Background: Cereal crops are a primary energy source for humans. Grain size and weight affect both evolutionary fitness and grain yield of cereals. Although studies on gene mining and molecular mechanisms controlling grain size and weight are constantly emerging in cereal crops, only a few systematic reviews on the underlying molecular mechanisms and their breeding applications are available so far. Aim of review: This review provides a general state-of-the-art overview of molecular mechanisms and targeted strategies for improving grain size and weight of cereals as well as insights for future yield-improving biotechnology-assisted breeding. Key scientific concepts of review: In this review, the evolution of research on grain size and weight over the last 20 years is traced based on a bibliometric analysis of 1158 publications and the main signaling pathways and transcriptional factors involved are summarized. In addition, the roles of post-transcriptional regulation and photosynthetic product accumulation affecting grain size and weight in maize and rice are outlined. State-of-the-art strategies for discovering novel genes related to grain size and weight in maize and other cereal crops as well as advanced breeding biotechnology strategies being used for improving yield including marker-assisted selection, genomic selection, transgenic breeding, and genome editing are also discussed.

The Western Route of the South-to-North Water Diversion Project is the final link to be constructed for the four-horizontal three-vertical framework of China’s national water network. The adjustable water volume in the water source areas is a focal and challenging issue in the planning and demonstration of the Western Route. This study raised ten questions related to the adjustable water volume in the water source areas of the Western Route from the aspects of water resource background conditions and evolution trends, ecological and socio-economic water demands, scales of reservoirs and water conveyance projects, as well as impacts of water diversion on water resource development and utilization, aquatic ecological environment, hydropower, and navigation in the water source areas and their lower reaches. Based on a distributed hydrological model of the upper reaches of the Yangtze River, a simulation and analysis model for calculating the adjustable water volume in water source areas of the Western Route is established to quantitatively answer the ten questions raised. The main conclusions are as follows. First, under the premise of solely satisfying ecological and socio-economic water demands within and outside the river channels of the water source areas, a plan that combines upper and lower routes presents an average adjustable water volume of 1.59×1010 m³ over multiple years, while a plan for solely constructing a lower route can reach an average adjustable water volume of 1.74×1010 m³ by constructing the Gangtuo Reservoir for coordinated operation. Second, comprehensively considering the impacts of water diversion on water balance, hydropower, and navigation, as well as their acceptability in the water source areas, the adjustable water volume is between 1.22×1010 and 1.26×1010 m³. Third, in the long term, the plan should focus on connecting and integrating the water network in the southwestern region with the main framework and major streams of the national water network of China. This will reduce the impacts of water diversion by the Western Route Project, increase the overall water supply capacity of the project, and promote its adaptability to external conditions such as climate change.

The molecular mechanisms driving the onset and metastasis of prostate cancer remain poorly understood. Actin, under the control of actin-binding proteins (ABPs), plays a crucial role in shaping the cellular cytoskeleton, which in turn supports the morphological alterations in normal cells, as well as the invasive spread of tumor cells. Previous research indicates that ABPs of various types serve distinct functions, and any disruptions in their activities could predispose individuals to prostate cancer. These ABPs are intricately implicated in the initiation and advancement of prostate cancer through a complex array of intracellular processes, such as severing, linking, nucleating, inducing branching, assembling, facilitating actin filament elongation, terminating elongation, and promoting actin molecule aggregation. As such, this review synthesizes existing literature on several ABPs linked to prostate cancer, including cofilin, filamin A, and fascin, with the aim of shedding light on the molecular mechanisms through which ABPs influence prostate cancer development and identifying potential therapeutic targets. Ultimately, this comprehensive examination seeks to contribute to the understanding and management of prostate diseases.

Background/purpose(s)The continuously increasing carbapenem resistance within Enterobacterales and Pseudomonas poses a threat to public health, nevertheless, the molecular characteristics of which in southern China still remain limited. And carbapenemase identification is a key factor in effective early therapy of carbapenem-resistant bacteria infections. We aimed to determine the molecular characteristics of these pathogens and compare commercial combined disc tests (CDTs) with the modified carbapenem inactivation method (mCIM) and EDTA-CIM (eCIM) in detecting and distinguishing carbapenemases using whole genome sequencing (WGS).MethodsA total of 78 Enterobacterales, 30 Pseudomonas were obtained from two tertiary hospitals in southern China. Susceptibility tests were conducted using an automated VITEK2 compact system with confirmation via the Kirby–Bauer method. The WGS was conducted on all clinical isolates and the molecular characteristics were analyzed by screening the whole genome sequences. CDTs with or without cloxacillin, mCIM, and eCIM, were performed and compared by taking WGS results as the benchmark.ResultsA total of 103 carbapenem non-susceptible and 5 carbapenem susceptible bacteria were determined, with Klebsiella pneumoniae (42.7%), Pseudomonas aeruginosa (23.3%) and Escherichia coli (18.4%) being most prevalent. Carbapenemase genes were detected in 58 (56.3%) of the 103 carbapenem-non-susceptible clinical isolates, including 46 NDM, 6 KPC, 3 IMP, 1 IPM+VIM,1NDM+KPC, and 1 OXA-181. Carbapenemase-producing isolates were detected more frequently in Enterobacterales (76.3%). Among K. pneumoniae, the major sequence types were st307 and st11, while among E. coli and P. aeruginosa, the most prevalent ones were st410 and st242 respectively. For carbapenemase detection in Enterobacterales, the mCIM method achieved 100.00% (95% CI, 92.13–100.00%) sensitivity and 94.44% (70.63–99.71%) specificity (kappa, 0.96); for Pseudomonas, detection sensitivity was 100% (5.46–100.00%), and 100% (84.50–100.00%) specificity (kappa, 0.65). Commercial CDT carbapenemase detection sensitivity for Enterobacterales was 96.49% (86.84–99.39%), and 95.24% (74.13–99.75%) specificity (kappa, 0.90); for Pseudomonas, carbapenemase detection sensitivity was 100.00% (5.46–100.00%) and 37.93% (21.30–57.64%) specificity (kappa, 0.04). When cloxacillin testing was added, CDT specificity reached 84.61% (64.27–94.95%).ConclusionThe molecular epidemiology of carbapenem-non-susceptible isolates from pediatric patients in Southern China exhibited distinctive characteristics. Both the mCIM–eCIM combination and CDT methods effectively detected and differentiated carbapenemases among Enterobacterales isolates, and the former performed better than CDT among Pseudomonas.

Abstract Sparganii Rhizoma-Curcumae Rhizoma (SR-CR) is a classic drug pair for the treatment of castration-resistant prostate cancer (CRPC), but its mechanism has not been clarified. The study aims to elucidate the potential mechanism of SR-CR in the management of CRPC. The present study employed the TCMSP as well as the SwissTargetPrediction platform to retrieve the chemical composition and targets of SR-CR. The therapeutic targets of CRPC were identified through screening the GeneCards, Disgenet, and OMIM databases. Subsequently, the Venny online platform was utilized to identify the shared targets between the SR-CR and CRPC. The shared targets were enrichment analysis using the Bioconductor and Kyoto encyclopedia of genes and genomes (KEGG) databases. The active ingredients and core targets were verified through molecular docking and were validated using PC3 cells in the experimental validation phase. A total of 7 active ingredients and 1126 disease targets were screened from SR-CR, leading to a total of 59 shared targets. Gene Ontology (GO) analysis resulted in 1309 GO entries. KEGG pathways analysis yielded 121 pathways, primarily involving cancer-related signaling pathways. The results from molecular docking revealed stable binding interactions between the core ingredients and the core targets. In vitro cellular assays further demonstrated that SR-CR effectively suppressed the activation of the Prostate cancer signaling pathway in PC3 cells, leading to the inhibition of cell proliferation and promotion of apoptosis. The SR-CR exert therapeutic effects on CRPC by inhibiting cell proliferation and promoting apoptosis through the Prostate cancer signaling pathway.

