IntroductionBroadly neutralising antibodies are promising candidates for preventing and treating Human Immunodeficiency Virus/Acquired Immunodeficiency Syndrome (HIV/AIDS), as an alternative to or in combination with antiretroviral therapy (ART). These mAbs bind to sites on the virus essential for virus attachment and entry, thereby inhibiting entry into the host cell. However, the cost and availability of monoclonal antibodies, especially combinations of antibodies, hampers implementation of anti-HIV bNAb therapies in low- to middle- income countries (LMICs) where HIV-1 prevalence is highest.MethodsWe have produced three HIV broadly neutralizing antibodies (bNAbs), 10-1074, VRC01 and 3BNC117 in the Nicotiana benthamiana transient expression system. The impact of specific modifications to enhance potency and efficacy were assessed. To prolong half-life and increase bioavailability, a M252Y/S254T/T256E (YTE) or M428L/N434S (LS) mutation was introduced. To increase antibody dependent cellular cytotoxicity (ADCC), we expressed an afucosylated version of each antibody using a glycoengineered plant line.ResultsThe majority of bNAbs and their variants could be expressed at yields of up to 47 mg/kg. Neither the expression system nor the modifications impacted the neutralization potential of the bNAbs. Afucosylated bNAbs exhibit enhanced ability to bind to FcγRIIIa and trigger ADCC, regardless of the presence of Fc amino acid mutations. Lastly, we demonstrated that Fc-modified variants expressed in plants show enhanced binding to FcRn, which results in a favourable in vivo pharmacokinetic profile compared to their unmodified counterparts. ConclusionTobacco plants are suitable expression hosts for anti-HIV bNAbs with increased efficacy and an improved pharmacokinetic profile.

The dawn of antibody therapy was heralded by the rise of IgG therapeutics. However, other antibody classes are at our disposal—one of the most exciting is IgA and is the most abundant antibody class within humans. Unlike IgG, it is uniquely specialized for mucosal applications due to its ability to form complex Secretory IgA (SIgA) molecules. Since the mucosa is constantly exposed to potential infectious agents, SIgA is pivotal to disease prevention as an important component of the mucosal barrier. Compared to IgG, SIgA has proven superior effectiveness in mucosal surfaces, such as the airway epithelium or the harsh gut environment. Despite this, hurdles associated with low yield and challenging purification have blocked SIgA therapeutic advancement. However, as a result of new antibody engineering strategies, we are approaching the next generation of (IgA-based) antibody therapies. Strategies include fine-tuning SIgA assembly, exploring different production platforms, genetic engineering to improve purification, and glycoengineering of different components. Due to its stability in mucosal environments, SIgA therapeutics would revolutionize passive mucosal immunotherapy—an avenue still underexploited by current therapeutics. This chapter will focus on the current perspectives of SIgA engineering and explore different approaches to unlocking the full therapeutic potential of SIgAs.

Molecular pharming in plants offers exciting possibilities to address global access to modern biologics. However, differences in the

Nicotiana tabacum (the tobacco plant ) has numerous advantages for molecular farming, including rapid growth, large biomass and the possibility of both cross- and self-fertilization. In addition, genetic transformation and tissue culture protocols for regeneration of transgenic plants are well-established. Here, we describe the production of transgenic tobacco using Agrobacterium tumefaciens and the analysis of recombinant proteins, either in crude plant extracts or after purification, by enzyme-linked immunosorbent assays, sodium dodecyl sulfate polyacrylamide gel electrophoresis with western blotting and surface plasmon resonance.

Deforestation and forest degradation lead to an increase in the level of carbon in the atmosphere and disrupted the global carbon cycle. The tropical forest has received a lot of interest since it contributes around 60% of the total global forest carbon. By enhancing carbon sink, tropical forests have a great potential in mitigating climate change. Assessing aboveground biomass (AGB) and carbon stock through field inventories is crucial for this purpose as it provides the most accurate result. The research was conducted at Danum Valley Conservation Area and INFAPRO in Sabah, Malaysia. An earlier study over 35 years ago at this site suggests that restored forests accrue AGB at twice the rate of regenerating forests, though the cause of this difference between treatments is unclear. Thus, this study will focus on three principal study sites which are restored, naturally regenerating and old-growth forests to determine the forest’s potential to sequester and store carbon in the forest ecosystem. These three sites were chosen because it is a well-established plot from the earlier inventory over the last seven years. The field measuring method is a non-destructive methodology. Tree parameters such as diameter at breast height (DBH), tree height and tree species diversity were collected for calculating AGB using a species-specific allometric equation. Results showed a positive correlation between tree species, diameter at breast height, and biomass/carbon stock across three different forest treatments. The active restoration increases up to 151% carbon stock whilst the old-growth forest increased by 34% and natural regeneration increased by 73%, which active restoration can be the best solution for forest treatment. The outcome of this study will increase the ability of forest authorities and the Malaysian government to effective monitoring of carbon stock for establishing reliable standard guidelines in measuring deforestation and forest degradation toward achieving sustainable forest management.

