Rater comments tend to be qualitatively analyzed to indicate raters' application of rating scales. This study applied natural language processing (NLP) techniques to quantify meaningful, behavioral information from a corpus of rater comments and triangulated that information with a many-facet Rasch measurement (MFRM) analysis of rater scores. The data consisted of ratings on 987 essays by 36 raters (a total of 3948 analytic scores and 1974 rater comments) on a post-admission English Placement Test (EPT) at a large US university. We computed a set of comment-based features based on the analytic components and evaluative language the raters used to infer whether raters were aligned to the scale. For data triangulation, we performed correlation analyses between the MFRM measures of rater performance and the comment-based measures. Although the EPT raters showed overall satisfactory performance, we found meaningful associations between rater comments and performance features. In particular, raters with higher precision and fit to what the Rasch model predicts used more analytic components and used evaluative language more similar to the scale descriptors. These findings suggest that NLP techniques have the potential to help language testers analyze rater comments and understand rater behavior.

This study aimed to answer an ongoing validity question related to the use of nonstandard English accents in international tests of English proficiency and associated issues of test fairness. More specifically, we examined (1) the extent to which different or shared English accents had an impact on listeners' performances on the Duolingo listening tests and (2) the extent to which different English accents affected listeners' performances on two different task types. Speakers from four interlanguage English accent varieties (Chinese, Spanish, Indian English [Hindi], and Korean) produced speech samples for "yes/no" vocabulary and dictation Duolingo listening tasks. Listeners who spoke with these same four English accents were then recruited to take the Duolingo listening test items. Results suggested that there is a shared first language (L1) benefit effect overall, with comparable test scores between shared-L1 and inner-circle L1 accents, and no significant differences in listeners' listening performance scores across highly intelligible accent varieties. No task type effect was found. The findings provide guidance to better understand fairness, equality, and practicality of designing and administering high-stakes English tests targeting a diversity of accents.

Abstract Chimeric antigen receptor T (CAR-T) cell therapy has demonstrated promising efficacy in early trials for relapsed/refractory diffuse large B cell lymphoma (DLBCL). However, its efficacy in treating primary refractory DLBCL has not been comprehensively investigated, and the underlying resistance mechanisms remain unclear. Here, we report the outcomes of a phase I, open-label, single-arm clinical trial of relmacabtagene autoleucel (relma-cel), a CD19-targeted CAR-T cell product, with safety and efficacy as primary endpoints. Among the 12 enrolled patients, 8 experienced grade 4 hematologic toxicity of treatment-emergent adverse event. No grade ≥3 cytokine release syndrome or neurotoxicity occurred. Single-cell RNA sequencing revealed an increase proportion of C1QB-expressing macrophages in patients with progressive disease before CAR-T cell therapy. Cholesterol efflux from M2 macrophages was found to inhibit CAR-T cells cytotoxicity by inducing an immunosuppressive state in CD8+ T cells, leading to their exhaustion. Possible interactions between macrophages and CD8+ T cells, mediating lipid metabolism (AFR1-FAS), immune checkpoint activation, and T cell exhaustion (LGALS9-HAVCR2, CD86-CTLA4, and NECTIN2-TIGIT) were enhanced during disease progression. These findings suggest that cholesterol efflux from macrophages may trigger CD8+ T cell exhaustion, providing a rationale for metabolic reprogramming to counteract CAR-T treatment failure. This article investigated the effects of airborne-particle abrasion and nanosilica (nano-Si) infiltration treatment on the surface characteristics of dental zirconia. A total of 15 unsintered zirconia ceramic green bodies (10 × 10 × 3 mm) were divided into three groups (n = 5): Group C, no treatment after sintering; Group S, airborne-particle abrasion with 50-µm aluminum oxide particles after sintering; and Group N, infiltration of nano-Si followed by sintering and hydrofluoric acid (HF) etching. The zirconia disks' surface roughness was analyzed by atomic force microscopy (AFM). The surface morphology of the specimens was analyzed using scanning electron microscopy (SEM), and the chemical composition was analyzed by energy-dispersive x-ray (EDX). Data were statistically analyzed by Kruskal-Wallis test (P < .05). Zirconia surface treatments by infiltration of nano-Si, sintering, and HF etching showed multiple changes in the surface features. The surface roughness of Groups C, S, and N were 0.88 ± 0.07 µm, 1.26 ± 0.10 µm, and 1.69 ± 0.15 µm, respectively. The surface roughness of Group N was significantly higher than that of Groups C and S (P < .05). EDX analysis showed peaks that corresponded to silica (Si) after infiltration with colloidal Si that disappeared following acid etching. Infiltrating nano-Si increases the surface roughness of zirconia. The formation of retentive nanopores on the surface potentially improves the zirconia-resin cement bonding strengths. ABSTRACT: Meat qualities of free-range chicken (Xuan-Zhou) (XZ-FRC) are closely associated with slaughter age and directly influence the economic benefits of supplier and consumer's preference. Understanding of the relationship between meat qualities and ages will be of prime important to explore a better slaughter age of XZ-FRC. In this study, the quality traits of breast and thigh muscles from XZ-FRCs at 9 to 14 wk were analyzed to establish a relatively reliable method for selecting a better slaughter age. The results showed that the effects of slaughter ages on color (CIE L*, a* and b* values), shear force, centrifugal loss, and flavor of XZ-FRCs were significant (P < 0.05). There were greater differences in meat qualities, whatever breast or thigh muscles, between same or different ages. Eleven feature indexes used for colligation evaluation of slaughter age were selected by combining the quality characteristics and data analysis. The score of colligation evaluation for XZ-FRCs at 12 wk was higher than that at 9 and 14 wk, suggesting that the 12 wk was an optimal slaughter age. This work would provide a reference method that helps the producers of livestock and poultry to select a better slaughter age.