Abstract Background Alzheimer’s disease (AD), affecting many elders worldwide, is characterized by A-beta and tau-related cognitive decline. Accumulating evidence suggests that brain iron accumulation is an important characteristic of AD. However, the function and mechanism of the iron-mediated gut-brain axis on AD is still unclear. Methods A Caenorhabditis elegans model with tau-overexpression and a high-Fe diet mouse model of cognitive impairment was used for probiotic function evaluation. With the use of qPCR, and immunoblotting, the probiotic regulated differential expression of AD markers and iron related transporting genes was determined. Colorimetric kits, IHC staining, and immunofluorescence have been performed to explore the probiotic mechanism on the development of gut-brain links and brain iron accumulation. Results In the present study, a high-Fe diet mouse model was used for evaluation in which cognitive impairment, higher A-beta, tau and phosphorylated (p)-tau expression, and dysfunctional phosphate distribution were observed. Considering the close crosstalk between intestine and brain, probiotics were then employed to delay the process of cognitive impairment in the HFe mouse model. Pediococcus acidilactici (PA), but not Bacillus subtilis (BN) administration in HFe-fed mice reduced brain iron accumulation, enhanced global alkaline phosphatase (AP) activity, accelerated dephosphorylation, lowered phosphate levels and increased brain urate production. In addition, because PA regulated cognitive behavior in HFe fed mice, we used the transgenic Caenorhabditis elegans with over-expressed human p-tau for model, and then PA fed worms became more active and longer lived than E.coli fed worms, as well as p-tau was down-regulated. These results suggest that brain iron accumulation influences AD risk proteins and various metabolites. Furthermore, PA was shown to reverse tau-induced pathogenesis via iron transporters and AP-urate interaction. Conclusions PA administration studies demonstrate that PA is an important mediator of tau protein reduction, p-tau expression and neurodegenerative behavior both in Caenorhabditis elegans and iron-overload mice. Finally, our results provide candidates for AP modulation strategies as preventive tools for promoting brain health. Video Abstract

Abstract Past studies have shown that the Gelsolin-like actin-capping protein (CAPG) regulates cell migration and proliferation and is strongly associated with tumor progression. We present the first study of the mechanism of action of CAPG in early gastric cancer (EGC). We demonstrate that CAPG expression is upregulated in gastric cancer (GC) especially EGC. CAPG promotes GC proliferation, migration, invasion, and metastasis in vivo and in vitro. More importantly, CAPG plays a role in GC by involving the Wnt/β-catenin signaling pathway. Our findings suggest that CAPG may function as a novel biomarker for EGC.

Objective: Prostate cancer, marked by a high incidence and mortality rate, presents a significant challenge, especially in the context of castration-resistant prostate cancer (CRPC) with limited treatment options due to drug resistance. This study aims to explore the anti-tumor effects of Xihuang Pills (XHP) on CRPC, focusing on metabolic reprogramming and the Wnt/β-catenin pathway. Methods: In vitro and in vivo biofunctional assays were employed to assess the efficacy and mechanisms of XHP. Subcutaneous xenografts of PC3 in mice served as an in vivo model to evaluate XHP's anti-tumor activity. Tumor volume, weight, proliferation, and apoptosis were monitored. Various assays, including CCK8, TUNEL assay, QRT-PCR, and Western Blotting, were conducted to measure metabolic reprogramming, proliferation, apoptosis, and cell cycle in prostate cancer cells. RNA-seq analysis predicted XHP's impact on prostate cancer, validating the expression of Wnt/β-catenin-related proteins and mRNA. Additionally, 58 compounds in XHP were identified via LC-MS/MS, and molecular docking analysis connected these compounds to key genes. Results: In vitro and in vivo experiments demonstrated that XHP significantly inhibited CRPC cell viability, induced apoptosis, and suppressed invasion and migration. mRNA sequencing revealed differentially expressed genes, with functional enrichment analysis indicating modulation of key biological processes. XHP treatment downregulated Wnt signaling pathway-related genes, including CCND2, PRKCG, and CCN4. Moreover, XHP effectively inhibited glucose uptake and lactate production, leading to reduced HIF-1α and glycolytic enzymes (GLUT1, HK2, PKM2), suggesting its potential in attenuating the Warburg effect. Molecular docking analysis suggested a plausible interaction between XHP's active compounds and Wnt1 protein, indicating a mechanism through which XHP modulates the Wnt/β-catenin pathway. Conclusion: XHP demonstrated remarkable efficacy in suppressing the growth, proliferation, apoptosis, migration, and invasiveness of prostate tumors. The interaction between XHP's active constituents and Wnt1 was evident, leading to the inhibition of Wnt1 and downstream anti-carcinogenic factors, thereby influencing the β-catenin/HIF-1α-mediated glycolysis.

Environmental exposures during pregnancy have been associated with adverse obstetric outcomes. However, limited and inconsistent evidence exists regarding the association between air temperature exposure and the risk of preeclampsia (PE). This study aimed to evaluate the correlation between ambient temperature exposure during pregnancy and PE risk, as well as identify the specific time window of temperature exposure that increases PE risk. A population-based cohort study was conducted from January 2012 to April 2022 in Guangzhou, China. Pregnant women were recruited in early pregnancy and followed until delivery. A total of 3,314 PE patients and 114,201 normal pregnancies were included. Ambient temperature exposures at different gestational weeks were recorded for each participant. Logistic regression models were used to evaluate the correlation between ambient temperature exposure and PE risk. Stratified analyses were conducted based on maternal age and pre-pregnancy BMI. Distributed lag models were employed to identify the time window of temperature exposure related to PE. Exposure to extreme high temperature (aOR = 1.24, 95 % CI 1.12–1.38) and moderate high temperature (aOR = 1.22, 95 % CI 1.10–1.35) during early pregnancy was associated with an increased risk of PE. Furthermore, women with higher pre-pregnancy BMI had a higher risk of developing PE when exposed to high temperature during early pregnancy compared to normal-weight women. The time window of temperature exposure related to PE was identified as pregnancy weeks 1 to 8. This study provides evidence for the association of high temperature exposure during early pregnancy with the risk of PE, as well as identifies the specific time window of temperature exposure related to PE. These findings have implications for developing potential strategies to protect pregnant women, particularly those with higher pre-pregnancy BMI, from the adverse effects of extreme temperatures during early pregnancy.

Long non-coding RNAs (lncRNAs) are emerging as important players in gene regulation and cardiovascular diseases. However, the roles of lncRNAs in atherosclerosis are poorly understood. In the present study, we found that the levels of NIPA1-SO were decreased while those of NIPA1 were increased in human atherosclerotic plaques. Furthermore, NIPA1-SO negatively regulated NIPA1 expression in human umbilical vein endothelial cells (HUVECs). Mechanistically, NIPA1-SO interacted with the transcription factor FUBP1 and the NIPA1 gene. The effect of NIPA1-SO on NIPA1 protein levels was reversed by the knockdown of FUBP1. NIPA1-SO overexpression increased, whilst NIPA1-SO knockdown decreased BMPR2 levels; these effects were enhanced by the knockdown of NIPA1. The overexpression of NIPA1-SO reduced while NIPA1-SO knockdown increased monocyte adhesion to HUVECs; these effects were diminished by the knockdown of BMPR2. The lentivirus-mediated-overexpression of NIPA1-SO or gene-targeted knockout of NIPA1 in low-density lipoprotein receptor-deficient mice reduced monocyte-endothelium adhesion and atherosclerotic lesion formation. Collectively, these findings revealed a novel anti-atherosclerotic role for the lncRNA NIPA1-SO and highlighted its inhibitory effects on vascular inflammation and intracellular cholesterol accumulation by binding to FUBP1 and consequently repressing NIPA1 expression.