Ultra-hypofractionated radiotherapy (UHF-RT) is widely utilized in men with localized prostate cancer (PCa). There are limited data in Asian cohorts. We report the outcomes of a single-arm, phase II trial of UHF-RT from an Asian center.We recruited men with histologically confirmed, nonmetastatic localized PCa. UHF-RT regimens were 36.25 Gy (Cohort A) and 37.5 Gy (Cohort B) delivered in five fractions every other day over 1.5-2.5 weeks. Primary endpoint was physician-scored late genitourinary (GU) and gastrointestinal (GI) adverse events (AEs). Quality-of-life (QoL) was assessed by Expanded Prostate Cancer Index Composite (EPIC) at baseline, 1- and 2-year post-UHF-RT.Between March 2014 and August 2019, 105 men were recruited; four were subsequently excluded from analysis. Median age was 68.0 (Interquartile range (IQR): 63.8-73.0) years. 26 (24.8%) and 68 (64.8%) men had NCCN-defined low-and intermediate-risk PCa, respectively. No late ≥G3 GU or GI toxicities were reported in both cohorts. Peak incidence of acute ≥G2 GU AEs at 14 days post-UHF-RT was 23.6% (17/72) and 24.0% (6/25) in Cohorts A and B, respectively; ≥G2 GI AEs were observed in 9.7% (7/72) and 36.0% (9/25), respectively. Late ≥G2 GU and GI AEs occurred in 4.7% and 3.1% of Cohort A patients, and 5.0% in Cohort B at 12 months, with no AEs at 24 months. EPIC scores changed minimally across all domains. At a median follow-up of 44.9 months, we recorded one (1.3%) biochemical relapse by the Phoenix criteria (Cohort A).UHF-RT is well tolerated in Asian men and can be a recommended fractionation schema for localized PCa.

Dengue fever (DF) is an important public health problem, and it is now endemic in more than 100 countries worldwide. Dengue associated neurological complication is estimated to be affecting 0.5 to 6.2% of patients. Even though this is rare, neurological manifestation of DF is an increasingly recognized entity in recent years due to significant mortality and morbidity reported/seen. Reported central nervous system manifestations due to dengue include encephalitis, encephalopathy, myelitis, myositis, acute disseminated encephalomyelitis, Guillain-Barré syndrome, stroke and etc. We report here a case of acute necrotizing encephalopathy secondary to DF in a previously healthy 12-year-old girl.

Passive immunization with antibodies is a promising approach against enterotoxigenic Escherichia coli diarrhea, a prevalent disease in LMICs. The objective of this study was to investigate expression of a monoclonal anti-ETEC CfaE secretory IgA antibody in N. benthamiana plants, with a view to facilitating access to ETEC passive immunotherapy. SIgA1 and SIgA2 forms of mAb 68–81 were produced by co-expressing the light and engineered heavy chains with J chain and secretory component in N. benthamiana. Antibody expression and assembly were compared with CHO-derived antibodies by SDS-PAGE, western blotting, size-exclusion chromatography and LC-MS peptide mapping. N-linked glycosylation was assessed by rapid fluorescence/mass spectrometry and LC-ESI-MS. Susceptibility to gastric digestion was assessed in an in vitro model. Antibody function was compared for antigen binding, a Caco-2 cell-based ETEC adhesion assay, an ETEC hemagglutination inhibition assay and a murine in vivo challenge study. SIgA1 assembly appeared superior to SIgA2 in plants. Both sub-classes exhibited resistance to degradation by simulated gastric fluid, comparable to CHO-produced 68–61 SIgA1. The plant expressed SIgAs had more homogeneous N-glycosylation than CHO-derived SIgAs, but no alteration of in vitro functional activity was observed, including antibodies expressed in a plant line engineered for mammalian-like N glycosylation. The plant-derived SIgA2 mAb demonstrated protection against diarrhea in a murine infection model. Although antibody yield and purification need to be optimized, anti-ETEC SIgA antibodies produced in a low-cost plant platform are functionally equivalent to CHO antibodies, and provide promise for passive immunotherapy in LMICs.

