

Three dimensional (3D) bioprinting is a powerful tool, that was recently applied to tissue engineering. This technique allows the precise deposition of cells encapsulated in supportive bioinks to fabricate complex scaffolds, which are used to repair targeted tissues. Here, we review the recent developments in the application of 3D bioprinting to dental tissue engineering. These tissues, including teeth, periodontal ligament, alveolar bones, and dental pulp, present cell types and mechanical properties with great heterogeneity, which is challenging to reproduce in vitro. After highlighting the different bioprinting methods used in regenerative dentistry, we reviewed the great variety of bioink formulations and their effects on cells, which have been established to support the development of these tissues. We discussed the different advances achieved in the fabrication of each dental tissue to provide an overview of the current state of the methods. We conclude with the remaining challenges and future needs.

Renal cell carcinoma (RCC) poses a serious threat to human health, which urgently requires a method that can quickly distinguish between human normal renal tissue (NRT) and RCC for the purpose of accurate detection in clinical practice. The significant difference in cell morphology between NRT and RCC tissue underlies the great potential of the bioelectrical impedance analysis (BIA) to distinguish two types of human tissues. The study aims to achieve such discrimination through comparison of their dielectric properties within the frequency range from 10 Hz to 100 MHz. The dielectric properties of 69 cases of human normal and cancer renal tissue were measured 15 min after tissue isolation in a strictly controlled environment (37°C, 90% humidity). In addition to the impedance parameters (resistivity, conductivity and relative permittivity), the characteristic parameters extracted from the Cole curve were also compared between NRT and RCC. Furthermore, a novel index, distinguishing coefficient (DC), was used to obtain the optimal frequency for discrimination between NRT and RCC. The limitations of autologous bone grafts necessitate the development of advanced biomimetic biomaterials for efficient cranial defect restoration. The cranial bones are typical flat bones with sandwich structures, consisting of a diploe in the middle region and 2 outer compact tables. In this study, we originally developed 2 types of flat-bone-mimetic β-tricalcium phosphate bioceramic scaffolds (Gyr-Comp and Gyr-Tub) by high-precision vat-photopolymerization-based 3-dimensional printing. Both scaffolds had 2 outer layers and an inner layer with gyroid pores mimicking the diploe structure. The outer layers of Gyr-Comp scaffolds simulated the low porosity of outer tables, while those of Gyr-Tub scaffolds mimicked the tubular pore structure in the tables of flat bones. The Gyr-Comp and Gyr-Tub scaffolds possessed higher compressive strength and noticeably promoted in vitro cell proliferation, osteogenic differentiation, and angiogenic activities compared with conventional scaffolds with cross-hatch structures. After implantation into rabbit cranial defects for 12 weeks, Gyr-Tub achieved the best repairing effects by accelerating the generation of bone tissues and blood vessels. This work provides an advanced strategy to prepare biomimetic biomaterials that fit the structural and functional needs of efficacious bone regeneration.

A cerebral contrast-enhanced electrical impedance tomography perfusion method is developed for acute ischemic stroke during intravenous thrombolytic therapy. Several clinical contrast agents with stable impedance characteristics and high-conductivity contrast were screened experimentally as electrical impedance contrast agent candidates. Myofibroblasts, the primary effector cells for implant-induced fibrosis, contribute to this process by secreting excessive collagen-rich matrix and contracting. Thus, approaches that suppress myofibroblasts may achieve desirable suppression effects in the fibrotic process. As one of the important physical properties of materials, material topographical structures have been proven to affect various aspects of cell behaviors, so is it possible to manipulate the formation of myofibroblasts by tailoring the topographical properties of medical devices? In this study, polycaprolactone (PCL) surfaces with typical micropatterns (micro column and micro pit) were fabricated. The regulatory effects of surface micropatterns on the myofibroblastic differentiation of fibroblasts were investigated. Compared to the flat surfaces and surfaces with micro pit, surfaces with micro columns triggered the F- to G-actin transition, inhibiting the nuclear transfer of myocardin-related transcription factor-A. Subsequently, the downstream gene α-smooth muscle actin, which is a marker of myofibroblasts, was suppressed. Further in vivo investigation showed that PCL implants with micro-column-patterned surfaces inhibited the formation of peri-implant fibrotic capsules. Our results demonstrate that surface topographical properties are a potent regulator of fibroblast differentiation into myofibroblasts and highlight the antifibrotic potential of modifying surfaces with micro-column patterns.