ABSTRACTGenetically modified (GM) soybeans provide a huge amount of food for human consumption and animal feed. However, the possibility of unexpected effects of transgenesis has increased food safety concerns. High-throughput sequencing profiling provides a potential approach to directly evaluate unintended effects caused by foreign genes. In this study, we performed transcriptomic analyses to evaluate differentially expressed genes (DEGs) in individual soybean tissues, including cotyledon (C), germ (G), hypocotyl (H), and radicle (R), instead of using the whole seed, from four GM and three non-GM soybean lines. A total of 3,351 DEGs were identified among the three non-GM soybean lines. When the GM lines were compared with their non-GM parents, 1,836 to 4,551 DEGs were identified. Furthermore, Gene Ontology (GO) analysis of the DEGs showed more abundant categories of GO items (199) among non-GM lines than between GM lines and the non-GM natural varieties (166). Results of Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis showed that most KEGG pathways were the same for the two types of comparisons. The study successfully employed RNA sequencing to assess the differences in gene expression among four tissues of seven soybean varieties, and the results suggest that transgenes do not induce massive transcriptomic alterations in transgenic soybeans compared with those that exist among natural varieties. This work offers empirical evidence to investigate the genomic-level disparities induced by genetic modification in soybeans, specifically focusing on seed tissues.

To meet the demand of accurate water level prediction of the reservoir in Liuxihe River Basin, this paper proposes an improved long short-term memory (LSTM) neural network based on the Bayesian optimization algorithm and wavelet decomposition coupling. Based on the improved model, the water levels of Liuxihe Reservoir and Huanglongdai Reservoir are simulated and predicted by the 1 h prediction length, and the prediction accuracy of the improved model is verified separately by the 3, 6 and 12 h prediction lengths. The results show that: first, Bayesian optimization coupling can significantly reduce the average absolute error and root mean square error of the model and improve the overall prediction accuracy, but this algorithm is insufficient in the optimization of model extremum; Wavelet decomposition coupling can significantly reduce the outliers in model prediction and improve the accuracy of extremum, but it plays relatively weaker role in the overall optimization of the model. Second, by the prediction lengths of 1, 3, 6 and 12 h, the improved model based on the LSTM neural network and coupled with Bayesian optimization and wavelet decomposition is superior to Bayesian optimization and wavelet decomposition coupling model in overall prediction accuracy and prediction accuracy of extremum. Abstract Background Staphylococcus aureus, one of the most prevalent opportunistic pathogens, mainly colonizes the nasal cavity and is a risk factor for severe infections. Virulence factors and accessory gene regulator (agr) are key to the severity and diversity of staphylococcal infection. In this study, we aimed to characterise S. aureus agr-types and virulence genes and correlated them with genetic background and antibiotic-resistant phenotypes. Results Agr types were identified in 704 isolates (98.5%), with only 11 isolates were negative for agr type. Most of our isolates were classified as agr type I, followed by types III, II and IV. The enterotoxin c gene (sec) was detected in 48.6% of isolates, showing the highest prevalence among the five enterotoxin genes detected. The positivity rates for the lukS/F-PV and tsst genes were 4% and 2.2%, respectively, while neither sed nor SasX were detected. ST45, ST59, ST338, ST188, ST6, ST7, ST22, ST25, ST398, and ST944 belonged to agr I group, while ST5 and ST15 belonged to agr II group. ST30 and ST1 were classified into agr III group, and ST121 was assigned into agr IV group. The tsst gene was found exclusively within agr I and III types belonging to ST7 and ST30 isolates, while the lukS/F-PV was predominantly carried by agr I type isolates primarily within CC59 and CC22 clones. Among the methicillin-resistant S. aureus (MRSA) isolates, 89.7% belonged to agr I group, and 97.8% of rifampicin-resistant or intermediate isolates were assigned to agr I group. MRSA isolates harboured more tested virulence genes compared to methicillin-susceptible S. aureus isolates. Conclusions We characterized the distributions of agr types and eight major virulence genes of 715 S. aureus isolates, and our findings revealed clear associations between agr types and STs, as well as virulence genes, and drug resistant phenotypes.

Compound flooding from rainfall and storm tides is prone to occur in coastal cities. The identification of them is essential for controlling urban flooding. First, the dependence between rainfall and storm tides is quantified by Kendall's τ, Spearman's ρ, and tail dependence coefficient. Then, a bivariate copula-based probability distribution model is built to calculate the joint and conditional probability of rainfall and storm tides. Finally, MK and SQMK methods are employed to detect the trends of the dependence and joint probability. The results show that: (1) The dependence between strong rainfall and corresponding storm tides is much higher than that of small rainfall and storm tides, and the effect of tropical cyclones may be one of the reasons. (2) The dependence between rainfall and storm tides is the largest in October and the smallest in July. More attention should be paid to the compound flooding caused by rainfall and storm tides in October for Haikou. (3) The upper tail dependence coefficient of the rainfall and storm tides is significantly greater than the lower tail dependence coefficient and exhibits a significant positive trend. The results can provide additional insights into the effect of rainfall and storm tides for coastal flood management. HIGHLIGHTS The compound effects of rainfall and storm tides were investigated from different perspectives.; Trends of the compound risk were analyzed by the copula and Mann–Kendall method.; Significant increasing trends of joint probability were detected after 1988–1992.; Compound event of rainfall and storm tides is more likely to occur when a high storm tide event has occurred.;

The drainage capacity of stormwater inlets, which serve as the connection between the surface and the underground drainage system, directly affects surface runoff and the drainage capacity of underground drainage systems. However, in reality, stormwater inlets are often blocked due to the accumulation of leaves, human waste disposal and other factors, resulting in a greatly reduced drainage capacity of the drainage network and, in turn, urban waterlogging disasters. In view of the problem of stormwater inlet blockage, employing a typical waterlogging point in the Lianjiang Middle Road area of Fuzhou city as the research object, the stormwater inlet equivalent drainage method was adopted in this paper to characterize the drainage capacity of the pipe network and enable the control of the stormwater inlet blockage state. Coupled with the stormwater inlet drainage equation, an improved ITF-FLOOD two-dimensional hydrodynamic model was constructed, and the influence of stormwater inlet blockage on urban waterlogging under different rainfall return periods was simulated and analyzed. With increasing rainfall return period, the influences of stormwater inlet blockage on both the maximum area and the depth of accumulated water in the study area gradually decreased compared with those of a nonblocked stormwater inlet, and the growth proportions decreased from 43.35% and 34.58% under the 1-year rainfall scenario to 3.34% and 9.76% under the 50-year rainfall scenario, respectively. However, in terms of the change in the accumulated water level, stormwater inlet blockage will cause an increase, and the influence will always be significant. Overall, stormwater inlet blockage aggravated the waterlogging risk and the extent of waterlogging. Therefore, the results provided a reference for the construction of an urban waterlogging model and have certain guiding significance for waterlogging prevention and control in the study area prone to stormwater inlet blockage.

Helicobacter pylori ( H pylori ), a gastric bacterium, has been extensively studied for its association with gastritis, peptic ulcers, and gastric cancer. However, recent evidence suggests its potential implications beyond the stomach, linking it to other gastrointestinal malignancies, such as esophageal cancer, liver cancer, pancreatic cancer, gallbladder cancer, and colorectal cancer. In light of the expanding research landscape and the increasing interest in exploring H pylori broader role in gastrointestinal tumorigenesis, this comprehensive review aims to elucidate the relationship between H pylori and gastrointestinal tumors. This review encompasses recent epidemiological studies, research progress, and emerging perspectives, providing a comprehensive assessment of the relationship between H pylori and gastrointestinal tumors. The findings highlight the captivating world of H pylori and its intricate involvement in gastrointestinal malignancies.

