BACKGROUND: Alpha-1 antitrypsin deficiency (AATD) is related to developing lung and liver disease, but no large-scale studies examine its association with birth outcomes. OBJECTIVE: We investigated the risk of pregnancy complications and adverse birth outcomes in mothers and children with AATD. METHODS: Using a large cohort data of Danish mothers and children with AATD from 1973 to 2013 (n = 2,027,229), with 559 cases (305 mothers and 254 children). BackgroundNeonatal per- and polyfluoroalkyl substance (PFAS) exposure can disrupt hormonal homeostasis and induce neuro- and immunotoxicity in children. In this exploratory study, we investigated associations between PFAS levels in neonatal dried blood spots and retinoblastoma risk.Materials and methodsThis study included 501 retinoblastoma cases born from 1983 to 2011 and 899 controls frequency-matched by birth year (20:1 matching ratio), born to 755 US-born and 366 Mexico-born mothers in California. Perfluorooctanesulfonic acid (PFOS), perflurooctanoic acid (PFOA), and perfluorononanoic acid (PFNA) feature intensities were identified from neonatal blood spots from California newborn Genetic Disease Screening Program. Using logistic regression, we assessed whether an interquartile range (IQR) increase of PFAS levels or having above-mean levels of PFAS in blood affects retinoblastoma risk overall or its subtypes (i.e., unilateral, bilateral). We assessed children of US-born and Mexico-born mothers, separately.Results and discussionAmong all children, above-mean PFOS levels at birth increased the odds of retinoblastoma overall by 29% (95% Confidence Interval (CI): 1.00, 1.67) and unilateral retinoblastoma by 42% (95% CI: 1.03, 1.97). For children of Mexico-born mothers, we estimated the highest odds of retinoblastoma overall (adjusted odds ratio (aOR): 1.67; 95% CI: 1.06, 2.66) and bilateral retinoblastoma (aOR: 2.06; 95% CI: 1.12, 3.92) with above-mean PFOS levels. Among children of US-born mothers, higher PFOS levels increased the odds of unilateral retinoblastoma by 15% (95% CI: 0.99, 1.35) for each IQR increase and by 71% among children with above-mean PFOS levels (95% CI: 1.04, 2.90). In addition, for children of US-born mothers, PFOA increased the odds of retinoblastoma overall (aOR: 1.41; 95% CI: 1.00, 2.02 for above-mean levels, aOR: 1.06; 95% CI: 0.98, 1.16 per IQR increase). PFNA was not associated with retinoblastoma risk.ConclusionsOur results suggested that PFOS and PFOA might contribute to retinoblastoma risk in children born in California.

BackgroundRetinoblastoma is rare but nevertheless the most common pediatric eye cancer that occurs in children under age 5. High-resolution metabolomics (HRM) is a powerful analytical approach to profile metabolic features and pathways or identify metabolite biomarkers. To date, no studies have used pre-diagnosis blood samples from retinoblastoma cases and compared them to healthy controls to elucidate early perturbations in tumor pathways.ObjectivesHere, we report on metabolic profiles of neonatal blood comparing cases later in childhood diagnosed with retinoblastoma and controls.MethodsWe employed untargeted metabolomics analysis using neonatal dried blood spots for 1327 children (474 retinoblastoma cases and 853 healthy controls) born in California from 1983 to 2011. Cases were selected from the California Cancer Registry and controls, frequency matched to cases by birth year, from California birth rolls. We performed high-resolution metabolomics to extract metabolic features, partial least squares discriminant analysis (PLS-DA) and logistic regression to identify features associated with disease, and Mummichog pathway analysis to characterize enriched biological pathways.ResultsPLS-DA identified 1917 discriminative features associated with retinoblastoma and Mummichog identified 14 retinoblastoma-related enriched pathways including linoleate metabolism, pentose phosphate pathway, pyrimidine metabolism, fructose and mannose metabolism, vitamin A metabolism, as well as fatty acid and lipid metabolism.InterpretationOur findings linked a retinoblastoma diagnosis in early life to newborn blood metabolome perturbations indicating alterations in inflammatory pathways and energy metabolism. Neonatal blood spots may provide a venue for early detection for this or potentially other childhood cancers.