Electrical impedance tomography (EIT) has been widely used in biomedical research because of its advantages of real-time imaging and nature of being non-invasive and radiation-free. Additionally, it can reconstruct the distribution or changes in electrical properties in the sensing area. Recently, with the significant advancements in the use of deep learning in intelligent medical imaging, EIT image reconstruction based on deep learning has received considerable attention. This study introduces the basic principles of EIT and summarizes the application progress of deep learning in EIT image reconstruction with regards to three aspects: a single network reconstruction, deep learning combined with traditional algorithm reconstruction, and multiple network hybrid reconstruction. In future, optimizing the datasets may be the main challenge in applying deep learning for EIT image reconstruction. Adopting a better network structure, focusing on the joint reconstruction of EIT and traditional algorithms, and using multimodal deep learning-based EIT may be the solution to existing problems. In general, deep learning offers a fresh approach for improving the performance of EIT image reconstruction and could be the foundation for building an intelligent integrated EIT diagnostic system in the future.

Background and objectiveThe purpose of this study was to eliminate the interferences of electrical impedance tomography (EIT) on synchronous recording electroencephalography (EEG) for seizure detection.MethodsThe simulated EIT signal generated by COMSOL Multiphysics was superimposed on the clinical EEG signal obtained from the CHB-MIT Scalp EEG Database, and then the spectrum features of superimposed mixed signals were analyzed. According to the spectrum analysis, in addition to high-frequency interference at 51.2 kHz related to the drive current, there was also low-frequency interference caused by switching of electrode pairs, which were used to inject drive current. A low pass filter and a comb filter were used to suppress the high-frequency interference and low-frequency interference, respectively. Simulation results suggested the low-pass filter and comb filter working together effectively filtered out the interference of EIT on EEG in the process of synchronous monitoring.ResultsAs a result, the normal EEG and epileptic EEG could be recognized effectively. Pearson correlation analysis further confirmed the interference of EIT on EEG was effectively suppressed.ConclusionsThis study provides a simple and effective interference suppression method for the synchronous monitoring of EIT and EEG, which could be served as a reference for the synchronous monitoring of EEG and other medical electromagnetic devices.

Fibronectin (Fn) is significant to the performance of biomaterials, and the chemistry of biomaterial surface play important roles in Fn adsorption and subsequent cell behavior. However, the “molecular scale” mechanism is still unclear. Herein, we combined experimental strategies with molecular simulations to solve this problem. We prepared self-assembled monolayers with varying chemistries, i.e., SAMs-CH3, SAMs-NH2, SAMs-COOH and SAMs-OH, and characterized Fn adsorption and cell behaviors on them. Next, Monte Carlo method and all-atom molecular dynamics simulations were employed to reveal the orientation/conformation of Fn on surfaces. We found that SAMs-CH3 strongly adsorbed Fn via hydrophobic interactions, but show poor bioactivity as the low exposure of RGD/PHSRN motifs and the deformation of Fn. SAMs-NH2 and SAMs-COOH could adsorb Fn efficiently via vdW interactions, electrostatic interactions, hydrogen bonds and salt bridges. Fn exhibited excellent bioactivity for cell adhesion, proliferation and osteogenic differentiation as high exposure of bioactive motifs on SAMs-NH2, or as the activation of other inferior cell-binding motifs on SAMs-COOH. SAMs-OH showed poor Fn adsorption as the water film. However, the adsorbed Fn displayed non-negligible bioactivity due to high exposure of PHSRN motif and large degree of protein flexibility. We believe that the revealed mechanism presents great potential to rationally design Fn-activating biomaterials.

BackgroundReal-time detection of cerebral blood perfusion can prevent adverse reactions, such as cerebral infarction and neuronal apoptosis. Our previous clinical trial have shown that the infusion of therapeutic fluid can significantly change the impedance distribution in the brain. However, whether this alteration implicates the cerebral blood perfusion remains unclear. To explore the feasibility of monitoring cerebral blood perfusion, the present pilot study established a novel cerebral contrast-enhanced electrical impedance tomography (C-EIT) technique.Materials and methodsRabbits were randomly divided into two groups: the internal carotid artery non-occlusion (ICAN) and internal carotid artery occlusion (ICAO) groups. Both of groups were injected with glucose, an electrical impedance-enhanced contrast agent, through the right internal carotid artery under EIT monitoring. The C-EIT reconstruction images of the rabbits brain were analyzed according to the collected raw data. The paired and independent t-tests were used to analyze the remodeled impedance values of the left and right cerebral hemispheres within and between studied groups, respectively. Moreover, pathological examinations of brain were performed immediately after C-EIT monitoring.ResultsAccording to the reconstructed images, the impedance value of the left cerebral hemisphere in the ICAN group did not change significantly, whereas the impedance value of the right cerebral hemisphere gradually increased, reaching a peak at approximately 10 s followed by gradually decreased. In the ICAO group, the impedance values of both cerebral hemispheres increased gradually and then began to decrease after reaching the peak value. According to the paired t-test, there was a significant difference (P < 0.001) in the remodeling impedance values between the left and right hemispheres in the ICAN group, and there was also a significant difference (P < 0.001) in the ICAO group. According to the independent t-test, there was a significant difference (P < 0.001) of the left hemispheres between the ICAN and ICAO groups.ConclusionThe cerebral C-EIT proposed in this pilot study can reflect cerebral blood perfusion. This method has potential in various applications in the brain in the future, including disease progression monitoring, collateral circulation judgment, tumor-specific detection, and brain function research.