Abstract It is long been suggested that one‐carbon metabolism (OCM) is associated with Alzheimer's disease (AD), whereas the potential mechanisms remain poorly understood. Taking advantage of chemical biology, that mitochondrial serine hydroxymethyltransferase (SHMT2) directly regulated the translation of ADAM metallopeptidase domain 10 (ADAM10), a therapeutic target for AD is reported. That the small‐molecule kenpaullone (KEN) promoted ADAM10 translation via the 5′ untranslated region (5′UTR) and improved cognitive functions in APP/PS1 mice is found. SHMT2, which is identified as a target gene of KEN and the 5′UTR‐interacting RNA binding protein (RBP), mediated KEN‐induced ADAM10 translation in vitro and in vivo. SHMT2 controls AD signaling pathways through binding to a large number of RNAs and enhances the 5′UTR activity of ADAM10 by direct interaction with GAGGG motif, whereas this motif affected ribosomal scanning of eukaryotic initiation factor 2 (eIF2) in the 5′UTR. Together, KEN exhibits therapeutic potential for AD by linking OCM with RNA processing, in which the metabolic enzyme SHMT2 “moonlighted” as RBP by binding to GAGGG motif and promoting the 5′UTR‐dependent ADAM10 translation initiation.

Land Use/Land Cover (LULC) is one of the most significant human variables influencing the efficiency of Ecosystem Services (ESs) in terrestrial ecosystems. Theoretical and technical assistance for regional sustainable land use planning and management, as well as ecosystem conservation and restoration, is provided by investigating the influence of changes in the LULC pattern on the efficiency of ESs. This research focuses on the interactions between socioeconomic activities and natural ecological processes in the Three Gorges Reservoir Area (TGRA). We use LULC data from the TGRA for the years 1990, 2000, 2010, and 2020. The study includes the analysis and calculation of the spatiotemporal evolution features of the current LULC pattern and the efficiency of ESs, including their spatiotemporal distribution. Considering the TGRA's national development orientation and guidance, three potential LULC patterns are constructed under various develop-ment scenarios. To calculate the efficiency of ESs, the GeoSOS-FLUS future LULC simulation model is linked, and several methodologies such as INVEST, RUSLE, and CASA are used. The goal is to investigate the influence of future changes in LULC patterns on ESs efficiency. The findings show the following: (1) From 1990 to 2020, the values of water conservation services in the TGRA decreased and subsequently increased. High-value areas are primarily located in the reservoir's centre and eastern sections, whereas low-value areas are mostly found in the western section. Soil conservation service values initially declined and later climbed. The TGRA's carbon storage services have in-creased yearly, from 552.64 g/m2 in 2000 to 615.92 g/m2 in 2020. (2) In the ecological protection scenario, carbon storage and soil erosion increased compared to the ecosystem services in 2020. The ecological system service benefits are greater when compared to the natural development scenario. (3) The four ESs show positive spatial correlations across all three scenarios, and local spatial au-tocorrelation analysis findings demonstrate that carbon storage, water yield, and habitat quality have comparable spatial distributions across all three scenarios. To some extent, high-value areas for water conservation, soil retention, carbon storage, and habitat quality overlap.

Abstract Reconfigurable modular microfluidics presents an opportunity for flexibly constructing prototypes of advanced microfluidic systems. Nevertheless, the strategy of directly integrating modules cannot easily fulfill the requirements of common applications, e.g., the incorporation of materials with biochemical compatibility and optical transparency and the execution of small batch production of disposable chips for laboratory trials and initial tests. Here, we propose a manufacturing scheme inspired by the movable type printing technique to realize 3D free-assembly modular microfluidics. Double-layer 3D microfluidic structures can be produced by replicating the assembled molds. A library of modularized molds is presented for flow control, droplet generation and manipulation and cell trapping and coculture. In addition, a variety of modularized attachments, including valves, light sources and microscopic cameras, have been developed with the capability to be mounted onto chips on demand. Microfluidic systems, including those for concentration gradient generation, droplet-based microfluidics, cell trapping and drug screening, are demonstrated. This scheme enables rapid prototyping of microfluidic systems and construction of on-chip research platforms, with the intent of achieving high efficiency of proof-of-concept tests and small batch manufacturing.

The construction of new towns is one of the main measures to evacuate urban populations and promote regional coordination and urban–rural integration in China. Mining the spatio-temporal pattern of new town hot spots based on multivariate data and analyzing the influencing factors of new town construction hot spots can provide a strategic basis for new town construction, but few researchers have extracted and analyzed the influencing factors of new town internal hot spots and their classification. In order to define the key points of Wuhan’s new town construction and promote the construction of new cities in an orderly and efficient manner, this paper first constructs a space-time cube based on the luminous remote sensing data from 2010 to 2019, extracts hot spots and emerging hot spots in Wuhan New City, selects 14 influencing factor indicators such as population density, and uses bivariate Moran’s index to analyze the influencing factors of hot spots, indicating that the number of bus stops and vegetation coverage rate are the most significant. Secondly, the disorderly multivariate logistic regression model is used to analyze the influencing factors of emerging hot spots. The results show that population density, vegetation coverage, road density, distance to water bodies, and distance to train stations are the most significant factors. Finally, based on the analysis results, some relevant suggestions for the construction of Wuhan New City are proposed, providing theoretical support for the planning and policy guidance of new cities, and offering reference for the construction of new towns in other cities, promoting the construction of high-quality cities.

Lignin is a crucial substance in the formation of the secondary cell wall in plants. It is widely distributed in various plant tissues and plays a significant role in various biological processes. However, the number of copies, characteristics, and expression patterns of genes involved in lignin biosynthesis in maize are not fully understood. In this study, bioinformatic analysis and gene expression analysis were used to discover the lignin synthetic genes, and two representative maize inbred lines were used for stem strength phenotypic analysis and gene identification. Finally, 10 gene families harboring 117 related genes involved in the lignin synthesis pathway were retrieved in the maize genome. These genes have a high number of copies and are typically clustered on chromosomes. By examining the lignin content of stems and the expression patterns of stem-specific genes in two representative maize inbred lines, we identified three potential stem lodging resistance genes and their interactions with transcription factors. This study provides a foundation for further research on the regulation of lignin biosynthesis and maize lodging resistance genes.

Protein ubiquitination is an enzymatic cascade reaction and serves as an important protein post-translational modification (PTM) that is involved in the vast majority of cellular life activities. The key enzyme in the ubiquitination process is E3 ubiquitin ligase (E3), which catalyzes the binding of ubiquitin (Ub) to the protein substrate and influences substrate specificity. In recent years, the relationship between the subfamily of neuron-expressed developmental downregulation 4 (NEDD4), which belongs to the E3 ligase system, and digestive diseases has drawn widespread attention. Numerous studies have shown that NEDD4 and NEDD4L of the NEDD4 family can regulate the digestive function, as well as a series of related physiological and pathological processes, by controlling the subsequent degradation of proteins such as PTEN, c-Myc, and P21, along with substrate ubiquitination. In this article, we reviewed the appropriate functions of NEDD4 and NEDD4L in digestive diseases including cell proliferation, invasion, metastasis, chemotherapeutic drug resistance, and multiple signaling pathways, based on the currently available research evidence for the purpose of providing new ideas for the prevention and treatment of digestive diseases.

Objective: Wolfram Syndrome (WS) is a rare autosomal recessive neurodegenerative disorder caused by mutations in WFS1 or CISD2 (WFS2). We present a rare case report of pregnancy with WFS1 spectrum disorder (WFS1-SD) in our hospital and reviewed literature to provide the management of pregnancy in these patients through multi-disciplinary cooperation. Case report: A 31-year-old (gravida 6, para 1) woman with WFS1-SD conceived naturally. During the pregnancy, she adjusted insulin intermittently to control blood glucose and monitored intraocular pressure changes under the guidance of doctors without any complications. Cesarean section was delivered at 37+4 weeks of gestation due to breech position and uterine scar and the neonatal weight was 3200 g. Apgar score 10 at 1 min, 10 at 5-min and 10 at 10 min, respectively. This rare case had a good maternal and infant outcome under multidisciplinary management. Conclusion: WS is an extremely rare disease. Limited information is available on the impact and management of WS on maternal physiologic adaptation and fetal outcome. This case provide a guide for clinicians to raise awareness of this rare disease and strengthen the management of pregnancy in these patients.