Abstract GAS41, a member of the human YEATS domain family, plays a pivotal role in human cancer development. It serves as a highly promising epigenetic reader, facilitating precise regulation of cell growth and development by recognizing essential histone modifications, including histone acetylation, benzoylation, succinylation, and crotonylation. Functional readouts of these histone modifications often coincide with cancer progression. In addition, GAS41 functions as a novel oncogene, participating in numerous signaling pathways. Here, we summarize the epigenetic functions of GAS41 and its role in the carcinoma progression. Moving forward, elucidating the downstream target oncogenes regulated by GAS41 and the developing small molecule inhibitors based on the distinctive YEATS recognition properties will be pivotal in advancing this research field.

In this article, we report the results of a study through which we collected English language teachers’ needs and wants to design an online language assessment course. Through a mixed-methods approach, we asked 20 teachers from four Latin American countries what they wanted to learn in the course. The teachers wanted a course in which they could address the challenges they faced in assessment; discuss and develop new ways to assess; and learn about authentic, valid, and ethical assessment. Therefore, the findings suggest that the teachers wanted a course that mixed theory, practice, and principles of assessment. Additionally, the course should address emerging topics in English language assessment, namely bilingual assessment and the assessment of learners with special educational needs.

Teprotumumab-trbw,a monoclonal antibody that acts on the insulin growth factor-Ⅰ receptor, was approved in 2020 for the treatment of thyroid-associated ophthalmopathy, but little is known about it in China. It is hoped to provide guidance for clinical use through the review of its molecular structure, pharmacokinetics, therapeutic mechanism, clinical research and safety. It inhibits immune inflammation by blocking thyroid-stimulating hormone receptor /insulin growth factor-Ⅰ receptor crosstalk signaling, so as to reduce the production of hyaluronic acid and inflammatory factors in response. It can also promote the apoptosis of retro-orbital fibroblasts/adipocytes and inhibit the expression of genes related to the synthesis of thyroid hormones, thereby significantly improving the clinical symptoms such as exophthalmos and diplopia. The common adverse reactions of Teprotumumab-trbw are muscle spasm, hyperglycemia, hearing loss and so on. Teprotumumab-trbw is effective and durable in the treatment of thyroid-associated ophthalmopathy, and patients with secondary treatment can also benefit from it, which provides a new way and hope for the treatment of thyroid-associated ophthalmopathy.

Bacterial wilt disease caused by Ralstonia solanacearum is a widespread, severe plant disease. Tomato (Solanum lycopersicum), one of the most important vegetable crops worldwide, is particularly susceptible to this disease. Biological control offers numerous advantages, making it a highly favorable approach for managing bacterial wilt. In this study, the results demonstrate that treatment with the biological control strain Bacillus subtilis R31 significantly reduced the incidence of tomato bacterial wilt. In addition, R31 directly inhibits the growth of R. solanacearum, and lipopeptides play an important role in this effect. The results also show that R31 can stably colonize the rhizosphere soil and root tissues of tomato plants for a long time, reduce the R. solanacearum population in the rhizosphere soil, and alter the microbial community that interacts with R. solanacearum. This study provides an important theoretical basis for elucidating the mechanism of B. subtilis as a biological control agent against bacterial wilt and lays the foundation for the optimization and promotion of other agents such as R31.