Background HIV remains one of the most important health issues worldwide, with almost 40 million people living with HIV. Although patients develop antibodies against the virus, its high mutation rate allows evasion of immune responses. Some patients, however, produce antibodies that are able to bind to, and neutralise different strains of HIV. One such ‘broadly neutralising’ antibody is ‘N6’. Identified in 2016, N6 can neutralise 98% of HIV-1 isolates with a median IC50 of 0.066 µg/mL. This neutralisation breadth makes N6 a very promising therapeutic candidate. Results N6 was expressed in a glycoengineered line of N. benthamiana plants (pN6) and compared to the mammalian cell-expressed equivalent (mN6). Expression at 49 mg/kg (fresh leaf tissue) was achieved in plants, although extraction and purification are more challenging than for most plant-expressed antibodies. N-glycoanalysis demonstrated the absence of xylosylation and a reduction in α(1,3)-fucosylation that are typically found in plant glycoproteins. The N6 light chain contains a potential N-glycosylation site, which was modified and displayed more α(1,3)-fucose than the heavy chain. The binding kinetics of pN6 and mN6, measured by surface plasmon resonance, were similar for HIV gp120. pN6 had a tenfold higher affinity for FcγRIIIa, which was reflected in an antibody-dependent cellular cytotoxicity assay, where pN6 induced a more potent response from effector cells than that of mN6. pN6 demonstrated the same potency and breadth of neutralisation as mN6, against a panel of HIV strains. Conclusions The successful expression of N6 in tobacco supports the prospect of developing a low-cost, low-tech production platform for a monoclonal antibody cocktail to control HIV in low-to middle income countries. Summary In this study, a strategy based on polymeric immunoglobulin G scaffolds (PIGS) was used to produce a vaccine candidate for Mycobacterium tuberculosis. A genetic fusion construct comprising genes encoding the mycobacterial Ag85B antigen, an immunoglobulin γ‐chain fragment and the tailpiece from immunoglobulin μ chain was engineered. Expression was attempted in Chinese Hamster Ovary (CHO) cells and in Nicotiana benthamiana. The recombinant protein assembled into polymeric structures (TB‐PIGS) in N. benthamiana, similar in size to polymeric IgM. These complexes were subsequently shown to bind to the complement protein C1q and FcγRs with increased affinity. Modification of the N‐glycans linked to TB‐PIGS by removal of xylose and fucose residues that are normally found in plant glycosylated proteins also resulted in increased affinity for low‐affinity FcγRs. Immunization studies in mice indicated that TB‐PIGS are highly immunogenic with and without adjuvant. However, they did not improve protective efficacy in mice against challenge with M. tuberculosis compared to conventional vaccination with BCG, suggesting that additional or alternative antigens may be needed to protect against this disease. Nevertheless, these results establish a novel platform for producing polymeric antigen‐IgG γ‐chain molecules with inherent functional characteristics that are desirable in vaccines.