Abstract Manipulating grain size demonstrates great potential for yield promotion in cereals since it is tightly associated with grain weight. Several pathways modulating grain size have been elaborated in rice, but possible crosstalk between the ingredients is rarely studied. OsmiR396 negatively regulates grain size through targeting OsGRF4 (GS2) and OsGRF8, and proves to be multi-functioning. Here we showed that expression of GS3 gene, a Gγ-protein encoding gene, that negatively regulates grain size, was greatly down-regulated in the young embryos of MIM396, GRF8OE and GS2OE plants, indicating possible regulation of GS3 gene by OsmiR396/GRF module. Meanwhile, multiple biochemical assays proved possible transcriptional regulation of OsGRF4 and OsGRF8 proteins on GS3 gene. Further genetic relation analysis revealed tight genetic association between not only OsmiR396 and GS3 gene, but also GS2 and GS3 gene. Moreover, we revealed possible regulation of GS2 on four other grain size-regulating G protein encoding genes. Thus, the OsmiR396 pathway and the G protein pathway cross talks to regulate grain size. Therefore, we established a bridge linking the miRNA-transcription factors pathway and the G-protein signaling pathway that regulates grain size in rice.

Abstract Plants are affected by many environmental factors during their various stages of growth, among which salt stress is a key factor. WRKY transcription factors play important roles in the response to stress in plants. In this study, SmWRKY40 from eggplant (Solanum melongena L.) was found to belong to the subfamily of WRKY transcription factor group II, closely related to the evolution of wild tomato ScWRKY40 (Solanum chilense). The expression of SmWRKY40 could be induced by several abiotic stresses (drought, salt, and high temperature) and ABA to different degrees, with salt stress being the most significant. In Arabidopsis thaliana, the seed germination rate of SmWRKY40 overexpression seedlings was significantly higher than those of the wild type under high concentrations of NaCl and ABA, and root elongation of overexpression lines was also longer than wild type under NaCl treatments. SmWRKY40 overexpression lines were found to enhance Arabidopsis tolerance to salt with lower ROS, MDA, higher soluble protein, proline accumulation, and more active antioxidant enzymes. The expression level of genes related to stress and ABA signaling displayed significant differences in SmWRKY40 overexpression line than that of WT. These results indicate that SmWRKY40 regulates ABA and salt stress responses in Arabidopsis.

Abstract Leaf morphology is a crucial agronomic characteristic of rice that influences crop yield directly. One primary cause of rice leaf rolling can be attributed to alterations in bulliform cells. Several HD-ZIP IV genes have been identified to be epidemical characterized and function in leaf rolling in rice. Still others need to be studied to fully understand the overall function of HD-ZIP IV family. Among the nine ROC genes encoding HD-ZIP IV family transcription factors in rice, ROC1 exhibits the highest expression in the leaves. Overexpression of ROC1 decreased the size of bulliform cells, and thus resulted in adaxially rolled leaves. To the contrary, knockout of ROC1 (ROC1KO) through Crispr-cas9 system enlarged bulliform cells, and thus led to abaxially rolled leaves. Moreover, ROC1KO plants were sensitive to drought. ROC1 could form homodimers on its own, and heterodimers with ROC5 and ROC8 respectively. Compared to ROC1KO plants, leaves of the ROC1 and ROC8 double knocked out plants (ROC1/8DKO) were more severely rolled abaxially due to enlarged bulliform cells, and ROC1/8DKO plants were more drought sensitive. However, overexpression of ROC8 could not restore the abaxial leaf phenotype of ROC1KO plants. Therefore, we proved that ROC1, a member of the HD-ZIP IV family, regulated leaf rolling and drought stress response through tight association with ROC5 and ROC8.