Abstract Chimeric antigen receptor (CAR) T-cell therapy has emerged as a promising novel therapeutic approach. However, primary and secondary resistance to CAR-T cell therapy is commonly encountered in various clinical trials. Despite the comprehensive studies to elucidate the mechanisms of resistance, effective resolution in clinical practice is still elusive. Inadequate persistence and subsequent loss of infused CAR-T cells are proposed major resistance mechanism associated with CAR-T cell treatment failure. Thus, we generated CAR-T cells armored with IL-7 to prolong the persistence of infused T-cells, particularly CD4 + T cells, and enhanced anti-tumor response. IL-7 increased CAR-T-cell persistence in vivo and contributed to the distinct T-cell cytotoxicity profile. Using mass cytometry (CyTOF), we further assessed the phenotypic and metabolic profiles of IL-7-secreting CAR-T cells, along with conventional CAR-T cells at the single-cell level. With in-depth analysis, we found that IL-7 maintained CAR-T cells in a less differentiated T-cell state, regulated distinct metabolic activity, and prevented CAR-T-cell exhaustion, which could be essential for CAR-T cells to maintain their metabolic fitness and anti-tumor response. Our findings thus provided clinical rationale to exploit IL-7 signaling for modulation and metabolic reprogramming of T-cell function to enhance CAR-T cell persistence and induce durable remission upon CAR-T cell therapy.

Thyroid-associated ophthalmopathy(TAO)is an organ-specific autoimmune disease, which will cause a series of symptoms to significantly reduce the health level and life quality of patients. The pathogenesis of TAO has not been fully clarified. At present, there is a lack of unified and mature treatment scheme of it. Indeed, T-helper 17 lymphocyte(Th17)cells, regulatory T(Treg)cells and their imbalance are closely related to the immunological pathogenesis of TAO. It is currently believed that the cytokines secreted by Th17 cells can not only promote the inflammatory response of TAO and the fibrosis of orbital connective tissue, but also inhibit the adipogenic differentiation of TAO orbital connective tissue. In addition, Treg cells mainly exert immunosuppressive effect on TAO and delay the disease progression. At the same time, there is a dynamic balance relationship between Th17 and Treg cells, the imbalance of Th17/Treg cells can trigger the occurrence and development of TAO. This paper mainly expounds the influence mechanism of Th17, Treg cells and their balance on TAO, and analyzes the reasons for the differences between different research results, so as to provide some reference for the study of the pathogenesis and clinical treatment of TAO.

During colonization of soil and plants, biocontrol bacteria can effectively regulate the physiological metabolism of plants and induce disease resistance. To illustrate the influence of Bacillus subtilis R31 on the quality, transcriptome and metabolome of sweet corn, field studies were conducted at a corn experimental base in Zhuhai City. The results show that, after application of B. subtilis R31, sweet corn was more fruitful, with a 18.3 cm ear length, 5.0 cm ear diameter, 0.4 bald head, 403.9 g fresh weight of single bud, 272.0 g net weight of single ear, and 16.5 kernels sweetness. Combined transcriptomic and metabolomic analyses indicate that differentially expressed genes related to plant-pathogen interactions, MAPK signaling pathway-plant, phenylpropanoid biosynthesis, and flavonoid biosynthesis were significantly enriched. Moreover, the 110 upregulated DAMs were mainly involved in the flavonoid biosynthesis and flavone and flavonol biosynthesis pathways. Our study provides a foundation for investigating the molecular mechanisms by which biocontrol bacteria enhance crop nutrition and taste through biological means or genetic engineering at the molecular level.