The temperature dependence of the dielectric properties of blood is important for studying the biological effects of electromagnetic fields, electromagnetic protection, disease diagnosis, and treatment. However, owing to the limitations of measurement methods, there are still some uncertainties regarding the temperature characteristics of the dielectric properties of blood at low and medium frequencies. In this study, we designed a composite impedance measurement box with high heat transfer efficiency that allowed for a four/two-electrode measurement method. Four-electrode measurements were carried out at 10 Hz-1 MHz to overcome the influence of electrode polarization, and two-electrode measurements were carried out at 100 Hz-100 MHz to avoid the influence of distribution parameters, and the data was integrated to achieve dielectric measurements at 10 Hz-100 MHz. At the same time, the temperature of fresh blood from rabbits was controlled at 17–39°C in combination with a temperature-controlled water sink. The results showed that the temperature coefficient for the real part of the resistivity of blood remained constant from 10 Hz to 100 kHz (−2.42%/°C) and then gradually decreased to −0.26%/°C. The temperature coefficient of the imaginary part was positive and bimodal from 6.31 kHz to 100 MHz, with peaks of 5.22%/°C and 4.14%/°C at 126 kHz and 39.8 MHz, respectively. Finally, a third-order function model was developed to describe the dielectric spectra at these temperatures, in which the resistivity parameter in each dispersion zone decreased linearly with temperature and each characteristic frequency increased linearly with temperature. The model could estimate the dielectric properties at any frequency and temperature in this range, and the maximum error was less than 1.39%, thus laying the foundation for subsequent studies.

Introduction Liver resection is the mainstay of curative-intent treatment for hepatocellular carcinoma (HCC), but the postoperative 5-year recurrence rate reaches 70%, and there are no adjuvant or neoadjuvant therapies recommended by major HCC guidelines that can reduce the risk of recurrence. In the recent decade, significant progress has been achieved in the systemic treatment of HCC, mainly from immune checkpoint inhibitors (ICIs) and targeted therapy. In other malignancies, ICIs in the neoadjuvant setting have shown better outcomes than in the adjuvant setting. On the other hand, the addition of radiation to ICIs incrementally improves the systemic response to ICIs. Neoadjuvant therapy of ICIs plus stereotactic body radiotherapy (SBRT) has shown promising results in several types of solid tumours but not HCC.Methods and analysis Here, we describe a phase Ib clinical trial of neoadjuvant SBRT plus PD-1 (tislelizumab) prior to hepatic resection in HCC patients. Prior to resection, eligible HCC patients will receive 8 Gy×3 fractions of SBRT together with two cycles of tislelizumab with an interval of 3 weeks. HCC resection is scheduled 4 weeks after the second dose of tislelizumab, followed by adjuvant tislelizumab for 1 year. We plan to enrol 20 participants in this trial. The primary study endpoints include the delay of surgery, tumour response and safety and tolerability of the sequential SBRT/tislelizumab. Other endpoints are the disease-free survival and overall survival rates every 3 or 6 months after the surgery.Ethics and dissemination This trial was approved by the Ethics Committee of Shandong Cancer Hospital and Institute (SDZLEC2022-021-01). The final results of this trial will be published in a peer-reviewed journal after completion.Trial registration number NCT05185531.

