Abstract

A body shape index (ABSI) has been proven to be related to a population's CVD incidence. However, the application of this indicator has produced different results. This study aimed to evaluate the applicability of the ABSI in predicting the incidence of CVD in rural Xinjiang, China, and compare it with waist circumference (WC), waist-to-hip ratio (WHR), waist-to-height ratio (WHtR), and body mass index (BMI). 5375 people aged 18 years or older were included in the study. We used the Cox proportional hazard model to evaluate the relationship between WC, WHR, WHtR, BMI, and ABSI and the incidence of CVD, the area under the curve (AUC) to evaluate the predictive power of each anthropometric index for the incidence of CVD, and restricted cubic splines are used to analyse the trend relationship between anthropometric indicators and the incidence of CVD. After multivariate adjustment, standardised WC, WHR, WHtR, BMI, and ABSI all positively correlated with the incidence of CVD. WC had the highest HR (95% CI) value, 1.64 (1.51–1.78), and AUC (95% CI) value, 0.7743 (0.7537–0.7949). ABSI had the lowest HR (95% CI) value, 1.21(1.10–1.32), and AUC (95% CI) value, 0.7419 (0.7208–0.7630). In the sex-specific sensitivity analysis, the predictive ability of traditional anthropometric indicators for the incidence of CVD is higher than that of ABSI. In the rural areas of Xinjiang, the traditional anthropometric indicators of WC had better ability to predict the incidence of CVD than ABSI. [ABSTRACT FROM AUTHOR]

24. Biotin labeling and receptor analysis of huwentoxin-I

Abstract

Huwentoxin-I (HWTX-I) is the highest content peptidic neurotoxin in the crude venom of spider Selenocosmia huwena. This toxin blocks the nerve–muscle conduction of isolated phrenic nerve–hemidiaphragm preparation reversibly, with the action site in the presynaptic membrane. In order to further investigate the action mechanism of HWTX-I, the present work optimized the experimental conditions for the biotin labeling of HWTX-I and used the site-selectively biotinylated toxin to affinity purify and identify its receptor proteins from the plasma membrane in rat phrenic nerve–diaphragm synapses. The experiments demonstrated that when the biotinylation reagent reacted with HWTX-I at a molar ratio of 2 : 1 on ice for 2 h, most HWTX-I molecules were labeled by a single biotin group, with a yield of 57%. Tandem mass spectrometry and electrophysiological experiments indicated that the single biotin labeling occurred at the Lys13 residue of HWTX-I and this biotinylation did not significantly affect the bioactivity of HWTX-I. After the affinity purified protein fraction was ultrafiltered, digested with trypsin and analyzed by CapLC-MS/MS, several membrane proteins including voltage-gated calcium channel proteins that HWTX-I potentially interacted with were identified and their roles in the action mechanism of HWTX-I were discussed.

Abstract

Objective: To analyze the trend in the prevalence and mortality of prostate cancer in Shihezi, Xinjiang from 2009 to 2017 and provide some evidence for the prevention and control of the malignance., Methods: We collected the data on the cancer registries in the Shihezi area between 2009 and 2017, calculated the incidence and mortality rates of prostate cancer, and analyzed the annual percent change (APC) and prevalence trend of the disease., Results: The crude incidence rate, age-standardized incidence rate by Chinese standard population (ASIRC), age-standardized incidence rate by world standard population (ASIRW) and cumulative incidence rate of prostate cancer (in 0－74-year-olds) in Shihezi between 2009 and 2017 were 16.94, 10.33, 8.98 and 2.29 per 100 000, respectively. The crude mortality rate, age-standardized mortality rate by Chinese standard population (ASMRC), age-standardized mortality rate by world standard population (ASMRW) and cumulative incidence rate (in 0－74-year-olds) were 9.03, 5.39 and 4.72 and 0.49 per 100 000, respectively. Both the incidence and mortality rates showed an increasing trend from 2009 to 2017, with an APC of 16.69% (P < 0.05) and 19.71% (P < 0.05), respectively. From 2011 to 2017, the increase rates of incidence and mortality of prostate cancer in the >60-year-olds were 86.20% and 89.30%, with the peak values shifted from the 70－74 to the 80－84 years old males., Conclusions: The incidence and mortality of prostate cancer in Shihezi showed an increasing trend from 2009 to 2017, chiefly in the males aged over 60 years, with the peak value moving towards an older age.