Background: An increasing number and proportion of cancer patients with apparently localised disease are treated with chemotherapy and radiation therapy in contemporary oncology practice. In a pilot study of radiation-induced sarcoma (RIS) patients, we demonstrated that chemotherapy was associated with a reduced time to development of RIS. We now present a multi-centre collaborative study to validate this association. Methods: This was a retrospective cohort study of RIS cases across five large international sarcoma centres between 1 January 2000 to 31 December 2014. The primary endpoint was time to development of RIS. Results: We identified 419 patients with RIS. Chemotherapy for the first malignancy was associated with a shorter time to RIS development (HR 1.37; 95% CI: 1.08–1.72; P=0.009). Degeneracy in the genetic code allows multiple codon sequences to encode the same protein. Codon usage bias in genes is the term given to the preferred use of particular synonymous codons. Synonymous codon substitutions had been regarded as "silent" as the primary structure of the protein was not affected; however, it is now accepted that synonymous substitutions can have a significant effect on heterologous protein expression. Codon optimization, the process of altering codons within the gene sequence to improve recombinant protein expression, has become widely practised. Multiple inter-linked factors affecting protein expression need to be taken into consideration when optimizing a gene sequence. Over the years, various computer programmes have been developed to aid in the gene sequence optimization process. However, as the rulebook for altering codon usage to affect protein expression is still not completely understood, it is difficult to predict which strategy, if any, will design the "optimal" gene sequence. In this review, codon usage bias and factors affecting codon selection will be discussed and the evidence for codon optimization impact will be reviewed for recombinant protein expression using plants as a case study. These developments will be relevant to all recombinant expression systems; however, molecular pharming in plants is an area which has consistently encountered difficulties with low levels of recombinant protein expression, and should benefit from an evidence based rational approach to synthetic gene design. The sinterability of manganese-doped 8 mol% yttria-stabilized zirconia ranging from 0.5–5 wt% was investigated. The role of dopants in the samples was studied in terms of phase stability, microstructures, and mechanical properties. Accordingly, 0.5, 1, 3, and 5 wt% manganese were doped into yttria-stabilized zirconia and heated at temperatures ranging from 1300 °C to 1550 °C. The study revealed that the milling process reduced powder particle size from 0.54 μm to 0.16 μm. The grain size of the samples increased with increasing sintering temperature, and adding manganese dopants promoted further grain growth. When sintered at 1450 °C, 3 wt% manganese enhanced the densification of the sample to a relative density of 95.8% and Vickers hardness peaked at 14.4 GPa. The samples with manganese dopants exhibited fracture toughness values of 4.1–4.6 MPa · m 1/2 when sintered at temperatures above 1450 °C, regardless of dopant content. Zircon was found on the surface of the sintered sample with over 3 wt% manganese addition but did not disrupt zirconia phase stability. The study showed that the mechanical properties of yttria-stabilized zirconia improved when manganese dopants were added.

Biotechnology has transformed the potential for plants to be a manufacturing source of pharmaceutical compounds. Now, with transgenic and transient expression techniques, virtually any biologic, including vaccines and therapeutics, could be manufactured in plants. However, uncertainty over the regulatory path for such new pharmaceuticals has been a deterrent. Consideration has been given to using alternative regulatory paths, including those for nutraceuticals or cosmetic agents. This review will consider these possibilities, and discuss the difficulties in establishing regulatory guidelines for new pharmaceutical manufacturing technologies.

Summary Plants can provide a cost‐effective and scalable technology for production of therapeutic monoclonal antibodies, with the potential for precise engineering of glycosylation. Glycan structures in the antibody Fc region influence binding properties to Fc receptors, which opens opportunities for modulation of antibody effector functions. To test the impact of glycosylation in detail, on binding to human Fc receptors, different glycovariants of VRC01, a broadly neutralizing HIV monoclonal antibody, were generated in Nicotiana benthamiana and characterized. These include glycovariants lacking plant characteristic α1,3‐fucose and β1,2‐xylose residues and glycans extended with terminal β1,4‐galactose. Surface plasmon resonance‐based assays were established for kinetic/affinity evaluation of antibody–FcγR interactions, and revealed that antibodies with typical plant glycosylation have a limited capacity to engage FcγRI, FcγRIIa, FcγRIIb and FcγRIIIa; however, the binding characteristics can be restored and even improved with targeted glycoengineering. All plant‐made glycovariants had a slightly reduced affinity to the neonatal Fc receptor (FcRn) compared with HEK cell‐derived antibody. However, this was independent of plant glycosylation, but related to the oxidation status of two methionine residues in the Fc region. This points towards a need for process optimization to control oxidation levels and improve the quality of plant‐produced antibodies.