Polyimide foam (PIF) is a rising star in high-end applications such as aerospace thermal insulation and military sound absorption. However, the basic rule on molecular backbone design and uniform pore formation of PIF still need to be explored. In this work, polyester ammonium salt (PEAS) precursor powders are synthesized between alcoholysis ester of 3, 3′, 4, 4′-benzophenone tetracarboxylic dianhydride (BTDE) and aromatic diamines with different chain flexibility and conformation symmetry. Then, a standard “stepwise heating” thermo-foaming approach is used to prepare PIF with comprehensive properties. A rational thermo-foaming program is designed based on in situ observation of pore formation during heating. The fabricated PIFs have uniform pore structure, and PIFBTDA-PDA shows the smallest size (147 μm) and narrow distribution. Interestingly, PIFBTDA-PDA also presents a balanced strain recovery rate (SR = 91%) and mechanical robustness (0.051 MPa at 25% strain) and its pore structure maintains regularity after 10 compression–recovery cycles, mainly due to high rigidity of the chains. Furthermore, all the PIFs possess lightweight feature (15–20 kg∙m−3), good heat resistance (Tg at 270–340 °C), thermal stability (T5% at 480–530 °C), thermal insulation properties (λ = 0.046–0.053 W∙m−1K−1 at 20 °C, λ = 0.078–0.089 W∙m−1K−1 at 200 °C), and excellent flame retardancy (LOI > 40%). The reported monomer-mediated pore-structure control strategy can provide guidelines for the preparation of high-performance PIF and its industrial applications.

Molecularly imprinted polymers (MIPs) are synthetic polymers with specific binding sites that present high affinity and spatial and chemical complementarities to a targeted analyte. They mimic the molecular recognition seen naturally in the antibody/antigen complementarity. Because of their specificity, MIPs can be included in sensors as a recognition element coupled to a transducer part that converts the interaction of MIP/analyte into a quantifiable signal. Such sensors have important applications in the biomedical field in diagnosis and drug discovery, and are a necessary complement of tissue engineering for analyzing the functionalities of the engineered tissues. Therefore, in this review, we provide an overview of MIP sensors that have been used for the detection of skeletal- and cardiac-muscle-related analytes. We organized this review by targeted analytes in alphabetical order. Thus, after an introduction to the fabrication of MIPs, we highlight different types of MIP sensors with an emphasis on recent works and show their great diversity, their fabrication, their linear range for a given analyte, their limit of detection (LOD), specificity, and reproducibility. We conclude the review with future developments and perspectives.

Optimizing the concentration of different functional peptides on a surface can be a complex process. Here, the authors report on the use of a click immobilization strategy to create gradients of two different functional peptides on a surface to screen different density functions for rapid optimization.

Background: Hepatocellular carcinoma (HCC) is one of the most common malignancies in China. Most HCC patients are firstly diagnosed at an advanced stage, and systemic treatments are the mainstay of treatment. Summary: In recent years, immune checkpoint inhibitors have made a breakthrough in the systemic treatment of middle-advanced HCC, breaking the single therapeutic pattern of molecular targeted agents. To better guide the clinical treatment for effective and safe use of immunotherapeutic drugs, the Chinese Association of Liver Cancer and Chinese Medical Doctor Association has gathered multidisciplinary experts and scholars in relevant fields to formulate the “Chinese Clinical Expert Consensus on Immunotherapy for Hepatocellular Carcinoma (2021)” based on current clinical studies and clinical medication experience for reference in China. Key messages: The consensus contained 17 recommendations, including the preferred regimen for first- and second-line immunotherapy, evaluation and monitoring before/during/after treatment, management of complications, precautions for special patients and potential population for immunotherapy.

Biodegradable polylactic acid (PLA) foams with open-cell structures are good candidates for oil–water separation. However, the foaming of PLA with high-expansion and uniform cell morphology by the traditional supercritical carbon dioxide microcellular foaming method remains a big challenge due to its low melting strength. Herein, a green facile strategy for the fabrication of open-cell fully biodegradable PLA-based foams is proposed by introducing the unique stereocomplexation mechanism between PLLA and synthesized star-shaped PDLA for the first time. A series of star-shaped PDLA with eight arms (8-s-PDLA) was synthesized with different molecular weights and added into the PLLA as modifiers. PLLA/8-s-PDLA foams with open-cells structure and high expansion ratios were fabricated by microcellular foaming with green supercritical carbon dioxide. In detail, the influences of induced 8-s-PDLA on the crystallization behavior, rheological properties, cell morphology and consequential oil–water separation performance of PLA-based foam were investigated systemically. The addition of 8-s-PDLA induced the formation of SC-PLA, enhancing crystallization by acting as nucleation sites and improving the melting strength through acting as physical cross-linking points. The further microcellular foaming of PLLA/8-s-PDLA resulted in open-cell foams of high porosity and high expansion ratios. With an optimized foaming condition, the PLLA/8-s-PDLA-13K foam exhibited an average cell size of about 61.7 μm and expansion ratio of 24. Furthermore, due to the high porosity of the interconnected open cells, the high-absorption performance of the carbon tetrachloride was up to 37 g/g. This work provides a facile green fabrication strategy for the development of environmentally friendly PLA foams with stable open-cell structures and high expansion ratios for oil–water separation.