Plant cell elongation depends on well-defined gene regulations, adequate nutrients, and timely cell wall modifications. Anther size is positively correlated with the number and viability of pollen grains, while little is known about molecular mechanisms underlying anther cell elongation. Here, we found that properly activated cell elongation regulators at transcriptional levels in loss-of-function ZmMs33 mutant (ms33-6038) anthers failed to promote maize anther elongation. ZmMs33 deficiency disrupted metabolic homeostasis mainly by inhibiting both photosynthesis in anther endothecium and lipid accumulation in anther tapetum. Importantly, ms33-6038 anthers displayed ectopic, premature and excessive secondary cell wall thickening in anther middle layer, which constrained cell elongation structurally and blocked nutrient flows across different anther wall layers. The metabolic disorder was only found in ms33-6038 mutant rather than several representative male-sterility lines at transcriptional and post-translational levels. Collectively, the disordered metabolisms and blocked nutrient flows defeated the activated cell elongation regulators, and finally inhibited anther elongation and growth with a unique “idling effect” in ms33-6038 mutant.

Here, two multi-principal element alloys (MEPAs): CoCrFeNi and CoCrFeNiGd0.05 are fabricated by high-energy ball milling and spark plasma sintering. The effect of Gd on microstructure and mechanical properties of the CoCrFeNi MPEA is investigated. The addition of Gd to the CoCrFeNi base alloy leads to precipitation of a GdNi5-type phase with hexagonal structure (HS) and minor amounts of a Gd-rich oxide phase from the FCC matrix. Compared to the CoCrFeNi base alloy, the grain size of CoCrFeNiGd0.05 is smaller, and the tensile yield strength of CoCrFeNiGd0.05 significantly increases from 546 MPa to 859 MPa, along with minor sacrifices in plasticity where the elongation only slightly decreases from 20.8% to 19.4%. The enhanced strength of Gd-containing MPEA is mainly attributed to grain boundary strengthening from the ultrafine-grained microstructure and the precipitation strengthening contributed by the HS phase. Transmission electron microscopy (TEM) results demonstrate that the GdNi5-type intermetallic phase is coherent with the FCC matrix and shows the following orientation relationship: {111¯}FCC//{2¯119¯}HS. The high plasticity of the composite-structured CoCrFeNiGd0.05 alloy is attributed to the homogenous distribution of the ultrafine HS phase and the coherent HS-FCC interface facilitating dislocation slipping.

Abstract Background and Aims Recent studies have highlighted the biological significance of pyroptosis in cancer development. Nevertheless, it is still uncertain if pyroptosis also plays a part in immune modulation and the creation of the tumor microenvironment (TME). Methods The pyroptosis regulatory genes (PRGs) were comprehensively assessed in 1938 head and neck cancer samples, and systematically correlated these modification patterns with the infiltration characteristics of TME cells. The unsupervised consensus analysis method was used to identify specific pyroptosis clusters. The single‐sample gene set enrichment analysis and CIBERSOFT algorithms were used to evaluate the infiltration levels of various immune cell subsets. A principal component analysis algorithm was used to construct the pyrolysis potential index (PPI) to quantify the pyrolysis regulation patterns in head and neck squamous cell carcinoma (HNSC). Results Pyrophosphate regulatory genes (PRGs) are often upregulated in tumors due to mutations. PRGs relate to various clinical outcomes and pathways. Molecular subtyping identified pyroptosis patterns, which align with three tumor immunophenotypes: immune‐inflamed, immune‐excluded, and immune‐desert. The PPI measures pyrolysis roles, showing higher PPI in tumor samples linked to subtypes and clinical characteristics. Lower PPI correlates with longer survival, increased immune activity, more tumor mutations, high PD‐L1 expression, and mutations in significant genes like PIK3CA. Such patients also experience enhanced immune responses in immunotherapy trials. Conclusion We conducted a comprehensive examination of pyroptosis in HNSC and developed a PPI indicator that shows a strong correlation with the variety and intricacy of the TME.

Stalk lodging seriously affects yield and quality of crops, and it can be caused by several factors, such as environments, developmental stages, and internal chemical components of plant stalks. Breeding of stalk lodging-resistant varieties is thus an important task for maize breeders. To better understand the genetic basis underlying stalk lodging resistance, several methods such as quantitative trait locus (QTL) mapping and genome-wide association study (GWAS) have been used to mine potential gene resources. Based on different types of genetic populations and mapping methods, many significant loci associated with stalk lodging resistance have been identified so far. However, few work has been performed to compare and integrate these reported genetic loci. In this study, we first collected hundreds of QTLs and quantitative trait nucleotides (QTNs) related to stalk lodging traits in maize. Then we mapped and integrated the QTLs and QTNs in maize genome to identify overlapped hotspot regions. Based on the genomic confidence intervals harboring these overlapped hotspot regions, we predicted candidate genes related to stalk lodging traits. Meanwhile, we mapped reported genes to these hotspot regions. Finally, we constructed molecular regulatory networks underlying stalk lodging resistance in maize. Collectively, this study provides not only useful genetic loci for deeply exploring molecular mechanisms of stalk lodging resistance traits, but also potential candidate genes and targeted strategies for improving stalk lodging resistance to increase crop yields in future.

Ethanol fractional precipitation can initially separate polysaccharides according to the structure, which exhibits strong correlation with the biological activities. This study aimed to investigate the impact of varying ethanol concentrations on the structural characteristics, and the antitumor and antioxidant activities of polysaccharides derived from Dendrobium officinale through ethanol fractional precipitation, as well as their internal relationships. The polysaccharides acquired by absolute alcohol additions at a final liquor-ethanol volume ratio of 1:1, 1:2, and 1:4 were named DOP-1, DOP-2, and DOP-4, and the supernatant was named DOP-S. The results of the structural analysis revealed that the increase in ethanol concentrations resulted in a reduction in the molecular weights and the acetylation degree of the polysaccharides, as well as a decrease in mannose content and an increase in glucose content. In vitro experiments demonstrated that DOP-S exhibited optimal antitumor and antioxidant activities. Animal experiments further confirmed that DOP-S suppressed the growth of solid tumors significantly, enhanced lymphocytes, mediated immune ability, and improved the activity of antioxidant enzymes. These findings would establish a theoretical foundation and provide technical support for further advances and applications of polysaccharides derived from D. officinale in the fields of food and medicine.

This study investigated the impact of soil colloidal characteristics on the transfer patterns of different Cu and Cd speciation in contaminated soil treated with three different amendments: lime (L), zero-valent iron (ZVI), and attapulgite (ATP). It seeks to clarify the activation hazards and aging processes of these modifications on Cu and Cd. Compared with the control (CK), the available Cu concentrations treated with amendments reduced in the short term (6 months) by 96.49%, 5.54%, and 89.78%, respectively, and Cd declined by 55.43%, 32.31%, and 93.80%, respectively. Over a 12-year period, there was no significant change in the immobile effect with L, while Cu and Cd fell by 19.06% and 40.65% with ZVI and by 7.63% and 40.78% with ATP. Short- and long-term increases in the readily reducible iron and manganese oxide fraction of Cu and Cd were accompanied by a considerable rise in the concentrations of amorphous iron oxide in the soil and colloid after amendment treatment. This suggested that Cu and Cd were immobilized and stabilized in part by amorphous iron oxide.