Adding particles to the Ni - P coating for forming a composite coating could further improve the mechanical properties, corrosion resistance and lubricating properties of the coating. Hence, Ni - P/WS2 composite coatings were prepared by chemical coprecipitation on the surface of Q235 steel, and the coatings were post - treated by heating. Subsequently, the influence mechanism of WS2 content in plating solution and heat treatment after precipitation on the hardness and self - lubrocating performance of Ni - P/WS2 composite coating was studied by electron microprobe (EPMA), X - ray diffractometer (XRD), scanning electron microscope (SEM) and hardness tester. Results showed that the content of P in the plating solution was stable, which was not affected by the concentration of WS2 particles in plating solution, and it was beneficial to precipitation of a proper amount of body centered cubic Ni3P hardening phase in the subsequent heat treatment, causing that the surface hardness of the composite coating was effectively improved. Moreover, WS2 particles in the coating played a lubricating role on the surface of the coating, which could effectively reduce the friction coefficient of the coating surface, and the heat treatment enabled the precipitation of body centered cubic Ni3P phase in the coating. Thus, the surface support for WS2 particles was enhanced, which was conducive to increase the self - lubrication of the coating surface and effectively reduce the friction coefficient.

Abstract Background Epstein-Barr virus (EBV) represents an important pathogenic factor of lymphoma and is significantly associated with poor clinical outcome of diffuse large B-cell lymphoma (DLBCL). Circular RNAs (circRNAs) play an essential role in lymphoma progression. However, the underlying mechanism of circRNA on DLBCL progression related to EBV remains largely unknown. Methods CircRNA was screened by high-throughput sequencing in tumor samples of 12 patients with DLBCL according to EBV infection status. Expression of circEAF2, as well as the relationship with clinical characteristics and prognosis, were further analyzed in tumor samples of 100 DLBCL patients using quantitative real-time PCR. Gain- and loss-of-function experiments were conducted to investigate the biological functions of circEAF2 both in vitro and in vivo. The underlying mechanism of circRNA on DLBCL progression were further determined by RNA sequencing, RNA pull down assay, dual-luciferase reporter assay, rescue experiments and western blotting. Results We identified a novel circRNA circEAF2, which was downregulated in EBV + DLBCL and negatively correlated with EBV infection and DLBCL progression. In EBV-positive B lymphoma cells, circEAF2 overexpression induced lymphoma cell apoptosis and sensitized lymphoma cells to epirubicin. As mechanism of action, circEAF2 specifically targeted EBV-encoded miR-BART19-3p, upregulated APC, and suppressed downstream β-catenin expression, resulting in inactivation of Wnt signaling pathway and inhibition of EBV + DLBCL cell proliferation. In EBV-positive B-lymphoma murine models, xenografted tumors with circEAF2 overexpression presented decreased Ki-67 positivity, increased cell apoptosis and retarded tumor growth. Conclusions CircEAF2 counteracted EBV + DLBCL progression via miR-BART19-3p/APC/β-catenin axis, referring circEAF2 as a potential prognostic biomarker. Therapeutic targeting EBV-encoded miRNA may be a promising strategy in treating EBV-associated lymphoid malignancies.

Elicited imitation (EI) has gained popularity with recent interests in the quest for efficient and flexible measures of second language (L2) proficiency. Despite the surge of interests, questions remain as to what specific linguistic knowledge, skills, and strategies EI measures. To contribute to this line of inquiry, this study explored the nature of strategy use and its effect on EI performance to elucidate the constructs of EI. Twenty-four L2 learners and eight native speakers of Chinese completed an EI test of Chinese and a strategy use questionnaire after the test. Qualitative analyses of the questionnaire responses revealed that participants mainly employed five types of strategies, including approach strategies, cognitive strategies, metacognitive strategies, communication strategies, and test-wiseness strategies. While native speakers reported the least number of strategies, higher-proficiency L2 learners reported more strategies than lower-proficiency L2 learners. We further subjected strategy use, along with participant proficiency level, item length, and item complexity level, to linear mixed-effects regression analyses. The results showed that participant proficiency level, item length, and item complexity level explained the largest test score variance; in contrast, strategy use of different types only accounted for a smaller proportion. The total number of cognitive strategies had a significant, positive effect on EI performance whereas the total number of metacognitive strategies had a significant, negative effect. These findings offer some insights into the nature of speech comprehension and production on EI and provide validity evidence for the use of EI as a language proficiency measure.