Objective To analyze the value of placental volume combined with serum glucose transporter 3 (GLUT3) in predicting fetal growth restriction. Methods One hundred pregnant women with fetal growth restriction who underwent MRI examination in our hospital from December 2018 to May 2020 were selected as observation group, and 100 normal pregnant women as control group. The placental volumes of the subjects were measured by MRI examination, and the serum GLUT3 level was mea-sured. The combined diagnosis model of placental volume and serum GLUT3 level was established by logistic regression model. The area under the receiver operating characteristic curve (AUC) was applied to evaluate the value of placental volume and serum GLUT3 level used alone or in combination in predicting fetal growth restriction. Bartonella spp. are globally distributed gram-negative facultative intracellular bacteria that infect a wide range of hosts. Rodents are natural reservoirs of many Bartonella species, some of which are also pathogenic to humans. The rapid development of transportation and tourism has highlighted the risk of Bartonella transmission to humans. Thus, it is essential to maintain surveillance of Bartonella spp. infections in rodents. In China, Bartonella spp. infections have been monitored in various areas; however, these have not included the Hulunbuir border regions. In the present study, we monitored the prevalence and genetics of rodent-associated Bartonella spp. in the Hulunbuir border regions. Eleven rodent species were captured at five ports. Eight species were confirmed as Bartonella-positive using quantitative PCR assay, with an overall positivity rate of 20.05 %. Lasiopodomys brandtii was the predominant rodent species captured for Bartonella detection. Sequencing and phylogenetic analysis (using the maximum likelihood method) revealed the presence of three Bartonella species in these rodents, including two pathogenic to humans, namely, Bartonella alsatica and Bartonella grahamii. B. grahamii was the predominant Bartonella species identified in the rodents. Taken together, these results highlight the prevalence and genetic diversity of Bartonella spp. in rodents in the Hulunbuir border regions, indicating the need for risk assessment of human spillover.

Using agricultural wastes in asphalt pavements presents a promising solution to enhance pavement performance and utilize agricultural wastes simultaneously. In the paper, the fibers are made from recycled cotton stalks. Chemical modifiers are chosen to enhance the adherence of fiber asphalt binders. Different modified fibers are added to the SBR-modified emulsified asphalt. The modified asphalt binders are then utilized to prepare asphalt mixtures. Comparative studies on construction performance, road performance, and durability were carried out. Results showed that water consumption and bitumen aggregate ratio at the micro-surfacing were increased after adding fiber. Fibers with low hygroscopicity, rough surface, and high asphalt absorption rate had better adhesion properties. The long-term performance of a micro-surfacing mixture was most remarkably improved by the CH3COOH-modified cotton straw fibers, demonstrating a 10.07 % drop in rutting depth, a 9.13 % decrease in abrasion value, and a 41.13 % gain in shear resistance compared to the micro-surfacing mixtures with unmodified fibers. The study concluded that modified fiber micro-surfacing asphalt mixes are more suitable for seasonal frozen regions than the conventional mixes. The findings offer a theory foundation for the future development of cotton straw fibers in road engineering.

Abstract Background Stigma exsertion is an essential agricultural trait that can promote cross-pollination to improve hybrid seed production efficiency. However, the molecular mechanism controlling stigma exsertion remains unknown. Results In this study, the Nicotiana tabacum cv. K326 and its two homonuclear-heteroplasmic lines, MSK326 (male-sterile) and MSK326SE (male-sterile and stigma exserted), were used to investigate the mechanism of tobacco stigma exsertion. A comparison of the flowers between the three lines showed that the stigma exsertion of MSK326SE was mainly due to corolla shortening. Therefore, the corollas of the three lines were sampled and presented for RNA-seq analysis, which found 338 candidate genes that may cause corolla shortening. These genes were equally expressed in K326 and MSK326, but differentially expressed in MSK326SE. Among these 338 genes, 15 were involved in hormone synthesis or signal transduction pathways. Consistently, the content of auxin, dihydrozeatin, gibberellin, and jasmonic acid was significantly decreased in the MSK326SE corolla, whereas abscisic acid levels were significantly increased. Additionally, seven genes involved in cell division, cell cycle, or cell expansion were identified. Protein-protein interaction network analysis identified 45 nodes and 79 protein interactions, and the largest module contained 20 nodes and 52 protein interactions, mainly involved in the hormone signal transduction and pathogen defensive pathways. Furthermore, a putative hub gene coding a serine/threonine-protein kinase was identified for the network. Conclusions Our results suggest that hormones may play a key role in regulating tobacco stigma exsertion induced by corolla shortening.