Montane tropical soils are a large carbon (C) reservoir, acting as both a source and a sink of CO2. Enhanced CO2 emissions originate, in large part, from the decomposition and losses of soil organic matter (SOM) following anthropogenic disturbances. Therefore, quantitative knowledge of the stabilization and decomposition of SOM is necessary in order to understand, assess and predict the impact of land management in the tropics. In particular, labile SOM is an early and sensitive indicator of how SOM responds to changes in land use and management practices, which could have major implications for long-term carbon storage and rising atmospheric CO2 concentrations. The aim of this study was to investigate the impacts of grazing and fire history on soil C dynamics in the Peruvian montane grasslands, an understudied ecosystem, which covers approximately a quarter of the land area in Peru. A density fractionation method was used to quantify the labile and stable organic matter pools, along with soil CO2 flux and decomposition measurements. Grazing and burning together significantly increased soil CO2 fluxes and decomposition rates and reduced temperature as a driver. Although there was no significant effect of land use on total soil C stocks, the combination of burning and grazing decreased the proportion of C in the free light fraction (LF), especially at the lower depths (10–20 and 20–30 cm). In the control soils, 20 % of the material recovered was in the free LF, which contained 30 % of the soil C content. In comparison, the burnt–grazed soil had the smallest recovery of the free LF (10 %) and a significantly lower C content (14 %). The burnt soils had a much higher proportion of C in the occluded LF (12 %) compared to the not-burnt soils (7 %) and there was no significant difference among the treatments in the heavy fraction (F) ( ∼ 70 %). The synergistic effect of burning and grazing caused changes to the soil C dynamics. CO2 fluxes were increased and the dominant temperature driver was obscured by some other process, such as changes in plant C and N allocation. In addition, the free LF was reduced when these two anthropogenic activities took place on the same site – most likely a result of reduced detritus being incorporated into the soil. A positive finding from this study is that the total soil C stocks were not significantly affected and the long-term (+10 years) C storage in the occluded LF and heavy F were not negatively impacted. Possibly this is because of low-intensity fire, fire-resilient grasses and because the grazing pressure is below the threshold necessary to cause severe degradation.

Key message The potent anti-HIV microbicide griffithsin was expressed to high levels in tobacco chloroplasts, enabling efficient purification from both fresh and dried biomass, thus providing storable material for inexpensive production and scale-up on demand. Abstract The global HIV epidemic continues to grow, with 1.8 million new infections occurring per year. In the absence of a cure and an AIDS vaccine, there is a pressing need to prevent new infections in order to curb the disease. Topical microbicides that block viral entry into human cells can potentially prevent HIV infection. The antiviral lectin griffithsin has been identified as a highly potent inhibitor of HIV entry into human cells. Here we have explored the possibility to use transplastomic plants as an inexpensive production platform for griffithsin. We show that griffithsin accumulates in stably transformed tobacco chloroplasts to up to 5% of the total soluble protein of the plant. Griffithsin can be easily purified from leaf material and shows similarly high virus neutralization activity as griffithsin protein recombinantly expressed in bacteria. We also show that dried tobacco provides a storable source material for griffithsin purification, thus enabling quick scale-up of production on demand. Protein microbicides against HIV can help to prevent infection but they are required in large, repetitive doses. This makes current fermenter-based production systems prohibitively expensive. Plants are advantageous as production platforms because they offer a safe, economical and scalable alternative, and cereals such as rice are particularly attractive because they could allow pharmaceutical proteins to be produced economically and on a large scale in developing countries. Pharmaceutical proteins can also be stored as unprocessed seed, circumventing the need for a cold chain. Here, we report the development of transgenic rice plants expressing the HIV-neutralizing antibody 2G12 in the endosperm. Surprisingly for an antibody expressed in plants, the heavy chain was predominantly aglycosylated. Nevertheless, the heavy and light chains assembled into functional antibodies with more potent HIV-neutralizing activity than other plant-derived forms of 2G12 bearing typical high-mannose or plant complex-type glycans. Immunolocalization experiments showed that the assembled antibody accumulated predominantly in protein storage vacuoles but also induced the formation of novel, spherical storage compartments surrounded by ribosomes indicating that they originated from the endoplasmic reticulum. The comparison of wild-type and transgenic plants at the transcriptomic and proteomic levels indicated that endogenous genes related to starch biosynthesis were down-regulated in the endosperm of the transgenic plants, whereas genes encoding prolamin and glutaredoxin-C8 were up-regulated. Our data provide insight into factors that affect the functional efficacy of neutralizing antibodies in plants and the impact of recombinant proteins on endogenous gene expression. Eyelid reconstruction is complex and challenging since it is not only for structural and functional restoration, but also for an acceptable aesthetic result. In full thickness eyelid injuries, it will involve both anterior and posterior lamella. Therefore, when reconstructing the defect, it requires at least two layers; one will be a flap with blood supply, and the other can be a free graft. In this case, a rotational advancement cheek flap and composite graft were used to reconstruct the lower eyelid.