Abstract Long noncoding RNAs (lncRNAs) play an essential role in tumor progression. Few researches focused on the clinical and biological relevance of lncRNAs in peripheral T cell lymphoma (PTCL). In this research, a novel lncRNA (ENST00000503502) was identified overexpressed in the main subtypes of PTCL, and designated as T cell lymphoma-associated lncRNA1 (TCLlnc1). Serum TCLlnc1 was associated with extranodal involvement, high-risk International Prognostic Index, and poor prognosis of the patients. Both in vitro and in vivo, overexpression of TCLlnc1 promoted T-lymphoma cell proliferation and migration, both of which were counteracted by the knockdown of TCLlnc1 using small interfering RNAs. As the mechanism of action, TCLlnc1 directly interacted with transcription activator heterogeneous nuclear ribonucleoprotein D (HNRNPD) and Y-box binding protein-1 (YBX1) by acting as a modular scaffold. TCLlnc1/HNRNPD/YBX1 complex upregulated transcription of TGFB2 and TGFBR1 genes, activated the tumor growth factor-β signaling pathway, resulting in lymphoma progression, and might be a potential target in PTCL.

BACKGROUND: The citrus red mite, Panonychus citri (McGregor), a global pest of citrus, has developed different levels of resistance to various acaricides in the field. Abamectin is one of the most important insecticides/acaricides worldwide, targetting a wide number of insect and mite pests. The evolution of abamectin resistance in P. citri is threatening the sustainable use of abamectin for mite control. RESULTS: The abamectin resistant strain (NN‐Aba), derived from a field strain NN by consistent selection with abamectin, showed 4279‐fold resistance to abamectin compared to a relatively susceptible strain (SS) of P. citri. Cross‐resistance of NN‐Aba was observed between abamectin and emamectin benzoate, pyridaben, fenpropathrin and cyflumetofen. Inheritance analyses indicated that abamectin resistance in the NN‐Aba strain was autosomal, incompletely recessive and polygenic. The synergy experiment showed that abamectin toxicity was synergized by piperonyl butoxide (PBO), diethyl maleate (DEM) and tributyl phosphorotrithiotate (TPP) in the NN‐Aba strain, and synergy ratios were 2.72‐, 2.48‐ and 2.13‐fold, respectively. The glutathione‐S‐transferases activity in the NN‐Aba strain were significantly increased by 2.08‐fold compared with the SS strain. CONCLUSION: The abamectin resistance was autosomal, incompletely recessive and polygenic in P. citri. The NN‐Aba strain showed cross‐resistance to various acaricides with different modes of action. Metabolic detoxification mechanism participated in abamectin resistance in NN‐Aba strain. Abstract Background Cyclophosphamide, doxorubicin, vincristine, and prednisolone (CHOP)/CHOP-like chemotherapy is widely used in peripheral T cell lymphoma (PTCL). Here we conducted a phase 2, multicenter, randomized, controlled trial, comparing the efficacy and safety of CEOP/IVE/GDP alternating regimen with CEOP in newly diagnosed PTCL. Methods PTCL patients, except for anaplastic large cell lymphoma-anaplastic lymphoma kinase positive, were 1:1 randomly assigned to receive CEOP/IVE/GDP (CEOP, cyclophosphamide 750 mg/m2, epirubicin 70 mg/m2, vincristine 1.4 mg/m2 [maximum 2 mg] on day 1, and prednisone 60 mg/m2 [maximum 100 mg] on days 1–5 every 21 days, at the first and fourth cycle; IVE, ifosfamide 2000 mg/m2 on days 1–3, epirubicin 70 mg/m2 on day 1, and etoposide 100 mg/m2 on days 1–3 every 21 days, at the second and fifth cycle; and GDP, gemcitabine 1000 mg/m2 on days 1 and 8, cisplatin 25 mg/m2 on days 1–3, and dexamethasone 40 mg on days 1–4 every 21 days, at the third and sixth cycle) and CEOP (every 21 days for 6 cycles). Analysis of efficacy and safety was of the intent-to-treatment population. The primary endpoint was a complete response rate at the end of treatment. Meanwhile, whole exome sequencing and targeted sequencing were performed in 62 patients with available tumor samples to explore prognostic biomarkers in this cohort as an exploratory post hoc analysis. Results Among 106 patients, 53 each were enrolled to CEOP/IVE/GDP and CEOP. With 51 evaluable patients each in two groups, a complete response rate of the CEOP/IVE/GDP group was similar to that of the CEOP group (37.3% vs. 31.4%, p = 0.532). There was no difference in median progression-free survival (PFS; 15.4 months vs. 9.2 months, p = 0.122) or overall survival (OS; 24.3 months vs. 21.9 months, p = 0.178). Grade 3–4 hematological and non-hematological adverse events were comparable. Histone modification genes were most frequently mutated (25/62, 40.3%), namely KMT2D, KMT2A, SETD2, EP300, and CREBBP. With the rapid expansion of the Internet, intelligent question answering for information retrieval has once again gained widespread attention. However, current question answering models mainly focus on the general and common-sense questions in open domains and are incapable to effectively solve more complex professional domain questions. This paper proposed an integrated framework for Chinese intelligent question answering in restricted domains. The proposed model fused convolutional neural network and bidirectional long short-term memory network which performs efficient semantic analysis on the question pairs to extract more effective features of the text. Meanwhile, the coattention mechanism and attention mechanism were combined to obtain the semantic interaction and feature representation of the question pair for providing complete information for subsequent calculations. In addition, we introduced the method of question pair matching to implement the Chinese intelligent question answering in a restricted domain. Experiments were tested and evaluated on the open-source CCKS2018 dataset and our private self-built inverted pendulum control question answering (IPC-QA) dataset for automation control virtual learning environment. Experimental results confirm that the proposed models are efficient and achieve a high precision of 0.86042 and 0.8031 on CCKS2018 and IPC-QA respectively.