Abstract Background The brown planthopper (BPH) is a kind of piercing-sucking insect specific to rice, with the damage tops the list of pathogens and insects in recent years. microRNAs (miRNAs) are pivotal regulators of plant–environment interactions, while the mechanism underlying their function against insects is largely unknown. Results Here, we confirmed that OsmiR319, an ancient and conserved miRNA, negatively regulated resistance to BPHs, with overexpression of OsmiR319 susceptible to BPH, while suppression of OsmiR319 resistant to BPH in comparison with wild type. Meanwhile, we identified several targets of OsmiR319 that may mediate BPH resistance. Among them, OsPCF5 was the most obviously induced by BPH feeding, and over expression of OsPCF5 was resistance to BPH. In addition, various biochemical assays verified that OsPCF5 interacted with several MYB proteins, such as OsMYB22, OsMYB30, and OsMYB30C.Genetically, we revealed that both OsMYB22 and OsMYB30C positively regulated BPH resistance. Genetic interaction analyses confirmed that OsMYB22 and OsMYB30C both function in the same genetic pathway with OsmiR319b to mediate BPH resistance. Conclusions Altogether, we revealed that OsPCF5 regulates BPH resistance via association with several MYB proteins downstream of OsmiR319, these MYB proteins might function as regulators of BPH resistance through regulating the phenylpropane synthesis.

Abstract Background With the development of single-cell technology, many cell traits can be measured. Furthermore, the multi-omics profiling technology could jointly measure two or more traits in a single cell simultaneously. In order to process the various data accumulated rapidly, computational methods for multimodal data integration are needed. Results Here, we present inClust+, a deep generative framework for the multi-omics. It’s built on previous inClust that is specific for transcriptome data, and augmented with two mask modules designed for multimodal data processing: an input-mask module in front of the encoder and an output-mask module behind the decoder. InClust+ was first used to integrate scRNA-seq and MERFISH data from similar cell populations, and to impute MERFISH data based on scRNA-seq data. Then, inClust+ was shown to have the capability to integrate the multimodal data (e.g. tri-modal data with gene expression, chromatin accessibility and protein abundance) with batch effect. Finally, inClust+ was used to integrate an unlabeled monomodal scRNA-seq dataset and two labeled multimodal CITE-seq datasets, transfer labels from CITE-seq datasets to scRNA-seq dataset, and generate the missing modality of protein abundance in monomodal scRNA-seq data. In the above examples, the performance of inClust+ is better than or comparable to the most recent tools in the corresponding task. Conclusions The inClust+ is a suitable framework for handling multimodal data. Meanwhile, the successful implementation of mask in inClust+ means that it can be applied to other deep learning methods with similar encoder-decoder architecture to broaden the application scope of these models.