In the present work, pure forsterite (Mg2SiO4) powder was successfully synthesised via ball milling and subsequent heat treatment as an evident from X-ray diffraction analysis. The synthesised powder were formed by powder agglomerates that were made up of loosely packed fine particles. Then, the forsterite green bodies were sintered at 1200 and 1400°C with two different ramp rates (2 and 10°C per minute) via conventional pressureless sintering. No decomposition of forsterite phase was observed at both ramp rates. This study revealed that lower ramp rate (2°C per minute) possessed minor improvement on the mechanical properties only at lower sintering temperature (1200°C), whereas 10°C per minute ramp rate gave a significant enhancement on the mechanical properties of forsterite at 1400°C. A high fracture toughness of ∼4·9 MPa m1/2 and Vickers hardness of ∼7·1 GPa were obtained for forsterite samples sintered at 1400°C with a ramp rate of 10°C per minute.

The loop tack, peel strength and shear strength of cross-linked epoxidized natural rubber (ENR-50)/acrylonitrile-butadiene rubber (NBR) blend adhesives were studied in the presence of coumarone-indene resin. Benzoyl peroxide was used as the cross-linking agent with dosage ranging from 1 to 5 parts per hundred rubber by weight (phr). Toluene was used as the solvent throughout the investigation. A SHEEN hand coater was used to coat the adhesive on a polyethylene terephthalate substrate at 60 and 120 μm coating thickness. The adhesive was cured at 80°C for 30 min prior to testing on a Lloyd adhesion tester operating at various testing rates from 10 to 60 cm min−1. Results show that loop tack and peel strength of the ENR-50/NBR adhesives pass through a maximum value at 4 phr of benzoyl peroxide dosage. The loop tack, peel strength, and shear strength of crosslinked epoxidized natural rubber (ENR 25)/ethylene-propylene-diene rubber (EPDM) blend adhesives were investigated. Coumarone-indene resin, toluene, and benzoyl peroxide were used as the tackifier, solvent, and crosslinking agent, respectively, throughout the experiment. The adhesive was coated on a polyethylene terephthalate (PET) substrate using a SHEEN hand coater at 60 μm and 120 μm coating thickness. It was cured at 80°C for 30 minutes before testing on a Lloyd adhesion tester operating at testing rates from 10 to 60 cm min−1. Results show that loop tack and peel strength of the ENR 25/EPDM adhesive pass through a maximum value at 2 parts per hundred parts of rubber (phr) of benzoyl peroxide content. This observation is attributed to the increase in crosslinking which enhances the cohesive strength of the adhesive. Further addition of the crosslinking agent decreases the tack and peel strength due to the decrease in wettability of the over-crosslinked adhesive. Shear strength, however, increases steadily with benzoyl peroxide content, an observation which is associated with the steady increase in the cohesive strength. The adhesion properties increase with increasing coating thickness and testing rate.

Four major developments have taken place in the world of Molecular Pharming recently. In the USA, the DARPA initiative challenged plant biotechnology companies to develop strategies for the large-scale manufacture of influenza vaccines, resulting in a successful Phase I clinical trial; in Europe the Pharma-Planta academic consortium gained regulatory approval for a plant-derived monoclonal antibody and completed a first-in-human phase I clinical trial; the Dutch pharmaceutical company Synthon acquired the assets of Biolex Therapeutics, an established Molecular Pharming company with several clinical candidates produced in their proprietary LEX system based on aquatic plants; and finally, the Israeli biotechnology company Protalix Biotherapeutics won FDA approval for the commercial release of a recombinant form of the enzyme glucocerebrosidase produced in carrot cells, the first plant biotechnology-derived biopharmaceutical in the world approved for the market. Commercial momentum is gathering pace with additional candidates now undergoing or awaiting approval for phase III clinical trials. Filling the product pipeline is vital to establish commercial sustainability, and the selection of appropriate target products for Molecular Pharming will be a critical factor. An interesting feature of the four stories outlined above is that they span the use of very different platform technologies addressing different types of molecules which aim to satisfy distinct market demands. In each case, Molecular Pharming was an economically and technically suitable approach, but this decisionmaking process is not necessarily straightforward. Although the various technologies available to Molecular Pharming are broad ranging and flexible, competing technologies are better established, so there needs to be a compelling reason to move into plants. It is most unlikely that plant biotechnology will be the answer for the whole biologics field. In this article, we discuss the current plant biotechnology approaches that appear to hold the greatest promise and in doing so attempt to define the product areas that are most likely to benefit from different Molecular Pharming technologies.