Metallized polymer film is becoming increasingly common in cutting-edge technology fields, and interface adhesion is the key factor affecting service performance. In this article, we used all-wet process to prepare Ag/Polyimide composite film with ultra-high interface adhesion (Over 25 N/cm). The influence of etching and reduction on interface adhesion was analyzed using central composite experiments and response surface methodology. Microcracks on polyimide (PI) surface were the basis for the formation of mechanical interlocks. And combined with SEM, EDS, and XPS data, the effect of Ag structure and composition on interface adhesion during reduction was summarized. The role of Ag aggregations at interface was explained, and concluded the detachment of Ag aggregations during peeling and high temperature was the main reason of failure. RFID tag, Circuit, and Cu/Ag/PI film (Resistivity: 9.3 × 10−6 Ω·cm) were made, which confirmed the compatibility and usability of Ag/PI film.

Offshore wind forms (OWF), as frontier fields of new energy, are developing rapidly. However, the potential impacts of large-scale development of OWF on marine ecology remain unclear. To assess the effects of construction and operation of OWF on fishery resource, surveys were conducted in the Guishan OWF, 2021. The community structure, dominant species, species replacement ratio, resource density and spatial distribution of fishery resource both in construction, operation and surrounding areas were analyzed before and after construction of the OWF. The results showed that 84, 88, 105 and 99 species mainly including fish and crustaceans were identified in July, August, September and December, respectively. The species replacement ratios were 41.0%~93.4% between any two surveys. The Shannon-Wiener index and richness index in December were the maximum values; the highest Pielou index was in July. Compared with the baseline, the construction of OWF changed the structure of community and dominant species, decreased the fishery biodiversity and resource density. There is an artificial reef effect in the pile foundations of OWF. This study will provide the basic data and decision guidance for the long-term effects of construction and operation of OWF on fishery resource.

AIM: To evaluate the effects of virtual reality (VR) training on different parameters of vision. METHODS: Sixty individuals ranged 18-60 years old with asthenopia were randomly divided into short-term (n=40) and long-term (n=20) treatment groups. They were given a specially designed VR training device only once for 15min or 3-4 times a day for 15min each time for 1mo. The visual acuity, spherical equivalent, accommodative range, accommodative facility, pupil size, and visual fatigue were evaluated before (control) and after VR training. RESULTS: The visual acuity, accommodative range, and accommodative facility increased in subjects of the short-term treatment group, whereas their pupil size contracted significantly. No significant changes in spherical equivalent and visual fatigue were observed. The changes in distant vision and corrected visual acuity were positively correlated with those in pupil size, but not with spherical equivalent. The accommodative range and accommodative facility improved significantly in subjects of the long-term treatment group. No significant changes in visual acuity, spherical equivalent, pupil size, and visual fatigue were noted. CONCLUSION: VR training can improve the accommodative range and accommodative facility of human eyes. Although short-term VR training can transiently improve vision, which probably due to bright light adaptation, there is no evidence that it can improve myopia.

A broadband absorber with high optical transparency is proposed and realized using an optimization-designed method. The absorber’s reflection, lossy, and dielectric layers are considered for obtaining high optical transparency. First, to maintain microwave absorption, the artificial structures of the reflection layer are designed on the micron scale. Low square resistance and high optical transparency are achieved simultaneously. Then, considering the artificial structure fabrication process [fabricated on polyethylene terephthalate (PET)], high light transmittance polymethyl methacrylate (PMMA) was selected as the material of the dielectric layer. The permittivity of PMMA is similar to that of PET. The light reflection of layer-to-layer is reduced, and the light transmittance is improved. Finally, under the same microwave absorption band and absorption efficiency, the topology structures of the lossy layer are optimized to increase the duty ratio and the light transmittance. Through the above-mentioned design, high optical transparency and broadband microwave absorption are achieved simultaneously. The simulated and measured results prove that more than 90% of the absorption rate is obtained from 4.4 to 11.5 GHz (89.3%) and the average light transmittance reached 80%. The results verify the optimization-designed method’s effectiveness and provide technical support for transparent absorbers to be better applied to optical windows.

BackgroundTo determine the relationship between the number of controllable unhealthy lifestyles on the risk of the first occurrence of ischemic stroke after the disease in middle-aged and elderly people in the community, and to provide data support and basis for community physicians to guide hypertensive patients to control modifiable risk factors to prevent the first occurrence of ischemic stroke.MethodsThe relationship between the number of unhealthy lifestyles and the risk of hypertension was analyzed by binary logistic regression in 584 subjects using a medical record control study. A retrospective cohort study of 629 hypertensive patients was used to analyze the relationship between the number of unhealthy lifestyles and the risk of the first occurrence of ischemic stroke within 5 years of developing hypertensive disease using Cox proportional risk regression models.ResultsLogistic regression model analysis showed that taking an unhealthy lifestyle as a reference, the OR (95% CI) values of, 2, 3, 4 and 5 unhealthy lifestyle were 4.050 (2.595–6.324), 4 (2.251–7.108), 9.297 (3.81–22.686), and 16.806 (4.388–64.365), respectively. Cox Proportional risk regression model analysis showed that the risk of ischemic stroke within 5 years after developing hypertension was referenced to 5 unhealthy lifestyles, and the HR (95% CI) for 3, 2, and 1 unhealthy lifestyle were 0.134 (0.023–0.793), 0.118 (0.025–0.564), and 0.046 (0.008–0.256), respectively.ConclusionThe number of controllable unhealthy lifestyles in middle-aged and elderly people was positively associated with the risk of hypertension and first ischemic stroke after hypertension, and there was a dose-effect relationship between them. The risk of hypertension and first ischemic stroke within 5 years after hypertension onset increased with the number of unhealthy lifestyles.

Pre-eclampsia (PE) is a severe pregnancy disorder that poses a significant health risk to both mother and fetus, with no preventive or therapeutic measures. Our previous research suggested an association between elevated SERPINA5 levels and PE features. This study investigated whether SERPINA5 could be a potential therapeutic target for PE. We established PE-like features in pregnant rats using L-NAME (75 mg/kg/d) treatment. Adenoviruses carrying overexpressed or suppressed SERPINA5 genes were intravenously injected into these PE rats on the fifth and seventh days of pregnancy. We evaluated the rats’ systolic blood pressure, urine protein concentration, and placental and fetal metrics and histology. Placental gene expression following SERPINA5 overexpression was evaluated using mRNA sequencing. The L-NAME-induced PE rat model observed a significant increase in placental and peripheral SERPINA5 levels. The overexpression of SERPINA5 exacerbated L-NAME-induced hypertension and proteinuria in pregnant rats. A histology examination revealed a smaller placental junctional zone in L-NAME + overexpressing rats. Placental gene expression analysis in the L-NAME + overexpressing group indicated increased coagulation activation. L-NAME-induced hypertension and proteinuria were mitigated when SERPINA5 expression was suppressed. Additionally, placental development was improved in the SERPINA5-suppressed group. Our findings suggested that SERPINA5 may worsen L-NAME-induced PE-like features by promoting the activation of the coagulation cascade. Therefore, reducing SERPINA5 expression could potentially serve as a therapeutic strategy for PE.