AbstractBackground This study aimed to explore the prognostic role of pan-immune-inflammation value (PIV) and develop a new risk model to guide individualized adjuvant systemic treatment following radiofrequency ablation (RFA) for early-stage hepatocellular carcinoma (HCC).Materials and methods Patients with early-stage HCC treated by RFA were randomly divided into training cohort A (n = 65) and testing cohort B (n = 68). Another 265 counterparts were enrolled into external validating cohort C. Various immune-inflammatory biomarkers (IIBs) were screened in cohort A. Prognostic role of PIV was evaluated and validated in cohort B and C, respectively. A nomogram risk model was built in cohort C and validated in pooled cohort D. Clinical benefits of adjuvant anti-angiogenesis therapy plus immune checkpoint inhibitor (AA-ICI) following RFA was assessed in low- and high-risk groups.Results The cutoff point of PIV was 120. High PIV was an independent predictor of unfavorable recurrence-free survival (RFS) and overall survival (OS). ABSTRACTThis study aimed to provide data for the clinical features of invasive pneumococcal disease (IPD) and the molecular characteristics of Streptococcus pneumoniae isolates from paediatric patients in China. We conducted a multi-centre prospective study for IPD in 19 hospitals across China from January 2019 to December 2021. Data of demographic characteristics, risk factors for IPD, death, and disability was collected and analysed. Serotypes, antibiotic susceptibility, and multi-locus sequence typing (MLST) of pneumococcal isolates were also detected. A total of 478 IPD cases and 355 pneumococcal isolates were enrolled. Among the patients, 260 were male, and the median age was 35 months (interquartile range, 12–46 months). Septicaemia (37.7%), meningitis (32.4%), and pneumonia (27.8%) were common disease types, and 46 (9.6%) patients died from IPD. Thirty-four serotypes were detected, 19F (24.2%), 14 (17.7%), 23F (14.9%), 6B (10.4%) and 19A (9.6%) were common serotypes. Pneumococcal isolates were highly resistant to macrolides (98.3%), tetracycline (94.1%), and trimethoprim/sulfamethoxazole (70.7%). Non-sensitive rates of penicillin were 6.2% and 83.3% in non-meningitis and meningitis isolates. 19F-ST271, 19A-ST320 and 14-ST876 showed high resistance to antibiotics. This multi-centre study reports the clinical features of IPD and demonstrates serotype distribution and antibiotic resistance of pneumococcal isolates in Chinese children. There exists the potential to reduce IPD by improved uptake of pneumococcal vaccination, and continued surveillance is warranted.

Accurate monitoring of alpine wetland areas and scientific evaluation of alpine wetland restorability play important roles in revealing the changing mechanism of the ecological environment. However, because of the complexity and lack of spatial data, few studies on evaluation index system of restorability of alpine wetland considered wild animal habitat, such as habitat suitability index of black necked crane. Based on Landsat images of seven periods from 1990 to 2020, this study adopts the methods of spectral mixture analysis and change vector analysis to quantitatively demonstrate the variations and distribution patterns of alpine wetland ecosystem in the eastern edge of Qinghai–Tibet Plateau. The study focuses on determining the abundance of water, vegetation and soil, which the interannual variations represent as the shapes of ‘√’, mirror ‘N’ and normal ‘N’, respectively. In terms of natural ecological condition and human activity interference, we construct a comprehensive evaluation model of restorability of alpine wetland, separate the restorability district and construct the Bayesian network model to discuss the causal relationship between the restorability value and evaluation factor. Results show that the regions of alpine wetland in the study area changed most drastically from 1995 to 2000 and from 2000 to 2005. In addition, the total areas classified as suitable, relatively suitable and suitable for restoration account for 63.69 % of the study area, with a large potential for wetland restorability. Moreover, the prioritisation scheme of wetland restoration action includes the valley swamps of Heihe River in the middle, followed by the flat wetlands and grasslands in the south and lastly by uneven wetlands in the north.

Abstract Advancing fast‐charging technology is an important strategy for the development of alkali metal ion batteries (AMIBs). The exploitation of a new generation of anode material system with high‐rate performance, high capacity, and low risk of lithium/sodium/potassium plating is critical to realize fast‐charging capability of AMIBs while maintaining high energy density and safety. Among them, phosphorus‐based anodes including phosphorus anodes and metal phosphide anodes have attracted wide attention, due to their high theoretical capacities, safe reaction voltages, and natural abundance. In this review, we summarize the research progress of different phosphorus‐based anodes for fast‐charging AMIBs, including material properties, mechanisms for storing alkali metal ions, key challenges and solution strategies for achieving fast‐charging capability. Moreover, the future development directions of phosphorus‐based anodes in fast‐charging AMIBs are highlighted.