Barium Strontium Titanate (BST) thin film has been prepared by using sol-gel method. The samples are prepared with 2 different deposition layers (1 layer and 4 layer) and annealing temperature (600°C and 800°C) with Ba0.5Sr0.5TiO3 solution. Physical and electrical characterization of all the samples is done. The results showed that the grain size and surface roughness of the samples increased as the deposition layer and annealing temperature increased. In addition, the dielectric constant of the samples also increased as the deposition layer and annealing temperature increased. Thus, the physical and electrical characteristics of the thin films are related one to another.

Biotechnology has transformed the potential for plants to be a manufacturing source of pharmaceutical compounds. Now, with transgenic and transient expression techniques, virtually any biologic, including vaccines and therapeutics, could be manufactured in plants. However, uncertainty over the regulatory path for such new pharmaceuticals has been a deterrent. Consideration has been given to using alternative regulatory paths, including those for nutraceuticals or cosmetic agents. This review will consider these possibilities, and discuss the difficulties in establishing regulatory guidelines for new pharmaceutical manufacturing technologies.

Degeneracy in the genetic code allows multiple codon sequences to encode the same protein. Codon usage bias in genes is the term given to the preferred use of particular synonymous codons. Synonymous codon substitutions had been regarded as "silent" as the primary structure of the protein was not affected; however, it is now accepted that synonymous substitutions can have a significant effect on heterologous protein expression. Codon optimization, the process of altering codons within the gene sequence to improve recombinant protein expression, has become widely practised. Multiple inter-linked factors affecting protein expression need to be taken into consideration when optimizing a gene sequence. Over the years, various computer programmes have been developed to aid in the gene sequence optimization process. However, as the rulebook for altering codon usage to affect protein expression is still not completely understood, it is difficult to predict which strategy, if any, will design the "optimal" gene sequence. In this review, codon usage bias and factors affecting codon selection will be discussed and the evidence for codon optimization impact will be reviewed for recombinant protein expression using plants as a case study. These developments will be relevant to all recombinant expression systems; however, molecular pharming in plants is an area which has consistently encountered difficulties with low levels of recombinant protein expression, and should benefit from an evidence based rational approach to synthetic gene design. Phase equilibrium calculations are often involved in the design, simulation, and optimization of chemical processes. Reported methods for these calculations are based on either equation-solving or Gibbs free energy minimization approaches. The main objective of this work is to compare selected methods for these two approaches, in terms of reliability to find the correct solution, computational time, and number of K-value evaluations. For this, four equation-solving and three free minimization methods have been selected and applied to commonly encountered vapour–liquid equilibrium (VLE), liquid–liquid equilibrium (LLE), and vapour–liquid–liquid equilibrium (VLLE) examples involving multiple components and popular thermodynamic models. Detailed results show that the equation-solving method based on the Rachford–Rice formulation accompanied by mean value theorem and Wegstein's projection is reliable and efficient for two-phase equilibrium calculations not having local minima. When there are multiple minima and for three-phase equilibrium, the stochastic method, genetic algorithm (GA) followed by modified simplex method of Nelder and Mead (NM) is more reliable and desirable. Generic programs for numerical methods are ineffective for phase equilibrium calculations. These findings are of interest and value to researchers and engineers working on phase equilibrium calculations and/or developing thermodynamic models for phase behaviour.

We consider a fully connected queueing network in which customers have one direct and many alternative routes through the network, and where customer routeing is dynamic. We obtain an asymptotically optimal routeing policy, taking the limit as the number of queues of the network increases. We observe that good policies route customers directly, unless there is a danger of servers becoming idle, in which case customers should be routed alternatively so as to avoid such idleness, and this leads to policies that perform well in moderate-sized networks.

Rabies post-exposure prophylaxis (PEP) currently comprises administration of rabies vaccine together with rabies immunoglobulin (RIG) of either equine or human origin. In the developing world, RIG preparations are expensive, often in short supply, and of variable efficacy. Therefore, we are seeking to develop a monoclonal antibody cocktail to replace RIG. Here, we describe the cloning, engineering and production in plants of a candidate monoclonal antibody (E559) for inclusion in such a cocktail. The murine constant domains of E559 were replaced with human IgG1κ constant domains and the resulting chimeric mouse-human genes were cloned into plant expression vectors for stable nuclear transformation of Nicotiana tabacum. The plant-expressed, chimeric antibody was purified and biochemically characterized, was demonstrated to neutralize rabies virus in a fluorescent antibody virus neutralization assay, and conferred protection in a hamster challenge model.