Dressing materials with wet adhesive and antibacterial features are in high demand for the healing of infected wounds. Here, we report the development of an antibacterial peptide (KRWWKWWRRC)-modified collagen nanosheet (KCN) by a two-step spin-coating process. The two-dimensional nanosheet demonstrates promising mechanical strength of ~31 ​MPa at a thickness of 87 ​nm, which is comparable to human skin tissue. The KCN with high transparency and flexibility could firmly adhere to the complex surface of biological tissues in wet environments. In vitro cytotoxicity and antibacterial experiments showed that the KCN is biocompatible and has high antibacterial activity (over 99%). Moreover, a rat skin infection wound model demonstrated that the KCN showed rapid wound closure and low inflammation, and wound closure could be achieved on day 14 after surgery with good re-epithelialization. Our findings reveal a promising nanosheet-based strategy for treating bacteria-infected soft-tissue wounds.

Bone is an important part of the human body structure and plays a vital role in human health. A microfluidic chip that can simulate the structure and function of bone will provide a platform for bone-related biomedical research. Hydroxyapatite (HA), a bioactive ceramic material, has a similar structure and composition to bone mineralization products. In this study, we used HA as a microfluidic chip component to provide a highly bionic bone environment. HA substrates with different microchannel structures were printed by using ceramic stereolithography (SLA) technology, and the minimum trench width was 50 μm. The HA substrate with microchannels was sealed by a thin polydimethylsiloxane (PDMS) layer to make a HA-PDMS microfluidic chip. Cell culture experiments demonstrated that compared with PDMS, HA was more conducive to the proliferation and osteogenic differentiation of the human foetal osteoblast cell line (hFOB). In addition, the concentration gradient of the model drug doxorubicin hydrochloride (DOX) was successfully generated on a Christmas tree structure HA-PDMS chip, and the half maximal inhibitory concentration (IC50) of DOX was determined. The findings of this study indicate that the HA-PDMS microfluidic chip has great potential in the field of high-throughput bone-related drug screening and bone-related research.

Electrical impedance tomography (EIT) is a noninvasive functional diagnostic technique that has been successfully applied to human lung and brain. EIT reconstruction is an ill-conditioned problem: the regularization parameters establish a trade-off between acceptable data fitting and acceptable solution stability. This trade-off is necessary to select the optimal regularization parameters for each data frame to continuously obtain high-quality images in real-time monitoring. However, using the objective regularization parameter selection method before each image reconstruction may reduce the reconstruction efficiency, and using heuristic selection for all images may cause significant quality degradation, thereby creating challenges in long-term clinical EIT monitoring. This study explores a robust EIT target adaptive differential iterative reconstruction algorithm that does not require the advance selection of the optimal regularization parameter to facilitate the clinical application of EIT long-term bedside monitoring. Specifically, second-order Taylor approximation expansion and Euler–Lagrange theorem were used to derive the conductivity differential iteration relationship. Furthermore, the reconstruction quality and generalization ability of the proposed algorithm were validated based on comparisons with the L-curve and the generalized cross-validation parameter selection methods through simulations, phantom experiments, and human lung recruitment data. Compared with the L-curve and generalized cross-validation methods, the TADI algorithm reduces the position error by 33.07% and 47.17%, the ringing by 26.49% and 32.69%, and the shape deformation by 18.34% and 19.40% on average, respectively. The results revealed that the TADI algorithm could achieve a significant improvement over the existing state-of-the-art methods in terms of stability, consistency, and efficiency.

Electrospun fibers have received significant interests for various application areas such as filtration, composites and biomedical products due to their large surface area, good continuity, high porosity and many other unique properties. In bio-related applications, electrospun fibers have been used for in-situ drug delivery, tissue engineering scaffolds and wound dressing. In more recent years, there has been a drive toward novel electrospun fibers with added functionalities. Nanoengineering of electrospun fibers has introduced many of such novel properties. Through this review, researchers are provided with a state of the art overview of nanoenhanced electrospun fibers with added functionalities. Examples of some nanoengineered fibers include; surface functionalization, multi-component fibers, porous nanofibers, the creation of surface nano-topographies, and the incorporation of nanoparticles to create hierarchical fibrous structures for tailoring of physicochemical properties with a special focus on biomedical applications.