In the context of global climate change and the water-carbon peak target, improving water security in arid regions is a persistent challenge in global water resources management. Water diversion projects can serve as an important measure to effectively alleviate the uneven distribution of water resources, achieve rational allocation and efficient utilization of water resources. However, how to achieve the maximization of comprehensive benefits during the process of water allocation is also an urgent problem that needs to be solved. This study focuses on the Middle Route Project of the South to North Water Diversion Project in China, selecting four important municipalities and provinces during 2015 to 2021, namely Beijing, Tianjin, Hebei Province, and Henan Province, based on the actual benefits of the water receiving areas of the middle line project. Nine representative indicators related to social, economic, and ecological benefits were selected to evaluate the optimal combination of water resource allocation in the water receiving areas along the central line, in order to achieve the maximum comprehensive benefits and solve the problems of high water safety guarantee requirements and difficult balanced water distribution in urban agglomerations in the water receiving areas. Through the calculation of the Markovsky theoretical model, the results show that when 79.9% of the water conveyance is used to generate social benefits, 15.8% of the water conveyance is used to generate ecological benefits, and 4.5% of the water conveyance is used to generate economic benefits, the project achieves the maximum comprehensive benefits. This computational model method can be used to provide technical support and scientific reference for the optimal allocation of water resources in cross regional water transfer projects.

Glycosyltransferase family 1 (GT1) is a large group of proteins that play critical roles in secondary metabolite biosynthesis in plants. However, the GT1 family is not well studied in maize. In this study, 107 GT1 unigenes were identified in the maize reference genome and classified into 16 groups according to their phylogenetic relationship. GT1s are unevenly distributed across all ten maize chromosomes, occurring as gene clusters in some chromosomes. Collinearity analysis revealed that gene duplication events, whole-genome or segmental duplication, and tandem duplication occurred at a similar frequency, indicating that both types of gene duplication play notable roles in the expansion of the GT1 gene family. Expression analysis showed GT1s expressing in all tissues with specific expression patterns of each GT1, suggesting that they might participate in multiple biological processes during the whole growth and development stages. Furthermore, 16 GT1s were identified to have similar expression patterns to those of anthocyanidin synthase (ANS), the critical enzyme in anthocyanin biosynthesis. Molecular docking was carried out to examine the affinity of GT1s with substrates in anthocyanin biosynthesis. This study provides valuable information on the GT1s of maize and will promote the development of research on their biological functions in the biosynthesis of other secondary metabolites.

In order to solve the problem of external corrosion of deep well casing in oil and gas fields, a new type of high-temperature-resistant zinc alloy sacrificial anode material was used. The temperature and corrosion resistance of the new anode material and TP140 casing were investigated by simulating the high-temperature working conditions of a deep well in an oil field using high-temperature and high-pressure corrosion tests and electrochemical tests. The results showed that at 100–120 °C, the corrosion rate of TP140 protected by a sacrificial anode was only one-tenth of that under unprotected conditions, and the minimum corrosion rate of TP140 protected by a sacrificial anode at 100 °C was 0.0089 mm/a. The results of the dynamic potential polarization curve showed that the corresponding corrosion current density of TP140 first increased and then decreased with the increase in temperature. The self-corrosion potential in sacrificial anode materials first increased and then decreased with the increase in temperature, and the potential difference with TP140 gradually decreased.

The immune functions of the body are intricately intertwined with the onset and advancement of tumors, and immunotherapy mediated by bioactive compounds has exhibited initial effectiveness in overcoming chemotherapy resistance and inhibiting tumor growth. However, the comprehensive interpretation of the roles played by immunologic components in the process of combating tumors remains to be elucidated. In this study, the Codonopsis pilosula glucofructan (CPG) prepared in our previous research was employed as an immunopotentiator, and the impacts of CPG on both the humoral and cellular immunity of S180 tumor-bearing mice were investigated. Results showed that CPG administration of 100 mg/kg could effectively inhibit tumor growth in mice with an inhibitory ratio of 45.37% and significantly improve the expression of Interleukin-2 (IL-2), Interferon-γ (IFN-γ), and Tumor Necrosis Factor-α (TNF-α). Additionally, CPG clearly enhanced B-cell-mediated humoral immunity and immune-cell-mediated cellular immunity, and, finally, induced S180 cell apoptosis by arresting cells in the G0/G1 phase, which might result from the IL-17 signaling pathway. These data may help to improve comprehension surrounding the roles of humoral and cellular immunity in anti-tumor immune responses.

Polycyclic aromatic hydrocarbons (PAHs) are common carcinogens. Benzo(a)pyrene is one of the most difficult high-molecular-weight (HMW) PAHs to remove. Biodegradation has become an ideal method to eliminate PAH pollutants from the environment. The existing research is mostly limited to low-molecular-weight PAHs; there is little understanding of HMW PAHs, particularly benzo(a)pyrene. Research into the biodegradation of HMW PAHs contributes to the development of microbial metabolic mechanisms and also provides new systems for environmental treatments. Pseudomonas benzopyrenica BaP3 is a highly efficient benzo(a)pyrene-degrading strain that is isolated from soil samples, but its mechanism of degradation remains unknown. In this study, we aimed to clarify the high degradation efficiency mechanism of BaP3. The genes encoding Rhd1 and Rhd2 in strain BaP3 were characterized, and the results revealed that rhd1 was the critical factor for high degradation efficiency. Molecular docking and enzyme activity determinations confirmed this conclusion. A recombinant strain that could completely mineralize benzo(a)pyrene was also proposed for the first time. We explained the mechanism of the high-efficiency benzo(a)pyrene degradation ability of BaP3 to improve understanding of the degradation mechanism of highly toxic PAHs and to provide new solutions to practical applications via synthetic biology.

The urban fringe is the transitional area from rural form to urban form, and it is also the urban space reserve land in the Territorial Spatial Plan. However, few researchers predict its overall evolution and guide the implementation of the Territorial Spatial Plan. This study attempts to explore the dynamic evolution law of urban fringe, analyze its driving factors, predict its future development, and put forward management suggestions for the implementation of the Territorial Spatial Plan. In this paper, the land use data of Wuhan in 2000, 2010 and 2020 are applied to delimit the urban fringe area of Wuhan by means of a sliding t-test. Fifteen driving factors are selected from three dimensions, natural factors, socio-economic factors and traffic accessibility, and brought into the Logistic model to explore the driving factors of its spatial evolution. The CA–Markov model is used to predict the fringe area of Wuhan in 2035. The results show that the transformation of rural hinterland into urban fringe is obviously affected by the distance from railway stations, highways, commercial centers and urban main roads. It is predicted that the outer boundary of Wuhan’s fringe area in 2035 will be basically the same as the planned urban development boundary. In order to realize the intention of land space planning, the development and construction of the northwest of the Huangpi District, the East Lake Scenic Area, and the west side of the Jiangxia District should be restricted. From the perspective of the evolution of the fringe area, this paper puts forward some management suggestions for the implementation of the Territorial Spatial Plan and makes a beneficial attempt in theory and method to understand the development characteristics of the fringe area and promote the implementation of the Territorial Spatial Plan.

PurposeSalmonella infection is a key global public health concern and has lead to an increased economic burden on society. We investigated the epidemiological characteristics and antimicrobial resistance profiles of clinically isolated Salmonella strains in Guangzhou Women and Children's Medical Center.Patients and methodsThis was a retrospective study of 1,338 Salmonella strains collected from children in Guangzhou Women and Children's Medical Center during 2016 to 2021.ResultsThe results revealed that 1,338 cases of Salmonella were mainly isolated from feces and blood samples. The age distribution was dominated by infants under 3 years old. The seasonal distribution was high in summer and autumn. 48 serotypes were detected, and S. typhimurium (78.7%) was the predominant serogroup. The results of antimicrobial susceptibility showed that the highest resistance was observed in ampicillin (84.5%), while lower resistance was observed in piperacillin/tazobactam, cefoperazone/sulbactam and ciprofloxacin. The antimicrobial resistance rate of fecal isolates was higher than that of blood isolates. The five-year average detection rate of multi-drug resistant Salmonella was 8.5% (114/1338) and the MDR rate of S. typhimurium was the lowest (6.9%; 73/1053).ConclusionWe concluded that antibacterial treatment should be carefully selected according to serotype and antimicrobial sensitivity results in children. Antimicrobial resistance monitoring for multi-drug resistant Salmonella is still required.