In China, baicalin is the main active component of Scutellaria baicalensis , which has been used in the treatment of inflammation-related diseases, such as inflammation-induced acute lung injury. However, its specific mechanism remains unclear. This study examined the protective effect of baicalin on LPS-induced inflammation injury of alveolar epithelial cell line A549 and explored its protective mechanism. Compared with the LPS-induced group, the proliferation inhibition rates of alveolar type II epithelial cell line A549 intervened by different concentrations of baicalin decreased significantly, as did the levels of inflammatory factors IL-6, IL-1β, prostaglandin 2 and TNF-α in the supernatant. The expression levels of inflammatory proteins inducible NO synthase (iNOS), NF-κB65, phosphorylated ERK (p-ERK1/2), and phosphorylated c-Jun N-terminal kinase (p-JNK1) significantly decreased, as did the protein expression of follistatin-like protein 1 (FSTL1). In contrast, expression of miR-200b-3p significantly increased in a dose-dependent manner. These results suggested that baicalin could significantly inhibit the expression of inflammation-related proteins and improve LPS-induced inflammatory injury in alveolar type II epithelial cells. The mechanism may be related to the inhibition of ERK/JNK inflammatory pathway activation by increasing the expression of miR-200b-3p. Thus, FSTL1 is the regulatory target of miR-200b-3p.

Abstract Background Osteosarcoma (OS) is the most common bone malignancy prevalent in children and young adults. MicroRNA-133b (miR-133b), through directly targeting the fibroblast growth factor receptor 1 (FGFR1), is increasingly recognized as a tumor suppressor in different types of cancers. However, little is known on the biological and functional significance of miR-133b/FGFR1 regulation in osteosarcoma. Methods The expressions of miR-133b and FGFR1 were examined by RT-qPCR and compared between 30 paired normal bone tissues and OS tissues, and also between normal osteoblasts and three OS cells lines, MG-63, U2OS, and SAOS-2. Using U2OS and MG-63 as the model system, the functional significance of miR-133b and FGFR1 was assessed on cell viability, proliferation, apoptosis, migration/invasion, and epithelial–mesenchymal transition (EMT) by overexpressing miR-133b and down-regulating FGFR1 expression, respectively. Furthermore, the signaling cascades controlled by miR-133b/FGFR1 were examined. Results miR-133b was significantly down-regulated while FGFR1 robustly up-regulated in OS tissues and OS cell lines, when compared to normal bone tissues and normal osteoblasts, respectively. Low miR-133b expression and high FGFR1 expression were associated with location of the malignant lesion, advanced clinical stage, and distant metastasis. FGFR1 was a direct target of miR-133b. Overexpressing miRNA-133b or knocking down FGFR1 significantly reduced the viability, proliferation, migration/invasion, and EMT, but promoted apoptosis of both MG-63 and U2OS cells. Both the Ras/MAPK and PI3K/Akt intracellular signaling cascades were inhibited in response to overexpressing miRNA-133b or knocking down FGFR1 in OS cells. Conclusion miR-133b, by targeting FGFR1, presents a plethora of tumor suppressor activities in OS cells. Boosting miR-133b expression or reducing FGFR1 expression may benefit OS therapy.