Integrating wind power with energy storage technologies is crucial for frequency regulation in modern power systems, ensuring the reliable and cost-effective operation of power systems while promoting the widespread adoption of renewable energy sources. Power systems are changing rapidly, with increased renewable energy integration and evolving system architectures. These transformations bring forth challenges like low inertia and unpredictable behavior of generation and load components. As a result, frequency regulation (FR) becomes increasingly important to ensure grid stability. Energy Storage Systems (ESS) with their adaptable capabilities offer valuable solutions to enhance the adaptability and controllability of power systems, especially within wind farms. This research provides an updated analysis of critical frequency stability challenges, examines state-of-the-art control techniques, and investigates the barriers that hinder wind power integration. Moreover, it introduces emerging ESS technologies and explores their potential applications in supporting wind power integration. Furthermore, this paper offers suggestions and future research directions for scientists exploring the utilization of storage technologies in frequency regulation within power systems characterized by significant penetration of wind power.

IntroductionLocalized nitrogen (N) supply affects rice N uptake by influencing N release, and few studies have examined the effects of root zone N distribution and root growth on rice yield under localized N supply (LNS).MethodsA two-year field experiment was conducted with six treatments: no N application, farmers’ fertilizer practice (FFP), and four LNS treatments, including two types of N fertilizer with urea (U) and controlled release urea (CRU) were mechanically side deep fertilized (SDF) or root zone fertilized (RZF) at 10 cm soil depth (US, UR, CRUS and CRUR treatments, respectively).ResultsCompared with FFP, the dry matter accumulation, N uptake, and yield of LNS increased by 27%, 21%, and 17%, respectively. For N fertilizer type, compared with U, the NH4+-N concentration, total root surface area, volume, average diameter, and root biomass of CRU were significantly increased by 50%, 43%, 53%, and 23%, respectively, which resulted in a significant increase in yield by 12%. Regarding the N application methods, the total surface area, volume, average diameter, and root biomass of SDF were significantly increased by 32%, 24%, 10%, and 25% compared with RZF, respectively. However, the NH4+-N under RZF was more stable and lasted longer, with a significant increase in NH4+-N concentration of 21% compared to the SDF. Moreover, CRUR increased yield, N agronomic use efficiency, and gross return by 3.15%, 5.62%, and 2.81%, respectively, compared to CRUS.ConclusionCRU should be selected as the recommended N fertilizer types, and the combination of CRU and RZF was the most effective choice for rice production.

Anthropogenic emissions of polycyclic aromatic hydrocarbons (PAHs) cause severe ecological impacts by contaminating natural water bodies, affecting various biological groups, and altering interspecies relationships and ecological functions. This study examined the effects of two typical PAHs, phenanthrene (Phe) and naphthalene (Nap), on the anti-grazing defense mechanisms of Tetradesmus obliquus, a primary producer in freshwater food chains. Four non-lethal concentrations (0.01, 0.1, 1, and 10 mg L–1) of Phe and Nap were tested and the population growth, photosynthetic capacity, pigment content, and morphological defense of T. obliquus were analyzed. The results indicated that Phe and Nap inhibited both the growth rate and formation of defensive colonies of T. obliquus induced by Daphnia grazing cues, and the inhibition ratio increased with concentration. Phe and Nap significantly shortened the defense colony formation time of T. obliquus. Phe and Nap significantly suppressed photosynthesis in the early stages; however, the photosynthetic efficiency recovered over time. These findings highlight the high sensitivity of grazing-induced colony formation in T. obliquus to Phe and Nap at non-lethal concentrations, which could affect the interactions between phytoplankton and zooplankton in aquatic ecosystems. Our study underscores the influence of Phe and Nap on the defense mechanisms of phytoplankton and the consequential effects on ecological interactions within freshwater ecosystems, providing insight into the complex impacts of pollutants on phytoplankton-zooplankton relationships. Therefore, it is necessary to consider interspecific interactions when assessing the potential negative effects of environmental pollutants on aquatic ecosystems.