To meet the requirement of the regulations and to reduce the pollution to the targeted level, minimization of emission level has been added into the dispatch strategies of generators by formulating emission constrained economic dispatch (ECED). Besides the conventional method by using the Lagrange Multiplier several evolutionary computation techniques such as Genetic Algorithm, Particle Swarm Optimisation, Ant Colony and Differential Evolution have been gaining popularity in solving general economic dispatch problems due to their desirable characteristics such as non-gradient dependent and ability to search for global optima. The effectiveness of these stochastic search techniques however is heavily dependent on the genetic operators and their parameters. The paper presents the study on sensitivity analysis of the parameters of Genetic Algorithm (GA) for the ECED problem. The results discuss the range of parameters suitable to be employed for the optimization and compare the difference between conventional economic dispatch and the ECED solutions.

Dedicated rapid access palliative radiation therapy improves patients' access to care, allowing more timely treatment which would positively impact on quality of life. The TomoTherapy (Accuray, Sunnyvale, CA) system provides megavoltage (MV) fan-beam computed tomography (FBCT) as the image guidance technique, and a module called “statRT” that allows the use of these MV FBCT images for direct planning. The possibility of using this imaging modality for palliative radiotherapy treatment planning is assessed against accepted planning CT standards by performing tests following AAPM TG 66 and an end-to-end measurement. Results have shown that MV FBCT images acquired by TomoTherapy are of sufficient quality for the purpose of target delineation and dose calculation for palliative treatments. Large image noise and extended scan acquisition time are the two main drawbacks, so this imaging modality should only be used for palliative treatments at areas with well-known, easily distinguishable, and relatively immobile targets such as spine and whole brain.

The age standardised incidence rate (ASR) of breast cancer in Malaysia which is a high middle- income country is similar to Indonesia, a low middle-income country. (Globocan 2008) It is however unknown whether the presentation of breast cancer differs between these two countries.We compared the stage, age at presentation, and pathological characteristics of breast cancer between two tertiary hospitals in Indonesia and Malaysia; Dharmais Cancer Centre (DCC), which is the national cancer referral centre in Indonesia, and University Malaya Medical Centre (UMMC), which is an academic hospital with established breast oncology services in Kuala Lumpur. One thousand, one hundred and fourteen consecutive women (477 in UMMC: 637 in DCC) who were newly diagnosed with breast cancer between January and December, 2010 were included. Patient's age, TNM stage at presentation, and pathological characteristics were compared. Estrogen receptor (ER) and progesterone receptor (PR) were considered positive if 10% or greater of invasive cell nuclei were stained while HER2 was considered positive with an immunohistochemistry staining intensity of 3+ . Logistic regression analyses were performed to identify differences.Median age at diagnosis was 52 years in UMMC and 47 years in DCC, whereby patients in DCC were more likely to be very young at diagnosis (aged35 years) compared to their counterparts in UMMC (Odds ratio (OR): 2.09; 95%CI: 1.32-3.31). Approximately one third of patients in UMMC presented with TNM stage III or IV, compared to 63% in DCC. Patients in DCC were three times more likely to present with metastatic breast cancer compared to patients in UMMC (OR: 3.01; 95% CI: 2.02-4.48). The percentage of low grade tumours in DCC was higher than in UMMC (28% vs 11% respectively), and the difference persisted even after multivariate adjustment. Although the frequency of ER and PR positivity appeared to be higher in UMMC (65% and 55% respectively) compared to DCC (48% and 40% respectively), these differences were not statistically significant following adjustment for age, stage, HER2 status and grade. The frequency of HER2 positivity was 45% in DCC compared to 26% in UMMC, and remained significantly higher even after multivariate adjustment (multivariate OR:1.76; 95%CI:1.25-2.47, in DCC compared to UMMC). The proportion of triple negative breast cancer was however similar in the two centres (19% in UMMC vs 21% in DCC).Indonesian women with breast cancer seem to present at a younger age and at later stages compared to Malaysian women. Their tumors were more likely to be of low grade and HER2 positive, even after adjustment for other factors, while hormone receptor positivity proved similar in the two groups. The higher HER2 positivity rate in Indonesian patients warrants further study.