This paper provides a narrative review of empirical research on the assessment of speaking proficiency published in selected journals in the field of language assessment. A total of 104 published articles on speaking assessment were collected and systematically analyzed within an argument-based validation framework (Chapelle et al., 2008). We examined how the published research is represented in the six inferences of this framework, the topics that were covered by each article, and the research methods that were employed in collecting the backings to support the assumptions underlying each inference. Our analysis results revealed that: (a) most of the collected articles could be categorized into the three inferences of evaluation, generalization, and explanation; (b) the topics most frequently explored by speaking assessment researchers included the constructs of speaking ability, rater effects, and factors that affect spoken performance, among others; (c) quantitative methods were more frequently employed to interrogate the inferences of evaluation and generalization whereas qualitative methods were more frequently utilized to investigate the explanation inference. The paper concludes with a discussion of the implications of this study in relation to gaining a more nuanced understanding of task- or domain-specific speaking abilities, understanding speaking assessment in classroom contexts, and strengthening the interfaces between speaking assessment, and teaching and learning practices.

Gastric cancer is one of the most frequently diagnosed cancer and poses a serious threat to health system in the world. Upregulation of meningioma-associated protein (MAC30) has been found in many solid tumors and can regulate the proliferation, differentiation, and apoptosis of different tumor cells. Quantitative polymerase chain reaction (qPCR) was used to detect the expression of MAC30 in 68 patients with gastric cancer and their adjacent tissues. Lentiviral vector pGCSIL-shMAC30-GFP of the RNA interference (RNAi) of the MAC30 gene was transfected into gastric cancer BGC-823 cell line and the expression of lentivirus label protein GFP was observed via fluorescence microscope, while cell proliferation and apoptosis were determined with flow cytometry and MTT assay, respectively. Also, related protein expressions on Wnt/β-catenin signaling pathway were analyzed by Western blot method. The expression of MAC30 was abnormally elevated in gastric cancer tissues, while interfering of its expression could significantly inhibit the proliferation of gastric cancer BGC-823 cell line. However, the promotion of apoptosis by mitochondrial pathway was mediated by Bax/Bcl-2 upregulation. Present work showed the effect of downregulated MAC30 expression on proliferation and apoptosis of gastric cancer cell through Wnt/β-catenin signaling pathway. Thus, this investigation provides an experimental basis for future development of chemotherapeutic agent on gastric cancer.

Abstract White grubs are largely unsolved problems in vegetable and tuber production in East Africa. Novel Rwandan as well as international entomopathogenic nematodes (EPNs) were screened in nine laboratory bioassays and two small-scale field trials in 2014 and 2015. Soil-based laboratory bioassays revealed that all EPNs were able to infect Anomala graueri larvae (Coleoptera: Scarabeidae), although a relatively high number of infective juveniles were needed. Rwandan EPNs were as infectious as their corresponding international strains. At 100 infective juveniles per larva, the Heterorhabditis bacteriophora strains (Rwanda14-N-C4a and international H06) as well as the international Steinernema carpocapsae All caused 18 to 22% grub mortality within 7 days compared to the control. At 1000 infective juveniles, both H. bacteriophora strains as well as the Rwandan S. carpocapsae RW14-G-R3a-2 killed 34 up to 58%. The Rwandan Steinernema RW14-M-C2a-3 least performed in the bioassays (2 to 6%). In two Irish potato fields, the into-soil-applied 1 × 109 infective juvenile Steinernema RW14-M-C2b-1 per hectare reduced 29 ± 33% and 96 ± 3% of grubs within 30 and 60 days, respectively. About 1 up to 2.5 × 109 infective juvenile Steinernema longicaudum X7 per hectare reduced 77 up to 85% of grubs until day 30, respectively, and 82 up to 95% until day 60. Avermectin + Chlorpyrifos tuber coatings reduced 39% of grubs, and Fipronil + Chlorpyrifos reduced 27%, whereas handpicking did not help much. In conclusion, at least Steinernema RW14-M-C2b-1 and S. longicaudum X7 are promising for managing white grubs in tuber production, this is at a rate of at least 1.5 × 109 EPNs per hectare, but H. bacteriophora RW14-N-C4a needs further field research. Findings will support the biocontrol product development in Rwanda, including registration if any would be needed for native macrobial biocontrol agents.