Microbial remediation is an eco-friendly and promising approach for the restoration of sites contaminated by petroleum hydrocarbons (PHCs). The degradation of total petroleum hydrocarbons (TPHs), semi volatile organic compounds (SVOCs) and volatile organic compounds (VOCs) of the soil samples collected from a petrochemical site by indigenous microbiome and exogenous microbes (Saccharomyces cerevisiae ATCC 204508/S288c, Candida utilis AS2.281, Rhodotorula benthica CBS9124, Lactobacillus plantarum S1L6, Bacillus thuringiensis GDMCC1.817) was evaluated. Community structure and function of soil microbiome and the mechanism involved in degradation were also revealed. After bioremediation for two weeks, the concentration of TPHs in soil samples was reduced from 17,800 to 13,100 mg/kg. The biodegradation efficiencies of naphthalene, benzo[a]anthracene, benzo[b]fluoranthene, benzo[a]pyrene, indeno[1,2,3-cd]pyrene, dibenzo[a,h]anthracene, 1,2,3-trichloropropane, 1,2-dichloropropane, ethylbenzene and benzene in soil samples with the addition of S. cerevisiae were 38.0%, 35.7%, 36.2%, 40.4%, 33.6%, 36.2%, 12.0%, 43.9%, 43.3% and 43.0%, respectively. The microbial diversity and community structure were improved during the biodegradation process. S. cerevisiae supplemented soil samples exhibited the highest relative abundance of the genus Acinetobacter for bacteria and Saccharomyces for yeast. The findings offer insight into the correlation between microbes and the degradation of PHC-based pollutants during the bioremediation process.

The mitotic regulator, Aurora kinase B (AURKB), is frequently overexpressed in malignancy and is a target for therapeutic intervention. The compound, LXY18, is a potent, orally available small molecule that inhibits the proper localization of AURKB during late mitosis, without affecting its kinase activity. In this study, we demonstrate that LXY18 elicits apoptosis in cancer cells derived from various indications, but not in non-transformed cell lines. The apoptosis is p53-independent, triggered by a prolonged mitotic arrest and occurs predominantly in mitosis. Some additional cells succumb post-mitotic slippage. We also demonstrate that cancer cell lines refractory to AURKB kinase inhibitors are sensitive to LXY18. The mitotic proteins MKLP2, NEK6, NEK7 and NEK9 are known regulators of AURKB localization during the onset of anaphase. LXY18 fails to inhibit the catalytic activity of these AURKB localization factors. Overall, our findings suggest a novel activity for LXY18 that produces a prolonged mitotic arrest and lethality in cancer cells, leaving non-transformed cells healthy. This new activity suggests that the compound may be a promising drug candidate for cancer treatment and that it can also be used as a tool compound to further dissect the regulatory network controlling AURKB localization.

Background. Carbapenem-resistant Enterobacteriaceae (CRE) are spreading worldwide, posing a serious public health concern. However, the data on CRE strains that cause infections in children in Guangzhou remain limited. Therefore, this study aimed to investigate the epidemiology of CRE, drug resistance, and resistance mechanisms in children in Guangzhou, Southern China. Methods. In total, 54 nonrepetitive CRE strains were collected in pediatric patients at three centers in Guangzhou, Southern China, from January 2016 to August 2018. CRE isolates were used for further studies on antimicrobial susceptibility, the modified Hodge test (MHT), the modified carbapenem inactivation method (mCIM), and drug resistance genes. Multilocus sequence typing (MLST) was used to elucidate the molecular epidemiology of K. pneumoniae. Results. The isolated CRE strains include 34 K. pneumoniae (63.0%), 10 E. coli (18.5%), 4 Enterobacter cloacae (7.4%), and 6 Proteus mirabilis (11.1%) strains. The strains were isolated mainly from the blood (31.5%, n = 17), sputum (31.5%, n = 17), and urine (16.7%, n = 9). All CRE isolates were highly resistant to the third- or fourth-generation cephalosporins, carbapenems, and β-lactam + β-lactamase inhibitors (94.4%–96.3%). In addition, the resistance rates to amikacin, ciprofloxacin, levofloxacin, tigecycline, and colistin were 5.6%, 14.8%, 16.7%, 9.3%, and 0%, respectively. Carbapenemase was detected in 35 (64.8%) of the CRE isolates. The most dominant carbapenemase gene was blaNDM (n = 17, 48.6%), followed by blaIMP (n = 13, 37.1%) and blaOXA-23 (n = 4, 11.4%). Other carbapenemase genes (blaKPC, blasim, blaAim, blaGES, blaGim, blaOXA-2, and blaOXA-48) and the mcr-1 gene were not detected. MLST analysis showed high diversity among the K. pneumoniae, and ST45 (11.7%, 4/34) was the dominant sequence type. Conclusion. This study revealed blaNDM was the most dominant carbapenemase gene in children in Guangzhou. Infection control measures need to be taken for the prevention and treatment of CRE infections.

The formation and transformation of secondary iron (oxyhydr)oxides and their role in the stabilization of copper (Cu) and zinc (Zn) in acid mine drainage (AMD) after limestone treatment are worth studying to better understand the impacts of limestone AMD treatment. In this study, the wastewater from a copper mine ditch was sampled. Two different doses of limestone (S: 5.33 g L−1 and SS: 8.00 g L−1) were applied to adjust the pH range of the sampled AMD. The concentrations of Fe, Cu and Zn in the supernatant and the levels of iron (oxyhydr)oxides and heavy metals in AMD sediments were dynamically monitored for 300 days to analyze the transformation of the secondary iron mineral phase and the role iron (oxyhydr)oxides play in the removal and stabilization of Cu and Zn. The results showed that the pH rose rapidly to 6.82, decreased to 5.82 on the 150th day, and finally decreased to approximately 4.63 by the 300th day, when the dosage of limestone (S) was 5.33 g L−1. Goethite was the main form of iron oxides in the sediments. As the incubation time increased, so did the content of crystalline Fe (oxyhydr)oxides. In addition, the Cu and Zn content in the fraction of crystalline Fe (oxyhydr)oxides increased as the corresponding iron (oxyhydr)oxide increased. When the high dosage of limestone (8.00 g L−1 or SS) was applied, the pH remained at approximately at 7.46 during the whole period and goethite and lepidocrocite were present in the sediment. Amorphous/ poorly crystalline Fe-oxyhydroxide was the main product after SS limestone dosage, indicating that the risk of Cu and Zn reactivation in the sediment was higher with a higher limestone treatment dosage.

Drought has become one of the main challenges facing global agricultural production and crop safety. Drought stress will lead to the termination of crop photosynthesis, which will seriously affect the growth and development of crops. We aimed to study a method for identificaton of the drought stress in tomato seedlings using chlorophyll fluorescence imaging. In this study, chlorophyll fluorescence parameters and there corresponding chlorophyll fluorescence images of 4 different drought stress levels were collected. Then three feature optimization algorithms which were Successive Projections Algorithm, Iteratively Retains Informative Variables and Variable Iterative Space Shrinkage Approac were used to choose important parameters. A total of five common parameters were obtained, and the corresponding chlorophyll fluorescence images of the five common parameters were selected. And two types of image features were used to study and analyze drought stress classes: histogram features and texture features. The Pearson correlations of the features were calculated and the high correlated features were input into three models, which were Linear Discriminant Analysis (LDA), Support Vector Machines (SVM) and k-Nearest Neighbor (KNN), to identify drought stress classes. The recognition accuracy rate of LDA, SVM and KNN were 86.8%, 87.1% and 76.5% respectively. Our experiment results showed that the five common fluorescence parameters and there corresponding image features could be used to evaluate the drought stress classes of tomato seedlings, and had a good evaluation effect. This research provideed a new method for monitoring drought stress classes and had considerable prospects for non-destructive diagnosis of plant drought stress.
